Abstract

International audience; “FruitBreedomics” European project has been designed by an international consortium including scientists, stakeholders and breeding companies. One of its aims is to increase the efficiency of breeding programs in apple and peach by developing Marker Assisted Breeding (MAB) using novel genomic tools. In the frame of this project, six peach mapping populations (two intraspecific F2, two interspecific F1 derived from P. davidiana, one self pollinated derived from Bolinha and one advanced pseudo-backcross) segregating for mendelian traits and/or Quantitative Trait Loci (QTLs) for pest resistance and fruit quality were genotyped using an Infinium II Illumina bead-chip containing 8144 SNPs derived from the sequencing of the peach genome. Mapping results were contrasted depending on the population considered. Relevancy of using general medium-range SNP bead- chips for breeding purposes is discussed in this study

Articles correspondant à plusieurs colloques organisés dans le cadre des séminaires du CTPS en 2012.; National audience; The Plum pox virus (PPV), the causal agent of the sharka disease, is the most detrimental virus on stone-fruit trees, worldwide. Infected fruits are not marketable. To date, no stone fruit cultivar (peach, apricot, plum) is resistant. However, sources of resistance have been identified and mapped in apricot and Prunus davidiana, a wild peach-related species. Several of the mapped QTL co-localize with candidate genes previously identified. In the present study, resistance in P. davidiana was confirmed in an F2 population and two new QTL were identified. Quantitative analysis of the apricot cultivars ‘Goldrich’ and ‘SEO’ resistance was also performed. Molecular markers targeting these genes were developed as a tool for marker-assisted selection. This study made it also possible to refine boundaries of the genomic region controlling PPV resistance as well as confirming the robustness and stability of the resistance trait in both species: P. davidiana and P. armeniaca.; La sharka, maladie provoquée par le Plum Pox Virus (PPV), affecte gravement les arbres fruitiers à noyaux du genre Prunus. Le PPV est largement répandu en Europe de l’Est et dans les régions méditerranéennes. Il affecte, entre autre, fortement la qualité des fruits, les rendant impropres à la commercialisation. A ce jour, aucun cultivar de pêchers n’est résistant et la majorité des variétés cultivées d'abricotiers sont également sensibles. Cependant, des sources de résistance ont été identifiées chez Prunus davidiana, espèce apparentée au pêcher et chez quelques cultivars d’abricotiers. L’étude de la ségrégation de ce caractère dans des populations F1 et F2 a permis d’identifier plusieurs QTL chez chacune de ces espèces et de développer des marqueurs moléculaires utilisables en Sélection Assistée par Marqueurs. De plus, nous avons pu vérifier la robustesse de ces QTL dans différents fonds génétiques et la stabilité de la résistance selon l’inoculum et le type d’inoculations utilisés.

Abstract

National audience; In spite of the worldwide decline in peach production, a constant stream of new varieties are being provide to fruit growers. For the greater part most of these new varieties being produced by private peach breeders, and as a consequence very few are selected on the basis of their resistance to pests or diseases, while the demand of consumers continues to be directed towards a quality fruit product which is free of pesticide residues. Within the framework of one INRA multidisciplinary group (Avignon-Bordeaux), we have developed for several years a wide applied breeding program aiming at improving the resistance of the peach tree to three of its main enemies: the green peach aphid (Myzus persicae), peach powdery mildew (Sphaerotheca pannosa var. persicae) and sharka disease (Plum Pox Virus). This work is globally conducted in a research context oriented towards varietal innovation including fruit quality and durable resistances building. In this way, two complementary approaches have been preferentially held for respectively improving the genetic gain by time unit and a better understanding of peach-enemies relationships. First, a genetic approach integrating the quantitative trait loci or major genes mapping for the development of molecular assisted selection. Second, a functional approach (i) leaning on the study of the insect behaviour and the plant metabolites involved in the resistance to M. persicae (ii) coupled to a candidate-genes research mainly developped for PPV resistance. Whole of first results and perspectives are discussed.

Abstract

National audience; Integrated production of fruit and vegetable species is one major field of research at Avignon INRA centre. Breeding for durable resistance to the green peach aphid, Myzus persicae (Sulzer) and other enemies of peach crop (Prunus persica L. Batsch) has been initiated fifteen years ago. Several sources of resistance are available among wild and domesticated peaches. Because all resistance genitors used in these programmes are of poor agronomic value, improvement of fruit quality is a long-lasting process. In order to avoid the erosion of the level of plant resistance during this process, the detailed analysis of the underlying mechanisms was undertaken. Here, we present some aspects of the work carried out on the genetic and physiological mechanisms of peach resistance to M. persicae. In addition to the analysis of the plant-aphid interaction under the influence of plant genetic factors, the effects of technical operations, such as winter pruning or nitrogen fertilization, on the aphid population dynamics are being investigated. Varietal resistance combined to cultural practices can be considered as an ultimate strategy to increase the efficacy of aphid control in orchards.

National audience; Les bases fonctionnelles des interactions entre plante et insectes piqueurs suceurs tels les pucerons restent mal connues malgré l’impact de ces ravageurs en agriculture. Dans le cadre de la caractérisation fonctionnelle de l’interaction entre le puceron Myzus persicae, espèce modèle en écologie, et son hôte primaire, le pêcher (Prunus persica L. Batsch), espèce modèle pour l’étude des Rosacées, nous avons mis en oeuvre une démarche d’analyse du métabolome sur les apex de pêcher, site d’alimentation privilégié de l’insecte. Nous avons étudié, par spectroscopie de RMN du proton et analyses HPLC ciblées sur les principaux pools de métabolites primaires et secondaires hydrosolubles, les modifications de profil métabolique induites par l’infestation dans les apex de deux variétés de pêcher, sensible (GF305) ou résistante (Rubira) à M. persicae [1,2]. L’analyse en composante principale de la signature spectrale obtenue par RMN et les analyses en HPLC révèlent de profondes modifications des profils métaboliques chez la variété résistante 48 heures après l’infestation : on observe notamment une baisse de la teneur de la plupart des sucres et acides organiques, de certains acides aminés (glutamine, proline, thréonine) alors que les teneurs en acides aminés ramifiés (leucine, isoleucine, valine) et aromatiques (phénylalanine, tyrosine) et en métabolites secondaires (composés phénoliques et cyanogéniques) augmentent. Les modifications observées chez la variété sensible sont faibles et pour la plupart divergent de celles observées chez la variété résistante. Ces résultats permettent d’émettre des hypothèses sur les voies métaboliques impliquées dans la réponse à l’infestation et sur la nature des effecteurs pouvant jouer un rôle dans la résistance induite chez Rubira. Cette étude met en lumière l’apport de l’approche métabolomique pour l’étude des interactions plante – insectes piqueurs suceurs.
