This interventional clinical trial aimed to assess the potential impact of Spirulina platensis supplementation on pregnant and lactating ewes living in a Moroccan endemic fluorosis area. Forty-eight ewes were divided into four equal groups: Groups I and II served as controls belonging respectively to fluorosis-free and endemic fluorosis areas, Groups III and IV received respectively 250 and 500 mg*kg−1 BW/day of S. platensis , during late pregnancy and early lactation. The results revealed that ewes reared in fluorosis-free areas exhibited significantly lower plasma fluoride and significantly higher haemoglobin levels compared to endemic fluorosis areas (P < 0.0001). However, supplementation with 500 mg*kg−1 BW*day−1 of S. platensis significantly improved these two parameters compared to Group II (P < 0.0001). Ewes in the endemic area also displayed increased oxidative stress (P < 0.05), characterized by decreased ascorbic acid levels and catalase activity, as well as elevated levels of reduced glutathione and malondialdehyde. Supplementation with 500 mg*kg−1 BW*day−1 of S. platensis enhanced the antioxidant status (P < 0.05) by increasing ascorbic acid levels and catalase activity and decreasing levels of reduced glutathione and malondialdehyde. Moreover, this dose yielded similar average daily gains compared to lambs of ewes living in fluorosis-free area. Context: The increasing intensification of pasture-based systems has subjected the cows to different productive environments, which could affect physiological responses and, consequently, their productive performance. Aims: The aim was to determine the effect of contrasting housing facilities (outdoor soil-bedded vs compost-bedded pack barn) used in partial confinement in pasture-based systems, on the metabolic adaptation during early lactation and its impact on productive and reproductive performance of autumn- and spring-calving dairy cows, having as a positive control a zero-grazing confined system in compost barn. Methods: Multiparous Holstein dairy cows that calved in autumn (n = 36) and spring (n = 48) were distributed in the following three treatments from calving to 90 days in milk (DIM): outdoor soil-bedded–grazing (OD-GRZ), compost barn–grazing (CB-GRZ), compost barn–total mixed ration (CB-TMR). Milk production, total milksolids (TMS), body condition score, non-esterified fatty acids (NEFA), beta-hydroxybutyrate (BHB), cholesterol, insulin, insulin-like growth factor-1 (IGF-1), and the proportion of cows with corpus luteum were determined in repeated measurements. Key results: In both calving seasons, milk production and TMS yields did not differ between OD-GRZ and CB-GRZ, but were greater in CB-TMR (P < 0.0001). In autumn, NEFA and BHB were not affected by treatments, but cholesterol increased faster in CB-TMR (P = 0.0500). In spring, NEFA and cholesterol concentrations were not affected by treatment, but BHB remained greater in OD-GRZ until 90 DIM than in the other treatments. IGF-1 and insulin did not differ between treatments in autumn, but in spring, IGF-1 was greater in CB-TMR (P < 0.0001). The proportion of cows with corpus luteum was not different between pasture-based treatments, but both were greater than CB-TMR up to 40 DIM in autumn (P = 0.0489) and during the entire study in spring CB-TMR (P = 0.0285). Conclusions: Although no differences were found among housing facilities in partial confinement, except that in spring OD-GRZ cows had greater BHB concentrations, confined cows prioritised milk production instead of reproductive function, despite presenting better energy indicators than did pasture-based cows. Implications: Outdoor housing combined with grazing can increase the risk of greater BHB concentrations, indicative of subclinical ketosis, under heat-stress conditions, when compared with indoor housing. Confined cows increased milk production but had a delay in the resumption of ovarian cyclicity, even having a better energy status than pasture-based cows. The thermal stress adversely affects crossbred dairy cattle production, especially in a hot, humid tropical state like Kerala. So, assessment of the intensity of stress and its relationship with the physiological alterations are crucial in adopting proper mitigation strategies to attain sustainable production. Hence this study was formulated to investigate the impact of heat stress on physio-biochemical parameters during early lactation of crossbred dairy cattle. The study comprised two sets of animals: six animals calved in month of December (control, season I) and six animals calved in the month of March (heat stressed,season II). Significant elevations (p < 0.05) in all the climatic and physiological parameters were noted in the second season. The heat stress did not result in significant differences (p > 0.05)in various blood and serum biochemical parameters such as potassium (K+), bicarbonate (HCO3−), calcium (Ca2+), phosphorus (P), Globulin, total erythrocyte count (TEC), Total leucocyte count (TLC), haemoglobin (Hb), the volume of packed red cells (VPRC), aspartate aminotransferase (AST), glucose, β hydroxyl butyrate (BHB), non-esterified fatty acid (NEFA) and triglyceride (TG).Significant differences (p < 0.05) were noted in sodium (Na+) on day zero postpartum and chloride (Cl−)on day 45 postpartum. Furthermore, there was a significant decrease (P < 0.05) in partial pressure of carbon dioxide (PCO2), gamma-glutamyl transferase (GGT), total protein, albumin, triiodothyronine (T3) and thyroxine (T4) during heat stressed period while cortisol and partial pressure of oxygen (PO2) levels increased. This study also revealed a highly significant reduction (p < 0.01) in milk yield, indicating the animals maintained their positive energy balance at the expense of milk production during the study period. ABSTRACT: In a previously established animal model, 38 multiparous Holstein cows were assigned to 2 groups fed different diets to achieve either a normal (NBCS) or high (HBCS) body condition score (BCS) and backfat thickness (BFT) until dry-off at −49 d before calving (NBCS: BCS 3.75 [3.82 ± 0.33] and BFT >1.4 cm [2.36 ± 0.35], mean ± SD). The groups were also stratified for comparable milk yields (NBCS: 10,361 ± 302 kg; HBCS: 10,315 ± 437 kg; mean ± SD). The cows were then fed the same diet during the dry period and subsequent lactation, maintaining the differences in BFT and BCS throughout the study. Using the serum metabolomics data, we created a classification model that identified different metabotypes. Machine learning classifiers revealed a distinct cluster labeled HBCS-PN (HBCS predicted normal BCS) among over-conditioned cows. These cows showed higher feed intake and better energy balance than the HBCS-PH (high BCS predicted high BCS) group, while milk yield was similar. The aim of this study was to investigate the changes in the hepatic transcriptome of cows differing in serum-metabotype postpartum. We performed hepatic transcriptome analysis in cows from 3 metabolic clusters: HBCS-PH (n = 8), HBCS-PN (n = 6), and normal BCS predicted normal BCS (NBCS-PN, n = 8) on d 21 (±2) postpartum. Liver tissue from cows expressed a total of 13,118 genes aligned with the bovine genome. A total of 48 differentially expressed genes (DEG; false discovery rate ≤0.1 and fold-change >1.5) were found between NBCS-PN and HBCS-PH cows, whereas 24 DEG (14 downregulated and 10 upregulated) were found between HBCS-PN and HBCS-PH cows. The downregulated DEG (n = 31) in NBCS-PN cows compared with HBCS-PH cows are involved in biosynthetic processes such as lipid, lipoprotein, and cholesterol synthesis (e.g., APOA1, MKX, RPL3L, CANT1, CHPF, FUT1, ZNF696), cell organization, biogenesis, and localization (e.g., SLC12A8, APOA1, BRME1, RPL3L, STAG3, FBXW5, TMEM120A, SLC16A5, FGF21), catabolic processes (e.g., BREH1, MIOX, APOBEC2, FBXW5, NUDT16), and response to external stimuli (e.g., APOA1, FGF21, TMEM120A, FNDC4), whereas upregulated DEG (n = 17) are related to signal transduction and cell motility (e.g., RASSF2, ASPN, SGK1, KIF7, ZEB2, MAOA, ACKR4, TCAF1), suggesting altered metabolic adaptations during lactation. Our results showed 24 DEG between HBCS-PN and HBCS-PH in the liver. The expression of SLC12A8, SLC16A5, FBXW5, OSGIN1, LAMA3, KDELR3, OR4X17, and INHBE, which are responsible for regulating cellular processes was downregulated in HBCS-PN cows compared with HBCS-PH cows. In particular, the downregulation of SLC12A8 and SLC16A5 expression in HBCS-PN cows indicates lower metabolic load and reduced need for NAD+ biosynthesis to support mitochondrial respiratory processes. The upregulation of MAOA, ACKR4, KIF27, SFRP1, and CAV2 in the liver of HBCS-PN cows may indicate adaptive mechanisms to maintain normal liver function in response to increased metabolic demands from over-conditioning. These molecular differences underscore the existence of distinct metabolic types in cows and provide evidence for the role of the liver in shaping different metabolic patterns.

ABSTRACT: The availability of certain macronutrients is likely to influence the capacity of the immune system. Therefore, we investigated the acute phase response to intramammary (i.mam.) lipopolysaccharide (LPS) in dairy cows fed a nitrogenic diet (n = 10) high in crude protein, a glucogenic diet (n = 11) high in carbohydrates and glucogenic precursors, or a lipogenic diet (n = 11) high in lipids. Thirty-two dairy cows were fed one of the dietary concentrates directly after calving until the end of trial at 27 ± 3 days in milk (mean ± standard deviation). In wk 3 of lactation, 20 µg of LPS was i.mam. injected in one quarter, and sterile NaCl (0.9%) in the contralateral quarter. Milk samples of the LPS-challenged and control quarter were taken hourly from before (0 h) until 9 h after LPS challenge and analyzed for milk amyloid A (MAA), haptoglobin (HP), and IL-8. In addition, blood samples were taken in the morning, and composite milk samples at morning and evening milkings, from 1 d before until 3 d after LPS challenge, and again on d 9, to determine serum amyloid A (SAA) and HP in blood, and MAA and HP in milk. The mRNA abundance of various immunological and metabolic factors in blood leukocytes was quantified by quantitative reverse-transcription PCR from samples taken at −18, −1, 6, 9, and 23 h relative to LPS application. The dietary concentrates did not affect any of the parameters in blood, milk, and leukocytes. The IL-8 was increased from 2 h, HP from 2 to 3 h, and MAA from 6 h relative to the LPS administration in the milk of the challenged quarter and remained elevated until 9 h. The MAA and HP were also increased at 9 h after LPS challenge in whole-udder composite milk, whereas HP and SAA in blood were increased only after 23 h. All 4 parameters were decreased again on d 9. Similar for all groups, the mRNA abundance of HP and the heat shock protein family A increased after the LPS challenge, whereas the mRNA expression of the tumor necrosis factor α and the leukocyte integrin β 2 subunit (CD18) were decreased at 6 h after LPS challenge. The glucose transporter (GLUT)1 mRNA abundance decreased after LPS, whereas that of the GLUT3 increased, and that of the GLUT4 was not detectable. The mRNA abundance of GAPDH was increased at 9 h after LPS and remained elevated. The acute phase protein response was detected earlier in milk compared with blood indicating mammary production. However, immunological responses to LPS were not affected by the availability of specific macronutrients provided by the different diets.

The aim of this study was to evaluate the association between time and amplitude of elevated milk BHB (EMB) occurring within 42 d in milk (DIM) and subsequent reproductive performance of lactating Holstein cows. ABSTRACT: Early lactation metabolic imbalance is an important physiological change affecting the health, production, and reproduction of dairy cows. The aims of this study were (1) to evaluate the potential of test-day (TD) variables with or without milk fatty acids (FA) content to classify metabolically imbalanced cows and (2) to evaluate the robustness of the metabolic classification with external data. A data set was compiled from 3 experiments containing plasma β-hydroxybutyrate, nonesterified FA, glucose, insulin-like growth factor-I, FA proportions in milk fat, and TD variables collected from 244 lactations in wk 2 after calving. Based on the plasma metabolites, 3 metabolic clusters were identified using fuzzy c-means clustering and the probabilistic membership value of each cow to the 3 clusters was determined. Comparing the mean concentration of the plasma metabolites, the clusters were differentiated into metabolically imbalanced, moderately impacted, and balanced. Following this, the 2 metabolic status groups identified were imbalanced cows (n = 42), which were separated from what we refer to as “others” (n = 202) based on the membership value of each cow for the imbalanced cluster using a threshold of 0.5. The following 2 FA data sets were composed: (1) FA (groups) having high prediction accuracy by Fourier-transform infrared spectroscopy and, thus, have practical significance, and (2) FA (groups) formerly identified as associated with metabolic changes in early lactation. Metabolic status prediction models were built using FA alone or combined with TD variables as predictors of metabolic groups. Comparison was made among models and external evaluations were performed using an independent data set of 115 lactations. The area under the receiver operating characteristics curve of the models was between 75 and 91%, indicating their moderate to high accuracy as a diagnostic test for metabolic imbalance. The addition of FA groups to the TD models enhanced the accuracy of the models. Models with FA and TD variables showed high sensitivities (80–88%). Specificities of these models (73–79%) were also moderate and acceptable. The accuracy of the FA models on the external data set was high (area under the receiver operating characteristics curve between 76 and 84). The persistently good performance of models with Fourier-transform infrared spectroscopy-quantifiable FA on the external data set showed their robustness and potential for routine screening of metabolically imbalanced cows in early lactation.

Simple Summary: Early lactation health disorders and their treatment are an important cause of welfare issues and production losses in dairy herds. These losses could be mitigated if cows at greater risk of health problems can be identified prior to calving. Cow mounted devices designed to monitor certain aspects of cow behavior may offer the opportunity to predict prior to calving which cows are likely to be at greater risk post calving by providing an alert based on each cow's behavior. This study attempted to determine the level and accuracy of alerts provided by one such monitoring system. Results revealed an alert level of 38.7% with an accuracy of prediction of 62.5%. These findings reflect the level of early health disorders observed in commercial dairy herds. Further research is required to improve the accuracy of prediction for cows receiving treatments for ill health and to develop effective measures to prevent the occurrence of those treatments. Identifying cows that are at greater risk for disease prior to calving would be a valuable addition to transition management. Prior to the commercial release of software features in an automated behavioral monitoring system, designed to identify cows in the dry period at greater risk of disease postpartum, a retrospective analysis was carried out in five dairy herds to evaluate whether the software could identify prepartum cows that subsequently received health treatments postpartum and whether prepartum alerts (transition alerts) are associated with a reduction in milk production in the subsequent lactation. Herd management and production records were analyzed for cows receiving treatment in the first 21 d of lactation (days in milk, DIM) for clinical mastitis, reproductive tract disease (metritis, retained fetal membranes), metabolic disease (hypocalcemia, ketosis and displaced abomasum) and for cows exiting the herd by 60 DIM. Data was gathered for 986 cows, 382 (38.7%) of which received a transition alert and 604 (61.3%) that did not. During the first 21 DIM 312 (31.6%) cows went on to receive a disease treatment, of these 51.9% (n = 162/312) were transition alert cows and 48.1% (n = 150/312) non-transition alert cows, while 8.6% (n = 33/382) alert cows exited the herd by 60 DIM compared to 4.8% (n = 29/604) of cows that did not receive an alert. A cow receiving a transition alert (OR = 1.76, 95% confidence interval (CI) = 1.27–2.44) and increasing parity (OR = 2.03, 95% CI = 1.44–2.86) were both associated with increased risk of receiving a disease treatment in the first 21 DIM. The occurrence of a transition alert was negatively associated with both week 4 milk yield (daily average yield in fourth week of lactation) and predicted 305 d yield. In order to examine the effects of feeding different levels of lysophospholipid (LPL) on early lactating Holstein cows, 15 cows were assigned in a completely randomized design with three experimental treatments including: 1) control ration; 2 and 3) control ration plus 0.1% and 0.15% of dry matter (DM) LPL, respectively. The experiment was conducted for 35 days (14 days for adaptation and 21 days for sample collection). Cows fed twice a day at 7:00 and 16:00 and received total mix ration (TMR). Daily feed intake and milk yield were measured. At the end of the experiment (day 35), the blood samples and rumen liquor were collected from each cow. The supplementation of LPL had no effect on DM intake and digestibility but increased digestibility of fat (P=0.0346). Addition of LPL to the diet slightly improved milk production (P=0.0673). The level of alanine aminotransferase (ALT) enzyme decreased with increasing LPL (P=0.0324) but activity of carboxymethyl cellulase enzyme activity increased (P=0.0421). Increasing LPL in the ration increased the amount of acetate (P=0.0452) and valerate (P=0.0033) produced in the rumen. The LPL significantly increased the population of cellulolytic bacteria (P=0.0386). In general, supplementing the ration of lactating cows with LPL, especially in the early lactation, increased feed efficiency in a dose-response manner, milk production and improved serum parameters, maintains liver health, improved ruminal bacterial population and fermentation parameters, and increased ruminal and hepatic enzymes activity. It has been demonstrated that canola meal (CM) is used more efficiently than soybean meal (SBM) by lactating dairy cows. This study was conducted to determine the effects of replacing SBM with CM on production performance, blood metabolites, and ruminal fermentation parameters of high-producing dairy cows in early lactation. Twelve lactating Holstein cows (4 primiparous and 8 multiparous; 36 ± 8 days in milk) were used in a 3 × 3 Latin square design with three treatments and four replicates (cows) over three 21-day periods. Cows were fed diets formulated with incremental amounts of CM as 0% (CM-0), 50% (CM-50), or 100% (CM-100) of SBM replaced by CM. Experimental diets were formulated to be isonitrogenous (16.8% crude protein) and isoenergetic (NEL = 1.65 Mcal/kg DM). Dry matter intake (23.2 kg/d), milk composition, energy-corrected milk production (34.5 kg/d), total VFA, and blood metabolites were not affected by treatment. However, cows fed CM diets produced an average 2.9 kg/d more milk than control cows (CM-0). Apparent N efficiency was greatest in cows fed CM-50. Compared with the control, crude protein digestibility and ruminal ammonia concentration were greater and lower for cows fed CM-50 or CM-100, respectively. Enzyme adenosine deaminase (ADA) is a marker of inflammation in domestic animals, but it is unclear whether it is a reliable marker of oxidative stress, especially in the transition period in dairy cows. This study aims to assess if ADA and redox status measurements in saliva provide the same utility to detect disease condition as that obtained from serum. Sixty-eight multiparous Holstein cows, between 2 and 3 weeks postpartum were selected. Five study groups were established: control (healthy), and cows with ketosis, mastitis, laminitis, and metritis. The parameters measured were ADA activity, total oxidants (TOS), antioxidants (TAC), and OSi ratio. Regarding redox status, no significant differences arise in both saliva and serum being the correlations negative and not significant. In saliva, ADA activity in healthy cows differs from those with pathological processes, having the lowest activities. In serum, ADA activity is similar in the healthy and ketosis cows, showing the lowest activities meanwhile animals with mastitis, laminitis, or metritis have significantly higher activities. In conclusion, the measurement of ADA activities and redox status in saliva does not give consistent results, being preferable to measure them in serum during the transition period. ABSTRACT: Energy and nutrient deficiency in dairy cows in early lactation is considered to contribute to their increased susceptibility to mastitis. We have tested the hypothesis that feeding diets with high contents of either nitrogenic, glucogenic, or lipogenic components in early lactation affects both the endocrine and metabolic status, as well as the mammary immune competence. After calving, cows were fed increasing amounts of concentrate up to 10 kg/d rich in crude protein (nitrogenic, n = 10), glucogenic precursors (glucogenic, n = 11), or lipids (lipogenic, n = 11). In wk 3, one udder quarter was challenged with lipopolysaccharide (LPS) from Escherichia coli. Blood and milk were sampled on the day before LPS challenge (d −1), and on d 0, 1, 2, 3, and 9 after LPS challenge. On the day of LPS challenge additional samples were taken hourly for quarter milk and every 3 h for blood. Urea concentrations were higher in plasma and milk of cows fed the nitrogenic diet. However, plasma concentrations of glucose, cholesterol, triglycerides, β-hydroxybutyrate, nonesterified fatty acids, as well as insulin, glucagon, and insulin-like growth factor-1 were not affected by the different diets. The mammary immune challenge induced a substantial increase of somatic cell count (SCC) in the treated quarter, and a transient decrease of total milk yield and white blood cells similar in all diet groups for one day. The absolute phagocytosis of blood leukocytes was decreased; however, the phagocytosis per cell was increased in glucogenic-fed cows at 6 h after LPS challenge. During mammary inflammation an insulin resistance, shown by increased plasma glucose, insulin, and glucagon, developed similarly in all diet groups. β-hydroxybutyrate and nonesterified fatty acids were decreased at 1 d after LPS challenge in glucogenic-fed cows only. Cholesterol did not change, and triglycerides only decreased significantly in lipogenic-fed cows 6 h after challenge. On d 9 after LPS challenge, SCC and milk yield and metabolic factors were recovered in all groups. In conclusion, the endocrine and metabolic situation, and the immune response to intramammary LPS of dairy cows during early lactation was not substantially influenced by the elevated supply of nitrogenic, glucogenic, or lipogenic components due to the provided feed in this study.

Although there is evidence that ketosis negatively affects fertility, the effect of late and early ketosis on the reproductive performance of lactating cows has not been systematically investigated. The aim of this study was to evaluate the association between time and amplitude of elevated milk BHB (EMB) occurring within 42 d in milk (DIM) and subsequent reproductive performance of lactating Holstein cows. The dairy herd information data of 30,413 cows with 2 test-day milk BHB recordings during early lactation periods 1 and 2 (5–14 and 15–42 DIM, respectively) assessed as negative (<0.15 mmol/L), suspect (0.15–0.19 mmol/L), or positive (≥0.2 mmol/L) for EMB were used in this study. Based on the time and amplitude of milk BHB, cows were grouped into 7 groups: (1) healthy cows negative in both periods 1 and 2 were classified as NEG; (2) suspect in period 1 and negative in period 2: EARLY_SUSP; (3) suspect in period 1 and suspect/positive in period 2: EARLY_SUSP_Pro; (4) positive in period 1 and negative in period 2: EARLY_POS; (5) positive in period 1 and suspect/positive in period 2: EARLY_POS_Pro; (6) negative in period 1 and suspect in period 2: LATE_SUSP; and (7) negative in period 1 and positive in period 2: LATE_POS. The overall prevalence of EMB within 42 DIM was 27.4%, with the highest prevalence being EARLY_SUSP (10.49%). Cows in EARLY_POS and EARLY_POS_Pro, but not other EMB categories, had a longer interval from calving to first service compared with NEG cows. For the reproductive parameters, first service to conception interval, days open and calving interval, cows in all EMB groups except EARLY_SUSP had longer intervals compared with NEG cows. These data indicate that there is a negative association between EMB within 42 d and reproductive performance after the voluntary waiting period. The intriguing findings of this study are the unaltered reproductive performance of EARLY_SUSP cows, and the negative association between late EMB and reproductive performance. Early lactation metabolic imbalance is an important physiological change affecting the health, production, and reproduction of dairy cows. The aims of this study were (1) to evaluate the potential of test-day (TD) variables with or without milk fatty acids (FA) content to classify metabolically imbalanced cows and (2) to evaluate the robustness of the metabolic classification with external data. A data set was compiled from 3 experiments containing plasma β-hydroxybutyrate, nonesterified FA, glucose, insulin-like growth factor-I, FA proportions in milk fat, and TD variables collected from 244 lactations in wk 2 after calving. Based on the plasma metabolites, 3 metabolic clusters were identified using fuzzy c-means clustering and the probabilistic membership value of each cow to the 3 clusters was determined. Comparing the mean concentration of the plasma metabolites, the clusters were differentiated into metabolically imbalanced, moderately impacted, and balanced. Following this, the 2 metabolic status groups identified were imbalanced cows (n = 42), which were separated from what we refer to as "others" (n = 202) based on the membership value of each cow for the imbalanced cluster using a threshold of 0.5. The following 2 FA data sets were composed: (1) FA (groups) having high prediction accuracy by Fourier-transform infrared spectroscopy and, thus, have practical significance, and (2) FA (groups) formerly identified as associated with metabolic changes in early lactation. Metabolic status prediction models were built using FA alone or combined with TD variables as predictors of metabolic groups. Comparison was made among models and external evaluations were performed using an independent data set of 115 lactations. The area under the receiver operating characteristics curve of the models was between 75 and 91%, indicating their moderate to high accuracy as a diagnostic test for metabolic imbalance. The addition of FA groups to the TD models enhanced the accuracy of the models. Models with FA and TD variables showed high sensitivities (80–88%). Specificities of these models (73–79%) were also moderate and acceptable. The accuracy of the FA models on the external data set was high (area under the receiver operating characteristics curve between 76 and 84). The persistently good performance of models with Fourier-transform infrared spectroscopy-quantifiable FA on the external data set showed their robustness and potential for routine screening of metabolically imbalanced cows in early lactation. Background and Aim: It is known that during the early postpartum and lactation periods in dairy cows, metabolic disorders develop, that is, ketosis, which can lead to secondary damage to internal organs. Therefore, it is important to address the issues of changing the lactating cows' clinical, laboratory, and physiological parameters regarding the development of hepatocardial syndrome. This study aimed to provide clinical and diagnostic justification for developing hepatocardial syndrome in highly productive dairy cows. Materials and Methods: The study was conducted on 20 black and white cows in the early postpartum period (7–10 days after birth), with a milk production level of >4500 kg of milk during the previous lactation period, a positive result in the formol colloid sedimentary test, the presence of deafness and splitting of heart sounds, changes in the size, or increased pain sensitivity of the percussion field of the liver. Clinically healthy dairy cows in the early postpartum period were used as controls (n = 24). Clinical, electrocardiographic, echocardiographic, and biochemical parameters were also evaluated. Results: Dairy cows with hepatocardial syndrome developed arterial hypertension and sinus tachycardia, which led to a significant decrease in PQ and QT intervals at ECG. A significant increase in the diastolic size of the interventricular septum, systolic size of the free wall of the left ventricle, and diastolic and systolic sizes of the left ventricle and a significant decrease in the shortening fraction of the left ventricular myocardium were observed in the cows due to the development of hepatocardial syndrome. The affected cows demonstrated a significant increase in serum activity of gamma-glutamyl transferase, alanine aminotransferase, lactate dehydrogenase, creatine phosphokinase, alkaline phosphatase, troponin, malondialdehyde, diene conjugates, and ceruloplasmin and a decrease in glucose concentration. In addition, they demonstrated decreased activity of superoxide dismutase, catalase, glutathione peroxidase, and glutathione reductase. Conclusion: Hepatocardial syndrome in dairy cows occurs due to ketosis, characterized by arterial hypertension, sinus tachycardia, a moderate decrease in myocardial contractility, oxidative stress, and cytolysis of cardiomyocytes and hepatocytes. Therefore, the control and prevention of the development of hepatocardial syndrome will make it possible to maintain the productive health and longevity of dairy cows.

Simple Summary: In this study, 24 first-lactation Holstein cows were evaluated to investigate how different levels of diet-induced subacute rumen acidosis (SARA) during early lactation (up to 70 days in milk; DIM) affect claw health. Claw health was monitored by recording claw lesions at three hoof trimming visits (before calving, at 70 DIM, and three months thereafter), and by assessing locomotion scores (LCS) 1 to 5 at two-week intervals. An intraruminal sensor monitored pH continuously, enabling the determination of the number of days the cows experienced SARA (pH below 5.8 for more than 330 min in 24-h), which allowed us to derive a severity index for SARA. In this manner the cows were assigned to three groups retrospectively: light, moderate, and severe SARA. A statistically significant increase in lameness incidence was observed when comparing light and severe SARA groups, but no effect was observed on LCS and claw lesion prevalence. This study aimed to evaluate the effects of diet-induced subacute rumen acidosis (SARA) severity during transition and the early lactation period on claw health in 24 first-lactation Holstein heifers. All heifers were fed a 30% concentrate (in dry matter) close-up ration three weeks before calving, then switched to a high-concentrate ration (60% dry matter), which was fed until the 70th day in milk (DIM) to induce SARA. Thereafter, all cows were fed the same post-SARA ration with around 36% concentrate in dry matter. Hoof trimming was performed before calving (visit 1), at 70 (visit 2) and at 160 DIM (visit 3). All claw lesions were recorded, and a Cow Claw Score (CCS) was calculated for each cow. Locomotion scores (LCS 1–5) were assessed at two-week intervals. Intraruminal sensors for continuous pH measurements were used to determine SARA (pH below 5.8 for more than 330 min in 24 h). The cluster analysis grouped the cows retrospectively into light (≤11%; n = 9), moderate (>11–<30%; n = 7), and severe (>30%; n = 8) SARA groups, based on the percentage of days individual cows experienced SARA. Statistically significant differences were found between SARA groups light and severe in terms of lameness incidence (p = 0.023), but not for LCS and claw lesion prevalence. Further, the analysis of maximum likelihood estimates revealed that for each day experiencing SARA, the likelihood of becoming lame increased by 2.52% (p = 0.0257). A significant increase in white line lesion prevalence was observed between visits 2 and 3 in the severe SARA group. The mean CCS in severe SARA group cows were higher at each visit compared to cows in the other two groups, but without statistical significance. Overall, this is the first study indicating that first-lactation cows fed a similar high-concentrate diet but with a higher severity of SARA tended to have poorer claw health, albeit with only partial statistical evidence. The objective of this experiment was to investigate the effect of reducing early lactation milking frequency on milk yield and persistency through lactation and early lactation fat mobilization, measured by body condition score (BCS) and BHB in milk. We hypothesized that milking cows twice per day in early lactation before milking them 3 times per day for the remaining lactation would cause less fat mobilization in early lactation, a lower peak milk yield but improved persistency throughout lactation compared with milking cows 3 times per day for the entire lactation. The experiment took place on 2 commercial dairy farms in Denmark. All cows calving in a period of nine months (n = 239) in their current first and later parities were randomly allocated at dry-off to 1 of 3 treatments based on expected calving date. The treatments were (1) cows milked 2 times per day for 1 week after calving, (2) cows milked 2 times per day for 4 weeks after calving and (3) cows milked 2 times per day for 7 weeks after calving. All cows were then milked 3 times per day for the remaining lactation. Milk yield peaked 3.3 and 3.6 d later and milk yield persistency improved with 18 and 19 g per day when cows were milked 2 times per day for 4 and 7 weeks, respectively, compared with milking 2 times per day for 1 week after calving. We found a significant highest milk BHB in treatment 2, but the underlying effect of milking cows 2 times per day for 4 weeks compared with 1 or 7 weeks was unclear. In conclusion, we did not confirm our hypothesis that milking cows 2 times per day compared to 3 times in early lactation would reduce fat mobilization and reduce peak yield. We did, however, find an improved milk yield persistency, which partially offset a numerical reduction in peak yield, and hence there was no significant effect of reducing early lactation milking frequency on total lactation (305 DIM) milk yield. Simple Summary: Studies with rodent models have shown that fibroblast growth factor 21 (FGF21) is a metabolic regulator induced in the liver in response to different stress conditions, such as energy and nutrient deprivation, inflammation, and metabolic disorders. Recently, it has been found that hepatic FGF21 expression is strongly upregulated in dairy cows during early lactation. However, the function of FGF21 in cows has not yet been established. Therefore, the aim of the present study was to gain knowledge about the physiological role played by FGF21 in cows during this period. To this end, out of 30 cows, 8 cows with the highest hepatic FGF21 expression were compared to 8 cows with the lowest hepatic FGF21 expression. Cows with high and low hepatic FGF21 expression did not differ in milk yield, feed intake, nor energy balance. Transcriptomics screening, targeted plasma metabolomics, and analyses of antioxidant parameters indicated that high hepatic FGF21 expression was related to endoplasmic reticulum stress and induction of the antioxidative system in the livers of dairy cows. Therefore, the data of this study suggest that FGF21 plays an important role in the adaptation to cellular stress conditions in early lactation when cows are typically confronted with several stress stimuli. Induction of FGF21 expression in the liver and a significant increase in plasma FGF21 concentration have been demonstrated in cows during early lactation, but knowledge about the function of FGF21 in dairy cows remains limited. In order to improve the understanding of the physiological role of FGF21 in dairy cows, the present study aimed to investigate differences in metabolic pathways between dairy cows with high and low hepatic expression of FGF21 at week 1 of lactation (n = 8/group) by liver transcriptomics, targeted plasma metabolomics, and analysis of inflammatory and oxidative stress-related parameters. Dry matter intake, energy balance, milk yield, and energy-corrected milk yield at days 8–14 postpartum did not differ between cows with high and low hepatic FGF21 expression. However, cows with high FGF21 expression showed an upregulation of genes involved in endoplasmic reticulum stress, inflammation, and nuclear factor E2-related factor 2 (Nrf2)-dependent cytoprotection compared to cows with low FGF21 expression at week 1 postpartum (p < 0.05). Concentrations of important antioxidants (tocopherols, β-carotene, and glutathione) in the liver and plasma, trolox equivalent antioxidant capacity in plasma, concentrations of oxidative stress-related compounds (thiobarbituric acid-reactive substances and protein carbonyls), and levels of most acute phase proteins at week 1 postpartum did not differ between cows with high or low FGF21 expression. Moreover, among a total of >200 metabolites assayed in the plasma, concentrations of only 7 metabolites were different between cows with high or low FGF21 expression (p < 0.05). Overall, the results showed that cows with high and low FGF21 hepatic expression had only moderate differences in metabolism, but FGF21 might be important in the adaptation of dairy cows to stress conditions during early lactation. Dairy cows undergo dramatic metabolic changes and impaired immune system function in the transition period; thus, they are very susceptible to peripartum disease. Vitamin D as an immune-modulatory agent may have protective effects against oxidative stress and numerous diseases. Our objective in this study was to investigate the changes in 25(OH)D concentrations in dairy cattle during the transition period and its association with some inflammatory and oxidative stress markers. Twenty-three healthy cows were selected from two commercial farms and were sampled 1 week prepartum and 1 week and 3 weeks postpartum. 25(OH)D, IL-4, IL-10, INF-γ, Hap, IgG, MDA, and TAC amounts were measured. Time had significant effects on 25(OH)D concentrations and its concentration was decreased 1 week after parturition and then increased. The consumption of vitamin D for calcium homeostasis and the proper immune system functioning may be the reason for decreasing in its amounts after parturition. Pearson test showed that in 1 week prepartum, 25(OH)D levels tended to be significantly correlated with IL-4 (P = 0.076) and IgG (P = 0.058) levels. No significant correlation was observed between 25(OH)D levels and other measured variables in the first and third weeks after parturition. It should be noted that the amount of 25(OH)D in the first week after calving reaches to its lowest level, which may leads to impaired immune system function and increased susceptibility of dairy cows to disease. ABSTRACT: A total of 72 dairy ewes of 2 breeds (MN, Manchega, 72.4 ± 1.9 kg of body weight, n = 36; LC, Lacaune, 77.7 ± 2.3 kg of body weight; n = 36) were used to evaluate the lactational effects of melatonin implants in early lactation and under the short-day photoperiod conditions of autumn (experiment was centered on the winter solstice). Ewes lambed in autumn and were penned indoors in 12 balanced groups of 6 ewes by breed, body weight, age, and number of lambs, and randomly assigned to a 2 × 2 × 3 factorial design (treatment × breed × replicate). Ewes suckled their lambs for 28 d. Treatments were (1) melatonin (MEL), which received 1 subcutaneous implant of melatonin (18 mg/ewe) in the ear base at 35 ± 1 d (1 wk after lamb weaning), and (2) control, which did not receive any treatment. Ewes were fed ad libitum a total mixed ration (forage:concentrate, 60:40) and machine milked twice daily. Daily milk yield was automatically recorded from d 29 to 105 of lactation and sampled every 2 wk for composition. Jugular blood was sampled for plasma hormone analyses at 30, 50, 80, 110, and 124 d of lactation. Body reserves were assessed every 2 wk. Feed intake was measured by pen during 3 separated periods after the start of the treatments (wk 2 to 3, wk 6 to 7, and wk 10 to 11). Feed intake, and milk yield and composition varied by breed, but no MEL effects were detected on dry matter intake, milk yield, milk composition, or fat and protein standardized milk in either breed. As a result of the unique composition of the implants and the variable body weights of the ewes, the MEL treatment dose (on average, 0.24 mg/kg of body weight) was 6.8% greater in the MN (lighter) than in the LC (heavier) ewes. Plasmatic melatonin markedly increased in the MEL-treated ewes (on average, 111%), but despite the amount of MEL used, the MN responded greatly compared with the LC ewes (150 vs. 63%, respectively). No differences in basal plasmatic melatonin were detected between breeds (6.4 ± 1.1 pg/mL, on average), indicating the greater responsiveness to the implants of the lighter MN ewes. Plasmatic prolactin tended to decrease in the MEL-treated ewes (−35%, on average), but the effect was significant only in the MN ewes (−54%), in agreement with their greater response to MEL. No effects of MEL treatment were detected on plasmatic IGF-I in either breed. Moreover, body reserves did not vary by effect of MEL treatment or breed throughout the experiment. In conclusion, the use of exogenous melatonin as MEL implants, together with the endogenous melatonin naturally produced under short-day photoperiod conditions, had no effects on the early-lactation performances of dairy sheep, despite their breed and level of production.

Identifying cows that are at greater risk for disease prior to calving would be a valuable addition to transition management. Prior to the commercial release of software features in an automated behavioral monitoring system, designed to identify cows in the dry period at greater risk of disease postpartum, a retrospective analysis was carried out in five dairy herds to evaluate whether the software could identify prepartum cows that subsequently received health treatments postpartum and whether prepartum alerts (transition alerts) are associated with a reduction in milk production in the subsequent lactation. Herd management and production records were analyzed for cows receiving treatment in the first 21 d of lactation (days in milk, DIM) for clinical mastitis, reproductive tract disease (metritis, retained fetal membranes), metabolic disease (hypocalcemia, ketosis and displaced abomasum) and for cows exiting the herd by 60 DIM. Data was gathered for 986 cows, 382 (38.7%) of which received a transition alert and 604 (61.3%) that did not. During the first 21 DIM 312 (31.6%) cows went on to receive a disease treatment, of these 51.9% (n = 162/312) were transition alert cows and 48.1% (n = 150/312) non-transition alert cows, while 8.6% (n = 33/382) alert cows exited the herd by 60 DIM compared to 4.8% (n = 29/604) of cows that did not receive an alert. A cow receiving a transition alert (OR = 1.76, 95% confidence interval (CI) = 1.27–2.44) and increasing parity (OR = 2.03, 95% CI = 1.44–2.86) were both associated with increased risk of receiving a disease treatment in the first 21 DIM. The occurrence of a transition alert was negatively associated with both week 4 milk yield (daily average yield in fourth week of lactation) and predicted 305 d yield. Transition alerts correctly predicted 62.5% (95% CI: 59.3–65.5) of treatments with a sensitivity of 42.4% (95% CI: 37.4–45.5) and a specificity of 75.2% (95% CI: 71.5–78.6). Associations were identified between postpartum health and production outcomes and prepartum behavioral measures from an automated activity monitoring system.

The objective of this experiment was to investigate the effect of feeding strategy on milk production, composition and selected processability parameters in the initial stages of lactation. Twenty Holstein Friesian cows were allocated to one of two dietary treatments (n = 10; 7 multiparous and 3 primiparous) in a randomised complete block design for 21 d from day 10 to day 31 post-calving. Treatment 1 (pasture-based system [PBS]) was a pasture/pasture silage-based diet where cows were offered ad libitum grazed pasture or pasture silage (when weather did not permit grazing) plus 3 kg DM/d or 5 kg DM/d concentrate supplementation, respectively. On average, cows grazed pasture for 7.5 d and were fed pasture silage indoors for 13.5 d. Treatment 2 (TMR) was a total mixed ration (TMR) diet made up of concentrate, plus maize silage, pasture silage, beet pulp, soya bean meal and straw. Multiparous cows were blocked on calving date and balanced for parity and milk yield. Primiparous cows were balanced for live weight. Milk attributes pertinent to composition and functionality (e.g., fatty acids and rennet coagulation time [RCT]) were examined over a 21-d experimental period from day 10 to day 31 post-calving. Cows offered PBS tended to have a lower test day milk yield (PBS = 24.2 kg/cow vs. TMR = 26.8 kg/cow, P = 0.09) and a greater milk urea nitrogen (MUN) content compared to TMR (PBS = 0.030 g/100 g milk vs. TMR = 0.013 g/100 g milk, P < 0.001). Most notably, PBS-derived milks had a greater (P < 0.001) concentration of cis-9 trans-11 conjugated linoleic acid (CLA) compared to TMR. In conclusion, milk produced during early lactation from both feeding strategies was suitable for processing. Feeding a TMR compared with ad libitum pasture/pasture silage had no impact on average milk pH, casein concentration or RCT. Cows fed a pasture/pasture silage-based diet produced milk with a desirable RCT for milk processing, while the higher MUN content from cows offered PBS did not impact the processability of milk. Furthermore, milk from cows offered PBS had greater concentrations of cis-9 trans-11 CLA, which may offer human health benefits.

The experiment was to study the effect of different proportions of fatty acids in early-lactating diets on the production performance and serum indexes of dairy cows. A total of 160 early-lactating cows were randomly divided into four groups, with 40 replicates in each group and one cows in each replicate. (C16: 0): (C18: 1)=72.20: 15.30 in the basic diet of control group. The (C16: 0): (C18: 1) diets of trial group I, group II and group III were 60.60: 26.10, 56.00: 30.80 and 43.50: 2.00, respectively. The test period was 21 d. The results showed that compared with control group, the dry matter intake of group II and group III was extremely increased (P<0.01), while that of group I was extremely decreased (P<0.01), the milk yield of group II was significantly decreased (P<0.05), the feed conversion efficiency of group II and group III were extremely decreased (P<0.01). Compared with control group, the body condition scores of cows in group II and group III on 0,7,14 d postpartum were extremely increased (P<0.01). Compared with control group, the content of BHBA and T4 in serum of group II and group III were extremely decreased (P<0.01), and the content of GLU were extremely increased (P<0.01). The levels of serum Insulin and T3 in each test group were extremely increased (P<0.01), the serum GH content in group I and group III were significantly increased (P<0.05), the content of serum BUN in group II was extremely decreased (P<0.01), the content of CREA in serum of group I and group II were extremely decreased (P<0.01). Compared with control group, the serum AST activity of dairy cows in group II and group 川 was significantly decreased (P<0.05). Compared with control group, the content of serum MDA in group II and group 川 were extremely increased (P<0.01), the activity of serum SOD in group I were extremely decreased (P<0.01), and the activity of serum GSH-Px in group 川 was significantly decreased (P<0.05). The study indicates that when the fatty acid composition of the diet C16: 0: C18: 1=72.20: 15.30, the milk yield of cows is better, and when (C16: 0)：(C18: 1)= 56.00: 30.80, the effect of postpartum body fat mobilization to maintain the body condition is better. Background and Aim: It is known that during the early postpartum and lactation periods in dairy cows, metabolic disorders develop, that is, ketosis, which can lead to secondary damage to internal organs. Therefore, it is important to address the issues of changing the lactating cows' clinical, laboratory, and physiological parameters regarding the development of hepatocardial syndrome. This study aimed to provide clinical and diagnostic justification for developing hepatocardial syndrome in highly productive dairy cows. Materials and Methods: The study was conducted on 20 black and white cows in the early postpartum period (7-10 days after birth), with a milk production level of >4500 kg of milk during the previous lactation period, a positive result in the formol colloid sedimentary test, the presence of deafness and splitting of heart sounds, changes in the size, or increased pain sensitivity of the percussion field of the liver. Clinically healthy dairy cows in the early postpartum period were used as controls (n = 24). Clinical, electrocardiographic, echocardiographic, and biochemical parameters were also evaluated. Results: Dairy cows with hepatocardial syndrome developed arterial hypertension and sinus tachycardia, which led to a significant decrease in PQ and QT intervals at ECG. A significant increase in the diastolic size of the interventricular septum, systolic size of the free wall of the left ventricle, and diastolic and systolic sizes of the left ventricle and a significant decrease in the shortening fraction of the left ventricular myocardium were observed in the cows due to the development of hepatocardial syndrome. The affected cows demonstrated a significant increase in serum activity of gamma-glutamyl transferase, alanine aminotransferase, lactate dehydrogenase, creatine phosphokinase, alkaline phosphatase, troponin, malondialdehyde, diene conjugates, and ceruloplasmin and a decrease in glucose concentration. In addition, they demonstrated decreased activity of superoxide dismutase, catalase, glutathione peroxidase, and glutathione reductase. Conclusion: Hepatocardial syndrome in dairy cows occurs due to ketosis, characterized by arterial hypertension, sinus tachycardia, a moderate decrease in myocardial contractility, oxidative stress, and cytolysis of cardiomyocytes and hepatocytes. This study aimed to investigate the effects of dietary supplementation with different levels of calcium propionate on the lactation performance, blood energy metabolite parameters, and milk metabolites of dairy cows in early lactation. Thirty-two multiparous Holstein cows were randomly divided into 4 groups, which were orally drenched with 0, 200, 350, and 500 g/d calcium propionate per cow supplemented to a basal diet for 5 weeks from calving. The milk and blood of the dairy cows were sampled and measured every week. The milk samples from the last week were used for the metabolomic analysis via liquid chromatography–mass spectrometry (LC-MS). The results showed that the calcium propionate supplementation quadratically increased the dry matter intake, energy-corrected milk yield, and 4% fat-corrected milk yield; linearly reduced the milk protein and milk lactose concentrations; and quadratically decreased the somatic cell count in the milk. With the increase in calcium propionate, the serum glucose content showed a linear increase, while the serum insulin content showed a quadratic increase. The diets supplemented with calcium propionate quadratically decreased the β-hydroxybutyric acid and linearly decreased the non-esterified fatty acid content in the serum. The metabolomic analysis revealed that eighteen different metabolites were identified in the milk samples of the dairy cows supplemented with calcium propionate at 350 g/d, which decreased the abundance of genistein and uridine 5-monophosphate and increased the abundance of adenosine, uracil, protoporphyrin IX, and sphingomyelin (d 18:1/18:0) compared with the control group. The milk metabolic analysis indicated that the calcium propionate effectively improved the milk synthesis and alleviated the mobilization of adipose tissue and bone calcium. In summary, the calcium propionate could improve the lactation performance and energy status and promote the milk metabolic profile of dairy cows in early lactation. Simple Summary: The active forms of niacin that represent niacin status are nicotinamide adenine dinucleotide (NAD), nicotinamide adenine dinucleotide phosphate (NADP) and the NAD:NADP ratio. Previous studies have shown metabolic changes in the function of niacin form and dose, but it has not been determined whether there are changes in the function of active form of niacin that indicate the vitamin status in the body. In this study, we examined differences in NAD, NADP and NAD:NADP concentration in blood and their relationship with metabolic parameters in cows receiving and not receiving additional niacin in food. We concluded that NAD and NADP are good indicators of the ability of an additional niacin source to create functional cofactors due to their concentration changes, while the NAD:NADP ratio is a good indicator of the biological effects of additional niacin due to correlation with many metabolites. Previous experimental models on cows have examined the difference in the metabolic adaptation in cows after niacin administration, without identifying the most important mediators between niacin administration and its biological effects, namely active forms of niacin. All tissues in the body convert absorbed niacin into its main metabolically active form, the coenzyme nicotinamide adenine dinucleotide (NAD) and nicotinamide adenine dinucleotide phosphate (NADP). The aim of this study was to determine the influence of niacin administration in periparturient period on NAD, NADP and the NAD:NADP ratio and to determine relationship between these indicators of an active form of niacin with metabolic parameters in cow blood. The study included 90 healthy cows: 45 cows receiving niacin and 45 cows were negative control. The niacin group was treated with nicotinic acid for two weeks before, as well as two weeks after parturition. Nicotinic acid was applied per os with feed. In cows receiving niacin, there was a significantly higher concentration of NAD and NADP, but the NAD:NADP ratio did not differ compared with control. All three indicators were able to separate cows who received and who did not receive additional niacin. NAD and NADP are good indicators of the availability of niacin from additional sources. The NAD:NADP ratio is a good indicator of the biological effect of applied niacin on metabolites in cows due to its correlation with a number of metabolites: positive correlation with glucose, insulin, glucose to insulin ratio and the revised quantitative insulin sensitivity check index (RQUICKIBHB) of insulin resistance, triglycerides and cholesterol, and a negative correlation with nonesterified fatty acid (NEFA), beta hydroxybutyrate (BHB), gamma-glutamyltranspherase (GGT) and urea in cows receiving niacin. The same amount of added niacin in feed can produce different concentrations of NAD, NADP and NAD:NADP in the blood, and this was not related to their concentration before the addition of niacin. The change in the concentration of the active form of niacin (NAD, NADP and NAD:NADP) further correlates with the concentration of metabolic parameters, which indicates that the intensity of the biological effect of additional niacin can be accurately determined only if we know the concentrations of its active forms in blood. Research data on milk composition in cetaceans are scattered and fragmentary. This paper analyses the gross and mineral composition, the fatty acids profile and the fat globule size of bottlenose dolphin (Tursiops truncatus) colostrum and milk at early lactation. The milk samplings were carried out on three lactating female of bottlenose dolphins at the 1st, 4-5 and 20- 42 day post partum. High percentages of dry matter (51.88%), fat (26.08%) and protein (13.83%) were found in the colostrum on the first day, while there was a tendency for these components to decrease in the milk. The average diameter the milk fat globule (7.07 μm) of this species was assessed for the first time. The milk was rich in unsaturated fatty acids, which were more than twice compared to the saturated fatty acids (unsaturated / saturated ratio = 2.6). The main fatty acids of dolphin milk were C16: 0, C16:1 n-7, C18: 1n-9, C20: 1 n-11 C20: 5 and C22:6n-3. Several FA tended to increase from colostrum to milk: C12: 0 doubled, C20: 1 n-7 and C22:1 n-11 tripled, C20: 1 n-9 quintupled in later milk samples. Differently, other FA tended to decrease: C18: 3 n-3 was approximately 4 times lower and C24: 0 and C24: 1 halved in milk compared to colostrum. In conclusion, this paper contributes to the knowledge of the nutritional value of milk from bottlenose dolphins maintained under human care. Higher content of nutritional components such as fat and protein was found in the first day colostrum compared to milk. This study aimed to access the effect of heat stress on milk yield, antioxidative levels, and serum metabolites in primiparous and multiparous Holstein dairy cows during the early lactation stage. A total of 200 cows were selected based on their month of calving (June, temperature humidity index (THI) = 66.72; July, THI = 70.30; August, THI = 69.32; September, THI = 67.20; October, THI = 59.45). Blood samples were collected on days 0, 21, 50, 80, and 100 after calving for serum oxidative status analysis and milk yield was recorded daily. The lower average daily milk yield was recorded among the cows that calved in June and July (P < 0.05), and the average daily milk yield of multiparous cows was higher than that of primiparous cows that calved in the same month (P < 0.05) from d1 to d100, suggesting that seasonal (June, July) heat stress negatively affected milk yield in both primiparous and multiparous cows at early lactation. The study also indicated that there was seasonal variation in most of the serum metabolites across the studied months. The study shows that heat stress (average THI = 70.30) was higher among the cows calving in June vis-à-vis those calving in October and differences were also observed among the primiparous cows and multiparous cows, respectively. These metabolites (e.g., glycine, serine, etc.) which showed significant variations were mainly involved in the pathways of aminoacyl-tRNA biosynthesis, glyoxylate and dicarboxylate metabolism, and the metabolism of glycine, serine and threonine. These data suggested that heat stress negatively affected the elevation of the serum oxidative and antioxidative index and thus badly influence milk yield. Dietary supplementation with calcium propionate can effectively alleviate negative energy balance and hypocalcemia of dairy cows in early lactation. The objective of this study was to investigate the effects of calcium propionate feeding levels on the immune function, liver function, and fecal microbial composition of dairy cows in early lactation. Thirty-two multiparous Holstein cows were randomly assigned to four treatments after calving. Treatments were a basal diet plus 0, 200, 350, and 500 g calcium propionate per cow per day throughout a 5-week trial period. Cows were milked three times a day, and blood was sampled to measure immune function and liver function on d 7, 21, and 35. The rectal contents were sampled and collected on d 35 to analyze the microbial composition using 16S rRNA gene sequencing. The results indicated that increasing amounts of calcium propionate did not affected the serum concentrations of total protein, IgG, IgM, and calcium, but the concentrations of albumin and IgA changed quadratically. With the increase of calcium propionate, the activity of serum alanine transaminase and aspartate aminotransferase increased linearly, in contrast, the activity of alkaline phosphatase decreased linearly. Moreover, dietary supplementation with increasing levels of calcium propionate tended to quadratically decrease the relative abundance of Firmicutes while quadratically increased the abundance of Bacteroidetes, and consequently linearly decreased the Firmicutes/Bacteroidetes ratio in the rectal microbiota. Additionally, the supplementation of calcium propionate increased the relative abundances of Ruminococcaceae_UCG-005 and Prevotellaceae_UCG-004 linearly, and Ruminococcaceae_UCG-014 quadratically, but decreased the relative abundances of Lachnospiraceae_NK3A20_group and Family_XIII_AD3011_group quadratically. Compared with the CON group, the calcium propionate supplementation significantly decreased the relative abundance of Acetitomaculum but increased the abundances of Rikenellaceae_RC9_gut_group and Alistipes. In summary, these results suggested that the supplementation of calcium propionate to dairy cows in early lactation could beneficially alter the rectal microbiota.

This study aimed to evaluate the effects of diet-induced subacute rumen acidosis (SARA) severity during transition and the early lactation period on claw health in 24 first-lactation Holstein heifers. All heifers were fed a 30% concentrate (in dry matter) close-up ration three weeks before calving, then switched to a high-concentrate ration (60% dry matter), which was fed until the 70th day in milk (DIM) to induce SARA. Thereafter, all cows were fed the same post-SARA ration with around 36% concentrate in dry matter. Hoof trimming was performed before calving (visit 1), at 70 (visit 2) and at 160 DIM (visit 3). All claw lesions were recorded, and a Cow Claw Score (CCS) was calculated for each cow. Locomotion scores (LCS 1–5) were assessed at two-week intervals. Intraruminal sensors for continuous pH measurements were used to determine SARA (pH below 5.8 for more than 330 min in 24 h). The cluster analysis grouped the cows retrospectively into light (≤11%; n = 9), moderate (>11–n = 7), and severe (>30%; n = 8) SARA groups, based on the percentage of days individual cows experienced SARA. Statistically significant differences were found between SARA groups light and severe in terms of lameness incidence (p = 0.023), but not for LCS and claw lesion prevalence. Further, the analysis of maximum likelihood estimates revealed that for each day experiencing SARA, the likelihood of becoming lame increased by 2.52% (p = 0.0257). A significant increase in white line lesion prevalence was observed between visits 2 and 3 in the severe SARA group. The mean CCS in severe SARA group cows were higher at each visit compared to cows in the other two groups, but without statistical significance. Overall, this is the first study indicating that first-lactation cows fed a similar high-concentrate diet but with a higher severity of SARA tended to have poorer claw health, albeit with only partial statistical evidence.

Calcium propionate is one kind of good source for preventing and treating hypocalcemia and ketosis for dairy cows in early lactation. However, little is known about the effects of different feeding levels of calcium propionate on the ruminal bacterial community of early lactation dairy cows. This study aimed to explore the effects of different calcium propionate feeding levels on the ruminal fermentation and bacterial community composition of early lactation dairy cows. Twenty-four multiparous cows were randomly allocated into control (CON), low calcium propionate (LCaP), medium calcium propionate (MCaP), and high calcium propionate (HCaP) groups with six cows per group after calving. The CON group cows were fed the normal total mixed ration (TMR), and the cows of the LCaP, MCaP, and HCaP groups were fed TMR supplemented with 200, 350, and 500 g/day calcium propionate for 35 days after calving, respectively. The rumen fermentation parameters were measured every week, and the ruminal bacterial community composition of the last week was analyzed by 16S rRNA gene sequencing. Under the same diet, the rumen pH showed no difference among the four groups, but the content of microbial crude protein (MCP) and ammonia nitrogen quadratically decreased and linearly increased with calcium propionate supplementation, respectively. The feeding of calcium propionate linearly increased the concentrations of total volatile fatty acid (VFA), acetate, propionate, butyrate, iso-valerate, and valerate in the rumen. In all the treatment groups, Bacteroidetes , Firmicutes , and Proteobacteria were the dominant phyla, and Prevotella_1 and Succiniclasticum were the dominant genera in the rumen. Compared with the CON group, the addition of calcium propionate to the early lactation dairy cows quadratically improved the alpha diversity index of Chao1 estimator and observed species, but had little effect on the relative abundance of the major bacterial at phyla and genera level. These results suggested different levels of calcium propionate supplementation improved the rumen fermentation and the ruminal bacterial diversity but had little impact on the major ruminal bacterial community composition of dairy cows in early lactation. In this study, it was aimed to identify the predominant coagulase-negative staphylococci (CNS) species in subclinical, clinical, and persistent infections in Konya province, to determine various virulence and resistance mechanisms of CNS species within the agelactation level. The research has been carried out in a dairy herd that is free from brucellosis and tuberculosis and uses an automated herd management system. Thirty-two heifers at the beginning of lactation with milk yields of more than 20 liters have been observed in terms of udder infections during the first 1-4 months of lactation. The prevalence of intramammary infections (IMI) and of CNS-caused IMI has been 75% and 53%, respectively. The rate of CNS was 61% among all isolates, with the rate of the major pathogen 26%, and the rate of the other pathogen 12%. S. chromogenes has been the most frequently isolated CNS species (45.5%). In conclusion, CNS may cause udder infections at the herd level in heifers during the early lactation period, which may cause persistent infections with different mechanisms, and S. chromogenes may emerge as a new contagious mastitis agent in the future. CNSs should be identified at the species level rather than the group level in order to develop effective control programs. A total of 72 dairy ewes of 2 breeds (MN, Manchega, 72.4 ± 1.9 kg of body weight, n = 36; LC, Lacaune, 77.7 ± 2.3 kg of body weight; n = 36) were used to evaluate the lactational effects of melatonin implants in early lactation and under the short-day photoperiod conditions of autumn (experiment was centered on the winter solstice). Ewes lambed in autumn and were penned indoors in 12 balanced groups of 6 ewes by breed, body weight, age, and number of lambs, and randomly assigned to a 2 × 2 × 3 factorial design (treatment × breed × replicate). Ewes suckled their lambs for 28 d. Treatments were (1) melatonin (MEL), which received 1 subcutaneous implant of melatonin (18 mg/ewe) in the ear base at 35 ± 1 d (1 wk after lamb weaning), and (2) control, which did not receive any treatment. Ewes were fed ad libitum a total mixed ration (forage:concentrate, 60:40) and machine milked twice daily. Daily milk yield was automatically recorded from d 29 to 105 of lactation and sampled every 2 wk for composition. Jugular blood was sampled for plasma hormone analyses at 30, 50, 80, 110, and 124 d of lactation. Body reserves were assessed every 2 wk. Feed intake was measured by pen during 3 separated periods after the start of the treatments (wk 2 to 3, wk 6 to 7, and wk 10 to 11). Feed intake, and milk yield and composition varied by breed, but no MEL effects were detected on dry matter intake, milk yield, milk composition, or fat and protein standardized milk in either breed. As a result of the unique composition of the implants and the variable body weights of the ewes, the MEL treatment dose (on average, 0.24 mg/kg of body weight) was 6.8% greater in the MN (lighter) than in the LC (heavier) ewes. Plasmatic melatonin markedly increased in the MEL-treated ewes (on average, 111%), but despite the amount of MEL used, the MN responded greatly compared with the LC ewes (150 vs. 63%, respectively). No differences in basal plasmatic melatonin were detected between breeds (6.4 ± 1.1 pg/mL, on average), indicating the greater responsiveness to the implants of the lighter MN ewes. Plasmatic prolactin tended to decrease in the MEL-treated ewes (−35%, on average), but the effect was significant only in the MN ewes (−54%), in agreement with their greater response to MEL. No effects of MEL treatment were detected on plasmatic IGF-I in either breed. Moreover, body reserves did not vary by effect of MEL treatment or breed throughout the experiment. This study aimed to evaluate differences in productive performance between primiparous and multiparous Murrah buffaloes and the interrelationships between metabolic traits during the transition period and early lactation. Thirty pregnant buffaloes were monitored during the transition period and at the beginning of the lactation. Animals were randomly assigned within the two experimental groups considering the calving number and the estimated calving date: primiparous (n = 15) and multiparous buffaloes (n = 15). The buffaloes were monitored every week during the last 30 days of pregnancy, and the first 63 days postpartum. Buffaloes were kept in the same environment condition, and management practices. Multiparous buffaloes, at the postpartum period, showed higher milk fat, protein, lactose, total dry extract production, non-fat dry extract contents, and higher milk urea nitrogen and casein contents than primiparous buffaloes. Primiparous buffaloes showed higher urine pH and hematocrit concentration than the multiparous group at the prepartum period and higher leukocyte and lymphocytes concentrations at the postpartum. During the transition period, primiparous buffaloes exhibited negative interrelationships between metabolic traits and productive performance related to variations in their metabolic status. These results may indicate that multiparous buffaloes fewer sensitive to variations of metabolic status during the transition period.

Calcium propionate is one kind of good source for preventing and treating hypocalcemia and ketosis for dairy cows in early lactation. However, little is known about the effects of different feeding levels of calcium propionate on the ruminal bacterial community of early lactation dairy cows. This study aimed to explore the effects of different calcium propionate feeding levels on the ruminal fermentation and bacterial community composition of early lactation dairy cows. Twenty-four multiparous cows were randomly allocated into control (CON), low calcium propionate (LCaP), medium calcium propionate (MCaP), and high calcium propionate (HCaP) groups with six cows per group after calving. The CON group cows were fed the normal total mixed ration (TMR), and the cows of the LCaP, MCaP, and HCaP groups were fed TMR supplemented with 200, 350, and 500 g/day calcium propionate for 35 days after calving, respectively. The rumen fermentation parameters were measured every week, and the ruminal bacterial community composition of the last week was analyzed by 16S rRNA gene sequencing. Under the same diet, the rumen pH showed no difference among the four groups, but the content of microbial crude protein (MCP) and ammonia nitrogen quadratically decreased and linearly increased with calcium propionate supplementation, respectively. The feeding of calcium propionate linearly increased the concentrations of total volatile fatty acid (VFA), acetate, propionate, butyrate, iso-valerate, and valerate in the rumen. In all the treatment groups, Bacteroidetes, Firmicutes, and Proteobacteria were the dominant phyla, and Prevotella_1 and Succiniclasticum were the dominant genera in the rumen. Compared with the CON group, the addition of calcium propionate to the early lactation dairy cows quadratically improved the alpha diversity index of Chao1 estimator and observed species, but had little effect on the relative abundance of the major bacterial at phyla and genera level. These results suggested different levels of calcium propionate supplementation improved the rumen fermentation and the ruminal bacterial diversity but had little impact on the major ruminal bacterial community composition of dairy cows in early lactation.

The objectives of this study were to induce hindgut and metabolic acidosis via abomasal infusion of corn starch and β-hydroxybutyrate (BHB), respectively, and to determine the effects of these physiological states in early-lactation dairy cows. In a 6 × 6 Latin square design, 6 rumen-fistulated Holstein-Friesian dairy cows (66 ± 18 d in milk) were subjected to 5 d of continuous abomasal infusion treatments followed by 2 d of rest. The abomasal infusion treatments followed a 3 × 2 factorial design, with 3 levels of corn starch and 2 levels of BHB. The infusions were water as control, 1.5 kg of corn starch/d, 3.0 kg of corn starch/d, 8.0 mol BHB/d, 1.5 kg of corn starch/d + 8.0 mol BHB/d, or 3.0 kg of corn starch/d + 8.0 mol BHB/d. A total mixed ration consisting of 35.0% grass silage, 37.4% corn silage, and 27.6% concentrate (on a dry matter basis) was fed at 90% of ad libitum intake of individual cows. The experiment was conducted in climate respiration chambers to facilitate determination of energy and N balance. Fecal pH decreased with each level of corn starch infused into the abomasum and was 6.49, 6.00, and 5.15 with 0.0, 1.5, and 3.0 kg of corn starch/d, respectively, suggesting that hindgut acidosis was induced with corn starch infusion. No systemic inflammatory response was observed and the permeability of the intestine or hindgut epithelium was not affected by the more acidic conditions. This induced hindgut acidosis was associated with decreased digestibility of nutrients, except for crude fat and NDF, which were not affected. Induced hindgut acidosis did not affect milk production and composition and energy balance, but increased milk N efficiency. Abomasal infusion of BHB resulted in a compensated metabolic acidosis, which was characterized by a clear disturbance of acid-base status (i.e., decreased blood total CO 2 , HCO 3 , and base excess, and a tendency for decreased urinary pH), whereas blood pH remained within a physiologically normal range. Abomasal infusion of BHB resulted in increased concentrations of BHB in milk and plasma, but both remained well below the critical threshold values for subclinical ketosis. Induced compensated metabolic acidosis, as a result of abomasally infused BHB, increased energy retained as body fat, did not affect milk production and composition or inflammatory response, but increased intestinal permeability. Induction of FGF21 expression in the liver and a significant increase in plasma FGF21 concentration have been demonstrated in cows during early lactation, but knowledge about the function of FGF21 in dairy cows remains limited. In order to improve the understanding of the physiological role of FGF21 in dairy cows, the present study aimed to investigate differences in metabolic pathways between dairy cows with high and low hepatic expression of FGF21 at week 1 of lactation (n = 8/group) by liver transcriptomics, targeted plasma metabolomics, and analysis of inflammatory and oxidative stress-related parameters. Dry matter intake, energy balance, milk yield, and energy-corrected milk yield at days 8–14 postpartum did not differ between cows with high and low hepatic FGF21 expression. However, cows with high FGF21 expression showed an upregulation of genes involved in endoplasmic reticulum stress, inflammation, and nuclear factor E2-related factor 2 (Nrf2)-dependent cytoprotection compared to cows with low FGF21 expression at week 1 postpartum (p < 0.05). Concentrations of important antioxidants (tocopherols, β-carotene, and glutathione) in the liver and plasma, trolox equivalent antioxidant capacity in plasma, concentrations of oxidative stress-related compounds (thiobarbituric acid-reactive substances and protein carbonyls), and levels of most acute phase proteins at week 1 postpartum did not differ between cows with high or low FGF21 expression. Moreover, among a total of >200 metabolites assayed in the plasma, concentrations of only 7 metabolites were different between cows with high or low FGF21 expression (p < 0.05). Overall, the results showed that cows with high and low FGF21 hepatic expression had only moderate differences in metabolism, but FGF21 might be important in the adaptation of dairy cows to stress conditions during early lactation.

Chromium may regulate dairy cow metabolism; a chelated formation of chromium methionine (Cr-Met) is available to the feed industry. The objective of this study was to investigate the effect of Cr-Met supplementation on lactation performance, hepatic respiratory rate and anti-oxidative capacity in early-lactating Holstein dairy cows. 64 multiparous cows were assigned to 16 blocks based on parity and milk yield and then the four cows in a block were randomly allocated to four treatment groups with 0, 4, 8 or 16 g/d of Cr-Met per cow supplemented to a basal diet. Cows were moved from an open dry lot to a naturally ventilated tie stall barn 2 weeks before treatment to adapt to this facility, fed and milked at 0630, 1400, and 1930 h every day. The experiment lasted for 12 weeks. Milk yield and composition were recorded weekly. Dry matter intake was measured every 2 weeks for a total of six times throughout the trial. The milk production of high producing European dairy cattle breeds in tropical environment is not in satisfaction level. In general, cows develop negative energy balance at the early lactation resulting poor production performances. The aim of the study was to assess the effect of total mixed ration feeding systems (TMR) over conventional feeding system to increase the production performances of cows in early lactation. Napier (Pennisetum purpureum) based conventional feeding system (CFS) versus two different types of total mixed rations as TMR1 (with the same ingredients and same quantity in CFS) and TMR2 (16% dry matter of napier in TMR1 was replaced with legume and non-legume) were studied on production performances of 15 Holstein Friesian cows in early lactation (n=5). The highest average daily milk yield was observed in TMR2 fed cows (17.88±3.03 L/d) while the lowest was observed in CFS fed cows (12.23±0.88 L/d, P<0.0001). The percentage of average daily weight gain was differed (P=0.01) among the treatments (CFS=1.53%, TMR1=2.79%, and TMR2=4.32%). The body condition score of the cows fed with TMR1 and TMR2 was increased by 0.1 and 0.25, respectively at the end of the experimental period whereas it was unchanged in the cows fed with CFS. Results revealed that feeding of TMR for Holstein Friesian cows in early lactation is a better way to achieve increased milk production with a better body condition in the tropics. This study aimed to investigate the effects of dietary supplementation with different levels of calcium propionate on the lactation performance, blood energy metabolite parameters, and milk metabolites of dairy cows in early lactation. Thirty-two multiparous Holstein cows were randomly divided into 4 groups, which were orally drenched with 0, 200, 350, and 500 g/d calcium propionate per cow supplemented to a basal diet for 5 weeks from calving. The milk and blood of the dairy cows were sampled and measured every week. The milk samples from the last week were used for the metabolomic analysis via liquid chromatography–mass spectrometry (LC-MS). The results showed that the calcium propionate supplementation quadratically increased the dry matter intake, energy-corrected milk yield, and 4% fat-corrected milk yield; linearly reduced the milk protein and milk lactose concentrations; and quadratically decreased the somatic cell count in the milk. With the increase in calcium propionate, the serum glucose content showed a linear increase, while the serum insulin content showed a quadratic increase. The diets supplemented with calcium propionate quadratically decreased the β-hydroxybutyric acid and linearly decreased the non-esterified fatty acid content in the serum. The metabolomic analysis revealed that eighteen different metabolites were identified in the milk samples of the dairy cows supplemented with calcium propionate at 350 g/d, which decreased the abundance of genistein and uridine 5-monophosphate and increased the abundance of adenosine, uracil, protoporphyrin IX, and sphingomyelin (d 18:1/18:0) compared with the control group. The milk metabolic analysis indicated that the calcium propionate effectively improved the milk synthesis and alleviated the mobilization of adipose tissue and bone calcium. In summary, the calcium propionate could improve the lactation performance and energy status and promote the milk metabolic profile of dairy cows in early lactation. Calcium propionate (350 g/d) is a well-recommended supplement for dairy cows for alleviating negative energy balance and hypocalcemia in early lactation.

Previous experimental models on cows have examined the difference in the metabolic adaptation in cows after niacin administration, without identifying the most important mediators between niacin administration and its biological effects, namely active forms of niacin. All tissues in the body convert absorbed niacin into its main metabolically active form, the coenzyme nicotinamide adenine dinucleotide (NAD) and nicotinamide adenine dinucleotide phosphate (NADP). The aim of this study was to determine the influence of niacin administration in periparturient period on NAD, NADP and the NAD:NADP ratio and to determine relationship between these indicators of an active form of niacin with metabolic parameters in cow blood. The study included 90 healthy cows: 45 cows receiving niacin and 45 cows were negative control. The niacin group was treated with nicotinic acid for two weeks before, as well as two weeks after parturition. Nicotinic acid was applied per os with feed. In cows receiving niacin, there was a significantly higher concentration of NAD and NADP, but the NAD:NADP ratio did not differ compared with control. All three indicators were able to separate cows who received and who did not receive additional niacin. NAD and NADP are good indicators of the availability of niacin from additional sources. The NAD:NADP ratio is a good indicator of the biological effect of applied niacin on metabolites in cows due to its correlation with a number of metabolites: positive correlation with glucose, insulin, glucose to insulin ratio and the revised quantitative insulin sensitivity check index (RQUICKIBHB) of insulin resistance, triglycerides and cholesterol, and a negative correlation with nonesterified fatty acid (NEFA), beta hydroxybutyrate (BHB), gamma-glutamyltranspherase (GGT) and urea in cows receiving niacin. The same amount of added niacin in feed can produce different concentrations of NAD, NADP and NAD:NADP in the blood, and this was not related to their concentration before the addition of niacin. The change in the concentration of the active form of niacin (NAD, NADP and NAD:NADP) further correlates with the concentration of metabolic parameters, which indicates that the intensity of the biological effect of additional niacin can be accurately determined only if we know the concentrations of its active forms in blood. Under basal conditions (without additional niacin), active forms of niacin that already exist in the blood do not show significant correlations with metabolic parameters.
