Abstract Background To ensure workforce stability in China’s healthcare system and maintain high-quality care, it is essential to comprehensively understand the interplay of factors contributing to turnover intention of healthcare workers. This study aims to examine the associations between potential factors and turnover intention in healthcare workers and explore the mediating effect of job satisfaction and work engagement in the association between them. Methods In this cross-sectional study, a random sample of 1060 healthcare workers working in 98 public medical institutions were recruited to rate their turnover intention in 2018 in Shenzhen, China. Information on socio-demographic characteristics, job-related factors, turnover intention, job satisfaction, work engagement, work stress and doctor-patient relationship of participants were collected. Pearson’s chi-squared tests and binary logistic regression analyses were performed to explore the association between these factor and turnover intention. Mediation analysis was used to explore the roles of potential mediators and moderators. Results The results showed that age (OR: 0.35, 95%CI: 0.16 to 0.72), tenure (OR: 0.98, 95%CI: 0.96 to 0.99), administrative positions (OR: 0.33, 95%CI: 0.16 to 0.63), and night shift frequency (OR: 1.84, 95%CI: 1.26 to 2.67) were significantly associated with turnover intention. We identified the mediating effect of job satisfaction and work engagement in the relationship between administrative positions and turnover intention, while the suppressing effect in the relationship between professional titles and turnover intention. Additionally, we found that monthly income plays a moderating role in the relationship between work engagement and turnover intention, and in the association between professional titles and turnover intention. Conclusions Greater job satisfaction and engagement, along with reasonable remuneration, were found to be associated with lower turnover intention among healthcare workers. Employers should proactively monitor the dynamic interactions among these factors and then develop more tailored interventions in order to alleviate the ongoing loss of healthcare workers.

In this study, medium-high temperature Daqu (MHTD), the primary saccharification and fermentation agent in the production of strong-aroma Baijiu, was investigated across different fermentation stages. Employing national standard methods, artificial intelligence sensing technology, and high-throughput sequencing techniques, a comprehensive analysis was conducted to investigate the physicochemical properties, flavor, taste, and changes in fungal community structure during the MHTD fermentation process. The correlation between quality indicators of MHTD and dominant fungal genera was also explored. The results revealed that, during fermentation, the moisture content, levels of organic sulfur compounds, terpenes, hydroxides, and astringency gradually decreased in MHTD, while saltiness steadily increased. The fungal community structure of MHTD was established in the early stages of fermentation. However, as fermentation progressed, fungal community diversity gradually decreased, accompanied by significant changes in the relative abundance of certain genera. In order to analyze the high-temperature Daqu bacterial taxa and their physicochemical properties in Linyi area, Shandong Province, Illumina MiSeq high-throughput sequencing technology and pure culture technology were used to analyze the bacterial taxa of high-temperature Daqu in Lanling wine, and then the physicochemical indexes were determined by conventional detection methods in this study. Finally, the correlation between bacterial taxa and physicochemical indexes in Daqu was explored. Inspired by the efficient swimming capabilities of swordfish, a novel wireless soft swordfish‐like robot with programmable magnetization has been developed, integrating direct‐ink‐writing (DIW) 3D printing and assembly technology. This 20 mm long robot features a streamlined form and magnetically programmable movements, enabling biomimetic locomotion patterns such as straight‐line swimming and turning swimming. The robot includes a silicone‐based torso (body, abdomen, and pectoral fin) and a crescent‐shaped tail fin made from a magnetically programmable polymer embedded with neodymium‐iron‐boron (NdFeB) particles. The tail fin, fabricated by multi‐material alternating printing to achieve a gradient magnetism distribution, is controlled by an external magnetic field to mimic the rapid oscillation of a swordfish's tail, achieving a swimming speed of 0.51 BL/ s. The tail fin's asymmetric oscillation amplitudes, adjusted by magnetic field control, allow the robot to transition seamlessly from high‐speed straight swimming to agile turning. The robot can perform tracking swimming along specific planned paths, such as “C” and “Z” shaped trajectories. Potential applications include environmental monitoring and targeted drug release. The multi‐material 3D printing technology enhances the robot's efficiency and sensitivity in simulating natural biological movements, extending to the design and development of various flexible devices and soft robots.

IntroductionGastric glomus tumor is a rare submucosal mesenchymal tumor with no distinct features on endoscopy. In clinical practice, it is often treated with laparoscopic partial gastrectomy. Here, we report a case of gastric glomus tumor successfully resected using a combination of gastroscopy and laparoscopy.Case descriptionThe patient was an elderly male who underwent gastroscopy, which revealed a submucosal mass in the gastric antrum. The lesion was suspected to be a stromal tumor. Further evaluation with computed tomography (CT) imaging indicated a space-occupying lesion in the gastric antrum, with the possibility of benign pathology. Endoscopic ultrasonography revealed that the tumor originated from the muscular layer and was approximately 28.8 mm ×22.5 mm. Blood flow was detected behind the lesion, suggesting the possibility of a gastric glomus tumor. The tumor was removed using a combination of gastroscopy and laparoscopy. Postoperative pathology suggested that it was a benign gastric glomus tumor. The patient recovered uneventfully and was discharged 7 days later.ConclusionGastric glomus tumor is a rare submucosal tumor that should be included in the differential diagnosis of gastric submucosal tumors. A combined approach using gastroscopy and laparoscopy offers a minimally invasive and effective treatment option.

Abstract Solar wind irradiation, as a crucial space weathering mechanism, alters the microscopic characteristics and reflectance spectrum of the lunar regolith, and its cumulative effect is strongly related to the exposure time. Ilmenite is highly resistant to solar wind irradiation. Here, we combined transmission electron microscopy, energy-dispersive X-ray spectroscopy, and electron energy loss spectroscopy to systematically illustrate the diverse space-weathered rims on the shoveled and drilled Chang’e-5 ilmenites resulting from varying degrees of solar wind irradiation, revealing that solar wind plays a key role in the early-stage alteration of exposed lunar soil. Summary: Background: Oesophagogastroduodenoscopy (OGD) quality and identification of the early upper gastrointestinal (UGI) neoplasm play an important role in detecting the UGI neoplasm. However, the optimal method for quality control in daily OGD procedures is currently lacking. We aimed to evaluate the efficacy of a real-time intelligent quality-control system (IQCS), which combines OGD quality control with lesion detection of early UGI neoplasms. Methods: We performed a multicentre, single-blinded, randomised controlled trial at 6 hospitals in China. Patients aged 40–80 years old who underwent painless OGD were screened for enrolment in this study. Patients with a history of advanced UGI cancer, stenosis, or obstruction in UGI tract were excluded. Eligible subjects were randomly assigned (1:1) to either the routine or IQCS group to undergo standard OGD examination and OGD examination aided by IQCS, respectively. Patients were masked to the randomisation status. The primary outcome was the detection of early UGI neoplasms. All analyses were done on a per-protocol basis. This trial is registered with ClinicalTrials.gov, NCT04720924. Findings: Between January 16, 2021 and December 23, 2022, 1840 patients were randomised (IQCS group: 919, routine group: 921). The full analysis set consisted of 914 in the IQCS group and 915 in the routine group. The early UGI neoplasms detection rate in the IQCS group (6.1%, 56/914) was significantly higher than in the routine group (2.3%, 21/915; P = 0.0001). Phellodendron chinense is an extraordinary medicinal plant, and its stem bark is a famous traditional Chinese medicine in China, containing a number of phytochemicals with biological and pharmacological functions. Studies have shown that the medicinal ingredients in P. chinense from various regions are different, as soil types might be important factors affecting the growth and quality of P. chinense. In this study, the effects of acidic purple soil, alkaline purple soil, yellow soil and red soil on the growth, nutrient element accumulation and partitioning in P. chinense seedlings were studied. Our results showed that the biomass of leaves and stems in acidic purple soil was significantly higher than in other soils, and the root biomass was significantly lower than that in yellow soil. C, N, P and Ca contents cultured in acidic purple soil were the highest compared to other soils, and K and Mg content were the highest in yellow soil and red soil, respectively. The distribution of carbon between different organs can reflect environmental stress. N has become a limiting factor for the growth and development of P. chinense seedlings. C:N and C:P ratios in the leaves were significantly lower than those in the stem and roots. Among four soil types, the highest N:P ratio was 1.65 in P. chinense seedlings. These results suggest that four soil types may significantly affect the accumulation and distribution of biomass and nutrient elements in P. chinense seedlings. The characteristic patterns in different organs have different responses. Acidic purple soil is more suitable for the growth of P. chinense. These findings will help to understand the distribution characteristics and requirements of nutrient elements in P. chinense seedlings, and further provide a theoretical basis and reference for site selection and cultivation strategy.

To explore the microbial diversity and flavor profiles of stinky acid, we utilized high-throughput sequencing, culture-based techniques, and bionic E-sensory technologies. The results revealed a significant correlation between the acidity levels of stinky acid and the richness of its microbial community. Ten core bacterial genera and three core fungal genera exhibited ubiquity across all stinky acid samples. Through E-nose analysis, it was found that sulfides constituted the principal odor compounds responsible for stinky acid's distinct aroma. Further insights arose from the correlation analysis, indicating the potential contribution of Debaryomyces yeast to the sour taste profile. Meanwhile, three genera—Rhizopus and Thermoascus and Companilactobacillus—were identified as contributors to aromatic constituents. Interestingly, the findings indicated that Rhizopus and Thermoascus could reduce the intensity of the pungent odor of stinky acid. In summary, this investigation's outcomes offer new insights into the complex bacterial diversity of stinky acid.

The continuous growth of global trade and maritime transport has significantly heightened the challenges of managing ship traffic in port waters, particularly within fairways. Effective traffic management in these channels is crucial not only for ensuring navigational safety but also for optimizing port efficiency. A deep understanding of ship behavior within fairways is essential for effective traffic management. This paper applies machine learning techniques, including Decision Tree, Random Forest, and Gradient Boosting Regression, to model and analyze the behavior of various types of ships at specific moments within fairways. The study focuses on predicting four key behavioral parameters: latitude, longitude, speed, and heading. The experimental results reveal that the Random Forest model achieves adjusted R2 scores of 0.9999 for both longitude and latitude, 0.9957 for speed, and 0.9727 for heading. All three models perform well in accurately predicting ship positions at different times, with the Random Forest model particularly excelling in speed and heading predictions. It effectively captures the behavior of ships within fairways and provides accurate predictions for different types and sizes of vessels, especially in terms of speed and heading variations as they approach or leave berths. This model offers valuable support for predicting ship behavior, enhancing ship traffic management, optimizing port scheduling, and detecting anomalies.

In this study, the physicochemical characteristics and bacterial taxa of 11 fen-flavor Daqu samples from Changzhi, Shanxi Province were analyzed using a combination of physicochemical analysis and MiSeq high-throughput sequencing, and the correlation between them was revealed. On the basis of using electronic tongue and electronic nose to evaluate the quality of Zhaguangjiao, MiSeq high-throughput sequencing technology was used to analyze the bacterial diversity and explore the effect of different salt concentration on its quality. The results showed that with the increase of salt concentration, the taste and flavor indexes changed regularly. At the gate level, Firmicutes (84.87%) gradually increased, while Proteobacteria (13.65%) decreased gradually. At the genus level, the relative content of Lactobacillus increased gradually, and the average relative content was as high as 60.20%. When the salt concentration was 5%, the flora structure of Zhaguangjiao tended to be stable, and strength of good indicators (umami) in flavor and taste was moderate, while the relative intensity of defective indicators (sourness) was generally low. Thus, it can be seen that adding 5% salt to the Zhaguangjiao can significantly improve the quality of the Zhaguangjiao.

In this study, to reveal the physicochemical properties and microbial taxa of Hong Qu produced in Shandong, China three Hong Qu samples were collected from a Shandong Qu-making company, and based on the determination of their physicochemical indexes and fermentation characteristics, the microbial taxa were resolved using MiSeq high-throughput sequencing technology, and the Bacillus and lactic acid bacteria contained in them were isolated and identified. To explore the effects of three pepper varieties on bacterial taxa and quality of Zhaguangjiao, Zhaguangjiao prepared using Capsicum frutescens Linn., Capsicum annuum L., and Capsicum annuum L. var. grossum (L.) Sendt. for the test group, while two control groups without the addition of any vegetables and with the addition of only Apium graveolens var. rapaceum were set up, and based on that, the quality of Zhaguangjiao was digitally evaluated and its bacterial taxa were resolved by high-throughput sequencing. Finally, the correlations between Zhaguangjiao quality and dominant bacterial genera were revealed. It was found that, compared with Zhaguangjiao prepared using Capsicum frutescens Linn., both the bacterial diversity and abundance of Zhaguangjiao prepared using Capsicum annuum L. and Capsicum annuum L. var. grossum (L.) Sendt. were higher. Zhaguangjiao prepared using Capsicum annuum L. and Capsicum annuum L. var. grossum (L.) Sendt. were dominated by Lactobacillus with average relative abundances of 77.53% and 94.41%, respectively, while Zhaguangjiao prepared using Capsicum frutescens Linn. was dominated by Pseudomonas with an average relative abundance of 67.35%. Compared with Zhaguangjiao prepared using Capsicum frutescens Linn., the sourness and content of aromatic compounds of Zhaguangjiao prepared using Capsicum frutescens Linn. was dominated by Pseudomonas were significantly higher, while the contents of organic sulfides, terpenes and nitrogen oxides among volatile substances were significantly lower. After Pearson correlation analysis, it was found that elevating the abundance of Lactobacillus in the fermentation system of Zhaguangjiao could obviously suppress the undesirable flavor and improve the content of aroma substances in Zhaguangjiao. Abstract Purpose The microbial community in the pit mud correlated closely with the quality of the final product of Chinese strong-flavored Baijiu (CSFB). However, environmental conditions and brewing processes can vary by region and distilleries. This may lead to differences in microbial composition and function in pit mud. Therefore, revealing the features of the pit mud microbial community structure and functions of different distilleries will provide key information for understanding the diversity and difference of microbes in the brewing of CSFB, which will be beneficial for the improvement of the quality of pit mud and CSFB in the future. Methods and results Illumina MiSeq sequencing of 16S rRNA gene amplicons was used to analyze the similarities and differences in microbial community structure and function in pit muds of different distilleries located in Shihezi (Xinjiang), Xiangyang (Hubei), and Yibin (Sichuan). At the genus level, Clostridium, Lactobacillus, Aminobacterium, Petrimonas, Syntrophomonas, Methanoculleus, Syntrophaceticus, Sedimentibacter, Caloramator, Ruminococcus, Bacillus, Methanosarcina, and Garciella were the dominated genera of pit muds. There were great differences in the composition of microorganisms in pit muds used by different distilleries. The significantly enriched prokaryotic microbiotas of pit muds collected in the distilleries of Xiangyang were mainly affiliated with Bacillus, Lactobacillus, and Croceifilum, and the relative abundance of methanogens, such as Methanomicrobia and Methanobacteria, were only significantly enriched in the pit mud collected from the distilleries of Yibin (P < 0.05). Functional analysis indicated that the difference of microbial composition in pit mud will further lead to significant differences in various metabolic functions. Conclusion The compositions and functions of dominant microorganisms in pit mud used for the production of CSFB by different enterprises across regions in China were greatly different, and there was a close relationship between the compositions and functions of microorganisms in pit mud. Therefore, it may be an effective method to improve CSFB fermentation processes by directionally regulating the microbial community functions of pit mud using specific strains.

In order to realize the efficient removal of sulfamethazine (SMZ) and overcome the recycling difficulty of catalysts in a heterogeneous catalytic ozonation process, low-cost natural zeolite loaded with CeO2 and Fe3O4 (FC/HZ) was synthesized by the co-precipitation method. The FC/HZ catalyst could completely degrade SMZ and realize 39.6% removal of TOC through catalytic ozonation. Moreover, the FC/HZ catalyst exhibited high catalytic ozonation activity under a wide pH range and possessed excellent recyclability (>90%) and stability with a magnet. Based on experiment and material characterization, the possible catalytic mechanism and degradation pathway of SMZ was revealed. In brief, this study offered an effective strategy for preparing recyclable catalysts and eliminating SMZ in aqueous solution.

To improve the accuracy of color image completion with missing entries, we present a recovery method based on generalized higher-order scalars. We extend the traditional second-order matrix model to a more comprehensive higher-order matrix equivalent, called the “t-matrix” model, which incorporates a pixel neighborhood expansion strategy to characterize the local pixel constraints. This “t-matrix” model is then used to extend some commonly used matrix and tensor completion algorithms to their higher-order versions. We perform extensive experiments on various algorithms using simulated data and publicly available images. The results show that our generalized matrix completion model and the corresponding algorithm compare favorably with their lower-order tensor and conventional matrix counterparts.

In order to study the power generation characteristics of a self-biased dielectric elastomer generator (DEG), a charge pump circuit applied to a DEG was designed based on the charge pump principle after analyzing the structure and power generation mechanism of DEG. A self-biased DEG model was established in MATLAB/Simulink software. The model was simulated and the factors on the output voltage of DEG, such as the series and capacitance of the charge pump circuit, were studied. Furthermore, the corresponding test platform was built. Then, the comparative analysis between the test results and simulation results was studied. The test and simulation results show that the combination of the charge pump circuit and DEG could realize the function of self-biased power generation. The larger the charge pump series, the higher the output voltage of DEG. The test study found that the output voltage of DEG was higher when the charge pump series was 3. The larger the capacitance of the charge pump, the lower the output voltage of DEG. When the capacitance of the charge pump was too large, the boosted voltage of DEG was not obvious. However, if the input voltage was low, the output voltage of DEG was high when the charge pump capacitance value was large. The above results provide the basis for further study of the self-biased DEG.

The substitution of noble metals with cost-effective copper nanoparticles (Cu NPs) in the preparation of metal/semiconductor composite catalysts holds significant environmental and economic implications for the degradation of various organic pollutants. However, the development of highly active and stable Cu NPs catalysts has emerged as a key challenge in the progression of non-noble metal catalysts. In this study, the reducibility of glutathione (GSH) was employed to reduce Cu2+ to Cu NPs, resulting in the formation of stable Cu-GSH nanoparticles through S–H bonds. An electrostatic self-assembly strategy was used to load Cu-GSH onto WO3 nanorods, thereby designing a Cu GSH/WO3 catalytic material with highly efficient charge transport efficiency. Under visible light irradiation, 3 wt% Cu GSH/WO3 demonstrated excellent degradation performance for organic pollutants, achieving the degradation of 99.8% of Rh B and 98.6% of TC within 60 minutes. Experimental results from photoelectrochemical (PEC) and electron spin resonance (ESR) analyses indicated that Cu GSH functions as an efficient electron trap, which triggers electron flow driven by the Schottky barrier, capturing the photoexcited electrons from WO3. This greatly enhances the separation efficiency of WO3 carriers and extends the lifetime of the carriers. The ubiquitous presence of magnetite in lunar soil has been hypothesized in previous Apollo Mössbauer spectroscopy and electron spin resonance studies, but there is currently no mineralogical evidence to prove it. Here, we report a large number of submicroscopic magnetite particles embedded within iron-sulfide on the surface of Chang'e-5 glass, with a close positive correlation between magnetite content and the TiO2 content of the surrounding glass. The morphology and mineralogy of the iron-sulfide grains suggest that these magnetite particles formed via an impact process between iron-sulfide droplets and silicate glass melt, and ilmenite is necessary for magnetite formation. Magnetite in lunar glass is a potential candidate for the "magnetite-like" phase detected in the Apollo era and suggests that impact-induced submicroscopic magnetite may be ubiquitous in high-Ti regions of the Moon. Background and aimMagnifying image-enhanced endoscopy was demonstrated to have higher diagnostic accuracy than white-light endoscopy. However, differentiating early gastric cancers (EGCs) from benign lesions is difficult for beginners. We aimed to determine whether the computer-aided model for the diagnosis of gastric lesions can be applied to videos rather than still images.MethodsA total of 719 magnifying optical enhancement images of EGCs, 1,490 optical enhancement images of the benign gastric lesions, and 1,514 images of background mucosa were retrospectively collected to train and develop a computer-aided diagnostic model. Subsequently, 101 video segments and 671 independent images were used for validation, and error frames were labeled to retrain the model. Finally, a total of 117 unaltered full-length videos were utilized to test the model and compared with those diagnostic results made by independent endoscopists.ResultsExcept for atrophy combined with intestinal metaplasia (IM) and low-grade neoplasia, the diagnostic accuracy was 0.90 (85/94). The sensitivity, specificity, PLR, NLR, and overall accuracy of the model to distinguish EGC from non-cancerous lesions were 0.91 (48/53), 0.78 (50/64), 4.14, 0.12, and 0.84 (98/117), respectively. No significant difference was observed in the overall diagnostic accuracy between the computer-aided model and experts. A good level of kappa values was found between the model and experts, which meant that the kappa value was 0.63.ConclusionsThe performance of the computer-aided model for the diagnosis of EGC is comparable to that of experts. Magnifying the optical enhancement model alone may not be able to deal with all lesions in the stomach, especially when near the focus on severe atrophy with IM. These results warrant further validation in prospective studies with more patients. Abstract Background Aortic intramural hematoma (IMH) is a subset of acute aortic syndrome, and its prognosis may differ between races. This study aimed to study the prognosis of Chinese type B IMH patients and to find out risk factors. Methods A total of 71 type B IMH patients with or without penetrating atherosclerosis ulcer (PAU) administrated in our center between September 2013 and October 2017 were retrospectively studied. Both clinical and imaging data were collected and analyzed. The primary end point was aorta-related death, and the secondary end point was progression, which was defined as enlargement of aorta, increased aortic wall thickness, and aortic dissection or aneurysm formation. Kaplan-Meier survival analysis and Cox regression analysis were used for prognostic analysis. Results Among these 71 patients, 21 had simple type B IMH, when 50 had type B IMH in association with PAU. Twenty-five patients received optimal medical therapy (OMT) alone, while 46 patients received surgery and OMT. The mean follow-up time was 27.5 ± 13.5 months. For type B IMH patients, association with PAU indicated poor prognosis and required more intensive management (HR = 16.68, 1.96~141.87), while maximum aortic diameter (MAD) was an independent risk factor (HR = 1.096, 1.016~1.182). For patients with PAU-IMH, MAD was an independent risk factor (HR = 1.04, 1.021~1.194), while surgical treatment was independent protective factor (HR = 0.172, 0.042~0.696). Conclusion Association with PAU and MAD were independent risk factors for type B IMH patients. Surgery may improve the outcomes for type B IMH in association with PAU.

ABSTRACT Bifidobacterium longum predominates in the human gut throughout the life span, from birth to old age, and could alter the intestinal microbial population and immune function in the elderly. We investigated the intestinal bacterial diversity in the elderly, and further evaluated the genetic diversity and population structure of B. longum. The results revealed a distinct difference in gut bacterial populations between the elderly from Xiangyang and its neighboring region, Enshi city. A total of 62 bifidobacterial strains were isolated, 30 of which were found to be B. longum. The multilocus sequence typing (MLST) analysis also revealed that 437 B. longum isolates from diverse regions worldwide, including the 30 isolated in this study, could be classified into 341 sequence types (STs). They could be further clustered into 10 clonal complexes and 127 singleton STs, indicating a highly genetic diversity among B. longum isolates. Two putative clone complexes (CCs) containing the isolates from Xiangyang were found to be geographically specific, and a 213-bp recombination fragment was detected. Phylogenetic trees divided these 437 isolates into three lineages, corresponding to the three subspecies of B. longum. It is noteworthy that two isolates from the elderly were identified to be B. longum subsp. suis, while the others were B. longum subsp. longum. Together, our study characterized the intestinal bacterial diversity and evolution of B. longum in the elderly, and it could contribute to further studies on the genotyping and discrimination of B. longum. IMPORTANCE Bifidobacterium longum are common inhabitants of the human gut throughout the life span, and have been associated with health-promoting effects, yet little is known about the genotype profile and evolution of these isolates. Our study showed that there was significant difference in gut bacterial community and abundance of B. longum between the elderly from two neighboring cities. Furthermore, the possible geographically specific STs, CCs, and intraspecies recombination fragment were found among the B. longum isolates from elderly.

Microorganisms in pit mud are the essential factor determining the style of strong flavor Baijiu. The spatial distribution characteristics of fungal communities and aroma in the pit mud for strong flavor Baijiu from Xinjiang, China, were investigated using Illumina MiSeq high-throughput sequencing and electronic nose technology. A total of 138 fungal genera affiliated with 10 fungal phyla were identified from 27 pit mud samples; of these, Saccharomycopsis, Aspergillus, and Apiotrichum were the core fungal communities, and Aspergillus and Apiotrichum were the hubs that maintain the structural stability of fungal communities in pit mud. The fungal richness and diversity, as well as aroma of pit mud, showed no significant spatial heterogeneity, but divergences in pit mud at different depths were mainly in pH, total acid, and high abundance fungi. Moisture, NH4+, and lactate were the main physicochemical factors involved in the maintenance of fungal stability and quality in pit mud, whereas pH had only a weak effect on fungi in pit mud. In addition, the fungal communities of pit mud were not significantly associated with the aroma. The results of this study provide a foundation for exploring the functional microorganisms and dissecting the brewing mechanism of strong flavor Baijiu in Xinjiang, and also contributes to the improvement of pit mud quality by bioaugmentation and controlling environmental physicochemical factors.

Fen flavor Baijiu needs two rounds of fermentation, which will obtain Dacha after initial fermentation and Ercha after secondary fermentation. The quality of Baijiu is closely related to the microbes within fermented grains. However, the bacterial diversity in Dacha and Ercha fermented grains of Fen flavor Baijiu has not been reported. In the present study, the structure and diversity of bacteria communities within fermented grains of Fen flavor Baijiu were analyzed and evaluated using MiSeq platform’s HTS with a sequencing target of the V3-V4 region of the 16S rRNA gene. Through the analysis of physical and chemical indexes and electronic senses, the relationship between bacterial flora, organic acid, taste, and aroma in fermented grains was clarified. The results indicated that Lactobacillus was the main bacteria in Dacha, and the mean relative content was 97.53%. The bacteria within Ercha samples were Pseudomonas and Bacillus, mean relative content was 37.16 and 28.02%, respectively. The diversity of bacterial communities in Ercha samples was significantly greater than that in Dacha samples. The correlation between Lactobacillus and organic acids, especially lactic acid, led to the difference between Dacha and Ercha organic acids, which also made the pH value of Dacha lower and the sour taste significantly higher than Ercha. Lactobacillus was significantly positively correlated with a variety of aromas, which made Dacha the response value of aromas higher. In addition, Bacillus had a significant positive correlation with bitterness and aromatic compounds, which led to a higher response value of bitterness in Ercha and made it present an aromatic aroma. This study provides an in-depth analysis of the difference between different stages of Fen flavor Baijiu, and theoretical support for the standard production and improvement in quality of Fen flavor Baijiu in the future.

Rice wine koji, a traditional homemade starter culture in China, is nutritious and delicious. The final quality of rice wine koji is closely related to the structure of its microbial community. However, the diversity of natural microorganisms in rice wine koji from different regions has not been evaluated. In this study, the microbial population of 92 naturally fermented rice koji samples collected from Hubei, Guangxi, and Sichuan was systematically analyzed by high-throughput sequencing. From all the rice wine koji samples, 22 phyla and 479 bacterial genera were identified. Weissella, Pediococcus, Lactobacillus, Enterobacter, Lactococcus, Pantoea, Bacillus, Staphylococcus, and Leuconostoc were the dominant genera in rice wine koji. The bacterial community structure of rice wine koji samples from different regions was significantly different (p < 0.05). The bacterial community composition of the samples from Hubei and Guangxi was similar, but significantly different from that of SC samples (p < 0.05). These differences may be caused by variations in geography, environment, or manufacturing. In addition, the results of microbial phenotype prediction by BugBase and bacterial functional potential prediction by PICRUSt showed that eight of the nine predicted phenotypic functions of rice wine koji samples from different regions were significantly different (p < 0.05) and that vigorous bacterial metabolism occurred in rice wine koji samples.
