Abstract Automated and non-invasive mammalian cell analysis is currently lagging behind due to a lack of methods suitable for a variety of cell lines and applications. Here, we report the development of a high throughput non-invasive method for tracking mammalian cell growth and performance based on plate reader measurements. We show the method to be suitable for both suspension and adhesion cell lines, and we demonstrate it can be adopted when cells are grown under different environmental conditions. We establish that the method is suitable to inform on effective drug treatments to be used depending on the cell line considered, and that it can support characterisation of engineered mammalian cells over time. This work provides the scientific community with an innovative approach to mammalian cell screening, also contributing to the current efforts towards high throughput and automated mammalian cell engineering.

Objective: Buyang-Huanwu Decoction is often used as an adjuvant therapy for ischemic stroke in clinical practice. At the same time, various laboratories have found that it has the potential to treat ischemic heart failure, lower extremity varicose veins, pulmonary hypertension, pulmonary fibrosis, and hyperlipidemia. Previous studies in our laboratory have found that different concentrations of ethanol were used to separate the polar components of Buyang-Huanwu decoction, and the therapeutic effects of each group on pulmonary fibrosis and hyperlipidemia were different. Therefore, we analyzed and explored Buyang Huanwu decoction and its products. Methods: The isolated components were obtained by treatment with absolute ethanol, 30% (v/v), 50% (v/v), 75% (v/v), and 90% (v/v) ethanol, and the components were analyzed by UPLC-MS. Results: The separation method used in our laboratory could well separate the components of Buyang Huanwu Decoction, and simply enrich carbohydrates, organic acids, phenylpropanoid compounds, lipids, nucleotides, and lignans. Conclusions: The products treated with different concentrations of ethanol have different therapeutic potentials. The high content components of 30% ethanol eluate and 50% ethanol eluate have good therapeutic effects on osteoarthritis and bone loss, while the high content components of the precipitate components and 75% eluate are more likely to be related to glucose and lipid metabolism disorders and cardiovascular diseases. SUMMARY: While it is known that increased dissolved CO2 concentrations and rising sea surface temperature (ocean warming) can act interactively on marine phytoplankton, the ultimate molecular mechanisms underlying this interaction on a long‐term evolutionary scale are relatively unexplored. Here, we performed transcriptomics and quantitative metabolomics analyses, along with a physiological trait analysis, on the marine diatom Thalassiosira weissflogii adapted for approximately 3.5 years to warming and/or high CO2 conditions. We show that long‐term warming has more pronounced impacts than elevated CO2 on gene expression, resulting in a greater number of differentially expressed genes (DEGs). The largest number of DEGs was observed in populations adapted to warming + high CO2, indicating a potential synergistic interaction between these factors. We further identified the metabolic pathways in which the DEGs function and the metabolites with significantly changed abundances. We found that ribosome biosynthesis‐related pathways were upregulated to meet the increased material and energy demands after warming or warming in combination with high CO2. This resulted in the upregulation of energy metabolism pathways such as glycolysis, photorespiration, the tricarboxylic acid cycle, and the oxidative pentose phosphate pathway, as well as the associated metabolites. These metabolic changes help compensate for reduced photochemical efficiency and photosynthesis. Our study emphasizes that the upregulation of ribosome biosynthesis plays an essential role in facilitating the adaptation of phytoplankton to global ocean changes and elucidates the interactive effects of warming and high CO2 on the adaptation of marine phytoplankton in the context of global change. Although the diverse impacts of elevated dissolved CO2 and warming on organisms within various trophic levels in marine food webs are well documented, we have yet to explore the biological links across different levels of biological organization from primary producers to secondary producers on an evolutionary time scale in a high‐CO2 ocean. Here, we cultured a model marine diatom Phaeodactylum tricornutum (primary producer) in predicted future high‐CO2 and/or warming conditions for ~ 1250 d with an experimental evolution approach and then fed them to the clam Coelomactra antiquata (secondary producer). We present an in‐depth multi‐omics analysis along the methylome (primary producer)–transcriptome (primary producer)–metabolome (primary producer)–metabolome (secondary producer) continuum. Our results showed that the downregulated terpenoid backbone biosynthesis in the methylome and transcriptome lead to decreased pyruvate levels and upregulation of some pathways (such as phenylalanine metabolism) in the metabolome of the primary producer in the long‐term warming conditions. These changes in metabolomic profile in the primary producer were then transferred to the secondary producer, resulting in changes in abundance of some metabolites, such as decreases in pyruvate, and in pyruvaldhyde (also known as methylglyoxal), and increases in 2‐hydroxylamino‐4,6‐dinitrotoluene. Our study provides a new insight into the molecular mechanisms underlying the trophic transfer from primary to secondary producers in a future high‐CO2 ocean and may provide more accurate projections of marine ecosystem services and functions over the next century. Genetic changes together with epigenetic modifications such as DNA methylation have been demonstrated to regulate many biological processes and thereby govern the response of organisms to environmental changes. However, how DNA methylation might act cooperatively with gene transcription and thereby mediate the long-term adaptive responses of marine microalgae to global change is virtually unknown. Here we performed a transcriptomic analysis, and a whole-genome bisulfite sequencing, along with phenotypic analysis of a model marine diatom Phaeodactylum tricornutum adapted for 2 years to high CO2 and/or warming conditions. Our results show that the methylated islands (peaks of methylation) mCHH were positively correlated with expression of genes in the subregion of the gene body when the populations were grown under high CO2 or its combination with warming for ~2 years. We further identified the differentially expressed genes (DEGs), and hence the metabolic pathways in which they function, at the transcriptomics level in differentially methylated regions (DMRs). Although DEGs in DMRs contributed only 18–24% of the total DEGs, we found that those DEGs acted cooperatively with DNA methylation and then regulated key processes such as central carbon metabolism, amino acid metabolism, ribosome biogenesis, terpenoid backbone biosynthesis, and degradation of misfolded proteins. Taken together, by integrating transcriptomic, epigenetic, and phenotypic analysis, our study provides evidence for DNA methylation acting cooperatively with gene transcription to contribute to the adaptation of microalgae to global changes. The multifaceted nature of firm innovation has prevented researchers from fully explaining the relationship between firm innovation and green management. This study, building on the Schumpeterian theory of innovation, explores this relationship by examining three major types of firm innovation—strategic innovation, managerial innovation, and product innovation—and their respective relationships with green management, considering several dimensions of environmental turbulence as distinctive boundary conditions. We propose that both strategic innovation and managerial innovation facilitate green management, which in turn mediates these effects on new product performance. The results of a survey of 303 Chinese firms provide strong support for this mediating logic. Moreover, we find that market turbulence weakens the effect of strategic innovation on green management whereas technological turbulence strengthens such effect but the effect of managerial innovation on green management is not influenced by environmental turbulence. Our research contributes to the innovation as well as green management and sustainability literatures by offering a framework in which to analyze firm innovation and green management and by showing how firms pursue sustainability and prosperity under specific environmental conditions.
