Antibiotics are currently used for the treatment of Helicobacter pylori (H. pylori), which is confirmed to be the major cause of gastric disorders. However, the long-term consumption of antibiotics has already caused antibiotic resistance and side effects in vivo. Therefore, there is an emerging need for searching for safe and effective anti-H. pylori agents. Inspired by the excellent bioactivities of cinnamic acid, a series of cinnamic acid derivatives (compounds 1–30) were synthesized and determined for H. pylori inhibition. The initial screening revealed that compound 23, a 2,4-dinitro cinnamic acid derivative containing 4-methoxyphenol, showed excellent H. pylori inhibition with an MIC value of 4 μM. Further studies indicated that compound 23 showed anti-bacterial activity and had a bactericidal effect on H. pylori due to the destruction of the bacterial structure. Molecular docking analysis revealed that the 2,4-dinitro groups in cinnamic acid moiety formed hydrogen bonding with amino acid residues in an active pocket of H. pylori protein. Interestingly, the ester moiety fitted into the hydrophobic pocket, attaining additional stability to compound 23. Above all, the present study reveals that compound 23 could be considered a promising anti-H. pylori agent to treat H. pylori causing gastritis.

Carbon capture, utilization, and storage (CCUS) has been widely applied to enhance oil recovery (CO2-EOR). A thorough investigation of the impact of injecting CO2 into a heterogeneous reservoir is critical to understanding the overall reservoir robustness and storage performance. We conducted fifteen flow-through tests on Morrow B sandstone that allowed for chemical reactions between a CO2-rich brackish solution and the sandstones, and four creep/flow-through tests that simultaneously allowed for chemical reactions and stress monitoring. From fluid chemistry and X-ray computed tomography, we found that the dissolution of disseminated cements and the precipitation of iron-rich clays did not significantly affect the permeability and geomechanical properties. Minor changes in mechanical properties from Brazilian and creep tests indicated that the matrix structure was well-supported by early diagenetic quartz overgrowth cement and the reservoir’s compaction history at deep burial depths. However, one sample experienced a dissolution of poikilotopic calcite, leading to a permeability increase and significant tensile strength degradation due to pore opening, which overcame the effect of the early diagenetic cements. We concluded that the Morrow B sandstone reservoir is robust for CO2 injection. Most importantly, cement timing, the abundance and texture of reactive minerals, and the reservoir’s burial history are critical in predicting reservoir robustness and storage capacity for CO2 injection.

Resveratrol (RES) is a natural polyphenolic compound with a broad range of physiological and pharmacological properties. Previous studies have shown that RES also plays an important role in protecting and promoting early bone metabolism and differentiation. The accumulation of cadmium (Cd), one of the world’s most poisonous substances, can inhibit skeletal growth and bone maturation, thus causing osteoporosis. However, whether RES can prevent the Cd-induced inhibition of osteogenic differentiation remains unknown. In this study, we found that RES promoted the early maturity of osteoblastic MC3T3-E1 cells, as demonstrated by the significantly increased mRNA and protein expression of a range of differentiation markers, including alkaline phosphatase (ALP), collagen 1 (COL1), bone morphogenetic protein-2 (BMP-2), and runt-related transcription factor 2 (RUNX2). In contrast, we found that cadmium chloride (CdCl2) inhibited the viability and osteogenic maturity of MC3T3-E1 cells. We also demonstrated that RES pretreatment for 30 min provided significant protection against Cd-induced apoptosis and attenuated the inhibition of osteogenic differentiation induced by Cd by modulating ERK1/2 and JNK signaling. In conclusion, our results indicate that RES is a potential femoral protectant that not only enhance the viability and early differentiation of osteoblasts, but also protect osteoblasts from cadmium damage.

Cadmium is an environmental metal pollutant that has been a focus of research in recent years, which is reported to cause bone disease; however, its skeletal toxicity and the mechanism involved are not yet fully known. Therefore, this study used MC3T3-E1 subclone 14 cells to determine the mechanism of cadmium toxicity on bone. Cadmium chloride (Cd) significantly reduced cell viability in a concentration-dependent manner. Exposure to Cd inhibited osteoblast-related proteins (Runx2, Col-1, STC2) and decreased alkaline phosphatase (ALP) activity. Cd caused Exportin-1 accumulation and induced DNA damage. Cd significantly down-regulated caspase 9 and induced cleaved-PARP, cleaved-caspase 3 protein level. Treatment with JNK inhibitor, SP600125, suppressed cadmium-induced elevation in the ratio of phosphorylation of JNK to JNK. Inhibition of caspase with pan-caspase inhibitor, Z-VAD-FMK, prevented MC3T3-E1 subclone 14 cells from cadmium-induced reduction of Runx2, STC2, caspase 9, and accumulation of cleaved PARP and cleaved caspase 3. Cd-induced cell survival enhanced by SP600125 but rescued by Z-VAD-FMK or KPT-335. These results suggest that cadmium cytotoxicity on bone involved exportin 1 accumulation, phosphorylation of JNK, induction of DNA damage and pro-apoptosis, which was induced by activation of caspase-dependent pathways.

Mineral reactive surface area (RSA) is one of the key factors that control mineral reactions, as it describes how much mineral is accessible and can participate in reactions. This work aims to evaluate the impact of mineral RSA on numerical simulations for CO2 storage at depleted oil fields. The Farnsworth Unit (FWU) in northern Texas was chosen as a case study. A simplified model was used to screen representative cases from 87 RSA combinations to reduce the computational cost. Three selected cases with low, mid, and high RSA values were used for the FWU model. Results suggest that the impact of RSA values on CO2 mineral trapping is more complex than it is on individual reactions. While the low RSA case predicted negligible porosity change and an insignificant amount of CO2 mineral trapping for the FWU model, the mid and high RSA cases forecasted up to 1.19% and 5.04% of porosity reduction due to mineral reactions, and 2.46% and 9.44% of total CO2 trapped in minerals by the end of the 600-year simulation, respectively. The presence of hydrocarbons affects geochemical reactions and can lead to net CO2 mineral trapping, whereas mineral dissolution is forecasted when hydrocarbons are removed from the system.
