Soil salinity is a major abiotic stress limiting crop production globally. Oat (Avena sativa) is an annual cereal with a strong salt tolerance, a high yield, and nutritional quality, although the mechanisms underlying its salt stress response remain largely unknown. We examined the physiological and transcriptomic responses of A. sativa seedlings to salt stress in tolerant cultivar Qingyongjiu 195 and sensitive cultivar 709. Under salt stress, Qingyongjiu 195 maintained a higher photosynthetic efficiency, antioxidant enzymes activity, and leaf K+ accumulation but a lower Na+ uptake than 709. RNA-seq revealed 6616 differentially expressed genes (DEGs), including 4265 up- and 2351 downregulated. These were enriched in pathways like plant–pathogen interaction, phenylpropanoid biosynthesis, and MAPK signaling. We specifically highlight DEGs involved in photosynthesis (chlG, CP47 psbB, COX2, LHCB) and antioxidants (trxA, GroES). Qingyongjiu 195 also appeared to enhance K+ uptake via KAT1 and AKT2 and sequester Na+ in vacuoles via NHX2. Additionally, HKT restricted Na+ while promoting K+ transport to shoots, maintaining K+/Na+. The expression levels of CAX, ACA, CML, CaM, and CDPK in Qingyongjiu 195 were higher than those in 709. Oats regulated Ca2+ concentration through CAX and ACA after salt stress, decoded Ca2+ signals through CML, and then transferred Ca2+ signals to downstream receptors through the Ca2+ sensors CaM and CDPK, thereby activating K+/Na+ transporters, such as SOS1 and NHX, etc. Our results shed light on plant salt stress response mechanisms and provide transcriptomic resources for molecular breeding in improving salt tolerance in oats. Poor selectivity to tumor cells is a major drawback in the clinical application of the antitumor drug doxorubicin (DOX). Peptide–drug conjugates (PDCs) constructed by modifying antitumor drugs with peptide ligands that have high affinity to certain overexpressed receptors in tumor cells are increasingly assessed for their possibility of tumor-selective drug delivery. However, peptide ligands composed of natural L-configuration amino acids have the defects of easy enzymatic degradation and insufficient biological stability. In this study, two new PDCs (LT7-SS-DOX and DT7-SS-DOX) were designed and synthesized by conjugating a transferrin receptor (TfR) peptide ligand LT7 (HAIYPRH) and its retro-inverso analog DT7 (hrpyiah), respectively, with DOX via a disulfide bond linker. Both conjugates exhibited targeted antiproliferative effects on TfR overexpressed tumor cells and little toxicity to TfR low-expressed normal cells compared with free DOX. Moreover, the DT7-SS-DOX conjugate possessed higher serum stability, more sustained reduction-triggered drug release characteristics, and stronger in vitro antiproliferative activity as compared to LT7-SS-DOX. In conclusion, the coupling of antitumor drugs with the DT7 peptide ligand can be used as a promising strategy for the further development of stable and efficient PDCs with the potential to facilitate TfR-targeted drug delivery. Summary: Abiotic stresses such as salinity, heat and drought seriously impair plant growth and development, causing a significant loss in crop yield and ornamental value. Biotechnology approaches manipulating specific genes prove to be effective strategies in crop trait modification. The Arabidopsis vacuolar pyrophosphatase gene AVP1, the rice SUMO E3 ligase gene OsSIZ1 and the cyanobacterium flavodoxin gene Fld have previously been implicated in regulating plant stress responses and conferring enhanced tolerance to different abiotic stresses when individually overexpressed in various plant species. We have explored the feasibility of combining multiple favourable traits brought by individual genes to acquire superior plant performance. To this end, we have simultaneously introduced AVP1, OsSIZ1 and Fld in creeping bentgrass. Transgenic (TG) plants overexpressing these three genes performed significantly better than wild type controls and the TGs expressing individual genes under both normal and various abiotic stress conditions, exhibited significantly enhanced plant growth and tolerance to drought, salinity and heat stresses as well as nitrogen and phosphate starvation, which were associated with altered physiological and biochemical characteristics and delicately fine‐tuned expression of genes involved in plant stress responses. Our results suggest that AVP1, OsSIZ1 and Fld function synergistically to regulate plant development and plant stress response, leading to superior overall performance under both normal and adverse environments. The information obtained provides new insights into gene stacking as an effective approach for plant genetic engineering. Leaf blight disease (LBD) caused by Pantoea agglomerans is a newly emerged oat disease in China that causes great economic and yield losses of oat production. Laboratory and field experiments were conducted to evaluate the efficacy of 13 antimicrobial agents against P. agglomerans and recommend agents with better control efficacy for use. The antibacterial potential of four concentrations of each agent was evaluated under in vitro conditions. Zhongshengmycin and ethylicin were the most effective in inhibiting the growth of P. agglomerans; their bacteriostatic rates (85.0% and 84.8%, respectively) were higher while EC50 lower (0.31 and 0.90, respectively) than other agents. The greenhouse and field experiment suggested that the greatest fresh weight per plant (57.26 g and 57.93 g), forage yield (22.53 t/ha and 22.21 t/ha) and control efficacy (>75%) were obtained after spraying twice with zhongshengmycin or ethylicin in the LBD occurrence phase, with lower yields and control by kasugamycin + dicopper chloride trihydroxide, kasugamycin + oxine copper, benziothiazolinone, copper hydroxide, phenanthrenecarboxylicacid, copper, picoxystrobin, chloroisobromine cyanuric acid, oxathiapiprolin or azoxystrobin. Large-size images cannot be directly put into GPU for training and need to be cropped to patches due to GPU memory limitation. The commonly used cropping methods before are random cropping and sequential cropping, which are crude and fatally inefficient. First, categories of datasets are often imbalanced, and just simple cropping misses an excellent opportunity to make the data distribution balanced. Second, the training needs to crop a large number of patches to cover all patterns, which greatly increases the training time. This problem is of great practical hazards but is often overlooked by previous works. The optimal solution is to generate valuable patches. Valuable patches refer to the value to network training, i.e., the value of this patch for the convergence of the network, and the improvement of the accuracy. To this end, we propose a data-related patch proposal strategy to sample high valuable patches. The core idea is to score each patch according to the accuracy of each category, so as to perform balanced sampling. Compared with random cropping or sequential cropping, our method can improve the segmentation accuracy and accelerate the training vastly. Lead sulfide colloidal quantum dots (PbS CQDs) are promising optoelectronic materials due to their unique properties, such as tunable band gap and strong absorption, which are of immense interest for application in photodetectors and solar cells. However, the tunable band gap of PbS CQDs would only cover visible short-wave infrared; the ability to detect longer wavelengths, such as mid- and long-wave infrared, is limited because they are restricted by the band gap of the bulk material. In this paper, a novel photodetector based on the synergistic effect of PbS CQDs and bismuth telluride (Bi2Te3) was developed for the detection of a mid-wave infrared band at room temperature. The device demonstrated good performance in the visible-near infrared band (i.e., between 660 and 850 nm) with detectivity of 1.6 × 1010 Jones at room temperature. It also exhibited photoelectric response in the mid-wave infrared band (i.e., between 4.6 and 5.1 μm). The facile fabrication process and excellent performance (with a response of up to 5.1 μm) of the hybrid Bi2Te3/PbS CQDS photodetector are highly attractive for many important applications that require high sensitivity and broadband light detection. Oat has been gaining renewed interest due to its role in a healthy human diet, in animal feed, and as a source of high value compounds with industrial applications. Nitrogen fertilization and planting density are two of the most important crop management practices that affect the formation of yield components and final yield of oat. A 2 year 3 × 5 factorial field experiment was conducted to investigate the effects of nitrogen (N) fertilizer and planting density on the flag leaf photosynthetic characteristics, grain yield, and yield components under rainfed conditions. The experiment consisted of three sowing densities (60, 180, and 300 kg·ha−1) and five nitrogen fertilizer rates (0, 45, 90, 135, and 180 kg·ha−1). Results showed that the grain yield was significantly (p < 0.05) correlated with the leaf net photosynthesis rate (Pn), stomatal conductance (Gs), intercellular CO2 concentration (Ci), transpiration rate (Tr), water-use efficiency (WUE), stomatal limitation value (Ls), chlorophyll content (SPAD value), leaf area index (LAI), panicle length, number of spikelets per panicle (NSP), number of grains per panicle (NGP), weight of grains per panicle (WGP), and 1000-kernel weight. Among the yield components, grain yield was driven by number of spikelets per panicle (NSP) and number of grains per panicle (NGP) at low (or high) planting density with low N supply, whereas, at high N supply, 1000-kernel weight was also an important factor for yield. Nitrogen fertilizer and sowing density had significant (p < 0.05) effects on the flag leaf photosynthetic characteristics, grain yield, and yield components of oat. The yield components increased and then decreased with the increase in nitrogen fertilizer, while they decreased with the increase in planting density. The maximum values (p < 0.05) of grain yield were observed in the nitrogen fertilization of 90 kg·ha−1 and sowing density of 180 kg·ha−1 treatment in both growing seasons, mainly contributing to the improved leaf photosynthesis traits (Pn, Gs, Tr, Ls, SPAD, and LAI). The combination of nitrogen fertilization of 90 kg·ha−1 and sowing density of 180 kg·ha−1 is suitable for oat production on a cool semiarid plateau or other agroecozones with similar environmental conditions. Soil desertification is a serious problem in arid northwestern China that threatens ecological sustainability. Artemisia sphaerocephala, a dominant shrub species, play an important role in the conservation of water and the restoration of soil in the desert ecosystem. However, the poor establishment of A. sphaerocephala often limits plant revegetation, and the optimal population density for sustainable growth is largely unknown. Here, we determined key soil properties and plant growth characteristics associated with different population densities of A. sphaerocephala (including from 1.1, 2.1, 3.1, 3.9 to 5.3 plants per m2) in the resource-limited Alashan desert of northwestern China. The results showed that plant population density was the primary factor determining the revegetation of A. sphaerocephala, followed by soil water availability. Soil N, P and K content, and soil fractal dimensions also contributed to the vegetation and productivity. Soil nutrients were mostly accumulated in the topsoil layers, coincidental with the root distribution pattern in which 57% to 82% of total roots were distributed in the top 20 cm soil layer. The concentrations of soil nutrients in higher population densities (3.9 to 5.3 plants per m2) were greater than those in lower population densities (1.1 to 2.1 plants per m2), suggesting that A. sphaerocephala may have the ability to promote nutrient cycling in the desert ecosystem. To evaluate the effects of temperature and lactic acid bacteria (LAB) inoculants on oat silage in Loess Plateau of China, oat was harvested at dough stage, inoculated without (Control) or with LAB inoculants Synlac I (SLI, Lactobacillus plantarum and Pedioccocus acidilactici) and a selected strain HT1 (L. rhamnosus) and ensiled at 25°C (T25), 35°C (T35) and 45°C (T45). The fermentation quality was measured after 60 d of ensiling and the aerobic exposure was conducted at 30°C for 9 d. The results showed that control silage (stored at 25°C) had better fermentation quality than that ensiled at 35°C or 45°C. High temperature of 45°C resulted in sharp decreases in LAB counts and lactic acid concentration and increases in pH and NH3-N concentration in the control group. Inoculation improved the fermentation quality, and HT1 was more effective than SLI at 35°C and 45°C, while SLI showed better performance at 25°C. All silages displayed mild fluctuation for all treatments at the first 3 d of aerobic exposure, and significant differences were observed among treatments after that. Both control and inoculated silages stored at 25°C showed a sharp pH increase, while HT1 treated silages stored at 35°C and 45°C maintained stable pH and better fermentation quality during the aerobic exposure. In conclusion, SLI was suitable for oat silage fermentation at normal atmospheric temperature (25°C), while HT1 was more effective in improving DM recovery, fermentation quality and aerobic stability of oat silage at high temperature during summer in the Loess Plateau of China. Drought severely limits the growth and development of oat (Avena sativa) seedlings. As an osmotic regulator simulating a drought environment, Polyethylene glycol (PEG) has been widely linked in response to plant drought tolerance. However, the underlying mechanism of oats' response to PEG stress is still largely unknown. Here, we investigated the physiological and transcriptome variables of the drought-resistant oat variety DA92-2F6, and the drought-susceptible variety Longyan 3 under 15% PEG-6000 drought stress to better understand the underlying drought tolerance molecular mechanisms. The physiological results showed that except for the cell membrane permeability, the antioxidant enzyme, osmotic adjustment substance, and photosynthetic efficiency were significantly higher in the DA92-2F6 after 7 d stress. Further, 12 cDNA libraries and 123,223 unigenes were obtained by RNA-seq. A total of 33,857 differentially expressed genes (DEGs) were detected, of which two co-upregulated and three co-downregulated in four comparisons. We highlighted an analysis of the DEGs in phytohormone signal transduction pathway. The auxin, cytokinin, and brassinosteroid signaling pathways, were suppressed in Longyan 3, while abscisic acid and jasmonic acid signaling pathways were mainly activated in DA92-2F6 under drought stress. The upregulated of PP2C, ABF, SNRK2, GID1, JAZ, and MYC2 genes may enhance the drought tolerance of DA92-2F6. Many species of seed-borne fungi are closely allied with seed varieties and growing regions, including many seed-borne pathogens, but their species richness and distribution remain largely unknown. This study was conducted to explore the seed-borne fungal composition, abundance and diversity in Avena sativa (B7) and A. nuda (B2) seed samples collected from Baicheng (BB), Dingxi (DB) and Haibei (HB) city, using Illumina sequencing techniques. Our results show that a total of 543,707 sequences were obtained and these were assigned to 244 operational taxonomic units (OTUs) with 97% similarity. Oat varieties and growing locations had a significant difference on seed-borne fungal diversity. HB had a higher fungal diversity than BB and DB, Shannon diversity and ACE richness index of fungal in HB seeds was significantly higher than in BB and DB (P < 0.05). In different varieties, both taxon richness and evenness of B7 seeds was significantly higher than B2 (P < 0.05). A total of 4 fungal phyla and 26 fungal genera were detected. Ascomycota was the dominant phylum and Alternaria sp. was the most abundant genus in B2 and B7 oat seeds from different regions. Mycosphaerella sp. had a higher abundance in HB7 and DB7, respectively, Epicoccum sp. had a higher abundance in HB7 and BB7. The results of alpha and beta diversity analysis revealed the presence of different effects in fungal communities of different varieties and regions of oat, especially in seed pathogenic fungi distribution. Structural equation modeling also explained oat varieties and growing regions have significant influences on seed-borne fungal abundance, composition and diversity. In this study, a peptide–drug conjugate was designed and synthesized by connecting a transferrin receptor (TfR)‐targeted binding peptide analog BP9a (CAHLHNRS) with doxorubicin (DOX) through N‐succinimidyl‐3‐maleimidopropionate (SMP) as the cross‐linker. Confocal laser scanning microscopy results indicated that free DOX mainly accumulated in the nuclei of both TfR overexpressed HepG2 hepatoma cells and L‐O2 normal liver cells expressing low level of TfR; most of the BP9a‐DOX conjugate displayed cytoplasmic location, and its cellular uptake by HepG2 cells was obviously reduced by TfR blockage test. Nevertheless, the cellular uptake of this conjugate by L‐O2 cells was much less than that of free DOX. Meanwhile, the BP9a‐DOX conjugate exhibited lower in vitro antiproliferative activity against HepG2 cells than free DOX, but its cytotoxic effect on L‐O2 cells was decreased compared with that of free DOX. Erect milkvetch (Astragalus adsurgens Pall.) is a major legume forage plant widely grown in Northern China. However, the lack of molecular markers has limited its research into its genetic diversity and work on germplasm improvement. In this study, a total of 39,163 EST-SSR loci were identified from 30,262 unigene sequences in the erect milkvetch transcriptome using Illumina sequencing. Moreover, 22,367 EST-SSR primer pairs (PPs) were successfully designed. In addition, 100 PPs were synthesized and preliminarily screened in two accessions; of these, 90 were determined to be clear and stable EST-SSR markers. Fifty-one PPs were randomly selected in order to assess the genetic diversity of 27 erect milkvetch accessions. The average polymorphism information content of the 51 PPs was 0.682. Greater genetic diversity was detected in accessions from Inner Mongolia and in the group of landrace and wild erect milkvetch accessions. Nearly 90% of the 390 million ha of grasslands in northern China are degraded. 'Grazing exclusion' has been implemented as a nature‐based solution to rejuvenate degraded grasslands, but the effectiveness of the rejuvenation processes is uncertain. Here, we investigated the effects of grazing exclusion on aboveground plant community traits, soil physiochemical and biological properties, and the mechanisms responsible for enhanced grassland rejuvenation. A meta‐analysis across various studies was used to assess the effectiveness. On average, grazing exclusion improved vegetation coverage by 18.5 percentage points and increased aboveground biomass by 1.13 t ha−1 and root biomass by 1.27 t ha−1, which represent an increase of 84%, 246%, and 31%, respectively, compared with continuous grazing practices. Grazing exclusion reduced soil bulk density by 13.7% and increased soil water content by 68.9%. Grasslands under grazing exclusion increased soil organic carbon (SOC) in the 0‐ to 15‐cm depth by 3.95 (±0.35 Std err) t ha−1 and total soil N, available N, and total soil P in the 0‐ to 40‐cm depth by 2.39 (±0.14), 0.83 (±0.37), and 1.96 (±0.44) t ha−1, respectively, compared with continuous grazing; these values represent an increase of 31%, 25%, 23%, and 14%, respectively. Prolonging the duration (years) of grazing practices enlarged the differences in SOC and soil N content between grazing exclusion and continuous grazing. Grazing exclusion has improved plant community traits and enhanced soil physiochemical and biological properties of degraded grasslands, and thus, this 'nature‐based' approach can serve as an effective means to rejuvenate degraded grasslands. Oat is a main feed crop in high-altitude areas of western China, but few studies have been done on its silage making. The aim of this study was to evaluate the effect of silage additives on fermentation, aerobic stability, and nutritive value of different oat varieties (OV) grown in the Qinghai-Tibet Plateau of China. Two OV (Avena sativa L. cv. Longyan No.1 (OVL1) and Avena sativa L. cv. Longyan No.3 (OVL3)) were planted in a randomized complete block design, harvested at early dough stage with 32.6% and 34.1% DM, respectively. The fresh material was chopped to 2-cm length and treated with additives (0, Sila-Mix (MIX), Sila-Max (MAX) in a 2 × 3 factorial arrangement of treatments with three replicates. Both additives contained a mixture of lactic acid bacteria and supplied a final application rate of 2.5 × 108 of lactic acid bacteria per kg of fresh forage weight. After 60 d of ensiling, the number of lactic acid bacteria in treated silages was about 10-fold greater than the control and generally resulted in a lower pH and ammonia-nitrogen (P < 0.001), greater total acids and ratios of lactic acid/acetic acid (P < 0.001), and DM recovery (P = 0.028). Treatment with additives also decreased (P < 0.001) the number of yeasts, which resulted in marked (P < 0.001) improvements in aerobic stability with the effect being greatest with MAX. Both additives improved (P = 0.036) the 48-h in situ DM digestion in OVL1, but not in OVL3 (P = 0.052). Treatment with both additives also increased (P = 0.003) NDF digestion in OVL1 while it was improved (P < 0.001) only by MAX in OVL3. In contrast, the additives did not affect (P = 0.088) in situ hemicellulose digestion in OVL1, but it was improved (P = 0.048) by MIX and further improved (P = 0.002) by MAX in OVL3. Treatment with MAX improved yields of digestible DM and digestible NDF in both varieties. Dry matter recovery was not affected (P = 0.121) by variety. Compared to CTRL, silage treated with MAX had a greater (P = 0.015) DM recovery (96.7% vs. 93.9%). Inoculation improved (P < 0.001) aerobic stability. The MAX was the most effective for both varieties, while MIX was intermediate and was more effective in OVL3 than OVL1 silage. The results also showed that in Qinghai-Tibet Plateau, compared to OVL1, OVL3 resulted in greater (P = 0.002) yields of digestible nutrients; specifically, treated with MAX improved silage fermentation efficiency, DM recovery, and provided excellent aerobic stability for feeding to ruminants. A field grazing experiment was conducted in Jingtai County, Gansu Province, China, from 2003 to 2010. The grazing intensity treatments included light grazing (LG, 0.45 sheep unit/ha), moderate grazing (MG, 0.75 sheep unit/ha), heavy grazing (HG, 1.50 sheep unit/ha), and no grazing (NG). Soil physical, chemical, and biological properties were measured. The results showed that, with increasing grazing intensities, the concentrations of total nitrogen, available nitrogen, organic matter, available phosphorus, available potassium, urease, catalase, and alkaline phosphatase in the soil decreased significantly with LG being mostly the greatest (P < 0.05). Soil bulk density for HG and NG was greater than that for LG and MG (P < 0.05). The concentrations of copper, manganese, and zinc were lower than their critical levels for plant growth, whereas iron was greater than its critical level. To solve the problem of template update in biometric recognition system at regular time, this paper presented a template update method in biometric recognition system.Firstly, it described the self-update and co-update algorithms.Then, it made a theoretical analysis of self and co-update performance by exploiting the path-based clustering approach.Finally, it proposed the update method based on path-based clustering and unsupervised template.The experiments on the large DIEE multimodal data set show that the proposed method can update the template in biometric recognition system effectively.Moreover, the results show that the performance of co-update is superior to that of self-update by a simulative model. Abstract: Ethnopharmacological relevance: Jasminum officinale L. var. grandiflorum (JOG) is a folk medicine used for the treatment of hepatitis in south of China. Phytochemical studies showed that secoiridoid glycosides are the typical constituents of this plant. Aim of the study: The present study was undertaken to evaluate the effect of oleuropein (Ole) derived from the flowers of JOG on hepatitis B virus (HBV) replication in HepG2 2.2.15 cell line in vitro and duck hepatitis B virus (DHBV) replication in ducklings in vivo. Material and methods: The extracellular hepatitis B e antigen (HBeAg) and hepatitis B surface antigen (HBsAg) concentrations in cell culture medium were determined by ELISA. DHBV in duck serum was analyzed by dot blot. Results: Ole blocks effectively HBsAg secretion in HepG2 2.2.15 cells in a dose-dependent manner (IC50 =23.2μg/ml). Ole (80mg/kg, intraperitoneally, twice daily) also reduced viremia in DHBV-infected ducks. Conclusion: Ole therefore warrants further investigation as a potential therapeutic agent for HBV infection. The characteristics of selectivity for K+ over Na+ by the roots of the halophyte Puccinellia tenuiflora were investigated in comparison with the glycophyte wheat (Triticum aestivum). Under various NaCl concentrations, the concentrations of K+ in the shoots of P. tenuiflora were 16–24% lower than those of wheat, whereas the concentrations of K+ in Puccinellia roots were 2.8–4.0 times higher than those of wheat. In 200 mM NaCl, the concentrations of Na+ in shoots of P. tenuiflora and wheat were similar under high K+ levels, but the concentrations of Na+ in wheat were 1.6 times higher than those in Puccinellia under low K+ levels. The concentrations of K+ in roots of P. tenuiflora were 1.5–2.0 times higher than those of wheat under low K+ levels. Formulas are given for calculating net selective absorption (SA) capacity and selective transport (ST) capacity by roots for K+ over Na+. We interpret SA as the net capacity of selectively absorbing K+ over Na+ by epidermal and cortical cells of whole plant roots into the root symplast. ST could reflect the net capacity of selection for K+ over Na+ transport from whole root stelar symplast to the xylem vessels. The lower ST value of P. tenuiflora might be the reason for accumulation of K+ in its roots. The SA values of P. tenuiflora and wheat were approximately equivalent in the low-affinity K+ uptake range. The SA values of the former were about two times higher than that of the latter in the high-affinity K+ uptake range, showing the root high-affinity K+ uptake system of the halophyte P. tenuiflora has a stronger capacity for K+ uptake.
