Neural stem progenitor cell (NSPC) transplantation has been regarded as a promising therapeutic method for spinal cord injury (SCI) repair. However, different NSPCs may have different therapeutic effects, and it is therefore important to identify the optimal NSPC type. In our study, we compared the transcriptomes of human fetal brain-derived NSPCs (BNSPCs), spinal cord-derived NSPCs (SCNSPCs) and H9 embryonic stem-cell derived NSPCs (H9–NSPCs) in vitro and subsequently we transplanted each NSPC type on a collagen scaffold into a T8-9 complete SCI rat model in vivo. In vitro data showed that SCNSPCs had more highly expressed genes involved in nerve-related functions than the other two cell types. In vivo, compared with BNSPCs and H9–NSPCs, SCNSPCs exhibited the best therapeutic effects; in fact, SCNSPCs facilitated electrophysiological and hindlimb functional recovery. This study demonstrates that SCNSPCs may be an appropriate candidate cell type for SCI repair, which is of great clinical significance.

Abstract Background Recently, neural stem cell (NSC) therapy has shown promise for the treatment of many neurological diseases. Enhancing the quality of implanted cells and improving therapeutic efficacy are currently research hotspots. It has been reported that collagen sponge material provided sufficient room for cell growth in all directions and promoted the absorption of nutrients and removal of wastes. And also, the Rotary Cell Culture System (RCCS), which mimics the microgravity environment, can be used to culture cells for tissue engineering. Materials and methods We performed the mRNA and miRNA sequencing to elucidate the regulatory mechanism of NSCs cultured on the collagen sponge in the RCCS system. The luciferase assay and Western blot revealed a direct regulatory role between let-7i-5p and neurotrophic receptor tyrosine kinase 3 (NTRK3; also called TrkC). And then, the neural differentiation markers Tuj1 and Map2 were detected by immunofluorescence staining. In the meantime, the migratory ability of NSCs was detected both in vitro and in spinal cord injury animals. Results In this study, we demonstrated that the expression of NTRK3 was elevated in NSCs cultured on collagen sponge in the RCCS system. Furthermore, increased NTRK3 expression was regulated by the downregulation of let-7i-5p. Compared to traditionally cultured NSCs, the NSCs cultured on collagen sponge in the RCCS system exhibited better neuronal differentiation and migratory ability, especially in the presence of NT-3. Conclusions As the biological properties and quality of transplanted cells are critical for therapeutic success, the RCCS system combined with the collagen sponge culture system shows promise for applications in clinical practice in the future.

This paper proposes a reverse procedure detection method for space target streaks based on motion parameter estimation. According to the phase shift characteristics of the spectrum, the interframe phase difference is used to obtain the displacement vectors of the target streak and image background, and then the morphological parameters of the target streak are estimated. In addition, a streak window is designed on the basis of the parameters, utilizing the local grayscale correlation in two image frames to carry out target searching and positioning with the interval of the target displacement vector. Images from different scenes are used to evaluate the adaptability of our method, and the experimental results suggest that the expected targets can be detected successfully with subpixel precision in all telescope operating modes. The proposed frequency-domain reverse procedure detection method avoids the problem of fake target confirmation caused by blind detection in traditional methods and overcomes the interference produced when the target streak passes through a bright star. Moreover, first target tracking and then positioning, which cannot be performed with time-domain methods, can be achieved. Our proposed method is attractive for multiple applications, such as space target cataloging, early warning and orbit prediction.

Energy evolution process of rock deformation is conducive to essentially reveal the rock failure mechanism and is of great significance to uncover the breeding of dynamic disasters in rock engineering. To characterize the damage evolution of energy dissipation during rock failure, the digital image correlation (DIC) technique is proposed to describe the rock failure mechanics and its energy evolution process. The uniaxial compression experiment of sandstone specimen was carried out, and the whole field deformation and failure characteristics of the rock had been captured by the DIC system. Measurement accuracy was verified by the fiber Bragg grating (FBG) sensor, the elastic region of the specimen was divided according to the location of strain localization band (SLB), and the evolution process of elastic strain energy of the rock was analyzed. The results show that the time history development of rock strain obtained by the FBG and DIC system matches identically, and the deviation of peak axial strain of both means is less than 5%, which verifies the applicability of DIC system. The uncoordinated evolution of rock deformation displacement field is discussed to reveal the crack development and failure form of the sandstone specimen under uniaxial compression. The energy evolution of the elastic region of the specimen is revealed, and the development of releasable elastic strain energy would be divided into three stages, which correspond to the stress–strain characteristics of rock failure mechanics. This study could provide an alternative analytical method for the experimental rock mechanics research studies.

In this study, a multi-physics coal and gas outburst experimental apparatus is developed to allow investigating the gas-driven mechanism behind the disaster caused by coal and gas outbursts in tectonic regions. The apparatus can simulate the coal and gas outbursts under different initial geo-stresses, gas pressures and temperatures. By integrating acoustic emission sensors in the holes on axial platens, acoustic signals from coal samples during the whole test can be monitored. A series of testability experiments were conducted on the developed apparatus to verify its performance. The damage characteristics and outburst occurrence of raw coal under different gas pressures were experimentally studied and that provided a more complete theoretical basis for coal and gas outburst. The degree of fragmentation of coal samples and the mass proportion distributions of the outburst pulverized coal after outbursts under different gas pressures were analyzed, as a reference for exploring the evolutionary characteristics of coal and gas outburst with gas-driven action.

Abstract Background Emerging evidence suggests that miR-124 performs important biological functions in neural stem cells (NSCs); it regulates NSC behavior and promotes the differentiation of NSCs into neurons, but the exact molecular mechanism remains unknown. And also, the role of miR-124 during spinal cord injury regeneration is unclear. Materials and methods In order to explore the function of miR-124 in neural differentiation, the molecular markers (Tuj1, Map2, and GFAP) correlated with the differentiation of NSCs were detected by immunofluorescence staining both in cultured mouse spinal cord progenitor cells (SC-NPCs) and in spinal cord injury (SCI) animal models. The migration ability and apoptosis of cultured SC-NPCs were also evaluated by Transwell migration assay and TUNEL assay. In addition, the relative expression of lnRNA Neat1- and Wnt/β-catenin signaling-related genes were detected by quantitative real-time PCR. Results In this study, we revealed that lncRNA Neat1 is involved in regulating Wnt/β-catenin signaling that is activated by miR-124 in SC-NPCs. LncRNA Neat1 was also found to play an important role in regulating neuronal differentiation, apoptosis, and migration of SC-NPCs. Furthermore, we demonstrated that overexpression of miR-124 resulted in elevated Neat1 expression, accompanied with the functional recovery of locomotion in a mouse model of spinal cord injury. Conclusions Our results confirm the therapeutic effectiveness of miR-124 on the functional recovery of injured spinal cord, supporting the rationale and feasibility of miR-124 for spinal cord injury treatment in future clinical therapy. Furthermore, we concluded that the miR-124-Neat1-Wnt/β-catenin signaling axis is involved in regulating the cell function of SC-NPCs, and this may offer novel therapeutic avenues for future treatment of SCI.

Coal seam permeability is one of the key factors influencing the gas extraction efficiency, which is of great significance to reduce coal and gas dynamic disasters in gassy coal mines. Hydraulic slotting technique is an effective method to stimulate the coal reservoir, but the selection of slotting key parameters has great impact on gas extraction efficiency. For this reason, the hydraulic slotting model was established by using FLAC3D software to analyze the stress distribution before and after slotting. Then, the influence of borehole diameter, slotting width, and slotting length on coal seam stress relief is also discussed. The results show that the slotting width has a great influence on the stress relief of the coal seam, while the borehole diameter and slotting length have no obvious influence on that. Based on the results of numerical simulation, field tests were carried out in Sangshuping NO.2 coal mine. The results show that the coal seam stress can be fully released, resulting in the improvement of coal seam permeability. The gas extraction efficiency can be highly enhanced by hydraulic slotting. This research achievement provides the guidance basis for high-stress water jet slotting technology with adaptive selection of slotting parameters in different geological conditions.

Many insects spin cocoons to protect the pupae from unfavorable environments and predators. After emerging from the pupa, the moths must escape from the sealed cocoons. Previous works identified cocoonase as the active enzyme loosening the cocoon to form an escape-hatch. Here, using bioinformatics tools, we show that cocoonase is specific to Lepidoptera and that it probably existed before the occurrence of lepidopteran insects spinning cocoons. Despite differences in cocooning behavior, we further show that cocoonase evolved by purification selection in Lepidoptera and that the selection is more intense in lepidopteran insects spinning sealed cocoons. Experimentally, we applied gene editing techniques to the silkworm Bombyx mori, which spins a dense and sealed cocoon, as a model of lepidopteran insects spinning sealed cocoons. We knocked out cocoonase using the CRISPR/Cas9 system. The adults of homozygous knock-out mutants were completely formed and viable but stayed trapped and died naturally in the cocoon. This is the first experimental and phenotypic evidence that cocoonase is the determining factor for breaking the cocoon. This work led to a novel silkworm strain yielding permanently intact cocoons and provides a new strategy for controlling the pests that form cocoons.

Abstract In this study, we upregulated insulin-like growth factor-1 (IGF1) expression specifically in skeletal muscle by engineering an enhancer into its non-coding regions and verified the expected phenotype in a mouse model. To select an appropriate site for introducing a skeletal muscle-specific myosin light chain (MLC) enhancer, three candidate sites that exhibited the least evolutionary conservation were chosen and validated in C2C12 single-cell colonies harbouring the MLC enhancer at each site. IGF1 was dramatically upregulated in only the site 2 single-cell colony series, and it exhibited functional activity leading to the formation of extra myotubes. Therefore, we chose site 2 to generate a genetically modified (GM) mouse model with the MLC enhancer incorporated by CRISPR/Cas9 technology. The GM mice exhibited dramatically elevated IGF1 levels, which stimulated downstream pathways in skeletal muscle. Female GM mice exhibited more conspicuous muscle hypertrophy than male GM mice. The GM mice possessed similar circulating IGF1 levels and tibia length as their WT littermates; they also did not exhibit heart abnormalities. Our findings demonstrate that genetically modifying a non-coding region is a feasible method to upregulate gene expression and obtain animals with desirable traits.

Space object recognition in high Earth orbits (between 2000 km and 36,000 km) is affected by moonlight and clouds, resulting in some bright or saturated image areas and uneven image backgrounds. It is difficult to separate dim objects from complex backgrounds with gray thresholding methods alone. In this paper, we present a segmentation method of star images with complex backgrounds based on correlation between space objects and one-dimensional (1D) Gaussian morphology, and the focus is shifted from gray thresholding to correlation thresholding. We build 1D Gaussian functions with five consecutive column data of an image as a group based on minimum mean square error rules, and the correlation coefficients between the column data and functions are used to extract objects and stars. Then, lateral correlation is repeated around the identified objects and stars to ensure their complete outlines, and false alarms are removed by setting two values, the standard deviation and the ratio of mean square error and variance. We analyze the selection process of each thresholding, and experimental results demonstrate that our proposed correlation segmentation method has obvious advantages in complex backgrounds, which is attractive for object detection and tracking on a cloudy and bright moonlit night.

Impedance spectroscopy has been widely used to extract the electron-hole recombination rate constant in organic photovoltaic cells (OPVs). This technique is typically performed on OPVs held at open-circuit. Under these conditions, the analysis is simplified with recombination as the only pathway for the decay of excess charge carriers; transit provides no net change in the charge density. In this work, we generalize the application and interpretation of impedance spectroscopy for bulk heter-ojunction OPVs at any operating voltage. This, in conjunction with reverse bias external quantum efficiency measurements, permits the extraction of both recombination and transit rate constants. Using this approach, the transit and recombination rate constants are determined for OPVs with a variety of electron donor-acceptor pairings and compositions. It is found that neither rate constant individually is sufficient to characterize the efficiency of charge collection in an OPV. It is demonstrated that a large recombination rate constant can be accompanied by a large transit rate constant, thus fast recombination is not necessarily detrimental to OPV performance. Extracting the transit and recombination rate constants permits a detailed understanding of how OPV architecture and processing conditions impact the transient behavior of charge carriers, elucidating the origin of optimum device configurations. Metal-oxides have been used as interlayers at the anode-organic interface in organic photovoltaic cells (OPVs) to increase the open-circuit voltage (VOC). We examine the role of MoOx in determining the maximum VOC in a planar heterojunction OPV and find that the interlayer strongly affects the temperature dependence of VOC. Boron subphthalocyanine chloride (SubPc)-C60 OPVs that contain no interlayer show a maximum VOC of 1.2V at low temperature, while those with MoOx show no saturation, reaching VOC>1.4 V. We propose that the MoOx-SubPc interface forms a Schottky junction that provides an additional contribution to VOC at low temperature. [ABSTRACT FROM AUTHOR]
