Abstract Background In clinical practice, fluctuating vision or decreased quality of vision is a common complaint in DED patients. Our study was designed to investigate the change in dynamic optical quality in dry eye patients after the use of artificial tears. Methods Fifty-nine patients with dry eye disease (DED) and 31 control subjects were included in this prospective case-control study. There was no significant difference in age and sex between these two groups (P = 0.342, P = 0.847, respectively). Clinical evaluation of the ocular surface included Ocular Surface Disease Index (OSDI), tear film break-up time (TBUT), lipid layer thickness (LLT), and Schirmer I test. DED patients were divided into two groups, mild (31 patients) and severe (28 patients). The optical quality of the tear film was measured with the Optical Quality Analysis System (OQAS) using the mean objective scatter index (mean OSI), standard deviation of objective scatter index (SD-OSI) and modulation transfer function cut-off (MTF cut-off). After baseline examinations, one drop of artificial tears (ATs, carboxymethylcellulose ophthalmic solution, 0.5%) was instilled in both eyes, and optical quality parameters were measured again at 5 and 30 min following application of ATs. Background Deep learning has been widely used for glaucoma diagnosis. However, there is no clinically validated algorithm for glaucoma incidence and progression prediction. This study aims to develop a clinically feasible deep-learning system for predicting and stratifying the risk of glaucoma onset and progression based on color fundus photographs (CFPs), with clinical validation of performance in external population cohorts.Methods We established data sets of CFPs and visual fields collected from longitudinal cohorts. The mean follow-up duration was 3 to 5 years across the data sets. Artificial intelligence (AI) models were developed to predict future glaucoma incidence and progression based on the CFPs of 17,497 eyes in 9346 patients. The area under the receiver operating characteristic (AUROC) curve, sensitivity, and specificity of the AI models were calculated with reference to the labels provided by experienced ophthalmologists. Incidence and progression of glaucoma were determined based on longitudinal CFP images or visual fields, respectively.Results The AI model to predict glaucoma incidence achieved an AUROC of 0.90 (0.81–0.99) in the validation set and demonstrated good generalizability, with AUROCs of 0.89 (0.83–0.95) and 0.88 (0.79–0.97) in external test sets 1 and 2, respectively. The AI model to predict glaucoma progression achieved an AUROC of 0.91 (0.88–0.94) in the validation set, and also demonstrated outstanding predictive performance with AUROCs of 0.87 (0.81–0.92) and 0.88 (0.83–0.94) in external test sets 1 and 2, respectively.Conclusion Our study demonstrates the feasibility of deep-learning algorithms in the early detection and prediction of glaucoma progression.FUNDING National Natural Science Foundation of China (NSFC); the High-level Hospital Construction Project, Zhongshan Ophthalmic Center, Sun Yat-sen University; the Science and Technology Program of Guangzhou, China (2021), the Science and Technology Development Fund (FDCT) of Macau, and FDCT-NSFC.

In order to study the energy evolution and infrared radiation response characteristics during the deformation and failure process of coal with different bedding angles, uniaxial compression tests with four different bedding angles (0°, 30°, 60° and 90°) were carried out on a strong burst liability coal samples from 3-1 coal seam of Hongqinghe coal mine. The infrared temperature field characteristics of the coal samples during deformation and damage were monitored using an infrared thermal imaging camera. The test results show that as the bedding angle increases the compressive strength and strain energy of the sample show a “V” shaped trend, reaching the minimum value at 60°. The bedding has a significant impact on the elastic strain Ue in the pre-peak stage and dissipation energy Ud in the post-peak stage; The coal samples showed an overall warming trend with different bedding angles, with an average infrared temperature precursor point of 0.84σp.The bedding structure seriously affects the infrared radiation temperature and the difference of failure patterns lead to different temperature ranges. The 0° and 30° specimens are dominated by shear-tension composite failure, with a high temperature rise of 1.12 and 1.30 ℃, respectively, while the 60° specimen is dominated by single shear failure, with a low temperature rise of 0.46 ℃. The failure of 90° specimen is tensile failure, and the temperature rise is the lowest 0.4 ℃. Abstract Background The loach (Misgurnus anguillicaudatus) is a significant freshwater economic fish in China, renowned for its tender meat, delicious taste, and high nutritional value. It is widely distributed across the country, except for the western plateau. However, the loach is currently at risk of population decline and degradation of wildlife resources. Research on genetic diversity provides a crucial foundation for the conservation and development of these fish resources. To enhance the protection and utilization of wild loach germplasm resources, we analyzed the genetic structure and diversity of 60 wild loach populations from Xiangtan City (XT), Shaoyang City (SY), and Yueyang City (XY) in Hunan, Guilin City (GL) and Guiping City (GP) in Guangxi, and Wuhan City (WH) in Hubei. Additionally, we mapped the DNA fingerprints of these 60 wild loaches using 12 high-quality SNP sites. Results Whole genome resequencing (WGRS) was conducted on 60 loaches from six regions, yielding a total of 1047.17 Gb of raw data and 1046.98 Gb of clean data. From this 2,812,906 high-quality single nucleotide polymorphisms (SNPs) were identified, of which 10,022 core SNPs were selected to analyze the population genetic structure, and 12 core SNPs were used to assess the genetic diversity and DNA fingerprint. The analysis revealed that the 60 loach samples could be grouped into three clusters: Cluster A comprised the XT, SY, and XY groups; Cluster B included the WH group; and Cluster C consisted of the GL and GP groups. The mean nucleic acid diversity (Pi), heterozygosity (Ho), and expected heterozygosity (He) of SNP markers were 0.130, 0.140, and 0.123, respectively. The average inbreeding coefficient was 0.552, indicating high levels of inbreeding. Conclusions Whole genome resequencing is an effective high-throughput approach for identifying SNP information in loach germplasm and describing genetic relationships within this genus. DNA fingerprinting based on SNP marker technology can accurately assess the genetic structure and variation within natural loach populations, making it a valuable tool for strain and variation identification. Our findings provide a scientific basis for the conservation, development, and optimal breeding of native loach germplasm resources and contribute to expanding the genetic diversity database of wild loach populations.

Abstract The nanoscale fibrillar morphology, featuring long-range structural order, provides abundant interfaces for efficient exciton dissociation and high-quality pathways for effective charge transport, is a promising morphology for high performance organic solar cells. Here, we synthesize a thiophene terminated non-fullerene acceptor, L8-ThCl, to induce the fibrillization of both polymer donor and host acceptor, that surpasses the 20% efficiency milestone of organic solar cells. After adding L8-ThCl, the original weak and less continuous nanofibrils of polymer donors, i.e. PM6 or D18, are well enlarged and refined, whilst the host acceptor L8-BO also assembles into nanofibrils with enhanced structural order. By adapting the layer-by-layer deposition method, the enhanced structural order can be retained to significantly boost the power conversion efficiency, with specific values of 19.4% and 20.1% for the PM6:L8-ThCl/L8-BO:L8-ThCl and D18:L8-ThCl/L8-BO:L8-ThCl devices, with the latter being certified 20.0%, which is the highest certified efficiency reported so far for single-junction organic solar cells.

Heat stress is a major environmental stressor that affects fish metabolism, growth, and death rates. This research examined the impact of dietary rutin addition (0, 100, 300, or 500 mg/kg) for 60 days on juvenile hybrid fish (Carassius auratus cuvieri, WCC, ♀ × Carassius auratus red var, RCC, ♂, WR) (27.97 ± 0.56 g) under heat stress conditions (32 ± 1 °C for 48 h). The analysis focused on blood parameters, oxidative stress biomarkers, and hepatic hsp70 and hsp90 gene expression in WR. The results demonstrated that rutin supplementation elevated blood glucose levels and the liver activities of glutathione reductase, glutathione peroxidase, catalase, and total superoxide dismutase in comparison to the control group. Additionally, rutin supplementation also significantly reduced serum cortisol and hepatic malondialdehyde levels while upregulating hepatic hsp70 and hsp90 gene expression. These findings suggest that rutin supplementation enhances antioxidant responses and alleviates the impact of heat stress on fish physiological and biochemical markers and heat shock protein gene expression.

Abstract Conjugated polymers are generally featured with low structural order due to their aromatic and irregular structural units, which limits their light absorption and charge mobility in organic solar cells. In this work, we report a conjugated molecule INMB-F that can act as a molecular bridge via electrostatic force to enhance the intermolecular stacking of BDT-based polymer donors toward efficient and stable organic solar cells. Molecular dynamics simulations and synchrotron X-ray measurements reveal that the electronegative INMB-F adsorb on the electropositive main chain of polymer donors to increase the donor-donor interactions, leading to enhanced structural order with shortened π-π stacking distance and consequently enhanced charge transport ability. Casting the non-fullerene acceptor layer on top of the INMB-F modified donor layer to fabricate solar cells via layer-by-layer deposition evidences significant power conversion efficiency boosts in a range of photovoltaic systems. A power conversion efficiency of 19.4% (certified 18.96%) is realized in PM6/L8-BO binary devices, which is one of the highest reported efficiencies of this material system. The enhanced structural order of polymer donors by INMB-F also leads to a six-fold enhancement of the operational stability of PM6/L8-BO organic solar cells.

Abstract Background Distant hybridization is an important breeding technique for creating new strains with superior traits by integrating two different genomes. Successful hybridization of Megalobrama amblycephala (Blunt snout bream, BSB, 2n = 48) and Culter alburnus (Topmouth culter, TC, 2n = 48) was achieved to establish hybrid lineages (BT and TB), which provide valuable materials for exploring the mechanisms of distant hybridization fertility. In this study, the gonadal tissue transcriptomes of BSB, TC, BT-F1, and TB-F1 were sequenced using Illumina high-throughput sequencing technology to analyze the reproductive characteristics of BT and TB. Results Differential gene expression analysis showed that the differentially expressed genes in BT vs BSB and BT vs TC were mainly enriched in signaling pathways not directly associated with meiosis. While, the differentially expressed genes of TB vs BSB and TB vs TC were mainly enriched in pathways related to meiosis, and most of them were down-regulated, indicating that meiosis is suppressed in TB. Under-dominance (UD) genes were enriched in pathways related to meiosis and DNA repair in TB. Over-dominance (OD) genes were enriched in MAPK signaling pathway, expression level dominance-BSB (ELD-B) genes were enriched in pathways related to steroid hormone synthesis and expression level dominance-TC (ELD-T) genes were not significantly enriched in any pathway in both BT and TB. Conclusions These results suggest that meiotic progression may not be affected in BT, whereas it is clearly inhibited in TB. Offspring of M. amblycephala maternal parent may have better genomic compatibility and fertility. Our study provides important information on the molecular mechanisms of breaking reproductive isolation in distantly hybridized fertile lineages.

Nonylphenol (NP) is a known endocrine-disrupting chemical (EDC) that has been shown to affect bone development in mammals. However, the detrimental impacts of NP on the skeletal growth and development of aquatic species, especially bony fish, remain poorly understood. Bone morphogenic proteins (BMPs), essential for bone formation and osteoblast differentiation, act through the BMP-Smad signaling pathway. In this study, two BMP genes, BMP2 and BMP4, were cloned and characterized in the red crucian carp (Carassius auratus red var.). The full-length cDNAs of BMP2 and BMP4 were 2029 bp and 2095 bp, respectively, encoding polypeptides of 411 and 433 amino acids, and share a typical TGF-β domain with other BMPs. The tissue expression patterns of both genes were identified, showing ubiquitous expression across all studied tissues. Additionally, the exposure of embryos or adult fish to NP stress resulted in a downregulation of BMP2, BMP4, and other genes associated with the BMP-Smad signaling pathway. Moreover, the combined treatment of adult fish with NP and the specific BMP receptor inhibitor significantly reduced these genes’ expression. These findings elucidate the mechanism of NP stress on BMP2 and BMP4, suggesting a role for the BMP-Smad signaling pathway in the response to endocrine-disrupting chemicals in fish.

Figla is one of the earliest expressed genes in the oocyte during ovarian development. In this study, Figla was characterized in C. maculata, one of the main aquaculture species in China, and designated as CmFigla. The length of CmFigla cDNA was 1303 bp, encoding 197 amino acids that contained a conserved bHLH domain. CmFigla revealed a female-biased expression patterns in the gonads of adult fish, and CmFigla expression was far higher in ovaries than that in testes at all gonadal development stages, especially at 60~180 days post-fertilization (dpf). Furthermore, a noteworthy inverse relationship was observed between CmFigla expression and the methylation of its promoter in the adult gonads. Gonads at 90 dpf were used for in situ hybridization (ISH), and CmFigla transcripts were mainly concentrated in oogonia and the primary oocytes in ovaries, but undetectable in the testes. These results indicated that Figla would play vital roles in the ovarian development in C. maculata. Additionally, the frame-shift mutations of CmFigla were successfully constructed through the CRISPR/Cas9 system, which established a positive foundation for further investigation on the role of Figla in the ovarian development of C. maculata. Our study provides valuable clues for exploring the regulatory mechanism of Figla in the fish ovarian development and maintenance, which would be useful for the sex control and reproduction of fish in aquaculture.

Abstract Background The influence of environmental factors, especially temperature, on sex ratio is of great significance to elucidate the mechanism of sex determination. However, the molecular mechanisms by which temperature affects sex determination remains unclear, although a few candidate genes have been found to play a role in the process. In this study, we conducted transcriptome analysis of the effects induced by high temperature on zebrafish during gonad differentiation period. Results Totals of 1171, 1022 and 2921 differentially expressed genes (DEGs) between high temperature and normal temperature were identified at 35, 45 and 60 days post-fertilization (dpf) respectively, revealing that heat shock proteins (HSPs) and DNA methyltransferases (DNMTs) were involved in the heat-exposed sex reversal. The Gene Ontology (GO) terms and the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway that were enriched in individuals after heat treatment included Fanconi anemia (FA) pathway, cell cycle, oocyte meiosis and homologous recombination. Conclusions Our study provides the results of comparative transcriptome analyses between high temperature and normal temperature, and reveals that the molecular mechanism of heat-induced masculinization in zebrafish is strongly related to the expression of HSPs and DNMTs and FA pathway during gonad differentiation.

Abstract Crop diseases have a devastating effect on agricultural production, and serious diseases can lead to harvest failure entirely. Recent developments in deep learning have greatly improved the accuracy of image identification. In this study, we investigated the transfer learning of deep convolutional neural networks and modified the network structure to improve the learning capability of plant lesion characteristics. The MobileNet with squeeze‐and‐excitation (SE) block was selected in our approach. Integrating the merits of both, the pre‐trained MobileNet and SE block were fused to form a new network, which we termed the SE‐MobileNet, and was used to identify the plant diseases. In particular, the transfer learning was performed twice to obtain the optimum model. The first phase trained the model for the extended layers while the bottom convolution layers were frozen with the pre‐trained weights on ImageNet; by loading the model trained in the first phase, the second phase retrained the model using the target dataset. The proposed procedure provides a significant increase in efficiency relative to other state‐of‐the‐art methods. It reaches an average accuracy of 99.78% in the public dataset with clear backdrops. Even under multiple classes and heterogeneous background conditions, the average accuracy realises 99.33% for identifying the rice disease types. The experimental findings show the feasibility and effectiveness of the proposed procedure.

Leptin receptor (LEPR) is a type of transmembrane receptor and a member of the class I cytokine receptor family that plays a decisive role in the physiological functions mediated by leptin. In this study, a fish LEPR gene (CiLEPR) was cloned from Ctenopharyngodon idella. The open reading frame (ORF) sequence of the CiLEPR gene included 3504 bp, which encoded 1078 amino acids that formed six functional domains, including five FN3 domains and a lep_receptor_Ig domain. Phylogenetic analysis showed that the CiLEPR amino acid sequence was closely related to that of LEPR from Hypophthalmichthys molitrix. The qRT-PCR results showed that the CiLEPR gene was widely expressed in all investigated tissues of C. idella and was relatively highly expressed in the gills. The results of the circadian rhythm experiment revealed that the intestinal expression of the CiLEPR gene changed regularly over time. In C. idella intestinal cell experiments, it was observed that recombinant leptin could upregulate the expression levels of CiLeptin, CiLEPR and CiPepT1 in the intestines of C. idella. The results of experiments in which fish were fed with various protein sources demonstrated that the mRNA expression levels of CiLeptin, CiLEPR and CiPepT1 in the fish meal group were higher than those in the soybean meal group at 28 d. The data indicated that C. idella fed diets with medium amounts of dietary protein (37%) expressed the highest mRNA levels of CiLeptin, CiLEPR and CiPepT1 after 4 weeks. These results provide a basis for further studies on the nutritional regulation and physiological functions of leptin receptors in fish.

To achieve high-accuracy tracking of a dual-linear-motor-driven (DLMD) gantry, high-level synchronization between redundant actuators becomes a non-negligible factor and also a difficult issue to be solved priorly. Especially, when both X and Y axes are simultaneously operating to accomplish complex tasks efficiently, additional coupling effects will be generated by the dynamic load presented on the crossbeam, which makes the synchronization issue more complicated than the case with static load. However, due to the absence of an accurate model to fully reveal the complete coupling characteristics, existing approaches to this issue still have inherent limitations. Therefore, this paper focuses on the systematic physical modeling and synchronization control of a DLMD gantry with a dynamic load presented on the crossbeam. A complete coupling mathematical model is established first, by fully considering two linear motions (X-axis and Y-axis) and also including the additional rotational motion of the crossbeam. Built upon the effective model information, corresponding solutions by compensating the dynamic load effects and actively controlling the rotational dynamics to regulate the internal forces have been proposed, leading to a novel adaptive robust synchronization control method. The results of comparative experiments verify the effectiveness and superiority of the proposed method in dealing with the synchronization issue subjected to dynamic load effects.

Enhancers are cis-acting elements that can promote the expression of target genes and respond to estrogen to induce the transcription of eRNAs, which are closely associated with cancer development. Further study on eRNAs may lead to a better understanding of the significance of transcriptional regulation and the progression of malignant tumors. SMAD7 enhancer RNA (SMAD7e) is an estrogen-responsive eRNA. However, the relationship between SMAD7e and bladder cancer remains unclear. SMAD7e was significantly upregulated in bladder cancer tissues and estrogen-stimulated cells. Knockdown of SMAD7e by CRISPR-Cas13a suppressed cell proliferation and migration, and induced cell apoptosis and inhibited cell invasion. Estrogen caused overexpression of SMAD7e and played a facilitating role in bladder cancer cells. Furthermore, knockdown of SMAD7e by CRISPR-Cas13a prevented the cancer-promoting effects of estrogen on bladder cancer both in vitro and in vivo. The present study suggested the crucial role of SMAD7e in bladder cancer. Estrogen might promote the development of bladder cancer by inducing SMAD7e production. These findings may provide a potential target for CRISPR-mediated gene therapy for bladder cancer in the future.

Background: The purpose of this study was to review recent research related to the analgesic effect of ablation therapy combined with cementoplasty, as well as to identify the duration of analgesic effect and risk for cement leaks. Methods: A systematic literature search using PubMed, Web of Science, and annual meeting proceedings of the oncology society and other organizations were conducted. Results: Twelve retrospective studies met the inclusion criteria. Four of the studies included in the review assessed the changes immediately after treatment. Five studies were subjected to analyses of analgesic effect of combined percutaneous thermal ablation and Cementoplasty at 24 weeks after treatment. Incidences of leakage of bone cement during surgery were detected in 4 out of 12 studies. The change of mean pain scores at 1 days, at 1 week, and at 4 weeks, 12 weeks, and 24 weeks after treatment were −3.90 (95% CI: −4.80 to −3.00), −4.55 (95% CI:−5.46 to −3.64), −4.78 (95% CI: −5.70 to −3.86), −5.16 (95% CI: −6.39 to −3.92), and −5.91 (95% CI: −6.63 to −5.19). The relative risk of cement leakage was 0.10 (95% CI: −6.63 to −5.19). Conclusions: Our systematic review suggested that thermal ablation combined with cementoplasty could be a safe and effective intervention for the management of bone metastases-induced pain. Pasting properties of barley starch are important characteristics from a processing standpoint. The isolation of starch form barley grains is time consuming thus the whole grain flour is always used. To compare pasting properties of starch with those of the whole grain flour, we used a Rapid Visco Analyser (RVA) to measure pasting properties of three types of samples: grain flour and starches isolated using two different extraction methods. We also investigated compositional, morphological and structural properties of the two starch samples. Significant differences in pasting properties were found among the three sample types, but most of the parameters of pasting properties displayed significant correlations between flour and starch. No significant differences were found in amylose/amylopectin ratio, granule morphology, granule size distribution and crystal structure between starches extracted using two different methods. However, the starch isolated from water homogenization had a higher protein content and lower total starch, amylose and amylopectin contents than the starch extracted with homogenized extraction under alkaline conditions. We concluded that the whole grain flour can be used to predict the pasting properties in breeding programs.

Amylopectin concentration in barley endosperm has important effects on grain quality and end-use. In this study, quantitative trait locus (QTL) analysis together with genome-wide association studies (GWAS) were performed to identify markers linked to grain amylopectin content respectively using a doubled haploid (DH) population of 178 lines and a collection of 185 diverse barley germplasms both genotyped by genotyping-by-sequencing (GBS). A stable QTL on chromosome 7H and 11 associated single nucleotide polymorphisms (SNPs) were detected. In the co-localized region, the SSIIa (SSII-3) gene was predicted as the candidate gene. Then we isolated and characterized biparental SSIIa alleles of the DH population, investigated the expression pattern by quantitative real-time PCR (qRT-PCR), and revealed that a 33-bp deletion in exon 2 is responsible for reducing SSIIa transcript, thus resulting in a reduced amylopectin content. A sequence-based molecular marker was developed for the SSIIa allele and validated the effectivity, which would provide help for barley breeding.

With the substantial increase of vehicles on road, driving safety and transportation efficiency have become increasingly concerned focus from drivers, passengers, and governments. Wireless networks constructed by vehicles and infrastructures provide abundant information to share for the sake of both enhanced safety and network efficiency. This paper presents the systematic research to enhance the vehicle safety by wireless communication, in the aspects of information acquisition through vehicle sensing, vehicle-to-vehicle (V2V) routing protocol for the highly dynamic vehicle network, vehicle-to-infrastructure (V2I) routing protocol for a tradeoff in real-time performance and load balance, and hardware implementation of V2V system with on-road test. Simulations and experimental result validate the feasibility of the algorithms and communication system.
