Self‐propelling active particles are an exciting and interdisciplinary emerging area of research with projected biomedical and environmental applications. Due to their autonomous motion, control over these active particles that are free to travel along individual trajectories, is challenging. This work uses optically patterned electrodes on a photoconductive substrate using a digital micromirror device (DMD) to dynamically control the region of movement of self‐propelling particles (i.e., metallo‐dielectric Janus particles (JPs)). This extends previous studies where only a passive micromotor is optoelectronically manipulated with a translocating optical pattern that illuminates the particle. In contrast, the current system uses the optically patterned electrode merely to define the region within which the JPs moved autonomously. Interestingly, the JPs avoid crossing the optical region's edge, which enables constraint of the area of motion and to dynamically shape the JP trajectory. Using the DMD system to simultaneously manipulate several JPs enables to self‐assemble the JPs into stable active structures (JPs ring) with precise control over the number of participating JPs and passive particles. Since the optoelectronic system is amenable to closed‐loop operation using real‐time image analysis, it enables exploitation of these active particles as active microrobots that can be operated in a programmable and parallelized manner. Bioinspired artificial ionic components are extensively utilized to mimic biological systems, as the vast majority of biological signaling is mediated by ions and molecules. Particular attention is given to nanoscale fluidic components where the ion transport can be regulated by the induced ion permselectivity. As a step from fundamentals toward ion-controlled devices, this study presents the use of ionic diodes made of oppositely charged polyelectrolytes, as a gate for low-abundance molecules. The use of ionic diodes that exhibited nonlinear current–voltage responses enabled realization of a basic Boolean operation of an ionic OR logic gate. Aside from the electrical response, the asymmetric ion transport through the diode was shown to affect the transport of low-abundance molecules across the diode, only allowing crossing when the diode was forward-biased. Integration of multiple diodes enabled implementation of an OR logic operation on both the voltage and the molecule transport, while obtaining electrical and optical output readouts that were associated with low and high logic levels. Similarly to electronics, implementation of logic gates opens up new functionalities of on-chip ionic computation via integrated circuits consisting of multiple basic logic gates. Electrically powered micro- and nanomotors are promising tools for in vitro single-cell analysis. In particular, single cells can be trapped, transported, and electroporated by a Janus particle (JP) using an externally applied electric field. However, while dielectrophoretic (DEP)-based cargo manipulation can be achieved at high-solution conductivity, electrical propulsion of these micromotors becomes ineffective at solution conductivities exceeding "0.3 mS cm-1. Here, JP cargo manipulation and transport capabilities to conductive near-physiological (<6 mS cm-1) solutions are extended successfully by combining magnetic field-based micromotor propulsion and navigation with DEP-based manipulation of various synthetic and biological cargos. Combination of a rotating magnetic field and electric field results in enhanced micromotor mobility and steering control through tuning of the electric field frequency. In addition, the micromotor's ability of identifying apoptotic cell among viable and necrotic cells based on their dielectrophoretic difference is demonstrated, thus, enabling to analyze the apoptotic status in the single-cell samples for drug discovery, cell therapeutics, and immunotherapy. The ability to trap and transport live cells towards regions containing doxorubicin-loaded liposomes is also demonstrated. This hybrid micromotor approach for label-free trapping, transporting, and sensing of selected cells within conductive solutions opens new opportunities in drug delivery and single-cell analysis, where close-to-physiological media conditions are necessary. Organ/organoid-on-a-chip (OoC) technologies aim to replicate aspects of the in vivo environment in vitro, at the scale of microns. Mimicking the spatial in vivo structure is important and can provide a deeper understanding of the cell–cell interactions and the mechanisms that lead to normal/abnormal function of a given organ. It is also important for disease models and drug/toxin testing. Incorporating active fluid flow in chip models enables many more possibilities. Active flow can provide physical cues, improve intercellular communication, and allow for the dynamic control of the environment, by enabling the efficient introduction of biological factors, drugs, or toxins. All of this is in addition to the fundamental role of flow in supplying nutrition and removing waste metabolites. This review presents an overview of the different types of fluid flow and how they are incorporated in various OoC models. The review then describes various methods and techniques of incorporating perfusion networks into OoC models, including self-assembly, bioprinting techniques, and utilizing sacrificial gels. The second part of the review focuses on the replication of spermatogenesis in vitro; the complex process whereby spermatogonial stem cells differentiate into mature sperm. A general overview is given of the various approaches that have been used. The few studies that incorporated microfluidics or vasculature are also described. Finally, a future perspective is given on elements from perfusion-based models that are currently used in models of other organs and can be applied to the field of in vitro spermatogenesis. For example, adopting tubular blood vessel models to mimic the morphology of the seminiferous tubules and incorporating vasculature in testis-on-a-chip models. Improving these models would improve our understanding of the process of spermatogenesis. It may also potentially provide novel therapeutic strategies for pre-pubertal cancer patients who need aggressive chemotherapy that can render them sterile, as well asfor a subset of non-obstructive azoospermic patients with maturation arrest, whose testes do not produce sperm but still contain some of the progenitor cells. Herein, we demonstrate digital microfluidics-like manipulations of preconcentrated biomolecule plugs within a continuous flow that is different from the commonly known digital microfluidics involving discrete (i.e. droplets) media. This is realized using one- and two-dimensional arrays of individually addressable ion-permselective membranes with interconnecting microfluidic channels. The location of powered electrodes, dictates which of the membranes are active and generates either enrichment/depletion diffusion layers, which, in turn, control the location of the preconcentrated plug. An array of such powered membranes enables formation of multiple preconcentrated plugs of the same biosample as well as of preconcentrated plugs of multiple biosample types introduced via different inlets in a selective manner. Moreover, digital-microfluidics operations such as up-down and left–right translation, merging, and splitting, can be realized, but on preconcentrated biomolecule plugs instead of on discrete droplets. This technology, based on nanoscale electrokinetics of ion transport through permselective medium, opens future opportunities for smart and programmable digital-like manipulations of preconcentrated biological particle plugs for various on-chip biological applications. We report the first nanofluidic inductor (L) to complement the known nanofluidic capacitors (C), resistors (R), and diodes for ion currents. Under negative bias, the nanopore behaves like a parallel RC circuit at low frequencies; however, under positive bias, the asymptotic dynamics is that of a serial RL circuit. This new ionic circuit element can lead to nanofluidic RLC or diode-inductor oscillator circuits and new intrapore biosensing/rapid sequencing strategies. A universal theory, with explicit estimates for the capacitance and inductance at opposite biases, is derived to collapse the rectified dynamics of all conic nanopores to facilitate design of this new nanofluidic circuit. Ion current rectification inversion is observed in a funnel-shaped nanochannel above a threshold voltage roughly corresponding to the under-limiting to over-limiting current transition. Previous experimental studies have examined rectification at either low-voltages (under-limiting current region) for conical nanopores/funnel-shaped nanochannels or at high-voltages (over-limiting region) for straight nanochannels with asymmetric entrances or asymmetric interfacing microchannels. The observed rectification inversion occurs because the system resistance is shifted, beyond a threshold voltage, from being controlled by intra-channel ion concentration-polarization to that controlled by external concentration-polarization. Additionally, strong hysteresis effects, due to residual concentration-polarization, manifest themselves through the dependence of the transient current rectification on voltage scan rate. Herein, we studied localized electroporation and gene transfection of mammalian cells using a metallodielectric hybrid micromotor that is magnetically and electrically powered. Much like nanochannel-based, local electroporation of single cells, the presented micromotor was expected to increase reversible electroporation yield, relative to standard electroporation, as only a small portion of the cell's membrane (in contact with the micromotor) is affected. In contrast to methods in which the entire membrane of all cells within the sample are electroporated, the presented micromotor can perform, via magnetic steering, localized, spatially precise electroporation of the target cells that it traps and transports. In order to minimize nonselective electrical lysis of all cells within the chamber, resulting from extended exposure to an electrical field, magnetic propulsion was used to approach the immediate vicinity of the targeted cell, after which short-duration, electric-driven propulsion was activated to enable contact with the cell, followed by electroporation. In addition to local injection of fluorescent dye molecules, we demonstrated that the micromotor can enhance the introduction of plasmids into the suspension cells because of the dielectrophoretic accumulation of the plasmids in between the Janus particle and the attached cell prior to the electroporation step. Here, we chose a different strategy involving the simultaneous operation of many micromotors that are self-propelling, without external steering, and pair with cells in an autonomic manner. The locally electroporated suspension cells that are considered to be very difficult to transfect were shown to express the transfected gene, which is of significant importance for molecular biology research. When an electrorheological fluid is located between two electrodes and an electrical voltage is applied to them, the particles in the fluid move and form chains along the electric field lines. This phenomenon is called the electrorheological effect. The exact behavior of the particles has not yet been studied completely. Some optical investigations of particle motion or behavior have been performed, but did not take into account the high dynamic range directly after the application of an electric field. This study is intended to help explain how the particles behave when they encounter an electric field and then try to align themselves with it. There is an investigation into how these chains develop in a microchannel within milliseconds. For this purpose, the particle behavior of the electrorheological fluid is investigated with high dynamic imaging using a microscope. A high-speed camera records videos of the first milliseconds at 3000 fps synchronously with the application of an electric field. The results provide a better understanding of the chain formation and particle behavior of the electrorheological effect in the high dynamic range and can be used for the design of electrorheological applications as well as simulations of the particle movement. The electrorheological effect produced by an ER-Fluid with an applied electric field is not yet fully understood. However, it is significant to understand how the viscosity increases, and thus, the ER-Effect works to optimally design ER-Applications. In this article, the ER-Effect and its response to different voltage forms with varying frequencies within a miniature fluidic device are described and visualized using a microscope. The different pressure drops across the channel are measured. Problems such as long saturation times using microchannels in combination with low flow velocities are described and explained. An electric frequency dependence of the chain formation has been discovered and that the use of alternating current offers superior performance in comparison with direct current. The optimal operating point for achieving the largest possible pressure difference is from 5 to 10 Hz for square wave voltages. In this frequency range, the performance of the electrorheological effect is greater than using direct current. From approximately 10 Hz, the power decreases with increasing electric frequency. Above 20 Hz, direct current is superior to alternating current voltage, so is not recommended for use in this range. [ABSTRACT FROM AUTHOR]

Abstract: A class of supercolloidal particles that controllably spin about their central axis in AC electric fields is reported. The rational design of these “microspinners” enables their rotation in a switchable manner, which gives rise to several interesting and programmable behaviors. It is shown that due to their complex shape and discrete metallic patches on their surfaces, these microspinners convert electrical energy into active motion via the interplay of four mechanisms at different electric field frequency ranges. These mechanisms of rotation include (in order of increasing frequency): electrohydrodynamic flows, reversed electrohydrodynamic flows, induced charge electrophoresis, and self‐dielectrophoresis. As the primary mechanism powering their motion transitions from one phenomenon to the next, these microspinners display three directional spin inversions (i.e., from clockwise to anticlockwise, or vice versa). To understand the mechanisms involved, this experimental study is coupled with scaling analyses. Due to their frequency‐switchable rotation, these microspinners have potential for applications such as interlocking gears in colloidal micromachines. Moreover, the principles used to power their switchable motion can be extended to design other types of supercolloidal particles that harvest electrical energy for motion via multiple electrokinetic mechanisms. Utilization of active colloids to transport both biological and inorganic cargo has been widely examined in the context of applications ranging from targeted drug delivery to sample analysis. In general, carriers are customized to load one specific target via a mechanism distinct from that driving the transport. Here we unify these tasks and extend loading capabilities to include on-demand selection of multiple nano/micro-sized targets without the need for pre-labelling or surface functionalization. An externally applied electric field is singularly used to drive the active cargo carrier and transform it into a mobile floating electrode that can attract (trap) or repel specific targets from its surface by dielectrophoresis, enabling dynamic control of target selection, loading and rate of transport via the electric field parameters. In the future, dynamic selectivity could be combined with directed motion to develop building blocks for bottom-up fabrication in applications such as additive manufacturing and soft robotics. Photothermal convection has been a major obstacle for stable particle trapping in plasmonic optical tweezer at high optical power. Here, we demonstrate a strategy to suppress the plasmonic photothermal convection by using vanishingly small thermal expansion coefficient of water at low temperature. A simple square nanoplasmonic array is illuminated with a loosely Gaussian beam to produce a two dimensional optical lattice for trapping of micro particles. We observe stable particle trapping due to near-field optical gradient forces at elevated optical power at low temperature. In contrast, for the same optical power at room temperature, the particles are convected away from the center of the optical lattice without their accumulation. This technique will greatly increase usable optical power and enhance the trapping capability of plasmonic optical tweezer. [ABSTRACT FROM AUTHOR]

Background: Myxozoa is a speciose group of endoparasitic cnidarians that can cause severe ecological and economic effects. Although highly reduced compared to free-living cnidarians, myxozoans have retained the phylum-defining stinging organelles, known as cnidae or polar capsules, which are essential to initiating host infection. To explore the adaptations of myxozoan polar capsules, we compared the structure, firing process and content release mechanism of polar tubules in myxospores of three Myxobolus species including M. cerebralis, the causative agent of whirling disease. Results: We found novel functions and morphologies in myxozoan polar tubules. High-speed video analysis of the firing process of capsules from the three Myxobolus species showed that all polar tubules rapidly extended and then contracted, an elasticity phenomenon that is unknown in free-living cnidarians. Interestingly, the duration of the tubule release differed among the three species by more than two orders of magnitude, ranging from 0.35 to 10 s. By dye-labeling the polar capsules prior to firing, we discovered that two of the species could release their entire capsule content, a delivery process not previously known from myxozoans. Having the role of content delivery and not simply anchoring suggests that cytotoxic or proteolytic compounds may be present in the capsule. Moreover, while free-living cnidarians inject most of the toxic content through the distal tip of the tubule, our video and ultrastructure analyses of the myxozoan tubules revealed patterns of double spirals of nodules and pores along parts of the tubules, and showed that the distal tip of the tubules was sealed. This helical pattern and distribution of openings may minimize the tubule mechanical weakness and improve resistance to the stress impose by firing. The finding that myxozoan tubule characteristics are very different from those of free-living cnidarians is suggestive of their adaptation to parasitic life. Conclusions: These findings show that myxozoan polar tubules have more functions than previously assumed, and provide insight into their evolution from free-living ancestors. [ABSTRACT FROM AUTHOR]

Previously, metallodielectric Janus particles have been shown to travel with their dielectric hemisphere forward under low frequency applied electric fields as a result of asymmetric induced-charge electroosmotic flow. Here, it is demonstrated that at high frequencies, well beyond the charge relaxation time of the electric double layer induced around the particle, rather than the velocity decaying to zero, the Janus particles reverse direction, traveling with their metallic hemisphere forward. It is proposed that such motion is the result of a surface force, arising from localized nonuniform electric field gradients, induced by the dual symmetry-breaking of an asymmetric particle adjacent to a wall, which act on the induced dipole of the particle to drive net motion even in a uniform AC field. Although the field is external, since the driving gradient is induced on the particle level, it may be considered an active colloid. We have thus termed this propulsion mechanism "self-dielectrophoresis", to distinguish from traditional dielectrophoresis where the driving nonuniform field is externally fixed and the particle direction is restricted. It is demonstrated theoretically and experimentally that the critical frequency at which the particle reverses direction can be characterized by a nondimensional parameter which is a function of electrolyte concentration and particle size. Here, we report the characterization of the transport of micro- and nanospheres in a simple two-dimensional square nanoscale plasmonic optical lattice. The optical potential was created by exciting plasmon resonance by way of illuminating an array of gold nanodiscs with a loosely focused Gaussian beam. This optical potential produced both in-lattice particle transport behavior, which was due to near-field optical gradient forces, and high-velocity (~μm/s) out-of-lattice particle transport. As a comparison, the natural convection velocity field from a delocalized temperature profile produced by the photothermal heating of the nanoplasmonic array was computed in numerical simulations. We studied the choked flow behavior of gas flow in a single microchannel at inlet pressures of up to 2.2 MPa. Fair agreement was obtained between the basic compressible flow theory, known as Fanno flow, numerical simulations, and experiments for the mass flow rate dependency on the inlet pressure. Investigation of the effect of varying the inlet temperature on the choked flow behavior revealed that the power law scaling of the maximum flow rate on the temperature varies with inlet pressure. The current study extends previous analytical and numerical solutions of chronopotentiometric response of one-dimensional systems consisting of three layers to the more realistic two-dimensional (2D) heterogeneous ion-permselective medium. An analytical solution for the transient concentration-polarization problem, under the local electroneutrality approximation and assumption of ideal permselectivity, was obtained using the Laplace transform and separation of variables technique. Then the 2D electric potential was obtained numerically and was compared to the full Poisson-Nernst-Planck solution. It was then shown that the resultant voltage drop across the system varies between the initial Ohmic response and that of the steady state accounting for concentration polarization. Also, the field-focusing effect in a 2D system is shown to result in a faster depletion of ions at the permselective interface. [ABSTRACT FROM AUTHOR]
