Abstract In this study, we concentrate on the seismic signature of subducted sediments and suggest the formation of the L‐discontinuity beneath the ancient craton related to migrated sediment dehydration. We first determined the single‐crystal elasticity of topaz, the product of sediment dehydration, at high pressures and temperatures by Brillouin scattering. Using the derived elastic parameters, we establish the velocity and density profiles of subducted sediments in the upper mantle. According to our modeling results, 8.5–17.5 vol.% sediments intruding into the upper mantle will induce a 2%–4% low‐VS anomaly at 210–260 km. Meanwhile, continuous heating will lead to the dehydration of phengite in sediments. The dehydration of this amount sediments can generate a 3%–6% ISS with negative Clapeyron slopes, satisfying the observed L‐discontinuity in northern Finland and northern America without the anisotropy changes but accompanied by low‐velocity anomalies. Our study thus provides new insights into the origin of the L‐discontinuity.

Polycyclic aromatic hydrocarbons (PAHs) and dioxins are potential causes of multiple diseases by activating the aryl hydrocarbon receptor (AhR) pathway. Health risk assessment of chemicals primarily relies on the relative potency factor (RPF), although its accuracy may be limited when solely using EC50 values. The induction of cytochrome P4501A1 (CYP1A1) serves as a biomarker for AhR activation and is an integrator of dioxin-like toxicity. Here, we present a method for evaluating the risks associated with AhR activation using mathematical models of dose-CYP1A1 induction. The dose-effect curves for certain PAHs and dioxins, including Ant, BghiP, 1,2,3,4,7,8-HxCDD, and others, exhibited a non-classical S-shaped form. The toxic equivalent factor (TEF) profiles revealed a broad range of toxic equivalent factor values. The TEFs for PAHs ranged from approximately 0.01 to 6, with higher values being observed when the concentration was less than 10−10 M, with the exceptions of Ace, Phe, and BghiP. Most congeners of dioxins got the lowest TEF value at around 10−10 M, ranging from 0.04 to 1.00. The binding affinity of AhR to ligands did not display a strong correlation with the EC50 of CYP1A1 expression, suggesting that the AhR-mediated effects of PAHs and dioxins are not fixed but instead fluctuate with the dose. Air samples acquired from a parking area were used to compare the proficiency of RPF and our current approach. In the current method, naphthalene and chrysene were the primary contributors of PAHs to AhR-mediated risks in parking lots air samples, respectively. However, the contributions of naphthalene and chrysene could be disregarded in the RPF approach.

Polybrominated biphenyl ethers (PBDEs) have been pervasively used as an additive flame retardant in various fields. The nature of environmental persistence, bioaccumulation, and potential toxicity attracted attention worldwide and became a threat to both the ecosystem and human health. Originally, PBDEs are produced in chemical factories. PBDEs exposure was mainly by ingestion of contaminated food products, followed by inhalation of particles containing deposited PBDEs. This review concluded the liver injuries, thyroid hormone disorder, neurotoxicity, genotoxicity and immune system disorders as five major toxicities via overexposure to PBDEs and their toxic mechanism. PBDEs can cause severe consequences by inducing mitochondria dysfunction and oxidative stress, disrupting neurotransmitter metabolism and compromising the integrity of DNA, etc. This review presented an in-depth analysis of the most recent toxicological evidence to offer suggestions for future studies to control human overexposure to PBDEs.

Abstract This study investigated two random forest (RF) models for the non‐iterative parametrization of surface‐layer turbulent fluxes: (a) the RF scheme, a calculation model that is directly trained using correlated variables, and (b) the RF_Li10 scheme, a random forest correction model based on the Li10 scheme (Li et al., 2010, https://doi.org/10.1007/s10546-010-9523-y). A comparison between these two new models and the Li10 scheme against an iterative scheme revealed the hierarchy of maximum relative errors in estimating stability parameters, as well as momentum and heat transfer coefficients. This hierarchy is as follows: the Li10 scheme is the greatest, followed by the RF scheme, with the RF_Li10 scheme exhibiting the least errors.

Resorcinol-bis(diphenyl)-phosphate (RDP), an emerging organophosphate flame retardant, is increasingly used as a primary alternative for decabromodiphenyl ether and is frequently detected in global environmental matrices. However, the long-term effects of its exposure to humans remain largely unknown. To investigate its intergenerational transfer capacity and health risks, female Sprague Dawley rats were orally exposed to RDP from the beginning of pregnancy to the end of the lactation period. The RDP content, gut microbiota homeostasis, and metabolic levels were determined. RDP accumulation occurred in the livers of maternal rats and offspring and increased with exposure time. 16S rRNA gene sequencing showed that exposure to RDP during pregnancy and/or lactation significantly disrupted gut microbiota homeostasis, as evidenced by decreased abundance and diversity. In particular, the abundance of Turicibacter, Adlercreutzia, and YRC22 decreased, correlating significantly with glycollipic metabolism. This finding was consistent with the reduced levels of short-chain fatty acids, the crucial gut microbial metabolites. Meanwhile, RDP exposure resulted in changes in gut microbiome-related metabolism. Nine critical overlapping KEGG metabolic pathways were identified, and the levels of related differential metabolites decreased. Our results suggest that the significant adverse impacts of RDP on gut microbiota homeostasis and metabolic function may increase the long-term risks related to inflammation, obesity, and metabolic diseases.

Decabromodiphenyl ethane (DBDPE) as a widely used brominated flame retardant is harmful to human health due to its toxicity, including cardiovascular toxicity, reproductive toxicity, and hepatotoxicity. However, the knowledge of the long-term effects and structural and metabolic function influence on gut microbiota from DBDPE exposure remains limited. This study was mainly aimed at the gut microbiome and fecal metabolome of female rats and their offspring exposed to DBDPE in early life. 16S rRNA gene sequencing demonstrated that maternal DBDPE exposure could increase the α-diversity of gut microbiota in immature offspring while decreasing the abundance of Bifidobacterium, Clostridium, Muribaculum, Escherichia, and Lactobacillus in adult offspring. The nonmetric multidimensional scaling showed a consistency in the alternation of β-diversity between pregnant rats and their adult offspring. Furthermore, the short-chain fatty acids produced by gut microbiota dramatically increased in adult offspring after maternal DBDPE exposure, revealing that DBDPE treatment disrupted the gut microbial compositions and altered the gut community's metabolic functions. Untargeted metabolomics identified 41 differential metabolites and seven metabolic pathways between adult offspring from various groups. Targeted metabolomic showed that maternal high dose DBDPE exposure obviously decreased the level of glutathione, taurine, and l-carnitine in their adult offspring, which verified the correlation between weight loss and amino acid metabolites. An interesting link between some gut bacteria (especially the Firmicutes) and fecal metabolites demonstrated the shifts in gut microbiota may drive the metabolic process of fecal metabolites. The current findings provide new insight into long-term effects on human health.

This review explored the potential of edible insects to address the challenges of malnutrition and food security. Although grain production in China has met the Food and Agriculture Organization standards, the shortage of protein supply is still a big issue. Moreover, expanding livestock farming is considered unsustainable and environmentally unfriendly. Edible insects have become an alternative with higher sustainable and ecological properties. There are 324 species of insects currently consumed in China, and they have high nutritional value, with a rich source of protein and unsaturated fatty acids. Insect farming provides numerous benefits, including green feeds for livestock, poultry, and aquaculture, sustainable organic waste management, as well as industrial and pharmaceutical raw materials. The food toxicological evaluations conducted in China indicated that edible insects are safe for general consumption by the Chinese, but allergies and other related food safety issues should not be ignored. Consumer acceptance is another barrier to overcome, with different schemas between China and Western countries. More research on the potential functions of edible insects and their product development may enhance their acceptance in China. Overall, incorporating edible insects into our diet is a promising solution to address challenges related to protein supply and food security. To ensure safety and sustainability, appropriate legislation, quality regulations, large-scale insect farms, and acceptable processing techniques are necessary. Moreover, more scientific research and social awareness are required to promote the culture and utilization of edible insects in China.

Abstract Background The PIG-A gene mutation assay is a valuable tool for measuring in vivo gene mutations in blood cells. The human PIG-A assay, used as a potential genotoxicity biomarker, is minimally invasive, sensitive, and cost-efficient; however, the relationship between carcinogen exposure and PIG-A mutations is not well understood. Methods We investigated the genotoxic effect of red blood cells using PIG-A assay and lymphocyte cytokinesis-block micronucleus test in barbecue restaurant workers (N = 70) exposed to polycyclic aromatic hydrocarbons (PAHs) and self-identified healthy control subjects (N = 56). Urinary PAH metabolites were measured to evaluate internal exposure levels. Results Multivariate Poisson regression showed that the PAH-exposed workers exhibited significantly higher PIG-A mutant frequency (MF) (8.04 ± 6.81 × 10− 6) than did the controls (5.56 ± 5.26 × 10− 6) (RR = 0.707, 95% CI: 0.615–0.812, P

BackgroundNeuronal intranuclear inclusion disease (NIID) is a rare chronic progressive neurodegenerative disease, with complex and diverse clinical manifestations and pathological eosinophilic hyaline intranuclear inclusions in the central and peripheral nervous systems and visceral organs. Improvements in diagnostic methods such as skin biopsy and gene testing are helpful in revealing the clinical and genetic characters of NIID.Materials and methodsWe presented two cases of NIID diagnosed by using NOTCH2NLC gene testing and skin biopsy. Diffusion weighted imaging (DWI) showed high linear intensity in corticomedullary junction. We also reviewed all the published NIID cases with positive NOTCH2NLC GGC repeat expansion and skin biopsy results in PubMed.ResultsPatient 1 was a 63-year-old male who carried 148 GGC repeats and presented with progressive tremor and limb weakness. Patient 2 was a 62-year-old woman who carried 131 GGC repeats and presented with tremors, memory loss and headaches. The most common clinical manifestation of 63 NIID patients in this study was cognitive impairment, followed by tremors. In our study, almost all the patients were from East Asia, the male to female ratio was 1:1.26, with an age of onset of 54.12 ± 14.12 years, and an age of diagnosis of 60.03 ± 12.21 years. Symmetrical high signal intensity at the corticomedullary junction on DWI were revealed in 80.96% of the patients. For the GGC repeat numbers, the majority of GGC repeats were in the 80–119 intervals, with few GGC repeats above 160. The number of GGC repetitions was significantly higher in patients presented with muscle weakness than in other clinical manifestations.ConclusionNIID is a neurodegenerative disease caused by aberrant polyglycine (polyG) protein aggregation. NIID mostly occurs in the elderly population in East Asia, with cognitive dysfunction as the most common symptom. Staging NIID based on clinical presentation is inappropriate because most patients with NIID have overlapping symptoms. In our study, there was no significant correlation between the number of GGC repeats and different phenotypes except for muscle weakness. Abnormal trinucleotides repeat and PolyG protein aggregation maybe common pathogenic mechanism in neurodegenerative diseases and cerebrovascular diseases, which needs to be confirmed by more studies.

This study simulated the spatial distribution of wind speeds and wind energy density by using the WRF model, and we used the WRF-simulated results to evaluate the sea surface wind speeds retrieved from the HY-2C and CFOSAT satellite-borne microwave scatterometers over the Yellow Sea region. The main conclusions were as follows: (1) The combination of the MRF boundary layer parameterization scheme, the MM5 near-surface parameterization scheme, and the Global Data Assimilation System (GDAS) initial field demonstrated the best performance in simulating the 10 m wind speed in the Yellow Sea region, with a root-mean-square error (RMSE) of 1.57, bias of 1.24 m/s, and mean absolute percentage error (MAPE) of 17%. (2) The MAPE of the HY-2C inversion data was 9%, while the CFOSAT inversion data had an MAPE of 6%. The sea surface wind speeds derived from the HY-2C and CFOSAT satellite scatterometer inversions demonstrated high accuracy and applicability in this region. (3) The wind speed was found to increase with altitude over the Yellow Sea, with higher wind speeds observed in the southern region compared to the northern region. The wind power density increased with altitude, and the wind power density in the southern area of the Yellow Sea was higher than in the northern region. (4) The CFOSAT satellite inversion products were in good agreement with the WRF simulation results under low wind speed conditions. In contrast, the HY-2C satellite inversion products showed better agreement under moderate wind speed conditions. Under high wind speed conditions, both satellite inversion products exhibited minor deviations, but the HY-2C product had an overall overestimation, while the CFOSAT product remained within the range of −1 to 1 m/s. (6) The wind power density increased with the satellite-inverted 10 m wind speed. When the 10 m wind speed was less than 9 m/s, the wind power density exhibited a roughly cubic trend of increase. However, when the 10 m wind speed exceeded 9 m/s, the wind power density no longer increased with the rise in 10 m wind speed. These findings provide valuable insights into wind energy resources in the Yellow Sea region and demonstrate the effectiveness of satellite scatterometer inversions for wind speed estimation. The results have implications for renewable energy planning and management in the area.

Polycyclic aromatic hydrocarbons (PAHs) derived from the incomplete combustion of organic materials are associated with adverse health effects. However, little is known about PAH exposure levels and their influencing factors on a large scale in developing countries. In this study, urinary monohydroxylated metabolites of PAHs (OH-PAHs), including the metabolites of naphthalene, fluorene, phenanthrene, pyrene, chrysene, and benzo[a]pyrene, were measured in 1154 samples in the general population nationwide from 26 provincial capitals in China. Concentrations of OH-PAHs ranged from 1.39 to 228 μg/L. OH-Nap, metabolite of naphthalene, was the predominant compound, accounting for 65.1% of totals. People in eastern, southwest and northeast China, such as Shanghai, Kunming, Nanning, and Changchun, suffered more PAH exposure than other regions which might associate with sampling time, living habits of the subjects, and the imbalance of economic development and energy consumption across regions. Urinary OH-PAH concentrations were associated with body mass index, gender, and age, and smoking was the main correlating factor. Inhalation and diet might be the main exposure route of human exposure to PAHs, especially for smokers by inhalation. Hazard indices showed that no subject was exposed to PAHs with potential non-carcinogenic risk. Furthermore, the carcinogenic risk was the most significant health effects, with almost all subjects having carcinogenic risk values higher than the acceptable level of 10−6. Naphthalene and phenanthrene were the main contributors. The results also suggested a possible relationship between PAH exposure and lung cancer in the Chinese population. This first nationwide study on human internal exposure to PAHs provides a large body of scientific information for governmental decision-making about associated human health and the prevention of human exposure to PAHs.

Due to the prohibition of polybrominated diphenyl ethers, organophosphate flame retardants (OPFRs) and tetrabromobisphenol A (TBBPA) have become emerging flame retardants. However, knowledge about their occurrence, especially their transformation products, is still limited. This study collected sediment samples from two rivers, i.e., Lianjiang River (located at an e-waste dismantling area) and Xiaoqing River (situated at a flame retardant production base), to investigate the occurrence, composition, and spatiality distribution of OPFRs, TBBPA, and their transformation products. Both targets were detected in the Lianjiang River in the range of 220–1.4 × 104 and 108–3.1 × 103 ng/g dw (dry weight) for OPFRs and TBBPA, and 0.11–2.35 and 4.8–414 ng/g dw for their respective transformation products, respectively. The concentrations of OPFRs and TBBPA in the Xiaoqing River ranged from 4.15 to 31.5 and 0.76–2.51 ng/g dw, respectively, and no transformation products were detected. Different compositional characteristics of OPFRs and distinct spatial distribution from mainstream and tributary observed between the two rivers are attributed to the difference in the local industries. Spatial distribution and principal component analysis indicated that e-waste dismantling activities could be a vital source of local pollution. Besides, the confluence of tributaries seemed to determine the contaminant levels in the Xiaoqing River. Also, concentration ratios and Spearman’s correlation between metabolites and parent chemicals were analyzed. Low concentration ratios (3.6 ×10–4 to 0.16) indicated a low transformation degree, and Spearman’s correlation analysis suggested transformation products were partly stemming from commercial products. Considering the limited study of these transformation products, more studies on their sources, transform mechanism, and toxicity are required.

Abstract Background Alzheimer's disease (AD) is the most common degenerative disease characterized by cognitive impairment, memory decline, and language disorder for which there is no effective treatment. Neurogenesis has been indicated in AD and may play an important role in the pathogenesis of AD. Targeting this pathway is a new idea for the treatment of the disease. A recent study reveals that the cellular prion protein (PrP), a receptor for Aβ oligomers, regulates neurogenesis, and its elevated expression is related to cell differentiation. The aim of the present study was to investigate the neuroprotective effects of 6D11 (PrP monoclonal antibody) via neurogenesis promotion in APP/PS1 transgenic mice and Aβ‐induced cell model of AD. Methods In the present study, 9‐month‐old male APP/PS1 mice were injected with 6D11. Then, the Morris water maze was used to examine the spatial learning and memory abilities of the mice in both groups, and immunostained was used to assess the level of Aβ, neurogenesis, and neural stem cells (NSCs) differentiation. Results 6D11 attenuated cognitive deficits in APP/PS1 transgenic mice, which was accompanied by a decrease of the deposition of Aβ. In addition, 6D11 treatment promoted differentiation of the existing hippocampal cells to neurons. Conclusions Our findings confirmed that 6D11 has a therapeutic effect in APP/PS1 transgenic AD mouse model and Aβ‐induced AD cell model, and the effect exerted via increase of neurogenesis and cell differentiation by transduction of Aβ peptide signal.

A model-free fractional-order sliding mode control (MFFOSMC) method based on a non-linear disturbance observer is proposed for the electric drive system in this paper. Firstly, the ultra-local model is established by using the mathematical model of electric drive system under parameter perturbation. Then, aiming at reducing the chattering of the sliding mode controller and improving the transient response, a model-free fractional-order sliding mode controller is designed based on fractional-order theory. Next, considering that the traditional sliding mode control can only suppress matched disturbance and that it is sensitive to mismatched disturbance, a non-linear disturbance observer is used to estimate disturbance, and the estimated variables are used in the design of a sliding mode surface to improve the tracking accuracy of the system. Finally, the experiment is completed on an asynchronous motor drive platform. Compared with the model-free integer-order sliding mode control (MFIOSMC), the results show that the proposed method has good dynamic response and strong robustness. Meanwhile, the proposed method reduces the dependence on mathematical models.

Several in vitro methods for simulating human gastrointestinal digestion have been validated for predicting the bioavailability of heavy metals, but the methods for successfully predicting the bioavailability of organic pollutants are still limited. In this study, we used an adapted fasting in vitro digestion method (Fa-VDM) from the Simulator of the Human Intestinal Microbial Ecosystem and four other in vitro methods comprising In Vitro Gastrointestinal, a physiologically-based extraction test, the unified BARGE method, and Deutsches Institut für Normung e.V. in order to measure the bioaccessibility of polybrominated diphenyl ethers (PBDEs) in soils from an e-waste dismantling town, China, with a Standard Reference Material (SRM2585) as the control. Furthermore, the bioaccessibility data were compared with the bioavailability measured using female C57BL/6 mice. The bioavailability of PBDEs in the soils and SRM2585 were 1.7% to 38.1% and 3.9% to 48.8%, respectively, and the bioaccessibility determined using Fa-VDM were 1.6–55.4% and 6.7–32.1%. There were negative and parabolic correlations between octanol/water partition coefficient for PBDEs and the bioavailability and bioaccessibility, respectively, whereas the H/C ratios and organic matter contents of the soils did not correlate with them. The bioaccessibility data determined by Fa-VDM were generally higher than those obtained using the other four methods, mainly due to the higher bile concentration and larger liquid to solid ratio in the digestion solution in Fa-VDM. There was a significant linear relationship between the results according to the in vivo and in vitro method of Fa-VDM where the slopes varied from 0.83 to 1.16 (R2 > 0.73) and intercepts from 0.3%–7.7% for BDE47, 99, 100, and 153 measured using Fa-VDM, thereby indicating that the bioaccessibility assessed by this method can potentially be used to predict the bioavailability of moderately brominated congeners in soils. Persistent halogenated organic pollutants (HOPs) are a class of toxic chemicals, which may have adverse effects on fetuses via transplacental transfer from their mothers. Here, we review reported internal exposure levels of various HOPs (organochlorinated pesticides, polychlorinated biphenyls, polybrominated diphenyl ethers, short- and medium-chain chlorinated paraffins, and per- and poly-fluoroalkyl substances) in placenta, and both maternal and umbilical cord sera. We also present analyses of the transplacental transfer and placental distribution characteristics of each class of compounds, and discuss effects of several factors on the transfer and accumulation efficiencies of HOPs, as well as the main mechanisms of HOPs’ transfer across the placental barrier. Reported compound-specific transplacental transfer efficiencies and distribution efficiencies, expressed as umbilical cord:maternal serum and placental:maternal serum concentration ratios (RCM and RPM, respectively), are summarized. Average published RCM values of the HOPs range from 0.24 to 3.08 (lipid-adjusted) and from 0.04 to 3.1 (based on wet weights), and are highest for perfluoroalkylcarboxylates (PFCAs) and tetrabromobisphenol A. Average published RPM values range from 0.14 to 1.02 (lipid-adjusted) and from 0.30 to 1.4 (based on wet weights). The broad RCM and RPM ranges may reflect effects of various factors, inter alia physicochemical properties of HOPs, metabolic capacities of mothers and fetuses, placental maturity, and differential expression of influx/efflux transporters in the placenta. Generally, HOPs’ RCM values decline linearly with molecular size, and are curvilinearly related to solubility. Plasma protein binding affinity and the difference between maternal and fetal metabolic capacities may also affect some HOPs’ transfer efficiencies. HOPs’ molecular size may be influential. Transplacental transport of HOPs likely occurs mostly through passive diffusion, although influx/efflux transporters expressed on maternal and/or fetal sides of the placenta may also facilitate or hinder their transport. Overall, the review highlights clear gaps in our understanding of mechanisms involved in HOPs’ transplacental transport.

Previous research has shown the absorption of polybrominated diphenyl ethers (PBDEs) in the human gastrointestinal tract, but limited attention has been given to the influence of nutrients on PBDE absorption from food matrices. We investigated the effects of nutrients (oil, starch, protein, and dietary fiber) on the absorption and transport of PBDEs in a Caco-2 cell model and bioaccessibility of PBDEs by an in vitro gastrointestinal digestion method. The results showed that the accumulation ratios of PBDE congeners in Caco-2 cells were higher in the nutrient addition groups (oil: 26.7–50.6%, starch: 27.0–58.7%, protein: 12.1–44.1%, and dietary fiber: 28.2–55.1%) than the control group (7.17–36.1%), whereas the transport ratios were lower (oil: 2.30–7.20%, starch: 1.55–9.15%, protein: 1.04–8.78%, and dietary fiber: 0.85–7.04%) than control group (3.78–11.1%). Additionally, the PBDE bioaccessibility could be increased by adding the nutrients, particularly oil and starch. This study clarified the differences in PBDE absorption in the presence of nutrients using the in vitro digestion and Caco-2 cell model. The findings showed that nutrients were an important factor that promoted PBDE absorption in the gastrointestinal tract. Therefore, it is important to focus on a novel dietary strategy of food consumption with contaminant compounds to protect human health.

Many hazardous substances can be released during incineration of municipal solid waste (MSW), which pose a potential threat to human health. As additives, phthalates (PAEs) and bisphenols (BPs), which are widely used in daily goods, are likely to be present in the released hazardous substances. In the present study, we investigated the urinary levels of phthalate metabolites (mPAEs) and BPs in workers in an MSW incineration plant (the exposed group) and in residents 8 km away (the control group) in Shenzhen, China. The results showed that the median total urinary concentration of mPAEs in workers was significantly higher than that in residents (1.02 × 103 vs. 375 ng/mL). However, there was no significant difference between workers and residents for BPs. Among the mPAEs measured, the most abundant compound was mono-n-butyl phthalate in both exposed and control groups. Monoethyl phthalate and monomethyl phthalate might be potential markers for MSW incineration because of significantly high levels in the exposed group. The workers engaged in different types of workshops showed no significant differences in the urinary levels of mPAEs, also for BPs. It was worth noting that 70.8% of workers were at risk of the non-carcinogenic effects caused by PAEs with diethylhexyl phthalate having the highest risk. Actions should be taken to reduce the risks caused by these hazardous chemicals.

Extensive electronic waste (e-waste) recycling might be an important emission source of polycyclic aromatic hydrocarbons (PAHs) mixture, which might induce negative effects on the employees. In the present work, atmospheric pollution patterns of PAHs and their derivatives were determined in five different workshops to dismantle waste printed circuit boards (WPCBs) via thermal treatment. The results showed that mass concentrations of PAHs, chlorinated PAHs (ClPAHs), brominated PAHs (BrPAHs), oxy-PAHs (OPAHs) as well as carbazole (CBZ) were ranged from 1.53 × 104–2.02 × 105, 32.3–364, 8.29–1.13 × 103, 923–1.39 × 104 and 225–1.95 × 103 pg·m−3, respectively. Electric heating furnaces (EHF) workshops emitted relatively higher contaminants than other disposal sectors. OPAHs was found to be the most predominant derivatives of PAHs with 9,10-anthraquinone (83.0%) has the absolute superior in EHFTV, while benzo(a)anthracene-7,12-dione (>45.0%) was found to be the highest congener in other workshops, respectively. 9,10-Cl2Phe exhibited the largest contributions to the ΣClPAHs whereas the composition profiles of BrPAHs varied among five workshops. In addition to direct chlorination of parent PAHs, thermal degradation of halogenated flame retardants incorporated into plastic materials might dominate the generation of Cl/BrPAHs from e-waste dismantling activities. The specific isomeric ratios of BrPAHs (3-BrFlu/1-BrPyr and 1-BrPyr/3-BrFlu) might be used to discriminate other emission sources from pyrolysis of WPCBs. However, their specific application as novel tracers for source identification should be further verified with more studies. The emitted PAHs mixture with their derivatives in all dismantling workshops posed carcinogenic risks to these dismantling workers via inhalation, particularly the workshop using electric heating furnaces to treat router. Nevertheless, new loadings of PAHs derivatives observed from e-waste dismantling activities, as well as their comprehensive health risk assessment provides us with a fresh perspective on the source appointment and potential adverse consequences of PAHs. More attention needs to be paid to the potential carcinogenic risks of exposure to PAHs and their derivatives from e-waste dismantling processes.

The gut microbiota has been shown to be highly involved in many vital physiological processes that play key roles in human health. The intergenerational transfer of Dechlorane Plus (DP) and the complex interaction between DP and microbiota has been poorly studied. Additionally, the structural and functional effects of DP on the gut microbiota have not been studied. This study aimed to investigate the DP transfer in Sprague-Dawley rats during pregnancy and the effects of DP exposure on gut microbiota, as detected by 16S rRNA gene sequencing. The results showed that excretion in feces is a very important elimination pathway of orally dosed DP. The main intergenerational transfer pathway of DP might be via lactation rather than transplacental transport. The 16S rRNA sequencing revealed that DP exposure could decrease the richness and diversity of gut microbiota, especially at the genus level. Furthermore, in DP exposure groups, the gut microbiota production of metabolites of short-chain fatty acids was dramatically increased. The results demonstrated that DP exposure not only altered the gut microbiota structures, but also immensely influenced metabolic functions, causing long-term impact to offspring. This data indicates that more attention should be paid to the long-term health effects related to DP exposure.

Electronic waste (e-waste) dismantling is an important source of flame retardant emissions, and may have potentially adverse effects on surrounding area. This study investigated their influence on the surrounding area and the human health risks after an industrial park was built in 2015 and environmentally friendly technologies were introduced at an e-waste dismantling site in South China. The concentrations of flame retardants, including polybrominated diphenyl ethers (PBDEs), polybromobenzenes (PBBzs), Dechlorane plus (DP), and organophosphate esters (OPEs), in the soils were measured. The results showed that soil contamination was greater in the industrial park than in the surrounding area. The PBDE concentrations were the highest with BDE209, a daca-BDE, being the dominant congener, followed by OPEs, where triphenyl phosphate levels were the highest. Furthermore, triphenyl phosphate can be used as an indicator of flame retardant emissions during e-waste dismantling in this region. The fanti value of DP was stable at around 0.75. The principal component analysis showed that direct emission was the major source of pollutants in the industrial park. The direct emission proportion decreased in the surrounding area, but migration and transformation increased. None of the chemicals posed a non-carcinogenic risks to children and adults via oral uptake or dermal contact when the absorption factors of the chemicals were included in the estimation. However, the total hazard quotients for children were close to a unit in the industrial park, of which, the PBDE and OPE proportions accounted for 84.2% and 15.8% of the total, respectively. However, the PBBz and DP percentages were negligible. Therefore, PBDEs and OPEs should be given more attention in the future.

In this study, 96 pairs of hair and urine samples were collected from e-waste (EW) dismantling workers of an industrial park, as well as residents living in surrounding areas. The concentrations of polycyclic aromatic hydrocarbons (PAHs) and hydroxylated PAH metabolites (OH-PAHs) were analyzed . The results show that concentrations of Σ15PAHs ranged from 6.24 to 692 ng/g dry weight (dw) and Σ12OH-PAHs from undetected to 187 ng/g dw in hair external (hair-Ex), and ranged from 31.7 to 738 ng/g dw and 21.6 to 1887 ng/g dw in hair internal (hair-In). There was no significant difference in exposure levels between EW dismantling workers and residents of the surrounding area. For the parent PAHs, the concentrations of Σ15PAHs of hair-In were comparable with those of hair-Ex for all populations except for the child residents. On the contrary, for the OH-PAHs, the concentrations of Σ12OH-PAHs of hair-In were 9–37 times higher than those of hair-Ex for populations. Moreover, the congener profiles of OH-PAHs of hair-In were different from those of hair-Ex, but similar to that of urine. Particularly, 3-OH-Bap, which is a carcinogenic metabolite, was only detected in the hair-In. These results indicate that OH-PAHs in hair-In, just like in urine, are mainly derived from endogenous metabolism and could be considered as reliable biomarkers for PAHs exposure. However, there was almost no significant correlations between hair-In and urine for OH-PAHs. This indicates that more attention should be paid to OH-PAHs when conducting PAHs exposure risk assessment using hair, which will help to obtain more reliable and comprehensive information on health risk assessments. Electroencephalography neurofeedback (EEG-NFB) training can induce changes in the power of targeted EEG bands. The objective of this study is to enhance and evaluate the specific changes of EEG power spectral density that the brain-machine interface (BMI) users can reliably generate for power augmentation through EEG-NFB training. First, we constructed an EEG-NFB training system for power augmentation. Then, three subjects were assigned to three NFB training stages, based on a 6-day consecutive training session as one stage. The subjects received real-time feedback from their EEG signals by a robotic arm while conducting flexion and extension movement with their elbow and shoulder joints, respectively. EEG signals were compared with each NFB training stage. The training results showed that EEG beta (12–40 Hz) power increased after the NFB training for both the elbow and the shoulder joints’ movements. EEG beta power showed sustained improvements during the 3-stage training, which revealed that even the short-term training could improve EEG signals significantly. Moreover, the training effect of the shoulder joints was more obvious than that of the elbow joints. These results suggest that NFB training can improve EEG signals and clarify the specific EEG changes during the movement. Our results may even provide insights into how the neural effects of NFB can be better applied to the BMI power augmentation system and improve the performance of healthy individuals.

Brain–Machine Interfaces (BMIs) have attracted much attention in recent decades, mainly for their applications involving severely disabled people. Recently, research has been directed at enhancing the ability of healthy people by connecting their brains to external devices. However, there are currently no successful research reports focused on robotic power augmentation using electroencephalography (EEG) signals for the shoulder joint. In this study, a method is proposed to estimate the shoulder’s electromyography (EMG) signals from EEG signals based on the concept of a virtual flexor–extensor muscle. In addition, the EMG signal of the deltoid muscle is used as the virtual EMG signal to establish the EMG estimation model and evaluate the experimental results. Thus, the shoulder’s power can be augmented by estimated virtual EMG signals for the people wearing an EMG-based power augmentation exoskeleton robot. The estimated EMG signal is expressed via a linear combination of the features of EEG signals extracted by Independent Component Analysis, Short-time Fourier Transform, and Principal Component Analysis. The proposed method was verified experimentally, and the average of the estimation correlation coefficient across different subjects was 0.78 (±0.037). These results demonstrate the feasibility and potential of using EEG signals to provide power augmentation through BMI technology.

Chronic inflammation drives the development of colorectal cancer (CRC), where tumor-infiltrating immune cells interact with cancer cells in a dynamic crosstalk. Mast cells (MC), one of earliest recruited immune cells, accumulate in CRC tissues and their density is correlated with cancer progression. However, the exact contribution of MC in CRC and their interaction with colon cancer cells is poorly understood. Here, we investigated the impact of primary human MC and their mediators on colon cancer growth using 2D and 3D coculture models. Primary human MC were generated from peripheral CD34+ stem cells. Transwell chambers were used to analyze MC chemotaxis to colon cancer. Colon cancer cells HT29 and Caco2 differentially recruited MC by releasing CCL15 or SCF, respectively. Using BrdU proliferation assays, we demonstrated that MC can directly support colon cancer proliferation and this effect was mediated by their cellular crosstalk. 3D coculture models with cancer spheroids further confirmed the pro-tumor effect of MC on colon cancer growth, where direct cell-cell contact is dispensable and increased production of multiple soluble mediators was detected. Moreover, TLR2 stimulation of MC promoted stronger growth of colon cancer spheroids. By examining the transcriptome profile of colon cancer-cocultured MC versus control MC, we identified several MC marker genes, which were deregulated in expression. Our study provides an advanced in vitro model to investigate the role of human MC in cancer. Our data support the detrimental role of MC in CRC development and provide a molecular insight into the cellular crosstalk between MC and colon cancer cells.

Mast cells (MC) accumulate in colorectal cancer (CRC) and the relationship between MC density and cancer progression has been well recognized. MC can be either pro-tumor or anti-tumor players, depending on the local factors present in the tumor microenvironment. Upon malignant transformation, cancer cells express high levels of sialic acids on cell membrane or by secretion. Siglecs are a family of immunoglobulin-like receptors that bind sialic acids and each subtype has a distinct pattern of expression on immune cells. Among them, Siglec-6 is expressed predominately by MC. However, the function of Siglec-6 in MC is largely unexplored and whether it is expressed by CRC-associated MC remains unknown. In this study, we explored the function of Siglec-6 in CD34+ derived human MC. MC activation was initiated by IgE crosslinking with or without preincubation of anti-Siglec-6 Ab. Siglec-6 engagement significantly attenuated IgE-dependent MC degranulation as measured by ß-hexosaminidase release and CD63 expression. Interestingly, the production of GM-CSF was also shown reduced upon Siglec-6 engagement. To mimic the milieu of CRC, we cultured primary human MC with colon cancer cells or under hypoxia and Siglec-6 was then measured on these conditioned MC. Coculture with colon cancer cells (HT29 and Caco2) induced upregulation of Siglec-6 on MC. In comparison, normal colon cells (CCD841) had no effect. Also, a time-dependent increase of Siglec-6 by MC was observed under 1% O2. Immunohistochemistry of CRC tissue showed expression of Siglec-6 by MC in submucosa. Lectin immunochemistry revealed the presence of actual ligands for Siglec-6 in human CRC tissues. Together, our findings illustrate that Siglec-6 is a functionally inhibitory receptor on MC and suggest that Siglec-6 expression may be relevant for MC activity in the tumor microenvironment of CRC.

Cancer-associated antigen 215 (CA215) is an immunoglobulin molecule expressed by numerous tumor types. Membrane-bound and soluble CA215 have been detected in the majority of cancer cells and rarely identified in normal tissues. In addition, CA215C is a carbohydrate-associated epitope in the variable region of CA215, which is specifically recognized by the monoclonal antibody, RP215. However, CA215C is not a suitable vaccine candidate as it is a thymus-independent antigen. In the present study, RP215 was used as a target to screen short peptide mimics of CA215C from a phage display peptide library. Following three rounds of screening, 30 positive phage clones that specifically bound to RP215 were identified and sequenced. The result of amino-acid sequence analysis revealed five conserved sequence groups for seventeen of the positive phage clones. The sequences of phage clones 2, 13 and 42 were selected for peptide synthesis and binding analysis. The synthetic peptides R2 and R42 specifically bound RP215. Antisera from mice immunized with R2-BSA or R42-BSA bound purified CA215C and innate CA215C expressed on human hepatic and rectal carcinoma tissues, as demonstrated by immunohistochemistry. Furthermore, R2-BSA and R42-BSA antisera inhibited RP215 binding to cancer tissues. These results revealed that R2-BSA and R42-BSA antisera had similar characteristics to RP215 and that the synthetic peptides R2 and R42 may mimic the CA215C epitope. Isolated cortical vein thrombosis (ICoVT) is a rare form of cerebral venous thrombosis and is easily misdiagnosed as brain tumor due to its atypical clinical presentations and radiological findings similar to brain tumors. The present study focused on 4 patients with ICoVT, 2 men and 2 women. In the 2 male patients, the onset of disease was insidious, with progressive numbness and weakness in limbs as the major symptoms. By contrast, 2 female patients suffered from acute onset of symptoms, presenting with headache and seizures. Brain computed tomography and magnetic resonance imaging showed large hemorrhagic lesions surrounded by massive edemas in the 4 patients. Brain biopsies were performed in the 4 patients due to concern for brain tumors. However, the pathological results supported the diagnosis of ICoVT, and the subsequent anticoagulant treatment administered was effective for all 4 patients. In conclusion, ICoVT can be easily misdiagnosed as brain tumor because of the atypical clinical and imaging features. The results suggested that the possibility of ICoVT always be considered in patients with neuroimaging showing cortical hemorrhagic lesions with massive edemas. [ABSTRACT FROM AUTHOR]

Muscle carnitine deficiency usually results in a lipid storage myopathy, but more rarely, neuropathy occurs in this condition. We report herein a 29-year-old man with muscle carnitine deficiency who developed not only a lipid storage myopathy, but also a severe sensory neuropathy. Oral therapy with levo-carnitine (3 g per day) for 3 months produced a remarkable improvement of the myopathy and sensory neuropathy. Six months later, he remained in good condition under strict dietary control. This report emphasizes that severe neuropathy may occur in some patients with muscle carnitine deficiency, and highlights the need for the neurologist’s familiarity with those afflicted to achieve optimal clinical management. We have shown that quetiapine, a new antipsychotic drug, protects cultured cells against oxidative stress-related cytotoxicities induced by amyloid β (Aβ)25-35, and that quetiapine prevents memory impairment and decreases Aβ plaques in the brains of amyloid precursor protein (APP)/presenilin-1 (PS-1) double-mutant mice. The aim of this study was to understand why quetiapine has these protective effects. Because the cytotoxicity of both Aβ(25-35) and Aβ(1-40) requires fibril formation, our first experiments determined the effect of quetiapine on Aβ(25-35) aggregation. Quetiapine inhibited Aβ(25-35) aggregation in cell-free aqueous solutions and blocked the fibrillar aggregation of Aβ(25-35), as observed under an electron microscope. We then investigated why quetiapine inhibits Aβ(25-35) aggregation. During the aggregation of Aβ(25-35), a hydroxyl radical (OH•) was released, which in turn amplified Aβ(25-35) aggregation. Quetiapine blocked OH•-induced Aβ(25-35) aggregation and scavenged the OH• produced in the Fenton system and in the Aβ(25-35) solution, as analyzed using electron paramagnetic resonance spectroscopy. Furthermore, new compounds formed by quetiapine and OH• were observed in MS analysis. Finally, we applied Aβ(25-35) to PC12 cells to observe the effect of quetiapine on living cells. Aβ(25-35) increased levels of intracellular reactive oxygen species and calcium in PC12 cells and caused cell death, but these toxic effects were prevented by quetiapine. These results demonstrate an anti-oxidative stress mechanism of quetiapine, which contributes to its protective effects observed in our previous studies and explains the effectiveness of this drug for Alzheimer’s disease patients with psychiatric and behavioral complications. The diurnal variation of atmospheric carbonyls and VOCs in a forest in south China were studied in summer 2004. Twenty kinds of carbonyls and eight kinds of VOCs were identified and quantified. Formaldehyde and acetaldehyde were the two most abundant carbonyls, while the most abundant VOCs were isoprene, followed by o-xylene. Most C3-C10 carbonyls had higher concentrations from 09:00 to 15:00, and their levels were lower during night-time and often reached the lowest in early morning. Formaldehyde and acetaldehyde, however, showed two high levels in their diurnal patterns partly due to their different sources and sinks. The VOCs had different diurnal patterns compared to most carbonyls. The highest concentrations were observed from 03:00 to 06:00 for 1-butene, from 06:00 to 12:00 for isoprene, and from 12:00 to 15:00 for α-pinene. The highest levels for aromatic hydrocarbons occurred during midnight and the lowest in late afternoon. According to the study, emissions from vegetation and photo-oxidation of gas-phase hydrocarbons were the main sources for some carbonyls and VOCs in this region. Other compounds, such as formaldehyde, acetaldehyde and BTEX, showed anthropogenic sources. [ABSTRACT FROM AUTHOR]
