Objective To investigate the clinical and electroencephalographic (EEG) features of children with eating-induced reflex epilepsy. Methods A retrospective analysis was performed for the clinical features, EEG findings, treatment, and prognosis of three children with eating-induced reflex epilepsy who attended Department of Pediatrics in our hospital from 2014 to 2024. Results There were two boys among the three children. Two children had normal intellectual development before the onset of the disease, while one was diagnosed with intellectual disability. All three children had normal cranial MRI results, and whole-exome genetic testing and copy number variation testing were performed for two children and their families, which yielded negative results. In case 1, the child had focal seizures and later developed into focal secondary bilateral tonic-clonic seizures, and EEG monitoring showed bilateral Rolandic discharges in the interictal period, but without eating-induced seizures. In case 2, the child had focal epileptic spasms during wakefulness and generalized myoclonic seizures during sleep, and EEG monitoring showed the coexistence of multifocal discharges and generalized discharges in the interictal period, drinking water induced three clusters of predominantly right-sided generalized multiphase slow waves with epileptic spasms, as well as several spontaneous generalized myo-clonic seizures during sleep. In case 3, the child had coexistence of focal epileptic spasms and generalized tonic seizures, and EEG monitoring showed the coexistence of multifocal discharges and generalized discharges, eating induced a cluster of predominantly left posterior multiphase slow waves with epileptic spasms. All three children received more than two anti-seizure medications (ASMs) and failed to achieve the control of seizures, and they presented with varying degrees of intellectual disability after disease onset. Conclusion Eating-induced reflex epilepsy is slightly more common in boys and is often accompanied by intellectual disability after disease onset. The coexistence of spontaneous seizures and induced seizures is often observed in this disease, with epileptic spasms being the more common type of eating-induced seizures. EEG manifestations in the interictal period include focal discharges, generalized discharges, or the coexistence of both types of discharges. ASMs have a poor therapeutic effect on this disease.

Centromere protein M (CENPM) is essential for chromosome separation during mitosis. However, its roles in lung adenocarcinoma (LUAD) progression and metastasis remain unknown. In this study, we aimed to explore the effects of CENPM on LUAD progression as well as the underlying mechanisms. We analyzed the expression of CENPM and its correlation with clinicopathological characteristics using GEO LUAD chip datasets and TCGA dataset. We further investigated the impact of CENPM on LUAD in vitro and in vivo. In silico analysis and qRT-PCR revealed that CENPM is upregulated in LUAD compared with that in normal lung tissues. Via gain/loss-of-function assays, we further found that CENPM promotes the LUAD cell cycle, cell proliferation, migration and invasion, and inhibits cell apoptosis in vitro. The in vivo study showed that loss of CENPM inhibits the growth of A549 xenografts. Furthermore, we found that CENPM can promote the phosphorylation of mTOR rather than directly affect the mTOR content. Inhibition of mTOR activity abrogates the promoting effects of CENPM on cell cycle progression, cell proliferation, migration and invasion. Taken together, these results show that CENPM plays an important role in the growth and metastasis of LUAD and may be a promising therapeutic target in LUAD.

Continuous cultivation of faba beans often results in a high occurrence of Fusarium wilt. Nevertheless, this issue can be successfully managed through wheat‐faba bean intercropping. Our objective is to elucidate the function of non‐host wheat in combating faba bean Fusarium wilt. We assessed the impact of wheat on the development of faba bean Fusarium wilt through field and pot experiments, and examined and identified the compounds in the root exudates of wheat. The influence of wheat root exudates on Fusarium commun mycelial growth and spore multiplication was investigated through indoor culture experiments. Wheat‐faba bean intercropping significantly reduced the occurrence of Fusarium wilt in field trials. Root separation experiments in pots indicated that wheat root exudates might play a vital role in this outcome. The exogenous addition of wheat root exudates to faba bean grown alone notably decreased the occurrence and severity of Fusarium wilt. Furthermore, the exogenous addition of wheat root exudates effectively hindered the growth and reproduction of Fusarium commun's mycelium. And, wheat root exudates contained six compounds with relatively high concentrations, including salicylic acid, p‐hydroxybenzoic acid, tartaric acid, malic acid, glutamic acid, and threonine, all of which inhibited the growth and spore formation of Fusarium commun. In a wheat‐faba bean intercropping system, these six compounds found in the root exudate of non‐host wheat can help control Fusarium wilt in faba bean by inhibiting the mycelial growth and sporulation of Fusarium commun. A spiral steel silo has become the preferred choice for building steel silos because of its short construction period, low cost, and high strength. Owing to the harsh outdoor welding conditions of the spiral steel silo and the high risk of manual welding, in this paper, an automatic welding robot is proposed, and its key technology is studied. The welding process parameters consisted of welding voltage, current, and the distance between the conductive nozzle and the base material, which were determined through experiments. The key technology of seam tracking on the roof of a spiral steel silo was studied. Finally, a weld bead tracking experiment and prototype welding experiment were carried out. The results suggest that the prototype can track the weld bead stably and accurately, and the welding effect can meet welding process requirements when the welding current is 200 A, the voltage is 20 V, and the distance between the welding nozzle and the base material is 12 mm. Background: More than 50 loci are associated with spinocerebellar ataxia (SCA), and the most frequent subtypes share nucleotide repeats expansion, especially CAG expansion. Objective: The objective of this study was to confirm a novel SCA subtype caused by CAG expansion. Methods: We performed long‐read whole‐genome sequencing combined with linkage analysis in a five‐generation Chinese family, and the finding was validated in another pedigree. The three‐dimensional structure and function of THAP11 mutant protein were predicted. Polyglutamine (polyQ) toxicity of THAP11 gene with CAG expansion was assessed in skin fibroblasts of patients, human embryonic kidney 293 and Neuro‐2a cells. Results: We identified THAP11 as the novel causative SCA gene with CAG repeats ranging from 45 to 100 in patients with ataxia and from 20 to 38 in healthy control subjects. Among the patients, the number of CAA interruptions within CAG repeats was decreased to 3 (up to 5–6 in controls), whereas the number of 3′ pure CAG repeats was up to 32 to 87 (4–16 in controls), suggesting that the toxicity of polyQ protein was length dependent on the pure CAG repeats. Intracellular aggregates were observed in cultured skin fibroblasts from patients. THAP11 polyQ protein was more intensely distributed in the cytoplasm of cultured skin fibroblasts from patients, which was replicated with in vitro cultured neuro‐2a transfected with 54 or 100 CAG repeats. Conclusions: This study identified a novel SCA subtype caused by intragenic CAG repeat expansion in THAP11 with intracellular aggregation of THAP11 polyQ protein. Our findings extended the spectrum of polyQ diseases and offered a new perspective in understanding polyQ‐mediated toxic aggregation. © 2023 The Authors. This systematic review and meta‐analysis aimed to evaluate the efficacy of vigabatrin (VGB) in treating infantile epileptic spasms syndrome (IESS). Databases of PubMed, Embase, Web of Science, MEDLINE, and Cochrane Library were systematically searched. All the relevant randomized controlled trials (RCTs) and observational studies (OSs) of VGB for IESS were included and analyzed separately. The primary outcome was the cessation of epileptic spasms (ES). Five RCTs and nine OSs compared the efficacy of VGB vs hormonal monotherapy for IESS. Meta‐analysis of the five RCTs showed that hormonal monotherapy was significantly better than VGB monotherapy (OR = 0.37, 95% CI = 0.20‐0.67) for patients with new‐onset IESS. Meta‐analysis of the nine OSs agrees with the result from RCTs (OR = 0.61, 95% CI = 0.43‐0.85). VGB was more effective in patients with TSC than in those with other etiologies (five OSs, OR = 5.59, 95% CI = 2.17‐14.41). There was no significant difference in the efficiency of VGB combined with hormonal therapy vs hormonal monotherapy for IESS (two RCTs, OR = 0.75, 95% CI = 0.09‐6.45). Hormonal monotherapy is better than VGB monotherapy for non‐TSC‐associated IESS. But for patients with IESS due to TSC, VGB is the first choice. The microstructure of 3D five-directional braided composites (5DBCs) comprises the following three constituents: yarn, matrix and interface. The interface is the bridge between yarn and the matrix, and it controls the mechanical properties of composites. In this paper, a meso-scale finite element (FE) model containing an interface is proposed to analyze the off-axial tensile behavior of 5DBCs. Hashin criteria and maximum stress criteria with a gradual degradation scheme are employed to capture the damage evolution behavior of yarn and the matrix; zero-thickness cohesive elements ruled by a bilinear traction–separation constitutive relation are applied to simulate the interface debonding. These constitutive models are coded into a user-defined material subroutine VUMAT and implemented to attain a numerical solution based on ABAQUS/Explicit solver. The meso-scale FE model is verified with available experimental data in the on-axial loading condition and then utilized to predict the mechanical response in diverse off-axial tensile cases. The damage evolution process of a 5DBC specimen under typical off-axial loading is provided, and the corresponding failure mechanism is analyzed in detail. Aims: Vagus nerve stimulation (VNS) is a neuromodulation therapy for children with drug‐resistant epilepsy (DRE). The efficacy of VNS is heterogeneous. A prediction model is needed to predict the efficacy before implantation. Methods: We collected data from children with DRE who underwent VNS implantation and received regular programming for at least 1 year. Preoperative clinical information and scalp video electroencephalography (EEG) were available in 88 children. Synchronization features, including phase lag index (PLI), weighted phase lag index (wPLI), and phase‐locking value (PLV), were compared between responders and non‐responders. We further adapted a support vector machine (SVM) classifier selected from 25 clinical and 18 synchronization features to build a prediction model for efficacy in a discovery cohort (n = 70) and was tested in an independent validation cohort (n = 18). Results: In the discovery cohort, the average interictal awake PLI in the high beta band was significantly higher in responders than non‐responders (p < 0.05). The SVM classifier generated from integrating both clinical and synchronization features had the best prediction efficacy, demonstrating an accuracy of 75.7%, precision of 80.8% and area under the receiver operating characteristic (AUC) of 0.766 on 10‐fold cross‐validation. In the validation cohort, the prediction model demonstrated an accuracy of 61.1%. Conclusion: This study established the first prediction model integrating clinical and baseline synchronization features for preoperative VNS responder screening among children with DRE. With further optimization of the model, we hope to provide an effective and convenient method for identifying responders before VNS implantation. [ABSTRACT FROM AUTHOR]

Objective: This study aimed to delineate the detailed characteristics of startle-induced epileptic spasms (ES) and explore the brain regions where startle-induced ES originated. Methods: Among 581 patients with ES registered in our database, 30 were diagnosed with startle-induced ES according to video-electroencephalogram (EEG) and seizure semiology and were included in this study. Patients' clinical characteristics and ictal high-frequency oscillations (HFOs) were analyzed. Results: Mean age at the onset of startle-induced ES was 28.1 months. Half of the patients had structural etiology, two of whom were diagnosed with co-existing structural and genetic etiologies. The focal neuroimaging abnormalities were predominant in the frontal cortex (9/15, 60.0%). Fifteen patients (50%) had prominent interictal epileptiform discharges in the frontal and anterior temporal. Ictal HFOs counts of the startle-induced ES in the anterior region were significantly higher than those in the posterior regions (p < 0.05). Five patients (16.7%) became seizure-free ≥6 months, and ten (33.3%) showed startle-induced ES cessation ≥6 months. All patients except one had mild to severe psychomotor developmental delay after the onset of seizures. Conclusion: Patients with startle-induced ES typically had brain lesions and showed drug-resistant. The neuroimaging and EEG findings, including ictal HFOs, support that startle-induced ES often originates from the frontal cortex. [ABSTRACT FROM AUTHOR]
