An earthquake and tsunami on 1 January 2024, caused extensive damage across Ishikawa Prefecture, Japan. This study systematically examined the recovery process. It aimed to analyze the initial stages of recovery and highlight the lessons learned from these efforts. By collecting data from Ishikawa's post-disaster records, including government reports, reconstruction plans, local news, and observational records, we compiled the first "Early Reconstruction Timeline for Ishikawa". This timeline divided the recovery process into four phases: disaster occurrence and emergency response (January), reconstruction preparation (February–April), the beginning of early reconstruction (May–September), and the beginning of mid-reconstruction to the present (October–Present). Analysis of this timeline revealed several significant findings. First, Ishikawa's reconstruction efforts were structured into three key phases: 'Post-Disaster Relief and Emergency Response', 'Post-Disaster Reconstruction Preparation Period', and 'Infrastructure Reconstruction and Emergency Housing Construction Period'. This phased approach highlights an efficient and organized recovery process, distinguishing Ishikawa from other disaster-affected regions. Additionally, the housing reconstruction model showcased an innovative balance between emergency housing as a public resource and the specific needs of displaced residents. These findings not only establish a timeline-based framework for Ishikawa's reconstruction but also provide practical insights for guiding early post-disaster recovery efforts in various disaster-affected contexts worldwide. Since the 2011 Great East Japan Earthquake, 13 years have passed, and many areas have completed post-disaster reconstruction. Therefore, it is necessary to further summarise and analyse the experiences gained from this post-disaster reconstruction. This study conducted door-to-door interviews with residents of Ōtsuchi-chō, Iwate Prefecture, in the 13 years following the earthquake. It recorded the residents' perspectives on post-disaster reconstruction and its shortcomings. By collecting locally published materials like newspapers, and local government records and integrating them with interview records, the first "Great East Japan Earthquake Timeline" for Ōtsuchi-chō was compiled. We primarily divided this timeline into the following three phases: the disaster and early reconstruction period (2011), the mid-reconstruction period (2012–2015), and the late reconstruction period (2016–Present). This timeline has yielded the following results: firstly, some issues have arisen during this prolonged reconstruction process. For example, continuous relocation hinders community rebuilding, especially in the context of an ageing society. Secondly, the "Ōtsuchi-chō Reconstruction Timeline" was created, which can be used for post-disaster reconstruction in contemporary Japanese villages facing severe ageing. This timeline provides a clear understanding of how disaster-affected areas can rebuild housing and infrastructure, and recover economically. It offers guidance for the reconstruction of Japanese villages after a disaster. Therefore, post-disaster reconstruction in the region should accelerate infrastructure development and continuously listen to residents' voices, providing corresponding services to make reconstruction comprehensive and specific. Sit-to-stand (STS) transfer is an easy everyday activity for healthy people, but it is a challenging task for the elderly, and patients with lower-limb weaknesses. This article presents an intelligent assistive walker, which assists the user to accomplish safe STS transfers. An STS intention recognition method is proposed based on an optimized adaptive network-based fuzzy inference system (ANFIS), where the walker is able to accurately detect a voluntary STS and to apply an appropriate control accordingly. The walker is controlled by using a proxy-based controller with respect to the detected human intention, that is measured using thin-film force sensing resistors. During the STS transfer, the assistance force is generated on-line using a virtual spring-damper between the proxy and the linear actuator of the assistive walker. The stability of the proposed controller is theoretically analyzed, and verified by simulations. Experiments with five young healthy subjects and one elderly subject demonstrated that the proposed assistive walker is able to efficiently follow the user’s intention to complete stable and compliant STS transfers. Titanium dioxide nanoparticles (TiO2 NPs) are authorized food additives, and children have the highest exposure. Therefore, children are likely more susceptible to the adverse effects of TiO2 NPs than adults. Previous study showed that oral administration of 50 mg/kg body weight (bw) TiO2 NPs increase plasma glucose in mice. However, few studies have directly compared the adverse effects of exposure to TiO2 NPs on plasma glucose metabolism of different age groups. In this study, the developing (age 3 weeks) and adult mice (age 10 weeks) were orally administered with 50 mg/kg bw TiO2 NPs per day. The TiO2 NPs induced hyperglycemia earlier in the developing mice than in the adult mice. Then mechanisms were analyzed after mice were oral administration of TiO2 NPs for 8 weeks and 26 weeks, respectively. Results showed that the treatment with TiO2 NPs activated xenobiotic biodegradation in livers of both developing and adult mice at the early stage. However, only in the developing mice, TiO2 NPs induced endoplasmic reticulum (ER) stress in livers and increased reactive oxygen species in livers and sera in the early stage. The ER stress and ROS activated an inflammation response and mitogen‐activated protein kinase pathways, thereby inducing insulin resistance in the livers of developing mice at the early stage. The response of the adult mice was delayed, and these changes were observed in the late stage of the study. Zinc oxide nanoparticles (ZnO NPs) represent an important class of commercially applied materials. Recently, adverse effects of ZnO NPs were found in humans and animals following ingestion, although the effects on endocrine system disease remain unclear. In this study, ZnO NPs were orally administered to mice, and at doses of 25 mg/kg bw (body weight) ZnO NPs and above, plasma glucose increased significantly. The genome-wide effects of ZnO NPs were then investigated using RNA-sequencing technology. In the cluster analysis, the most significantly enriched Gene Ontology (GO) terms and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways concerned membranes and their close association with endoplasmic reticulum (ER) stress and reactive oxygen species (ROS) generation. Biochemical and gene and protein expression analyses revealed that ZnO NPs activated a xenobiotic biodegradation response and increased the expression of cytochrome P450 (CYP) enzymes in mice livers, leading to ER stress. The ER stress increased ROS generation. The high levels of ROS activated the MAPK and NF-κB pathways and induced an inflammation response, resulting in the phosphorylation of insulin receptor substrate 1. Thus, the insulin resistance that developed was the primary mechanism for the increase in the plasma glucose of mice treated orally with ZnO NPs. [ABSTRACT FROM AUTHOR]

Titanium dioxide nanoparticles (TiO2 NPs) are reported to increase plasma glucose levels in mice at specific doses. The production and accumulation of reactive oxygen species (ROS) is potentially the most important factor underlying the biological toxicity of TiO2 NPs but the underlying mechanisms are unclear at present. Data from genome‐wide analyses showed that TiO2 NPs induce endoplasmic reticulum (ER) stress and ROS generation, leading to the inference that TiO2 NP‐induced ER stress contributes to enhancement of ROS in mice. Resveratrol (Res) effectively relieved TiO2 NP‐induced ER stress and ROS generation by ameliorating expression of a common set of activated genes for both processes, signifying that ER stress and ROS are closely related. TiO2 NP‐induced ER stress occurred earlier than ROS generation. Upon treatment with 4‐phenylbutyric acid to relieve ER stress, plasma glucose levels tended toward normal and TiO2 NP increased ROS production was inhibited. These results suggest that TiO2 NP‐induced ER stress promotes the generation of ROS, in turn, triggering increased plasma glucose levels in mice. In addition, Res that displays the ability to reduce ER stress presents a dietary polyphenol antioxidant that can effectively prevent the toxicological effects of TiO2 NPs on plasma glucose metabolism. Inorganic mercury, though a key component of pediatric vaccines, is an environmental toxicant threatening human health via accumulating oxidative stress in part. Luteolin has been of great interest because of its antiinflammatory, anticarcinogenic and antioxidative effects. Here we hypothesized that luteolin would attenuate hepatotoxicity induced by acute inorganic mercury exposure. Kunming mice were treated with luteolin (100 mg/kg) 24 h after administration of 4 mg/kg mercuric chloride (HgCl2). The results showed that luteolin ameliorated HgCl2 induced anemia and hepatotoxicity, regulating radical oxygen species (ROS) production and hepatocyte viability in vitro and oxidative stress and apoptosis in vivo. Furthermore, luteolin reversed the changes in levels of inflammation- and apoptosis-related proteins involving NF-κB, TNF-α, Sirt1, mTOR, Bax, p53, and Bcl-2, and inhibited p38 MAPK activation. Luteolin enhanced antioxidant defense system based on Keap1, Nrf2, HO-1, NQO1, and KLF9. Moreover, luteolin did not affect miRNA-146a expression. Collectively, our findings, for the first time, elucidate a precise mechanism for attenuation of HgCl2-induced liver dysfunction by dietary luteolin via regulating Sirt1/Nrf2/TNF-α signaling pathway, and provide a foundation for further study of luteolin as a novel therapeutic agent against inorganic mercury poisoning. Elevated plasma free fatty acid (FFA) levels are associated with insulin resistance and can cause lipotoxicity in skeletal muscles. In response to FFAs, skeletal muscle can secrete a variety of cytokines. Irisin, one such muscle-secreted cytokine, can improve glucose tolerance, glucose uptake, and lipid metabolism. It is produced by the transmembrane protein fibronectin type Ⅲ domain containing 5 (FNDC5) by specific proteases. The purpose of this study was to investigate the regulatory mechanisms of the FNDC5 response to palmitate and their relationships with insulin resistance in C2C12 myotubes. RNA sequencing analysis results from C2C12 myotubes treated with palmitate showed that palmitate could activate the TGF-β signaling pathway. Palmitate directly affected the expression of Smad3, but not its phosphorylation level, in C2C12 myotubes. Furthermore, knockdown and knockout of Smad3 alleviated the inhibitory effect of palmitate on the expression of FNDC5. In contrast, overexpression of Smad3 aggravated the inhibition of FNDC5 expression. There is a Smad3 binding motif in the −660 bp to −649 bp region of the Fndc5 promoter. CRISPR/Cas9 knockout of this region also alleviated the inhibition of FNDC5 expression in response to palmitate. More importantly, inhibition of FNDC5 expression mediated by Smad3 led to a decrease in insulin sensitivity in C2C12 myotubes. Collectively, these findings suggest that palmitate could induce insulin resistance through Smad3-mediated down-regulation of the Fndc5 gene. • Palmitate had an inhibitory effect on the expression of Fndc5. • Smad3 had a certain enrichment in the −756 to −634 bp region of the Fndc5 promoter. • Palmitate might attenuate the insulin signaling pathway via Smad3-mediated down-regulation of Fndc5.
