Abstract Gastric cancer (GC) is globally recognized as the fifth most common cancer and the third leading cause of cancer-related mortality. Early metastasis in GC significantly contributes to its high mortality and unfavorable prognosis. However, the underlying mechanisms of this phenomenon remain largely unexplored. Among the various factors involved, AKR1C3 has emerged as a crucial component in the pathways of tumorigenesis and metastasis across multiple cancer types. Yet, the precise significance of AKR1C3 in GC patients’ prognosis and its role in GC progression remain elusive. This study illuminated the significant downregulation of AKR1C3 in GC tissues, linking it to an aggressive phenotype and poor prognosis. Interestingly, while AKR1C3 overexpression did not affect the proliferation of GC cells, it significantly inhibited their ability to invade and metastasize. The underlying mechanism appears to involve AKR1C3’s inhibition of the p-JNK pathway, which leads to reduced phosphorylation of IKKα/β and IKBα, lowering p-NF-κB levels and hindering its movement into the nucleus, thereby stifling the epithelial-mesenchymal transition (EMT) process in GC cells. These insights reveal AKR1C3’s tumor-suppressive effects in GC and suggest its potential as a diagnostic and prognostic biomarker, offering new avenues for targeted therapies in gastric cancer management.

According to the existing personnel structure, quantity, development strategy, and market demand of the Shuohuang Railway Company’s operation and maintenance project, the demand quantity of various employees of the company for the past three years is predicted, and a human resource optimization model based on existing human resources and future plans is established. Then, the optimal solutions of the two mathematical models were calculated and analyzed using LINGO software. Finally, combined with the actual situation, the optimal allocation of human resources for the operation and maintenance project of KY company was obtained. The following conclusions are obtained. (1) For the optimal allocation model of existing human resources, the maximum net profit of the optimal staffing model is CNY 3258000. (2) The human resources allocation cost of the minimum dismissal model is CNY 81000. (3) The human resources allocation cost of the lowest cost model is CNY 15500. The research results can effectively guide the human resource management of the operation and maintenance project of the Shuohuang Railway Company, and have important theoretical and practical significance for further analysis of human resources model and its optimal allocation method.

The combined finite-discrete element method (FDEM) and laboratory test were selected to study the extent of deformation range and time-dependent deformation of surrounding rock during tunnel excavation without support in a fault-crossing tunnel project. FDEM was found to accurately reflect the deformation and failure characteristics of different surrounding rocks during stress release, including conjugate shear and extrusion. Analysis of the results showed that the disturbance range of surrounding rocks could reach 1.5 to 2.5 times the tunnel diameter when crossing the fault zone. The rock surrounding the tunnel was found to incur significant conjugate shear deformation and extrusion deformation: conjugate shear deformation was identified as dominant in the deep rock mass, whereas extrusion deformation prevailed in the rock mass near the cave wall. The conjugate shear distribution was represented as a spiral line of deformation circling around the tunnel section, with an elliptical main deformation zone with its long axis parallel to the fault plane. Compared with the findings when crossing the intact rock mass, the deformation of the surrounding rocks when crossing the fault zone was characterized by rapid development, deep expansion area, and large deformation. The study conclusions were that supporting bolts and steel arches should be implemented timely when excavating fault zones and that both lithology and optimal construction timing were essential considerations in determining the length of the supporting bolts.
