Abstract Hydrogen-bonded organic frameworks (HOFs) are versatile materials with potential applications in proton conduction. Traditional approaches involve incorporating humidity control to address grain boundary challenges for proton conduction. This study finds vitrification as an alternative strategy to eliminate grain boundary effect in HOFs by rapidly melt quenching the kinetically stable HOF-SXU-8 to glassy state HOF-g. Notably, a remarkable enhancement in proton conductivity without humidity was achieved after vitrification, from 1.31 × 10−7 S cm−1 to 5.62× 10−2 S cm−1 at 100 °C. Long term stability test showed negligible performance degradation, and even at 30 °C, the proton conductivity remained at high level of 1.2 × 10−2 S cm−1. Molecule dynamics (MD) simulations and X-ray total scattering experiments reveal the HOF-g system is consisted of three kinds of clusters, i.e., 1,5-Naphthalenedisulfonic acid (1,5-NSA) anion clusters, N,N-dimethylformamide (DMF) molecule clusters, and H+-H2O clusters. In which, the H+ plays an important role to bridge these clusters and the high conductivity is mainly related to the H+ on H3O+. These findings provide valuable insights for optimizing HOFs, enabling efficient proton conduction, and advancing energy conversion and storage devices.

Starch is the most important component in the endosperm, and its synthesis is regulated by multiple transcription factors (TFs) in cereals. However, whether the functions of these TFs are conserved among cereals remains unclear. In this study, we cloned a B3 family TF in wheat, named TaABI19, based on its orthologous sequence in maize (Zea mays L.). Alignment of the DNA and protein sequences showed that ABI19 is conserved in maize and wheat (Triticum aestivum L.). We found that TaABI19 is highly expressed in young spikes and developing grains, and encodes a nucleus-localized transcriptional activator in wheat. The taabi19-b1 null mutants obtained by EMS exhibited a down-regulation of starch synthesis, shorter grain length and lower thousand-grain weight (TGW). Furthermore, we proved that TaABI19 could bind to the promoters of TaPBF homologous genes and enhance their expression. Haplotype association showed that TaABI19-B1 is significantly associated with TGW. We found that Hap2 and Hap3 were favored and had undergone positive selection in China’s wheat breeding programs. Less than 50% of the modern cultivars convey the favored haplotypes, indicating that TaABI19 still can be considered as a target locus for marker-assisted selection breeding to increase TGW in China.

Abstract Splicing factor 3B subunit 4 (SF3B4) plays important functional roles not only in pre-mRNA splicing, but also in the regulation of transcription, translation, and cell signaling, and its dysregulation contributes to various diseases including Nager syndrome and tumorigenesis. However, the role of SF3B4 and underlying mechanisms in clear cell renal cell carcinoma (ccRCC) remain obscure. In the present study, we found that the expression of SF3B4 was significantly elevated in ccRCC tissues and negatively correlated with the overall survival of ccRCC patients. Upregulation of SF3B4 promotes migration and invasion of ccRCC cells in vitro and in vivo. The promoting effect of SF3B4 on cell migration and invasion is mediated by Twist1, a key transcription factor to mediate EMT. Interestingly, SF3B4, a component of the pre-mRNA spliceosome, is able to promote KLF16 expression by facilitating the transport of KLF16 mRNA into the cytoplasm. Mechanistically, SF3B4 promotes the export of KLF16 mRNA from the nucleus to the cytoplasm and thus enhances KLF16 expression, and in turn elevated KLF16 directly binds to the Twist1 promoter to activate its transcription, leading to EMT and ccRCC progression. Our findings provide evidence that the SF3B4-KLF16-Twist1 axis plays important functional roles in the development and progression of ccRCC, and manipulating this pathway may be a novel therapeutic target for the treatment of ccRCC.

Abstract Background Both E2F transcription factor and cyclin-dependent kinases (CDKs), which increase or decrease E2F activity by phosphorylating E2F or its partner, are involved in the control of cell proliferation, and some circRNAs and miRNAs regulate the expression of E2F and CDKs. However, little is known about whether dysregulation among E2Fs, CDKs, circRNAs and miRNAs occurs in human PCa. Methods The expression levels of CDK13 in PCa tissues and different cell lines were determined by quantitative real-time PCR and Western blot analysis. In vitro and in vivo assays were preformed to explore the biological effects of CDK13 in PCa cells. Co-immunoprecipitation anlysis coupled with mass spectrometry was used to identify E2F5 interaction with CDK13. A CRISPR-Cas9 complex was used to activate endogenous CDK13 and circCDK13 expression. Furthermore, the mechanism of circCDK13 was investigated by using loss-of-function and gain-of-function assays in vitro and in vivo. Results Here we show that CDK13 is significantly upregulated in human PCa tissues. CDK13 depletion and overexpression in PCa cells decrease and increase, respectively, cell proliferation, and the pro-proliferation effect of CDK13 is strengthened by its interaction with E2F5. Mechanistically, transcriptional activation of endogenous CDK13, but not the forced expression of CDK13 by its expression vector, remarkably promotes E2F5 protein expression by facilitating circCDK13 formation. Further, the upregulation of E2F5 enhances CDK13 transcription and promotes circCDK13 biogenesis, which in turn sponges miR-212-5p/449a and thus relieves their repression of the E2F5 expression, subsequently leading to the upregulation of E2F5 expression and PCa cell proliferation. Conclusions These findings suggest that CDK13 upregulation-induced formation of the positive feedback loop among circCDK13, miR-212-5p/miR-449a and E2F5 is responsible for PCa development. Targeting this newly identified regulatory axis may provide therapeutic benefit against PCa progression and drug resistance.

The climatic variation of offshore wind energy has a close relationship with the long-term plan of energy utilization. However, the work on this aspect is scarce and mainly focuses on the variation of wind power density (WPD). There is little research on the climatic trends of effective wind speed occurrence (EWSO) and occurrence of energy level greater than 200 W/m2 (rich level occurrence, RLO), which are directly related to the available rate and richness of wind energy. Based on the ERA-Interim wind product from the ECMWF, this study calculated the climatic trends of series of key factors of wind energy in the global oceans, including the WPD, EWSO, and RLO. The results show that the wind energy exhibits a positive trend globally for the past 36 years, with overall annual increasing trends in WPD, EWSO, and RLO, of 0.698 (W/m2)/yr, 0.076%/yr, and 0.090%/yr separately. The annual trend exhibits evident regional differences. The areas with significant increasing trends are mainly distributed in the mid- low-latitude waters of global oceans and part of the southern hemisphere westerlies. The annual increasing trend of WPD is strongest in the southern westerlies, especially in the extratropical South Pacific (ETSP), of about 1.64 (W/m2)/yr. The annual increasing trends of EWSO and RLO are strongest in the tropical waters, especially the tropical Pacific Ocean (TPO), of 0.17%/yr and 0.19%/yr separately. The annual and seasonal WPD, EWSO, and RLO in most global oceans have significant increasing trends or no significant variation, meaning that the wind energy trends are rich or stable, which is beneficial for energy development. The climatic trends of wind energy are dominated by different time periods. There is no evident abrupt change of wind energy in the extratropical waters globally and tropical Atlantic Ocean (TAO). The abrupt periods of wind energy in the TIO and TPO occurred in the end of the 20th century and the beginning of the 21st century. The wind energy of the South China Sea, Arabian Sea, and the Bay of Bengal and nino3 index share a common period of approximately 5 years. The offshore wind energy was controlled by an oscillating phenomenon.

AIM: To investigate the diffusion changes in both the optic nerve and optic tract in orbital space-occupying lesion patients with decreased visual acuity, and its clinical significance using probabilistic diffusion tractography (PDT). METHODS: Twenty patients with orbital space-occupying lesions and 25 age- and gender-matched healthy persons were included. All patients and controls underwent routine orbital magnetic resonance imaging and diffusion tensor imaging (DTI), using a 3.0T magnetic resonance scanner (Trio Tim Siemens). After the image data were preprocessed, each DTI parameters of the optic nerve and optic tract was obtained by PDT, including fractional anisotropy (FA), mean diffusivity (MD), axial diffusivity (AD) and radial diffusivity (RD). The asymmetry index (AI) of each parameter was calculated. Compared the parameters of the affected side optic nerve and ipsilateral optic tract with the contralateral side by paired sample t-test; compared AI of parameters of optic nerve and optic tract between the patient group and the control group by independent sample t-test. Patients were divided into three subgroups according to the low vision grade standard of WHO, compared the FA and AI of FA between the three subgroups by single factor variance analysis. RESULTS: The affected side optic nerve presented significantly decreased FA, increased MD, AD, and RD values compared to the unaffected side (P0.05). In recent years, some laboratories have been able to prepare calcium hydrobromide (CaHBr) by melting hydride and anhydrous bromide or metal and bromide in a hydrogen atmosphere at 900°C and have studied some of its properties. But there are few theoretical studies, especially the theoretical studies of monolayer CaHBr. We use the first-principles method to calculate the structure, elastic properties, and lattice thermal conductivity of the monolayer CaHBr based on the Boltzmann transport equation. We obtain a stable crystal structure by the optimization of monolayer CaHBr. By calculating the elastic constant of monolayer CaHBr, its mechanical stability is proved, and the elastic limit of monolayer CaHBr is obtained by biaxial tensile strain on monolayer CaHBr. And the corresponding phonon spectra show no imaginary frequency, indicating the dynamic stability of the monolayer CaHBr. By the ShengBTE code, we calculate the lattice thermal conductivity of the monolayer CaHBr, the iterative solution of BTE and RTA at 300 K–1200 K is obtained, and the lattice thermal conductivity at room temperature is κιBTE=2.469 W/m⋅K and κιRTA=2.201 W/m⋅K, respectively. It can be seen that the lattice thermal conductivity of monolayer CaHBr is low. And by analyzing the phonon spectrum, the scattering rate, and the mean free path of the phonons, the lattice thermal conductivity of monolayer CaHBr mainly depends on the acoustic modes. We hope this study can provide theoretical guidance for the experiments and practical application of monolayer CaHBr.

Abstract Background Docetaxel-based chemotherapy failure in advanced prostate carcinoma has partly been attributed to the resistance of prostate cancer (PC) cells to docetaxel-induced apoptosis. Hence, there is an urgent need to identify mechanisms of docetaxel chemoresistance and to develop new combination therapies. Methods miR-193a-5p level was evaluated by qPCR in prostate tissues and cell lines, and its expression in the tissues was also examined by in situ hybridization. PC cell line (PC3 cell) was transfected with miR-193a-5p mimic or its inhibitor, and then cell apoptosis and the expression of its downstream genes Bach2 and HO-1 were detected by TUNEL staining and Western blotting. Luciferase reporter assay was used to detect the effect of miR-193a-5p and Bach2 on HO-1 expression. Xenograft animal model was used to test the effect of miR-193a-5p and docetaxel on PC3 xenograft growth. Results miR-193a-5p was upregulated in PC tissues and PC cell lines, with significant suppression of PC3 cell apoptosis induced by oxidative stress. Mechanistically, miR-193a-5p suppressed the expression of Bach2, a repressor of the HO-1 gene, by directly targeting the Bach2 mRNA 3′-UTR. Docetaxel treatment modestly decreased Bach2 expression and increased HO-1 level in PC3 cells, whereas a modest increase of HO-1 facilitated docetaxel-induced apoptosis. Notably, docetaxel-induced miR-193a-5p upregulation, which in turn inhibits Bach2 expression and thus relieves Bach2 repression of HO-1 expression, partly counteracted docetaxel-induced apoptosis, as evidenced by the increased Bcl-2 and decreased Bax expression. Accordingly, silencing of miR-193a-5p enhanced sensitization of PC3 cells to docetaxel-induced apoptosis. Finally, depletion of miR-193a-5p significantly reduced PC xenograft growth in vivo. Conclusions Silencing of miR-193a-5p or blockade of the miR-193a-5p-Bach2-HO-1 pathway may be a novel therapeutic approach for castration-resistant PC.

High-rise buildings are very sensitive to wind excitations, and wind-induced responses have always been the key factors for structural design. Facade openings have often been used as aerodynamic measures for wind-resistant design of high-rise buildings to meet the requirement of structural safety and comfort. Obvious wind speed amplifications can also be observed inside the openings. Therefore, implementing wind turbines in the openings is of great importance for the utilization of abundant wind energy resources in high-rise buildings and the development of green buildings. Based on numerical simulation and wind tunnel testing, the wind loads and wind speed amplifications on high-rise buildings with openings are investigated in detail. The three-dimensional numerical simulation for wind effects on high-rise building with openings was firstly carried out on FLUENT 15.0 platform by SST k−ε model. The mean wind pressure coefficients and the wind flow characteristics were obtained. The wind speed amplifications at the opening were analyzed, and the distribution law of wind speed in the openings is presented. Meanwhile, a series of wind tunnel tests were conducted to assess the mean and fluctuating wind pressure coefficients in high-rise building models with various opening rates. The variation of wind pressure distribution at typical measuring layers with wind direction was analyzed. Finally, the wind speed amplifications in the openings were studied and verified by the numerical simulation results.

Fast and accurate determination of effective bentonite content in used clay bonded sand is very important for selecting the correct mixing ratio and mixing process to obtain high-performance molding sand. Currently, the effective bentonite content is determined by testing the ethylene blue absorbed in used clay bonded sand, which is usually a manual operation with some disadvantages including complicated process, long testing time and low accuracy. A rapid automatic analyzer of the effective bentonite content in used clay bonded sand was developed based on image recognition technology. The instrument consists of auto stirring, auto liquid removal, auto titration, step-rotation and image acquisition components, and processor. The principle of the image recognition method is first to decompose the color images into three-channel gray images based on the photosensitive degree difference of the light blue and dark blue in the three channels of red, green and blue, then to make the gray values subtraction calculation and gray level transformation of the gray images, and finally, to extract the outer circle light blue halo and the inner circle blue spot and calculate their area ratio. The titration process can be judged to reach the end-point while the area ratio is higher than the setting value.

Air pollution has been classified as Group 1 carcinogenic to humans, but the underlying tumorigenesis remains unclear. In Xuanwei City of Yunnan Province, the lung cancer incidence is among the highest in China attributed to severe air pollution generated by combustion of smoky coal, providing a unique opportunity to dissect lung carcinogenesis of air pollution. Here we analyzed the somatic mutations of 164 non-small cell lung cancers (NSCLCs) from Xuanwei and control regions (CR) where smoky coal was not used. Whole genome sequencing revealed a mean of 289 somatic exonic mutations per tumor and the frequent C:G → A:T nucleotide substitutions in Xuanwei NSCLCs. Worldwide, cancer is a serious threat to the health of citizens of every country, with the incidence and mortality increasing year by year. Cisplatin is the first-line anticancer drug commonly used in clinics and is widely used for the treatment of solid tumors including lung, gastric, liver, bladder, and ovarian cancer. Although cisplatin-based chemotherapy has a high clinical response efficacy, patients will inevitably develop drug resistance after repeated use, leading to severe restrictions of its application. Circular RNAs (circRNAs) are a promising class of non-coding RNAs capable of promoting or suppressing cancer via functioning as miRNAs sponges. Recently, an increasing amount of evidence shows that circRNAs are closely related to the cisplatin resistance of cancers. Therefore, standing at the perspective of the cisplatin chemotherapy resistance, this paper reviews the research progress of circRNAs related to cisplatin resistance of various cancers. Selenium (Se) is an essential micronutrient for many organisms. Despite its importance, our current knowledge of the biogeochemical cycling of dissolved Se in tropical estuaries is limited, especially in Southeast Asia. To gain insights into Se cycling in tropical peat-draining rivers and estuaries, samples were collected from the Rajang, Maludam, Sebuyau, Simunjan, Sematan, Samunsam, and Lunda rivers and estuaries in western Sarawak, Malaysia, in March and September 2017 and analysed for various forms of Se (dissolved inorganic and organic). Mean total dissolved Se (TDSe), dissolved inorganic Se (DISe), and dissolved organic Se concentrations (DOSe) were 2.2 nmol L-1 (range: 0.7 to 5.7 nmol L-1), 0.18 nmol L-1 (range: less than the detection limit to 0.47 nmol L-1), and 2.0 nmol L-1 (range: 0.42 to 5.7 nmol L-1), respectively. In acidic, low-oxygen, organic-rich blackwater (peatland-draining) rivers, the concentrations of DISe were extremely low, whereas those of DOSe were high. In rivers and estuaries that drained peatland, DOSe / TDSe ratios ranged from 0.67 to 0.99, showing that DOSe dominated. The positive relationship between DISe and salinity and the negative relationship between DOSe and salinity indicate marine and terrestrial origins of DISe and DOSe, respectively. The positive correlations of DOSe with the humification index and humic-like chromophoric dissolved organic matter components in freshwater river reaches suggest that peat soils are probably the main source of DOSe. Discharges of water enriched with DOSe fractions associated with peatland-derived high-molecular-weight, high-aromaticity dissolved organic matter discharged from estuaries may promote productivity in the adjoining oligotrophic coastal waters. Background: The need for empathy and the difficulties of coping with mortality when caring for the dying and the bereaved can cause psychological, emotional, and spiritual strain. Objective: The aim of this study was to examine the effectiveness of art-therapy-based supervision in reducing burnout and death anxiety among end-of-life care workers in Hong Kong. Methods: Through a quasi-experimental des ign, 69 par t icipant s enrol led in a 6-week, 18-hour art-therapy-based supervision group, and another 63 enrolled in a 3-day, 18-hour standard skills-based supervision group (n=132). Preand post-intervention assessments were carried out with three outcome measures: the Maslach Burnout Inventory—General Survey, the Five Facet Mindfulness Questionnaire, and the Death Attitude Profile—Revised. The data was analysed using paired sample t-tests. Results: Significant reductions in exhaustion and death anxiety and significant increases in emotional awareness were observed for participants in the art-therapy-based supervision group. Conclusion: This study provides preliminary evidence that art-therapy-based supervision for end-of-life care workers can reduce burnout by enhancing emotional awareness and regulation, fostering meaning-making, and promoting reflection on death. A quiz concerning annular erythematous scaly lesions on the face of a 22-year old Chinese woman is presented.

The article presents a case study of a 68-year-old Chinese male who developed an expanding, painful, necrotic skin ulcer on his lower abdomen wall. The patient had undergone a skin biopsy from the ulcer border and was diagnosed with ost-surgical pyoderma gangrenosum (PSPG). He was treated with methylprednisolone and was completely healed.

Exploring the potential functions of nonconserved residues on the outer side of a-helices and systematically optimizing them are pivotal for their application in protein engineering. Based on the evolutionary structural conservation analysis of GH5_5 cellulases, a practical molecular improvement strategy was developed. Highly variable sites on the outer side of the a-helices of the GH5_5 cellulase from Aspergillus niger (AnCel5A) were screened, and 14 out of the 34 highly variable sites were confirmed to exert a positive effect on the activity. After the modular combination of the positive mutations, the catalytic efficiency of the mutants was further improved. By using CMC-Na as the substrate, the catalytic efficiency and specific activity of variant AnCel5A_N193A/T300P/D307P were approximately 2.0-fold that of AnCel5A (227 6 21 versus 451 6 43 ml/s/mg and 1,726 6 19 versus 3,472 6 42 U/mg, respectively). The half-life (t1/2) of variant AnCel5A_N193A/T300P/D307P at 75°C was 2.36 times that of AnCel5A. The role of these sites was successfully validated in other GH5_5 cellulases. Computational analyses revealed that the flexibility of the loop 6- loop 7-loop 8 region was responsible for the increased catalytic performance. This work not only illustrated the important role of rapidly evolving positions on the outer side of the a-helices of GH5_5 cellulases but also revealed new insights into engineering the proteins that nature left as clues for us to find.
