This study aims to investigate the primary carbides precipitation in H13 steel solidified at relatively high cooling rates, ranging from 300 to 6,000 °C·min-1, based on in situ observations with a high temperature confocal laser scanning microscope. In the cooling rate range investigated, the solidification microstructure becomes more refined as cooling rate increases and the relationship between the secondary dendrite arm spacing (SDAS), λ2, and cooling rate,.T, can be expressed as λ2=128.45.T -0.124. Regardless of cooling rates, two kinds of primary carbides, i.e., the Mo-Cr-rich and V-rich carbides, are precipitated along the interdendritic region and most of them are the Mo-Cr-rich carbides. The morphology of Mo-Cr-rich carbide is not obviously influenced by the cooling rate, but that of V-rich carbide is obviously affected. The increasing cooling rate markedly refines the primary carbides and reduces their volume fractions, but their precipitations cannot be inhibited even when the cooling rate is increased to 6,000 °C·min-1. Besides, the segregation ratios (SRs) of the carbides forming elements are not obviously affected by the cooling rate. However, compared with the conventionally cast ingot, the SDAS and primary carbides in the steel solidified at the investigated cooling rates are much finer, morphologies of the carbides have changed significantly, and SRs of the carbides forming elements are markedly greater. The variation of primary carbide characteristics with cooling rate is mainly due to the change in SDAS.

This study optimized the ultrasound image of carotid artery stenosis using Taguchi dynamic analysis and an indigenous water phantom. Eighteen combinations of seven essential factors of the ultrasound scan facility were organized according to Taguchi’s L18 orthogonal array. The seven factors were assigned as follows: (1) angle of probe; (2) signal gain; (3) resolution vs. speed; (4) dynamic range; (5) XRES; (6) zoom; (7) time gain compensation. An indigenous water phantom was customized to satisfy the quantified need in Taguchi’s analysis. Unlike the conventional dynamic Taguchi analysis, an innovative quantified index, the figure of merit (FOM), was proposed to integrate four specific quality characteristics, namely (i) average difference between the practical scan and theoretically preset area (78.5, 50.2 and 12.6 mm2) of stenosis, (ii) standard deviation of the average, (iii) practical scan’s sensitivity β to various stenosis diameters (10, 8, and 4 mm), and (iv) correlation coefficient r2 of the linear regressed sensitivity curve. The highest value (FOM = 0.413) was furnished by the optimal combination of factors on 18 groups under study, yielding high r2 and low β or standard deviation values and the best quality of ultrasound images for the further clinical diagnosis. The comparison between FOM and the conventional signal-to-noise (S/N) ratio in Taguchi’s analysis revealed that FOM compiled more quality characteristics that were superior by nature to fulfill the practical need in clinical diagnosis. The alternative choice in ultrasound scan optimization can be based on stenosis diameter variation from a different perspective to be explored in the follow-up study.

Circulating tumor cells (CTCs) have the potential to predict metastasis, and the capture of CTCs based on their surface markers is mostly applied for CTC detection. Considering that the CTCs with a metastatic phenotype preferably form a metastatic focus and that aptamers have the ability to bind targets with high specificity and affinity, we selected aptamers directed toward metastatic cells by subtractive Cell-SELEX technology using highly metastatic MDA-MB-231 cells as the target cell and low-metastatic MCF-7 cells as the negative cell for the capture of metastatic CTCs. Affinity and selectivity assays showed that aptamer M3 had the highest affinity, with a KD of 45.6 ± 1.2 nM, and had good specificity against several other types of metastatic cancer cells. Based on these findings, we developed an M3-based capture system for CTC enrichment, which has the capability to specifically capture the metastatic cells MDA-MB-231 mixed with non-metastatic MCF-7 cells and CTCs derived from the peripheral blood from metastatic breast cancer patients. A further comparative analysis with the anti-EpCAM probe showed that M3 probe captured epithelial feature-deletion metastatic cells. We developed an aptamer-based CTC capture system through the selection of aptamers by taking whole metastatic cells, not known molecules, as targets, which provided a new insight into CTC capture and Cell-SELEX application. Surface plasmons supported by various metallic nanostructures have given rise to several significant breakthroughs in the field of integrated photonic devices due to its ability to effectively confine and enhance optical field in subwavelength volume. In particular, the demand to actively control optical responses of plasmonic systems becomes urgent for the miniaturization of signal processing devices, surface-enhanced Raman scattering (SERS) substrates and biochemical sensors. In this paper, we systematically investigate the plasmon modes as well as their interaction in a layered nanostructure composed of a periodically-arranged radiative nanoring and a metallic ground plane, as well as a thin insulating spacer. A tunable transparent peak on the background of the broadband plasmon resonance emerges in the reflection spectrum as changing the periodicity of nanoparticle array, a plasmonic analogue of electromagnetically induced transparency (EIT). Owing to the structural symmetry of the rings, we demonstrate a new scheme of controlling the interference between localized and delocalized plasmons by means of incident polarization and believe that the proposed metasurface may find applications in optical switching if the polarization-controlled components are introduced. Background: The PIK3CA (H1047R) mutation is considered to be a potential predictive biomarker for EGFR-targeted therapies. In this study, we developed a novel PCR-PFLP approach to detect the PIK3CA (H1047R) mutation in high effectiveness.Methods: A 126-bp fragment of PIK3CA exon-20 was amplified by PCR, digested with FspI restriction endonuclease and separated by 3 % agarose gel electrophoresis for the PCR-RFLP analysis. The mutant sequence of the PIK3CA (H1047R) was spiked into the corresponding wild-type sequence in decreasing ratios for sensitivity analysis. Eight-six cases of formalin-fixed paraffin-embedded colorectal cancer (CRC) specimens were subjected to PCR-RFLP to evaluate the applicability of the method.Results: The PCR-RFLP method had a capability to detect as litter as 0.4 % of mutation, and revealed 16.3 % of the PIK3CA (H1047R) mutation in 86 CRC tissues, which was significantly higher than that discovered by DNA sequencing (9.3 %). A positive association between the PIK3CA (H1047R) mutation and the patients' age was first found, except for the negative relationship with the degree of tumor differentiation. In addition, the highly sensitive detection of a combinatorial mutation of PIK3CA, KRAS and BRAF was achieved using individual PCR-RFLP methods.Conclusions: We developed a sensitive, simple and rapid approach to detect the low-abundance PIK3CA (H1047R) mutation in real CRC specimens, providing an effective tool for guiding cancer targeted therapy. The chemical bath deposition (CBD) method is very crucial to the reaction rate. Generally, the rate can be controlled through tuning buffer agent concentration. CdS nanocrystals on the silicon nanoporous pillar array (CdS/Si-NPA) have been prepared through the CBD method. By varying the buffer agent concentration, the reaction rates can be tuned. The diffraction peaks of hexagonal CdS and Cd can be observed due to the reduction of Cd2+ caused by the silicon nanoporous pillar array. The average size of CdS nanocrystals is decreased with the increasing buffer agent concentration and the optical band gaps from CdS nanocrystals are increased. From the photoluminescence of CdS/Si-NPA, it can be observed that the blue emissions are independent of the buffer agent concentration and the green emissions show blue shift with the increasing buffer agent concentration. We theoretically investigate optical properties of a plasmonic metasurface consisting of a dipolar wire as the bright antenna stacked above a quadrupolar wire as the dark antenna. It is demonstrated that by adjusting the lateral displacement between the two resonators, the spectral feature of the metasurface can be evolved from the plasmonic electromagnetically-induced transparency to electromagnetically-induced absorption. The extracted physical parameters based on the two-coupled-oscillator model reveal that the near-field coupling strength plays a key role for the transition behavior in the plasmonic metasurface. In this paper, we demonstrate that an electromagnetically induced transparency (EIT)-like behavior is induced by the coupling of two magnetic resonances in a planar terahertz (THz) metamaterial structure, which consists of a U-shaped split ring (USR) as one bright resonator and a cut wire pair (CWP) as one dark resonator. It is found that the EIT-like spectral response is insensitive to the lateral distance between the two magnetic resonators, whereas an on-to-off modulation of the amplitude of the transparency window can be achieved by moving the USR vertically along the CWP. The underlying physical mechanism is mainly attributed to magneto-inductive excitation of the CWP. This investigation may provide an insight of the role of magnetic coupling in achieving EIT-like effect, inspiring interest in the developments of magnetically tunable transparency metamaterial for a wide range of optical devices with switching and modulation capabilities. In this paper, a plasmonic analogue of electromagnetically induced transparency (EIT) is demonstrated theoretically in a T-shaped silver nanohole array. A sharply narrow reflectance transparency window is clearly observed within the background spectrum of the broad dipole-like resonance at optical frequencies when structural asymmetry is introduced. Furthermore, the transparency peak exhibits highly sensitive response to the refractive index of surrounding medium and yield a sensitivity of 725 nm/refractive index unit (RIU), which ensures our proposed nanohole array as an excellent plasmonic sensor. In this paper, based on the constructive interference of plasmonic dipolar and quadrupolar modes, a classical analogue of electromagnetically induced absorption (EIA) is demonstrated theoretically in a stacked metamaterial consisting of a short metal strip (which acts as a bright resonator) and a long metal strip (acting as a dark resonator), which has been reported to support the electromagnetically induced transparency (EIT) effect. The transition from EIA to EIT can be clearly observed in the absorbance spectra via varying the vertical spacing between two resonant oscillators. With the help of the coupled two-oscillator model, the phase shift between the bright and dark resonance modes is calculated by fitting the simulated absorbance spectra, which reveals the physical mechanisms behind constructive and destructive interference effects in EIT/EIA metamaterials. CdS/Si multi-interface nanoheterojunction has been fabricated by depositing CdS nanocrystals (nc-CdS) on the silicon nanoporous pillar array (Si-NPA) using a chemical bath deposition method. Utilizing the reducibility of Si-NPA, Cd nanocrystals (nc-Cd) are incorporated into the Si-CdS interface. The structures of CdS/Si multi-interface nanoheterojunction with incorporation of nc-Cd into the interface (CdS:Cd/Si-NPA) has been concluded. The upper layer is a continuous CdS thin film, the intermediate layer is the interface region constructed by randomly and alternately dispersed nc-CdS and nc-Si with small amount of nc-Cd, and the bottom layer is the unfilled silicon nanoporous layer grown on sc-Si substrate. An obvious photovoltaic effect is observed in the prototype photovoltaic device based on CdS:Cd/Si-NPA. Compared with the previous work, the increased energy conversion efficiency has been ascribed to the incorporation of nc-Cd into the Si-CdS interface. [ABSTRACT FROM AUTHOR]
