Abstract Background The triglyceride glucose (TyG) index and triglyceride-to-high-density lipoprotein cholesterol (TG/HDL-C) ratio are recognized as simple non-insulin-based insulin resistance indices. Our study aimed to explore the relationship between these two indicators and heart failure (HF) in overweight or obesity individuals without diabetes. Methods This cross-sectional study selected 13,473 participants from the National Health and Nutrition Examination Survey (NHANES) 2001–2018 dataset. Weighted multivariable logistic regression and subgroup analysis were employed to evaluate the relationships between TyG index, TG/HDL-C ratio, and HF prevalence, respectively. Additionally, smooth curve fitting was utilized to analyze the dose–response relationships. Results A total of 13,473 obesity or overweight people without diabetes were included in this study through screening, among whom 291 (2.16%) had comorbid HF. Abstract A hallmark of tumor cells, including bladder cancer (BLCA) cells, is metabolic reprogramming toward aerobic glycolysis (Warburg effect). The classical oncogene MYC, which is crucial in regulating glycolysis, is amplified and activated in BLCA. However, direct targeting of the c-Myc oncoprotein, which regulates glycolytic metabolism, presents great challenges and necessitates the discovery of a more clarified regulatory mechanism to develop selective targeted therapy. In this study, a siRNA library targeting deubiquitinases identified a candidate enzyme named USP43, which may regulate glycolytic metabolism and c-Myc transcriptional activity. Further investigation using functional assays and molecular studies revealed a USP43/c-Myc positive feedback loop that contributes to the progression of BLCA. Moreover, USP43 stabilizes c-Myc by deubiquitinating c-Myc at K148 and K289 primarily through deubiquitinase activity. Additionally, upregulation of USP43 protein in BLCA increased the chance of interaction with c-Myc and interfered with FBXW7 access and degradation of c-Myc. These findings suggest that USP43 is a potential therapeutic target for indirectly targeting glycolytic metabolism and the c-Myc oncoprotein consequently enhancing the efficacy of bladder cancer treatment.

Abstract Clear cell renal cell carcinoma (ccRCC) is a type of kidney cancer that is both common and aggressive, with a rising incidence in recent decades. Hypoxia is a key factor that plays a vital role in the tumorigenesis and metastasis of malignancy. However, the precise mechanisms of hypoxia driving ccRCC progression were not totally uncovered. Our study found that hypoxia level was elevated in ccRCC and might be an independent risk factor of prognosis in ccRCC patients. We identified a key protein PLOD2 was induced under hypoxic conditions and strongly associated with poor prognosis in ccRCC patients. When PLOD2 was depleted, the proliferation and migration of ccRCC cells were reduced in vitro and in vivo, while overexpression of PLOD2 had the opposite effect. Mechanically, the study further revealed that PLOD2 was transcriptionally activated by HIF1A, which binds to a specific promoter region of the PLOD2 gene. PLOD2 was also shown to interact with EGFR, leading to the phosphorylation of the receptor. Furthermore, PLOD2 was responsible for binding to the extracellular domain of EGFR, which ultimately activated the AKT signaling pathway, thus promoting the malignant progression of ccRCC. Treatment with the PLOD2 inhibitor Minoxidil significantly suppressed ccRCC progression by inactivating the EGFR/AKT signaling axis. In summary, the findings of this study shed light on the molecular mechanisms behind PLOD2 expression in ccRCC and suggest that it may serve as a potential predictor and therapeutic target for the clinical prognosis and treatment of ccRCC.

At the beginning of the 21st century，a large number of oil and gas wells were put into operation in Bohai E oilfield.With the production of the oilfield entering the middle and late stage，the phenomenon of annulus pressure is common in gas wells，severe situations may develop into same pressure of tubing and casing， causing serious pipe corrosion hazards and serious safety issues such as oil and gas leakage and environmental pollution.In order to ensure the production safety of old wells and break through the technical limitations in stimulation operation，in this paper the influence of different factors on the corrosion rate of oil and gas well tubing was investigated， the statistics on the operation of five gas wells in Bohai E oilfield were conducted， the types of media that might cause corrosion were discussed.The integrity of the oil casing body was tested using different methods.Based on indoor experimental results and actual conditions， the corrosion rate of the oil casing in the above five gas wells was predicted， which would provide a basis for determining the degree of oil casing corrosion.

Mitochondria play a key role in the cerebral ischemia-reperfusion injury. Although the extracellular signal-regulated kinase 1/2 inhibitor PD98059 (PD) is a selective and reversible flavonoid that can protect the mitochondria in a rat model of cardiac arrest/cardiopulmonary resuscitation, its role requires further confirmation. In this study, we investigated whether PD could maintain mitochondrial homeostasis and decrease reactive oxygen species (ROS) production in neuroblastoma (SH-SY5Y) cells exposed to oxygen–glucose deprivation/reperfusion (OGD/R). PD improved the mitochondrial morphology and function, reversed the increase in ROS production and cell apoptosis, and reduced total-superoxide dismutase and Mn-superoxide dismutase activities induced by OGD/R. PD decreases ROS production and improves mitochondrial morphology and function, protecting SH-SY5Y cells against OGD/R-induced injury.

Polygonatum cyrtonema Hua is a traditional Chinese herb medicine, and it is widely distrihuted in China. The intrageneric taxonomy and phylogenetic relationships within Polygonatum have long heen controversial due to their morphological similarity and lacking special DNA barcodes. In this paper, the complete chloroplast genome is a relatively conserved quadripartite structure including a large single copy region of 84 711 bp, a small single copy region of 18 210 bp, and a pair of inverted repeats region of 26142 bp. A total of 342 simple sequence repeats were identified, and most of them were found to be composed of A/T, including 126 mono-nucleotides and 179 di-nucleotides. Nucleotide diversity was analyzed and eight highly variable regions (psbl~trnT-CGU, atpF~atpH, trnT-GGU~psbD, psaJ~rps20, trnL-UAG~ndhD, ndhG~ndhl, ndhA, and rpl32~ccsA) were identified as potential molecular markers. Phylogenetic analysis based on the whole chloroplast genome showed that P. cyrtonema, within the family Asparagaceae, is closely related to Polygonatum sibiricum and Polygonatum kingianum. The sequence matK, trnT-GGU & ccsA, and ndhG~ndhA were identified as three DNA barcodes. The assembly and comparative analysis of P. cyrtonema complete chloroplast genome will provide essential molecular information about the evolution and molecular biology for further study. An optimal, complete, continuous theory of the Luenberger dynamic compensator (or state estimator or state observer) is obtained for the recently studied class of heat-structure interaction partial differential equation (PDE) models, with structure subject to high Kelvin-Voigt damping, and feedback control exercised either at the interface between the two media or else at the external boundary of the physical domain in three different settings. It is a first, full investigation that opens the door to numerous and far reaching subsequent work. They will include physically relevant fluid-structure models, with wave- or plate-structures, possibly without Kelvin-Voigt damping, as explicitly noted in the text, all the way to achieving the ultimate discrete numerical theory, so critical in applications. While the general setting is functional analytic, delicate PDE-energy estimates dictate how to define the interface/boundary feedback control in each of the three cases.

The terminal alkyne C≡C stretch has a large Raman scattering cross section in the "silent" region for biomolecules. This has led to many Raman tag and probe studies using this moiety to study biomolecular systems. A computational investigation of these systems is vital to aid in the interpretation of these results. In this work, we develop a method for computing terminal alkyne vibrational frequencies and isotropic transition polarizabilities that can easily and accurately be applied to any terminal alkyne molecule. We apply the discrete variable representation method to a localized version of the C≡C stretch normal mode. The errors of (1) vibrational localization to the terminal alkyne moiety, (2) anharmonic normal mode isolation, and (3) discretization of the Born–Oppenheimer potential energy surface are quantified and found to be generally small and cancel each other. This results in a method with low error compared to other anharmonic vibrational methods like second-order vibrational perturbation theory and to experiments. Several density functionals are tested using the method, and TPSS-D3, an inexpensive nonempirical density functional with dispersion corrections, is found to perform surprisingly well. Diffuse basis functions are found to be important for the accuracy of computed frequencies. Finally, the computation of vibrational properties like isotropic transition polarizabilities and the universality of the localized normal mode for terminal alkynes are demonstrated. The development and utilization of perovskites are beneficial to improve or even change the optical properties of devices and obtain fascinating performances such as higher photoelectric conversion efficiency, better thermal stability, higher external quantum efficiency, more excellent remodeling, and flexibility. So, there are many articles on perovskite reviews having been reported from synthesis, properties to various applications (such as optoelectronic devices, electrical memristor, etc.). Based on the reported review of perovskites, this study will make a further supplement to the research progress of perovskites in visible light communication (VLC), optical neuromorphic devices, and highlight huge development prospects in these emerging fields in recent years. First, we briefly reviewed the preparation methods of common perovskite materials, followed by the optical and electrical characteristics. Then, the specific applications of optical properties based on perovskite materials are emphatically investigated, in addition to traditional photovoltaic devices, especially the latest cutting-edge fields of information encryption and decryption, VLC as well as optical memristive devices for photonic synapse and photonic neuromorphic computing. Finally, the main conclusions and prospects are given. Perovskite-based optical memristive devices are enabled to assist photonic neuromorphic calculations, showing huge potential application prospects in intelligent integrated chip fusing sensing, storage, and computing.
