Abstract Aquatic macrophytes form a three dimensional complex structure in the littoral zones of lakes, with many physical, chemical and biological gradients and interactions. This special habitat harbours a unique microalgal assemblage called metaphyton, that differs both from the phytoplankton of the pelagial and from the benthic assemblages whose elements are tightly attached to the substrates. Since metaphytic assemblages significantly contribute to the diversity of lakes’ phytoplankton, it is crucial to understand and disentangle those mechanisms that ensure their development. Therefore, we focused on the question of how a single solid physical structure contribute to maintaining metaphytic assemblages. Using a laboratory experiment we studied the floristic and functional differences of microalgal assemblages in microcosms that simulated the conditions that an open water, a complex natural macrophyte stand (Utricularia vulgaris L.), or an artificial substrate (cotton wool) provide for them. We inoculated the systems with a species rich (> 326 species) microalgal assemblage collected from a eutrophic oxbow lake, and studied the diversity, trait and functional group composition of the assemblages in a 24 day long experimental period. We found that both natural and artificial substrates ensured higher species richness than the open water environment. Functional richness in the open water environment was lower than in the aquaria containing natural macrophyte stand but higher than in which cotton wool was placed. This means that the artificial physical structure enhanced functional redundancy of the resident functional groups. Elongation measures of microalgal assemblages showed the highest variation in the microcosms that simulated the open water environment. Our results suggest that assembly of metaphytic algal communities is not a random process, instead a deterministic one driven by the niche characteristics of the complex three dimensional structure created by the stands of aquatic macrophytes.

Nowadays the increasing amount of saline wastewaters has given rise to various biological desalination processes, among which the application possibilities of microalgae represents a priority research area. Next to “real” aquatic species (members of phytoplankton or phytobenthon), species from ephemeral aquatic habitats or aeroterrestrial algae also could be good candidates of research studying salt tolerance or desalination ability, since salinity stress is often referred as “physiological drought” and species from ephemeral habitats can be characterized by high drought tolerance. In this study, the salinity tolerance, salt and nutrient removal ability of a Haematococcus lacustris strain from eastern Hungary were investigated. Vegetative cells showed low salt tolerance, survival was ensured by the formation of cysts up to a sodium-chloride concentration of 2,000 mg l−1. Although relatively moderate (a max. 30%) conductivity reduction and chloride removal were observed, notable (nearly 100%) nitrate and phosphate removal occurred even in the presence of 2,000 mg l−1 NaCl. Carotenoid accumulation was observed earlier and in higher extent in salt treated cultures than in drying out ones, although the amount of astaxanthin-esters was significantly higher in the cultures of drying out experiment than in the corresponding cultures of salt treatment characterized with similar chloride content. Our results suggest that algae isolates from ephemeral aquatic habitats endangered by regular drying out (exposed to special salt stress), could have notable salt tolerance and consequently successful applicability in nutrient removal processes from slightly saline wastewaters. The accumulation of valuable metabolites (such as astaxanthin) as a response to salinity stress, could enhance the economic value of the biomass.

Changing precipitation dynamics is one of the most important mechanisms that, by affecting the water regime, modifies the physical and chemical environment of aquatic assemblages. Thus, hydrological extremes can be considered as key drivers that shape algal assemblages and lead to diversity changes even in large rivers. Here, we investigated the long-term changes in the benthic diatom composition of the Rába River (River Raab), the watershed of which experienced continuous loss of precipitation in the last five years. We aim to answer the main question: Do one-off drought events and trend-like (continuous) precipitation decrease result in similar changes within diatom assemblages of this perennial river? Because, it has been already demonstrated that resilience and resistance of assemblages may vary depending on whether the drought occurs regularly or only occasionally. Our results demonstrated that one-off dry events hardly affected either assemblages’ composition or biodiversity. In contrast, continuously decreasing precipitation (drying period) had a pronounced effect on taxa and trait distribution and resulted in a significant decrease in taxonomic diversity and in functional richness. It is important to stress that the observed deteriorating diversity anticipates that a lasting drought period is likely to upset the ecological balance of the ecosystem and lead to remarkable natural damage. Since the climate scenarios project extremes in water regime in the near future, including longer periods of low precipitation, any knowledge that predicts changes in microflora can help to develop action plans by authorities to save lotic ecosystems.

Under increasing pressure of climatic change and anthropogenic eutrophication, water blooms, i.e. the formation of high phytoplankton biomass of a single or a few species, have become more and more frequent in lake ecosystems that is caused mostly by Cyanobacteria. The dynamics of phytoplankton under a cyanobacterial pressure may provide important information about what a competitive advantage of Cyanobacteria have over eukaryotic microalgae. The research, therefore focused on the relationship between cyanobacterial blooms and the accompanying species in hypertrophic shallow lakes where nutrients are in excess. The following hypotheses were selected for testing: i) Cyanobacterial blooms negatively affect the species richness and diversity of phytoplankton assemblages in the studied hypertrophic lakes. ii) Cyanobacterial blooms negatively affect the multi-trait functional diversity of phytoplankton assemblages in the studied hypertrophic lakes. iii) The formation of a cyanobacterial bloom causes a shift in the species and trait composition of phytoplankton assemblages. It was found that the species diversity of non-cyanobacterial assemblages was not affected negatively by increasing density of Cyanobacteria. Cyanobacterial biomass negatively affected the functional richness and functional dispersion of the assemblages, but both relationships were only marginally significant. Some temporal shift was detected in the community weighted mean values of width, colonial ability of accompanying microalgae, and also in flagellatedness and mixotrophic ability. We conclude that Cyanobacterial blooms basically alter dominance relations in the phytoplankton and reduce availability of light in waters. However, it does not necessarily coincide with the elimination of other taxa and reduction their species numbers or diversity. The formerly published data on the decrease in species richness can be likely explained by methodological deficiencies, which are responsible for reduced detectability of species. To better understand the seasonal dynamics and resilience of phytoplankton assemblages in hypertrophic water bodies the development of new methodologies by which diversity and succession of subordinate species can be evaluated is required.

Climate change is putting increasing pressure on flowing waters. Drastic water level fluctuations in rivers or drying up of small and medium-sized streams all contribute to the biodiversity crisis threatening freshwater ecosystems. Benthic diatoms are important elements of biofilm in small streams. However, knowledge on the relationship between benthic diatoms and flow intermittence is incomplete, especially in regions recently impacted by recurrent drying. Thus, we investigated benthic diatom flora of small intermittent, hilly streams in the warm temperate region of Europe (the Pannonian Ecoregion). Our hypotheses were addressed to compositional changes, biodiversity loss and diatom-based ecological assessment. The results revealed clear flow intermittence-induced differences in taxa and trait composition of diatoms. Altogether six species for the dry phase and three species in the aquatic phase were identified as indicative ones by using indicator value analyses. In contrast to water regime induced changes in assemblages, there was a seasonal overlap in taxa and trait composition. During the study period, the drying up of streams did not result in significant biodiversity loss either at taxa or trait levels. Functional dispersion, however, reduced significantly by summer. Overall, neither the hydrological regime nor seasonal changes had a significant effect on diatom-based quality indices, except for the Rott trophic index (TID index). The TID index values were significantly lower in dry phases than in aquatic ones. These results suggested that the drying up of streams has a very complex influence on benthic diatoms. It seems that taxonomical and functional redundancy can reduce the negative impact of short-time flow intermittence on assemblages. As a practical benefit, the results are the first to support the use of diatom-based quality indices in the assessment of flow intermittence in the temperate region.

Even though microscopic algae play pivotal role in the healthy functioning of freshwater ecosystems, recent water protection strategies rarely consider them and primarily focus on macroscopic organisms. Here, we studied the effect of protection level and utilization type of lowland standing waters on the composition and diversity of benthic diatom assemblages and on the diatom-based ecological status of waters. We hypothesized that (i) protected waters will sustain more diverse diatom assemblages and better ecological quality than not protected ones. We also hypothesized that (ii) the increase in number of utilization will affect negatively on biodiversity and on ecological quality. Clear taxonomic differences were revealed only in protected and not utilized waters while trait composition in protected waters was independent from the utilization type. Neither biodiversity nor ecological status of waters were influenced by protection level. The increase in number of utilization types, however, significantly decreased functional richness. Although high biodiversity of algae can effectively buffer the negative effects of climate change and anthropogenic impact, recent protection strategies are insufficient to support it.

Land-use imposes an important potential threat on the aquatic ecosystems of riverine habitats. In this study, DNA metabarcoding was used to assess the effect of land-use on diatom assemblages, with a special focus on cropland area as an integrative proxy for several direct-acting pressures. The so-called taxonomy-free approach was tested using exact sequence variants (ESVs) without pre-clustering and without assigning them to traditional taxonomy. Our hypothesis is that the taxonomy-free approach gives better interpretation of the effect of agricultural land-use and it provides a more efficient index to indicate agricultural stress than the classical method with taxonomical clustering. Classical ordination techniques (PCA, NMDS) were performed to study the effects of cropland area proportion on diatom assemblages and a modified version of the Zelinka-Marvan equation for the index development. Results showed that (i) although ESVs provided better results when studying land-use effects on diatoms, taxonomic assignment after analysis was necessary to give ecological interpretations and that (ii) a better performing index could be developed by using the taxonomy-free approach. By using ESVs without taxonomic assignment, information on the ecology of sequences belonging to the same species and of unassigned sequences could be kept. New types of clustering methods are welcome in the future of biomonitoring where the delimitation of taxonomic units should be refined based on a higher emphasis on their ecology rather than on morphological or genetical criteria.

Mass production of microorganisms, algae among them, for new bioactive compounds and renewable innovative products is a current issue in biotechnology. The greatest challenge of basic research on this topic is to find the best solution for both physiology and scalability. In this study, the main goal was to highlight the contradictions of physiological and technological optimization in the same, relatively small, laboratory scale. The green alga Monoraphidium pusillum (Printz) Komárková-Legnorová was cultured in a conventional Erlenmeyer flask (as air bubbled in a tank-type photobioreactor) and in a hybrid (fermenter type + helical tubular type) photobioreactor of the same volume (2.8 L). Higher cell numbers from 1.7–2.3-fold, 2–2.8-fold higher dry masses, and 1.9–2.6-fold higher total lipid contents (mg·L−1) were measured in the tank reactor than in the hybrid reactor. Cultures in the conventional tank reactor were characterized with better nutrient utilization (42.8–77.7% higher phosphate uptake) and more diverse lipid composition than in the hybrid reactor. The study highlights that well-scalable arrangements and settings could be not optimal (or unsuitable in some cases) from a physiological point of view. The results suggest certain developmental directions for complex, well-scalable devices and highlight the importance of testing the gained physiological optima on these systems.

Increased proliferation of algae is a current problem in natural and artificial water bodies. Controlling nutrients is the most sustainable treatment of increased algal proliferation, however in certain cases, it is not sufficiently available, or it does not provide results fast enough. Chemicals derived from natural sources, which could be effective in low concentrations and are biodegradable, may have an advantage over conventional chemical treatments. The main aim of the present study was to investigate the anti-cyanobacterial and anti-algal properties of allyl-isothiocyanate-containing essential oil produced from horseradish roots with a complex approach of the topic: on laboratory strains of cyanobacteria and eukaryotic algae, on microcosms containing natural phytoplankton assemblages, and on semi-natural biofilms. The results show that acute treatment can significantly reduce the viability of all the tested cyanobacteria and eukaryotic algae. Results of microcosm experiments with natural phytoplankton assemblages show that horseradish essential oil from 7.1 × 10−6% (v/v) is applicable to push back phytoplankton proliferation even in natural assemblages. The individual number in the biofilm was dropped down to one-fifth of the original individual number, so 7.1 × 10−6% (v/v) and higher concentration of the essential oil can be considered as a successful treatment against biofouling.

In wastewater, nutrient concentrations and salinity vary substantially, however, the optimal N:P ratio for the treatment using microalgae is not well described. In this study, the effects of higher and lower nitrate and phosphate contents and N:P ratios on growth, nutrient removal ability and halotolerance of the common green alga Coelastrum morus were investigated in model solutions. The results suggest that high nitrate content (above 100 mg L−1) with a similarly high phosphate concentration (resulting low N:P ratio) is not favorable for growth. The studied isolate can be considered as a halotolerant species, showing remarkable growth up to 1000 mg L−1 NaCl and it seems that despite the negative effects on growth, higher nutrient content contributes to higher halotolerance. A significant amount of nitrate removal was observed in media with different nutrient contents and N:P ratios with different salt concentrations. High N:P ratios favor phosphate removal, which is more inhibited by increasing NaCl concentration than nitrate uptake. Overall, with a relatively higher nutrient content and a favorable (5 or higher) N:P ratio, a common green algal species such as C. morus could be a promising candidate next to species from the Chlorellaceae and Scenedesmaceae families.

Cylindrospermopsin (CYN) is a toxic secondary metabolite produced by filamentous cyanobacteria which could work as an allelopathic substance, although its ecological role in cyanobacterial-algal assemblages is mostly unclear. The competition between the CYN-producing cyanobacterium Chrysosporum (Aphanizomenon) ovalisporum, and the benthic green alga Chlorococcum sp. was investigated in mixed cultures, and the effects of CYN-containing cyanobacterial crude extract on Chlorococcum sp. were tested by treatments with crude extracts containing total cell debris, and with cell debris free crude extracts, modelling the collapse of a cyanobacterial water bloom. The growth inhibition of Chlorococcum sp. increased with the increasing ratio of the cyanobacterium in mixed cultures (inhibition ranged from 26% to 87% compared to control). Interestingly, inhibition of the cyanobacterium growth also occurred in mixed cultures, and it was more pronounced than it was expected. The inhibitory effects of cyanobacterial crude extracts on Chlorococcum cultures were concentration-dependent. The presence of C. ovalisporum in mixed cultures did not cause significant differences in nutrient content compared to Chlorococcum control culture, so the growth inhibition of the green alga could be linked to the presence of CYN and/or other bioactive compounds.
