Sustainable agricultural practices are increasingly becoming a strategic asset for global and national environmental policies and economy. A big challenge is the selection of appropriate indicators to describe the complexity of the agroecosystem management. In the present work, the sustainability of grape production, in vineyard trials of Pinot blanc and Rhine Riesling, managed with integrated (INT) and organic management (organic, with cattle manure ORG1 and organic with green manure ORG2), was compared using a multi-indicator approach. The experiment was set in 2011 (1.5 ha in Trento, Italy) and carbon footprint (CF), nitrogen footprint (NF), water footprint (WF), soil microbial diversity (alpha diversity of bacteria, fungi, oomycetes communities) and soil C stock change, were evaluated in 2018. The CF was 0.213—0.227 kg CO2-eq/kg in the INT, 0.144—0.168 kg CO2-eq/kg in ORG1 and 0.134—0.147 kg CO2-eq/kg in ORG2. The NF was around 1 g Nr/kg for the INT, 0.4 g Nr/kg for ORG1 and 0.5 g Nr/kg for ORG2. The WF, excluding the pesticides impact on grey water, was 666—708 L/kg for INT, 605—655 L/kg for ORG1 and 529—580 L/kg for ORG2. The impact of farming practices on soil microbial alpha diversity showed no significant difference among treatments for oomycetes and significantly higher indexes for fungi and bacteria in the ORG1, with INT and ORG2, being similar. No difference in bulk organic C were observed among treatments. Overall, the multi-indicator approach allowed to demonstrate that the organic management was more beneficial for most of the environmental spheres of the agroecosystem compared to integrated management, without affecting the grape yield.

Many countries rely on statements issued by professional associations to delimit the scope of school psychological practice. It is, however, not always clear to what extent these statements match current practices and school psychologists' self-perception of their professional role, as empirical data is often unavailable or limited. This study aims to address this gap by collecting empirical data on the scope of school psychological practice in Germany. In a mixed method study, we first applied the Delphi technique to develop a questionnaire in collaboration with school psychological experts from different federal states of Germany. Second, we collected information on federal policies through semi-structured interviews with regional experts. Third, we invited a representative sample of school psychologists to complete the questionnaire developed through the Delphi technique. In this first report, we focus on the Delphi procedure and overall results of the survey describing the scope of school psychological practice in Germany at a country-wide level. These findings provide a detailed characterization of the broad and heterogeneous scope providing an evidence-base for future research and political decision-making.

Forest management practices may heavily affect epiphytic cryptogams. This study was conceived in March 2016, as soon as we were informed about an authorized logging for timber within a Mediterranean mixed oak forest in Tuscany (central Italy), which threatened a large population of the forest macrolichen Lobaria pulmonaria (L.) Hoffm., composed of hundreds of fertile thalli. Lobaria pulmonaria is often used as an ecological indicator of high quality habitats hosting rare lichens, and in general, cryptogams worthy of conservation. The species has suffered a general decline throughout Europe as a consequence of air pollution and intensive forest management, and currently it is red-listed in several countries, where it is considered also as a “flag species”. We estimated that 40% of the lichen biomass (8.5-12.3 kg ha-1) has been lost due to logging operations (in the core area, up to 1.8 kg every 100 m2), in particular large and fertile thalli. One year after the conclusion of logging operations, the analysis of chlorophyll fluorescence emission (indicator of the photosynthetic performance of the lichen photobionts), revealed a significant reduction of the vitality of the thalli on retained-isolated trees. The article provides issues for conservation in Mediterranean oak forests and outlines the outmost importance of ensuring the safeguard of forest ecosystems hosting fertile populations of this model species, especially in the case of unprotected forests.

Chitosan was subjected to a crosslinking reaction with three polyhydroxylated diacids (glucaric (GlcA), mannaric (ManA), and mucic (MucA) acids) that only differ in the spatial orientation of their hydroxyl groups. This work aimed to obtain experimental evidence of the impact of the three-dimensional arrangement of the crosslinkers on the resulting properties of the products. In all the cases, the products were hydrogels, and their chemical structures were fully elucidated by FT-IR spectroscopy and conductometric titration. Thermogravimetric and morphological studies were also carried out. The specific surface area of all the products was similar and higher than that of native chitosan. Moreover, all hydrogels were characterized in terms of viscoelastic properties and long-term stability under external perturbation. Furthermore, their lead adsorption efficiency and swelling capacity were assessed. Despite the resemblant chemical structure in all the hydrogels, Ch/ManA exhibited the highest lead adsorption capacity, (Ch/ManA: 93.8 mg g−1, Ch/GlcA: 82.9 mg g−1, Ch/MucA: 79.2 mg g−1), while Ch/GlcA exhibited a remarkably higher swelling capacity (i.e., ~30% more than Ch/MucA and ~40% more than Ch/ManA). The results obtained herein evidenced that the selection of the polyhydroxylated crosslinker with the appropriate three-dimensional structure could be crucial to finely adjust the final materials’ features.

Yb:CaF2 transparent ceramics represent a promising laser gain medium for ultra-short lasers due to their characteristics: low phonon energy, relatively high thermal conductivity, negative thermo-optical coefficient, and low refractive index. Compared to single crystals, Yb:CaF2 ceramics offer superior mechanical properties, lower cost, and it is easier to obtain large-sized samples with proper shape and uniform Yb3+ doping at high concentrations. The combination of air pre-sintering and Hot Isostatic Pressing (HIP) emerges as a viable strategy for achieving high optical quality and fine-grained structure of ceramics at lower sintering temperatures. The properties of the powders used in ceramic fabrication critically influence both optical quality and laser performance of Yb:CaF2 ceramics. In this study, the 5 atomic percentage (at.%) Yb:CaF2 transparent ceramics were fabricated by air pre-sintering and hot isostatic pressing (HIP) using nano-powders synthesized through the co-precipitation method. The co-precipitated powders were optimized by studying air calcination temperature (from 350 to 550 °C). The influence of calcination temperature on the microstructure and laser performance of Yb:CaF2 ceramics was studied in detail. The 5 at.% Yb:CaF2 transparent ceramics air pre-sintered at 625 °C from powders air calcined at 400 °C and HIP post-treated at 600 °C exhibited the highest in-line transmittance of 91.5% at 1200 nm (3.0 mm thickness) and the best laser performance. Specifically, a maximum output power of 0.47 W with a maximum slope efficiency of 9.2% at 1029 nm under quasi-CW (QCW) pumping was measured.

Biscogniauxia nummularia is a xylariaceous fungus known as a common endophyte of European beech, living in plant tissues without development of symptoms, or even inducing strip-cankers and wood decay on trees stressed by drought. We studied the presence of the fungus in apparently healthy beech trees, growing in two different bioclimatic zones characterized by Continental and Mediterranean climates. Asymptomatic twigs were collected in each zone over the season and evaluated for the presence of B. nummularia infections using both cultural and qPCR methods. Results from qPCR indicated differences in the detection of B. nummularia among the seasons and between the study sites. In both sites the highest frequency of detection was in summer. B. nummularia was more frequently detected in the Mediterranean bioclimatic area, where drought is more common. These results suggest that B. nummularia may be a possible bioindicator of beech health stands.

In addition to defensins, plants possess an array of defensin-like peptides that share many of their characteristics, as well as a role in plant’s innate immunity. Their involvement in the response to pathogens is well-known but the contribution in the plant response to abiotic stimuli is not fully understood. We have undertaken an in silico analysis to characterize all defensin family genes hitherto found in Arabidopsis, including genes encoding for defensin-like peptides, by detecting several peptides as candidates for further studies aiming to decipher specific responses to biotic and abiotic stresses, as well as to their crosstalk. We performed several analyses, including co-expression and cis-regulatory elements analyses, using transcriptomic data obtained from the ARS database, which integrates more than 20,000 Arabidopsis RNA-seq libraries.In silico analysis showed that jasmonates and ABA, together with transcription factors belonging to WRKY and AP2/EREBP families, modulate defensin and defensin-like gene expression. Indeed, the analysis performed in this study allowed to extract and organize omics data, which finally supported the inducible nature of defensins under both abiotic and biotic stresses. Moreover, in vivo expression analyses confirmed the heat and drought responsiveness of PDF1.4, ATTI1, PDF1.1, DEFL 206, defensin family genes selected for being upregulated by several abiotic conditions, at transcriptional level. Finally, the co-expression analysis provided information on other biological processes that may be correlated to the defensin induction, such as maintaining ROS homeostasis. Combining the comprehensive analysis of different transcriptional datasets with the integration of in vivo analyses emerged as a robust methodological approach to assess the proposed multi-stress responsive nature of defensin family genes.

Abstract Purpose When dealing with the health status of the knee articular surface, the entire osteochondral unit has gained increasing attention, and in particular the subchondral bone, which plays a key role in the integrity of the osteochondral unit. The aim of this article was to discuss the current evidence on the role of the subchondral bone. Methods Experts from different geographical regions were involved in performing a review on highly discussed topics about the subchondral bone, ranging from its etiopathogenetic role in joint degeneration processes to its prognostic role in chondral and osteochondral defects, up to treatment strategies to address both the subchondral bone and the articular surface. Discussion Subchondral bone has a central role both from an aetiologic point of view and as a diagnostic tool, and its status was found to be relevant also as a prognostic factor in the follow‐up of chondral treatment. Finally, the recognition of its importance in the natural history of these lesions led to consider subchondral bone as a treatment target, with the development of osteochondral scaffolds and procedures to specifically address osteochondral lesions. Conclusion Subchondral bone plays a central role in articular surface lesions from different points of view. Several aspects still need to be understood, but a growing interest in subchondral bone is to be expected in the upcoming future towards the optimization of joint preservation strategies. Increasing numbers of reports have revealed novel catalytically active cryptic guanylate cyclases (GCs) and adenylate cyclases (ACs) operating within complex proteins in prokaryotes and eukaryotes. Here we review the structural and functional aspects of some of these cyclases and provide examples that illustrate their roles in the regulation of the intramolecular functions of complex proteins, such as the phytosulfokine receptor (PSKR), and reassess their contribution to signal generation and tuning. Another multidomain protein, Arabidopsis thaliana K+ uptake permease (AtKUP5), also harbors multiple catalytically active sites including an N-terminal AC and C-terminal phosphodiesterase (PDE) with an abscisic acid-binding site. We argue that this architecture may enable the fine-tuning and/or sensing of K+ flux and integrate hormone responses to cAMP homeostasis. We also discuss how searches with motifs based on conserved amino acids in catalytic centers led to the discovery of GCs and ACs and propose how this approach can be applied to discover hitherto masked active sites in bacterial, fungal, and animal proteomes. Finally, we show that motif searches are a promising approach to discover ancient biological functions such as hormone or gas binding.

Promoting academic success among undergraduate students is crucial for tackling the need to foster employability competencies. Low levels of academic attainment in higher education, along with the increasing number of persons participating in tertiary education, represent crucial trends, which need to be studied in order to develop efficient retention practices. The current study aimed to investigate the relationship between relevant factors that can foster academic success: learning strategies, future orientation, and academic self-efficacy. To this purpose, a longitudinal study was performed on a sample of N = 87 undergraduate students from one of the largest Italian universities (63.4% males, 74.2% enrolled in the first year). Participants filled in an online questionnaire at two different time points, with a time lag of 12 months. Results of a moderated mediation model indicated that the relationship between learning strategies at Time 1 (T1) and Grade Point Average (GPA) at Time 2 (T2) was mediated by students' future orientation. Moreover, this association was moderated by T1 academic self-efficacy. These results suggest that learning strategies positively influence GPA through an enhanced future orientation, in particular when students report high or medium levels of self-efficacy. The current findings invite a thorough review of training interventions for improving academic achievement.
