In this work, we present the detection of proteins expressed by poxvirus with fiber-optic probes based on a semi-distributed interferometer (SDI) assisted by a fiber Bragg grating (FBG), performing the measurement directly into a wastewater sample. Modern biosafety applications benefit from real-time, dynamic-sensing technologies that can perform diagnostic tasks into a wide set of analytes, with a particular emphasis on wastewater, which appears to collect a significant number of viral titers in urban and indoor environments. The SDI/FBG probe can perform substantial progress in this field, as it embeds a dual sensitivity mechanism to refractive index changes (sensitivity up to 266.1 dB/RIU (refractive index units)) that can be exploited in biosensing, while simultaneously having the capability to measure the temperature (sensitivity 9.888 pm/°C), thus providing an intrinsic cross-sensitivity compensation. In addition, a standard FBG analyzer can be used as an interrogator, improving affordability and real-time detection over previous works. The probes have been functionalized with antibodies specific for L1, A27 and A33 vaccinia virus proteins, performing detection of a protein concentration in a scenario compatible with online viral threat detection. Direct detection of wastewater samples shows that the L1-functionalized sensor has a higher response, 9.1–11.3 times higher than A33 and A27, respectively, with a maximum response of up to 1.99 dB and excellent specificity. Dynamic detection in wastewater shows that the sensors have a response over multiple detection cycles, with a sensitivity of 0.024–0.153 dB for each 10-fold increase of concentration.

Communications in space are rapidly increasing in both capacity and extension. A satellite network in space calls for efficient and lightweight optical transmitters, suitable to communicate over distances going from the low-Earth-orbit to interplanetary links and beyond. To this aims, here we study the use of novel resources as an array of nanoptical coupler from guided sources and modulators to free space modes, that is phased with a suitable correction systems as well as efficient coding with error corrections capable of bearing large values of losses. We develop a method for designing such an array, which achieve a main lobe divergence of 1 µrad. We also discuss the lower and upper limits of the beam steering technique as a function of the array and emitter size, respectively. The optimization of grating couples (GCs) resulted an apodized structure with approximately 0.7 coupling efficiency and 70-µm beam waist. We also demonstrate the feasibility of phase correction and polarization modulation of the output mode by means of metalenses. The resulting link budget in terms of received photons on the Earth sets exceptional conditions for the digital optical communication system, for which a power efficient modulation scheme such as pulse position modulation (PPM) is considered. Here, we assess the PPM performance and we propose suitable channel coding schemes, essential to reduce the high bit error rate obtained in few-photon regime at the receiver.

In this paper, we consider the problem of detecting the channel activity in LoRaWAN networks. To improve the medium access performance, recent implementations of the chipsets for LoRaWAN end nodes are offering a feature called Channel Activity Detection (CAD), as the first step toward the implementation of the Carrier Sense Multiple Access (CSMA) in LoRaWAN networks. The algorithm underlying the CAD is at the moment left undisclosed. In this paper, we propose a possible implementation of the CAD algorithm; we then evaluate its performance in an additive white Gaussian noise (AWGN) channel, and we find the performance being completely satisfactory and matching the one of the undisclosed algorithm. The results we find are encouraging the community to include the CAD feature in open source implementations of a LoRaWAN receiver.

Abstract In this work, we present the development and biofunctionalization of a fiber-optic ball-resonator biosensor for the real-time detection of vaccinia poxvirus. We fabricated several ball-tip resonators, functionalized through a silanization process to immobilize two bioreceptors: the monoclonal anti-L1R antibody targeting the L1R protein, and the polyclonal rabbit serum antibodies targeting the whole vaccinia virus (VV) pathogen. Experimental measurements were carried out to detect VV in concentrations from 103 to 108 plaque-forming units (PFU), with a limit of detection of around 1.7–4.3 × 103 PFU and a log-quadratic pattern, with a response up to 5 × 10−4 RIU (refractive index units). The specificity was assessed against herpes simplex virus, used as a non-specific control, with the best results obtained with anti-L1R monoclonal antibodies, and through the detection of vaccinia virus/herpes simplex-1 combination. The obtained results provide a real-time viral recognition with a label-free sensing platform, having rapid response and ease of manufacturing, and paving the road to the seamless detection of poxviruses affecting different human and animal species using optical fibers.

Traditional multiple access schemes, as well as more recent preamble-based schemes, cannot achieve the extremely low latency, complexity, and collision probability required by the next-generation Internet-of-Things (IoT) networks to operate. To address such issues and further reduce the latency and packet loss, we introduce a novel semi-grant-free multiple access protocol for short packet transmission, the partial-information multiple access (PIMA) scheme. PIMA transmissions are organized in frames, and in the partial information acquisition (PIA) sub-frame of each frame, the base station (BS) estimates the number of active devices, i.e., the devices having packets waiting for transmission in their queue. Based on this estimate, the BS chooses both the total number of slots to be allocated in the data transmission (DT) sub-frame and the respective user-to-slot assignment. Although collisions may still occur due to multiple users assigned to the same slot, they are drastically reduced with respect to slotted ALOHA-based schemes, while achieving lower latency than both time-division multiple-access (TDMA) and preamble-based protocols, due to the extremely reduced overhead of the PIA sub-frame. We assess the performance of PIMA under various activation statistics, proving the robustness of the proposed solution to the traffic intensity, also with traffic bursts.

Vehicular wireless networks are one of the most valuable tools for monitoring platforms in the automotive domain. At the same time, Internet of Things (IoT) solutions are playing a crucial role in the same framework, allowing users to connect to vehicles in order to gather data related to their working cycle. Such tasks can be accomplished by resorting to either cellular or non-cellular wireless technologies. While the former can ensure low latency but require high running costs, the latter can be employed in quasi-real-time applications but definitely reduce costs. To this end, this paper proposes the results of two measurement campaigns aimed at assessing the performance of the long-range wide-area network (LoRaWAN) protocol when it is exploited as an enabling technology to provide vehicles with connectivity. Performances are evaluated in terms of packet loss (PL) and received signal strength indicator (RSSI) in wireless links. The two testing scenarios consisted of a transmitter installed on a motorbike running on an elliptical track and a receiver placed in the centre of the track, and a transmitter installed on the roof of a car and a receiver placed next to a straight road. Several speeds were tested, and all the spreading factors (SFs) foreseen by the protocol were examined, showing that the Doppler effect has a marginal influence on the receiving performance of the technology, and that, on the whole, performance is not significantly affected by the speed. Such results prove the feasibility of LoRaWAN links for vehicular network purposes.

ObjectivesTo investigate the effects of SARS-CoV-2 infection, as well as short- (within 48 hours) and long-term (within 30 days) adverse events (AEs) of SARS-CoV-2 vaccines, including arthritis flares in a large cohort of patients with inflammatory arthritis (IA).MethodsA retrospective cohort study comprising 362 patients: 94 (26%) rheumatoid arthritis, 158 (43.6%) psoriatic arthritis and 110 (30.4%) ankylosing spondylitis; and 165 healthy controls (HC) to ascertain the prevalence and severity of SARS-CoV-2 infection in patients with IA, the rate of AEs associated with SARS-CoV-2 vaccines and disease flares within a month of the vaccination. All patients provided informed consent and data about SARS-CoV-2 infection and/or vaccination status.ResultsOne-hundred-seventeen (32.3%) patients and 39 (23.6%) HC were affected by SARS-CoV-2 infection. Forty (34.2%) patients experienced an IA flare within one month of infection, of whom 3 (7.5%) needed to switch therapy. The prevalence of SARS-CoV-2 infection, disease severity, and hospitalization rate were not significantly different. At least one shot of SARS-CoV-2 vaccine was administered in 331 (91.4%) patients and 147 (89.1%) HC. Within 48 hours, 102 (30.8%) patients developed vaccine-related AEs; 52 (15.7%) patients with >1 vaccine dose experienced an IA flare-up, of whom 12 (23.1%) needed to switch therapy.ConclusionsA significantly higher rate of IA flare was observed among patients who contracted SARS-CoV-2 infection vs. those without infection. Patients with IA experienced flares after SARS-CoV-2 vaccination, though it was not statistically significant.

Inflammatory bowel disease (IBD), comprising mainly Crohn’s disease (CD) and ulcerative colitis (UC), is a chronic inflammatory disease of the gastrointestinal tract. In recent years, a wealth of data has been accumulated demonstrating the complex interplay of many different factors in the pathogenesis of IBD. Among these are factors impacting the epithelial barrier function, including vessel and extracellular matrix (ECM) formation, the gut microbiome (e.g., bacterial antigens), and, most importantly, the production of cytokines (pro- and anti-inflammatory) directly shaping the immune response. Patients failing to resolve the acute intestinal inflammation develop chronic inflammation. It has been shown that the expression of the matricellular protein periostin is enhanced during IBD and is one of the drivers of this disease. The C-C chemokine receptor 5 (CCR5) is engaged by the chemotactic mediators CCL3/MIP-1α, CCL4/MIP-1β, and CCL5/RANTES. CCR5 blockade has been reported to ameliorate inflammation in a murine IBD model. Thus, both periostin and CCR5 are involved in the development of IBD. In this study, we investigated the potential crosstalk between the two signaling systems and tested a highly potent CCL5 derivative acting as a CCR5 antagonist in a murine model of IBD. We observed that the absence of periostin influences the CCR5-expressing cell population of the gut. Our data further support the notion that targeted modulation of the periostin and CCR5 signaling systems bears therapeutic potential for IBD.

Introduction:The plasticity of the neural circuits after injuries has been extensively investigated over the last decades. Transcallosal microsurgery for lesions affecting the third ventricle offers an interesting opportunity to investigate the whole-brain white matter reorganization occurring after a selective resection of the genu of the corpus callosum (CC).MethodDiffusion MRI (dMRI) data and neuropsychological testing were collected pre- and postoperatively in six patients with colloid cysts, surgically treated with a transcallosal-transgenual approach. Longitudinal connectometry analysis on dMRI data and graph analysis on structural connectivity matrix were implemented to analyze how white matter pathways and structural network topology reorganize after surgery.ResultsAlthough a significant worsening in cognitive functions (e.g., executive and memory functioning) at early postoperative, a recovery to the preoperative status was observed at 6 months. Connectometry analysis, beyond the decrease of quantitative anisotropy (QA) near the resection cavity, showed an increase of QA in the body and forceps major CC subregions, as well as in the left intra-hemispheric corticocortical associative fibers. Accordingly, a reorganization of structural network topology was observed between centrality increasing in the left hemisphere nodes together with a rise in connectivity strength among mid and posterior CC subregions and cortical nodes.ConclusionA structural reorganization of intra- and inter-hemispheric connective fibers and structural network topology were observed following the resection of the genu of the CC. Beyond the postoperative transient cognitive impairment, it could be argued anterior CC resection does not preclude neural plasticity and may subserve the long-term postoperative cognitive recovery.

The large number of pathologies that position CCR5 as a central molecular determinant substantiates the studies aimed at understanding receptor-ligand interactions, as well as the development of compounds that efficiently block this receptor. This perspective focuses on CCR5 antagonism as the preferred landscape for therapeutic intervention, thus the receptor active site occupancy by known antagonists of different origins is overviewed. CCL5 is a natural agonist ligand for CCR5 and an extensively studied scaffold for CCR5 antagonists production through chemokine N-terminus modification. A retrospective 3D modeling analysis on recently developed CCL5 mutants and their contribution to enhanced anti-HIV-1 activity is reported here. These results allow us to prospect the development of conceptually novel amino acid substitutions outside the CCL5 N-terminus hotspot. CCR5 interaction improvement in regions distal to the chemokine N-terminus, as well as the stabilization of the chemokine hydrophobic core are strategies that influence binding affinity and stability beyond the agonist/antagonist dualism. Furthermore, the development of allosteric antagonists topologically remote from the orthosteric site (e.g., intracellular or membrane-embedded) is an intriguing new avenue in GPCR druggability and thus a conceivable novel direction for CCR5 blockade. Ultimately, the three-dimensional structure elucidation of the interaction between various ligands and CCR5 helps illuminate the active site occupancy and mechanism of action.

In time of COVID-19 biological detection technologies are of crucial relevance. We propose here the use of state of the art optical fiber biosensors to address two aspects of the fight against SARS-CoV-2 and other pandemic human coronaviruses (HCoVs). Fiber optic biosensors functionalized with HCoV spikes could be used to discover broadly neutralizing antibodies (bnAbs) effective against known HCoVs (SARS-CoV, MERS-CoV and SARS-CoV-2) and likely future ones. In turn, identified bnAbs, once immobilized onto fiber optic biosensors, should be capable to detect HCoVs as diagnostic and environmental sensing devices. The therapeutic and preventative value of bnAbs is immense as they can be used for passive immunization and for the educated development of a universal vaccine (active immunization). Hence, HCoV bnAbs represent an extremely important resource for future preparedness against coronavirus-borne pandemics. Furthermore, the assembly of bnAb-based biosensors constitutes an innovative approach to counteract public health threats, as it bears diagnostic competence additional to environmental detection of a range of pandemic strains. This concept can be extended to different pandemic viruses, as well as bio-warfare threats that entail existing, emerging and extinct viruses (e.g., the smallpox-causing Variola virus). We report here the forefront fiber optic biosensor technology that could be implemented to achieve these aims.

With the aim of rationally devising a refined and potent HIV-1 blocker, the cDNA of CCL5 5p12 5m, an extremely potent CCR5 antagonist, was fused to that of C37, a gp41-targeted fusion inhibitor. The resulting CCL5 5p12 5m-C37 fusion protein was expressed in E. coli and proved to be capable of inhibiting R5 HIV-1 strains with low to sub-picomolar IC50, maintaining its antagonism toward CCR5. In addition, CCL5 5p12 5m-C37 inhibits R5/X4 and X4 HIV-1 strains in the picomolar concentration range. The combination of CCL5 5p12 5m-C37 with tenofovir (TDF) exhibited a synergic effect, promoting this antiviral cocktail. Interestingly, a CCR5-targeted combination of maraviroc (MVC) with CCL5 5p12 5m-C37 led to a synergic effect that could be explained by an extensive engagement of different CCR5 conformational populations. Within the mechanism of HIV-1 entry, the CCL5 5p12 5m-C37 chimera may fit as a powerful blocker in several instances. In its possible consideration for systemic therapy or pre-exposure prophylaxis, this protein design represents an interesting lead in the combat of HIV-1 infection.

Learning from the lengthy fight against HIV-1, influenza, and Ebola virus infection, broadly neutralizing antibodies (bnAbs), directed at conserved regions of surface proteins crucial to virus entry (Env, hemagglutinin, and GP, respectively), are an essential resource for passive as well as active immunization. Rare in their emergence and antigen recognition mode, bnAbs are active toward a large set of different viral strains. Isolation, characterization and production of bnAbs lead to their possible use in passive immunotherapy and form the basis for an educated effort in the development of vaccines for universal coverage. SARS-CoV-2-specific antibodies targeting the spike receptor binding domain (RBD) may lead to antibody dependent enhancement (ADE) of infection, possibly hampering the field of vaccine development. This perspective points to the identification of conserved regions in the spike of SARS-CoV-2, SARS-CoV, and MERS-CoV through investigation, dissection and recombinant production of isolated moieties. These spike moieties should be capable of independent folding and allow the detection as well as the elicitation of bnAbs, thus setting the basis for an effective passive immunotherapy and the development of a universal vaccine against human epidemic coronaviruses (HCoVs). SARS, MERS and, most of all, COVID-19 demonstrate that humanity is the target of HCoV, preparedness for future hits is thus no longer an option.

Spiral waves are self-repeating waves that can form in excitable media, propagating outward from their center in a spiral pattern. Spiral waves have been observed in different natural phenomena and have been linked to medical conditions such as epilepsy and atrial fibrillation. We used a simple cellular automaton model to study propagation in excitable media, with a particular focus on understanding spiral wave behavior. The main ingredients of this cellular automaton model are an excitation condition and characteristic excitation and refractory periods. The literature shows that fixed excitation and refractory periods together with specific initial conditions generate stationary and stable spiral waves. In the present work, we allowed the activation and refractory periods to fluctuate uniformly over a range of values. Under these conditions, this very simplistic model can recreate the meandering and breakup of spiral waves that were observed in more elaborate models in the literature.

In this work, we investigate the effect of thermal treatment on CeO2 films fabricated by using atomic layer deposition (ALD) on titanium nitride (TiN) or on silicon (Si) substrates. In particular, we report on the structural, chemical and morphological properties of 25 nm thick ceria oxide with particular attention to the interface with the substrate. The annealing treatments have been performed in situ during the acquisition of X-Ray diffraction patterns to monitor the structural changes in the film. We find that ceria film is thermally stable up to annealing temperatures of 900 °C required for the complete crystallization. When ceria is deposited on TiN, the temperature has to be limited to 600 °C due to the thermal instability of the underlying TiN substrate with a broadening of the interface, while there are no changes detected inside the CeO2 films. As-deposited CeO2 films show a cubic fluorite polycrystalline structure with texturing. Further, after annealing at 900 °C an increase of grain dimensions and an enhanced preferential (200) orientation are evidenced. These findings are a strong indication that the texturing is an intrinsic property of the system more than a metastable condition due to the ALD deposition process. This result is interpreted in the light of the contributions of different energy components (surface energy and elastic modulus) which act dependently on the substrate properties, such as its nature and structure.

The development of direct probes of entanglement is integral to the rapidly expanding field of complex quantum materials. Here we test the robustness of entangled neutrons as a quantum probe by measuring the Clauser-Horne-Shimony-Holt contextuality witness while varying the beam properties. Specifically, we show that the mode entanglement of the spin and path subsystems of individual neutrons prepared in two different experiments using two different apparatuses persists even after varying the entanglement length, coherence length, and neutron energy difference of the paths. The two independent apparatuses acting as entangler-disentangler pairs are static-field magnetic Wollaston prisms and resonance-field radio-frequency flippers. Our results show that the spatial and energy properties of the neutron beam may be significantly altered without reducing the contextuality witness value below the Tsirelson bound, meaning that maximum entanglement is preserved. We also show that two paths may be considered distinguishable even when the path states significantly overlap. Therefore, we have shown that our experimental results are consistent with the distinguishable subsystem assumption down to a separation of less than 100 nm, proving entanglement and the contextual nature of reality on short length scales. This work is the key step in the realization of the modular, robust technique of entangled neutron scattering, which can extract entanglement information from a sample without the knowledge of the microscopic sample Hamiltonian: only semiquantitative knowledge of the correlation lengths of the relevant degrees of freedom and the timescales of the characteristic dynamics is required.

Chronic rhinosinusitis (CRS) is thought to be a multifactorial disease that includes a direct involvement of bacteria that trigger inflammation and contribute to CRS pathogenesis. Staphylococcus aureus infection and persistence is associated with chronic rhinosinusitis (CRS), and it may be particularly relevant in the form with nasal polyps (CRSwNP). The large array of exotoxins deployed by S. aureus is instrumental for the bacterium to warrant its infection and dissemination in different human body districts. Here, we analyze the common Th2 environment in CRSwNP and prospect a possible dynamic role played by S. aureus leukocidins in promoting this chronic inflammation, considering leukocidin ED (LukED) as a strong prototype candidate worth of therapeutic investigation. CCR5 is an essential target for LukED to exert its cytotoxicity towards T cells, macrophages and dendritic cells. Therefore, CCR5 blockade might be an interesting therapeutic option for CRS and, more specifically, persistent and relapsing CRSwNP. In this perspective, the arsenal of CCR5 antagonists being developed to inhibit HIV-1 entry (CCR5 being the major HIV-1 co-receptor) could be easily repurposed for CRS therapeutic investigation. Finally, direct targeting of LukED by neutralizing antibodies could represent an important additional solution to S. aureus infection.

CCR5 and its interaction with chemokine ligands have been crucial for understanding and tackling HIV-1 entry into target cells. However, over time, CCR5 has witnessed an impressive transition from being considered rather unimportant in physiology and pathology to becoming central in a growing number of pathophysiological conditions. It now turns out that the massive efforts devoted to combat HIV-1 entry by interfering with CCR5, and the subsequent production of chemokine ligand variants, small chemical compounds, and other molecular entities and strategies, may set the therapeutic standards for a wealth of different pathologies. Expressed on various cell types, CCR5 plays a vital role in the inflammatory response by directing cells to sites of inflammation. Aside HIV-1, CCR5 has been implicated in other infectious diseases and non-infectious diseases such as cancer, atherosclerosis, and inflammatory bowel disease. Individuals carrying the CCR5Δ32 mutation live a normal life and are warranted a natural barrier to HIV-1 infection. Therefore, CCR5 antagonism and gene-edited knockout of the receptor gained growing interest for the therapeutic role that CCR5 blockade may play in the attenuation of the severity or progression of numerous diseases.

The low-power wide-area network (LPWAN) paradigm is gradually gaining market acceptance. In particular, three prominent LPWAN technologies are emerging at the moment: LoRaWAN™ and SigFox™, which operate on unlicensed frequency bands, and NB-IoT, operating on licensed frequency bands. This paper deals with LoRaWAN™, and has the aim of describing a particularly interesting feature provided by the latest LoRaWAN™ specification—often neglected in the literature—i.e., the roaming capability between different operators of LoRaWAN™ networks, across the same country or even different countries. Recalling that LoRaWAN™ devices do not have a subscriber identification module (SIM) like cellular network terminals, at a first glance the implementation of roaming in LoRaWAN™ networks could seem intricate. The contribution of this paper consists in explaining the principles behind the implementation of a global LoRaWAN network, with particular focus on how to cope with the lack of the SIM in the architecture and how to realize roaming.

IgE is a highly elusive antibody class, yet a tremendously powerful elicitor of immune reactions. Despite huge efforts spent on the characterization and understanding of the IgE system many questions remain either unanswered or only marginally addressed. One above all relates to the role of IgE. A common doubt is based on whether IgE mode of action should only be relegated to anti-parasite immunity and allergic manifestations. In search for a hidden role of IgE, reports from several laboratories are described herein in which a natural IgE link to cancer or the experimental redirection of IgE against cancer have been investigated. Epidemiological and investigational studies are trying to elucidate a possible direct intervention of endogenous IgE against cancer, raising thus far no definitive evidence. Conversely, experimental approaches implementing several strategies and engineered IgE formats built up a series of convincing results indicating that cancer might be tackled by the effector functions of this immunoglobulin class. Because of its peculiar immune features, IgE may present a superior anti-tumor performance as compared to IgG. However, extreme care should be taken on how IgE-based anti-tumor approaches should be devised. Overall, IgE appears as a promising resource, likely destined to enrich the anti-cancer arsenal.

Objectives: To investigate the effects of SARS-CoV-2 infection, as well as short- (within 48 hours) and long-term (within 30 days) adverse events (AEs) of SARSCoV- 2 vaccines, including arthritis flares in a large cohort of patients with inflammatory arthritis (IA). Methods: A retrospective cohort study comprising 362 patients: 94 (26%) rheumatoid arthritis, 158 (43.6%) psoriatic arthritis and 110 (30.4%) ankylosing spondylitis; and 165 healthy controls (HC) to ascertain the prevalence and severity of SARS-CoV-2 infection in patients with IA, the rate of AEs associated with SARS-CoV-2 vaccines and disease flares within a month of the vaccination. All patients provided informed consent and data about SARS-CoV-2 infection and/or vaccination status. Results: One-hundred-seventeen (32.3%) patients and 39 (23.6%) HC were affected by SARS-CoV-2 infection. Forty (34.2%) patients experienced an IA flare within one month of infection, of whom 3 (7.5%) needed to switch therapy. The prevalence of SARS-CoV-2 infection, disease severity, and hospitalization rate were not significantly different. At least one shot of SARSCoV- 2 vaccine was administered in 331 (91.4%) patients and 147 (89.1%) HC. Within 48 hours, 102 (30.8%) patients developed vaccine-related AEs; 52 (15.7%) patients with >1 vaccine dose experienced an IA flare-up, of whom 12 (23.1%) needed to switch therapy. Conclusions: A significantly higher rate of IA flare was observed among patients who contracted SARS-CoV-2 infection vs. those without infection. Patients with IA experienced flares after SARS-CoV-2 vaccination, though it was not statistically significant. [ABSTRACT FROM AUTHOR]
