本研究では、生体適合性を有するゼラチンに放射線を照射することで、すい臓内に侵入することができるゼラチンナノ粒子を作製し、PETイメージングに有用な銅（Cu-64）を担持させることにより、すい臓がんの診断・治療に向けた新規腫瘍追跡型薬剤の開発を目的とした。その結果、0.1 wt.％ゼラチン水溶液にCo-60 γ線を1 kGy/h、25℃、空気飽和条件で0.5 kGy照射することにより、5 nmの平均粒径を有する放射線架橋ゼラチンナノ粒子の作製に成功した。さらに、ゼラチンナノ粒子へのCuの吸着を調べるため、放射性のCu-64の代わりに非放射性のCu-63を用いて担持能をICP-MSにより評価したところ、2.7 µ mol/Lの銅イオンがゼラチンナノ粒子に担持していることが分かった。, 令和2年度 日本化学会関東支部群馬地区研究交流発表会

医療・創薬分野において、放射線架橋ゼラチンゲルを用いたドラッグデリバリーシステムの研究開発が進展している。一方で、放射線架橋ゼラチンゲルの生成メカニズムは不明であり、ゲルの物性（膨潤度、弾性率、粒径など）を精密に制御するための高度な放射線改質技術が確立されていない。本研究では、ゼラチンの放射線架橋に関与すると考えられているフェニルアラニン（Phe）、チロシン（Tyr）、ヒスチジン（His）1)を主成分としたペプチドを設計・有機合成し、その反応解析からゼラチンの放射線架橋メカニズムの解明を目的とした。Pheを競争標準試薬とした競合反応により、各種ペプチドと水の放射線分解で生成するヒドロキシル（OH）ラジカルの反応速度定数を決定した。Pheを含むペプチドの反応速度定数が6.95×109 L/mol/sと最も大きかったことから、Phe残基がゼラチンの放射線架橋に最も寄与していると結論づけた。, 令和2年度 日本化学会関東支部群馬地区研究交流発表会

MRI (Magnetic Resonance Imaging) contrast reagents are generally constructed of gadolinium ions complexed with linear or cyclic organic ligands for higher contrast effect in the clinical use. However, these gadolinium-based contrast agents (GBCAs) for MRI with low molecular weight invade in the patient brain through blood brain barrier and blood cerebrospinal fluid barrier. The deposition of GBCAs with linear ligands to the brain have been reported by recent researches1, and U.S. Food and Drug Administration has placed a warning on all of them. The contrast agents having biocompatibility and rapid renal excretion without passing through the blood brain barrier and blood cerebrospinal fluid barrier have been expected for the general diagnosis. It is necessary to design a contrast agent from 1 to 20 nm in diameter for safer diagnostics. Background Evidence of eribulin therapy for metastatic breast cancer (MBC) in clinical practice is not well documented. Methods We retrospectively analyzed the safety and efficacy of eribulin in 29 MBC patients from 2011 to 2016 at Fukuoka University Hospital. Results The median patient age, number of courses, total dose, and relative dose intensity were as follows: 65 years, five courses, 8.6 mg/m2, and 75%, respectively. One patient achieved a complete response, (CR) six a partial response (PR), eight stable disease (SD) and 14 patients exhibited progressive disease. The objective response rate (ORR: CR + PR) was 24.1%, and the clinical benefit rate (CBR: CR + PR + SD) was 51.7%. The median progression‐free survival was 90 days (95% confidence interval [CI] 67–126) and median overall survival was 264 days (95% CI 198–357). In patients who previously received 2–4 regimens, the ORR was 28.5% and the CBR was 57.1%. In patients who received 5–12 regimens, the ORR was 20% and the CBR was 45%. Chemotherapy was administered to 20 patients (69%) after eribulin administration, and the median overall survival rate of cases that achieved greater than a PR was 1088 days. The most frequent treatment‐related grade 3/4 adverse events were neutropenia (55.2%), and febrile neutropenia (20.1%). Grade 3 peripheral neuropathy occurred in 13.8% of patients, but was not exacerbated even if present before treatment. Conclusion Eribulin is effective for MBC patients who have received multiple chemotherapies. Neutropenia and febrile neutropenia may develop after heavy prior therapy.
