Implantable and wearable bioelectronic systems can enable tailored therapies for the effective management of long-term diseases, thus minimising the risk of associated complications. In this context, glucose fuel cells hold great promise as in- or on-body energy harvesters for ultra-low-power bioelectronics and as self-powered glucose sensors. We report here the generation of gold nanostructures through a gold electrodeposition method in a soft template for the abiotic electrocatalysis of glucose in glucose fuel cells. Two different types of soft template were used: a lipid cubic phase-based soft template composed of Phytantriol and Brij®-56, and an emulsion-based soft template composed of hexane and sodium dodecyl sulphate (SDS). The resulting gold structures were first characterised by SAXS, SEM and TEM to elucidate their structure, and then their electrocatalytic activity towards glucose was compared in both a three-electrode set-up and in a fuel cell set-up. The Phytantriol/Brij®-56 template led to a nanofeather-like Au structure, while the hexane/SDS template led to a nanocoral-like Au structure. These templated electrodes exhibited similar electrochemical active surface areas (0.446 cm2 with a roughness factor (RF) of 14.2 for Phytantriol/Brij®-56 templated nanostructures and 0.421 cm2 with an RF of 13.4 for hexane/SDS templated nanostructures), and a sensitivity towards glucose of over 7 μA mM−1 cm−2. When tested as the anode of an abiotic glucose fuel cell (in a phosphate-buffered solution with a glucose concentration of 6 mM), a maximum power density of 7 μW cm−2 was reached; however the current density in the case of the fuel cell with the Phytantriol/Brij®-56 templated anode was approximately two times higher, reaching the value of 70 μA cm−2. Overall, this study demonstrates two simple, cost-effective and efficient strategies to manipulate the morphology of gold nanostructures, and thus their catalytic property, paving the way for the successful manufacturing of functional abiotic glucose fuel cells.

ABSTRACT Multiple processes exist in a cell to ensure continuous production of essential proteins either through cap-dependent or cap-independent translation processes. Viruses depend on the host translation machinery for viral protein synthesis. Therefore, viruses have evolved clever strategies to use the host translation machinery. Earlier studies have shown that genotype 1 hepatitis E virus (g1-HEV) uses both cap-dependent and cap-independent translation machineries for its translation and proliferation. Cap-independent translation in g1-HEV is driven by an 87-nucleotide-long RNA element that acts as a noncanonical, internal ribosome entry site-like (IRESl) element. Here, we have identified the RNA-protein interactome of the HEV IRESl element and characterized the functional significance of some of its components. Our study identifies the association of HEV IRESl with several host ribosomal proteins, demonstrates indispensable roles of ribosomal protein RPL5 and DHX9 (RNA helicase A) in mediating HEV IRESl activity, and establishes the latter as a bona fide internal translation initiation site. IMPORTANCE Protein synthesis is a fundamental process for survival and proliferation of all living organisms. The majority of cellular proteins are produced through cap-dependent translation. Cells also use a variety of cap-independent translation processes to synthesize essential proteins during stress. Viruses depend on the host cell translation machinery to synthesize their own proteins. Hepatitis E virus (HEV) is a major cause of hepatitis worldwide and has a capped positive-strand RNA genome. Viral nonstructural and structural proteins are synthesized through a cap-dependent translation process. An earlier study from our laboratory reported the presence of a fourth open reading frame (ORF) in genotype 1 HEV, which produces the ORF4 protein using a cap-independent internal ribosome entry site-like (IRESl) element. In the current study, we identified the host proteins that associate with the HEV-IRESl RNA and generated the RNA-protein interactome. Through a variety of experimental approaches, our data prove that HEV-IRESl is a bona fide internal translation initiation site.

Abstract Nutrient surplus and consequent free fatty acid accumulation in the liver cause hepatosteatosis. The exposure of free fatty acids to cultured hepatocyte and hepatocellular carcinoma cell lines induces cellular stress, organelle adaptation, and subsequent cell death. Despite many studies, the mechanism associated with lipotoxicity and subsequent cell death still remains poorly understood. Here, we have used the proteomics approach to circumvent the mechanism for lipotoxicity using hepatocellular carcinoma cells as a model. Our quantitative proteomics data revealed that ectopic lipids accumulation in cells severely affects the ubiquitin-proteasomal system. The palmitic acid (PA) partially lowered the expression of deubiquitinating enzyme USP7 which subsequently destabilizes p53 and promotes mitotic entry of cells. Our global phosphoproteomics analysis also provides strong evidence of an altered cell cycle checkpoint proteins’ expression that abrogates early G2/M checkpoints recovery with damaged DNA and induced mitotic catastrophe leading to hepatocyte death. We observe that palmitic acid prefers apoptosis-inducing factor (AIF) mediated cell death by depolarizing mitochondria and translocating AIF to the nucleus. In summary, the present study provides evidence of PA-induced hepatocellular death mediated by deubiquitinase USP7 downregulation and subsequent mitotic catastrophe.

ABSTRACT: Among all fuel cells, proton exchange membrane fuel cells (PEMFCs) are appealing energy conversion devices in portable and transportation applications because of their higher efficiency, excellent ability to reduce air pollution, lightweight, generation of less noise due to the absence of mechanical components, quick start-up time and non-involvement in the combustion process. This review article emphasizes the importance of perfluorosulfonic acid (PFSA) polymer-based proton exchange membranes (PEMs) for fuel cell applications and their recent achievements, difficulties, and prospects. The most widely used PEMs for low-temperature PEMFCs are PFSA polymer-based membranes and some non-fluorinated polymers such as sulfonated poly(ether ketone) (SPEEK), sulfonated polysulfone (SPSF), sulfonated polyimide (SPI), and sulfonated polystyrene (SPS). Among these PEMs, PFSA membranes (such as Nafion) are the most often utilized low-temperature PEM for fuel cell applications because of their outstanding chemical, mechanical, and thermal stability with higher proton conductivity under-hydrated conditions. Due to some limitations of PFSA membranes, such as poor conductivity in anhydrous conditions, high manufacturing costs, and degradation of their properties at higher temperatures, research on low-temperature PEM has been shifted to the development of polymer electrolyte composite membranes incorporating various multifunctional organic, inorganic, and hybrid fillers. The PFSA polymer composite membranes show better chemical, thermal, and oxidative stability with higher proton conductivity and fuel cell performance than pristine PFSA membranes. In addition, the PFSA polymer composite membranes maintain proton conductivity and fuel cell performance in low humidity conditions. These findings show that developing suitable PFSA polymer composite membranes could enhance the possibility of commercial applications of PEMFCs.

Abstract Global dysregulation of RNA splicing and imbalanced sphingolipid metabolism has emerged as promoters of cancer cell transformation. Here, we present specific signature of alternative splicing (AS) events of sphingolipid genes for each breast cancer subtype from the TCGA-BRCA dataset. We show that ceramide synthase 2 (CERS2) undergoes a unique cassette exon event specifically in Luminal B subtype tumors. We validated this exon 8 skipping event in Luminal B cancer cells compared to normal epithelial cells, and in patient-derived tumor tissues compared to matched normal tissues. Differential AS-based survival analysis shows that this AS event of CERS2 is a poor prognostic factor for Luminal B patients. As Exon 8 corresponds to catalytic Lag1p domain, overexpression of AS transcript of CERS2 in Luminal B cancer cells leads to a reduction in the level of very-long-chain ceramides compared to overexpression of protein-coding (PC) transcript of CERS2. We further demonstrate that this AS event-mediated decrease of very-long-chain ceramides leads to enhanced cancer cell proliferation and migration. Therefore, our results show subtype-specific AS of sphingolipid genes as a regulatory mechanism that deregulates sphingolipids like ceramides in breast tumors, and can be explored further as a suitable therapeutic target.

Employing epoxy polymer and pristine and aromatic amine and hydroxyl functionalized SWCNTs in poly(ether ether ketone) (PEEK), novel polymer nanocomposite models were created. Molecular dynamics (MD) simulations were used to explore the interfacial, mechanical, and structural features of single-walled carbon nanotube (SWCNT) reinforced PEEK/epoxy matrix (BM) systems. We used MD simulation techniques to show how covalently functionalizing SWCNTs and blending PEEK with a polymer containing hydroxy‑functional groups can improve the interfacial and mechanical properties of PEEK nanocomposites. This study showed that –CH2C6H4NH2, –CH2C6H4OH group grafted SWCNT improve the interfacial and mechanical properties of PEEK-based nanocomposites due to the formation of hydrogen bonds between the filler materials and the polymer matrix, as well as better filler dispersion into the polymer matrix. The improvement of properties of polymer nanocomposites depends on the extent of the interaction of nanoparticles with the polymer matrix. Aromatic amine and hydroxyl (-CH2C6H4NH2, and -CH2C6H4OH) functionalization of SWCNTs is required for their high-performance engineering applications. Because epoxy polymer has a great ability to form hydrogen bonds with fictionalized SWCNT, adding 10% epoxy polymer to the BM increased the interfacial strength, elastic modulus, and shear modulus of SWCNT composites. This study provides a novel technique to improve the properties of polymer nanocomposites by covalently functionalizing SWCNTs.

Abstract Purpose The study aims to search for potent cellulase producer from the gut of Gryllotalpa africana as well as to characterize and determine the effect of the purified enzyme on the cellulosic waste materials. Methods The potent cellulolytic strain was identified through morphological, biochemical, physiological, and molecular characterization like 16S rRNA and fatty acid methyl ester profile. After the optimization of cellulase production, the enzyme was purified through DEAE-Sepharose column chromatographic separation. The molecular weight of the purified enzyme was determined by SDS-PAGE analysis. The purified enzyme was characterized in terms of its activity and its effect on cotton fiber, and sawdust was also studied. Result The selected potent strain GAC 16.2 was identified as Acinetobacter junii that was capable to produce enhanced cellulase (112.38 ± 0.87 U/ml) at standardized optimum fermentation conditions. The molecular weight of the purified enzyme was determined as 55 kDa. The utmost activity of the purified enzyme was detected pH 7.0, temperature 50 °C, and in the presence of metal ions like Mg2+ and Mn2+. The substantive degradation of cotton fiber and sawdust has been observed in a reasonably short period. Conclusion Purified cellulase from the selected isolate A. junii GAC16.2, a gut isolate of G. africana, has the potentiality to degrade cellulosic substances. This property can make the isolate a potent candidature for industrial application, as well as an effective biotechnological tool for environmental monitoring through cellulosic waste management.

In agricultural soil, cadmium (Cd) pollution compromises soil health, reduces crop yield, and produces Cd-contaminated crops. Bio-based approaches are necessary as an eco-friendly and sustainable solution to mitigate Cd-polluted areas. A heavy metal-resistant rhizobacterial strain (AS10) has been isolated from a heavy metal-defiled rice field. The 16S rDNA sequence and MALDI-TOF MS analyses of ribosomal protein reveal its identity closely similar to Enterobacter cloacae. The strain was found to withstand up to 4000 μg/ml Cd2+, 3312 µg/ml Pb2+ and 1500 µg/ml As3+. The Cd2+ removal efficiency was recorded as high as 72.11% when grown in 4000 μg/ml Cd2+. The strain's Cd-accumulation efficiency was also apprehended by TEM-EDAX followed by XRD-XRF-FTIR analyses. Besides, the strain showed solubilization of inorganic phosphate, ACC deaminase activity, nitrogen fixation and IAA production ability. Added further, the strain, as an efficient bioinoculant, significantly improved rice plant growth at the seedling stage through Cd immobilization. It prevented the surge of stress ethylene and oxidative stress in rice seedlings, resulting in overall plant growth improvement. Hence, the strain AS10 as potent plant growth-promoting rhizobacteria (PGPR) may be beneficial, especially in heavy metal-contaminated crop fields.

Bone tissue engineering (BTE) is a promising alternative to repair bone defects using biomaterial scaffolds, cells, and growth factors to attain satisfactory outcomes. This review targets the fabrication of bone scaffolds, such as the conventional and electrohydrodynamic techniques, for the treatment of bone defects as an alternative to autograft, allograft, and xenograft sources. Additionally, the modern approaches to fabricating bone constructs by additive manufacturing, injection molding, microsphere-based sintering, and 4D printing techniques, providing a favorable environment for bone regeneration, function, and viability, are thoroughly discussed. The polymers used, fabrication methods, advantages, and limitations in bone tissue engineering application are also emphasized. This review also provides a future outlook regarding the potential of BTE as well as its possibilities in clinical trials.

Replication of a positive-strand RNA virus involves an RNA-protein complex consisting of viral genomic RNA, host RNA(s), virus-encoded proteins, and host proteins. Dissecting out individual components of the replication complex will help decode the mechanism of viral replication. 5′ and 3′ UTRs in positive-strand RNA viruses play essential regulatory roles in virus replication.

Phosphorus is a primary macronutrient required for normal plant health, metabolism and survival. It is present in soil in compound insoluble form for which plant cannot uptake it directly from the soil. Some phosphate solubilizing bacteria possess some important enzymes for phosphate solubilization as well as mineralization. Alkaline (or basic) phosphatase (EC 3.1.3.1) is a type of zinc containing dimeric hydrolase enzyme responsible for removing the phosphate groups from various kinds of molecules including nucleotides, proteins, and alkaloids. Unlike acid phosphatases alkaline phosphatases are most effective in an alkaline environment. Alkaline phosphatases (ALPs) are of immense importance in various agricultural industries including dairy industries for testing successful pasteurization process. In this present study, Pseudomonas aeruginosa phosphatase was selected for a detailed computational investigation to exploit its physicochemical characteristics, structural properties including 3D model, model quality analysis, phylogenetic assessment and functional analysis using a number of available standard bioinformatics tools. The protein having average molecular weight about 51 kDa, was found thermostable and alkaline in nature belonging to metalloenzyme superfamily. Specifically, the thermostable behavior of the protein is suitable for the dairy industry. Moreover, this theoretical overview will help researchers to get an idea about the predicted protein structure and it may also help to design genetically engineered phosphate solubilizing bacteria by designing specific primers.
