Abstract To explore the clinical characteristics of patients infected with SARS-CoV-2 nationwide, especially the effect factors of asymptomatic infection and disappearance of clinical symptoms. A total of 66,448 COVID-19 patients in China who have been diagnosed by nucleic acid test or rapid antigen test were surveyed online (December 24, 2022 to January 16, 2023). Our cross-sectional study used descriptive analyses and binary Logistics regression model to assess the correlation between the clinical characteristics and relative factors, including age, gender, pre-existing conditions, reinfection, vaccination and treatment. A total of 64,515 valid questionnaires were collected. Among included participants, 5969 of which were asymptomatic. The symptoms were mainly upper respiratory symptoms, including dry and itchy throat (64.16%), sore throat (59.95%), hoarseness (57.90%), nasal congestion (53.39%). In binary Logistics regression model, we found that male, no pre-existing conditions, reinfection and vaccination have positive correlations with the appearance of asymptomatic COVID-19 patients. In Cox proportional-hazards regression model, considering all clinical symptoms disappeared in 14 days as outcome, we found that ≤ 60 years old, male, no pre-existing conditions, vaccination and adopted treatment have positive correlations with rapid amelioration of clinical symptoms in COVID-19 patients. The clinical symptoms of the participants were mainly upper respiratory symptoms which were according with the infection of Omicron variant. Factors including age, gender, pre-existing conditions and reinfection could influence the clinical characteristics and prognosis of COVID-19 patients. Importantly, vaccination has positive significance for the prevention and treatment of COVID-19. Lastly, the use of Chinese medicine maybe beneficial to COVID-19 patients, however, reasonable guidance is necessary.

Lung cancer has surpassed other types of cancer to become the primary cause of cancer-related deaths. Surgery stands as the foremost clinical treatment strategy available for tackling this condition, but it receives a low efficiency for most patients. In recent years, some adjuvant therapies are employed to improve the lung cancer treatment efficiency, such as chemotherapy, targeted therapy and immunotherapy. However, these strategies have not significantly increased overall survival of patients. Additionally, the random distribution of drugs will induce severe side effects. Nanomedicines have got great attentions to boost drug effect and reduce adverse reactions, including liposome-based nanoparticles, polymeric nanoparticles, inorganic nanoparticles, and exosomes. Importantly, nanomedicines contribute to improving drug bioavailability, stability and residency in target regions. Benefiting from the physiological characteristics of lung, the inhaled pulmonary delivery strategy in combination with nanomedicine will provide a non-invasive and effective strategy for treating lung cancer. Furthermore, the use of targeting ligands enables precise delivery of loaded drugs to lung cancer cells. Inhaled nanomedicine exhibits unique distribution and sustained release behaviors in the alveoli, amplifying the therapeutic effect and reducing side effects. This review aims to discuss various inhaled methods of delivering nanomedicine to treat lung cancer and also summarizes the clearance mechanism of nanomedicine in the lung. Overall, this review focuses on the application of different inhalable nanomedicines, which may inspire the development of more effective treatments against lung cancer.

Abstract Land-use change is a direct driver of biodiversity loss, projection and future land use change often consider a topical issue in response to climate change. Yet few studies have projected land-use changes over Africa, owing to large uncertainties. We project changes in land-use and land-use transfer under future climate for three specified time periods: 2021–2040, 2041–2060, and 2081–2100, and compares the performance of various scenarios using observational land-use data for the year 2020 and projected land-use under seven Shared Socioeconomic Pathways Scenarios (SSP): SSP1-1.9, SSP1-2.6, SSP2-4.5, SSP3-7.0, SSP4-3.4, SSP4-6.0 and SSP5-8.5 from 2015 to 2100 in Africa. Background: Cardiovascular diseases (CVDs) remain the leading cause of global mortality, with dyslipidemia recognized as a significant risk factor. Dyslipidemia, characterized by elevated levels of total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), and triglycerides (TG), is associated with an increased lifetime risk of CVD. Although alfalfa’s cholesterol-lowering effects have been demonstrated in animal models, clinical trial data supporting its efficacy in human dyslipidemia treatment are limited. Methods: A randomized controlled trial was conducted from September 20, 2021, to January 12, 2023, involving patients diagnosed with dyslipidemia. Approved by the China Ethics Committee of Registering Clinical Trials, the study included multiple sites in China. Eligible participants were adults aged 18 years or older, meeting criteria from the Chinese Guidelines for the Prevention and Treatment of Adult Blood Lipid Disorders. Participants were randomly assigned to either a health education-only group or a combined alfalfa powder supplementation and health education group for a three-month intervention period. Results: A total of 160 eligible patients were enrolled and randomized into two groups: the Alfalfa group (n = 80) and the Education group (n = 80). The intervention resulted in significant improvements in key lipid parameters in the Alfalfa group, including reductions in TC, LDL-C, and Apo A1 levels. Although changes in HDL-C levels were not significantly different between the groups, the Alfalfa group exhibited a larger increase. Serum lipid metabolomics analysis using liquid chromatography-tandem mass spectrometry (LC-MS/MS) indicated that the combined intervention did not significantly alter the overall blood lipid metabolome but identified fifteen regulated differential metabolites, with the glycerophospholipid metabolism pathway significantly affecting lipid metabolism levels. Conclusion: Supplementation with alfalfa powder combined with health education significantly reduced TC, LDL-C, and Apo A1 levels in patients with dyslipidemia. KEGG pathway analysis suggests an association between the glycerophospholipid metabolism pathway and dyslipidemia. Further research is warranted to validate these findings and explore the underlying mechanisms.

IntroductionMultiple factors, including human disturbance and environmental change, have caused a significant global mangrove resource loss. Therefore, the Chinese government has restructured mangrove habitats and restored the ecosystem functionality through establishing naturally protected mangrove areas. Despite efforts spanning several years, over 90% of mangrove forests in China have been now integrated into the natural protected area system, with the health status and influencing factors of mangrove ecosystem remaining a pertinent subject for further exploration. Given the intricacies of mangrove ecosystems with complex nonlinear relationships among evaluation factors, it is imperative to adopt scientific methodologies to assess their health status.MethodsTo illustrate this, we conducted a mangrove ecosystem health assessment of natural protected areas of Guangdong Province, China. By employing a fuzzy comprehensive evaluation method and establishing an index system for mangrove ecosystem health assessment.ResultsThe results revealed the following findings. (1) Various factors exert differing degrees of influence on mangrove ecosystem health. Notably, the mangrove habitat pattern (weight 47.95%), bird diversity (weight 20.97%), mangrove community (weight 14.31%), water environment (weight 11.76%), and soil sedimentary environment (weight 5.01%) were identified. (2) Overall, the mangrove ecosystem within protected areas of Guangdong exhibited unhealthy signs. There were 4 healthy protected areas (20.00%), 4 areas classified as sub-healthy (20.00%), and 12 were deemed unhealthy (60.00%). (3) Key factors contributing to the compromised health of mangrove ecosystems included the homogenization of mangrove plants, decreased habitat diversity, and exogenous pollution.DiscussionBy planting local mangrove species, scientifically managing the ratio of mangrove cover area to mudflat area, and controlling pollution sources and treating pollutants, the structure of the mangrove ecosystem can be optimized, and the quality of mangrove forest can be improved. These findings can enhance mangrove ecosystem management practices, assist mangrove natural reserve managers in performing targeted mangrove ecological protection and restoration, promote effective management, and contribute to the realization of “harmonious symbiosis” between humanity and nature within mangrove ecosystems.

Power flow is an extensively used tool in various operations and planning of power systems. In this paper, we propose an efficient hybrid method for solving the power flow problems in ill-conditioned power systems. This method is implemented in a framework of two-part alternating iteration. In Part I of this method, PV bus voltages are regarded as constant and PQ bus voltages are updated by the Z-bus Gauss method. In Part II of this method, PQ bus loads are equivalent to dynamic admittances, and PV bus voltage angles are updated by Newton’s method. The values of PQ bus voltage magnitudes and PV bus voltage angles are passed between Part I and Part II. The proposed method is validated in well- and ill-conditioned systems and compared with several well-known power flow methods. Results show that the proposed method is robust and efficient to address the issues related with large-scale ill-conditioned power systems and it is not significantly affected by the considered initial guess.

Abstract In sustaining arch locked-segment-type slopes, natural soil arches play a key anti-sliding role in the slope's evolution. In this study, a self-developed model test device was used to simulate the whole process of deformation evolution of sustaining arch locked-segment-type slopes, and the formation of natural sustaining arch and its locking control effect on slope stability were studied. The test results show that the continuous formation and progressive destruction of the sustaining arch were observed. The sustaining arch formed in the second time has the best locking effect, and the anti-sliding force reaches its stress peak point. However, the slope is not in a critically unstable state, instead, the stress is continuously adjusted to form a larger range of soil arch to resist the slope thrust. Consequently, the slope destabilizes until the ultimate shear strength of arch foots is exceeded, at which point the critical arch height of the arch is reached. The critical arch height mechanical model for slope stability analysis was developed based on the soil arching effect and limit equilibrium theory. The applicability of the model was demonstrated by the physical test and Xintan slope data, which can provide some guidance for early warning of landslides.

Abstract Candidalysin, a cytolytic peptide toxin secreted by the human fungal pathogen Candida albicans, is critical for fungal pathogenesis. Yet, its intracellular targets have not been extensively mapped. Here, we performed a high-throughput enhanced yeast two-hybrid (HT-eY2H) screen to map the interactome of all eight Ece1 peptides with their direct human protein targets and identified a list of potential interacting proteins, some of which were shared between the peptides. CCNH, a regulatory subunit of the CDK-activating kinase (CAK) complex involved in DNA damage repair, was identified as one of the host targets of candidalysin. Mechanistic studies revealed that candidalysin triggers a significantly increased double-strand DNA breaks (DSBs), as evidenced by the formation of γ-H2AX foci and colocalization of CCNH and γ-H2AX. Importantly, candidalysin binds directly to CCNH to activate CAK to inhibit DNA damage repair pathway. Loss of CCNH alleviates DSBs formation under candidalysin treatment. Depletion of candidalysin-encoding gene fails to induce DSBs and stimulates CCNH upregulation in a murine model of oropharyngeal candidiasis. Collectively, our study reveals that a secreted fungal toxin acts to hijack the canonical DNA damage repair pathway by targeting CCNH and to promote fungal infection.

Abstract Background About 25% of pregnant women experience bleeding in the early stage, and half of them eventually progress to pregnancy loss. Progesterone serves as a useful biomarker to predict miscarriage in threatened miscarriage, yet its performance is still debated. Abstract Aedes sp. mosquitoes are changing their geographic range in response to climate change. This is of concern because these mosquitoes can carry dengue fever and other viral diseases. Changing weather patterns can also increase the numbers of Aedes mosquitoes, leading to greater human exposure and enhancing population health risks. We project the geographic distribution of Aedes and associated changes in populations exposed to dengue in Asian metropolitan areas under warming scenarios from 1.5°C to 5.0°C above pre‐industrial temperatures, using multi‐model ensembles. With global warming, the southern part of the Arabian Peninsula, the coast of the Arabian Sea in southern Iran, southern Pakistan in West Asia, the Korean Peninsula, most of the Japanese islands, and parts of North China in East Asia are projected to become suitable for dengue transmission. The numbers of metropolitan areas exposed to dengue is projected to change from 142 (48%) in the reference period (1995–2014) to 211 (71%) at 5.0°C warming. With the combined impact of socioeconomic and climate change, population exposure to dengue in Asian metropolitan areas is projected to increase from 263 (multi‐model range 252–268) million in 1995–2014 to 411 (394–432) million, 446 (420–490) million, 509 (475–601), 558 (493–685) and 587 (529–773) million, respectively, at 1.5°C, 2.0°C, 3.0°C, 4.0°C and 5°C warming, with an average of 2.9 million new people exposed to dengue fever in metropolitan areas each year.

A growing number of clinical and animal studies suggest that the nucleus accumbens (NAc), especially the shell, is involved in the pathogenesis of temporal lobe epilepsy (TLE). However, the role of parvalbumin (PV) GABAergic neurons in the NAc shell involved in TLE is still unclear. In this study, we induced a spontaneous TLE model by intrahippocampal administration of kainic acid (KA), which generally induce acute seizures in first 2 h (acute phase) and then lead to spontaneous recurrent seizures after two months (chronic phase). We found that chemogenetic activation of NAc shell PV neurons could alleviate TLE seizures by reducing the number and period of focal seizures (FSs) and secondary generalized seizures (sGSs), while selective inhibition of PV exacerbated seizure activity. Ruby-virus mapping results identified that the hippocampus (ventral and dorsal) is one of the projection targets of NAc shell PV neurons. Chemogenetic activation of the NAc-Hip PV projection fibers can mitigate seizures while inhibition has no effect on seizure ictogenesis. In summary, our findings reveal that PV neurons in the NAc shell could modulate the seizures in TLE via a long-range NAc-Hip circuit. All of these results enriched the investigation between NAc and epilepsy, offering new targets for future epileptogenesis research and precision therapy.

Abstract Drought has a paramount impact on global agriculture and food security. However, the study on future cropland areas that can incur drought is inadequate. This paper uses input parameters from 7 CMIP6 models for 7 future scenarios (SSP1‐1.9, SSP1‐2.6, SSP4‐3.4, SSP2‐4.5, SSP4‐6.0, SSP3‐7.0, and SSP5‐8.5) to measure South Asian cropland exposure to drought and its underlying factors. Some defined epochs such as 2021–2040 (near‐term), 2041–2060 (mid‐term), 2081–2100 (long‐term), and 1995–2014 (reference period) are designed to explore diverse outlooks of the change. The Standardized Precipitation Evapotranspiration Index and the Run theory methods are applied to detect drought. Results indicate an intensified cropland (under SSP4‐3.4, SSP3‐7.0, and SSP5‐8.5) in the Indo‐Gangetic Plain region of South Asia, where mostly the variation occurs among scenarios and periods. Notably, the future cropland exposed to drought will increase in the 2021–2040, and 2041–2060 periods, but it intends to decline during the 2081–2100. Relatively, the exposed cropland will upturn highest by 49.2% (SSP3‐7.0) in the mid‐term period and decrease by −8.2% (SSP5‐8.5) in the end future. Spatially, distributed cropland in the central, south‐west, and portion of the northeast of South Asia are subjective to be exposed largely, but it can drop greatly across the eastern part by the end future. Importantly, the climate change effect plays a grounding role in future exposure change over the region during the near to mid‐term periods, while the cropland change effect is predominant in the long‐term perspectives. However, these findings signify the urgency of policymaking focusing on drought mitigation to ensure food security.

Traditional fermented foods are known to offer cardiovascular health benefits. However, the potential of fermented Chinese chives (FCC) in reducing coronary heart disease (CHD) remains unclear. This study employed anaerobic fermentation to investigate Lactiplantibacillus plantarum (L. plantarum) P470 from FCC. The results indicated that L. plantarum P470 enhanced hydroxyl radical scavenging and exhibited anti-inflammatory effects on RAW264.7 macrophages in the fecal fermentation supernatant of CHD patients. These effects were attributed to the modulation of gut microbiota and metabolites, including short-chain fatty acids (SCFAs). Specifically, L. plantarum P470 increased the abundance of Bacteroides and Lactobacillus while decreasing Escherichia-Shigella, Enterobacter, Veillonella, Eggerthella, and Helicobacter in CHD patient fecal samples. Furthermore, L. plantarum P470 regulated the biosynthesis of unsaturated fatty acids and linoleic acid metabolism. These findings suggest that L. plantarum P470 from FCC can improve the fecal physiological status in patients with CHD by modulating intestinal microbiota, promoting SCFA production, and regulating lipid metabolism.

Papain-like cysteine proteases (PLCPs) play pivotal roles in plant defense against pathogen invasions. While pathogens can secrete effectors to target and inhibit PLCP activities, the roles of PLCPs in plant-virus interactions and the mechanisms through which viruses neutralize PLCP activities remain largely uncharted. Here, we demonstrate that the expression and activity of a maize PLCP CCP1 (Corn Cysteine Protease), is upregulated following sugarcane mosaic virus (SCMV) infection. Transient silencing of CCP1 led to a reduction in PLCP activities, thereby promoting SCMV infection in maize. Furthermore, the knockdown of CCP1 resulted in diminished salicylic acid (SA) levels and suppressed expression of SA-responsive pathogenesis-related genes. This suggests that CCP1 plays a role in modulating the SA signaling pathway. Interestingly, NIa-Pro, the primary protease of SCMV, was found to interact with CCP1, subsequently inhibiting its protease activity. A specific motif within NIa-Pro termed the inhibitor motif was identified as essential for its interaction with CCP1 and the suppression of its activity. We have also discovered that the key amino acids responsible for the interaction between NIa-Pro and CCP1 are crucial for the virulence of SCMV. In conclusion, our findings offer compelling evidence that SCMV undermines maize defense mechanisms through the interaction of NIa-Pro with CCP1. Together, these findings shed a new light on the mechanism(s) controlling the arms races between virus and plant.

Abstract Here, we propose a green and sustainable 3D porous aerogel based on citrus peel (CP), chitosan (CS), and bentonite (BT). This aerogel is prepared through a simple sol–gel and freeze-drying process and is designed for efficient capture of Cu(II) ions from water matrices. CCBA-2, with its abundance of active binding sites, exhibits an impressive Cu(II) adsorption yield of 861.58 mg/g. The adsorption isotherm and kinetics follow the Freundlich and pseudo-second-order models, respectively. In the presence of coexisting mixed-metal ions, CCBA-2 demonstrates a significantly higher selectivity coefficient (Kd Cu = 1138.5) for removing Cu(II) ions compared to other toxic metal ions. Furthermore, the adsorption of Cu(II) ions by CCBA-2 is not significantly affected by coexisting cations/anions, ionic strength, organic matter, or different water matrices. Dynamic fixed-bed column experiments show that the adsorption capacity of Cu(II) ions reaches 377.4 mg/g, and the Yoon-Nelson model accurately describes the adsorption process and breakthrough curve. Through experiments, FTIR, and XPS analyses, we propose a reasonable binding mechanism between CCBA-2 and metal cations, involving electrostatic attraction and chemical chelation between Cu(II) and the functional groups of the aerogel. CCBA-2 saturated with Cu(II) ions can be successfully regenerated by elution with 1 M HNO3, with only a slight decrease in adsorption efficiency (5.3%) after 5 adsorption–desorption cycles. Therefore, CCBA-2 offers a cost-effective and environmentally friendly material that can be considered as a viable alternative for the green and efficient removal of toxic Cu(II) ions from wastewater.

Abstract Objective Anterior nucleus of thalamus (ANT) has been widely accepted as a potential therapeutic target for drug‐resistant epilepsy. Although increased volume of the ANT was also reported in patients with absence epilepsy, the relationship between the ANT and absence epilepsy has been barely illustrated. Methods Using chemogenetics, we evaluated the effect of ANT parvalbumin (PV) neurons on pentylenetetrazole (PTZ)‐induced absence seizures in mice. Results We found that intraperitoneal injection of PTZ (30 mg/kg) can stably induce absence‐like seizures characterized by bilaterally synchronous spike–wave discharges (SWDs). Selective activation of PV neurons in the ANT by chemogenetics could aggravate the severity of absence seizures, whereas selective inhibition of that cannot reverse this condition and even promote absence seizures as well. Moreover, chemogenetic inhibition of ANT PV neurons without administration of PTZ was also sufficient to generate SWDs. Analysis of background EEG showed that chemogenetic activation or inhibition of ANT PV neurons could both significantly increase the EEG power of delta oscillation in the frontal cortex, which might mediate the pro‐seizure effect of ANT PV neurons. Significance Our findings indicated that either activation or inhibition of ANT PV neurons might disturb the intrinsic delta rhythms in the cortex and worsen absence seizures, which highlighted the importance of maintaining the activity of ANT PV neurons in absence seizure.

It has been strongly suggested that selenium deficiency and T-2 toxin contamination have a strong relationship with the occurrence and development of Kashin–Beck disease (KBD). In order to provide information for understanding the high prevalence of KBD in Tibet, this study collected the responses to a cubital venous blood and dietary questionnaire of 125 subjects including 75 KBD patients and 50 healthy controls in a KBD-prevalent county (Luolong County) in Tibet, China. A total of 10 household local families were randomly selected in this area, and local diet samples of brick tea, Zanba powder, milk residue, and hulless Barley were collected from these residents. Selenium content in blood was detected by inductively coupled plasma mass spectrometry (ICP-MS). The T-2 toxin contamination level in food sample was assayed using an ELISA kit. The selenium levels of patients and controls were 42.0 ± 19.8 and 56.06 ± 22.4 μg/L, respectively. The serum selenium level in controls was higher than that in patients, but there was no significant difference, and the serum selenium level both in patients and controls in Tibet was lower than the normal range. The results of the dietary survey showed that the number of respondents who consumed butter tea was large; 46.67% of patients indicated that they drank buttered tea every day, which was significantly higher than in controls. The contents of T-2 toxin in Zanba powder, milk residue, hulless barley and drinking water samples were below the detection limit (0.05 μg/kg); this result was labeled Tr. Unexpectedly, the contents of T-2 toxin in brick tea were higher, with average levels of 424 ± 56 μg/kg in Detong village and 396 ± 24 μg/kg in Langcuo village. For the first time, we report the presence of an extremely high concentration of T-2 toxin in brick tea of Tibet.

Abstract Water-induced strength deterioration of rock mass is a crucial factor for rock slope instability. To better show the degradation process of rock slope water–rock interaction, we used bentonite as a water-sensitive regulator to build a new rock-like material that matches the features of water-induced strength degradation based on the cement-gypsum bonded materials. Twenty-five schemes of the material mixture proportion were designed using the orthogonal design method considering four factors with five variable levels, and a variety of experiments were conducted to obtain physico-mechanical parameters. In addition, one group of rock-like material proportion was selected and applied to the large-scale physical model test. The experiment results reveal that: (1) The failure mode of this rock-like material is highly similar to that of natural rock masses, and the physico-mechanical parameters vary over a wide range; (2) The bentonite content has a significant influence on the density, elastic modulus, and tensile strength of rock-like materials; (3) It is feasible to obtain the regression equation based on the linear regression analysis to determine the proportion of rock-like material; (4) Through application, the new rock-like material can effectively simulate or reveal the startup mechanism and instability characteristics of rock slopes under water-induced degradation. These studies can serve as a guide for the fabrication of rock-like material in the other model tests.

Abstract Background Circulating tumor cells (CTCs) are important biological indicators of the lung cancer prognosis, and CTC counting and typing may provide helpful biological information for the diagnosis and treatment of lung cancer. Methods The CTC count in blood before and after radiotherapy was detected by the CanPatrol™ CTC analysis system, and the CTC subtypes and the expression of hTERT before and after radiotherapy were detected by multiple in situ hybridization. The CTC count was calculated as the number of cells per 5 mL of blood. Results The CTC positivity rate in patients with tumors before radiotherapy was 98.44%. Epithelial–mesenchymal CTCs (EMCTCs) were more common in patients with lung adenocarcinoma and squamous carcinoma than in patients with small cell lung cancer (P = 0.027). The total CTCs (TCTCs), EMCTCs, and mesenchymal CTCs (MCTCs) counts were significantly higher in patients with TNM stage III and IV tumors (P 1 (P = 0.022 and P = 0.024, respectively). We report on the theoretical design and experimental verification of a high efficiency microwave wireless power transmission (MWPT) system operating in the Fresnel region. To achieve high conversion efficiency over a transmit distance of 11 meters, the transmit reflector antenna was optimized to locate the focal point at 11 m. The size of the receive antenna was decided by calculating the field distribution and the received power at different positions in the receive antenna aperture. Furthermore, an accurate model of the diode is presented, which was imported into the ADS software for high-precision rectifying circuit design. As a result, an overall DC-DC conversion efficiency of 20% was achieved, as measured in an anechoic chamber at a given distance of 11 m. The experimental results validated the proposed method.

Water Resourcing in China has historically been a complex issue requiring the ability to deal with regular floods, droughts and diverse water needs. Climate change represents another challenge to this sector, albeit one that is not traditionally considered by water managers. In this sector in China water management is predominantly based on historic, seasonal and annual forecast data while multi-annual and (multi-)decadal data are seldom used. In this paper, we present the co-development of a climate service prototype designed to provide water managers with insights into the impacts of climate change on the Upper Yellow River region for the next century. The paper is an outcome from our project that encouraged water resource planners and water resource managers to utilise long-term climate information to understand the uncertainties and the challenges our changing climate is likely to have in the region. Using an interdisciplinary team and adopting a user-centred, co-production approach, a prototype web-based data visualisation tool was developed. The development of the prototype was based on a design specification constructed from the findings of detailed interviews that allowed it to be developed and tested under SARS-CoV-2 pandemic restrictions that prevented the typical development process to be undertaken. The developed prototype presents climate information and communicates uncertainties regarding climate change in the remainder of the century through data sets that are typically used by the water sector in China in a simple, easy to understand style. Models that estimate river levels under different extraction scenarios and results about estimated river level and flow, and flood risk are also presented. The prototype was shown to be successful, as key messages relating to the impact of climate change and the challenges for water resource management could be effectively communicated through the tool interface. Practical implications: Understanding the impacts of climate change on water resourcing is complicated and multifaceted. There is a need for better data about what water there is and how it is moving around between and within catchments. Estimates of past, present and future climate variables along with historical measurements of river flow can be used to help visualise some of the uncertainties and changes that may happen in the next 50 years. In addition, there is a need to understand changing water demands and water resource management practices. Current water resource management practices are based on historical conditions and assumptions that are less likely to hold true in a more variable and warmer climate. Communicating how future changes will impact future water resourcing is critical to water resources in a changing climate (Belcher et al. 2018).This research outlines the construction of a tool to visualise the impacts of climate change on water availability in part of China that is typically water scarce, using models developed using the Soil Water Assessment Toolkit (SWAT). A model of the Upper Yellow River (UYR) was developed to demonstrate the impact of climate change on river levels in the catchment based on climate variables. The rainfall-runoff model was based on climate predictions from the CMIP5 assessment HadGEM3-GC3.05 climate model and incorporated information about water resource allocations for different administrative regions of the catchment The general climate trend for the region is that it is expected to become significantly warmer. The total amount of precipitation is likely to be about the same, and yet it is expected that overall, the catchment will become significantly drier over time as winter shortens and summer lengthens. The outputs from the model reflect the changes in climate variables. The uncertainties were communicated via a Web based tool. Water resource managers in China helped to coproduce the tool by participating in workshops and providing feedback on prototypes. The workshops helped scientists and water resource managers to communicate about climate change impacts on water resources and water resource management.

Microbes are critical for flavor formation in fermented foods; however, their mechanisms of action are not fully understood. The microbial composition of 51 dairy and 47 vegetable products was functionally annotated and the carbohydrate-active enzyme (CAZyme) profiles of Lactiplantibacillus plantarum 84-3 (Lp84-3), isolated from dairy samples, can promote resistant starch (RS) degradation, were analyzed. Lactobacillus, Streptococcus, and Lactococcus were the predominant genera in dairy products, whereas the major genera in vegetables were Lactobacillus, Weissella, and Carnimonas. Phages from Siphoviridae, Myoviridae, and Herelleviridae were also present in dairy products. Additionally, the glycosyl hydrolase (GHs) family members GH1 and GH13 and the glycosyltransferase (GTs) family members GT2 and GT4 were abundant in Lp84-3. Moreover, Lp84-3 was enriched in butanoate metabolism enzymes and butanoate metabolite compounds. Therefore, fermented food microbes, especially Lp84-3, have an abundant repertoire of enzymes that promote flavor production, as starter improving the flavor of fermented dairy and vegetable products.

Abstract As an internal forcing of the earth climate system, soil moisture (SM) significantly influences the water and energy cycle by controlling evapotranspiration and terrestrial solar energy. The current study used observed precipitation, remotely sensed SM, and reanalysis of atmosphere and land parameters to assess the East Asian Monsoon (EAM) precipitation variability due to meridional SM oscillations across China. A generalized linear method, namely coupled manifold technique (CMT) for assessing the reciprocal forcing between two climate fields and numerical simulations are applied to SM and EAM precipitation. We find that the EAM precipitation interannual variability between north and south China significantly correlates with SM meridional oscillation. The CMT results further showed that SM forcing has a significant (99% confidence) influence on the EAM precipitation explaining about 0.40 of the variance ratio in north and south China. The EAM and SM composite analysis show that the wetter (drier) north (south) oscillates the EAM precipitation over the north (south) of China and vice versa due to SM thermal controls. We then used control and sensitivity simulations with SM observations to further validate the findings implying that SM can potentially improve the interannual EAM forecast skills. The model results show that a wetter (drier) north (south) results in negative (positive) sensible heat (latent heat) anomalies that impact the boundary layer and propagate to change the meridional atmospheric heating profile. When positive (negative) SM anomalies exist over northern (southern) China, the zonal easterlies and extratropical westerlies move to north China causing above-normal precipitation that descends into southern China, suppressing subtropical westerlies and precipitation in southern China. On the contrary, a dry (wet) north (south) favors intensified subtropical westerlies and precipitation in southern China. The findings have dire implications for the water and energy cycle of the region in the projected wetting and drying patterns of the north (south).

Cracks in rock masses have adverse effects on their mechanical characteristics and the stability of rock mass engineering. For this experiment, uniaxial compression and true triaxial compression unloading tests were conducted on mortar specimens. The strength of the specimens was the lowest when the dip angle of the crack was 45°. Under uniaxial compression, the failure mode was mainly tensile failure. With an increase in the crack dip angle of the specimens, the types of cracks changed from shear to tensile cracks. Under true triaxial compression, the confining pressure considerably increased the strength and deformation characteristics of the specimens and weakened the influence of the crack dip angle on the strength. The failure mode when the crack dip angle was 15° was mainly tensile. The failure mode when the crack dip angles were 45° and 75° was mainly tensile–shear composite. With the increase in the crack dip angle, the failure degree of the specimens was more severe. Under the triaxial unloading confining, the failure mode was mainly shear. Compared with uniaxial compression, the failure degree of the specimens was more severe under true triaxial compression and true triaxial unloading. The innovation of this experiment is the use of a special mold to prepare specimens. We recommend that scholars conduct experimental and numerical simulation studies on different crack geometries.

Abstract At first glance, assessing future landslide‐exposed population appears to be a straightforward task if landslide hazard estimates, climate change, and population projections are available. However, the intersection of landslide hazard with socioeconomic elements may result in significant variation of estimated landslide exposure due to considerable variations in population projections. This study aims to investigate the effects of different sources of population data on the evaluation of landslide‐exposed population in China under four Shared Socioeconomic Pathways (SSPs) scenarios. We utilize multiple global climate models (GCMs) from Coupled Model Intercomparison Project Phase 6 and six high‐resolution spatially explicit static and dynamic population data sets to drive available landslide models. The results indicate an overall rise in landslide hazard projections, with an increase in the potential impact area of 0.4%–2.7% and an increase in the landslide frequency of 4.7%–20.1%, depending on the SSPs scenarios and future periods. However, the likely changes in future landslide exposed population, as modeled by incorporating population data from different sources with landslide hazard, yield divergent outcomes depending on the population data source. Thus, some of the projections depict an increase in future landslide exposure, while others show a clear decrease. The nationwide divergence ranged from −64% to +48%. These divergent findings were mainly attributed to differences in population data and a lesser extent to variations in GCMs. The present findings highlight the need to pay closer attention to the dynamic evolution of the elements at risk and the associated data uncertainties.

ABSTRACT Clostridioides difficile is one of the leading pathogens causing nosocomial infection. The infection can range from mild to severe, and rapid identification is pivotal for early clinical diagnosis and appropriate treatment. Here, a genetic testing platform for toxins, referred to as OC-MAB (orthogonal CRISPR system combined with multiple recombinase polymerase amplification [RPA]), was developed to detect the C. difficile toxin genes tcdA and tcdB. While recognizing the amplified products of the tcdA gene and the tcdB gene, Cas13a and Cas12a could activate their cleavage activities to cut labeled RNA and DNA probes, respectively. The cleaved products were subsequently identified by dual-channel fluorescence using a quantitative PCR (qPCR) instrument. Finally, they could also be combined with labeled antibodies on immunochromatographic test strips to achieve visual detection. The OC-MAB platform exhibited ultrahigh sensitivity in detecting the tcdA and tcdB genes at levels of as low as 102 to 101 copies/mL. When testing 72 clinical stool samples, the sensitivity (95% confidence interval [CI], 0.90, 1) and specificity (95% CI, 0.84, 1) of the single-tube method based on the fluorescence readout was 100%, with a positive predictive value (PPA) value of 100% (95% CI, 0.90, 1) and a negative predictive value (NPA) value of 100% (95% CI, 0.84, 1), compared to the results of qPCR. Likewise, the sensitivity of the 2-step method based on the test strip readout was 100% (95% CI, 0.90, 1), while the specificity was 96.3% (95% CI, 0.79, 0.99), with a PPA of 98% (95% CI, 0.87, 0.99) and an NPA of 100% (95% CI, 0.90, 1). In short, orthogonal CRISPR technology is a promising tool for the detection of C. difficile toxin genes. IMPORTANCE C. difficile is currently the primary causative agent of hospital-acquired antibiotic-induced diarrhea, and timely and accurate diagnosis is crucial for hospital-acquired infection control and epidemiological investigation. Here, a new method for the identification of C. difficile was developed based on the recently popular CRISPR technology, and an orthogonal CRISPR dual system was utilized for the simultaneous detection of toxin genes A and B. It also uses a currently rare CRISPR dual-target lateral flow strip with powerful color-changing capabilities, which is appropriate for point-of-care testing (POCT).

Interference has recently become a critical factor in communication satellite performance, and the interference source location is one of the most important factors in resolving this issue. The article proposes an innovative method of interference source direction-finding suitable for communication satellites with an existing single antenna beam and single radio frequency (RF) channel, which utilizes the symmetry of the antenna pattern to search for interference sources. Compared to traditional position methods with time-frequency-synchronized multi-satellites or a directing antenna array in a single satellite, the method does not require any particular direction-finding payload in communication satellites and shares existing antennas and RF channels with communication systems in satellites. The ability to find the direction of the interference source is a software-defined function in the communication processor. The proposed method provides a novel way to solve the problems of interference source direction-finding with the least engineering complexity, and it has excellent coexistence with other existing systems in communication satellites. The computer simulation and out-field experiment results in this article show that the method has excellent performance with high direction-finding resolution within extensive coverage, offering significant value and bright prospects for resolving the growing interference issues in communication satellites.

Vegetation slope protection has become more popular in recent years as an environmentally friendly support approach for slope protection projects. Thus, it's critical to study how to properly plant vegetation to maximize its slope protection effect. As a result, this work uses arbor as the research object and 3D printing technology to construct a model of the arbor root. The slope protection effect of arbor was studied by using a self-designed landslide model test system and conducting systematic physical model tests with three different root arrangement modes and three different root spacings. The results reveal that: ①With tree roots supporting the slope, the peak value of anti-sliding force increases, the time to reach the peak value of anti-sliding force is substantially delayed, and anti-sliding force attenuation is reduced. ②Anti-arch arrangement>positive arch arrangement>linear arrangement>no root support in terms of anti-sliding force improvement. S=1.5D (root coverage range D=7 cm) has a similar improved impact to S= 2.0D, while S=2.5D has the highest peak value of anti-sliding force. ③In terms of the root arrangement mode, the anti-arch arrangement has the most obvious improvement effect on the slope displacement, while the positive arch arrangement and linear arrangement are close in terms of the improvement effect on the displacement. In terms of the root spacing, S=2.5D has the most obvious improvement effect on the slope displacement, while S=1.5D and S=2.0D are close in terms of the improvement effect on the slope. ④Under the linear arrangement, the slope's sliding range expands, whereas the positive arch and anti-arch arrangements effectively limit slope deformation. Root spacing has no obvious effect on the slope deformation field.In conclusion, managing the arrangement mode of trees and root spacing in the vegetation protection project can increase the slope protection effect.

The response of soil organic carbon to climate warming may be soil depth-dependent, but remains unquantified in situ. Here the authors show that warming induces more proportional soil carbon losses in topsoil than in subsoil, particularly from high-latitudinal carbon-rich soils.

The transition metal iron plays a crucial role in living cells. However, high levels of iron are potentially toxic through the production of reactive oxygen species (ROS), serving as a deterrent to the commensal fungus Candida albicans for colonization in the iron-rich gastrointestinal tract. We observe that the mutant lacking an iron-responsive transcription factor Hap43 is hyper-fit for colonization in murine gut. We demonstrate that high iron specifically triggers multiple post-translational modifications and proteasomal degradation of Hap43, a vital process guaranteeing the precision of intestinal ROS detoxification. Reduced levels of Hap43 de-repress the expression of antioxidant genes and therefore alleviate the deleterious ROS derived from iron metabolism. Our data reveal that Hap43 functions as a negative regulator for oxidative stress adaptation of C. albicans to gut colonization and thereby provide a new insight into understanding the interplay between iron homeostasis and fungal commensalism.

In the context of global climate change and urban expansion, urban residents are encountering greater rainstorm waterlogging risk. Quantifying population exposure to rainstorms is an important component of rainstorm waterlogging risk assessments. This study utilized a two-dimensional hydrodynamic model to simulate the inundation water depth and inundation area resulting from rainstorms, with return periods of 5, 10, 50, and 100 years, in the Xiong’an New Area, and overlaid the gridded population data in 2017 and in 2035 under SSP2 to assess the change in population exposure. The results show that the average inundation depth and area increase were from 0.11 m and 207.9 km2 to 0.18 m and 667.2 km2 as the rainstorm return period increased from once in 5 years to once in 100 years. The greatest water depths in the main urban areas were mainly located in the low-lying areas along the Daqing River. The total population exposed to rainstorm waterlogging for the 5-, 10-, 50-, and 100-year return periods was 0.31, 0.37, 0.50, and 0.53 million, respectively, in 2017. However, this is projected to rise significantly by 2035 under SSP2, increasing 2–4-fold compared with that in 2017 for the four return periods. Specifically, the projected population exposure is expected to be 0.7, 1.0, 1.8, and 2.0 million, respectively. The longer the return period, the greater the increase in population exposure. The proportion of the population exposed at the 0.05–0.2 m water depth to the total population exposure decreases as the return periods increases, whereas the proportion changes in the opposite direction at the 0.2–0.6 m and >0.6 m depth intervals. Spatially, high-exposure areas are concentrated in densely populated main urban regions in the Xiong’an New Area. In the future, more attention should be paid to densely populated low-lying areas and extreme recurrence rainstorm events for urban flood-risk management to ensure population safety and sustainable urban development.

Background: Uterine myoma, which seriously threatens women’s physical and mental health and quality of life, is the most common benign pelvic tumor in gynecology. At present, laparoscopic myomectomy (LM) has become an effective and commonly surgical method in clinical practice. The aim of this study, using meta-analysis method, is to analyze the advantages and disadvantages of baseball suture comparing with traditional one, and provide a theoretical basis for the selection of surgical suture methods. Methods: CNKI, WanFang Data, VIP, PubMed, web of science and the Cochrane Library are searched to collect original data and cases. Two researchers independently screen the literature according to the inclusion and exclusion criteria, extract the data and cross check them. Then RevMan 5.4 and STATA 14 are used for statistical analysis. The difference is statistically significant (p < 0.05). Results: Compared with the traditional suture method, the operation time of LM with baseball suture method is shorter, the amount of intraoperative bleeding is less, the time required for postoperative exhaust, first getting out of bed, hospitalization, and uterine incision suture is shorter, the incidence of needle eye bleeding is lower, the decline of average hemoglobin is lower, and the incidence of pelvic infection is lower, while the difference is statistically significant (p < 0.05). The incidence of postoperative morbidity is lower, while the difference is not statistically significant (p > 0.05). Conclusions: Baseball suture has progressiveness and important value in LM surgical suture. The hemostasis and healing effect is ideal, which can enable patients to recover more quickly, reduce the pain and economic burden caused by the operation, and also enable surgeons to complete the operation more conveniently and efficiently. Candida auris is a multidrug-resistant human fungal pathogen responsible for nosocomial outbreaks worldwide. Here, the authors identify differential innate immune responses induced by C. auris and Candida albicans in vitro and in vivo, which correlate with structural features of the cell wall.

Abstract The tensile strength of loess is closely related to geological disasters. As eco-friendly materials, biopolymers have an excellent strengthening effect on the mechanical properties of soil. The effect of different initial dry densities and xanthan gum (XG) contents on the microstructure and mechanical behavior of XG-treated loess was studied with a series of microscopic tests and splitting tensile tests based on the particle image velocimetry technique. The results show that the XG became concentrated and agglomerated during dehydration, forming bridge links between soil particles and covering their surfaces. The XG-treated loess had a significant concentration of micropores and mesopores, with greater peak pore size distribution values than untreated loess. The specimens’ load–displacement curves with different XG contents and initial dry densities showed strain-softening. The displacement vector field indicated that specimens’ primary cracks were radial–vertical, and the secondary cracks were well-developed. The strain-softening phenomenon was more significant with increased XG content and initial dry density, and the specimens’ splitting tensile strength and brittleness increased. XG treatment gave the soils stronger cementation and a denser structure, helping to increase strength and brittleness. This research provides a scientific basis and practical experience for applying XG in geotechnical engineering.

Summary: Mosaic symptoms are commonly observed in virus-infected plants. However, the underlying mechanism by which viruses cause mosaic symptoms as well as the key regulator(s) involved in this process remain unclear. Here, we investigate maize dwarf mosaic disease caused by sugarcane mosaic virus (SCMV). We find that the manifestation of mosaic symptoms in SCMV-infected maize plants requires light illumination and is correlated with mitochondrial reactive oxidative species (mROS) accumulation. The transcriptomic and metabolomic analyses results together with the genetic and cytopathological evidence indicate that malate and malate circulation pathways play essential roles in promoting mosaic symptom development. Specifically, at the pre-symptomatic infection stage or infection front, SCMV infection elevates the enzymatic activity of pyruvate orthophosphate dikinase by decreasing the phosphorylation of threonine527 under light, resulting in malate overproduction and subsequent mROS accumulation. Our findings indicate that activated malate circulation contributes to the manifestation of light-dependent mosaic symptoms via mROS.

PurposeClostridioides difficile (C. difficile) infection is the most common cause of nosocomial infection, which is a severe challenge in modern medical care. Currently, many laboratory diagnostic methods for C. difficile are available, such as PCR, culture-based tests, and antigen-based tests. However, these methods are not suitable for rapid point-of-care testing (POCT). Therefore, it is of great significance to develop a rapid, sensitive, and cost-effective method to detect C. difficile toxin genes.MethodsRecently, the development of clustered regularly interspaced short palindromic repeats (CRISPR) technology has emerged as a promising tool for rapid POCT. In this study, we developed a rapid and specific detection platform for dual C. difficile toxins by combining recombinase polymerase amplification (RPA) and CRISPR/Cas12a.ResultsThe platform includes multiplex RPA-cas12a-fluorescence assay and multiplex RPA-cas12a-LFS (Lateral flow strip) assay, through which the detection limit for tcdA and tcdB was 10 copies/μL and 1 copy/μL, respectively. The results can be more clearly distinguished using a violet flashlight, which realized a portable visual readout. The platform can be tested within 50 min. Furthermore, our method did not cross-react with other pathogens that cause intestinal diarrhea. The results of 10 clinical samples using our method was 100% consistent with those from real-time PCR detection.ConclusionIn conclusion, the CRISPR-based double toxin gene detection platform for C. difficile is an effective, specific, and sensitive detection method, which can be used as a powerful on-site detection tool for POCT in the future.

Objective: functional gastroduodenal disease is the main type of functional gastrointestinal disease in the clinical department of Gastroenterology and psychosomatic medicine at present, which accounts for a large proportion of outpatients in gastroenterology. The main manifestations are epigastric pain, dyspepsia, belching, chronic nausea, and vomiting. The purpose of this study is to explore the changes in brain function in patients with functional gastroduodenal diseases through experiments to reveal the possible central etiology and development process. Methods: the functional changes of the prefrontal lobe in patients with functional gastroduodenal diseases and normal controls were detected and analyzed by near-infrared brain imaging. At the same time, SCL-90 was used to evaluate the mental health status of patients with functional gastroduodenal diseases and normal controls. The changes in the autonomic nerve system in patients and normal controls were detected and compared by heart rate variability trend chart. Results: the activity of left prefrontal lobe areas s8-d8, s10-d4, s10-d10 and s10-d15 in patients with functional gastroduodenal disease was significantly lower than normal controls (p 0.05). Conclusion: the function of the left frontal lobe is decreased in patients with functional gastroduodenal disease. The autonomic nervous system may be related to the connection system between the brain center and internal organs.

Although the complete nucleotide sequence of strawberry vein banding virus (SVBV) has been determined and bioinformatic analysis has revealed that the SVBV genome could encode seven proteins, the precise function of each protein is unclear. This study provided evidence that the P1 protein of SVBV (SVBV-P1) possesses the following features. Bioinformatic and subcellular localization analyses showed that SVBV-P1 is localized in the cytoplasm and cell walls of epidermal cells in Nicotiana benthamiana, and it forms inclusion bodies associated with microtubules and the endoplasmic reticulum. Dilution experiments demonstrated that SVBV-P1 could move from the original agro-infiltrated cells to adjacent cells in N. benthamiana leaves. Further trans-complementation experiments demonstrated that SVBV-P1 could facilitate the intercellular movement of a movement-deficient potato virus X mutant in N. benthamiana leaves. Finally, yeast two-hybrid and bimolecular fluorescence complementation assays revealed that SVBV-P1 could interact with the SVBV coat protein, which is a major component of Caulimovirus virions. Results of the electrophoretic mobility shift assay indicated that SVBV-P1 lacks DNA-binding capability. In summary, the results suggest that SVBV-P1 is probably a movement protein of SVBV, providing new insights into the function of movement proteins of the Caulimovirus genus.

Abstract Background Promoters are important factors affecting gene expression in cells. The driven activities of viral promoters were generally assessed to screen available promoters for transgenic and research and biotech industries. In this study, we cloned a full-length promoter from a Chinese isolate of strawberry vein banding virus (SVBV) and produced several deletion mutants for evaluation of applications in production of reporter proteins in stable transgenic plants. Methods The full-length promoter of SVBV (SP1) and its three deletion mutants (SP2, SP3, and SP4) were amplified using polymerase chain reaction. The effects of SVBV SP1, SP2, SP3, and SP4 on gene expression were evaluated using β-glucuronidase (GUS) and green fluorescent protein (GFP) reporter genes. Results Transient expression assays showed that the SVBV SP1 promoter and its three deletion mutants all expressed the reporter genes, albeit at very different levels. Interestingly, transcriptional activity driven by the SP1 promoter was much higher than that of the cauliflower mosaic virus (CaMV) 35S promoter. After stable transformation of the GUS gene into Nicotiana tabacum plants, SVBV SP1-driven transgene expression was approximately 2.6-fold higher than CaMV 35S promoter-driven transgene expression. In addition, GUS gene expression levels were enhanced by co-inoculation of the plants with the SP1 promoter-driven vector carrying the GUS gene and the vector expressing SVBV open reading frame (ORF) V or ORF VI. Conclusions The SVBV SP1 promoter from the Chinese isolate evaluated in this study could successfully drive transient and stable expression in plants, it was a stronger promoter than the CaMV 35S and FLt-US promoters and may be more useful for the production of stable transgenic plants.

Abstract Strawberry vein banding virus (SVBV) is a double-stranded DNA virus. In our previous studies, we generated an infectious clone of SVBV, pSVBV, which causes light-green vein banding symptoms along the leaf veins in strawberry plants (Fragaria vesca). In this study, we constructed pSVBV-P1-MCS and pSVBV-P4-MCS, two recombinant virus vectors containing a multiple cloning site (MCS) downstream of the SVBV-encoded movement protein gene (P1) and coat protein gene (P4), respectively. At 35 days post-inoculation, the two SVBV-based vectors could produce light-green vein banding symptoms on the systemic leaves of strawberry plants, indicating that they could successfully cause infection. Furthermore, the infectivity rates of the recombinant virus vectors pSVBV-P1-MCS and pSVBV-P4-MCS were similar to that of the wild-type infectious clone pSVBV, indicating that the insertion of MCS did not affect the infectivity of SVBV-based vectors. Additionally, we engineered SVBV as a transient overexpression vector, which could be used for the overexpression of exogenous green fluorescent protein in strawberry plants. Collectively, these SVBV-based vectors provide a new approach for the analysis of gene functions in strawberry plants.

Abstract In order to understand the effects of different patterns of prefabricated fractures and grain size compositions on the fracture characteristics, acoustic emission characteristics and mechanical properties of the rock masses. We conducted compression shear experiments on square rock masses with different modes of prefabricated fractures and different grain size compositions. The experimental results showed that five fracture patterns were produced in specimens with different fracture patterns. The first fracture specimen, the fourth fracture specimen and the fifth fracture specimen were all brittle fractures. The other four specimens were not brittle fractures. The fracture patterns, fracture processes and mechanical characteristics of the different fracture pattern rock masses were revealed. The lowest peak shear stresses were found in specimens consisting of two grain size ranges and the highest peak shear stresses were found in specimens consisting of three grain size ranges. The highest shear displacements corresponding to the peak shear stresses were found in the specimens consisting of three particle size ranges. The effect of different grain size compositions on the peak shear stress and its corresponding shear displacement of the rock mass was revealed. Specimens consisting of one grain size range produced significant fracture and acoustic emission prior to the peak shear stress. The acoustic emission was jumped after the main fracture was formed. The specimen consisting of two grain size ranges produced fractures and strong acoustic emission characteristics after the peak shear stress. Thereafter, fracture reappeared and the acoustic emission signature increased again. As the specimen entered the residual strength phase, the acoustic emission was jumpy. Specimens consisting of three grain size ranges were brittle fractures with weak acoustic emission characteristics after the main fracture has formed. The cumulative energy of shear acoustic emission was the highest for a rock mass consisting of three grain size ranges. The rock mass consisting of three grain size ranges was also the strongest and most difficult to fracture because the grains were more fully embedded in each other.

Summary Fungal infections have emerged as a major global threat to human health because of the increasing incidence and mortality rates every year. The emergence of drug resistance and limited arsenal of antifungal agents further aggravates the current situation resulting in a growing challenge in medical mycology. Here, we identified that ponatinib, an FDA‐approved antitumour drug, significantly enhanced the activity of the azole fluconazole, the most widely used antifungal drug. Further detailed investigation of ponatinib revealed that its combination with fluconazole displayed broad‐spectrum synergistic interactions against a variety of human fungal pathogens such as Candida albicans, Saccharomyces cerevisiae and Cryptococcus neoformans. Mechanistic insights into the mode of action unravelled that ponatinib reduced the efflux of fluconazole via Pdr5 and suppressed the expression of the proton pump, Pma1. Taken together, our study identifies ponatinib as a novel antifungal that enhances drug activity of fluconazole against diverse fungal pathogens.

In the electronic brake system (EBS) of commercial vehicles, due to the compressibility of gas, it is difficult to achieve accurate control in the pneumatic pipeline. To address this issue, a vertical load estimator based on unscented particle filtering (UPF) was designed, which can estimate vertical load during the running of the vehicle. Then, the EBS dynamics model was established based on software, including a brake signal sensor, single-channel bridge control module, ABS solenoid valve, and dual-channel bridge control module. Finally, based on the characteristics of the EBS valve, the control algorithm of the valve was studied, and the algorithm was tested using a hardware-in-the-loop experiment. The experiment results showed that the designed algorithm could improve braking performance.

Intestinal diseases caused by sleep deprivation (SD) are severe public health threats worldwide. However, whether or not probiotics attenuate the intestinal damage associated with SD remains unclear. In this study, we used antibiotic pretreatment and fecal microbiota transplantation to investigate the protective role of Lactiplantibacillus plantarum (L. plantarum) 124 against SD-related intestinal barrier damage in C57BL/6 mice. Compared with those of a normal sleeping mouse, we observed that intestinal antioxidant capacity and anti-inflammatory cytokine levels were decreased, while pro-inflammatory cytokines were increased in sleep deprivation mice with an increasing duration of sleep deprivation. This resulted in decreased tight junction protein expression and increased intestinal barrier permeability. In contrast, intragastric administration with L. plantarum 124 reversed SD-associated intestinal oxidative stress, inflammation, colonic barrier damage, and the dysbiosis of the microbiota in the colon. In addition, L. plantarum 124 restored gut microbiota homeostasis via restoring abundance, including that of Dubosiella, Faecalibaculum, Bacillus, Lachnoclostridium, and Bifidobacterium. Further studies showed that gut microbiota mediated SD-associated intestinal damage and the treatment L. plantarum 124 in SD-associated colonic barrier damage. L. plantarum 124 is a potential candidate for alleviating SD-associated intestinal barrier damage. Overall, L. plantarum 124 consumption attenuates intestinal oxidative stress, inflammation, and intestinal barrier damage in SD-associated mice via the modulation of gut microbes.

Due to their superior antibacterial properties, biocompatibility and high conductivity, nanomaterials have shown a broad prospect in the biomedical field and have been widely used in the prevention and treatment of oral diseases. Also due to their small particle sizes and biodegradability, nanomaterials can provide solutions for tissue engineering, especially for oral tissue rehabilitation and regeneration. At present, research on nanomaterials in the field of dentistry focuses on the biological effects of various types of nanomaterials on different oral diseases and tissue engineering applications. In the current review, we have summarized the biological effects of nanoparticles on oral diseases, their potential action mechanisms and influencing factors. We have focused on the opportunities and challenges to various nanomaterial therapy strategies, with specific emphasis on overcoming the challenges through the development of biocompatible and smart nanomaterials. This review will provide references for potential clinical applications of novel nanomaterials in the field of oral medicine for the prevention, diagnosis and treatment of oral diseases.

Bread was prepared using wheat flour with grape seed proanthocyanidin (GSP) (0.4%). GSP improved the textural properties of bread including hardness, cohesiviness, gumminess and chewiness. At the last stage of fermentation, GSP reinforced the gluten microstructure with increased the disulfide bonds and hydrophobic interaction and α-helix in the secondary structures. Moreover, GSP addition could increase the total phenolics and antioxidative acitivity of the bread significantly. In addition, the degree of fermentation had a strong influence on the dough forces, and the reasonable control of bread fermentation time was beneficial to improve the bread quality, which provided a reference for the bread processing industry.

The instability of perilous rock is mostly manifested as sudden collapse and failure without obvious displacement characteristics. Therefore, it is difficult to achieve the purpose of monitoring and early warning by conventional displacement monitoring. But the existing stability monitoring indicators are mostly deformation, stress, and strain. There is a problem that the stability evaluation parameters are inconsistent with the monitoring parameters. Taking sliding-type perilous rock as the research object, the structural plane is assumed to be homogeneous and isotropic, and linear elastic deformation in the amplitude range. Based on the dynamic theory and limit equilibrium model, the quantitative relationship model can be established involving safety factors, natural vibration frequency, structural surface bonding area, elastic modulus, and mass. The remote laser vibrometer is used to monitor the natural vibration frequency of the sliding-type perilous rock on the slope of the Huangzangsi Hydrojunction, and the stability evaluation of the perilous rock is achieved based on the quantitative relationship model between the safety factor and the natural vibration frequency. In this way, the frequency of slipping perilous rock stability evaluation and safety factor can be monitored. The results are basically the same with the safety factor calculated by the limit equilibrium method, indicating that the method is correct and feasible. The research has high theoretical significance and practical value for the safety monitoring and advanced warning of sliding perilous rock.
