Linear low-density polyethylene (PE) containing an inclusion complex composed of [beta]-cyclodextrin and D-limonene (CD-lim) is proposed as an innovative food wrapping material with antimicrobial features. PE/CD-lim composites are prepared by conventional, high-volume processing, and the optical and mechanical properties of the developed formulations appear suitable for their use as food packaging materials. Compared to plain PE, the PE/CD-lim films have improved haze and UV-vis outcomes, but also decreased gloss values. Addition of CD-lim slightly increases water uptake, up to levels suitable for the foreseen application. It also allows to improve performance of PE as food packaging material, since the PE/CD-lim composites provide barrier to ultraviolet light, an added value for packaging of fatty acids-containing food items, whose shelf life is often limited by oxidation caused by UV light. Most importantly, PE/CD-lim composites exhibit significant antimicrobial properties against a wide range of microorganisms, including fungi and bacteria. Hence, PE films containing CD-lim display suitable properties to play a role as active food packaging films. The pursuit of novel or modified substances based on a natural origin, like flavonoids, is essential in addressing the increasing number of diseases and bacterial resistance to antibiotics, as well as in maintaining intestinal balance and enhancing overall gut health. The primary goal of this research was to evaluate the impact of specific flavonoid compounds—chalcones, flavanones, and flavones—substituted with -Br, -Cl, -CH3, and -NO2 on both pathogenic and probiotic microorganisms. Additionally, this study aimed to understand these compounds' influence on standardized normal and pathologically altered intestinal microbiomes. 8-Bromo-6-chloroflavone 4′-O-β-D-(4″-O-methyl)-glucopyranoside and 8-bromo-6-chloroflavanone showed the most promising results as bactericidal agents. They significantly limited or inhibited the growth of pathogenic bacteria without adversely affecting the probiotic's growth. Digestion in vitro studies indicated that 6-methyl-8-nitroflavone and 8-bromo-6-chloroflavone positively modulated the gut microbiome by increasing beneficial bacteria and reducing potentially pathogenic microbes. This effect was most notable in microbiomes characteristic of older individuals and those recovering from chemotherapy or antibiotic treatments. The study assessed the health-promoting properties and the content of minerals in the bark of bird cherry (Prunus padus L.), which was then used as an ingredient in functional teas. The infusions were made with the use of Matricaria chamomilla L., Tilia cordata Mill., and Calendula officinalis L., and then combined with the bark in various proportions. The prepared infusions were tested for antioxidant activity, ability to reduce copper ions and iron ions, as well as the ability to scavenge hydroxyl radicals. In the next stage, the antimicrobial activity and the ability to inhibit the enzyme cycloxygenase-2 were assessed. Bird cherry bark contains a high potassium content of 19.457 ± 762 mg/kg d.m. In all the tests evaluating the antioxidant activity, infusions from the bark of bird cherry alone and with its 30% addition had the strongest properties. The analyzed infusions also have the ability to reduce Cu(ii) ions; they are active to reduce Fe(iii) ions and scavenge hydroxyl radical. The highest antimicrobial activity was found for teas with 20 and 30% bark, especially against Listeria monocytogenes (25.0–27.0 mm) (±3.0). The bark infusion was also found to have the highest inhibitory activity against cyclooxygenase-2 (COX-2) – 77.0%.

The search for new substances of natural origin, such as flavonoids, is necessary in the fight against the growing number of diseases and bacterial resistance to antibiotics. In our research, we wanted to check the influence of flavonoids with chlorine or bromine atoms and a nitro group on pathogenic and probiotic bacteria. We synthesized flavonoids using Claisen–Schmidt condensation and its modifications, and through biotransformation via entomopathogenic filamentous fungi, we obtained their glycoside derivatives. Biotransformation yielded two new flavonoid glycosides: 8-amino-6-chloroflavone 4′-O-β-D-(4″-O-methyl)-glucopyranoside and 6-bromo-8-nitroflavone 4′-O-β-D-(4″-O-methyl)-glucopyranoside. Subsequently, we checked the antimicrobial properties of the aforementioned aglycon flavonoid compounds against pathogenic and probiotic bacteria and yeast. Our studies revealed that flavones have superior inhibitory effects compared to chalcones and flavanones. Notably, 6-chloro-8-nitroflavone showed potent inhibitory activity against pathogenic bacteria. Conversely, flavanones 6-chloro-8-nitroflavanone and 6-bromo-8-nitroflavanone stimulated the growth of probiotic bacteria (Lactobacillus acidophilus and Pediococcus pentosaceus). The aim of the study was to compare the content of selected phytochemicals as well as the antioxidant and antimicrobial potential of the leaves of Prunus padus L. and Prunus serotina L., as there is very little research on this subject in the literature. Therefore, it is used to deepen knowledge on this subject. In addition, an electrochemical test was also carried out, which was not yet available for the above plants. Antibacterial studies have also been deepened to include the analysis of new strains of bacteria and fungi, which has not been studied earlier. The water extracts of P. padus using the utra-performance liquid chromatography (UPLC) system showed a higher content of both phenolic acids and flavonols (651.77b ± 18.12 mg/100 g dw for acids and 3.85b ± 0.08 mg/100 g dw for flavonols, respectively). Ferulic and p-coumaric acids were the dominant polyphenols in leaves. Extracts from P. padus showed higher activity against DPPH radical, which was 6.62b ± 0.06 mg TE/1 g dw, as well as higher antioxidant capacity, measured using 2,2′-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) cation radical (37.39b ± 3.81 mg TE/g dw). The higher antioxidant potential of P. padus was confirmed based on the oxidizing potentials of electroactive compounds present in them. Stronger inhibition against Enterococcus faecium and Klebsiella pneumoniae was found for P. padus, whereas P. serotina extract was more potent against Enterococcus faecium bacterium. It has been shown that P. padus can be an attractive raw material with antioxidant and antimicrobial properties that can be used on a much wider scale in food technology than its current application.

The compounds present in hemp show multidirectional biological activity. It is related to the presence of secondary metabolites, mainly cannabinoids, terpenes, and flavonoids, and the synergy of their biological activity. The aim of this study was to assess the activity of the Henola Cannabis sativae extract and its combinations with selected carriers (polyvinyl caprolactam–polyvinyl acetate–polyethylene glycol graft copolymer, magnesium aluminometasilicate, and hydroxypropyl-β-cyclodextrin) in terms of antimicrobial, probiotic, and immunobiological effects. As a result of the conducted research, the antimicrobial activity of the extract was confirmed in relation to the following microorganisms: Clostridium difficile, Listeria monocytogenes, Enterococcus faecalis, Staphylococcus aureus, Staphylococcus pyrogenes, Escherichia coli, Klebsiella pneumoniae, Salmonella typhimurium, Pseudomonas aereuginosa, and Candida albicans (microorganism count was reduced from ~102 CFU mL−1 to <10 CFU mL−1 in most cases). Additionally, for the system with hydroxypropyl-β-cyclodextrin, a significant probiotic potential against bacterial strains was established for strains Lactobacillus acidophilus, Lactobacillus casei, Lactobacillus plantarum, Lactobacillus brevis, Lactobacillus rhamnosus, Lactobacillus reuteri, Pediococcus pentosaceus, Lactococcus lactis, Lactobacillus fermentum, and Streptococcus thermophilus (microorganism count was increased from ~102 to 104–107). Pomegranate flower extract, rich in anthocyanins, demonstrates beneficial health-promoting properties such as an anti-diabetic and antioxidant effect, among others. However, the potential health-promoting properties may be hindered by the low stability of anthocyanins. Therefore, the aim of our study was to assess whether stabilizing carriers, namely HP-γ-cyclodextrin (HP-γ-CD), α-cyclodextrin (α-CD), Methyl-β-cyclodextrin (Me-β-CD), Inulin (Inu) and Arabic gum (AGu) affect the antioxidant and antidiabetic activity of lyophilized pomegranate flower extract, how they influence stability, release profile, and whether the systems exhibit prebiotic activity. Interactions between pomegranate flower extract and these factors were analyzed using FT-IR. The structures were examined through microscopic imaging while for the prepared prebiotic systems, antidiabetic activity was determined and confirmed by the inhibition of α-amylase and α-glucosidase; antioxidant activity was expressed by DPPH and CUPRAC assays. The content of pelargonidin-3,5-glucoside in these systems was assessed using the HPLC method. The release profiles of pelargonidin-3,5-glucoside were examined in a medium at pH = 6.8 and pH = 1.2, and the stability was assessed after subjecting the systems to high temperatures (T = 90 °C). The prebiotic potential was evaluated for 10 prebiotic bacterial strains (Lactobacillus acidophilus, Lactobacillus casei, Lactobacillus plantarum, Lactobacillus brevis Lactobacillus rhamnosus gg, Lactobacillus reuteri, Pediococcus pentosaceus, Lactococcus lactis, Lactobacillus fermentum lf, Streptococcus thermophilus). As a result of the conducted research, better functionalities of the obtained systems containing Pomegranate flower extract were proven in terms of prebiotic and antidiabetic effects. Centella asiatica extract is a valued plant material with known anti-inflammatory and anti-microbiological properties. Using the Design of Experiment (DoE) approach, it was possible to obtain an optimized water/alcoholic extract from Centella asiatica, which allowed the preparation of the final material with biological activity in the wound healing process. Studies on the novel applications of Centella asiatica in conjunction with the multifunctional chitosan carrier have been motivated by the plant's substantial pharmacological activity and the need to develop new and effective methods for the treatment of chronic wounds. The controlled release of asiaticoside was made possible by the use of chitosan as a carrier. Based on the findings of investigations using the PAMPA skin assay, which is a model imitating the permeability of actives through skin, this compound, characterized by sustained release from the chitosan delivery system, was identified as being well able to permeate biological membranes such as skin. Chitosan and the lyophilized extract of Centella asiatica worked synergistically to block hyaluronidase, exert efficient microbiological activity and take part in the wound healing process, as proven in an in vitro model. A formulation containing 3% extract with 3% medium-molecular-weight chitosan was indicated as a potentially new treatment with high compliance and effectiveness for patients. Optimization of the chitosan-based hydrogel preparation ensured the required rheological properties necessary for the release of the bioactive from the chitosan delivery system and demonstrated a satisfactory antimicrobial activity. Dermocosmetics are products intended for sensitive and problematic skin. They are a good solution for additional care of skin affected by conditions such as atopic dermatitis, acne, psoriasis, or for couperose skin. The compounds of particular importance in the care of atopic skin include ceramides, ectoine, hyaluronic acid, vitamins A and E, as well as natural vegetable oils. The aim of this work was to develop a qualitative and quantitative approach and to investigate the characteristics of formulations such as a face cream, topical cream, body lotion, and cleansing foam dedicated to patients with atopic dermatitis. The finished cosmetic formulation was evaluated by performing formulation stability tests at different times and temperatures, together with packaging compatibility and organoleptic evaluation. Preservative tests were carried out to determine the efficacy of the preservative. In addition, dermatological and instrumental tests were done to verify the product properties. The newly developed cosmetic formulations showed good physical and chemical characteristics, typical of this type of product. They were also structurally and microbiologically stable during storage at various temperatures. Dermatological tests did not reveal any irritating or allergenic properties. The best data were obtained for the face cream--namely improvement of skin hydration by 21.88%, reduction of transepidermal water loss by 15.86%, alleviation of erythema by 15.77%, and of skin sensitivity by 17.20%. This work resulted in the preparation of innovative dermocosmetics to be used for atopic skin. This study investigated the ability of PVP/HPβCD-based electrospun nanofibers to enhance the dissolution rate of poorly soluble polydatin and resveratrol, the main active components of Polygoni cuspidati extract. To make a solid unit dosage form that would be easier to administer, extract-loaded nanofibers were ground. SEM examination was used to analyze the nanostructure of the fibers, and the results of the cross-section of the tablets showed that they had maintained their fibrous structure. The release of the active compounds (polydatin and resveratrol) in the mucoadhesive tablets was complete and prolonged in time. Additionally, the possibility of staying on the mucosa for a prolonged time has also been proven for both tablets from PVP/HPβCD-based nanofibers and powder. The appropriate physicochemical properties of the tablets, along with the proven antioxidant, anti-inflammatory, and antibacterial properties of P. cuspidati extract, highlight the particular benefits of the mucoadhesive formulation for use as a drug delivery system for periodontal diseases. Hot Melt Extrusion (HME) technology was developed to obtain blends containing lyophilized Scutellariae baicalensis root extract and chitosan in order to improve the rheological properties of the obtained blends, including tableting and compressibility properties. (Hydroxypropyl)methyl cellulose (HPMC) in 3 different ratios was used as amorphous matrix formers. The systems were characterized using X-ray powder diffraction (PXRD), Fourier Transform Infrared Spectroscopy with Attenuated Total Reflectance (FTIR-ATR), and in vitro release, permeability, and microbiological activity studies. Then, the extrudates were used to prepare tablets in order to give them the appropriate pharmaceutical form. HPMC-based systems released baicalin more slowly, resulting in delayed peaks in the acceptor fluid. This behavior can be explained by the fact that HPMC swells significantly, and the dissolved substance must have diffused through the polymer network before being released. The best tabletability properties are provided by the formulation containing the extrudate with lyophilized extract HPMC 50:50 w/w. These tablets offer a valuable baicalin release profile while maintaining good mucoadhesive properties that condition the tablet's retention in the application site and the effectiveness of therapy. Composites made of poly(l‐lactic acid) (PLLA) and β‐cyclodextrin/D‐limonene inclusion complex (CD‐Lim) are prepared to develop novel food packaging material with antibacterial properties. The composites are formulated with bio‐based materials that are also biodegradable. The addition of CD‐Lim to PLLA results in enhanced permeability and water uptake. Optical properties of PLLA/CD‐Lim composites also significantly vary compared to plain PLLA, with partial loss of transparency and gloss, but sizably increased barrier to UV light, which imparts protection from oxidation to lipid‐containing food. The mechanical properties of the composite films are also affected by composition. Scutellarie baicalensis radix, as a flavone-rich source, exhibits antibacterial, antifungal, antioxidant, and anti-inflammatory activity. It may be used as a therapeutic agent to treat various diseases, including vaginal infections. In this study, six binary mixtures of chitosan with stable S. baicalensis radix lyophilized extract were obtained and identified by spectral (ATR-FTIR, XRPD) and thermal (TG and DSC) methods. The changes in dissolution rates of active compounds and the significant increase in the biological properties towards metal chelating activity were observed, as well as the inhibition of hyaluronic acid degradation after mixing plant extract with chitosan. Moreover, the combination of S. baicalensis radix lyophilized extract with a carrier allowed us to obtain the binary systems with a higher antifungal activity than the pure extract, which may be effective in developing new strategies in the vaginal infections treatment, particularly vulvovaginal candidiasis. Cornelian cherry (Cornus mas L.) fruits are a valuable source of bioactive compounds that are responsible for the perception of bitter taste of chocolate products. The aim of the study was to validate the inhibitory effect of Cornus mas on the TAS2R3 and TAS2R13 bitter taste receptors and to assess the effect of masking the bitter taste of dark chocolate with the help of the sensory panel. Dark chocolate was prepared with an addition of 5% of freeze-dried cornelian cherry fruits and 108 CFU/g of Bacillus coagulans probiotic strains. Effect on the TAS2R receptors was evaluated in specially transfected HEK293T cells, and the inhibition ratio was measured using the calcium release test. Moreover, the total polyphenol content, antioxidant activity and simulated intestinal in vitro digestion were determined for the samples. The tested chocolate products were rich in chlorogenic, caffeic and sinapic acids. The addition of cornelian cherry positively affected the antioxidant activity. The phytochemicals of Cornus mas decreased the TAS2R13 activity by 132% after a 2-minute interaction and, % at the same time, inhibited the TAS2R3 activity by 11.5. The nutraceutical system of curcumin-piperine in 2-hydroxypropyl-β-cyclodextrin was prepared by using the kneading technique. Interactions between the components of the system were defined by X-ray powder diffraction (XRPD), differential scanning calorimetry (DSC), infrared spectroscopy (FT-IR), nuclear magnetic resonance (NMR). Application of hydroxypropyl-β-cyclodextrin as a carrier-solubiliser improved solubility of the curcumin–piperine system, its permeability through biological membranes (gastrointestinal tract, blood–brain barrier) as well as the antioxidant, antimicrobial and enzyme inhibitory activities against acetylcholinesterase and butyrylcholinesterase. Food can be a source of valuable peptides with high bioactivity, which regulate the functioning of cardiovascular and nervous systems. The aim of the study was to evaluate the possibility of usage ricotta after hydrolysis to obtain innovative chocolate desserts. It was shown that the hydrolysis of whey proteins in ricotta had insignificant effect on the texture indices of the products, except gumminess, as it declined to 16% in ricotta samples and to 7% in case of chocolate dessert samples. Confirmed was that the hydrolysis of the ricotta affected the activity of prepared desserts with respect to cholinesterases and angiotensin‐converting enzyme. Enzymatic hydrolysate of ricotta may be consider as a semifinished product of high functional activity, and its further application in dessert production allows to provide novel prohealth new products. Practical Application: Study results indicate new feasibilities of ricotta application as functional ingredient of new products––chocolate desserts. The results show that ricotta after the stage of enzyme hydrolysis of proteins might have noticeable effect on product functionality. A measurable benefit for the consumer is the receipt of a new product with favorable health‐promoting properties, and for the entrepreneur new possibilities to expand the range of functional products. One of the main goals of industrial biotechnology is to develop an effective method for ethanol production for fuel purposes using lignocellulosic biomass. Variability of lignocellulosic raw materials, selection of an effective method for the pretreatment of raw material, and selection of microorganisms with the ability to ferment not only hexoses but also pentoses and are moreover resistant to environmental stress generated by the products of lignocellulosic complex decomposition, are the challenges encountered in ethanol production. The use of agricultural wastelands and overgrowing plants that have little possibility of application in processes other than energy production seem to be an interesting alternative to conventional, but very often rather cultivation demanding energy crops. The aim of this study was to evaluate the possibility of using the stems of fireweed (Epilobium angustifolium L.), European goldenrod (Solidago virgaurea L.), and common broom (Cytisus scoparius L.) for ethanol production. The key elements studied were characteristics of the lignocellulosic complex structure, influence of the selected ionic liquids on the structural changes in biomass, and efficiency of enzymatic hydrolysis and ethanol fermentation processes. The results showed that under the assumed conditions the best effect was observed with the fireweed materials subjected to pretreatment with 1‐ethyl‐3‐methylimidazolium acetate and enzymatic hydrolysis with Viscozyme® preparation. The final concentration of ethanol obtained was 2.509 g L−1 with a yield of 92.3%. This was due to the highest share of cellulose (40.9%) in the whole lignocellulosic complex compared to other raw materials, which in combination with the selection of an appropriate ionic liquid and an enzymatic preparation, led to high bioprocess efficiency. The influence of ionizing radiation on the physicochemical properties of panipenem in the solid-state was studied. In the first part of our studies, we evaluated the possibility of applying radiosterilization to obtain sterile panipenem. Spectroscopic (FT-IR, Raman), chromatography (HPLC and HPLC-MS) and X-ray powder diffraction (XRPD) studies have shown that irradiation of panipenem with the 25 kGy, the dose required to achieve sterility, does not change the physicochemical properties of the studied compound. The antimicrobial activity of panipenem irradiated with the dose of 25 kGy was reduced for three species. This study aimed to investigate changes in the solubility and antimicrobial efficacy of cefuroxime axetil (CA) when incorporated into cyclodextrin (CD). While choosing the CD, the validated in silico model was used. A theoretical model based on docking and molecular mechanics/generalized born surface area was validated using a curated dataset of API (active pharmaceutical ingredient)-CD stability constants. The library of commonly used cyclodextrins was virtually screened, indicating CA -hydroxypropyl-βCD (HPβCD) as the most thermodynamically favored system. Solid-state CA-HPβCD system was prepared and characterized by differential scanning calorimetry (DSC), Fourier-transform infrared (FT-IR), and X-ray diffraction (XRPD) methods. The dissolution profiles of the CA and its cyclodextrin system were evaluated. Microbiological activity of the CA-HPβCD inclusion system was studied based on changes in minimal inhibitory concentration (MIC) values and related to ones of the pure CA. The theoretical model was successfully validated, obtaining an average correlation with experimental data R = 0.7. The dissolution study showed significantly improved dissolution profiles of CA-HPβCD compared to CA. HPβCD increases the antimicrobial efficacy of CA up to 4-fold compared to pure CA. The study presents the results of 4-year field trials; their purpose was to assess maize yield in relation to the depth of a mineral two-component (NP) fertiliser application in the soil layer, the type of nitrogen fertiliser (ammonium nitrate and urea) and the date of its application. The yield grain was significantly dependent on changing weather conditions in the growing seasons. Initial fertilisation with two-component NP fertiliser, regardless of the years, significantly affected the grain yield. Row fertilisation with two-component NP fertiliser increases the availability of phosphorus in the acid soil environment, elevating maize grain yield. The efficiency of row NP fertiliser application is determined by the natural soil richness in phosphorus. Maize grain yield depended more on the date of application than the form of nitrogen, and its application before sowing was more effective. The application of ammonium nitrate in the BBCH 15/16 stage significantly reduced the number of production ears per unit area compared to the pre-sowing application of this fertiliser. Prunus padus L. is a wild growing plant in various parts of the world with low soil requirements. This work evaluated the content of selected phytochemicals, antioxidant potential and determined the antimicrobial effect of individual anatomical parts of P. padus (i.e. leaves, bark, and fruits). It was shown that the examined anatomical parts of bird cherry (Prunus padus L.) contained polyphenols and low-molecular organic acids. The highest content of phenolic acids was found in P. padus leaves (1063 ± 26 mg/100 g dm). The analysis regarding flavonols confirmed that the highest content was found in the bark (1530 ± 61 mg/100 g dm). The content of low molecular weight organic acids, whose highest value was found in the fruits (1658 ± 27 mg/100 g dm) was also assessed. The highest antioxidant activity with DPPH radicals was confirmed for the bark (8.9 ± 0.03 mg TE/g dw). The FRAP test also showed the highest activity for the bark extract (380 ± 18 µM FeSO4/g dw). It was shown that signals located in the region of negative potentials in SWV (square wave voltammetry) indicated a high reducing ability of compounds present in P. padus, which means that they can be a very efficient protective barrier against oxidizing agents (i.e. the greatest intensity was for the leaves extract). The highest activity against Gram-negative bacteria was recorded in bird cherry leaves and the lowest in the bark. It showed that the leaves, fruits, and bird cherry bark can be a good source of antioxidant compounds and an attractive raw material with antimicrobial properties that can be widely applied, which is confirmed by the obtained results. One of the best sterilization methods for chemically unstable drugs, such as carbapenems, is irradiation. The chemical and physical properties of radiosterilized imipenem and cilastatin were analyzed using spectrophotometric and chromatographic methods. Unstable free radicals were presented in samples after irradiation; their mean lifetime was 142 ± 64 h. The antibacterial activity of cefozopran hydrochloride irradiated with a dose of 25 kGy changed for Klebsiella pneumoniae and Escherichia coli. Imipenem and cilastatin in the solid state are not resistant to radiation sterilization; therefore, this method cannot be used for sterilization of this compound. [ABSTRACT FROM AUTHOR]

As a systemic disease, diabetes mellitus (DM) is characterized by the disruption of many glucose metabolic pathways. Therefore, it seems critical to study new therapies to support treatment to develop therapeutic systems that can operate across a broad metabolic spectrum. The current state of knowledge indicates an essential role of the gut microbiota in the development and course of the disease. Cornus mas fruits have demonstrated a rich biological activity profile and potential for application in the treatment of DM. As part of a preliminary analysis, the activity of four cultivars of Cornus mas fruits was analyzed. The cultivar Wydubieckij was selected as having the highest activity in in vitro conditions for further prebiotic system preparation. The study aimed to develop a unique therapeutic system based, first of all, on the mechanism of α-glucosidase inhibition and the antioxidant effect resulting from the activity of the plant extract used, combined with the prebiotic effect of inulin. The obtained system was characterized in vitro in terms of antioxidant activity and enzyme inhibition capacity, and was then tested on diabetic rats. The study was coupled with an analysis of changes in the intestinal microflora. The system of prebiotic stabilized Cornus mas L. lyophilized extract with inulin offers valuable support for the prophylaxis and treatment of DM. [ABSTRACT FROM AUTHOR]

Abstract: A major problem with fermentative ethanol production is the formation of large amounts of numerous organic pollutants. In an industrial distillery, stillage, fermenter and condenser cooling water are the main sources of wastewater. However, the selection of a proper technology makes it possible to almost completely avoid emissions of such kind of wastewater to the environment. This study examines the effect of stillage recirculation on fuel ethanol production. It is based on the use of Saccharomyces cerevisiae and a granular starch hydrolyzing enzyme in a simultaneous saccharification and fermentation process using a native starch obtained from corn flour. It was shown that the yield of the ethanol production was not influenced by the recycled stillage, a mean yield being 83.38% of the theoretical value. No significant trend for change in the ethanol concentration or in the residual starch was observed during any particular run, even after the 75% of fresh water was replaced with stillage. Thus, by applying this new clean technology it is possible to significantly reduce the rate of water consumption and in this way the production of by-products such as stillage. Background. The most important innovations in boethanol production in the last decade were: simultaneous saccharification and fermentation processes (SSF), high gravity fermentation, the use of new enzyme preparation able to hydrolyse native granular starch and construction of genetically modified strains of microorganisms able to carry out simultaneous production of hydrolytic enzymes and fermentation of C6 and C5 sugars. The aim of this study was to assess the efficiency of ethanol fermentation using new type of amylolytic enzymes able to hydrolyse native corn starch in a SSF process. Material and methods. The simultaneous saccharification and fermentation of raw corn flour by fed-batch processes using Saccharomyces cerevisiae strain Red Star Ethanol Red and Stargen 001 enzyme preparation was performed. As experimental variable were investigated: fermentation temperature (35-37-40°C), rate of mash stirring (100 and 200 rpm), fermentation time (0-92 h) and dosage of corn flour (different portion and different time). Results. It was found that optimal temperature for fed-batch SSF process was 37°C at initial pH of 5.0. However, the yeast intensively fermented the saccharides also at 40°C. The fermentation stirring rate has significant effect on starch utilization and fermentation production. The prolongation of fermentation time over 72 h has no substantiation in additional ethanol production. In all experimental fermentations the level of produced organic acids was very low, significantly below toxic concentration for the yeast. Conclusions. It was stated that the use of new method of starch raw material preparation resulted in satisfied fermentation yield and allowed to reduce energy requirements for starch liquefaction. Simultaneous saccharification and ethanol fermentation of corn starch using enzyme preparation Stargen 001 was investigated. The experiments were conducted in an agro-distillery plant. The preparation of corn mashed based on the use of corn flour suspended in recycled liquid fraction of stillage and distillation condensates. It was shown that the final concentrations of ethanol after fermentation of 25% w/v mashes were of 8-10% w/v. The elimination of cooking for starch liquefaction allowed to reduce of energy requirements, nevertheless, made s serious problem with bacterial contamination of mashes. The concentration of lactic acid at the end of fermentation process reached up to 3.8% w/v. The SFF process connected with repeated stillage recycling allowed to remove water requirements and to perform this process as discard-less. [ABSTRACT FROM AUTHOR]

Blackberry fruits are recognized as functional foods while blackberry leaves are outside this classification and they also contain active compounds with health-promoting potential. Therefore, the aim of this study was the phytochemical analysis of blackberry leaves of varieties (Chester, Loch Ness, Loch Tay and Ruczaj) and screening of their biological activity (antioxidant potential, possibility of inhibition of enzymes, anti-inflammatory and microbial activity). The following compounds from selected groups: phenolic acids (caffeic acid, ellagic acid, gallic acid, syringic acid), flavonols (quercetin, kaempferol) and their glycosides (rutin, isoquercetin, hyperoside) and flavon-3-ols (catechin, epicatechin) were chromatographically determined in the aqueous and hydroalcoholic leaves extracts. All tested blackberry leaves extracts showed antioxidant effects, but the highest compounds content (TPC = 101.31 mg GAE/g) and antioxidant activity (e.g., DPPH IC50 = 57.37 μg/mL; ABTS IC50 = 24.83 μg/mL; CUPRAC IC50 = 62.73 μg/mL; FRAP IC50 = 39.99 μg/mL for hydroalcoholic extracts) was indicated for the Loch Tay variety. Blackberry leaf extracts' anti-inflammatory effect was also exceptionally high for the Loch Tay variety (IC50 = 129.30 μg/mL), while leaves extracts of the Loch Ness variety showed a significant potential for microbial activity against Lactobacillus spp. and Candida spp. Summarizing, the best multidirectional pro-health effect was noted for leaves extracts of Loch Tay variety. Progressive increase in bacterial resistance has caused an urgent need to introduce new antibiotics, one of them being oxazolidinones with their representative tedizolid. Despite the broad spectrum of activity of the parent tedizolid, it is characterized by low water solubility, which limits its use. The combination of the active molecule with a multifunctional excipient, which is cyclodextrins, allows preservation of its pharmacological activity and modification of its physicochemical properties. Therefore, the aim of the study was to change the dissolution rate and permeability through the model membrane of tedizolid by formation of solid dispersions with a cyclodextrin. The research included identification of tedizolid-hydroxypropyl-β-cyclodextrin (tedizolid/HP-β-CD) inclusion complex by thermal method (Differential Scanning Colorimetry), spectroscopic methods (powder X-ray diffraction, Fourier-Transform Infrared spectroscopy), and molecular docking. The second part of the research concerned the physicochemical properties (dissolution and permeability) and the biological properties of the system in terms of its microbiological activity. An increase in the dissolution rate was observed in the presence of cyclodextrin, while maintaining a high permeation coefficient and high microbiological activity. The proposed approach is an opportunity to develop drug delivery systems used in the treatment of resistant bacterial infections, in which, in addition to modifying the physicochemical properties caused by cyclodextrin, we observe a favorable change in the pharmacological potential of the bioactives. Honeys have a pleasant taste and a wide range of use. They are characterized by a relatively high consumption compared to bee pollen or beebread. Honeys are the most popular bee products. Considering health reasons, beebread exhibits the strongest properties as it has the highest nutritional value as well as strong detoxifying, antioxidant, and antiradical properties. Despite having such valuable properties, consumption of beebread is negligible; sometimes, it is limited only to supplementation in case of diseases. This paper proposes a new food product, that is, beebread caviar made from buckwheat honey. The expiry date and sensory and physicochemical quality of beebread caviar have been determined in this study. Beebread caviar, obtained by immobilization on alginate carrier, contained 0.34 mg GAE/mL extract. It remained stable until five days after preparation. Its total acidity was 33.7 mval/kg. Its extract content was 22.53%. Caviar had a high overall sensory score of 4.8 points on a 5-point scale. Beebread caviar can be successfully classified as probiotic food because beebread contains a large amount of lactic acid. In the form of caviar, a new, attractive, and convenient form of beebread consumption could become one of the products of comfortable and functional food. [ABSTRACT FROM AUTHOR]

Mulberry is one of the most beneficial plant of our planet for sustainable development. White mulberry (Morus alba L.) is widely recognized for its health-promoting properties. It is characterized by a high content of bioactive compounds, mainly flavonoids, and has a strong antioxidant effect, and thus can have a beneficial effect on health. The aim of the study was to evaluate the effect of freezing storage of bread with the addition of extract from mulberry leaves and fruits on the content of polyphenols, antioxidant activity and sensory properties. The stored bread with mulberry addition was characterized by high content of phenolic compounds, reducing and chelating activity and antiradical activity. The addition of mulberry had greater effect on the increase in the content of protocatechuic and chlorogenic acids, and isoquercetin among the flavonols. Bread enriched with mulberry was microbiologically clean and sensory accepted both after baking and after 30 days of storage under refrigerated conditions. White mulberry is a raw material which can be used as an addition to enrich refrigerated bread. The use of the extract and mulberry fruit to fortify bread is consistent with the principle of sustainable development due to the use of raw materials which are a good source of compounds contributing to the improvement of the well-being of the population. Currently, coniferous shoots are almost absent as a food ingredient despite their wide availability in many parts of the world. The aim of the study was to assess and compare the composition of selected plant metabolites, evaluate the antioxidant and antimicrobial properties of selected shoots collected in 2019 from the arboretum in Zielonka (Poland), including individual samples from Picea abies L. (PA), Larix decidua Mill (LD), Pinus sylvestris L. (PS), Pseudotsuga menziesii (PM) and Juniperus communis L. (JC). The present work has shown that aqueous extracts obtained from tested shoots are a rich source of phenols such as caffeic acid, ferulic acid, chlorogenic acid, 4-hydroxybenzoic acid and many others. Obtained extracts exhibit antioxidant and antimicrobial properties in vitro. The highest sum of the studied phenolic compounds was found in the PA sample (13,947.80 µg/g dw), while the lowest in PS (6123.57 µg/g dw). The samples were particularly rich in ferulic acid, chlorogenic acid and 4-hydroxybenzoic acid. The highest values regarding the Folin-Ciocâlteu reagent (FCR) and ferric reducing ability of plasma (FRAP) reducing ability tests, as well as the total flavonoid content assay, were obtained for the LD sample, although the LD (14.83 mg GAE/g dw) and PM (14.53 mg GAE/g dw) samples did not differ statistically in the FCR assay. With respect to free radical quenching measurements (DPPH), the PA (404.18-μM Trolox/g dw) and JC (384.30-μM Trolox/g dw) samples had the highest radical quenching ability and did not differ statistically. Calendulae flos is a valued plant material with known anti-inflammatory and antimicrobiological properties. The limitation for its use in the treatment of chronic wounds is the lack of adhesion to the required site of action. Obtaining the Calendulae flos lyophilized extract from water-ethanolic extract allowed to prepare valuable material whose biological activity in the wound healing process was confirmed by a positive result of the scratch test. The Calendulae flos lyophilized extract was standardized for the contents of the chlorogenic acid and the narcissin. The significant potency of the Calendulae flos pharmacological activity has become the reason for studies on its novel applications in combination with the multifunctional chitosan carrier, to create a new, valuable solution in the treatment of chronic wounds. The use of chitosan as a carrier allowed for the controlled release of the chlorogenic acid and the narcissin. These substances, characterized by prolonged release from the chitosan delivery system, were identified as well permeable, based on the results of the studies of the parallel artificial membrane permeability assay (PAMPA Skin) a model simulating permeability through membrane skin. The combination of the Calendulae flos lyophilized extract and the chitosan allowed for synergy of action towards hyaluronidase inhibition and effective microbiological activity. Optimization of the hypromellose hydrogel preparation ensuring the required rheological properties necessary for the release of the chlorogenic acid and the narcissin from the chitosan delivery system, as well as demonstrated antimicrobial activity allows indicating formulations of 3% Calendulae flos lyophilized extract with chitosan 80/500 in weight ratio 1:1 and 2% or 3% hypromellose as an important support with high compliance of response and effectiveness for patients suffering from chronic wounds. Chelidonium majus (also known as celandine) contains pharmacologically active compounds such as isoquinoline alkaloids (e.g., chelidonine, sanguinarine), flavonoids, saponins, carotenoids, and organic acids. Due to the presence of isoquinoline alkaloids, Chelidonii herba extracts are widely used as an antibacterial, antifungal, antiviral (including HSV-1 and HIV-1), and anti-inflammatory agent in the treatment of various diseases, while chitosan is a biocompatible and biodegradable carrier with valuable properties for mucoadhesive formulations preparation. Our work aimed to prepare mucoadhesive vaginal drug delivery systems composed of Chelidonii herba lyophilized extract and chitosan as an effective way to treat vaginitis. The pharmacological safety of usage of isoquinoline alkaloids, based on MTT test, were evaluated for the maximum doses 36.34 ± 0.29 µg/mL and 0.89 ± 1.16 µg/mL for chelidonine and sanguinarine, respectively. Dissolution rate profiles and permeability through artificial membranes for chelidonine and sanguinarine after their introduction into the chitosan system were studied. The low permeability for used save doses of isoquinoline alkaloids and results of microbiological studies allow confirmation that system Chelidonii herba lyophilized extract chitosan 80/500 1:1 (w/w) is a promising strategy for vaginal use. Ex vivo studies of mucoadhesive properties and evaluation of tableting features demonstrated that the formulation containing Chelidonii herba lyophilized extract (120.0 mg) with chitosan (80/500—100.0 mg) and polymer content (HPMC—100.0 mg, microcrystalline cellulose—50.0 mg, lactose monohydrate—30.0 mg and magnesium stearate—4.0 mg) is a vaginal dosage form with prolonging dissolution profile and high mucoadhesion properties (up to 4 h). Barley (Hordeum vulgare L.) is an annual plant cultivated in spring or autumn. Currently, over 70% of the cultivated barley grains are utilized for preparing fodder, while the rest is used for the production of malt and cereals in the food industry. The purpose of the present work was to evaluate the content of bioactive compounds, antioxidant potential, and cholinesterase inhibitory effect of the aqueous extracts of juvenile barley leaves. It was found that the barley cultivars differed in their content of the determined phytochemicals as well as their antioxidant potential and cholinesterase-inhibitory activity. The water extracts of young barley leaves contained phenolic acids as well as quercetin, rutin, and kaempferitrin. The extracts showed a higher inhibitory effect on 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) than on 2,2-diphenyl-1-picrylhydrazyl (DPPH) radicals. Based on the aqueous extracts analyzed, we found that winter cultivars were characterized by the highest iron-chelating activity. Furthermore, barley extracts showed a stronger inhibitory effect against acetylcholinesterase compared to butyrylcholinesterase. The results of the present work indicated that barley cultivars differed in their germination process. For the first time, the influence of ionising radiation on the physicochemical properties of ertapenem in solid state was studied. During our studies, we evaluated the possibility of applying radiosterilization to obtain sterile ertapenem. Spectroscopic (Fourier Transform Infrared (FT-IR)), thermal (differential scanning calorimetry (DSC), chromatography (High-Performance Liquid Chromatography (HPLC) and HPLC-MS), and X-ray powder diffraction (XRPD) studies shown that irradiation of ertapenem with the 25 kGy, the dose required to achieve sterility, does not change the physicochemical properties of the studied compound. The antimicrobial activity of ertapenem irradiated with the dose of 25 kGy was only reduced for one species. Based on the received results, we can conclude that radiostelization is a promising alternative method of obtaining sterile ertapenem. In our studies, ertapenem was also exposed to e-beam radiation with a dose of 400 kGy.
