• Drought-induced xylem embolism is a serious threat to plant survival under future climate scenarios. Hence, accurate quantification of species-specific vulnerability to xylem embolism is a key to predict the impact of climate change on vegetation. Low-cost hydraulic measurements of embolism rate have been suggested to be prone to artefacts, thus requiring validation by direct visualization of the functional status of xylem conduits using nondestructive imaging techniques, such as X-ray microtomography (microCT). • We measured the percentage loss of conductance (PLC) of excised stems of Laurus nobilis (laurel) dehydrated to different xylem pressures, and compared results with direct observation of gas-filled vs water-filled conduits at a synchrotron-based microCT facility using a phase contrast imaging modality. • Theoretical PLC calculated on the basis of microCT observations in stems of laurel dehydrated to different xylem pressures overall were in agreement with hydraulic measurements, revealing that this species suffers a 50% loss of xylem hydraulic conductance at xylem pressures averaging--3.5 MPa. • Our data support the validity of estimates of xylem vulnerability to embolism based on classical hydraulic techniques. ABSTRACTThe possible link between stomatal conductance (gL), leaf water potential (ΨL) and xylem cavitation was studied in leaves and shoots of detached branches as well as of whole plants of Laurus nobilis L. (Laurel). Shoot cavitation induced complete stomatal closure in air-dehydrated detached branches in less than 10 min. By contrast, a fine regulation of gL in whole plants was the consequence of ΨL reaching the cavitation threshold (ΨCAV) for shoots. A pulse of xylem cavitation in the shoots was paralleled by a decrease in gL of about 50%, while ΨL stabilized at values preventing further xylem cavitation. In these experiments, no root signals were likely to be sent to the leaves from the roots in response to soil dryness because branches were either detached or whole plants were growing in constantly wet soil. The stomatal response to increasing evaporative demand appeared therefore to be the result of hydraulic signals generated during shoot cavitation. Diurnal changes in percentage loss of hydraulic conductivity ( PLC), with recorded values being higher at midday than on the following morning, have been interpreted as evidence for the occurrence of cycles of xylem conduits' embolism and repair. Recent reports have suggested that diurnal PLC changes might arise as a consequence of an experimental artefact, that is, air entry into xylem conduits upon cutting stems, even if under water, while under substantial tension generated by transpiration. Rehydration procedures prior to hydraulic measurements have been recommended to avoid this artefact. In the present study, we show that xylem rehydration prior to hydraulic measurements might favour xylem refilling and embolism repair, thus leading to PLC values erroneously lower than those actually experienced by transpiring plants. When xylem tension relaxation procedures were performed on stems where refilling mechanisms had been previously inhibited by mechanical (girdling) or chemical (orthovanadate) treatment, PLC values measured in stems cut under native tension were the same as those measured after sample rehydration/relaxation. Our data call for renewed attention to the procedures of sample collection in the field and transport to the laboratory, and suggest that girdling might be a recommendable treatment prior to sample collection for PLC measurements. This work reports on experimental evidence for the role of ion-mediated changes of xylem hydraulic conductivity in the functional response of Solanum lycopersicum L. cv. Naomi to moderate salinity levels. Measurements were performed in fully developed 12-week-old plants grown in half-strength Hoagland solution (control, C-plants) or in the same solution added with 35mM NaCl (NaCl-plants). NaCl-plants produced a significantly less but heavier leaves and fruits but had similar gas-exchange ratesas control plants. Moreover, NaCl-plants showed higher vessel multiple fraction (F[sub vM]) than control plants. Xylem sap potassium and sodium concentrations were significantly higher in NaCl-plants than in control plants. When stems were perfused with10 mM NaCl or KCl, the hydraulic conductance of NaCl plants was nearly 1.5 times higher than in control plants. Accordingly, stem hydraulic conductance measured in planta was higher in NaCl- than in control plants. Our data suggest that tomato plants grown under moderate salinity upregulate xylem sap [Na[sup +]] and [K[sup +]], as well as sensitivity of xylem hydraulics to sap ionic content, thus, increasing water transport capacity. Leaf hydraulics of Aesculus hippocastanum L. were measured over the growing season and during extensive leaf mining by the larvae of an invasive moth ( Cameraria ohridella Deschka et Dimic) that specifically destroy the palisade tissue. Leaves showed seasonal changes in hydraulic resistance ( Rlamina) which were related to ontogeny. After leaf expansion was complete, the hydraulic resistance of leaves and the partitioning of resistances between vascular and extra-vascular compartments remained unchanged despite extensive disruption of the palisade by leafminers (up to 50%). This finding suggests that water flow from the petiole to the evaporation sites might not directly involve the palisade cells. The analysis of the temperature dependence of Rlamina in terms of Q10 revealed that at least one transmembrane step was involved in water transport outside the leaf vasculature. Anatomical analysis suggested that this symplastic step may be located at the bundle sheath where the apoplast is interrupted by hydrophobic thickening of cell walls. Our findings offer some support to the view of a compartmentalization of leaves into well-organized water pools so that the transpiration stream would involve veins, bundle sheath and spongy parenchyma, while the palisade tissue would be largely by-passed with the possible advantage of protecting cells from short-term fluctuations in water status. The ability of stems of Laurus nobilis(L.) to refill embolised xylem conduits was studied in plants both at optimal water supply (W) and under conditions of soil drought inducing xylem pressures (Px) of –1.54 (S1) and –2.35MPa (S2). Starch depolymerisation in wood parenchyma was measured as percentage of cells ‘with high starch content' (HSC-cells) counted under a microscope. HSC-cells decreased during embolism and increased again in refilled stems. A direct relationship was found between percentage of HSC-cells and Px, with HSC-cells between 65 and 75% of the total at Px≥–0.6MPa, at which recovery from PLC was recorded. At low transpiration, starch re-appeared in wood parenchyma cells but only in plants that showed diurnal stomatal opening (W- and S1-plants). In S2-plants showing diurnal stomatal closure and nocturnal opening with Pxbetween –1.2 to –2.4MPa, HSC-cells were only 25% and plants did not recover from PLC. This finding suggests that (i) the Pxthreshold for embolism repair was≥–0.6MPa, and (ii) impeded phloem loading limits starch content in wood parenchyma and embolism repair. We conclude that starch depolymerisation acts as a signal to phloem unloading sugars to embolised conduits thus generating the necessary osmotic gradients driving refilling. [ABSTRACT FROM AUTHOR]

Leaf veins undergo cavitation at water potentials (Ψleaf) commonly experienced by field-growing plants. Theoretically, embolism reversal should not be possible until xylem pressures rise by several kilopascals of atmospheric pressure, but recent evidence suggests that embolized conduits can be refilled even when surrounded by others at substantial tension (novel refilling). The present study reports ‘novel refilling’ occurring in leaf veins of sunflower ( Helianthus annuus L.) while at Ψleaf = −0.33 MPa. Sixty per cent loss of vein hydraulic conductance (Kvein) was recorded at Ψleaf < −0.65 MPa, while stem hydraulic conductance (Kstem) was unaffected even at Ψleaf = −1.1 MPa. Loss of Kvein was accompanied by stomatal closure. Water-stressed plants (Ψleaf = −1.1 MPa) were rehydrated overnight to different target water potentials achieved by using PEG at different concentrations as irrigation medium. Kvein recovered by 50% at Ψleaf = −0.47 MPa and vein refilling was complete at Ψleaf = −0.33 MPa, i.e. well below the theoretical limit for conduit refilling (−0.05 MPa as calculated for sunflower minor veins). Mercurials supplied to detached leaves had no effect on the refilling process. Upon rehydration, recovery of Kvein was not paralleled by recovery of whole-plant hydraulic conductance or leaf conductance to water vapour (gL), as a likely consequence of hydraulic failure of other components of the water pathway (root system or extravascular leaf compartments) and/or root-to-leaf chemical signalling. Spatial heterogeneity of gas exchange rates in the leaves of Nicotiana tabacum L. (tobacco) was investigated. Leaf conductance to water vapour was higher (by about 18%) at the apical regions of leaves than at the basal ones. Local, small-scale measurements of pressure–volume (PV) parameters and water status (performed with a dewpoint hygrometer) revealed that bulk leaf water potential, osmotic potential, turgor pressure and bulk modulus of elasticity were not significantly different in the leaf apex or base. Hydraulic measurements showed that the apical regions of the leaf blade were about 30% more conductive than the basal regions. Such differences were explained by analogous differences in terms of venation patterns. In fact, vein density turned out to be higher (by about 13%) near the leaf apex with respect to the leaf base. On the contrary, stomatal density was the same both in the apical and basal leaf portions. Our data suggest that spatial stomatal heterogeneity may arise from heterogenous distribution of local hydraulic resistances and would be addressed to maintaining local water potential above critical values, possibly triggering vein cavitation. Xylem embolism repair is preceded by starch depolymerisation in vessel-associated cells (VAC) of Laurus nobilis L. (laurel) twigs, but the primary signal triggering such a process is still unknown. We tested the hypothesis that conduit wall vibrations during cavitation may be sensed by VAC inducing starch-to-sugar conversion. Twigs of laurel from watered or stressed plants were exposed to ultrasound for 60min to simulate acoustic waves emitted by cavitating conduits. Preliminary tests showed that ultrasound caused no damage to cell membrane integrity nor did they cause xylem embolism. The number of VAC with high starch content (HSC-cells) was estimated microscopically by counting the cells with more than 50% of their lumen filled with starch granules. Sonication had no effect on HSC-cells in twigs from watered plants while it induced a drop in the percentage HSC-cells from 80 to 40% in twigs from stressed plants, at the ultrasound source location. No effect was recorded in these twigs 20mm from the ultrasound source. Sonication was a good simulator of cavitation in inducing starch depolymerisation which suggests a possible bio- physical nature for the signal initiating embolism repair. [ABSTRACT FROM AUTHOR]

The expression of OePIP1.1 and OePIP2.1 aquaporins in root, stem and leaf was higher in Olea europaea L. (olive) plants carrying a dwarfing (D) clone as rootstock, than in plants carrying a vigorous (M) clone as rootstock. The highest difference of expression between plants with D and M rootstocks was detected in the root and for the PIP2 gene, the transcripts of which show a high water channel activity in oocytes. Compared with plants with M rootstock, plants with D rootstock showed reduced root and canopy biomass and reduced hydraulic conductance of the bulk root system (Kroot). Hydraulic conductance of D roots was higher than that of M roots when Kroot was scaled by root DW, in agreement with a higher expression of PIP genes. Leaf hydraulic conductance scaled by unit leaf surface area was almost twice as high in leaves from D plants compared with that in leaves from M plants. Modification of root growth imposed by grafting M onto D and D onto M slightly changed expression levels of root OePIP1.1 gene, the transcripts of which show almost null water channel activity in oocytes, suggesting a role less evident for OePIP1.1 in root hydraulic reequilibration. Changes in hydraulic conductivity (Kh) were measured monthly in stems of Laurus nobilis between July 2005 and June 2006 and in March and June 2006 in stems of Prunus laurocerasus and Phillyrea latifolia. Percent enhancement of Kh (ΔKh) was recorded as KCl injected into stems compared with deionized water. Stems of L. nobilis perfused with increasing [KCl] saturated the hydraulic effect at 50 mM KCl, while P. laurocerasus and Ph. latifolia showed saturation at 17 and less than 5 mM KCl, respectively. Impressive seasonal changes in ΔKh were recorded in L. nobilis, from +120% in February to +20% during the growth period. ΔKh was negatively correlated to minimum air temperatures and was highest during winter frost. Negative correlation also existed between ΔKh and conduit lengths as detected during transition of plants from activity to winter rest and from winter to spring. In the winter, the thermal effect was prevailing. High cavitation-induced Kh loss coincided with high ΔKh. Similar but much smaller seasonal effects on ΔKh were recorded in the other two species where, however, also smaller seasonal changes in conduit lengths were recorded. We conclude that (1) the K+-induced effect on Kh was because of interference of the cation with pectins of intervessel pit membranes (increasingly crossed by the ionic solution in stems with shorter conduits); (2) winter frost might have an effect on ΔKh and (3) seasonal changes in ΔKh because of possible change in pectic composition may represent a mechanism to regulate water flows in planta. Changes in hydraulic conductivity (Kh) were measured in stems of Laurus nobilis L. during perfusion with KCl, NaCl or sucrose solutions. Ionic solutes induced marked increase of Kh with respect to deionized water but sucrose had no effect. The kinetics of KCl-induced Kh increase was measured together with changes in [K+] of the perfused solution. Kh increases were paralleled by increases in the [K+]out/[K+]in ratio. Samples of different lengths or with increasing percentage loss of conductivity (PLC) due to xylem cavitation were tested, with the aim of increasing radial flow through intervessel pits. KCl solutions enhanced the Kh of 12-cm-long samples with a concentration-dependent effect up to 100 mm KCl. ΔKh increased from 3 to 30% in 1.5- and 12-cm-long samples, respectively and remained constant for longer samples. Increasing PLC induced an exponential increase in ΔKh. PLC measured with KCl solutions was significantly less than that measured with deionized water, suggesting that measurements of PLC can be affected by the composition of the perfused solution. Experiments support the hypothesis that the ‘ionic effect’ is mediated by physico-chemical changes of pectins of the pit membranes and raise the possibility that plants might alter the ionic composition of the xylem sap to alleviate the hydraulic impact of cavitation. We investigated the hydraulic architecture of young olive trees either self-rooted or grafted on rootstocks with contrasting size-controlling potential. Clones of Olea europea L. (Olive) cv ‘Leccino’ inducing vigorous scion growth (Leccino ‘Minerva’, LM) or scion dwarfing (Leccino ‘Dwarf’, LD) were studied in different scion/rootstock combinations (LD, LM, LD/LD, LM/LM, LD/LM and LM/LD). Shoots growing on LD root systems developed about 50% less leaf surface area than shoots growing on LM root systems. Root systems accounted for 60–70% of plant hydraulic resistance (R), whereas hydraulic resistance of the graft union was negligible. Hydraulic conductance (K = 1/R) of LD root systems was up to 2.5 times less than that of LM root systems. Total leaf surface area (AL) was closely and positively related to root hydraulic conductance so that whole-plant hydraulic conductance scaled by AL did not differ between experimental groups. Accordingly, maximum transpiration rate and minimum leaf water potential did not differ significantly among experimental groups. The circadian regulation of leaf hydraulic conductance ( Kleaf) was investigated in Helianthus annuus L. (sunflower). Kleaf was measured with an high pressure flow meter during the light and dark period from plants growing at a photoperiod of 12 h. Kleaf was 4.0 e−4 kg s−1 m−2 MPa−1 during the light period (LL) and 30–40% less during the dark period (DL). When photoperiod was inverted and leaves were measured for Kleaf at their subjective light or dark periods, Kleaf adjusted to the new conditions requiring 48 h for increasing from dark to light values and 4 d for the opposite transition. Plants put in continuous dark showed Kleaf oscillating from light to dark values in phase with their subjective photoperiod indicating that Kleaf changes were induced by the circadian clock. Several cuts through the minor veins reduced leaf hydraulic resistance ( Rleaf) of both LL and DL to the same value (1.0 e + 3 MPa m2 s kg−1) that equalled the vascular resistance ( Rv). The contribution of the non-vascular leaf resistance ( Rnv) to Rleaf was of 71.9% in DL and of 58.4% in LL. The dominant Rnv was shown to be reversibly modulated by mercurials, suggesting that aquaporins play a role in diurnal changes of Kleaf. [ABSTRACT FROM AUTHOR]

We studied changes in the hydraulic conductance of leaves (Kleaf) between dawn and dusk during the growth period (July) and at midday at the beginning of autumn in four tree species. The main objectives of the study were to check the extent of diurnal and seasonal changes in Kleaf and the relationships between Kleaf, irradiance and leaf gas exchange. Two evergreen (Aleurites moluccana and Persea americana) and two deciduous trees (Platanus orientalis and Quercus rubra) were studied. Leaf hydraulic conductance was measured every 2 h between 0700 and 1900 h in July and compared with values measured between 0900 and 1300 h in October. Other variables measured were photosynthetically active radiation (PAR), leaf conductance to water vapor (gL) and water potential (ΨL). In July, Kleaf varied by up to 75% in Pe. americana on a diurnal basis and by at least 44% in Q. rubra. The diurnal time course of Kleaf showed a distinct increase between dawn and late morning (1100 h) and a subsequent decrease in the evening in A. moluccana and Pl. orientalis, whereas in the other two species, Kleaf was highest just after dawn and lowest in the evening. In October, Kleaf of all the species studied was lower than in July, with differences of 20 to 28% for A. moluccana and Pl. orientalis and of 66 to over 70% in Pe. americana and Q. rubra, respectively. Significant correlations were found between PAR and Kleaf (in all species) as well as between gL and Kleaf (in three out of four species). The impact of leaf vein cavitation and embolism on stomatal response and leaf hydraulic conductance was studied in potted plants of sunflower subjected to water limitation. Plant dehydration was achieved either by cutting well-watered plants near their base and leaving them dehydrating in air or by depriving intact plants of irrigation. The vein cavitation threshold ( ΨCAV) was estimated in terms of ultrasound acoustic emissions (UAE) from the leaf blade versus leaf water potential ( ΨL). This was found to be the same ( ΨCAV ≈ −0.6 MPa) for leaves of both cut and intact plants where stomata began to close in coincidence with starting vein cavitation. Vein embolism was detected by infiltrating leaves at different ΨL with 0.7 m M fluorescein and measuring the percentage fluorescent area as percentage of total leaf surface area. A distinct loss of vein functionality (up to 50%) was found to occur in leaves at progressively decreasing ΨL, starting when leaves reached ΨCAV. A linear positive relationship with high statistical significance was found to exist between gL and percentage leaf fluorescent area, thus indicating that stomata were sensitive to vein embolism. The hydraulic conductance ( KL) of the leaf was affected by leaf dehydration less than expected ( KL decreased by about 20% between near full turgor and ΨL = −1.3 MPa). When the extravascular leaf compartment was excluded either by killing cells by immersing leaves in 70% ethanol or by cutting the main leaf venous system through to allow flow to bypass it, KL turned out to increase 5.5 times, thus suggesting that the high dominance of the hydraulic resistance of the extravascular leaf compartment over the total leaf resistance might buffer or mask possibly large local changes in KL inducing stomatal closure. Changes in leaf hydraulic conductance (K) were measured using the vacuum chamber technique during dehydration and rehydration of potted plants of Ceratonia siliqua. K of whole, compound leaves as well as that of rachides and leaflets decreased by 20–30% at leaf water potentials (ΨL) of −1.5 and −2.0 MPa, i.e. at ΨL values commonly recorded in field‐growing plants of the species. Higher K losses (up to 50%) were measured for leaves at ΨL of −2.5 and −3.0 MPa, i.e. near or beyond the leaf turgor loss point. Leaves of plants rehydrated while in the dark for 30 min, 90 min and 12 h recovered from K loss with characteristic times and to extents inversely proportional to the initial water stress applied. Leaf conductance to water vapour of plants dehydrated to decreasing ΨL and rehydrated at low transpiration was inversely related to loss of K, thus suggesting that leaf vein embolism and refilling (and related changes in leaf hydraulics) may play a significant role in the stomatal response.
