Abstract

Kalanchoe beharensis, a vulnerable species according to the International Union for Conservation of Nature, is highly prized for its ornamental value and medicinal properties. Therefore, an efficient methodology to propagate this ecologically significant species would be of particular interest. The propagation of K. beharensis has traditionally been achieved by seed or cuttings, but these methods are limited in efficiency. Micropropagation provides a more efficient and controlled alternative by enabling the in vitro production of numerous plants in a small space and in a short period of time. Despite its advantages, no micropropagation protocol for K. beharensis has been reported in the literature. In this study, we report an efficient in vitro regeneration protocol for K. beharensis. In order to implement this, we evaluated the morphogenetic response of leaf and root explants in media supplemented with auxins, cytokinins, or a combination of both growth regulators. Surprisingly, the best results were observed in indole-3-acetic acid-supplemented media. Adventitious shoots were rooted in either hormone-free or auxin-supplemented media, with indole-3-acetic acid yielding the best results. Rooted plants were acclimatized in the greenhouse, achieving over 80% survival during acclimatization. This protocol improves multiplication rate, space utilization, and uniformity, providing a viable alternative to conventional propagation methods. [ABSTRACT FROM AUTHOR]

16. Acidogenic Fermentation of Cassava Wastewater: Effect of the Substrate-to-Microorganism Ratio and Temperature on Volatile Fatty Acids Production.

Abstract

The production of volatile fatty acids (VFAs) through the acidogenic fermentation of wastewater is an emerging technology that requires further research to optimize operational variables for specific substrates. Cassava wastewater, which is a byproduct of the cassava sour starch extraction process, has been minimally studied regarding its potential for VFA production through acidogenic fermentation. Batch reactors were used to evaluate the effects of the substrate-to-microorganism (S/M) ratio and temperature on VFA production from cassava wastewater. The results showed no statistically significant differences between the evaluated S/M ratios. The maximum total VFA concentration observed was 2214.64 mg of acetic acid (HAc)/L (0.32 gCODVFA/gCOD), which was achieved at a S/M ratio of 4 gCOD/gVS. This concentration was predominantly composed of acetic acid (42.7%), followed by butyric acid (30.1%) and propionic acid (24.6%), with a minor quantity of isovaleric acid (2.6%). The statistical analysis for the temperature variable showed significant differences between the evaluated conditions. The maximum concentration of total VFAs was 2650.19 mgHAc/L (0.45 gCODVFA/gCOD) at 34 ± 1 °C, with acetic (40.9%), butyric (29.8%), and propionic (29.3%) acids as primary metabolites. Leishmaniasis is a zoonotic disease which is caused by protozoan parasites of the genus Leishmania. Canids are the most important reservoir of the parasites; however, limited data are available on the species of Leishmania prevalent in these animals and their impact on human health. The objective of this study was to estimate the seroprevalence of leishmaniasis in dogs from an inter-Andean region of Colombia during July 2016–July 2017, and to describe the clinical and histopathological features of the disease.

Abstract

This paper proposes and develops a protocol for measuring the aerobic granulation potential of sludge, aiming to provide an affordable and simple alternative that can facilitate the development of aerobic granulation technology. In this sense, the protocol comprises a set of parameters and considerations that interact to create a controlled environment and stimulate cell population clustering. All of this is done in the context of procedural simplicity, low cost, and the speed at which results are obtained. The protocol is essentially a three-stage method: preparation of the substrate, adaptation of the inoculum, and implementation of the protocol. Simple parameters were measured to evaluate the granulation process: SVI, settling velocity, and morphological parameters. The protocol was validated according to optimal ranges and criteria previously established in the literature. For this purpose, an activated sludge inoculum from a domestic wastewater treatment plant was submitted to the protocol, obtaining an optimal response of the biomass (SVI5 =13.90 mL g−1, settling velocity= 25,79 m h−1, Diameter > 0.2 mm) in a relatively short time (7 d). The results show that this protocol can constitute a tool for evaluation and decision-making using traditional laboratory equipment and is applicable at different scales.

Abstract

Acidogenic fermentation is a technology that involves halting methanogenesis in the conventional anaerobic digestion process to produce mainly volatile fatty acids (VFAs). VFAs serve as direct precursors to energy-rich or higher value-added products upon undergoing additional processing. In this study, batch reactors were utilized to assess the individual and interaction effects of fermentation time and pH variables on VFA production from acidogenic fermentation of cassava wastewater through the establishment of a completely randomized design and a second-order response surface (rotatable central composite design), respectively. The maximum VFA production observed was 3444.04 mg of acetic acid (HAc)/L (0.58 gCODVFA/gCOD) in a fermentation time of 6 days, with acetic (48.5%), propionic (28.3%), and butyric (13.6%) acids identified as the main metabolites. Additionally, in the assessment of the effect of pH, the maximum VFA production reached 2547.72 mgHAc/L (0.34 gCODVFA/gCOD) at pH 5.9, and acetic acid was identified as the predominant organic acid. Statistically, the fermentation time and pH variables individually affect VFA production from cassava wastewater; however, the interaction between them generated a non-significant effect. Durante el siglo xiii, el desarrollo de las ciudades en Castilla transformó el panorama económico, político e ideológico. El Libro de Apolonio puede analizarse como un testimonio de estos cambios. El poema narra una historia predominantemente urbana; muchos de sus espacios y personajes pueblan y conforman ciudades, cuyas representaciones ayudan a codificar la ideología monárquica y legitimar el control de las urbes por la Corona, mediante su subordinación a los dominios del rey Apolonio quien, a su vez, personifica la figura del gobernante ideal para la supervivencia y prosperidad de las ciudades. De tal suerte, dichas representaciones buscan reafirmar el sistema monárquico como el único capaz de imponer orden, ley y justicia en la vida urbana, siempre que gobierne un monarca virtuoso y sus consejeros, capaces de proveer, también, justicia en aquellos espacios que impulsan el desarrollo de las ciudades: los mercados.

En este artículo se analizan los juicios y valoraciones expresados en torno a la obtención y el uso de la riqueza en el Libro del cavallero Zifar, así como las representaciones de la fortuna material dentro de las historias, las sentencias y los exempla del texto; asimismo, se intenta destacar la influencia que estos elementos tendrían en las prácticas, hábitos ndividuales y sociales de los escuchantes o lectores. Tanto juicios como representaciones pueden considerarse una política de la riqueza. El enfoque de este análisis pretende resaltar la relevancia de la riqueza material en esta obra, ya que el devenir de la historia principal está impulsado por el deseo de Zifar de recuperar el esplendor perdido de su linaje. El encumbramiento del personaje y su familia dependen directamente de la posesión de fortuna material, como ocurre normalmente en una sociedad estamental como la que se representa en el libro.

Abstract

Lathyrane-type diterpenes have a wide range of biological activities. Among them, euphoboetirane A (1) exerts neurogenesis-promoting activity. In order to increase the structural diversity of this type of lathyrane and explore its potential use in neurodegenerative disorders, the biotransformation of 1 by Streptomyces puniceus BC-5GB.11 has been investigated. The strain BC-5GB.11, isolated from surface sediments collected from the intertidal zone of the inner Bay of Cadiz, was identified as Streptomyces puniceus, as determined by phylogenetic analysis using 16S rRNA gene sequence. Biotransformation of 1 by BC-5GB.11 afforded five products (3–7), all of which were reported here for the first time. The main biotransformation pathways involved regioselective oxidation at non-activated carbons (3–5) and isomerization of the ∆12,13 double bond (6). Abstract

The microbiota is attributed to be important for initial soil formation under extreme climate conditions, but experimental evidence for its relevance is scarce. To fill this gap, we investigated the impact of in situ microbial communities and their interrelationship with biocrust and plants compared to abiotic controls on soil formation in initial arid and semiarid soils. Additionally, we assessed the response of bacterial communities to climate change. Topsoil and subsoil samples from arid and semiarid sites in the Chilean Coastal Cordillera were incubated for 16 weeks under diurnal temperature and moisture variations to simulate humid climate conditions as part of a climate change scenario. Our findings indicate that microorganism-plant interaction intensified aggregate formation and stabilized soil structure, facilitating initial soil formation. Interestingly, microorganisms alone or in conjunction with biocrust showed no discernible patterns compared to abiotic controls, potentially due to watermasking effects. Arid soils displayed reduced bacterial diversity and developed a new community structure dominated by Proteobacteria, Actinobacteriota, and Planctomycetota, while semiarid soils maintained a consistently dominant community of Acidobacteriota and Proteobacteria. This highlighted a sensitive and specialized bacterial community in arid soils, while semiarid soils exhibited a more complex and stable community. We conclude that microorganism-plant interaction has measurable impacts on initial soil formation in arid and semiarid regions on short time scales under climate change. Additionally, we propose that soil and climate legacies are decisive for the present soil microbial community structure and interactions, future soil development, and microbial responses. Abstract

Plant pathogenic infections causing substantial global food losses are a persistent challenge. This study investigates a potential biocontrol strategy against the necrotrophic fungus Botrytis cinerea using the endophytic fungus Sordaria tomento-alba isolated from Gliricidia sepium in Colombia. Today, synthetic fungicides dominate B. cinerea control, raising environmental and health concerns. S. tomento-alba exhibits notable in vitro effects, inhibiting B. cinerea growth by approximately 60% during co-culture and 50% in double disc co-culture. Additionally, it suppresses botryanes production and produces the compound heptacyclosordariolone, which has proven effective in inhibiting B. cinerea mycelial growth and spore germination in vitro. This biocontrol agent could be a potential eco-friendly alternative to replace synthetic fungicides. Our study provides insights into the chemical and biological mechanisms underpinning the antagonistic activity of S. tomento-alba, emphasizing the need for further research to understand its biosynthesis pathways and optimize its biocontrol potential. It also contributes molecular evidence of fungal interactions with implications for advanced forums in molecular studies in biology and chemistry, particularly in addressing plant pathogenic infections and promoting sustainable agriculture. Abstract

Spray coating and vacuum-assisted resin infusion processes were implemented in this work to develop multifunctional biocomposite laminates. The biocomposites were fabricated using a bio-based epoxy resin reinforced with natural sisal fibers coated with graphene nanoplatelets (GNPs). A systematic characterization of material properties was performed to evaluate the mechanical, thermomechanical, electrical, and piezoresistive behavior of biocomposites with different GNPs contents (0, 1, 4, 6, and 8 wt.%). The mechanical tests revealed that adding GNPs to biocomposites has a slight positive effect on their flexural properties compared to the neat biocomposites (without GNPs). The electrical and thermomechanical tests showed that the electrical conductivity and glass transition temperature of biocomposites containing GNPs were enhanced significantly, achieving average values of 5.19 x 10-4 S/m and 63.29°C (26%), respectively. Regarding electromechanical tests, the biocomposites with 8 wt.% GNPs exhibited an excellent piezoresistive behavior under monotonic loading conditions, achieving a gage factor (strain sensitivity) of 3.56. Bending tests with cyclic loading were also performed, and cyclic reproducibility of the piezoresistive response of the biocomposites after 10 cycles was demonstrated, evidencing that the incorporation of GNPs onto sisal fibers by spray-coating produces an effective formation of conductive networks into biocomposites suitable for sensing applications. Abstract

Background and Objectives: The efficacy of COVID‐19 convalescent plasma (CP) associates with high titres of antibodies. ConPlas‐19 clinical trial showed that CP reduces the risk of progression to severe COVID‐19 at 28 days. Here, we aim to study ConPlas‐19 donors and characteristics that associate with high anti‐SARS‐CoV‐2 antibody levels. Materials and Methods: Four‐hundred donors were enrolled in ConPlas‐19. The presence and titres of anti‐SARS‐CoV‐2 antibodies were evaluated by EUROIMMUN anti‐SARS‐CoV‐2 S1 IgG ELISA. Results: A majority of 80.3% of ConPlas‐19 donor candidates had positive EUROIMMUN test results (ratio ≥1.1), and of these, 51.4% had high antibody titres (ratio ≥3.5). Antibody levels decline over time, but nevertheless, out of 37 donors tested for an intended second CP donation, over 90% were still EUROIMMUN positive, and nearly 75% of those with high titres maintained high titres in the second sample. Donors with a greater probability of developing high titres of anti‐SARS‐CoV‐2 antibodies include those older than 40 years of age (RR 2.06; 95% CI 1.24–3.42), with more than 7 days of COVID‐19 symptoms (RR 1.89; 95% CI 1.05–3.43) and collected within 4 months from infection (RR 2.61; 95% CI 1.16–5.90). Male donors had a trend towards higher titres compared with women (RR 1.67; 95% CI 0.91–3.06). Conclusion: SARS‐CoV‐2 CP candidate donors' age, duration of COVID‐19 symptoms and time from infection to donation associate with the collection of CP with high antibody levels.
