This article examines the implementation, participation rates, and potential determinants of participation in the digital addiction prevention program "ready4life." A two-arm cluster-randomized trial recruited German vocational students via class-based strategies. Intervention group received 16 weeks of in-app coaching; the control group received health behavior information, with coaching offered after 12 months. Potential determinants of participation were analyzed based on class and individual characteristics. Out of 525 contacted schools, 35 participated, enrolling 376 classes. Implementation during the pandemic required flexible adjustments, with 49.7% of introductions conducted in person, 43.1% digitally via online streaming, and 7.2% received a video link via email. Despite challenges, 72.3% of the vocational students downloaded the app, and 46.7% gave informed consent. Participation rates were highest among (associate) professionals, vocational grammar school classes, classes introduced by females, younger individuals, members of the project team, and classes introduced face-to-face. Female gender, lower social competencies, lifetime cannabis use, higher problematic internet use, and higher perceived stress were associated with higher individual participation. The study highlights the importance of proactive outreach and personalized interventions for addiction prevention programs in vocational schools. While reached students aligned with the aims of the app, tailored recruitment strategies could enhance engagement among under-represented groups.

Abstract Background Social support is relevant to studying well-being, quality of life, and health during aging, particularly in people over 50. Therefore, brief instruments that allow its measurement within the clinical evaluation and research processes are necessary. The ENRICH Social Support Scale (ESSI) is a brief and easy-to-use instrument that measures the perception of social support; however, its psychometric properties in people over 50 in the Chilean context have yet to be tested. Method This study had a non-experimental, longitudinal panel design in which a sample of 303 people over 50 years of age (M = 55.14 years, SD = 5.19; 52% women) were administered a survey incorporating sociodemographic variables, the ESSI and the Newsom Social Negativity Scale. 277 participants completed the follow-up survey (M = 56.75 years, SD = 5.15; 54% women). Confirmatory factor analysis (CFA), structural equation modeling (SEM), invariance analysis, and internal consistency tests were performed to determine the psychometric properties. Results The six items from the scale load in a single-factor model obtained an excellent fit to the data and reliability coefficients (α = 0.902; ω = 0.904). The SEM analysis evidenced an inverse correlation between the ESSI and Newsom's social negativity scale, demonstrating evidence of construct validity. Furthermore, in the sex invariance analyses, the scalar level was reached, implying that the items’ meanings are the same for men and women. Finally, the residual level was reached in the temporal invariance analyses, which means that the scale items are consistent in time. Conclusions The ESSI is a reliable and valid tool to be used in the Chilean context both in the clinical setting and in research on people over 50. The evidence obtained supports its usability to appropriately measure the perception of social support, which is relevant considering that it has been associated with reports of higher quality of life and lower mortality and morbidity during aging.

Introduction Growing evidence in pediatric oncology underscores the significant benefits of physical activity (PA) during and after treatment (Benzing et al., 2021, 2022; Wurz et al., 2021). However, physical activity promotion in acute care settings within Swiss pediatric oncology units remains underdeveloped. To address this gap, the KiKli Fit program was developed and implemented at the Department of Pediatric Oncology and Hematology at the University Hospital Bern over the last 1.5 years. The aim of this research project was to accompany this process with implementation research to evaluate and support the integration of the developed PA program into clinical practice. Methods This study was conducted within the Consolidated Framework for Implementation Research (CFIR), which guided the PA program's development, implementation, and evaluation. Qualitative interviews were conducted with children, parents, nurses, physiotherapists, psychologists, doctors, a sport scientist leading the PA program, and two implementation leaders to gather comprehensive, multi-perspective insights. In total 15 interviews explored attitudes, perceived barriers, facilitators, and expectations regarding implementing the PA program in the pediatric oncology setting. Interview questions were tailored to each stakeholder group, ensuring relevance to their specific roles and experiences. Data was analyzed deductively using qualitative content analysis. Results Data analysis is currently ongoing. Preliminary findings indicate strong acceptance of the PA program among stakeholders, including patients, parents, and healthcare professionals. Children and their families expressed enthusiasm for engaging in structured PA, reporting perceived benefits such as reduced treatment-related side effects, increased PA levels, and improved self-esteem. Stakeholders also highlighted challenges, including resource limitations and logistical barriers, that must be addressed for successful program integration. Notably, healthcare professionals from other hospital units, particularly those caring for children with chronic illnesses, expressed interest in adapting the intervention to benefit additional clinical populations. Discussion/Conclusion Preliminary findings highlight the value of a participatory approach in implementing a PA program in pediatric oncology. High stakeholder acceptance and reported benefits, such as reduced side effects and improved self-esteem, demonstrate the program’s potential. The CFIR framework helped identify barriers, including logistical and resource constraints, which require attention for successful integration. Interest from other hospital units suggests the program’s broader applicability. Future efforts will focus on completing data analysis and exploring adaptations for other clinical settings. Introduction Pediatric cancer patients are particularly vulnerable to short- and long-term side and late-effects in physical and mental health caused by their disease and its treatment (Benzing et al., 2021, 2022; Robison & Hudson, 2013).Physical activity (PA) has been shown to benefit various physical and mental health domains. Emerging research in pediatric oncology increasingly demonstrates the positive impact of PA both during and after treatment. However, PA interventions in acute care settings within Swiss pediatric oncology remain rare. Therefore, the KiKli Fit project was initiated to design and implement a tailored PA program at the Department of Pediatric Oncology and Hematology at the University Hospital Bern. Methods First, a needs analysis was conducted through 14 qualitative interviews with patients to gather insights into their preferences and requirements for a PA intervention. Second, ward staff participated in an online questionnaire to identify structural and systemic opportunities and barriers to implementing such an intervention. Third, a group discussion involving ten ward staff members (including psychologists, nurses, doctors, and physiotherapists) was held to develop a concept for the PA program collaboratively. Building on this, a PA program was designed and implemented over 1.5-years. During this time, various formal and informal assessments were conducted to ensure quality assurance. Results Patients reported various negative side effects and low physical activity levels. Furthermore, they strongly desired greater variety during hospitalization and emphasized the need for a motivating and guided PA program. Similarly, hospital staff supported the implementation of such programs and advocated for the early integration of PA promotion into acute therapy. However, they identified several barriers to participation, including medical constraints, chemotherapy administration, fatigue, visits, or lack of motivation. Importantly, incorporating such programs into the routine of hospital care presents another significant challenge. Discussion/Conclusion The development and practice of the KiKli Fit program highlight the importance of involving all stakeholders to ensure a tailored and feasible intervention. The participatory approach, including input from patients and interdisciplinary staff, was essential for aligning the program with patient needs and hospital operations. Continuous communication and collaboration helped address challenges, such as logistical barriers, facilitating integration into hospital routines. Future efforts should focus on sustaining interdisciplinary collaboration and evaluating the long-term effects of the program to facilitate an integration of PA programs into standard care in Swiss Children’s hospitals. Introduction Young cancer patients (YCP) face high risks of acute and long-term physical and mental health effects resulting from their malignancy and its treatment (Benzing et al., 2022; Siegwart et al., 2020). Physical activity (PA) is increasingly recognized as a strategy to address physical, mental, cognitive, and social challenges faced by YCP and survivors (Braam et al., 2016). This systematic review aimed to summarize findings from randomized controlled trials (RCTs) and clinical control trials (CCTs) evaluating the impact of PA interventions on physical and mental health outcomes in YCP and survivors. Methods A systematic search was conducted in APA PsycINFO, PubMed, SPORTDiscus, Web of Science, Embase, and Scopus. We examined RCTs and CCTs investigating PA interventions in children and adolescents with cancer, during or after treatment, that reported outcomes related to physical or mental health and/or quality of life. Two authors independently screened studies for inclusion, extracted data, and assessed the risk of bias. Results Of 1,164 articles screened, 32 RCTs and CCTs met eligibility criteria, involving 1,315 participants aged 4–19 years. Studies included inpatients (18), survivors (11), or both (2). Chronic PA interventions, such as aerobic, resistance, or combined motor-cognitive training, were implemented. All studies reported physical health outcomes, primarily focusing on cardiorespiratory fitness, muscle strength, and fatigue, while 18 studies assessed psychological outcomes, including depression and quality of life. Four studies objectively evaluated cognitive functions. Benefits were most frequently observed (>50%) for strength, fatigue, and self-efficacy, with no negative effects reported. Discussion/Conclusion Despite variability in session durations and intervention lengths, this review highlights the broad benefits of PA for YCP and survivors, with no indication of negative effects. Most studies have focused on physical and mental health outcomes (Braam et al., 2016), with limited attention to cognitive performance and motor ability, despite the high risk of cognitive late effects in this population (Krull et al., 2018). High-quality RCTs are needed to better understand the role of PA in motor and cognitive development among YCP and survivors. Invasive parasites that expand their natural range can be a threat to wildlife biodiversity and may pose a health risk to non-adapted, naive host species. The invasive giant liver fluke, Fascioloides magna, native to North America, has extended its range in Europe and uses mainly red deer (Cervus elaphus) as definitive hosts. The penetration of the intestinal barrier by the young flukes to reach the liver via the abdominal cavity as well as the release of fluke metabolism products and excreta with the bile and/or changes in the microbial community of the biliary system may enable the translocation of intestinal bacteria across the intestinal barrier and, in turn, could be associated with inflammation and changes in the intestinal bacterial community. The gut commensal community plays a key role in host nutrition and interacts with cells of the immune system to maintain host health. For this study, the gut bacterial community of red deer infected with F. magna and of non-infected red deer from one of the largest forest ecosystems in Central Europe, located on the border between the Czech Republic and Germany, was investigated. The individual fluke burden was associated with changes in the gut microbial composition of the gut of infected individuals, whereas the diversity and composition of the gut bacteria were only slightly different between fluke-infected and uninfected deer. Several bacterial taxa at the genus level were unique to individuals carrying either one or many liver flukes. Our results suggest that the microbiota of red deer is stable to perturbation by low numbers of F. magna. However, a larger parasite burden may cause changes in the gut microbial composition in definitive hosts implying that non-invasive fecal microbiome assessments could serve as indicator for wildlife health monitoring.

Transhumanism is a school of thought that promotes the enhancement of humanity through technological intervention (e.g., cloning, gene therapies, uploading one’s mind to a computer, nanotechnology). Due to its aims of altering evolutionary processes (Bostrom, 2005), transhumanism is highly controversial (Sinicki, 2015). The ideology finds support from younger men, as well as those engaged in science-fiction literature (Gangadharbatla, 2020; Koverola et al., 2022). The present study aimed to investigate the role of gender and specific science fiction fan identities as predictors of transhumanism in three different samples of fandoms affiliated with science-fiction (e.g., anime fans, furries, and Star Wars fans) as well as in a control sample of college students. Participants (N = 6840) responded to a novel measure of transhumanist orientation in either an online or in-person survey. The findings indicated that men were the most likely to endorse transhumanism, as were fans of Star Wars and furries. Overall, the present study supports theorizing that transhumanism may be an influential motif in the science-fiction genre, as well as an appealing ideology for men.

There is an ongoing debate about whether digital media undermines or enhances the ability of stakeholders—in particular, secondary stakeholders—to influence a firm’s policies and practices. One prevailing perspective (Barnett, Henriques & Husted, 2020; Barnett, Henriques & Husted, 2024) is that digital media leads to slacktivism, a token display of support, which impedes the ability of stakeholders to act collectively and makes it easier for firms to dismiss stakeholder claims. Another perspective (Colleoni, Zyglidopoulos & Illia, 2024) argues that even though synchronicity may be difficult to achieve, digital media in the form of hashtags makes it easier for heterogeneous stakeholders to converge around a broader demand for societal change and indirectly pressure firms to change their policies and practices. In this exchange, we expand on Colleoni, Zyglidopoulos, and Illia’s (2024) arguments by encompassing other forms of digital activism beyond generic hashtags. Drawing from the logic of connective action, we argue that two universal pathways—connectivity and velocity—applicable across a wide range of digital activism, are reshaping and enhancing stakeholders’ ability to influence a firm’s social policies and practices. Chamois are mountain ungulates (Artiodactyla: Caprinae) which inhabit several medium- and high-altitude mountain ranges from southern Europe to the Near East. The first findings of Sarcocystis cysts in the musculature of chamois were reported in the 1970s. However, only limited work on the epidemiology of sarcocystosis and the identification of the species of Sarcocystis in chamois has been carried out in the past. The present study aimed to provide, for the first time, data on the prevalence and intensity of Sarcocystis spp. Infection in native Alpine chamois using a histology examination of heart and/or diaphragm tissue samples collected from 216 chamois (40 kids [Sarcocystis spp. infection (95%CI 72.5–84.4%). Sarcocysts were more frequently recorded in the heart vs. diaphragm (72.4% vs. 56.4%; p = 0.0021), and diaphragm positivity was associated with heart positivity (p = 0.0001). The sarcocyst prevalence (heart and/or diaphragm) was significantly (p < 0.001) lower in the kids than in the older chamois (27.1% vs. 88.6%, respectively); however, it did not differ between the sexes, regardless of the chamois’ age (p > 0.3). The intensity of infection was generally low (2 muscle cut) in both heart-positive and diaphragm-positive animals (94.7% and 93.7%, respectively). The heart tissue yielded higher sarcocyst counts than the diaphragm tissue (p < 0.001). Both the heart and diaphragm sarcocyst counts were significantly (p < 0.001) lower in the kids than in the older chamois. Sarcocystis spp. infection was demonstrated to be prevalent in chamois in Germany, but its intensity is apparently low. Further studies are desired to identify the species of Sarcocystis parasitizing the chamois using both phenotypic and molecular characteristics.

The biodiversity of parasitic mites of the Demodecidae, infesting mammalian carnivorans, is poorly understood. To date, 18 host-specific species have been described, including four each from domestic dogs and cats, and only 10 from wild carnivores, known from single or duplicate reports. No data is available on the level of infestation of wild populations, or the co-occurrence of different demodecids in the same host, as only single species have been identified in individual hosts. A convenient model for such studies turned out to be the European polecat Mustela putorius, in which a new genus and species, Miridex putorii, was recently described in the vibrissae region of the skin of the head. Our study reveals that M. putorii co-occur with other species: Demodex putorii sp. nov. (associated with the hairless skin of the head) and Demodex foetorii sp. nov. (associated with hairy skin). Thus, the present study provides descriptions of species new to science. It also provides the first analysis of the occurrence of Demodecidae in wild mammalian carnivorans at the species and individual animal level based on the co-occurrence of three demodecid mite species in the skin of the head in M. putorius. Demodecid mites were found in 75.7% of 37 polecats; M. putorii showed the highest prevalence (56.8%), and D. putorii showed the highest abundance in the skin (mean 10.8 mites in 9 cm2). The three species co-occurred in 5.4% of the polecats, and two species in 27.0%. Despite the very high abundances, infestation was not associated with gross skin pathology (a feature of stable parasite–host systems formed by long-term co-evolution). Linguatula serrata, commonly known as the dogs’ ‘tongue-worm’, is an arthropod endoparasite of the class Pentastomida infesting chiefly canids as definitive hosts and herbivores as intermediate hosts. Adult L. serrata usually reside in the upper respiratory tract, such as the nasal cavity and sinuses, and the larval stages are encapsulated in various visceral organs, respectively. This report presents the first case of a L. serrata nymphal pulmonary infestation in a cat from Albania and adds to the description of the overall rare cases of this parasitic infestation in domestic cats. Discussion of this case together with a comprehensive review of the literature allows us to conclude that cats are susceptible to the infestation with L. serrata when ingesting the parasite’s eggs and allow for the development of the nymphal stage of the parasite in several visceral organs. Therefore, cats may serve as accidental intermediate hosts in the life cycle of L. serrata but are unlikely to be of epidemiological relevance. There is no evidence that domestic cats can act as definitive hosts of L. serrata.

In the course of a survey of the endoparasites of Alpine ibex (Capra ibex) from Austria, examination of the lungs revealed male Protostrongylus nematodes presenting morphological characters which differed from those of the three Protostrongylus species previously reported from this host. Fragments of 16 adult male and 4 adult female nematodes extracted from the lung tissue of two female ibex following peptic digestion were subjected to close microscopic examination. Based on their morphology, the lungworms were identified as Protostrongylus caprae Zdzitowiecki et Boev, 1971. This species was originally described from Siberian ibex (Capra sibirica) in Asia and previously only reported from this host from Kazakhstan and Mongolia. The identification of P. caprae in Alpine ibex represents a new host and geographical record and reinforces the interest to further study the parasite diversity of wild ungulates for a better understanding of complex host-parasite associations and biogeography.

Abstract Humans are subject to a variety of cognitive biases, such as the framing-effect or the gambler's fallacy, that lead to decisions unfitting of a purely rational agent. Previous studies have shown that the ventromedial prefrontal cortex (vmPFC) plays a key role in making rational decisions and that stronger vmPFC activity is associated with attenuated cognitive biases. Accordingly, dysfunctions of the vmPFC are associated with impulsive decisions and pathological gambling. By applying a gambling paradigm in a between-subjects design with 33 healthy adults, we demonstrate that vmPFC excitation via transcranial direct current stimulation (tDCS) reduces the framing-effect and the gambler's fallacy compared to sham stimulation. Corresponding magnetoencephalographic data suggest improved inhibition of maladaptive options after excitatory vmPFC-tDCS. Our analyses suggest that the underlying mechanism might be improved reinforcement learning, as effects only emerge over time. These findings encourage further investigations of whether excitatory vmPFC-tDCS has clinical utility in treating pathological gambling or other behavioral addictions.

Abstract Background The zoonotic intracellular alpha-proteobacterium Anaplasma phagocytophilum is a tick-transmitted pathogen. The associations between vertebrate reservoirs and vectors are described as wide-ranging, and it was previously shown that the pathogenicity of A. phagocytophilum differs depending on the combination of pathogen variant and infected host species. This leads to the question of whether there are variations in particular gene loci associated with different virulence. Therefore, this study aims at clarifying existing host-variant combinations and detecting possible reservoir hosts. To understand these interactions, a complex toolset for molecular epidemiology, phylogeny and network theory was applied. Methods Sequences of up to four gene loci (msp4, msp2, groEL and 16S rRNA) were evaluated for different isolates from variable host species, including, for example, dogs, cattle and deer. Variant typing was conducted for each gene locus individually, and combinations of different gene loci were analysed to gain more detailed information about the genetic plasticity of A. phagocytophilum. Results were displayed as minimum spanning nets and correlation nets. Results The highest diversity of variants for all gene loci was observed in roe deer. In cattle, a reduced number of variants for 16S rRNA [only 16S-20(W) and 16S-22(Y)] but multiple variants of msp4 and groEL were found. For dogs, two msp4 variants [m4-20 and m4-2(B/C)] were found to be linked to different variants of the other three gene loci, creating two main combinations of gene loci variants. Cattle are placed centrally in the minimum spanning net analyses, indicating a crucial role in the transmission cycles by possibly bridging the vector-wildlife cycle to infections of humans and domestic animals. The minimum spanning nets confirmed previously described epidemiological cycles of the bacterium in Europe, showing separation of variants originating from wildlife animals only and a set of variants shared by wild and domestic animals. Conclusions In this comprehensive study of 1280 sequences, we found a high number of gene variants only occurring in specific hosts. Additionally, different hosts show unique but also shared variant combinations. The use of our four gene loci expand the knowledge of host–pathogen interactions and may be a starting point to predict future spread and infection risks of A. phagocytophilum in Europe. Network visualization is one of the most widely used tools in digital humanities research. The idea of uncertain or “fuzzy” data is also a core notion in digital humanities research. Yet network visualizations in digital humanities do not always prominently represent uncertainty. In this article, we present a mathematical and logical model of uncertainty as a range of values which can be used in network visualizations. We review some of the principles for visualizing uncertainty of different kinds, visual variables that can be used for representing uncertainty, and how these variables have been used to represent different data types in visualizations drawn from a range of non-humanities fields like climate science and bioinformatics. We then provide examples of two diagrams: one in which the variables displaying degrees of uncertainty are integrated/pinto the graph and one in which glyphs are added to represent data certainty and uncertainty. Finally, we discuss how probabilistic data and what-if scenarios could be used to expand the representation of uncertainty in humanities network visualizations.

Abstract Background Hippoboscid flies (Diptera: Hippoboscidae), also known as louse flies or keds, are obligate blood-sucking ectoparasites of animals, and accidentally of humans. The potential role of hippoboscids as vectors of human and veterinary pathogens is being increasingly investigated, but the presence and distribution of infectious agents in louse flies is still unknown in parts of Europe. Here, we report the use of molecular genetics to detect and characterize vector-borne pathogens in hippoboscid flies infesting domestic and wild animals in Austria. Methods Louse flies were collected from naturally infested cattle (n = 25), sheep (n = 3), and red deer (n = 12) across Austria between 2015 and 2019. Individual insects were morphologically identified to species level and subjected to DNA extraction for molecular pathogen screening and barcoding. Genomic DNA from each louse fly was screened for Borrelia spp., Bartonella spp., Trypanosomatida, Anaplasmataceae, Filarioidea and Piroplasmida. Obtained sequences of Trypanosomatida and Bartonella spp. were further characterized by phylogenetic and haplotype networking analyses. Results A total of 282 hippoboscid flies corresponding to three species were identified: Hippobosca equina (n = 62) collected from cattle, Melophagus ovinus (n = 100) from sheep and Lipoptena cervi (n = 120) from red deer (Cervus elaphus). Molecular screening revealed pathogen DNA in 54.3% of hippoboscids, including infections with single (63.39%), two (30.71%) and up to three (5.90%) distinct pathogens in the same individual. Bartonella DNA was detected in 36.9% of the louse flies. Lipoptena cervi were infected with 10 distinct and previously unreported Bartonella sp. haplotypes, some closely associated with strains of zoonotic potential. DNA of trypanosomatids was identified in 34% of hippoboscids, including the first description of Trypanosoma sp. in H. equina. Anaplasmataceae DNA (Wolbachia spp.) was detected only in M. ovinus (16%), while < 1% of the louse flies were positive for Borrelia spp. and Filarioidea. All hippoboscids were negative for Piroplasmida. Conclusions Molecular genetic screening confirmed the presence of several pathogens in hippoboscids infesting domestic and wild ruminants in Austria, including novel pathogen haplotypes of zoonotic potential (e.g. Bartonella spp.) and the first report of Trypanosoma sp. in H. equina, suggesting a potential role of this louse fly as vector of animal trypanosomatids. Experimental transmission studies and expanded monitoring of hippoboscid flies and hippoboscid-associated pathogens are warranted to clarify the competence of these ectoparasites as vectors of infectious agents in a One-Health context. Background and aimsChildren have been vastly overlooked in Internet Gaming Disorder (IGD) and Hazardous Gaming research so far. The diagnoses are listed in different ICD-11 chapters (addiction vs. problematic health condition) and are thus considered as distinct constructs. However, screening tools for children do not exist yet. We aimed to investigate the psychometric properties of an existing IGD screening tool modified to also assess Hazardous Gaming in children. Further, we aimed to compare the dissimilarity and overlap between (subclinical) IGD and Hazardous Gaming in children.MethodsThe study analyzed data from a mixed school and clinical sample. Data from N = 871 children aged between 8 and 12 years of age (M = 10.3, SD = 0.90) were analyzed. Data were collected via the Video Game Dependency Scale (CSAS) in its parent report version, which was adapted to assess Hazardous Gaming symptoms in addition to the IGD symptoms. Item analyses and reliability and factor analyses were conducted on the Hazardous Gaming version.ResultsThe results show that the adapted CSAS version that assesses Hazardous Gaming symptoms in children mostly shows acceptable psychometric properties. Explorative Factor Analysis (EFA) shows a two-factor structure with one factor of higher order. Additionally, results show that 35.2% of all children meeting the threshold for Hazardous Gaming exclusively meet criteria for Hazardous Gaming but not for (subclinical) IGD. Vice versa, 91.3% of children with IGD also meet the criteria for Hazardous Gaming.DiscussionHazardous Gaming and (subclinical) IGD are distinct constructs with some overlaps and might have a temporal relation. We recommend adding four items to assess Hazardous Gaming using the CSAS and further evaluate the validity. The assessment of Hazardous Gaming in children is crucial because it might occur earlier than subclinical or full-syndrome IGD.

Abstract The framing-effect is a bias that affects decision-making depending on whether the available options are presented with positive or negative connotations. Even when the outcome of two choices is equivalent, people have a strong tendency to avoid the negatively framed option. The ventromedial prefrontal cortex (vmPFC) is crucial for rational decision-making, and dysfunctions in this region have been linked to cognitive biases, impulsive behavior and gambling addiction. Using a financial decision-making task in combination with magnetoencephalographic neuroimaging, we show that excitatory compared to inhibitory non-invasive transcranial direct current stimulation (tDCS) of the vmPFC reduces framing-effects while improving the assessment of loss-probabilities, ultimately leading to increased overall gains. Behavioral and neural data consistently suggest that this improvement in rational decision-making is predominately due to an attenuation of biases towards negative affect (loss-aversion and risk-aversion). These findings recommend further research towards clinical applications of vmPFC-tDCS as in addictive disorders.

IntroductionStudies suggest an involvement of the ventromedial prefrontal cortex (vmPFC) in reward prediction and processing, with reward-based learning relying on neural activity in response to unpredicted rewards or non-rewards (reward prediction error, RPE). Here, we investigated the causal role of the vmPFC in reward prediction, processing, and RPE signaling by transiently modulating vmPFC excitability using transcranial Direct Current Stimulation (tDCS).MethodsParticipants received excitatory or inhibitory tDCS of the vmPFC before completing a gambling task, in which cues signaled varying reward probabilities and symbols provided feedback on monetary gain or loss. We collected self-reported and evaluative data on reward prediction and processing. In addition, cue-locked and feedback-locked neural activity via magnetoencephalography (MEG) and pupil diameter using eye-tracking were recorded.ResultsRegarding reward prediction (cue-locked analysis), vmPFC excitation (versus inhibition) resulted in increased prefrontal activation preceding loss predictions, increased pupil dilations, and tentatively more optimistic reward predictions. Regarding reward processing (feedback-locked analysis), vmPFC excitation (versus inhibition) resulted in increased pleasantness, increased vmPFC activation, especially for unpredicted gains (i.e., gain RPEs), decreased perseveration in choice behavior after negative feedback, and increased pupil dilations.DiscussionOur results support the pivotal role of the vmPFC in reward prediction and processing. Furthermore, they suggest that transient vmPFC excitation via tDCS induces a positive bias into the reward system that leads to enhanced anticipation and appraisal of positive outcomes and improves reward-based learning, as indicated by greater behavioral flexibility after losses and unpredicted outcomes, which can be seen as an improved reaction to the received feedback.

Campylobacter is the leading cause of bacterial gastroenteritis worldwide and an emerging and neglected pathogen in South America. This zoonotic pathogen colonizes the gastrointestinal tract of a wide range of mammals and birds, with poultry as the most important reservoir for human infections. Apart from its high morbidity rates, the emergence of resistant strains is of global concern. The aims of this work were to determine genetic diversity, presence of antimicrobial resistance determinants and virulence potential of Campylobacter spp. isolated from patients with acute gastrointestinal disease at ‘Clinica Alemana’, Santiago de Chile. The study considered the isolation of Campylobacter spp., from stool samples during a 20-month period (January 2020 to September 2021). We sequenced (NextSeq, Illumina) and performed an in-depth analysis of the genome sequences of 88 Campylobacter jejuni and 2 Campylobacter coli strains isolated from clinical samples in Chile. We identified a high genetic diversity among C. jejuni strains and the emergence of prevalent clonal complexes, which were not identified in our previous reports. While ~40% of strains harbored a mutation in the gyrA gene associated with fluoroquinolone resistance, no macrolide-resistance determinants were detected. Interestingly, gene clusters encoding virulence factors such as the T6SS or genes associated with long-term sequelae such as Guillain-Barré syndrome showed lineage-relatedness. In addition, our analysis revealed a high degree of variability regarding the presence of fT3SS and T6SS effector proteins in comparison to type strains 81-176, F38011, and NCTC 11168 and 488. Our study provides important insights into the molecular epidemiology of this emerging foodborne pathogen. In addition, the differences observed regarding the repertoire of fT3SS and T6SS effector proteins could have an impact on the pathogenic potential and transmissibility of these Latin American isolates, posing another challenge in characterizing the infection dynamics of this emergent and neglected bacterial pathogen.

Among carnivorans, mites of the family Demodecidae are mainly represented by the eight species of the genus Demodex known to cause demodecosis in domestic dog Canis lupus familiaris Linnaeus, 1758 and domestic cat Felis catus Linnaeus, 1758. However, nine other Demodex species from wild carnivorans are also known; in addition they are only known from few records. Previously unknown demodecid mites have been isolated from European polecats, Mustela putorius Linnaeus, 1758, originating from Germany. The specimens are characterized of by an aedeagus with a posterior end located between the opisthosoma and podosoma and an anterior end in the gnathosoma area, with a genital opening in the epistome area; aedeagus length corresponds to 53% (45–59%) of male body length. The mites were isolated from the head skin in 16 of 21 polecats examined (76.2%), mainly in the mystacial vibrissae area (84.8%) and less often in adjacent areas. However, the mite infestation did not appear to cause skin abnormality. Based on the morphological analysis of the adult mites and their morphological ontogenesis, including significant characteristics in demodecid taxonomy, the mite specimens have been classified as representatives of a new species and genus, described as Miridex putorii gen. nov., sp. nov.
