This study builds on the results of a previous study in which six commercial feed products based on organic acids were evaluated with respect to Salmonella contamination of piglets in an artificially challenged seeder model. In the present study, the efficacy of three of these commercial products was assessed for Salmonella reduction in fattening pigs on one closed farm with a natural high Salmonella prevalence. In each of four fattening compartments, one of the following feed treatments was evaluated during two consecutive fattening rounds: (i) butyric acid (active ingredients at 1.3 kg/ton of feed; supplement Al), (ii) a combination of short-chain organic acids (mixture of free acids and salts) and natural extracts (2.92 kg/ton; supplement A4), (iii) a 1:1 blend of two commercial products consisting of medium-chain fatty acids, lactic acid, and oregano oil (3.71 kg/ton; supplement A5+A6), and (iv) a control feed. On the farm, the Salmonella status of the fattening pigs was evaluated by taking fecal samples twice during the fattening period. At the slaughterhouse, samples were collected from the cecal contents and the ileocecal lymph nodes. Salmonella isolates were serotyped and characterized by pulsed-field gel electrophoresis. This farm had a particularly high number of pigs shedding Salmonella with a wide variety of sero- and pulsotypes. Only the feed blend based on the medium-chain fatty acids was able to significantly reduce Salmonella prevalence both on the farm and at the slaughterhouse. With this combined supplement, the Salmonella reduction in the feces at slaughter age, in cecal contents at slaughter, and the lymph nodes was 50, 36, and 67%, respectively, compared with the control animals. The aim of the study was to characterize isolates of Salmonella enterica serovar Enteritidis ( S. Enteritidis) obtained from humans and layer farms in Belgium collected during 2000-2010. Three periods were compared, namely (i) before implementation of vaccination (2000-2004), (ii) during voluntary vaccination (2005-2006) and (iii) during implementation of the national control program ( NCP) for Salmonella including mandatory vaccination against S. Enteritidis (2007-2010). The characteristics compared across time periods were distributions of phage type and multiple-locus variable number tandem-repeat assay ( MLVA). While PT4 and PT21 were predominantly isolated in Belgium in layers and humans before 2007, a significant reduction of those PTs was observed in both populations in the period 2007-2010. The relative proportion of PT4b, PT21c and PT6c was found to have increased considerably in the layer population since 2007. In the human population, PT8, PT1 and the group of 'other' PTs were more frequently isolated compared to the previous periods. When comparing the proportion of the predominant MLVA types Q2 and U2, no significant difference was found between the layer and human population in the three periods and between periods within each category (layer and human). A significant difference in isolate distribution among MLVA clusters I and II was found between human and layer isolates recovered during Period 3 and in the human population between Period 1 and 3. During the last few years, methicillin-resistant Staphylococcus aureus (MRSA) ST398 has been isolated frequently from livestock, especially from pigs and to a lesser extent from cattle and poultry. To gain insight into the distribution of this bacterium in pig farms versus multispecies farms, 30 Belgian farms (10 pig, 10 pig/poultry and 10 pig/cattle farms) were screened for the presence of MRSA. On each farm, 10 nasal swabs were taken from pigs. When present, cattle ( n = 10) were sampled in the nares and poultry ( n = 10) in the nares, earlobes and cloaca. A selection of the obtained isolates were further characterized using multilocus sequence typing (MLST), spa typing, SCC mec typing, pulsed field gel electrophoresis (PFGE), multiple-locus variable-number tandem repeat analysis (MLVA) and antimicrobial susceptibility testing. On 26 of 30 farms, MRSA was isolated from pigs. Furthermore, MRSA was also isolated from poultry and cattle on one pig/poultry and five pig/cattle farms, respectively. All tested MRSA isolates belonged to ST398. Eight spa types (t011, t034, t567, t571, t1451, t2974, t3423 and t5943) were detected, among which t011 predominated. SCC mec cassettes type IVa and V were present in 20% and 72% of the isolates, respectively. When combining the results of the two remaining typing methods, PFGE and MLVA, eighteen genotypes were obtained of which one genotype predominated (56% of the positive farms). All MRSA isolates were resistant to tetracycline. Resistance to trimethoprim, aminoglycosides, macrolides, lincosamides, fluoroquinolones and chloramphenicol was also observed. In conclusion, there was no effect of the farm type on the MRSA status of the pigs. A statistically significant difference was observed when comparing the pig/poultry or the pig/cattle MRSA status on the multispecies farms. Reducing Campylobacter shedding on the farm could result in a reduction of the number of hu-man campylobacteriosis cases. In this study, we first investigated if allicin, allyl disulfide, and garlic oil ex-tract were able to either prevent C. jejuni growth or kill C. jejuni in vitro. Allyl disulfide and garlic oil extract reduced C. jejuni numbers in vitro below a detectable level at a concentration of 50 mg/kg (no lower concen-trations were tested), whereas allicin reduced C. jejuni numbers below a detectable level at a concentration as low as 7.5 mg/kg. In further experiments we screened for the anti- C. jejuni activity of allicin in a fermentation system closely mimicking the broiler cecal environment using cecal microbiota and mucus isolated from C. jeju-ni-free broilers. During these fermentation experiments, allicin reduced C. jejuni numbers below a detectable level after 24 h at a concentration of 50 mg/kg. In con-trast, 25 mg/kg of allicin killed C. jejuni in the first 28 h of incubation, but anti-C. jejuni activity was lost af-ter 48 h of incubation, probably due to the presence of mucin in the growth medium. This had been confirmed in fermentation experiments in the presence of broiler cecal mucus. Based on these results, we performed an in vivo experiment to assess the prevention or reduction of cecal C. jejuni colonization in broiler chickens when allicin was added to drinking water. We demonstrated that allicin in drinking water did not have a statisti-cally significant effect on cecal C. jejuni colonization in broilers. It was assumed, based on in vitro experiments, that the activity of allicin was thwarted by the presence of mucin-containing mucus. Despite promising in vitro results, allicin was not capable of statistically influenc-ing C. jejuni colonization in a broiler flock, although a trend toward lower cecal C. jejuni numbers in allicin-treated broilers was observed. Campylobacter is the most commonly re-ported gastrointestinal bacterial pathogen in humans in many developed countries. During slaughter of broiler flocks, it is difficult to avoid contamination of broiler carcasses. This study aimed to quantify Campylobacter contamination on broiler carcasses at 5 points in the slaughter processing during the slaughter of a Cam-pylobacter-colonized flock by real-time PCR and con-ventional enumeration. In addition, the decontamina-tion effect of neutral electrolyzed oxidizing (EO) water and 1.5% lactic acid (pH 2.0) were evaluated. During processing, the Campylobacter counts on the carcasses declined toward the end of the processing line. The log counts on the carcasses as determined by quantita-tive real-time PCR (qPCR), decreased from 9.37 after scalding to 8.08 after the last cooling step. Enumera-tion of the Campylobacters on plates revealed the same trend, although the counts per carcass were generally 3 logs lower. After scalding, a mean of 6.86 log cfu/car-cass were counted, which decreased to 4.83 log cfu/car-cass after the last cooling step. Submerging carcasses after scalding in EO water gave a significant reduction of 1.31 log cfu/carcass by enumeration on plates and a not significant reduction of 0.53 log cfu/carcass by qPCR. Treatment of the carcasses after the inside-out-side bird washer led to reductions from 0.09 to 0.91 log cfu/carcass, although not significant. After submerging the carcasses in a 1.5% lactic acid solution, significant reductions of 1.62 and 1.24 log cfu/carcass by qPCR and enumeration, respectively, were observed. Spray-ing the carcasses with lactic acid led to nonsignificant reductions of 0.68 log cfu/carcass determined by qPCR and 0.26 log cfu/carcass by enumeration. Bacterial gastroenteritis caused by ther-motolerant Campylobacter species, mainly Campy-lobacter jejuni, has been the most reported zoonotic disease in many developed countries in recent years. Reducing Campylobacter shedding on the farm could result in a reduction of the number of campylobacterio-sis cases. In 2 independent broiler seeder experiments, in which broiler chickens were orally inoculated with 2 amounts of Enterococcus faecalis MB 5259, we es-tablished whether a live E. faecalis strain was capable of reducing cecal Campylobacter colonization in broiler chickens. In previous in vitro experiments it has been demonstrated that this E. faecalis MB 5259 displays anti- Campylobacter activity. The effect of pH and bile salts on E. faecalis MB 5259 showed that growth and survival of E. faecalis MB 5259 can be impaired dur-ing passage through the gastrointestinal tract of broiler chickens. Despite these results E. faecalis MB 5259 was capable of colonizing the broiler ceca. Contrary to the in vitro experiments, in which E. faecalis MB 5259 in-hibited C. jejuni MB 4185 growth, no inhibition was observed in the in vivo experiments independent of the inoculum size. Since 2007, a national Salmonella control program including obligatory vaccination has been on-going in Belgium. In this context, the aim of the pres-ent study was to investigate the diversity of Salmonella enterica serovar Enteritidis isolates on 5 persistently contaminated Belgian layer farms and to examine the potential sources and transmission routes of Salmonella Enteritidis contamination on the farms during succes-sive laying rounds. A collection of 346 Salmonella iso-lates originating from the sampled farms were char-acterized using a combination of multilocus variable number of tandem repeat analysis (MLVA) and phage typing (PT). On each farm, one or 2 dominant MLVA-PT types were found during successive laying cycles. The dominant MLVA type was different for each of the individual farms, but some farms shared the same dominant phage type. Isolates recovered from hens' feces and ceca, egg contents, eggshells, vermin (mice, rats, red mites, and flies), and pets (dog and cat feces) had the same MLVA-PT type also found in the inside henhouse environment of the respective layer farm. Per-sistent types were identified in the layer farm inside environment (henhouse and egg collecting area). Fur-thermore, this study demonstrated cross-contamination of Salmonella between henhouses and between the hen-house and the egg collecting area. Additional isolates with a different MLVA-PT type were also recovered, mainly from the egg collecting area. Salmonella represents a major challenge to the pig industry, as pork presents a risk for human salmonellosis. In this study, we have examined the effect of farm type on the prevalence of fattening pigs shedding Salmonella on 12 farms at risk for harboring Salmonella. On six open (grow-to-finish) and six closed (farrow-to-finish) farms, the prevalence of pigs shedding Salmonella was determined on two occasions approximately 2 months apart. The serovar, phage type, and antimicrobial resistance of the obtained Salmonella isolates were determined. On all farms, pigs shedding Salmonella were detected on at least one of the two sampling days. The mean within-herd prevalence was 7.8%. Closed farms were two times less likely to have pigs shedding Salmonella than open farms. On open farms, the odds of finding Salmonella shedding in pigs were 1.9 times higher when sampling was performed at slaughter age than when samples were taken halfway through the fattening period. Salmonella enterica serovar Typhimurium was the most predominant serotype, with a prevalence of 62 to 63% on both farm types. Of all the Salmonella Typhimurium isolates, 65% had the tetraresistant profile ASSuT (ampicillin, streptomycin, sulfonamide, and tetracycline) with or without additional resistance to trimethoprim-sulfonamide. Phage type DT120 seemed to be especially associated with this antimicrobial-resistant profile. The prevalence of Salmonella Typhimurium isolates showing resistance to ampicillin, streptomycin, tetracycline, sulfonamide, trimethoprim-sulfonamide, and lincomycin hydrochloride and spectinomycin sulfate tetrahydrate was significantly higher on open farms than on closed farms. Successively slaughtered poultry flocks were sampled for Salmonella to study the relationship between gastrointestinal colonization of the birds and contamination of the carcasses after slaughter. Samples from 56 broiler flocks and 16 spent layer and breeder flocks were collected in six slaughterhouses. Salmonella isolates were serotyped and further characterized by pulsed-field gel electrophoresis (PFGE). Although only 7 (13%) broiler flocks were colonized with Salmonella at slaughter, carcasses of 31 (55%) broiler flocks were contaminated after slaughter. Concerning the layer and breeder flocks, 11 (69%) flocks were colonized in the gastrointestinal tract, but after slaughter, carcasses of all flocks were contaminated. The Salmonella status determined at the farm did not always correlate to the status at slaughter. On the other hand, the slaughter of Salmonella colonized flocks did not always result in the contamination of the carcasses with the same PFGE types isolated from the gastrointestinal tract. When only uncolonized flocks were slaughtered, the carcasses of flocks were on some occasions still contaminated with Salmonella. This indicates possible cross-contamination from the slaughter equipment or transport crates. The possible colonization of the intestines and contamination of broilers after transport to the slaughterhouse with Campylobacter strains present in cleaned and disinfected transport containers was investigated. Seven broiler flocks with a Campylobacter-free status were sampled once just before loading at the farm and once just before slaughter. On both occasions, samples were also taken from the exterior of the birds and from the intestinal content. Transport containers used to transport the flock were sampled on the farm just before loading the birds. Campylobacters were enumerated and genotyped by flagellin gene A PCR-restriction fragment length polymorphism and pulsed-field gel electrophoresis. In total, 25 of the 35 sampled containers were Campylobacter contaminated, and 30 genotypes were found. Three broiler flocks became colonized on the farm between initial status determination and transport to the slaughterhouse, and three Campylobacter-free flocks were externally contaminated after transport. In none of the seven flocks was evidence found of intestinal colonization or cocolonization due to transport in Campylobacter-contaminated containers. [ABSTRACT FROM AUTHOR]

The relation between internal carriage and surface contamination with thermophilic Campylobacter species in broilers was examined by molecular typing methods. Samples from 39 flecks were collected in three Belgian poultry slaughterhouses. From each flock, crop swabs before slaughter and intestines and neck skins during slaughter were collected. A total of 309 isolates were identified at species level and further characterized by flagellin gene A PCR/restriction fragment length polymorphism and pulsed-field gel electrophoresis. Isolates were identified as Campylobacter jejuni (90%), Campylobacter coli (8.7%), and Campylobacter lari (2.2%), and 27 genotypes could be distinguished by combining the two molecular methods. Seventy-two percent of the flocks arriving at the abattoir were colonized with campylobacters. After slaughter, 79% of the flocks had contaminated neck skins. In six flocks, genotypes isolated from the neck skins were also found in the alimentary tract from previously slaughtered flocks. Four of these flocks were initially free of Campylobacter. Abstract: Many reports described the prevalence of methicillin-resistant Staphylococcus aureus (MRSA) in different livestock animals from one-species farms. However, in no published reports the prevalence on mixed poultry-pig farms was mentioned, nor the possible relation in MRSA colonization between those two species on one farm, and the possible role of the farmer in the dissemination of MRSA between those two species. Furthermore, no data is available on the optimal sampling site to detect MRSA in broilers. Therefore this study aimed to determine the most suitable sample location in broiler chickens for MRSA and the within flock prevalence of MRSA in various broiler flocks and compared this with the MRSA prevalence in pigs, the colonization of the farmer and the contamination in the barn environment in three mixed poultry-pig farms. MRSA was most frequently isolated from the cloaca and nose shell and to a lesser extent from the skin beneath the wing and the pharynx. The relative sensitivity of the different anatomical sites was, 44.4% for the cloaca, 33.3% for the nose shell, 16.7% for the skin beneath the wing and 5.6% for the pharynx. Based upon these relative sensitivities combining cloaca and nose shell would increase the chance of MRSA detection. A rather low within flock prevalence of MRSA varying between 0% and 28% was detected in broilers, whereas in pigs on the same farms the within herd prevalence varied between 82% and 92%. No MRSA contamination in the direct barn environment of the broilers was found, this in contrast to the environment of the pigs, indicating a relationship between MRSA prevalence and contamination in the environment. Two farmers were continuously colonized, while the third one was only once. In conclusion, a major difference was seen in MRSA occurrence between broilers and pigs from the same farm. This may suggest that broilers are naturally less susceptible to MRSA ST398 colonization than pigs. Conversely, short production time in broilers, vacancy of the barn environment during one week and the higher frequency of disinfection might also explain the lower prevalence in broilers. The farmer may play an important role in the dissemination of MRSA from pigs to poultry, especially in mixed farms where pigs are highly colonized and may act as a reservoir for MRSA ST398 carriage in humans. Salmonella contamination sources and transmission routes were studied in 5 Belgian poultry slaughterhouses. Samples from the slaughter and cutting line after cleaning and disinfection were collected, as well as neck skin samples and thighs during slaughter of the first flock. In total, 680 swab and water samples were taken from the slaughter line before slaughter. In all slaughterhouses, Salmonella was notwithstanding cleaning and disinfection still isolated from the slaughter line before start of activities. The prevalence of Salmonella in the plucking area was 10.4% (38/365) (hanging area: 5.0%, scalding tank: 5.8%, plucking machine: 17.0%); in the evisceration room, 1.5% (2/138); and in the cutting area, 2.0% (3/149). No Salmonella (0/28) was found in samples from the chilling line. On neck skin samples taken from the various lines, Salmonella prevalence was 16.1% (48/299) after plucking, 16.0% (48/300) after evisceration, 23.3% (70/300) after chilling; on thighs, prevalence was 10.0% (24/240). Nine Salmonella serotypes were identified of which Salmonella Infantis was the most common serovar (53.8%), especially in slaughterhouse A. Two contamination causes were identified; first, although all flocks had an official Salmonella negative status, this was in one case incorrect and led to an enormous contamination of the neck skins of the flock and the slaughterline (i.e., cooling water). Second, molecular typing revealed cross-contamination from flocks slaughtered 1 d before sampling. Salmonella was apparently not always eliminated by the cleaning and disinfection process and able to contaminate the carcasses of the first slaughtered flock. In conclusion, the results of this study provided practical insights for poultry production to further improve their Salmonella control, for example, Salmonella status determination closer to the slaughter date, to adapt cleaning and disinfection protocols especially for critical machinery and better hygienic designed equipment. In this study we investigated the prevalence and location of Listeria monocytogenes and hygiene indicator bacteria on beef and pig carcasses. Carcasses were sampled after slaughter and before cooling at eight and nine sites on the carcass, respectively. For each sample, detection and enumeration of Listeria was performed, as well as the enumeration of Total Aerobic Counts (TAC) and Enterobacteriaceae. The L. monocytogenes isolates were also typed to determine pulsotypes and clonal complexes (CC). L. monocytogenes was detected on 46% [95% CI: 35–56%] of beef and 22% [95% CI: 11–32%] of pig carcasses. Contamination levels at the different carcass sites differed considerably between beef and pigs. Genetic typing of strains suggests that carcass contamination originates from both incoming animals with transmission during slaughter practices as well as persistent (CC9) contamination from the slaughterhouse environment. These findings can be used to understand the complexity of introduction and persistence of this pathogen in slaughter facilities.
