Bile acids (BAs) are natural metabolites in mammals and have the potential to function as drugs against viral infection. However, the limited understanding of chenodeoxycholic acid (CDCA) receptors and downstream signaling, along with its lower suppression efficiency in inhibiting virus infection limits its clinical application. In this study, we demonstrate that farnesoid X receptor (FXR), the receptor of CDCA, negatively regulates interferon signaling, thereby contributing to the reduced effectiveness of CDCA against virus replication. FXR deficiency or pharmacological inhibition enhances interferon signaling activation to suppress virus infection. Mechanistically, FXR impairs the DNA binding and transcriptional abilities of activated interferon regulatory factor 3 (IRF3) through interaction. Reduced IRF3 transcriptional activity by FXR–IRF3 interaction significantly undermines the expression of Interferon Beta 1 (IFNB1) and the antiviral response of cells, especially upon the CDCA treatment. In FXR-deficient cells, or when combined with Z-guggulsterone (GUGG) treatment, CDCA exhibits a more potent ability to restrict virus infection. Thus, these findings suggest that FXR serves as a limiting factor for CDCA in inhibiting virus replication, which can be attributed to the “signaling-brake” roles of FXR in interferon signaling. Targeting FXR inhibition represents a promising pharmaceutical strategy for the clinical application of BAs metabolites as antiviral drugs.

Abstract Background While depression is increasing worldwide, some patients are diagnosed as having Major Depressive Disorder (MDD), but others are diagnosed with minor depression, however, the potential neuro mechanism is unknown. Methods Sixty-two patients with minor depression, 44 adolescents with MDD and 54 healthy adolescents participated in this study. Functional near-infrared spectroscopy (fNIRS), both HAMD and HAMA data were collected from all of the participants. Results The result indicates the pervasively decreased activation of BA, 11, 21, 45 and 46 were observed in the MDD group and reduced activation of BA 45 was observed in the minor depression group. However, cortical activation was not observed between the minor depression or MDD groups. Cortical activation was also not correlated with the depressive/anxious score in the minor and MDD groups separately. Conclusions Cortical activation was pervasively decreased in the MDD group and slightly reduced in the minor depression group, which may be a potential neural mechanism. As reduced cortical activation in minor depression, interventions in the early stages of minor depression may help slow or even modify the development of the illness.

ABSTRACT: The main purpose of the current study was to investigate the ameliorative effects of bovine milk osteopontin (bmOPN) on the gut dysfunction of pregnant rats fed a high-fat diet (HFD). Bovine milk osteopontin was supplemented at a dose of 6 mg/kg body weight. Bovine milk osteopontin supplementation during pregnancy reduced colonic inflammation of HFD dams, and it also increased the colonic expression of ZO-1 and claudin-4 of HFD dams. Bovine milk osteopontin significantly enriched the relative abundance of Bacteroidetes, whereas it decreased Proteobacteria, Helicobacteraceae, and Desulfovibrionaceae in feces of HFD dams. The levels of isobutyric acid and pentanoic acid in the HFD + bmOPN group were higher than that of the HFD group. Functional predication analysis of microbial genomes revealed that bmOPN supplementation to HFD pregnancies changed 4 Kyoto Encyclopedia of Genes and Genomes pathways including bile acid biosynthesis. Further, bmOPN enriched hepatic taurochenodeoxycholic acid and tauroursodeoxycholic acid plus taurohyodeoxycholic acid in the gut of HFD maternal rats. Our findings suggested that bmOPN improved the gut health of HFD pregnant rats partially through modulating bile acid biosynthesis.

In this study, the effects of Mg on the formation of He bubbles and diffusion behavior of He atoms in cubic silicon carbide (3C–SiC) were investigated by irradiation and annealing experiments as well as first-principles calculations. TEM results indicated that two damage bands were formed in He&Mg irradiated SiC. During annealing, Mg could prevent He atoms from diffusing to the surface, resulting in the formation of the He bubbles in the deeper areas far from Mg-implanted regions, which is helpful in avoiding surface blisters. First-principles calculations were then performed to explore the effects of Mg on the He behavior in SiC. The solution energy, binding energy charge density, bond length, and crystal orbital Hamiltonian population of these elements were calculated to identify their states. The results suggested that the binding capacity between He and Mg was weak, and Mg could increase the diffusion energy barrier of He. Ab initio molecular dynamics (AIMD) simulation showed that Mg could make He in a high-energy unstable state, and force He atom to move toward the vacancy away from Mg, which explains the experimental results.

Abstract Background The positive effects of exposing corals to microorganisms have been reported though how the benefits are conferred are poorly understood. Here, we isolated an actinobacterial strain (SCSIO 13291) from Pocillopora damicornis with capabilities to synthesize antioxidants, vitamins, and antibacterial and antiviral compounds supported with phenotypic and/or genomic evidence. Strain SCSIO 13291 was labeled with 5 (and − 6)-carboxytetramethylrhodamine, succinimidyl ester and the labeled cell suspension directly inoculated onto the coral polyp tissues when nubbins were under thermal stress in a mesocosm experiment. We then visualized the labelled bacterial cells and analyzed the coral physiological, transcriptome and microbiome to elucidate the effect this strain conferred on the coral holobiont under thermal stress. Results Subsequent microscopic observations confirmed the presence of the bacterium attached to the coral polyps. Addition of the SCSIO 13291 strain reduced signs of bleaching in the corals subjected to heat stress. At the same time, alterations in gene expression, which were involved in reactive oxygen species and light damage mitigation, attenuated apoptosis and exocytosis in addition to metabolite utilization, were observed in the coral host and Symbiodiniaceae populations. In addition, the coral associated bacterial community altered with a more stable ecological network for samples inoculated with the bacterial strain. Conclusions Our results provide insights into the benefits of a putative actinobacterial probiotic strain that mitigate coral bleaching signs. This study suggests that the inoculation of bacteria can potentially directly benefit the coral holobiont through conferring metabolic activities or through indirect mechanisms of suppling additional nutrient sources.

This study investigated the effects of exogenous nitric oxide (NO) on growth, antioxidant enzymes, and key nitrogen metabolism enzymes in pepper seedlings under high-temperature stress. In addition, targeted metabolomics was used to study the differential accumulation of amino acid metabolites, thereby providing theoretical support for the use of exogenous substances to mitigate high-temperature stress damage in plants. The results showed that high-temperature stress increased soluble sugar, soluble protein, amino acids, proline, malondialdehyde (MDA), and hydrogen peroxide (H2O2) content, electrolyte leakage, and superoxide anion (O2·-) production rate while altering the activities of antioxidant enzymes [superoxide dismutase (SOD), catalase (CAT), peroxidase (POD) and ascorbate peroxidase (APX)] and key nitrogen metabolism enzymes [nitrate reductase (NR), glutamine synthetase (GS), glutamate dehydrogenase (GDH), and nitric oxide synthase (NOS)]. c-PTIO (2-phenyl-4,4,5,5-tetramethylimidazoline-1-oxyl 3-oxide, an NO scavenger) exacerbates oxidative stress and further reduces NO content and enzyme activities. However, exogenous SNP (sodium nitroprusside, an NO donor) effectively alleviated these adverse effects by enhancing antioxidant defense mechanisms, increasing NO content, and normalizing amino acid metabolite levels (kynurenine, N-acetyl-L-tyrosine, L-methionine, urea, and creatine), thereby maintaining normal plant growth. These findings suggest that SNP can enhance stress tolerance in pepper seedlings by improving osmotic regulation, antioxidant capacity, and nitrogen metabolism, effectively mitigating the damage caused by high-temperature stress.

In order to study the influence of modular filled and composite material forms on the axial crushing and energy absorption properties of structures, modular filled composite structures were constructed, and innovatively applied to the inner side of double-hat beam (DHB) structures in automobiles. The modular filled structures comprise hexagonal, quadrilateral, and triangular sections. By analyzing the collision performance of modular filled DHB structures, significant enhancements were observed in both the sectional characteristics and the specific Mean Crushing Force of modular filled DHBs compared to the conventional double-hat beam structure. These advancements notably improved the plastic deformation characteristics of the structures. Additionally, dynamic weightlessness experiments were conducted to validate the accuracy of the simulation model. Among the proposed schemes, namely QU-5, HE-5, and TR-5, notable improvements in crashworthiness were identified. Specifically, crashworthiness indicators increased by 32.54%, 78.9%, and 116.53%. Compared with other thin-walled structures, modular filled composite DHBs have advantages in axial crushing and energy absorption. By optimizing layout characteristics, the modular filled structures will achieve significant lightweight and energy absorption performance improvements. This work has clear reference value for automotive engineers and scholars to further explore the axial crash safety, platform modularization, and lightweight design of vehicles.

Background: Although many circulating miRNAs (c-miRNAs) are associated with coronary artery disease (CAD), they are far from being the biomarker for CAD diagnosis or risk prediction. Therefore, novel c-miRNAs discovery and validation are still required, especially evaluating their prediction capacity. Objectives: Identify novel CAD-related c-miRNAs and evaluate its risk prediction capacity for CAD. Methods: miRNAs associated with CAD were preliminarily investigated in three paired samples representing pre-CAD stage and CAD stage of three female individuals using the Applied Biosystems miRNA TaqMan® Low-Density Array (TLDA). Then, the candidate miRNAs were further verified in an independent case-control study including 129 CAD patients and 76 controls, and their potential practical value in prediction for CAD was evaluated using a machine learning (ML) algorithm. The accuracy of classification and prediction was assessed with the area under the receiver operating characteristic curve (AUC). Results: TLDA analysis shows that miR-140-3p decreased significantly in CAD-stage (FC = −3.01, P = 0.007). t(1;19)(q23;p13) is one of the most common translocation genes in childhood acute lymphoblastic leukemia (ALL) and is also present in acute myeloid leukemia (AML) and mixed-phenotype acute leukemia (MPAL). This translocation results in the formation of the oncogenic E2A-PBX1 fusion protein, which contains a trans-activating domain from E2A and a DNA-binding homologous domain from PBX1. Despite its clear oncogenic potential, the pathogenesis of E2A-PBX1 fusion protein is not fully understood (especially in leukemias other than ALL), and effective targeted clinical therapies have not been developed. To address this, we established a stable and heritable zebrafish line expressing human E2A-PBX1 (hE2APBX1) for high-throughput drug screening. Blood phenotype analysis showed that hE2APBX1 expression induced myeloid hyperplasia by increasing myeloid differentiation propensity of hematopoietic stem cells (HSPCs) and myeloid proliferation in larvae, and progressed to AML in adults. Mechanistic studies revealed that hE2A-PBX1 activated the TNF/IL-17/MAPK signaling pathway in blood cells and induced myeloid hyperplasia by upregulating the expression of the runx1. Interestingly, through high-throughput drug screening, three small molecules targeting the TNF/IL-17/MAPK signaling pathway were identified, including OUL35, KJ-Pyr-9, and CID44216842, which not only alleviated the hE2A-PBX1- induced myeloid hyperplasia in zebrafish but also inhibited the growth and oncogenicity of human pre-B ALL cells with E2A-PBX1. Overall, this study provides a novel hE2A-PBX1 transgenic zebrafish leukemia model and identifies potential targeted therapeutic drugs, which may offer new insights into the treatment of E2A-PBX1 leukemia.

Different microenvironments shape distinct microbial communities that play a wide range of roles within coral holobionts. To better understand the capabilities of coral symbiotic microbiota, a more comprehensive characterization should be conducted. The gastrovascular cavity of coral is a distinct microenvironment that serves as a crucial link between the coral organism and the reef ecosystem. The species and functional compositions of microbiomes in the gastrovascular cavity are poorly studied. Therefore, this study was designed to investigate the microbial communities associated with mucus, tissue, skeleton, and gastrovascular cavity of Galaxea fascicularis to further explore their microscopic characteristics, species, and functional composition. The results revealed that the microbial cell abundance in the gastrovascular cavity was significantly higher than in the other three compartments, and the volume of microbial cells in the gastrovascular cavity, tissue, and mucus was significantly greater than that in the skeleton. There were significant differences in microbial community and functional composition among the four compartments of G. fascicularis. In particular, the microbial community in the gastrovascular cavity showed a relatively lower species diversity and the lowest network complexity compared to the other three compartments. Additionally, the potential functions of the microbiome in the gastrovascular cavity were enriched in substance metabolism, especially lipid metabolism. Microbial taxa and genes involved in nitrogen fixation, sulfur reduction and sulfur oxidation were less abundant in the gastrovascular cavity. Overall, our results suggested that the gastrovascular cavity was inhabited by a large number of microorganisms and it performs a strong selection process to retain microorganisms that are associated with substrate metabolism, which may be crucial to support the digestive function of the gastrovascular cavity. This also emphasizes the urgent need to explore coral microbial communities at the microscale to comprehend their role in coral health and resilience.

Abstract Mesenchymal stem cells (MSCs) are widely used in cell therapy, tissue engineering, and regenerative medicine because of their self-renewal, pluripotency, and immunomodulatory properties. The microenvironment in which MSCs are located significantly affects their physiological functions. The microenvironment directly or indirectly affects cell behavior through biophysical, biochemical, or other means. Among them, the mechanical signals provided to MSCs by the microenvironment have a particularly pronounced effect on their physiological functions and can affect osteogenic differentiation, chondrogenic differentiation, and senescence in MSCs. Mechanosensitive ion channels such as Piezo1 and Piezo2 are important in transducing mechanical signals, and these channels are widely distributed in sites such as skin, bladder, kidney, lung, sensory neurons, and dorsal root ganglia. Although there have been numerous studies on Piezo channels in MSCs in recent years, the function of Piezo channels in MSCs is still not well understood, and there has been no summary of their relationship to illustrate which physiological functions of MSCs are affected by Piezo channels and the possible underlying mechanisms. Therefore, based on the members, structures, and functions of Piezo ion channels and the fundamental information of MSCs, this paper focused on summarizing the advances in Piezo channels in MSCs from various tissue sources to provide new ideas for future research and practical applications of Piezo channels and MSCs.

Periodontitis is an inflammatory disease caused by bacterial infection directly, and the dysregulation of host immune-inflammatory response finally destroys periodontal tissues. Current treatment strategies for periodontitis mainly involve mechanical scaling/root planing (SRP), surgical procedures, and systemic or localized delivery of antimicrobial agents. However, SRP or surgical treatment alone has unsatisfactory long-term effects and is easy to relapse. In addition, the existing drugs for local periodontal therapy do not stay in the periodontal pocket long enough and have difficulties in maintaining a steady, effective concentration to obtain a therapeutic effect, and continuous administration always causes drug resistance. Many recent studies have shown that adding bio-functional materials and drug delivery systems upregulates the therapeutic effectiveness of periodontitis. This review focuses on the role of biomaterials in periodontitis treatment and presents an overview of antibacterial therapy, host modulatory therapy, periodontal regeneration, and multifunctional regulation of periodontitis therapy. Biomaterials provide advanced approaches for periodontal therapy, and it is foreseeable that further understanding and applications of biomaterials will promote the development of periodontal therapy.

Abstract The seismic vulnerability of interaction system of saturated soft soil and subway station structures was explored in this paper. The coupled nonlinear numerical models of interaction system were established using the u–p formulation of Biot′s theory to describe the saturated two-phase media. A refined finite element model of interaction system was developed to study its nonlinear seismic responses and seismic hazard mechanism. In this study, the multi-yield elastoplastic constitutive model was adopted for the soil, while a fiber section elastoplastic constitutive model was used for the structure. The seismic response of the structure was calculated by inputting the artificial seismic wave obtained from the power spectrum-triangular series method. The maximum inter-story drift angle was taken as a structural performance parameter for the subway station structure. The structural demand cloud was obtained under random ground motion sequences. Based on the probabilistic seismic demand model analysis method, the seismic vulnerability curve of the subway station structure was plotted, and the seismic vulnerability curve was analyzed as per the vulnerability of performance parameters. With the increase of soil strength, the vulnerability index of subway station structure under different peak acceleration ground motion decreased correspondingly. Based on the above vulnerability theory and analysis methods, it can be found from the above vulnerability theory and analysis methods that the subway station structure with established buried depth in saturated soft soil site exhibits a certain degree of safety and reliability, and can meet the seismic fortification goal of "no damage in small earthquakes, repairable in medium earthquakes and no collapse in large earthquakes". The results of vulnerability analysis are in line with the actual seismic survey, and the vulnerability analysis method proposed in this paper can be applied to the vulnerability analysis of underground structures on saturated soft soil foundation.

The scientific field concerned with the study of regeneration has developed rapidly in recent years. Stem cell therapy is a highly promising therapeutic modality for repairing tissue defects; however, several limitations exist, such as cytotoxicity, potential immune rejection, and ethical issues. Exosomes secreted by stem cells are cell-specific secreted vesicles that play a regulatory role in many biological functions in the human body; they not only have a series of functional roles of stem cells and exert the expected therapeutic effects, but they can also overcome the mass limitations of stem cells and are thus considered in the research as an alternative treatment strategy for stem cells. Since dental stem cell-derived exosomes (DSC-Exos) are easy to acquire and present modulating effects in several fields, including neurovascular regeneration and craniofacial soft and hard tissue regeneration processes, they are served as an emerging cell-free therapeutic strategy in various systematic diseases. There is a growing body of research on various types of DSC-Exos; however, they lack systematic elaboration and tabular summarization. Therefore, this review presents the isolation, characterization, and phenotypes of DSC-Exos and focuses on their current status of functions and mechanisms, as well as the multiple challenges prior to clinical applications.

Abstract Follicular atresia triggered by granulosa cell (GC) apoptosis severely reduces female fertility and accelerates reproductive aging. GC apoptosis is a complex process regulated by multiple factors, regulatory axes, and signaling pathways. Here, we report a novel, small regulatory network involved in GC apoptosis and follicular atresia. miR-187, a miRNA down-regulated during follicular atresia in sows, maintains TGFBR2 mRNA stability in sow GCs by directly binding to its 5’-UTR. miR-187 activates the transforming growth factor-β (TGF-β) signaling pathway and suppresses GC apoptosis via TGFBR2 activation. NORHA, a pro-apoptotic lncRNA expressed in sow GCs, inhibits TGFBR2-mediated activation of the TGF-β signaling pathway by sponging miR-187. In contrast, NORFA, a functional lncRNA associated with sow follicular atresia and GC apoptosis, enhances miR-187 and TGFBR2 expression by inhibiting NORHA and activating NFIX. Our findings define a simple regulatory network that controls GC apoptosis and follicular atresia, providing new insights into the mechanisms of GC apoptosis, follicular atresia, and female fertility.

The excavation and utilization of dormancy loci in breeding are effective endeavors for enhancing the resistance to pre-harvest sprouting (PHS) of wheat varieties. CH1539 is a wheat breeding line with high-level seed dormancy. To clarify the dormant loci carried by CH1539 and obtain linked molecular markers, in this study, a recombinant inbred line (RIL) population derived from the cross of weak dormant SY95-71 and strong dormant CH1539 was genotyped using the Wheat17K single-nucleotide polymorphism (SNP) array, and a high-density genetic map covering 21 chromosomes and consisting of 2437 SNP markers was constructed. Then, the germination percentage (GP) and germination index (GI) of the seeds from each RIL were estimated. Two QTLs for GP on chromosomes 5A and 6B, and four QTLs for GI on chromosomes 5A, 6B, 6D and 7A were identified. Among them, the QTL on chromosomes 6B controlling both GP and GI, temporarily named QGp/Gi.sxau-6B, is a major QTL for seed dormancy with the maximum phenotypic variance explained of 17.66~34.11%. One PCR-based diagnostic marker Ger6B-3 for QGp/Gi.sxau-6B was developed, and the genetic effect of QGp/Gi.sxau-6B on the RIL population and a set of wheat germplasm comprising 97 accessions was successfully confirmed. QGp/Gi.sxau-6B located in the 28.7~30.9 Mbp physical position is different from all the known dormancy loci on chromosomes 6B, and within the interval, there are 30 high-confidence annotated genes. Our results revealed a novel QTL QGp/Gi.sxau-6B whose CH1539 allele had a strong and broad effect on seed dormancy, which will be useful in further PHS-resistant wheat breeding.

Summary: Mitochondria play important roles in angiogenesis. However, the mechanisms remain elusive. In this study, we found that mitochondrial ubiquinol-cytochrome c reductase complex assembly factor 3 (UQCC3) is a key regulator of angiogenesis. TALEN-mediated knockout of Uqcc3 in mice caused embryonic lethality at 9.5–10.5 days postcoitum, and vessel density was dramatically reduced. Similarly, knockout of uqcc3 in zebrafish induced lethality post-fertilization and impaired vascular development. Knockout of UQCC3 resulted in slower tumor growth and angiogenesis. Mechanistically, UQCC3 was upregulated under hypoxia, promoted reactive oxygen species (ROS) generation, enhanced HIF-1α stability and increased VEGF expression. Finally, higher expression of UQCC3 was associated with poor prognosis in multiple types tumors, implying a role for UQCC3 in tumor progression. In conclusion, our findings highlight the important contribution of UQCC3 to angiogenesis under both physiological and pathological conditions, indicating the potential of UQCC3 as a therapeutic target for cancer.

To achieve multiple obstacle avoidance tasks consisting of zero-space obstacle avoidance and terminal obstacle avoidance with self-avoidance permission, we propose an algorithm for multiple obstacle avoidance tasks for dual-arm robots. First, we design a self-obstacle avoidance gradient optimization function to implement the self-obstacle avoidance task between the two arms of dual-arm robots by using the pseudo-distance as the basis for the collision judgment. Second, the terminal obstacle avoidance task for dual-arm robots is implemented by applying an adaptive terminal obstacle avoidance velocity. Third, a zero-space obstacle avoidance task for dual-arm robots is realized using an adaptive virtual repulsion force. Finally, a dual-arm robot consisting of two 6-DOF Jaco2 manipulators is simulated in a Matlab environment to verify the obstacle avoidance algorithm of dual-arm robots. The simulation results show that the algorithm proposed in this paper can complete the multi-obstacle avoidance tasks of terminal obstacle avoidance and zero-space obstacle avoidance simultaneously with the premise of realizing the self-obstacle avoidance of dual-arm robots, and has a minor terminal trajectory error, which proves that the algorithm is in the feasibility of completing terminal obstacle avoidance and zero-space obstacle avoidance with the premise of realizing dual-arm robots self-obstacle avoidance.

Abstract Background Peripheral atherosclerosis is a common macrovascular complication of diabetes, but the treatment is limited. Chinese herbal medicine is the complementary and alternative therapy to delay the progression of atherosclerosis and reduce blood glucose and lipids. Shen-Qi Xiao-Tan (SQXT) formula is one of the prescriptions commonly used to treat diabetic peripheral atherosclerosis, but there is still a lack of high-quality evidence-based evidence. Methods This is a randomized, double-blind, placebo-controlled add-on trial that is expected to enroll 114 diabetic patients with peripheral atherosclerosis. After a 2-week run-in period, participants will been randomly assigned in a 1:1 ratio and receive 12 weeks of usual treatment and SQXT formula (treatment group) or usual treatment and placebo (control group). The primary outcome is the change in carotid intima-media thickness from baseline to endpoint. The secondary outcomes are the structure and function of peripheral arteries, blood glucose and lipids, traditional Chinese medicine syndrome score, and quality of life, and safety and endpoint events are evaluated. To explore the therapeutic mechanism through oxidative stress, inflammation, and advanced glycation end products, and lipidomics will be used to screen for biomarkers for diagnosis and efficacy evaluation. Discussion The objective of this trial is to evaluate the efficacy, safety and therapeutic mechanism of SQXT formula in the treatment of diabetic peripheral atherosclerosis. It will obtain high-quality evidence-based evidence and promote the treatment of diabetic macroangiopathy and the research and development of new drugs. Trial registration This trial is registered on Chinese Clinical Trials.gov with number ChiCTR2100047189 on 10 Jun 2021, and has been approved by the Ethical Review Committee of Hospital of Chengdu University of Traditional Chinese Medicine with number 2020KL-080.

The regulation of apple (Malus domestica) fruit texture during ripening is complex and a fundamental determinant of its commercial quality. In climacteric fruit, ripening-related processes are regulated by ethylene (ET), and jasmonate (JA) is also involved in the ethylene biosynthesis pathway, mainly through the transcription factor MYC2. However, the molecular genetic mechanism for fruit ripening processes between the JA and ET signaling pathways still needs to be elucidated. In order to explore how JA regulates apple fruit ripening through ERF4, we used ‘Gala’ and ‘Ralls Janet’ fruit at different developmental stages as experimental materials to determine the fruit firmness and related gene expression analysis. Meanwhile, we carried out different hormone treatments on ‘Gala’ fruit at ripening stage. Here, we show that ERF4 is a core JA signaling hub protein JASMONATE ZIM-DOMAIN (JAZ) interactor that affects ethylene signaling pathways. During fruit development, ERF4 represses the expression of ACS1 and ACO1 by interacting with JAZ, as well as with the JA-activated transcription factor MYC2. Ripening is promoted in JAZ-suppressed apples. Thus, ERF4 acts as a molecular link between ethylene and JA hormone signals, and the natural variation of the ERF4 Ethylene-responsive binding factor-associated amphiphilic repression (EAR) motif decreases repression of ethylene biosynthesis genes.

For the demand of high reliability in the aerospace field, multi-phase electric machines have become a research hotspot for the fault-tolerant ability. Inter-turn short-circuit fault is one of the main fault forms in the electric machine. The fault mechanism, diagnosis method and fault-tolerant control strategy of inter-turn short-circuit fault of the open-winding five-phase permanent-magnet synchronous machine that is used as the starter/generator system are studied in this paper. Firstly, the fault mechanism of the inter-turn short-circuit fault (ITSCF) is analyzed. The characteristics of phase voltage, inter-turn short-circuit current and torque ripple are compared using the phase winding open-circuit method and the phase winding short-circuit method. Secondly, a fault diagnosis method based on the information fusion is proposed, which determines the fault phase firstly and then identifies the inter-turn short-circuit ratio. According to the fault mechanism, a fault-tolerant control method is proposed, in which the accurate inter-turn short-circuit ratio and inter-turn short-circuit current are not required. The proposed fault-tolerant control method has similar torque ripple compared with the control method using accurate inter-turn short-circuit ratio and inter-turn short-circuit current. Finally, the effectiveness of the proposed fault-tolerant control method has been verified by experimental results.

Here the authors report a 33-residue peptide tag which simultaneously increases the solubility and thermostability of multiple scFvs produced in Escherichia coli SHuffle strain.

Environmental heterogeneity in cold seeps is usually reflected by different faunal aggregates. The sediment microbiome, especially the geochemical cycling-associated communities, sustains the ecosystem through chemosynthesis. To date, few studies have paid attention to the structuring and functioning of geochemical cycling-associated communities relating to environmental heterogeneity in different faunal aggregates of cold seeps. In this study, we profiled the microbial community of four faunal aggregates in the Haima cold seep, South China Sea. Through a combination of geochemical and meta-omics approaches, we have found that geochemical variables, such as sulfate and calcium, exhibited a significant variation between different aggregates, indicating changes in the methane flux. Anaerobic methanotrophic archaea (ANME), sulfate-reducing, and sulfide-oxidizing bacteria (SRB and SOB) dominated the microbial community but varied in composition among the four aggregates. The diversity of archaea and bacteria exhibited a strong correlation between sulfate, calcium, and silicate. Interspecies co-exclusion inferred by molecular ecological network analysis increased from non-seep to clam aggregates and peaked at the mussel aggregate. The networked geochemical cycling-associated species showed an obvious aggregate-specific distribution pattern. Notably, hydrocarbon oxidation and sulfate reduction by ANME and SRB produced carbonate and sulfide, driving the alkalization of the sediment environment, which may impact the microbial communities. Collectively, these results highlighted that geofluid and microbial metabolism together resulted in environmental heterogeneity, which shaped the C and S cycling-associated microbial community.

Supercapacitors are extensively used in urban rail transit, electric vehicles, renewable energy storage, electronic products, and the military industry due to its long cycle life and high power density. Porous carbon materials are regarded as promising anode materials for supercapacitors due to their high specific surface areas and well-developed pore structures. However, the over-developed pore structure often results in poor conductivity and reduced cycle stability due to the destruction of a carbon skeleton. Herein, we introduce an advanced strategy for preparing porous carbon with high specific surface areas (3333 m2 g−1), high electrical conductivity (68.6 S m−1), and fast ion transport channels through secondary high-temperature carbonization treatment. As a result, the fabricated porous carbon anode delivers a high specific capacitance (199.2 F g−1 at 1 A g−1) and outstanding rate performance (136.3 F g−1 at 20 A g−1) in organic electrolyte. Furthermore, the assembled symmetrical supercapacitor achieves an energy density of 43.2 Wh kg−1 at 625.0 W kg−1, highlighting the potential of a secondary high-temperature carbonization strategy in practical applications.

Electric vehicles are powered by battery packs, which are usually composed of hundreds of units in series or in parallel. For power battery pack performance evaluation, many literatures have been published, including online performance assessment, life prediction and offline performance evaluation. Nevertheless, performance evaluation strategies that include both online and offline have not attracted sufficient attention. In this paper, we propose a performance evaluation method based on MCPE-DEKF, which can solve the problem of consistency analysis and sort of battery cells offline, as well as, implementing battery pack state estimation online. MCPE-DEKF is designed to enhanced the accuracy and adaptability for power battery pack SOC estimation. The pack SOC online estimation value from cells means model and the standard deviation of SOC estimation are combined with MCPE to determine their aggregation weights. An offline evaluation framework for cells grading and sorting problem is solved by MCPE methods. As the results show that, the RMSE of the online estimation for the battery pack is less than 2% and 7 mV for the SOC and the terminal voltage, respectively. The RMSE of the cell estimation is less than 0.3% and 6 mV, respectively. In terms of offline evaluation approaches for cells, we propose an approach with a fusion method to improve the reliability of sorting by integrating method 1 and method 4 (the second-best sorting method).

Abstract The ‘angiogenic switch’ is critical for tumor progression. However, the pathological details and molecular mechanisms remain incompletely characterized. In this study, we established mammal xenografts in zebrafish to visually investigate the first vessel growth (angiogenic switch) in real-time, by inoculating tumor cells into the perivitelline space of live optically transparent Transgenic (flk1:EGFP) zebrafish larvae. Using this model, we found that hypoxia and hypoxia-inducible factor (HIF) signaling were unnecessary for the angiogenic switch, whereas vascular endothelial growth factor A gene (Vegfa) played a crucial role. Mechanistically, transcriptome analysis showed that the angiogenic switch was characterized by inhibition of translation, but not hypoxia. Phosphorylation of eukaryotic translation initiation factor 2 alpha (Eif2α) and the expression of Vegfa were increased in the angiogenic switch microtumors, and 3D tumor spheroids, and puromycin-treated tumor cells. Vegfa overexpression promoted early onset of the angiogenic switch, whereas Vegfa knockout prevented the first tumor vessel from sprouting. Pretreatment of tumor cells with puromycin promoted the angiogenic switch in vivo similarly to Vegfa overexpression, whereas Vegfa knockdown suppressed the increase. This study provides direc and dynamic in vivo evidences that inhibition of translation, but not hypoxia or HIF signaling promotes the angiogenic switch in tumor by increasing Vegfa transcription.

ABSTRACT Deep-sea cold seeps are one of the most productive ecosystems that sustained by hydrocarbons carried by the fluid. Once the seep fluid ceases, the thriving autotrophic communities die out, terming as the extinct seep. But heterotrophic fauna can still survive even for thousands of years. The critical role of prokaryotes in active seeps are well defined, but their functions in extinct seeps are poorly understood to date. Here, we clarified the diversity, taxonomic specificity, interspecies correlation, and metabolic profiles of sediment prokaryotes at an extinct seep site of Haima cold seep, South China Sea. Alpha diversity of archaea significantly increased, while that of bacteria remained unchanged in extinct seep compared to active seep. However, archaea composition did not differ significantly at extinct seep from active or nonseep sites based on weighted-unifrac dissimilarity, while bacteria composition exhibited significant difference. Distribution of archaea and bacteria showed clear specificity to extinct seeps, indicating the unique life strategies here. Prokaryotes might live chemolithoautotrophically on cycling of inorganic carbon, sulfur, and nitrogen, or chemoorganotrophically on recycling of hydrocarbons. Notably, many of the extinct seep specific species and networked keystone lineages are classified as Proteobacteria. Regarding the functional diversity and metabolic flexibility of this clade, Proteobacteria is supposed to integrate the geochemical cycles and play a critical role in energy and resource supplement for microbiome in extinct seep. Collectively, our findings shed lights on the microbial ecology and functional diversity in extinct seeps, providing new understanding of biogeochemical cycling after fluid cessation. IMPORTANCE This research paper uncovered the potential mechanisms for microbiota mediated geochemical cycling in extinct cold seep, advancing our understanding in deep sea microbiology ecology.

Cyclists are vulnerable road users and often suffer head-neck injuries in car–cyclist accidents. Wearing a helmet is currently the most prevalent protection method against such injuries. Today, there is an ongoing debate about the ability of helmets to protect the cyclists’ head-neck from injury. In the current study, we numerically reconstructed five real-world car–cyclist impact accidents, incorporating previously developed finite element models of four cyclist helmets to evaluate their protective performances. We made comparative head-neck injury predictions for unhelmeted and helmeted cyclists. The results show that helmets could clearly lower the risk of severe (AIS 4+) brain injury and skull fracture, as assessed by the predicted head injury criterion (HIC), while a relatively limited decrease in AIS 4+ brain injury risk can be achieved in terms of the analysis of CSDM0.25. Assessment using the maximum principal strain (MPS0.98) and head impact power (HIP) criteria suggests that helmets could lower the risk of diffuse axonal injury and subdural hematoma of the cyclist. The helmet efficacy in neck protection depends on the impact scenario. Therefore, wearing a helmet does not seem to cause a significant neck injury risk level increase to the cyclist. Our work presents important insights into the helmet’s efficacy in protecting the head-neck of cyclists and motivates further optimization of protective equipment.

Sixteen groups of comprehensive tests were undertaken to examine the modifications in the mechanical characteristics of weak expansive soil resulting from the inclusion of a composite material comprising cement and jute fibers. The aim of the tests was to analyze the unconfined compressive strength and shear strength. The results reveal that (1) as the modified material content increased, the stress–strain curve of the modified soil progressively became steeper, with a more pronounced peak value. Under the same strain condition, the stress variation increased accordingly. After the stress reached its maximum, a sudden phenomenon of brittle axial compression failure occurred. (2) With the increase in cement content, the unconfined compressive strength and shear strength of the modified expansive soil gradually increased, reaching the highest shear strength when the cement content was 8%. With the increase in jute fiber content, the unconfined compressive strength and shear strength of modified expansive soil increased first and then decreased, reaching the highest value when the content was 0.5%. (3) The cohesion of modified expansive soil increased with the increase in cement content. When the cement content was 8%, the cohesion of the soil sample was as high as 298.29 kPa. The cohesion of modified expansive soil increased first and then decreased with the increase in fiber content. When the jute fiber content was 0.5%, the cohesion reached the maximum value. (4) The internal friction angle of the modified expansive soil increased with the increase in cement content. When the cement content was 8%, the internal friction angle of the soil sample increased by 2.64°. The cohesion of modified expansive soil increased first and then decreased with the increase in fiber content. When the jute fiber content was 0.5%, the internal friction angle reached the maximum value.

Background. The extensive burns devastate trauma. The research was designed to analyse the predictive value of early platelet (PLT) indices on the development of acute kidney injury (AKI) after severe burns. Methods and Results. 186 patients with extensive burns (burn area ≥30%) were eventually involved. Multivariate analyses pointed out that platelet distribution width (PDW) in the first 24 h after admission was an independent risk factor for AKI, severe AKI, and RRT requirement in patients with severe burns, and AKI risk showed an increase of 30.9% per increase of 1% in PDW (OR = 1.309, CI, 1.075–1.594, and P = 0.007). It was found that the area under the ROC curve (AUC) of PDW predicting AKI was 0.735 and that the AUC value was 0.81 for AKI after combining PDW and blood urea nitrogen (BUN). Based on the cut-off value PDW = 17.7%, patients were divided into high- (PDW ≥17.7%) and low-risk (PDW 17.7%, burn patients are not only at a higher risk for AKI but may also have higher AKI severity. Due to low cost and wide availability, PDW has the potential to be the tool that can predict AKI in extensive burn patients.

Soil moisture (SM) affects ecosystem homeostasis through interactions among land use type (LUT), precipitation, heat and human activity (HA). In the core areas of China Loess Plateau, we selected three sites along gradients of annual precipitation and annual accumulated temperature ≥ 0℃ and compare SM variations among three LUTs, including cropland, woodland, and rangeland, and in three precipitation years, including dry, wet, and normal year. The SM of three LUTs were sensitive to wet years in the north, dry years in the south, and both wet and dry years in the middle, respectively. From north to south, water use (WU) increased while water use efficiency (WUE) decreased. The WU for cropland was the highest and WUE was the lowest, while rangeland had the opposite tendency. The mean annual water deficiencies for cropland, rangeland and woodland were 42.41 mm, 21.38 mm, and 15.49 mm, respectively. HA for three LUTs increased as precipitation increased and the order was cropland > woodland > rangeland. The stability of SM to precipitation was as following order: rangeland > woodland > cropland. Rangeland was insensitive to precipitation, while cropland was sensitive to precipitation. Productivity of rangeland and cropland were stabilized mainly by increasing resistance to SM and the resilience to SM, respectively. To maximize the WUE of the rain fed system and enhance the stability of SM, the area ratios of cropland, woodland, and rangeland were recommended as 4:2:19, 3:2:5 and 3:2:4 in the northern, central, and southern regions, respectively.

Abstract The huge advantages of intelligent vehicles (IVs) in improving road safety and operating efficiency have become a research focus in the industry. IVs have made significant progress in recent years, but it is still face great challenges in order to be accepted by users on a large scale. In this regard, the authors propose that the research of IVs can be developed along the lines of safety, comfort and economy, gradually overcoming existing dilemmas. First of all, security is the most basic requirement of IVs. The authors sort out the key technologies and challenges of the basic architecture of IVs, and propose existing attack and defences strategies for IVs information security technology. Secondly, comfort is more about people's subjective feelings. From two aspects of physiological comfort and psychological comfort, the paper studies the anthropomorphic decision‐making to overcome the mechanized speed control, human computer interaction design, personalized driving style and ethical decision‐making methods. Then, aiming at the micro‐ and macro‐levels of economy, it outlines technologies such as economic driving behavioural, collaborative control of people, vehicles and roads and IVs sharing. Finally, the authors summarized the challenges and future development directions in the three stages of IVs development.

Abstract Transforming growth factor-β1 (TGF-β1) is essential for ovarian function and female fertility in mammals. Herein, we identified three completely linked variants, including two known variants referred to as c.1583A > G and c.1587A > G and the novel variant c.2074A > C in the porcine TGF-β1 3′-UTR. An important role of these variants in Yorkshire sow fertility was revealed. Variants c.1583A > G and c.1587A > G were located at the miRNA response element (MRE) of miR-2337 and affected miR-2337 regulation of TGF-β1 3′-UTR activity. Interestingly, miR-2337 induces, not reduces the transcription and production of TGF-β1 in granulosa cells (GCs). Mechanistically, miR-2337 enhances TGF-β1 promoter activity via the MRE motif in the core promoter region and alters histone modifications, including H3K4me2, H3K4me3, H3K9me2, and H3K9ac. In addition, miR-2337 controls TGF-β1-mediated activity of the TGF-β signaling pathway and GC apoptosis. Taken together, our findings identify miR-2337 as an endogenous small activating RNA (saRNA) of TGF-β1 in GCs, while miR-2337 is identified as a small activator of the TGF-β signaling pathway which is expected to be a new target for rescuing GC apoptosis and treating low fertility.

Marine oil spills are a significant concern worldwide, destroying the ecological environment and threatening the survival of marine life. Various oil-degrading bacteria have been widely reported in marine environments in response to marine oil pollution. However, little information is known about culturable oil-degrading bacteria in cold seep of the deep-sea environments, which are rich in hydrocarbons. This study enriched five oil-degrading consortia from sediments collected from the Haima cold seep areas of the South China Sea. Parvibaculum, Erythrobacter, Acinetobacter, Alcanivorax, Pseudomonas, Marinobacter, Halomonas, and Idiomarina were the dominant genera. Further results of bacterial growth and degradation ability tests indicated seven efficient alkane-degrading bacteria belonging to Acinetobacter, Alcanivorax, Kangiella, Limimaricola, Marinobacter, Flavobacterium, and Paracoccus, whose degradation rates were higher in crude oil (70.3–78.0%) than that in diesel oil (62.7–66.3%). From the view of carbon chain length, alkane degradation rates were medium chains > long chains > short chains. In addition, Kangiella aquimarina F7, Acinetobacter venetianus F1, Limimaricola variabilis F8, Marinobacter nauticus J5, Flavobacterium sediminis N3, and Paracoccus sediminilitoris N6 were first identified as oil-degrading bacteria from deep-sea environments. This study will provide insight into the bacterial community structures and oil-degrading bacterial diversity in the Haima cold seep areas, South China Sea, and offer bacterial resources to oil bioremediation applications.

ABSTRACT Improving the availability of representative isolates from the coral microbiome is essential for investigating symbiotic mechanisms and applying beneficial microorganisms to improve coral health. However, few studies have explored the diversity of bacteria which can be isolated from a single species. Here, we isolated a total of 395 bacterial strains affiliated with 49 families across nine classes from the coral Pocillopora damicornis. Identification results showed that most of the strains represent potential novel bacterial species or genera. We also sequenced and assembled the genomes of 118 of these isolates, and then the putative functions of these isolates were identified based on genetic signatures derived from the genomes and this information was combined with isolate-specific phenotypic data. Genomic information derived from the isolates identified putative functions including nitrification and denitrification, dimethylsulfoniopropionate transformation, and supply of fixed carbon, amino acids, and B vitamins which may support their eukaryotic partners. Furthermore, the isolates contained genes associated with chemotaxis, biofilm formation, quorum sensing, membrane transport, signal transduction, and eukaryote-like repeat-containing and cell-cell attachment proteins, all of which potentially help the bacterium establish association with the coral host. Our work expands on the existing culture collection of coral-associated bacteria and provides important information on the metabolic potential of these isolates which can be used to refine understanding of the role of bacteria in coral health and are now available to be applied to novel strategies aimed at improving coral resilience through microbiome manipulation. IMPORTANCE Microbes underpin the health of corals which are the building blocks of diverse and productive reef ecosystems. Studying the culturable fraction of coral-associated bacteria has received less attention in recent times than using culture-independent molecular methods. However, the genomic and phenotypic characterization of isolated strains allows assessment of their functional role in underpinning coral health and identification of beneficial microbes for microbiome manipulation. Here, we isolated 395 bacterial strains from tissues of Pocillopora damicornis with many representing potentially novel taxa and therefore providing a significant contribution to coral microbiology through greatly enlarging the existing cultured coral-associated bacterial bank. Through analysis of the genomes obtained in this study for the coral-associated bacteria and coral host, we elucidate putative metabolic linkages and symbiotic establishment. The results of this study will help to elucidate the role of specific isolates in coral health and provide beneficial microbes for efforts aimed at improving coral health.

BackgroundTreatment-resistant auditory verbal hallucinations (TRAVHs) might cause an increased risk of violence, suicide, and hospitalization in patients with schizophrenia (SCZ). Although neuroimaging studies have identified the neural correlation to the symptom of AVH, functional brain activity that correlates particularly in patients with TRAVH remains limited. Functional near-infrared spectroscopy (fNIRS) is a portable and suitable measurement, particularly in exploring brain activation during related tasks. Hence, our researchers aimed to explore the differences in the cerebral hemodynamic function in SCZ-TRAVH, patients with schizophrenia without AVH (SCZ-nAVH), and healthy controls (HCs), to examine neural abnormalities associated more specifically with TRAVH.MethodsA 52-channel functional near-infrared spectroscopy system was used to monitor hemodynamic changes in patients with SCZ-TRAVH (n = 38), patients with SCZ-nAVH (n = 35), and HC (n = 30) during a verbal fluency task (VFT). VFT performance, clinical history, and symptom severity were also noted. The original fNIRS data were analyzed using MATLAB to obtain the β values (the brain cortical activity response during the VFT task period); these were used to calculate Δβ (VFT β minus baseline β), which represents the degree of change in oxygenated hemoglobin caused by VFT task.ResultOur results showed that there were significant differences in Δβ values among the three groups at 26 channels (ch4, ch13-15, 18, 22, ch25–29, 32, ch35–39, ch43–51, F = 1.70 to 19.10, p < 0.043, FDR-corrected) distributed over the prefrontal–temporal cortical regions. The further pairwise comparisons showed that the Δβ values of 24 channels (ch13–15, 18, 22, 25, ch26–29, ch35–39, ch43–49, ch50–51) were significantly lower in the SCZ group (SCZ-TRAVH and/or SCZ-nAVH) than in the HC group (p < 0.026, FDR-corrected). Additionally, the abnormal activation in the ch22 of right postcentral gyrus was correlated, in turn, with severity of TRAVH.ConclusionOur findings indicate that specific regions of the prefrontal cortex may be associated with TRAVH, which may have implications for early intervention for psychosis.

Laminated shales are widely developed in the Dongying Sag and have attracted much attention as an oil reservoir. Macroscopically, these shales generally have multi-scale cyclicity, which is closely related to the development of laminae. Therefore, analyzing the origin of their cyclicity is helpful to understanding the formation mechanism of laminated shales and the vertical heterogeneity of shale reservoirs, which are of great significance for continental shale oil exploration and development. In this study, a gamma ray (GR) logging series, high-resolution elemental geochemical data, high-resolution core scanning photos and grayscale data, and mineralogical data were used to characterize the cyclicity of shale at different scales, and their relationship with different astronomical cycles was discussed. The results show that the Es3L and Es4U shale in the Dongying Sag has cyclicity from the meter-scale to the ten-meter scale and then to the hundred-meter scale, which is mainly manifested by periodic changes in organic matter abundance, mineral composition, element abundance, and grayscale. These cycles of different scales coincide with different astronomical periods. Specifically, the hundred-meter scale cyclicity is mainly controlled by the very long orbital period; the ten-meter scale cyclicity is mainly related to the eccentricity cycle; while the precession period is the main driver of the meter-scale cyclicity. Finally, we propose a simplified model for illustrating the formation of rhythmic organic-rich shale. This study is helpful to understanding the origin of continental organic-rich shale and predicting shale reservoir properties.

Increasing evidence shows that lncRNAs, an important kind of endogenous regulator, are involved in the regulation of follicular development and female fertility, but the mechanism remain largely unknown. In this study, we found that SDNOR, a recently identified antiapoptotic lncRNA, is a potential multifunctional regulator in porcine follicular granulosa cells (GCs) through RNA-seq and multi-dimension analyses. SDNOR-mediated regulatory networks were established and identified that SOX9, a transcription factor inhibited by SDNOR, mediates SDNOR’s regulation of the transcription of downstream targets. Functional analyses showed that loss of SDNOR significantly impairs GC morphology, inhibits cell proliferation and viability, reduces E2/P4 index, and suppresses the expression of crucial markers, including PCNA, Ki67, CDK2, CYP11A1, CYP19A1, and StAR. Additionally, after the detection of ROS, SOD, GSH-Px, and MDA, we found that SDNOR elevates the resistance of GCs to oxidative stress (OS) and also inhibits OS-induced apoptosis. Notably, GCs with high SDNOR levels are insensitive to oxidative stress, leading to lower apoptosis rates and higher environmental adaptability. In summary, our findings reveal the regulation of porcine GCs in response to oxidative stress from the perspective of lncRNA and demonstrate that SDNOR is an essential antioxidative lncRNA for maintaining the normal state and function of GCs.

The whole genome sequencing of tumor samples identifies thousands of somatic mutations. However, the function of these genes or mutations in regulating cancer progression remains unclear. We previously performed exome sequencing in patients with colorectal cancer, and identified one splicing mutation in C9orf9. The subsequent target sequencing of C9orf9 gene based on a validation cohort of 50 samples also found two function mutations, indicating that the loss of wild-type C9orf9 may participate in the tumorigenesis of colorectal cancer. In this research, we aimed to further confirm the function of C9orf9 in the CRC phenotype. Our Q-PCR analysis of the tumor and matched normal samples found that C9orf9 was downregulated in the CRC samples. Function assays revealed that C9orf9 exerts its tumor suppressor role mainly on cancer cell migration and invasion, and its loss was essential for certain tumor-microenvironment signals to induce EMT and metastasis in vivo. RNA-sequencing showed that stable-expressing C9orf9 can inhibit the expression of several metastasis-related genes and pathways, including vascular endothelial growth factor A (VEGFA), one of the essential endothelial cell mitogens which plays a critical role in normal physiological and tumor angiogenesis. Overall, our results showed that the loss of C9orf9 contributes to the malignant phenotype of CRC. C9orf9 may serve as a novel metastasis repressor for CRC.

In order to satisfy with the demand of high reliability in the aerospace field, a unified control scheme is proposed for five-phase open-winding permanent-magnet synchronous generator (FPOWPMSG) in this paper. Firstly, the mathematical model of FPOWPMSG in the phase coordinate system is established. Then, the proposed unified control scheme is presented in detail, which is mainly composed of the unified reference current generator, the current regulator, the synchronous electromotive force (EMF) compensator, and the piecewise nonlinear predictive compensator. In the unified reference current generator, normal operation, single-phase open-circuit, adjacent two-phase open-circuit, and non-adjacent two-phase open-circuit are taken into account by introducing a fault matrix. In the current regulator, the PI regulator is used. And the current tracking accuracy and the torque ripple are improved by both the synchronous EMF compensator and the piecewise nonlinear predictive compensator. Finally, the effectiveness of the proposed unified control scheme is validated by experimental results.

Reef-building corals are invertebrate animals that associate with diverse microorganisms, including Symbiodiniaceae, bacteria, fungi, and archaea. This symbiotic consortium, called the holobiont, is a dynamic system and rapidly responds to environmental temperatures. At present, the effects of temperature on bacteria-bacteria interactions in the coral-associated bacterial community are not clear. Antagonism is considered one of the potential structuring forces in coral microbial communities. Here, we examined the antagonistic interactions among 32 coral-associated bacteria and the physiological and biochemical characteristics of these isolates at different temperatures. The results showed that the antagonism breadth (i.e. the total number of antagonistic interactions) increased but antagonism intensity (i.e. the size of the inhibition zone) decreased at 32°C. The antagonistic interaction network was nested and sender-determined both at 25°C and 32°C, suggesting that the competition networks of coral-associated bacteria were more influenced by the antagonist strains than sensitive strains. Furthermore, we found that the elevated temperature increased the complexity of the antagonistic network. By evaluating the correlations between antagonism and the phylogenetic and phenotypic distances, we demonstrated that the antagonism probability correlated with the phylogenetic distance rather than phenotypic distance. Moreover, the antagonist strains have a wider metabolic niche space, i.e., grew on more carbon sources, than the antagonized strains at 25°C, while there was no difference at 32°C, suggesting the trade-off between antagonism and resource exploitation shifted in the antagonistic interactions under the higher temperature. These findings will be helpful for understanding the bacterial interactions in coral holobionts and the assembly of bacterial community in altered environments, especially under heat stress.

Diagnosis and treatment of the patients with major depression (MD) or the combined anxiety and depression (A&D) depend on the questionnaire, sometimes accompanied by tasks such as verbal fluency task (VFT). Functional near infrared spectroscopy (fNIRS) is emerging as an auxiliary diagnostic tool to evaluate brain function, providing an objective criterion to judge psychoses. At present, the conclusions derived from VFT or rest (non-task) studies are controversial. The purpose of this study is to evaluate if task performs better than non-task in separating healthy people from psychiatric patients. In this study, healthy controls (HCs) as well as the patients with MD or A&D were recruited (n = 10 for each group) to participate in the non-task and VFT tasks, respectively, and the brain oxygenation was longitudinally evaluated by using fNIRS. An approach of spectral analysis is used to analyze cerebral hemoglobin parameters (i.e., Oxy and Deoxy), characterizing the physiological fluctuations in the non-task and task states with magnitude spectrum and average power. Moreover, the standard deviation of oxygenation responses during the non-task was compared with the peak amplitude during the task, with the aim to explore the sensitivity of the VFT task to brain activation. The results show that there is no significant difference (p > 0.05) among the three groups in average power during non-task. The VFT task greatly enhanced the magnitude spectrum, leading to significant difference (p < 0.05) in average power between any of two groups (HC, MD, and A&D). Moreover, 40% patients with A&D have an intermediate peak (around 0.05 Hz) in the magnitude spectrum when performing the VFT task, indicating its advantage in characterizing A&D. We defined a rate of the non-task standard variation to the task peak amplitude (namely, SD-to-peak rate) and found that this rate is larger than 20% in 90% of the MD subjects. By contrast, only 40% HC subjects have an SD-to-peak rate larger than 20%. These results indicate that the non-task may not be sufficient to separate MD or A&D from HC. The VFT task could enhance the characteristics of the magnitude spectrum, but its intensity needs to be elevated so as to properly explore brain functions related to psychoses.

Systematically researching the association of the turbulentflow in cyclone with the particle revolution and high-speed self-rotation may provide new insights for improving the separation performance and expanding the application of cyclones. In this study, the effect of cyclone structural parameters and inlet gas velocity on the continuous phase flow field as well as the revolution and high-speed self-rotation of the dispersed phase particles. It was observed that the particle self-rotation speed increases with the increase of cylinder-to-cone ratio, the decrease of the cone angle, and the increase of the inlet gas velocity. With the cylinder-to-cone ratio, i=1, half-cone angle, α=4.7° and the inlet gas velocity of 20 m/s, the average particle self-rotation speed was determined to be 7124 rad/s. Meanwhile, it was also revealed that the cyclone structure and inlet gas velocity remarkably affected the velocity of the flow field, vorticity, pressure drop and residence time of revolution movement of particles within the cyclone. This study can provide theoretical fundamental for the industrial applications of cyclone in chemical engineering and environmental engineering related to the separation of pollutants on the surface of particles and in pores.

A novel pectic polysaccharide (HPP-1) with high immunomodulatory activity was extracted and isolated from the immature honey pomelo fruit (Citrus grandis). Characterization of its chemical structure indicated that HPP-1 had a molecular weight of 59,024 D. In addition, HPP-1 was primarily composed of rhamnose, arabinose, fucose, mannose, and galactose at a molar ratio of 1.00:11.12:2.26:0.56:6.40. Fourier-transform infrared spectroscopy, periodic acid oxidation, and Smith degradation results showed that HPP-1 had α- and β-glycosidic linkages and 1 → 2, 1 → 4, 1 → 6, and 1 → 3 glycosidic bonds. 13C NMR and 1H NMR analyses revealed that the main glycogroups included 1,4-D-GalA, 1,6-β-D-Gal, 1,6-β-D-Man, 1,3-α-L-Ara, and 1,2-α-L-Rha. Immunomodulatory bioactivity analysis using a macrophage RAW264.7 model in vitro revealed that NO, TNF-α, and IL-6 secretions were all considerably increased by HPP-1. Moreover, RT-PCR results showed that HPP-1-induced iNOS, TNF-α, and IL-6 expression was significantly increased in macrophages. HPP-1-mediated activation in macrophages was due to the stimulation of the NF-κB and MAPK signaling pathways based on western blot analyses. HPP-1 extracted from immature honey pomelo fruit has potential applications as an immunomodulatory supplement.

Many sets of Paleozoic marine organic-rich shale strata have developed in South China. However, the exploration and development results of these shale formations are quite different. Based on the data of core experiment analysis, drilling, fracturing test of typical wells, the reservoir differences and controlling factors of four sets of typical marine organic-rich shale in southern China are investigated. The four sets of shale have obvious differences in reservoir characteristics. Ordovician–Silurian shale mainly develops siliceous shale, mixed shale and argillaceous shale, with large pore diameter, high porosity, moderate thermal maturity, large pore volume and specific surface area. Cambrian shale mainly develops siliceous shale and mixed shale, with small pore diameter, low porosity, high thermal maturity and smaller pore volume and specific surface area than Ordovician–Silurian shale. Devonian–Carboniferous shale has similar mineral composition to Ordovician–Silurian shale, with small pore diameter, low porosity, moderate thermal maturity and similar pore volume and specific surface area to that of Cambrian shale. Permian shale has very complex mineral composition, with large pore diameter, low to medium thermal maturity and small specific surface area. Mineral composition, thermal maturity and tectonic preservation conditions are the main factors controlling shale reservoir development. Siliceous minerals in Cambrian shale and Ordovician–Silurian shale are mainly of biological origin, which make the support capacity better than Devonian–Carboniferous shale and Permian shale (siliceous minerals are mainly of terrigenous origin and biological origin). Thermal maturity of Ordovician–Silurian shale and Devonian–Carboniferous shale is moderate, with a large number of organic pores developed. Thermal maturity of Cambrian shale and Permian shale is respectively too high and too low, the development of organic pores is significantly weaker than the two sets of shale above. There are obvious differences in tectonic preservation conditions inside and outside the Sichuan Basin. Shale reservoirs inside the Sichuan Basin are characterized by overpressure due to stable tectonic activities, while shale reservoirs outside the Sichuan Basin are generally normal–pressure. Four sets of marine shale in South China all have certain resource potentials, but the exploration and development of shale gas is still constrained by complicated geological conditions, single economic shale formation, high exploration and development costs and other aspects. It is necessary for further research on shale gas accumulation theory, exploration and development technology and related policies to promote the development of China’s shale gas industry.

Deep learning is widely used in vision tasks, but feature extraction of IR small targets is difficult due to the inconspicuous contours and lack of color information. This paper proposes a new convolutional neural network–based (CNN-based) method for IR small target detection called DRUNet. The algorithm is divided into two parts: the feature extraction network and the prediction head. For the small IR targets, which are difficult to accurately label, Gaussian soft labels are added to supervise the training process and make the network converge faster. We use a simplified object keypoint similarity to evaluate the network accuracy by the ratio of the distance to the radius of the inner tangent circle of the target box and a fair method for evaluating the model inference speed after GPU preheating. The experimental results show that our proposed algorithm performs better when compared with commonly used algorithms in the field of small target detection. The model size is 10.5 M, and the test speed reaches 133 FPS under the RTX3090 experimental platform.

Anxiety and depression are widespread psychosis which are believed to affect cerebral metabolism, especially in frontal and temporal cortex. The comorbidity patients of anxiety and depression (A&D) have more serious clinical symptoms. Functional near-infrared spectroscopy (fNIRS) is a noninvasive modality used to monitor human brain oxygenation, and it could be considered as a potential tool to detect psychosis which may lead to abnormal cerebral oxygen status when the brain is activated. However, how sensitive the cerebral oxygenation response to the cortex activation and whether these responses are consistent at different stages of A&D or different regions still remains unclear. In this study, a conventional physiological paradigm for cortex activation, i.e., verbal fluency task (VFT), and a relatively new paradigm, i.e., high-level cognition task (HCT), were compared to detect A&D through a longitudinal measurement of cerebral oxygen status by fNIRS. The A&D patients at the acute, consolidation and maintenance stages as well as the healthy subjects participated in the VFT and HCT paradigms, respectively. For the VTF paradigm, the subject was instructed to answer questions of phrase constructions within 60 s. For the HCT paradigm, the subject was instructed to categorize items, logical reasoning, and comprehensive judgment and write down the answers within 60 s. For most of the subjects, the oxy-Hb is found to increase remarkably, accompanied with a relatively small reduction in deoxy-Hb when subject to both paradigms. The statistical analyses show a relatively large variability within any group, leading to the significant difference that was only found between A&D at the acute stage and healthy subjects in the temporal lobe region (p < 0.001). Nevertheless, HCT would activate more oxygen increment when compared with the VFT, with a large integral value in oxy-Hb. On average, the oxy-Hb integral value of the A&D patients differs substantially at different stages when subject to HCT paradigm. Moreover, the prefrontal lobe and temporal lobe responses were more consistent to the HCT paradigm rather than the VFT paradigm. Under the VFT paradigm, however, no remarkable difference in integral value was found among the three stages, either at the prefrontal lobe or at the temporal lobe. This study indicated that HCT, which is intensively involved in brain function, would activate more oxygenation changes in the cerebral cortex. Additionally, with good performance at distinguishing different stages according to the oxy-Hb criterion, the HCT has the potential to evaluate the therapeutic effects for A&D patients.

Single-ended resonant converters such as Class-E inverters have been widely considered as a potential topology for small- and medium-power wireless power transfer (WPT) applications, which feature compact circuits, low switching losses, and cost benefits, as they only use a low-side switch with a simple gate driver. However, there remains a practical challenge in the design of voltage stress, efficiency, and power density. In this paper, a single-ended resonant converter with a primary parallel resonant-matching network is investigated to absorb the bulky input-choke inductors of the Class-E inverters into the coil inductance. The analytical expressions for all the converter parameters are derived based on time-domain resonant waveforms, including: (1) analysis of critical zero-voltage switching (ZVS) conditions and (2) power transfer capabilities under the given maximum switch voltage stress. Furthermore, this paper elaborates on the design methodology of the proposed single-ended resonant converters, and an optimal operating point is chosen to ensure soft-switching operation and rated power. Finally, the accuracy of the proposed model is verified by simulation and experimental results.

Coupled inductors can effectively optimize the THD, loss, current ripple, and power density of multiphase interleaved totem-pole PFC converters. However, a coupled inductor will also worsen the zero-crossing distortion process. This paper first introduces the working principle of the interleaved totem-pole PFC converter with a coupled inductor based on a detailed analysis of modes, and then analyzes the influence on the zero-crossing process caused by the coupled inductor. To eliminate the zero-crossing distortion caused by the coupled inductor, an improved control strategy and its digital realization method of the totem-pole PFC converter is presented through the system model. Finally, the validity and correctness of the proposed circuit and control are verified in the 7.7 kW interleaved totem-pole PFC converter.
