One of the most common problem that researchers encounter when using fluorescence to visualize immunohistochemistry is the autofluorescence of the studied organ tissue sections and cell cultures. Autofluorescence quenching is necessary for a wide variety of organs and tissues, as well as for different methods of fixation and histochemical processing of sections. In addition to autofluorescence quenching, it is necessary to take into account the need for histological readability of tissue sections when using counterstains afterwards. Such protocol refinement for fluorescent immunohistochemistry for chicken, porcine and cattle tissues was carried out for the first time, as well as the use of a dimethyl sulfoxide (DMSO) solution with ethanol as Sudan Black B (SBB) solvent. Incubation of sections in SBB was chosen as the simplest and most nonspecific one. The most effective dissolution of the dye is achieved at a concentration of 0.3% SBB in a solution of 70% ethanol and absolutized DMSO in a 4:1 v/v ratio. The most thorough removal of SBB solution excess is achieved by rinsing the sections 5 times with 70% ethanol and then rinsing the sections with TBST (tris-buffered saline and Tween-20) buffer 5 times.

This article presents the results of the development of a mobile recommendation application to help library readers select references. One of the tasks of a librarian is to provide consultative assistance to the reader in selecting books. Working with a librarian can be replaced by the use of electronic library systems or special programs, which will allow you to solve the same task in less time. At the moment, there are websites and digital libraries that help the user to select books for further reading. They implement various algorithms that allow you to create personalized recommendations. Such systems are designed for electronic reading and are not linked to the databases of municipal library systems. Library readers can obtain recommendations when they choose literature for further reading using a mobile application. The paper considers the process of its creation: analysis of program requirements, design, implementation, testing and implementation. The application is developed for the Android operating system in the integrated development environment Android Studio in the Java programming language. The calculation of the technical and economic indicators of the project in its implementation has shown that the use of the application will provide sufficient economic effect. Its use can simplify the process of selecting books and significantly reduce the time to search for them, thus giving the librarian the opportunity to perform other tasks. The program does not require material costs for installation, which is an obvious advantage, and the functionality and system requirements allow its use on all mobile devices with the Android operating system.

Domestic tourism in our country is becoming an important tool for the development of regional economy. It covers all spheres of society, including culture, economy and social life. To stimulate domestic tourism by regions, it is necessary to update existing routes and create new ones. To develop new tourist routes, one of the effective approaches is the use of augmented reality technology. It allows to create interactive tourist routes that enrich the tourist experience of visitors to cities. The use of augmented reality technology allows to significantly diversify the leisure time of a tourist when visiting various cultural sites.

Clustering of k-means uses different variants of the algorithm of the same name to identify clusters. This paper deals with the performance study of the clustering algorithm using Euclidean distance metric and Mahalanobis metric. The choice of k-values as the initial estimate of the mean is considered in the second and fifth iterations. The BIRCH-3 and Mopsi-Finland datasets [1] are chosen as input data to investigate the performance of the metrics. The study shows the high efficiency of the k-means clustering algorithm using the Euclid metric depending on the random selection of the initial k values in the initial iterations of the algorithm. The use of Mahalanobis metric is more effective with an increasing number of iterations.

The study of the α diversity of the stand and the herb layer of all major forest types of primary pine and spruce forests in the Middle Urals was conducted. Four indices (Shannon's Diversity Index, Pielou's Evenness Index, Simpson's Diversity Index, Zhivotovsky's Diversity Index) were used to quantify α diversity. The indices were calculated based on the basal area (for the stand) and absolutely dry biomass (for the herb layer). It was shown that each forest type studied has its own characteristics of α diversity, while the relationship between the stand and the herb layer according to the dynamics of the calculated indices when changing habitats was not revealed.

The effect of light and root competition of the tree stand on the growth of the Pinus sibirica Du Tour undergrowth in post-fire birch forests of different ages is investigated using the example of mid-taiga forests of the Middle Urals. The studies were carried out on the basis of a microecosystem approach, using indices of light competition of the tree stand (Ilsc), root competition of the tree stand (Irsc) and integral competition of the tree stand (Irlsc). At the stage of the 13-year-old young birch stand with almost full illumination, the main factor determining the growth and development of Pinus sibirica undergrowth is root competition for soil nutrition. In the 32-year-old middle-aged birch stand, a joint, almost identical, effect of the factors of light and root competition on the growth of Pinus sibirica undergrowth is observed. In the mature 70-yearold birch stand, the main factor, influencing the growth of Pinus sibirica undergrowth, is the level of interception of photosynthetically active radiation by its canopy (light competition) with root competition is not expressed. In our opinion, this is due to the adaptive abilities and characteristics of the soil nutrition of the Pinus sibirica undergrowth and a change in the degree of soil saturation with organic compounds during the development of the post-fire birch forest.

The relevance of this work lies in the fact that for the country to be successful in sustainable development it is necessary to create quality documentation to ensure a balanced solution of social and economic problems, preservation of environmental potential and in general the state of the environment to further meet the needs of people. Today the issues of spatial planning are very topical, as over the past few decades a huge number of amendments and changes have been made to the land and urban planning legislation. At the moment, the processes of approval, development and approval of various spatial planning documents are undergoing changes in order to improve them. With regard to the territorial planning project, it is becoming necessary at the stage of drafting and obtaining a permit for further construction of any object. In cadastral activities, the above document is the basis for the state cadastral registration. The information provided in the territory planning project may not contradict the requirements of the documents of a higher level of influence. The main purpose of the territory planning project is to find the best options for the use of the territory of the settlement, which contributes to improving the living conditions of people living in such areas. The drafting of territory planning and land surveying may be carried out not only for land plots and construction projects, but also for linear objects.

Identifying the patterns of dynamics of natural populations of woody plants at the stage of their renewal is one of the main problems of forest ecology. The reforestation process is determined by both the dynamics of seed production and the specificity of seed dispersal of forest tree species. The five-needled pine renewal the Cembrae subsection with wingless seeds depends on birds of the genus Nucifraga sp., which store and spread their seeds. The aim of this study is to determine the relationship between seed production and regeneration of Pinus sibirica Du Tour using the thin-billed nutcracker (Nucifraga caryocatactes macrorhynchos Brehm C. L.) as a mediator. Long-term studies of these relationships have been carried out in the Siberian pine forests of the North Ural Mountains. As a result, a paradoxical conclusion is made that the number of Nucifraga birds during the period of storing and spreading of seeds in the previous year is a key factor in the abundance of Pinus sibirica seedlings. In turn, the number of seeds, both from the previous year and the current year, affects the number of Nucifraga birds.

The aim of the research is to reveal the relationship between the age and composition of the forest stand and the regeneration of Picea obovata Ledeb. and Abies sibirica Ledeb. Null hypothesis: we can estimate young generations of P. obovata and A. sibirica under the forest canopy based on the composition and age of the forest stand. The studies conducted in the most widespread forest type of the South Ural Mountains confirmed our null hypothesis about the possibility of modelling the regeneration of P. obovata and A. sibirica by the structure of the stand. The number of young generations of P. obovata and A. sibirica showed high sensitivity to the age and composition of the stand. The graphical models developed by us can be used to combine with forest management data in order to obtain large-scale data on the regeneration of woody plants in the primary spruce small-grass-green mossy forests and secondary birch and aspen forests of the South Ural Mountains. In order to expand the scope of application of the developed models, it is planned to conduct additional studies for other forest types of the Urals.

The largest part of the population of the Russian Federation (RF) is concentrated in settlements. The lands of settlements play an important role in the life of society, fulfilling the function of territorial and spatial basis. Occupying a relatively small area compared to the entire territory, these lands are the place of residence of the majority of the population. The lands occupied by settlements are intended for the location of cities, towns, rural settlements, and real estate objects that ensure the vital activity of the population. Land legislation singles out these lands as a separate category of lands. The spatial and land resources basis of urban and rural settlements consists of territories within the established borders (boundaries) of urban and rural settlements, as well as everything that is above and below the surface of these territories (Article 38 of the Town Planning Code). It should be noted that the inclusion of land plots within the boundaries of settlements does not entail the termination of the right of ownership, land tenure, land use and lease rights to these plots. However, in this case land tax rates are changed. Inclusion of land plots in the boundaries of settlements does not entail termination of the rights of owners of land plots, land users, landowners, and lessees of land plots. In modern conditions, the development of territories of settlements is an important area contributing to sustainable economic development in Russia.

Together with the development of professional disciplines in higher education, one of the most important trends is the introduction of research into the teaching and learning process. It allows students to develop their ability to work more deeply and comprehensively with scientific and methodological aids as well as with the legal and regulatory framework being the most essential and relevant to land surveying and cadastral relations. The application of different methodologies to scientific cognition helps to enhance students' research skills and prepare them for possible use in production processes in the future. An artificial land plot being the subject of this study shall be recognized as a land plot after putting into operation, but, based on its definition, continue to have the status of a structure. This fact predetermines its dualistic nature. As a consequence, effective regulation of the legal regime of artificial land plots is impossible without the cumulative impact of legislative norms reflecting the specifics of the relevant sphere of social relations. It is from this that the relevance and timeliness of the study follows.

Background and purpose: One of the most important mechanisms of tissue regeneration is the high functional activity of cells, including proliferation. Currently, there are practically no effective skin cell activators on the pharmaceutical market. The purpose of this work was to demonstrate the stimulating effect of spiroconjugated 1,2,3-triazolo[5,1-b]1,3,4-thiadiazine (STT) on the functional activity of fibroblasts. Experimental approach: STT containing ointment for dermal application was made. To assess in vivo effect of the STT a linear wound model in rats was tested. A combination of histological techniques and mechanical testing was employed to estimate the stimulating effect of STT on the functional activity of fibroblasts. Findings/Results: The STT significantly increased the number of fibroblasts as well as the density and order of produced collagen fibers in the dermis during the wound healing process. As a result, a tissue was formed at the site of damage with the structure corresponding to normal skin. In addition, skin functions were restored, in particular mechanically. Conclusion and implications: The results suggested the stimulating effect of the STT on fibroblast activity and demonstrated its potential for skin regeneration. [ABSTRACT FROM AUTHOR]

A propagation of ultrawideband electromagnetic pulses (UWB EMPs) through magnetized plasma has been experimentally studied using a "gigantic" coaxial line, which has been developed at IAP RAS for laboratory modeling of ionospheric effects. This coaxial line is 1.4 m in diameter and 10 m in length and is installed inside the chamber of the large-scale Krot plasma device. The line can be filled with rf inductively coupled plasma, magnetized or not. It allows one to explore the propagation of UWB EMPs in plasma along a long path without refraction and divergence and obtain a physical picture of EMP transformation. Under conditions where the duration of the UWB EMP is comparable to the period of electron plasma oscillations (fp−1), the period of cyclotron rotation of electrons (fc−1), or even significantly shorter, a complex of effects of transformation of the waveform and frequency spectrum of the pulse occurs. Without ambient magnetic field, a UWB EMP is distorted due to the effects of the cutoff and frequency dispersion. In dense magnetized plasma, i.e., when fp >> fc, the UWB EMP breaks into two wave packets, the high-frequency one (f > fp) and low-frequency one (f < fc). In rare magnetized plasma (fp << fc), the cyclotron absorption produces a long train of damped oscillations at a frequency close to the cyclotron frequency fc following the UWB EMP. The processes of natural reforestation play a crucial role for the preservation of primary coniferous forests and the success of their restoration after catastrophic impacts. The objective of our research was to identify the features of the natural renewal of the main coniferous forest-forming plants under the canopy of stands in the mountains of the Middle Urals, to establish dependencies on the forest type not only the total number of tree saplings, but also its age structure. The research was carried out in the mountainous part of the Sverdlovsk region (Russia) on the eastern macroslope of the Ural Mountains. We studied old-age pine forests, which belong to the 4 most common forest types. The tree saplings were counted into account on 2-4 subplots with a length of 20 meters and a width of 2 meters. It is established that under the canopy of all types of forest there is a different age coniferous saplings, despite significant differences in habitats. The most abundant pine saplings are found in pine forests with Vaccinium vitis-idaea, and the most abundant spruce saplings is found in pine forest with spruce and moss cover.

The adaptive capacity of ecosystems is a key to maintaining ecosystem functions and preserving biodiversity at all levels. The research was carried out in the western Southern Ural low-hill terrains (Russia). We analyzed distributions of above-ground biomass of herb layer species in the primary spruce, long-term secondary birch 20-35-year forests, stable-term secondary 20-year aspen forests. Species abundance distributions (SAD) analysis allowed us to study the species diversity and the species structure of communities, to understand the mechanism of adaptation of the plant community to disturbance and the role of species biodiversity in this process. It became clear that only with sufficient species diversity can SAD be maintained stable, and the adaptive potential and resistance of plant communities to external influences can be preserved. In General, the SAD analysis can be considered useful and informative for studying the adaptation of plant communities. In addition, it is universal and can be applied to any terrestrial and aquatic ecosystems. SAD can also help in designing artificial plant communities with increased resilience and high ecosystem functions.

The use of commercial land, land parcels or land in general refers to an objectively complex process being one of the main components of land relations revealing their content. The principle of rational land use acquires its objective character on a basis of clear interaction between technical, technological, natural-biological, socio-economic and environmental components that ensure effectiveness of law of continuous recovery of soil fertility and productivity of agricultural lands. Land use is a process providing implementation of these properties which manifests itself as growth of production.

This article discusses multiple aspects of militarization of life of the Soviet people in the 1920-1930s. The authors explore the reasons for this phenomenon, which lay not only in the foreign environment, and in the domestic politics, but also in the ideological foundations of the totalitarian state and society. Special attention is paid to the role of the Komsomol in the militarization of youth education, participation in the economic, political and social life, attempts of militarization of everyday life and private life in general. The article also reveals the role of the universal military duty and military service in preparation for a young person’s life, the participation of the army in dealing with economic and political problems. Attention is drawn to the militarization of culture and ideological indoctrination of the youth. The article investigates the tragic fate of the inter-war generation. This generation was at the same time the basis for the deployment of a wide social experiment to educate the new man at the heart of aspirations and expectations which would lay the ideas of socialism as a “bright future” of all mankind. These experiments, which began in the years of civil war, turned in many areas of culture: architecture, art, literature. They represent a strange mixture of modernism and Marxism. All these experiments were subordinated to the main goal – the education of the “new man”. Since the inevitability of military confrontation with the “capitalist encirclement”, it seems clear that the militarization of the country’s life is comprehensive.

Prime editing is a rapidly developing method of CRISPR/Cas-based genome editing. The increasing number of novel PE applications and improved versions demands constant analysis and evaluation. The present review covers the mechanism of prime editing, the optimization of the method and the possible next step in the evolution of CRISPR/Cas9-associated genome editing. The basic components of a prime editing system are a prime editor fusion protein, consisting of nickase and reverse transcriptase, and prime editing guide RNA, consisting of a protospacer, scaffold, primer binding site and reverse transcription template. Some prime editing systems include other parts, such as additional RNA molecules. All of these components were optimized to achieve better efficiency for different target organisms and/or compactization for viral delivery. Insights into prime editing mechanisms allowed us to increase the efficiency by recruiting mismatch repair inhibitors. However, the next step in prime editing evolution requires the incorporation of new mechanisms. Prime editors combined with integrases allow us to combine the precision of prime editing with the target insertion of large, several-kilobase-long DNA fragments. [ABSTRACT FROM AUTHOR]

One of the goals of higher professional education is to develop generic student competencies across a variety of disciplines that play a crucial role in education and that provide wider opportunities for graduates in finding good jobs and more chance of promotion. In this article a list of generic competencies developed in Russian universities is compared with a similar list developed by a consortium of Russian and European universities (project TUNING-RUSSIA). Then there is a second comparison with a list of competencies taken from the CDIO Syllabus. This comparison indicates the degree of similarity among the lists and the possible convergence among universities all over the world. The results are taken from a survey carried out among Russian employers, academics, and graduates. The survey asked to rate each listed competence by its importance and the degree of achieving goals in the process of the education.

In this contribution, a sufficiently simple, yet realistic model of TB propagation and control is studied in detail. We compute the controlled basic reproduction number R0(uB, uD) for the extended model including the action of two types of controls and analyze the sensitivity of R0(uB, uD) with respect to both controls following the recently devised approach. It is shown that the computed sensitivity coefficients admit a clear epidemiological interpretation and can be used in assessing the efficacy of the respective types of intervention.

The key role in the energy supply efficiency of smart cities is played by the electric power industry. The article provides an overview of the challenges of the electricity market in Europe and Russia and its main trends and directions. Changes in the organizational structure of the market, the role and tasks of each body managing the selected area of activity in the energy market are determined. Particular attention is paid to new opportunities provided to end-users of electricity in the smart cities. The article deals with the use of renewable sources and energy storages in accordance with new strategic programs in the EU and Russia. The research results and forecasts for the development of the electricity market and the use of different primary energy sources are shown. The main areas of development of the electricity market in the European Union and Russia are identified.The conclusion is drawn on the need to develop information systems that meet market changes and the business functions of such systems are determined.

The article studies geological and engineering conditions of the mineral deposits located in cryolithic zone. The demand to study and analyze geological and engineering conditions is justified by safety requirements established to development of the deposits. The article is focused on pebble-phosphate deposit located in Polar Urals. The geological and engineering conditions of this site fall within very severe category, which is connected with endogenetic and exogenic processes of its accumulation. It is the genesis of the deposit that generated the factors that complicate drilling-in conditions, included but not limited to weathering crusts with significant depths and discontinuity, frozen rock mass and its cavernous porosity.

The many recent crises in Belarus are often seen through the prism of democratization, post-communist transition, and nation- and identity-building. As a rule, it is put into the context of the 1989 democratization in Central and Eastern Europe and compared with similar societal mobilization in Georgia (2003), Ukraine (2004; 2014), and Kyrgyzstan (2005). This article, however, argues that while these theoretical approaches provide an important explanatory potential, they nevertheless fail to account for informal, hidden, and unstable processes presently unfolding in the Belarusian society, leading to profound change. We argue that, in the vulnerable, unpredictable, complex, and ambiguous (VUCA) world of today, our knowledge and ability to plan and achieve desirable outcomes are limited in contrast to a largely positivist or interpretivist epistemology of the mainstream theories, which conceive of the world as a closed system. In this article, we offer an alternative explanation of the many crises in Belarus by drawing on the insights of complexity-thinking to suggest that (hidden) transformative change in the country is now irreversible. The concept of a smart city is aimed at developing technologies and infrastructure based on the use of new technologies, large data centers, smart sensors and automated power grids. As technological advances in control systems increase the efficiency of sensors, users are calling for advances in sensor accuracy, reliability, dynamic characteristics (response time, dynamic linearity etc.), robustness, miniaturization and communication capability. The sensor industry is expected to experience sound growth during the five-year forecast period. It is possible satisfy the growing demand, in particular, by using automated systems of conceptual design of sensors, which can reduce the design time several times. Physical principles of operation of the sensors involved in information measuring and control systems of buildings are quite diverse. In this article, it is proposed to use a modified energy-information method for the conceptual design of sensors, invariant to the physical principle of operation. The possibility of using the operator method for the calculation and study of the dynamic characteristics of the designed elements of control systems of different physical nature is shown. The architecture of the expert system of evaluation of dynamic characteristics of sensitive elements of information-measuring devices of smart home is developed.

The purpose of this study is to develop a model of neuro-fuzzy regulation of the microclimate in the room. The proposed model consists of an artificial neural network serving to form a comfort index PMV, a fuzzy logic controller for regulating temperature and humidity in the room. This approach makes it easy to manage these parameters through an estimate of the PMV index, which indicates the level of thermal comfort in the room.

Fungi of genus Botrytis are important pathogens of legumes, causing gray mold and chocolate spot diseases. The use of molecular methods to identify pathogens has resulted in the discovery of several new Botrytis species and new associations of pathogens with diseases. Thus, chocolate spot of faba bean is now associated with at least four species: B. fabae, B. cinerea, B. pseudocinerea and B. fabiopsis. Species of Botrytis differ in host plant, pathogenicity, fungicide resistance and other relevant properties that affect disease control. The aim of this study was to identify the species of Botrytis isolated from different legume crops and to evaluate their in vitro pathogenicity. Between 2014 and 2019, 278 isolates of Botrytis were obtained from infected legumes in Latvia. A phylogenetic analysis was carried out by sequencing three nuclear genes, RPB2, HSP60, and G3PDH, considered to be diagnostic for species in this genus. A set of 21 representative isolates was selected for pathogenicity tests on detached leaves of faba bean, field pea, lupin and soybean using 5-mm mycelium-agar plugs. The diameter of the formed lesions under the inoculated plug was measured crosswise each day. The datasets were subjected to analysis of variance with the split-plot design of the experiment and repeated-measures model. Six species were identified: B. cinerea, B. fabae, B. pseudocinerea, B. fabiopsis, B. euroamericana and B. medusae. In addition to the expected combinations of host and pathogen, naturally occurring infections of B. fabiopsis were found on chickpea, B. euroamericana on faba bean and B. medusae in lupin seeds. Species and isolate had significant effects on pathogenicity on all crops tested. Several isolates were pathogenic on two or more host species: two of B. pseudocinerea, two of B. cinerea, two of B. fabiopsis and the one of B. medusae. One isolate of B. pseudocinerea and two of B. fabiopsis caused primary lesions on all five host species. The results show that these Botrytis species have a broad host range that should be borne in mind when planning crop sequences and rotations. [ABSTRACT FROM AUTHOR]

High-throughput protein assays are crucial for modern diagnostics, drug discovery, proteomics, and other fields of biology and medicine. It allows simultaneous detection of hundreds of analytes and miniaturization of both fabrication and analytical procedures. Photonic crystal surface mode (PC SM) imaging is an effective alternative to surface plasmon resonance (SPR) imaging used in conventional gold-coated, label-free biosensors. PC SM imaging is advantageous as a quick, label-free, and reproducible technique for multiplexed analysis of biomolecular interactions. PC SM sensors are characterized by a longer signal propagation at the cost of a lower spatial resolution, which makes them more sensitive than classical SPR imaging sensors. We describe an approach for designing label-free protein biosensing assays employing PC SM imaging in the microfluidic mode. Label-free, real-time detection of PC SM imaging biosensors using two-dimensional imaging of binding events has been designed to study arrays of model proteins (antibodies, immunoglobulin G-binding proteins, serum proteins, and DNA repair proteins) at 96 points prepared by automated spotting. The data prove feasibility of simultaneous PC SM imaging of multiple protein interactions. The results pave the way to further develop PC SM imaging as an advanced label-free microfluidic assay for the multiplexed detection of protein interactions. [ABSTRACT FROM AUTHOR]

The article presents the results of the study of fifty populations of common heather (Calluna vulgaris (L.) Hull) collected throughout its range. A phased comparative analysis (genetic, biochemical, anatomical, morphological, and ecological) was carried out with the estimation of indicators that included two key populations—Zavodouspenskoe (Pritobolye, Western Siberia) and Luga (Baltic, Eastern Europe). It was concluded that heather growing in Western Siberia should be identified as a separate taxonomic group, giving it the status of a subspecies. The gene pool of Pritobolye populations (including Zavodouspenskoe) is represented by the completely dominant (100%) monohaplotype S, which is not found anywhere else. The heather plant growing in Zavodouspenskoe has a longer lifespan. It is distinguished by larger linear leaf dimensions (length 2.06 ± 0.09 mm), thicker cuticle (4.77 ± 0.33 μm), increased number of trichomes (18.98 ± 0.56), and a reduced number of stomata (13.60 ± 0.63) than that growing in Luga. The new subspecies differs in biochemical composition: twice less content of epicatechin (average 1.992 ± 0.005 mg g−1), three times more myricetin (average 2.975 ± 0.005 mg g−1), twice as much chlorogenic acid (average 2.763 ± 0.004 mg g−1). An ecological feature is that C. vulgaris does not grow in the swamps of Western Siberia and has a small population. This species has a high horticultural potential and requires protection as its population in Western Siberia continues to decline rapidly. [ABSTRACT FROM AUTHOR]

One of the most critical problems throughout the world is air pollution, causing the death of millions of individuals annually, and it is reported that 90% of the global population breathes polluted air. Among the components of air pollution, the most harmful ones are the heavy metals, which can remain non-degraded in nature for a long time, bio-accumulate in living organisms, and be toxic or carcinogenic at low concentrations. Hence, monitoring and reducing heavy metal pollution in the air are high-priority research topics. Heavy metals can accumulate within various organs of plants grown in an environment with an increased level of heavy metal pollution. The metal analyses on these organs can provide insight into the heavy metal pollution in the air. In the present study, the concentrations of aluminum (Al), one of the most important heavy metals, were determined in the different organs of five plant species grown in regions with different traffic densities. Remarkable changes were observed in the Al concentrations in all the organs of species, which were examined here by organ and traffic density. Glycogen storage disease type 1 (GSD1) is a rare hereditary monogenic disease characterized by the disturbed glucose metabolism. The most widespread variant of GSD1 is GSD1a, which is a deficiency of glucose-6-phosphatase-ɑ. Glucose-6-phosphatase-ɑ is expressed only in liver, kidney, and intestine, and these organs are primarily affected by its deficiency, and long-term complications of GSD1a include hepatic tumors and chronic liver disease. This article is a brief overview of existing animal models for GSD1a, from the first mouse model of 1996 to modern CRISPR/Cas9-generated ones. First whole-body murine models demonstrated exact metabolic symptoms of GSD1a, but the animals did not survive weaning. The protocol for glucose treatment allowed prolonged survival of affected animals, but long-term complications, such as hepatic tumorigenesis, could not be investigated. Next, organ-specific knockout models were developed, and most of the metabolic research was performed on liver glucose-6-phosphate-deficient mice. Naturally occuring mutation was also discovered in dogs. All these models are widely used to study GSD1a from metabolic and physiological standpoints and to develop possible treatments involving gene therapy. Research performed using these models helped elucidate the role of glycogen and lipid accumulation, hypoxia, mitochondrial dysfunction, and autophagy impairment in long-term complications of GSD1a, including hepatic tumorigenesis. Recently, gene replacement therapy and genome editing were tested on described models, and some of the developed approaches have reached clinical trials. [ABSTRACT FROM AUTHOR]

This introduction to the Special Issue problematises the necessity to rethink governance through the lens of resilience and suggests a novel conceptualisation of resilience. Building the argument on complexity-thinking, this issue contends that in the context of change and complex life, challenges are most efficiently dealt with, at the source, 'locally', to make 'the global' more sustainable. Accordingly, the concept of resilience as self-governance is advanced in the introduction as an overriding framework to explore its constitutive elements—identity, 'good life', local coping strategies and support infrastructures—which, when mobilised, can turn community into 'peoplehood' in the face of adversity. This conceptualisation, we argue, explains what makes communities adapt and transform, and how they should be governed today. Central Eurasia, spanning from Belarus in the west, to Azerbaijan in the south and Tajikistan in the east, provides fertile grounds for exploring how resilience works in practice in times of complex change. By immersing into centuries-long traditions and philosophy, local experiences of survival, and visions for change, this introduction—along with the Special Issue—shows that governability at any level requires a substantive 'local' input to make 'the global' more enduring and resilient in a complex adaptive world.
