The high backpressure heating mode of direct air-cooling unit faces limitations due to the return water temperature of heat network and the coordinated heat exchanger unit control of air-cooling island. This limitation complicates the task of achieving a balance between operation economy and power generation output. To solve this inherent contradiction， Ebsilon Professional， a commercial software， is used to calculate and analyze the key parameters of the direct air-cooling exhaust steam extraction heat supply system. These parameters include the backpressure of the air-cooling island， steam turbine output and heat consumption， and suction coefficient of the injector under different loads and variable operating conditions. The calculations reveal that， during low load in winter， the optimal operating backpressure for the heat supply system falls within the range of 13~20 kPa， which allows for the gradient utilization of heating energy， ensures a balance in unit output， and meets the safety requirements for preventing freezing.

Background: The tumor immune microenvironment (TIME) is an important regulator of tumor progression, growth and metastasis. In addition, tumor metastasis is one of the principal obstacles to the treatment of colorectal cancer (CRC). Circular RNAs (circRNAs) have been recognized as important regulators in the development of malignancies. However, their specific roles and mechanisms in both CRC metastasis and TIME have not been thoroughly investigated. Methods: High-throughput next-generation sequencing technology and real-time fluorescence quantitative PCR technology were performed to identify differential circRNAs in CRC. Functional assays including transwell assay, wound healing assay, and metastasis models were conducted to assess the effect of circRNF216 on CRC metastasis. In addition, luciferase reporter, western blot, RNA immunoprecipitation (RIP), and fluorescent in situ hybridization (FISH) were performed to explore the underlying mechanism of circRNF216. The level of immune infiltration was assessed by bioinformatics analysis and flow cytometry in CRC model. Furthermore, rescue and mutation experiments were used for verification. Results: circRNF216 was identified as a putative tumor suppressor that is downregulated in CRC tissues and cells. Overexpression of circRNF216 inhibits metastasis in vitro and vivo. Mechanistically, circRNF216 acts as a competitive endogenous RNA (ceRNA) for miR-576-5p, alleviating miR-576-5p repression on its target ZC3H12C, which in turn downregulated N-cadherin. Additionally, circRNF216 could enhance the infiltration level of CD8+ T cells by upregulating ZC3H12C, ultimately inhibiting the development of CRC, which suggests that circRNF216 is a potential biomarker for the treatment of CRC. Conclusions: Here, we provide novel mechanistic insight revealing how circRNF216 functioned in CRC metastasis and TIME via the circRNF216/miR-576-5p/ZC3H12C pathway. A novel fluorescent sensor (LJ-1) as a Zn2+ and nitro oxide (NO) relay recognition probe was designed and synthesized based on the aminomethylnaphthol moiety. In PBS buffer, LJ-1 showed high selectivity and ratiometric fluorescence response to Zn2+ on the basis of the intermolecular charge transfer (ICT) mechanism. Spectroscopic titrations showed that LJ-1 could be recognized as a fluorescent turn-on probe with a 10−8 M detection level. Subsequently, the resultant LJ–Zn2+ complex could act as a turn-off probe for the detection of NO in several seconds at the 10−8 M detection level. Furthermore, the binding modes of Zn2+ and NO with LJ-1 were elucidated by a computational study using density functional theory (DFT). The results demonstrated that LJ-1 can serve as a ratiometric fluorescent probe for relay recognition of Zn2+ and NO in an aqueous medium at physiological pH value. [ABSTRACT FROM AUTHOR]

Nanomagnetic devices, such as nano-field effect transistor biosensors and radio frequency magnetic induction therapies, came into being with the development of medical nanomaterials. The application of nanomagnetic materials in the treatment of cancers is rapidly becoming increasingly important because of its ability to target therapy and diagnose early. In this review, an untechnical overview of the fundamental of magnetism in nanomaterials and an illustration of how these materials are applied are presented. The applications of nano-field effect transistor biosensors for the detection of tumor biomarker nanomaterials in the therapy and diagnosis of cancers and nanomagnetic materials are summarized in this paper. Periodontal disease is a complex problem which often interrelates with several serious systemic diseases. However, the satisfactory clinical therapy has yet to be achieved. Herein, serum albumin microspheres containing minocycline and zinc oxide nanoparticals (ZnO NPs) were prepared and incorporated in a Carbopol 940® hydrogel. Compared with 2% minocycline ointment (Perio®), the hydrogel has shown obvious therapy effects and the ability of gingival tissue self-repairing. The serum albumin microspheres containing 0.06% of minocycline and 0.025% of ZnO NPs presented an average size of 139 ± 0.42 nm using electrophoretic light scattering (n = 3). Photomicrographs obtained by TEM showed homogeneous and spherical-shaped particles. The encapsulation efficiency was 99.99% for minocycline and the slow-release time was more than 72 h with pH-sensitive property. The in vitro skin adhesion experiment showed that the largest bioadhesive force is 0.35 N. Moreover, the hydrogel showed broad-spectrum antimicrobial and effective antibacterial ability when concentration of the ZnO NPs was over 0.2 µg/mL. The cell survival rates were more than 85% below 0.8 mg/L of ZnO NPs, which proved its low toxicity and high security. [ABSTRACT FROM AUTHOR]

Several studies have reported a significant role of high mobility group box protein 1 (HMGB1) in lung cancer. Nevertheless, there is a lack of knowledge regarding the expression of HMGB1 and its correlation with the clinicopathological features of lung cancer. In addition, the potential molecular mechanisms underlying the role of HMGB1 in lung cancer are still unknown. We therefore investigated the clinicopathological and prognostic significance as well as the potential role of HMGB1 in the development and progression of lung cancer. HMGB1 expression in the tumor tissues of the cohort correlated with clinicopathological features. Moreover, lung cell migration and invasion were significantly increased after treatment with HMGB1. The matrix metalloproteinase-2 (MMP-2) expression and activity were upregulated after treatment with HMGB1, while the upregulated expression of MMP-2 stimulated by HMGB1 in lung cancer cells was significantly reduced with the blockage of si-p65. These results indicated that HMGB1 expression was significantly associated with lung cancer progression. We also showed that HMGB1 promoted lung cancer invasion and metastasis by upregulating the expression and activity of MMP-2 in an NF-κB-dependent manner. Ras-related protein (Rap)2a and Rap2b are members of the GTP-binding protein family, and serve an important function in tumor progression. However, the associations between Rap2c and cancer cell functions have not yet been reported. Osteosarcoma is a type of bone cancer; its high degree of invasion is considered to be a major treatment challenge. The present study first investigated the biological role of Rap2c in human osteosarcoma cells and investigated the underlying mechanism of Rap2c on osteosarcoma cell migration and invasion. The results of the present study demonstrated that Rap2c overexpression promoted the migratory and invasive ability of cancer cells, and increased the activity of matrix metalloproteinase-2 (MMP2). Correspondingly, the knockdown of Rap2c inhibited tumor cell migration and invasion, whereas alterations to Rap2c had no effect on osteosarcoma cell proliferation or rate of apoptosis. Furthermore, Rap2c overexpression may decrease the protein level of tissue inhibitor of metalloproteinases 2 and increase the phosphorylation level of protein kinase B (Akt). Collectively, these results indicated that Rap2c has a key function in tumor migration and invasion, and the Akt signaling pathway may be involved in Rap2c-induced MMP2 expression. Lung cancer has very high mortality and glycyrrhizin was found to significantly inhibit the growth of lung cancer cells in vitro and tissues in mice. However, the detailed inhibitory role of glycyrrhizin in the growth of lung cancer is still unclear. In this study, we first found that glycyrrhizin inhibited the growth of lung tumor in PDX mice. And high level of HMGB1 promoted the migration and invasion of lung cancer cells, which was suppressed by glycyrrhizin. Moreover, glycyrrhizin reduced the activity of JAK/STAT signaling pathway, which is the upstream regulator of HMGB1. Replication-competent adenovirus ( RCAd) has been used extensively in cancer gene therapy, and tumor-selection is critical for the use of replication-competent adenovirus. Here we investigated the anti-tumor characterization of oncolytic virus, whose E1 A gene is under the control of a renal cell carcinoma specific promoter - the G250 promoter. The constructed oncolytic virus G250- Ki67 is armed with transgene of Ki67-si RNA, and G250- ZD55- Ki67 also with E1 B-55 KD deleted. The tumor-specific expression of E1 A and Ki67 was demonstrated by Western blot and immunohistochemistry staining, and the tumor-specific cytotoxicity was assessed by crystal violet staining and cell viability assays. The G250- Ki67 and G250- ZD55- Ki67 adenoviruses could express E1 A protein in 786- O and OSRC cell lines but not in ACHN and HK-2 cell lines. The expression of Ki67 gene in 786- O and OSRC cell lines were suppressed by these adenoviruses. The cytotoxic effects induced by G250- ZD55- Ki67 and G250- Ki67 were more obvious on the 786- O cell lines than on the OSRC cell lines. Each group of adenoviruses could inhibit the proliferation of the 786- O cells and OSRC cells. However, the effects induced by G250- ZD55- Ki67 and G250- Ki67 on 786- O cells were stronger than on OSRC cells. Moreover, G250- ZD55- Ki67 had enhanced antitumor activities in these renal cancer cells compared with G250- Ki67. G250 promoter-derived CRAds carrying Ki67-si RNA could highly amplify and express Ki67-si RNA in renal cancer cells with expression of G250 antigen, inhibit renal cancer cells proliferation and induce apoptosis. These results demonstrated that the G250-specific oncolytic adenovirus expressing Ki67-si RNA is applicable for human renal clear cell cancer therapy. [ABSTRACT FROM AUTHOR]
