O circovírus é um DNA vírus, não envelopado, de fita simples e o agente etiológico da circovirose em aves, que acomete comumente os psitacídeos causando a Doença do Bico e das Penas dos Psitacídeos (Psittacine Beak and Feather Disease - PBFD). A enfermidade é altamente contagiosa e letal, sendo considerada exótica no Brasil. O objetivo da presente revisão é descrever as principais características virais e clínicas do circovírus na Ordem Psittaciformes. O circovírus é de fácil disseminação e sua transmissão ocorre principalmente por aerossóis e pelo pó das penas. Os principais sinais clínicos envolvem alterações e distrofias de penas e, em menor ocorrência, alterações em bico. Sua manifestação clínica ocorre de acordo com a idade e a condição imunológica da ave, podendo se apresentar na forma hiperaguda, aguda ou crônica. Devido sua característica imunossupressora, o óbito por infecções secundárias é comumente relatado. O principal método de diagnóstico laboratorial na atualidade é a reação em cadeia pela polimerase (PCR). Não existe tratamento efetivo para a circovirose (PBFD) e a eutanásia é indicada como forma de controle da doença. Medidas de quarentena e higiene rigorosa devem ser aplicadas para evitar a disseminação do agente em um plantel e, principalmente, o escape viral para vida livre. Isso se faz necessário sobretudo no Brasil, país que detém uma rica diversidade de espécies de psitacídeos.

Eight lovebirds of both sexes and different age were admitted showing alterations in behaviour and apathy. During the initial examination delamination of the beak was noted in all birds and discrete areas of alopecia in three of eight birds. Based on clinical experience, molecular diagnostic on PBFD, sequencing of obtained amplicons, and histopathological examination were performed. All birds tested positive on the presence of PBFD virus, despite the fact they did not exhibit expected clinical signs for PBFD. Sequencing results showed 100% match with sequences previously isolated from parrots with PBFD exhibiting classical PBFD manifestation. Histopathological examination showed similar findings as in previously described cases of PBFD in parrots. Other studies on psittacine birds correlate clinical manifestations and nucleotide variations with geographic localization. Our results indicate that the clinical manifestation of the disease is more dependent on bird species than on the genetic variation of the virus or the geographical distribution.

Background: Beak and feather disease virus (BFDV) is a circovirus that infects captive and wild psittacine birds, and is of conservation concern. The haemagglutination inhibition (HI) assay is used to determine antibody titres against BFDV, and the use of dried blood spots (DBS) on filter paper stored at room temperature has been suggested to be an equally valid technique to the use of frozen serum. However, research on other pathogens has found variable results when investigating the longevity of antibodies stored on DBS at room temperature. Consequently, we aimed to test the temporal stability of antibodies to BFDV in DBS samples stored long-term at room temperature. A further goal was to add to the current knowledge of antibody response to naturally acquired BFDV infection in crimson rosellas (Platycercus elegans). Methods: Blood was collected from wild P. elegans in Victoria, Australia, that had been live-trapped (n = 9) or necropsied (n = 11). BFDV virus load data were obtained from blood stored in ethanol by real-time quantitative PCR (qPCR); antibody titres were obtained by HI assay from either DBS or serum samples, which had been collected concurrently. All HI assays were performed commercially by the Veterinary Diagnostic Laboratory (VDL) in Charles Sturt University, Australia, who were blind to BFDV blood status. Results: HI titres from DBS stored at room temperature declined significantly over time (~80 weeks). By contrast, frozen serum samples assayed after 80 weeks in storage all had high HI titres, only varying up to one dilution step from the initial HI titres obtained from DBS at 3-6 weeks after sampling. Weak HI titres from DBS samples all came back negative when the test was repeated only nine weeks later. Novel high HI titres were reported in P. elegans, and while most birds with high antibody titres had corresponding negative qPCR results, a single subadult presented with high HI titres and virus load simultaneously. Conclusion: Detection of antibodies on filter paper stored at room temperature decreases over time, increasing the chances of false negatives in these samples, and in repeated testing of samples with weak HI titres. Consequently, serum should be the preferred sample type to use for seroepidemiological studies on BFDV in parrots and other bird species. When not possible, it may help to store DBS on filter paper at -20 °C or lower. However, prompt testing of DBS samples (e.g., <6 weeks in storage) is recommended pending further research on antibody temporal stability. Wildlife can be hosts for pathogens of conservation and zoonotic concern. Infection with one pathogen can increase the risk of co-infection with others, with resulting impacts on hosts, for example on body condition or survival. However, studies of co-infection in wild populations are rare. Globally, psittacine birds can be infected with beak and feather disease virus (BFDV), a circovirus of significant conservation concern, and with bacteria of the order Chlamydiales, including the zoonotic Chlamydia psittaci. These pathogens are common in Australian psittacines, and their putatively immunosuppressive effects would predict co-infection to be more common than expected due to chance alone. Here, we test this hypothesis across four abundant and widespread Australian parrot species: Crimson Rosella, Sulphur-crested Cockatoo, Galah and Blue-winged Parrot. Using wild-caught birds, we tested for pathogen DNA and, in the case of Chlamydiales, also antibodies, to investigate both current and previous infection status. We detected co-infection prevalence of up to 16.4% in the combined samples from the four species and up to 28.9% in Crimson Rosellas, the species with the largest sample size. Contrary to expectations, however, we found that the incidence of infections with the two pathogens was statistically independent. There was also no effect of co-infection on body condition or haematocrit. Our findings therefore suggest that while concurrent infection does occur, it is no more common than expected by chance. We discuss limitations of the data and suggest avenues for future research. Overall, our study provides insights that potentially benefit psittacine conservation, welfare, and human health. Beak and feather disease virus (BFDV), a circovirus, is the etiologic agent of psittacine beak and feather disease (PBFD), a progressive and often fatal disease in Psittaciformes. Even though neotropical psittacine species are more resistant to clinical infection than Old World species, BFDV is recognized as a threat to immunologically naïve wild psittacine flocks and its epidemiologic control is paramount for conservation efforts in Neotropical species. Samples were collected from multiple psittacine species, including Ara species, Amazona species, and the white-crowned parrot (Pionus senilis) from the only rescue center in Guatemala with formal psittacine rehabilitation and reintroduction programs. A total of 117 birds, with 101 adults and 16 juveniles of unknown sex, were tested for BFDV by means of a real-time polymerase chain reaction (PCR) assay. The BFDV prevalence found in this study was 0%, (95% confidence interval, 0%–6.0%). Seven 2–8-year-old scarlet macaws (Ara macao cyanoptera) with positive results from previous surveys by conventional PCR yielded negative results in this study, suggesting complete infection resolution. Indirect transmission of pathogens can pose major risks to wildlife, yet the presence and persistence of wildlife pathogens in the environment has been little studied. Beak and feather disease virus (BFDV) is of global conservation concern: it can infect all members of the Psittaciformes, one of the most threatened bird orders, with infection often being lethal. Indirect transmission of BFDV through contaminated nest hollows has been proposed as a major infection source. However, data on whether and for how long nest sites in the wild remain contaminated have been absent. We determined the BFDV status of birds (parents and nestlings) for 82 nests of Crimson Rosellas, Platycercus elegans and Eastern Rosellas, Platycercus eximius. In 11 of these nests (13.4%, 95% confidence interval 6.9–22.7), we found an infected parent or nestling. Using nest swabs, we then compared BFDV presence at three points in time (before, during and after breeding) in three groups of nest boxes. These were nest boxes occupied by infected birds, and two control groups (nest boxes occupied by uninfected birds, and unoccupied nest boxes). Detection of BFDV on nest swabs was strongly associated with the infection status of parents in each nest box and with the timing of breeding. During breeding, boxes occupied by BFDV-positive birds were significantly more likely to have BFDV-positive nest swabs than boxes occupied by BFDV-negative birds; nest swabs tested BFDV-positive in 80% (28.4–99.5) of nests with parental antigen excretion, 66.7% (9.4–99.2) of nests occupied by parents with BFDV-positive cloacal swabs and 66.7% (22.3–95.7) of nests occupied by parents with BFDV–positive blood. 0% (0–52.2) of nests with BFDV–positive nestlings had BFDV–positive nest swabs. Across all boxes occupied by BFDV-positive birds (parents or nestlings), no nest swabs were BFDV–positive before breeding, 36.4% (95% CI 10.9–69.2) were positive during breeding and 9.1% (0.2–41.3) remained positive after breeding. BFDV was present on nest swabs for up to 3.7 months. Our study provides novel insights into the potential role of nest cavities and other fomites in indirect transmission of BFDV, and possibly other pathogens, and offers a non-invasive method for surveillance of pathogens in wild bird populations.

Conservation efforts for the orange-bellied parrot (Neophema chrysogaster), one of the world's most critically endangered bird species, have been hampered by beak and feather disease virus (BFDV) spillover infection. To understand the vulnerability of orange-bellied parrots to potential reservoirs of infection we investigated geographic versus taxonomic structure in 160 full-genome and 319 partial Rep gene BFDV sequences from captive and wild orange-bellied parrots and other wild parrot species in Australia. We found that Australian BFDV populations are structured by host taxonomy. By identifying genetic stratification of BFDV in reservoir hosts we characterized three separate recent incursions of BFDV into orange-bellied parrots from other wild parrots, which demonstrates the susceptibility of critically endangered species to multiple threats of pathogen re-emergence. Our study highlighted how loss of endemic circulating BFDV in orange-bellied parrots precipitated repeated spillover into an immunologically naïve population, causing significant disease. Emerging infectious diseases (EIDs) are key contributors to the current global biodiversity crisis. Psittaciformes (parrots) are one of the most vulnerable avian taxa and psittacine beak and feather disease (PBFD) is the most common viral disease in wild parrots. PBFD is caused by the beak and feather disease virus (BFDV), which belongs to the Circoviridae family and comprises a circular, single-stranded DNA genome. BFDV is considered to have spread rapidly across the world and, in 2005, an outbreak of PBFD was documented in the recovering population of the Mauritius parakeet (Alexandrinus eques). The Mauritius parakeet was once the world’s rarest parrot and has been successfully recovered through 30 years of intensive conservation management. Molecular surveillance for the prevalence of BFDV was carried out across a 24-year sample archive spanning the period from 1993 to 2017, and DNA sequencing of positive individuals provided an opportunity to assess patterns of phylogenetic and haplotype diversity. Phylogenetic analyses show variation in the extent of viral diversification within the replicase protein (Rep). Timeseries of BFDV prevalence and number of haplotypes reveal that two subsequent waves of infection occurred in 2010/2011 and 2013/2014 following the initial outbreak in 2005. Continued disease surveillance to determine the frequency and intensity of subsequent waves of infection may benefit future translocation/reintroduction planning. The continued growth of the Mauritius parakeet population despite the presence of BFDV bodes well for its long-term persistence.

The establishment of viral pathogens in new host environments following spillover events probably requires adaptive changes within both the new host and pathogen. After many generations, signals for ancient cross-species transmission may become lost and a strictly host-adapted phylogeny may mimic true co-divergence while the virus may retain an inherent ability to jump host species. The mechanistic basis for such processes remains poorly understood. To study the dynamics of virus–host co-divergence and the arbitrary chances of spillover in various reservoir hosts with equal ecological opportunity, we examined structural constraints of capsid protein in extant populations of Beak and feather disease virus (BFDV) during known spillover events. By assessing reservoir-based genotype stratification, we identified co-divergence defying signatures in the evolution BFDV which highlighted primordial processes of cryptic host adaptation and competing forces of host co-divergence and cross-species transmission. We demonstrate that, despite extensive surface plasticity gathered over a longer span of evolution, structural constraints of the capsid protein allow opportunistic host switching in host-adapted populations. This study provides new insights into how small populations of endangered psittacine species may face multidirectional forces of infection from reservoirs with apparently co-diverging genotypes. The psittacine beak and feather disease (PBFD) is a globally widespread infectious bird disease that mainly affects species within the Order Psittaciformes (parrots and allies). The disease is caused by an avian circovirus (the beak and feather disease virus, BFDV), which is highly infectious and can lead to severe consequences in wild and captive populations during an outbreak. Both legal and illegal trading have spread the BFDV around the world, although little is known about its prevalence in invasive parrot populations. Here, we analyze the BFDV prevalence in sympatric invasive populations of rose-ringed (Psittacula krameri) and monk parakeets (Myiopsitta monachus) in Southern Spain. We PCR-screened 110 blood samples (55 individuals from each species) for BFDV and characterized the genotypes of five positives from each species. About 33% of rose-ringed parakeets and 37% of monk parakeets sampled were positive for BFDV, while neither species showed disease symptoms. The circovirus identified is a novel BFDV genotype common to both species, similar to the BFDV genotypes detected in several parrot species kept in captivity in Saudi Arabia, South Africa and China. Our data evidences the importance of an accurate evaluation of avian diseases in wild populations, since invasive parrots may be bringing BFDV without showing any visually detectable clinical sign. Further research on the BFDV prevalence and transmission (individual–individual, captive–wild and wild–captive) in different bird orders and countries is crucial to understand the dynamics of the viral infection and minimize its impact in captive and wild populations.

Beak and feather disease virus (BFDV) is a causative agent for psittacine beak and feather disease (PBFD), which shows a characteristic feather disorder in psittacine birds. Nineteen budgerigars, which were clinically suspected to have PBFD, were examined by two polymerase chain reactions (PCR), which target each of open reading frames (ORFs) V1 and C1. All of the 19 samples were detected BFDV by the PCR targeting ORF C1, whereas only two of them were detected by the PCR targeting ORF V1. It was assumed that BFDV derived from budgerigar (budgerigar BFDV) has two genotypes, which are tentatively classified as budgerigar BFDV genotype 1 and genotype 2 by the PCR amplification patterns. Whole genome sequences of six budgerigar BFDVs were determined to reveal the existence of two genotypes. In the phylogenic analysis, six budgerigar BFDV sequences formed a unique group branched from the other 23 published BFDV sequences. The budgerigar BFDV genotype 1 and genotype 2 were also segregated each other, and budgerigar BFDV genotype 2 was particularly distantly related with the other BFDVs. The hematological characteristics of juvenile long-billed corellas ( Cacatua tenurostris), with or without prior administration of a psittacine beak and feather disease vaccine, were studied for 97 days after experimental infection with beak and feather disease virus (BFDV). It was found that the pre-challenge hematological values were similar between vaccinated and non-vaccinated corellas. Most pre-challenge parameters were comparable to previously reported values of other cockatoos and psittacine birds. Significant differences were seen in both groups when comparing pre-challenge values with post-challenge values for total and differential leukocyte concentrations, but packed cell volume and total serum protein were not significantly affected by BFDV challenge. Abstract: PCR-based assays for the detection of BFDV DNA are in widespread use throughout the world. Quantitative real-time PCR assays are extremely sensitive and have the advantages over standard PCR assays that they do not require post-reaction processing to visualise PCR products and can quantify the amount of DNA present in a sample. This study describes a quantitative real-time PCR assay for the detection of BFDV DNA, using primers designed to amplify a conserved 81bp fragment of ORFV1 and SYTO9, a fluorescent intercalating dye. A synthetic oligonucleotide was used to make standard curves for the quantitation of viral load in blood and feather preparations. The assay was very sensitive, with a detection limit of 50copies/μL. The assay was developed using DNA extracts from the feathers of 10 different species of birds which had tested BFDV-positive previously and was validated with blood and feather samples from corellas vaccinated with an experimental BFDV vaccine, then challenged with live virus. Viral DNA was detected consistently in the blood of all control (non-vaccinated) birds and in some vaccinated birds. Contamination of the environment with feather dander from BFDV-infected birds meant that feather preparations used for the haemagglutination assay were unreliable for the detection and quantitation of viral excretion. Nonetheless, the assay should prove to be a useful and sensitive test for the detection of viral DNA in a range of samples. [Copyright &y& Elsevier]

Abstract: Currently, the only diagnostic test available routinely for the sero-diagnosis of BFDV is the haemagglutination-inhibition (HI) assay. This test, whilst useful and applicable to samples from a wide range of psittacine birds, is not an ideal assay; it requires erythrocytes from live animals, virus purified from the feathers of infected birds and polyclonal antibody preparations in order to perform the assay. Variations in these reagents make consistency between tests difficult to achieve, underscoring the need for a new test with standardised reagents for the sero-diagnosis of BFDV infection which has led to the development of an antibody response. The methods used to develop a novel “blocking” (or “competitive”) ELISA (bELISA) for the detection of anti-BFDV antibodies in psittacine sera are presented in this paper. The assay was developed using a baculovirus-expressed recombinant BFDV capsid protein and a newly developed monoclonal antibody raised against this protein. The assay was then validated with 160 samples from eastern long-billed corellas (Cacatua tenuiostris) vaccinated with the recombinant capsid protein and challenged with live virus and samples from 82 cockatiels known to be HI negative. This study describes for the first time the presence of circoviruses in ostrich tissue including embryos. A polymerase chain reaction (PCR) was used for the detection of the virus in liver samples. The use of a polymerase for low copy detection significantly increased the sensitivity of the test as well as a Southern blot. Viral DNA could be detected in chicks and eggs that did not hatch. The circovirus identified is a novel BFDV genotype common to both species, similar to the BFDV genotypes detected in several parrot species kept in captivity in Saudi Arabia, South Africa and China. Our data evidences the importance of an accurate evaluation of avian diseases in wild populations, since invasive parrots may be bringing BFDV without showing any visually detectable clinical sign. Further research on the BFDV prevalence and transmission (individual–individual, captive–wild and wild–captive) in different bird orders and countries is crucial to understand the dynamics of the viral infection and minimize its impact in captive and wild populations.
