Abstract For sustainable food production selection and breeding of feed efficient animals is crucial. The objective of this study was to evaluate whether multiparous dairy cows, ranked during their first lactation based on residual energy intake (REI) as efficient (low; L-REI) or inefficient (high; H-REI), differ in terms of nutrient use efficiency, methane emissions, rumen fermentation, and gut microbiota composition. Six L-REI and 6 H-REI cows were offered two diets with either a low or high proportion of concentrates (30 vs. 50% of DM) on two consecutive periods of 21 d. Gas exchanges, milk yield, feces and urine excretions were measured in open-circuit respiratory chambers. The results indicated that L-REI cows had higher methane yields (22.6 vs. 20.4 g/kg DM intake) and derived more energy (energy balance − 36.6 vs. − 16.9 MJ/d) and protein (N balance − 6.6 vs. 18.8 g/d) from the tissues to support similar milk yields compared to H-REI cows. Nutrient intake and digestibility were not affected by REI, and there were no interactions between REI and diet. Milk yield, milk production efficiency, and milk composition were not affected by REI except for milk urea concentration that was higher for L-REI cows (14.1 vs. 10.8 mg/100 ml). The rumen and fecal microbiota community structure and function were associated with both the diet and REI, but the diet effect was more pronounced. The current study identified several physiological mechanisms underlying the differences between high and low REI cows, but further studies are needed to distinguish the quantitative role of each mechanism.

The effects of grass silage and barley grain preservation methods on dairy cows were evaluated using four Nordic Red dairy cows placed in respiration chambers in a 4 × 4 Latin square. Silage was conserved using a formic acid-based product (AS) or a homofermentative lactic acid bacteria inoculant (IS), while grains were dried (DB) or crimped and ensiled (EB). Fermentation profile of silages and the chemical composition of the mixed diets were very similar. The dietary treatments did not affect feed intake, milk production, and rumen fermentation except molar proportion of butyrate, and energy metabolism. Digestibility of dry matter and organic matter were higher (p < 0.05) and that of crude protein was lower (p < 0.05) for AS than IS. Feeding EB compared to DB decreased (p < 0.05) diet organic matter and starch digestibility. The cows receiving AS tended (p = 0.06) to emit more methane per day than those receiving IS, but methane yield and intensity were not different between dietary treatments. Bacteria alpha diversity was higher (p < 0.01) in barley samples than grass silages and was not affected by the diet in rumen samples. All freshly prepared rations were dominated by Lactobacillaceae, Erwiniaceae, and Pseudomonadaceae but rations based on AS than IS remained more stable over 2 days. In conclusion, grass silage and barley grain preservation methods did not affect the measured parameters in dairy cows and the preservation method can be selected based on practical on-farm factors.

Negative energy status in early lactation is linked to a variety of metabolic disorders, reduced fertility, and decreased milk production. To improve the energy status of cows by breeding and management, the identification of negative energy status is crucial. While biomarkers such as non-esterified fatty acid (NEFA) concentration and beta-hydroxybutyrate (BHB) in blood plasma could be used to identify a negative energy state, measuring them directly from blood is both invasive and expensive. In this work, we developed prediction equations for blood plasma NEFA and BHB levels based on mid-IR spectral measurements of milk. The models were fitted using partial least squares regression and evaluated using both cross-validation and independent-herd validation. A total of 3 183 spectral records from 606 lactations originating from three different herds were utilised. R2 values of 0.53 (RMSE = 0.206 mmol/l, RMSE of cross-validation (RMSECV) 0.217 mmol/l) for NEFA and 0.63 (RMSE = 0.326 mmol/l, RMSECV = 0.353 mmol/l) for BHB were obtained. Furthermore, relatively similar prediction accuracies were found for BHB (RMSE of prediction (RMSEP) 0.411 mmol/l and 0.422 mmol/l) and NEFA (RMSEP = 0.186 mmol/l and 0.221 mmol/l) when model training was done using two herds and validated on the third herd. The results from the model fits confirm that it is possible to build blood plasma BHB and NEFA models based on mid-IR spectra that are sufficiently accurate for practical use.

Rumen microbiota modulation during the pre-weaning period has been suggested as means to affect animal performance later in life. In this follow-up study, we examined the post-weaning rumen microbiota development differences in monozygotic twin-heifers that were inoculated (T-group) or not inoculated (C-group) (n = 4 each) with fresh adult rumen liquid during their pre-weaning period. We also assessed the treatment effect on production parameters and methane emissions of cows during their 1st lactation period. The rumen microbiota was determined by the 16S rRNA gene, 18S rRNA gene, and ITS1 amplicon sequencing. Animal weight gain and rumen fermentation parameters were monitored from 2 to 12 months of age. Vegetable by-products have great potential for use as animal feeds and thus could improve the sustainability of the food system. The objective was to evaluate the milk production potential of potato by-product (PBP) replacing cereal grains in grass silage-based total mixed ration (TMR). Additionally, a laboratory scale experiment was conducted to assess the effect of PBP and chemical preservatives on the aerobic stability of TMRs. A change-over dairy cow feeding experiment was conducted to evaluate the inclusion of 135 g/kg (on a dry matter (DM) basis) of PBP in TMR. Additionally, the aerobic stability of TMR was evaluated by preparing TMRs with increasing levels of PBP and treated with different doses of formic and propionic acid or salt-based preservatives. The inclusion of PBP in dairy cow diets decreased feed intake, but organic matter digestibility was greater in the PBP diet, resulting in only slightly decreased production (milk protein 1435 vs. 1363 g/day, p < 0.05; milk lactose 1716 vs. 1606 g/day, p < 0.05). The aerobic stability of TMR was negatively affected by the inclusion of PBP, but it could to a limited extent be prolonged using chemical preservatives. The inclusion of PBP decreased feed intake and milk production to some extent when included in the diet of high-producing dairy cows. The utilization of PBP may, however, be justified from a sustainability point of view, as it provides a way to circulate the nutrients of a non-human edible feed material back into the food chain.

Efficient feed utilization in dairy cows is crucial for economic and environmental reasons. The rumen microbiota plays a significant role in feed efficiency, but studies utilizing microbial data to predict host phenotype are limited. In this study, 87 primiparous Nordic Red dairy cows were ranked for feed efficiency during their early lactation based on residual energy intake, and the rumen liquid microbial ecosystem was subsequently evaluated using 16S rRNA amplicon and metagenome sequencing. The study used amplicon data to build an extreme gradient boosting model, demonstrating that taxonomic microbial variation can predict efficiency (rtest = 0.55). Prediction interpreters and microbial network revealed that predictions were based on microbial consortia and the efficient animals had more of the highly interacting microbes and consortia. Rumen metagenome data was used to evaluate carbohydrate-active enzymes and metabolic pathway differences between efficiency phenotypes. The study showed that an efficient rumen had a higher abundance of glycoside hydrolases, while an inefficient rumen had more glycosyl transferases. Enrichment of metabolic pathways was observed in the inefficient group, while efficient animals emphasized bacterial environmental sensing and motility over microbial growth. The results suggest that inter-kingdom interactions should be further analyzed to understand their association with the feed efficiency of animals.

Microcrystalline cellulose (MCC) has several applications in food and pharmaceutical industries but the nutritional value for dairy cows and effects on in vivo digestion are not known. A feeding experiment was conducted using 24 dairy cows. The cows were offered MCC originating from unbleached softwood kraft pulp at 0, 10 or 100 g per kg diet dry matter (DM) to replace barley grain in the diet. The total daily DM intake was on average 25.6 kg and not significantly affected by the diet. Positive effects on rumen fermentation could not be demonstrated in a feeding situation where total mixed ration and a concentrate proportion of 0.50 on DM basis was used. Diet organic matter digestibility was not affected by MCC inclusion but fibre digestibility improved and the additional MCC fibre was virtually completely digested. The production potential of MCC was lower than that of barley grain as daily yields of energy-corrected milk, milk fat and protein, and milk protein concentration decreased when MCC replaced barley grain in the diet. Based on these results, MCC is not recommended as a dietary component for high-yielding dairy cows.

Grass was ensiled using an acid-based additive (AS) or homofermentative lactic acid bacteria (IS). In addition, barley grain was either dried (DB) or crimped and ensiled (EB). The feeds were fed as total mixed rations (TMR) in a 2 × 2 factorial arrangement to 16 Nordic Red dairy cows in four replicated Latin squares. The differences in the fermentation quality of the two grass silages were unexpectedly small. Dry matter intake was higher (p < 0.01) for cows fed AS than those fed IS but was not affected by barley preservation method. Ruminal molar proportion of butyrate tended to be higher in cows fed AS rather than IS (p < 0.10) in expense of acetate (p < 0.05). Barley preservation method did not affect rumen fermentation but modulated rumen bacterial community composition. Milk production was not affected by silage additive but tended (p < 0.10) to be higher (39.6 vs. 39.0 kg/d) for cows fed DB rather than EB. However, barley type did not affect energy corrected milk yield due to a tendency (p < 0.10) for higher milk fat content of cows fed EB rather than DB. Milk fat yield tended (p < 0.10) to be higher for AS-fed cows than IS-fed cows, and milk protein yield was higher for cows receiving DB rather than EB. The AS resulted in more aerobically stable TMR than IS and a minor advantage was found for DB compared to EB.

The relationship between heart girth, wither height, body length and body weight in 3- to 9.5-month-old pre-pubertal Finnish Ayrshire heifers gaining 600-650 g/d was analysed (experiment I). Regression analysis showed that heart girth was the trait most highly correlated to body weight (R2 = 0.969). Including body length or wither height as a second term in the regression, increased R2 values only slightly. When the relationship between heart girth and body weight was used to predict the body weight of heifers reared at two feeding levels (experiment II), the precision of prediction was affected by the plane of nutrition. Actual body weight for a given heart girth was slightly higher on the high than on the low feeding level. It is, nevertheless, concluded that the equations presented in the paper can be used to estimate accurately the body weight of pre-pubertal (95-140-cm heart girth) Ayrshire heifers gaining 550-700 g/d.
