Manganese (Mn), iron (Fe), copper (Cu), zinc (Zn), and selenium (Se) are essential micronutrients for human health. However, the genetic basis for the content of Mn, Fe, Cu, Zn, and Se in wheat grains remains unclear. A recombinant inbred lines (RIL) population derived from Yangmai 4/Yanzhan 1 (YM4/YZ1) with wheat 55K single nucleotide polymorphism (SNP) arrays and micronutrient content of two environments was used to construct a genetic linkage map and dissect the quantitative trait loci (QTL) for the content of Mn, Fe, Cu, Zn, and Se in wheat. A total of 8 QTL were detected and located on chromosomes 1A, 1B, 2D, 4D, 7A, and 7D, respectively. Among them, QFe.yaas-2D and QSe.yaas-2D were co-located on chromosome 2D, while QMn.yaas-4D and QZn.yaas-4D were co-located on chromosome 4D, which were in the dwarfing locus of Rht-D1 region. The positive alleles of QCu.yaas-1A, QMn.yaas-1B, and QZn.yaas-7D were contributed by YZ1 and explained 7.66–19.92% of the phenotypic variances, while the positive alleles of QFe.yaas-2D, QSe.yaas-2D, QMn.yaas-4D, QZn.yaas-4D, and QCu.yaas-7A were contributed by YM4 and explained 5.77–20.11% of the phenotypic variances. The positive alleles of QCu.yaas-1A, QMn.yaas-1B, and QMn/Zn.yaas-4D increased TGW by 3.52%, 3.45%, and 7.51% respectively, while the positive alleles of QFe/Se.yaas-2D decreased TGW by 6.45%. Six SNP markers flanked the target QTL were converted into Kompetitive allele specific PCR (KASP) markers, and their effects were validated in a panel of one hundred and forty-nine wheat advanced lines. Twenty-five advanced lines harboring at least five positive alleles were identified in the validation populations. A total of 60 and 51 high-confidence annotated genes for QFe/Se.yaas-2D and QMn/Zn.yaas-4D were identified using the International Wheat Genome Sequencing Consortium Reference Sequence v2.1 (IWGSC RefSeq v2.1), respectively. Some genes in these two regions were involved in stress tolerance, growth development, Zn synthesis in plants. These results provide the basis for fine-mapping the target QTL of micronutrient content and marker-assisted selection in grain quality breeding programs.

Purpose: To evaluate choroidal thickness and retinal nerve fiber layer (RNFL) thickness in different trimesters using enhanced depth imaging optical coherence tomography (EDI-OCT). Methods: A prospective comparative study included 45 healthy pregnant women in the first trimester, 45 women in the second, 45 women in the third and 45 healthy non-pregnant women as the control group. Macular choroidal thickness was measured at three locations: The subfoveal, 1 ​mm temporal, and 1 ​mm nasal from the fovea with EDI-OCT. Peripapillary choroidal thickness (PPCT) and RNFL thickness parameters were automatically calculated by the Spectralis OCT. Abstract Urban livability has become a major policy and practice priority in many parts of the world. However, its attainment remains challenging in many cities of developing and emerging economies. The lack of data with appropriate quality, coverage, and spatial and temporal resolution often complicates both the assessment of livability in such cities and the identification of priority areas for improvement. Here we develop a framework to mobilize and synthesize open-source data to analyze spatially urban livability patterns in Shanghai. The framework brings together diverse types of open-source data including housing characteristics, population distribution, transportation networks, and points of interest to identify city areas with low livability, and thus priority areas for improvement. Such findings can provide a comprehensive overview of the residential living conditions in Shanghai, as well as useful information to urban planners and decision-makers. Furthermore, subject to data availability, the proposed method has the potential for application in other cities.

Urban parks provide essential ecosystem services (ESs) that enhance human wellbeing. However, discrepancies often arise between objective assessments of these services and stakeholders’ subjective perceptions. This study addresses a research gap concerning the synergies and tradeoffs between objective evaluations and subjective perceptions of key ecosystem services across various spatial scales. We investigated six key ecosystem services in Century Park, Shanghai, across seven buffer radii (8–100 m). Objective data were obtained from park view images (PVIs) and spatial analysis, while subjective perceptions were gathered through a scoring survey of 33 stakeholders. The key finding is that a buffer radius of 35 m offers optimal synergy between objective and subjective assessments for most ESs, particularly in pollution mediation, temperature regulation, and cultural services. Professionals showed stronger alignment in regulatory services like pollution mediation and temperature regulation, while residents exhibited higher synergy in net primary production (NPP) beyond a 75 m radius. Notably, cultural services displayed nuanced differences, with professionals preferring simpler landscapes and residents demonstrating varied aesthetic preferences. These findings emphasize the importance of integrating objective data and human perceptions in urban green space planning and governance. By incorporating diverse stakeholders and identifying optimal buffer zones, planners and designers can effectively balance ESs with human experiences. This approach ultimately fosters more sustainable and wellbeing-centered urban environments.

As urbanization accelerates globally, urban areas have become major sources of greenhouse gas emissions. In this context, urban parks are crucial as significant components of carbon sinks. Using Shanghai Century Park as a case study, this study aims to develop an applicable and reliable workflow to accurately assess the carbon sequestration capacity of urban parks from a spatial–temporal perspective. Firstly, the random forest model is employed for biotope classification and mapping in the park based on multi-source data, including raw spectral bands, vegetation indices, and texture features. Subsequently, the Net Primary Productivity and biomass of different biotope types are calculated, enabling dynamic monitoring of the park’s carbon sequestration capacity from 2018 to 2023. Moreover, the study explores the main factors influencing changes in carbon sequestration capacity from the management perspective. The findings reveal: (1) The application of multi-source imagery data enhances the accuracy of biotope mapping, with winter imagery proving more precise in classification. (2) From 2018 to 2023, Century Park’s carbon sequestration capacity showed a fluctuating upward trend, with significant variations in the carbon sequestration abilities of different biotope types within the park. (3) Renovation and construction work related to biotope types significantly impacted the park’s carbon sequestration capacity. Finally, the study proposes optimization strategies focused on species selection and layout, planting density, and park management.

The relatively low export innovation capacity is not conductive to the steady transformation and upgrading of China’s manufacturing industry, and it is necessary to implement suitable policies to enhance export innovation capacity. This study empirically analyzes the data of 827,471 manufacturing enterprises from 2000 till 2013 to investigate the impact of subsidy policy on export innovation. The overall results show that China’s subsidy policy has a significant crowding-out effect on export innovation, and subsidy for relatively small enterprises is more conducive to promoting export innovation; however, enterprises’ independent investment does not own much impact on export innovation. The econometric results made from perspectives of the sub-region and the industry reveal that subsidy policy is not conducive to export innovation, independent investment is not beneficial to export innovation, and export innovation ability is positively correlated with enterprise scale, but the influence coefficient shows obvious differences.

High temperatures accelerate the accumulation of storage material in seeds, often leading to defects in grain filling. However, the mechanisms regulating grain filling at high temperatures remain unknown. Here, we want to explore the quality factors influenced by the environment and have identified a LATE EMBROYGENESIS ABUNDANT gene, OsLEA1b, a heat-stress-responsive gene in rice grain filling. OsLEA1b is highly expressed in the endosperm, and its coding protein localizes to the nucleus and cytoplasm. Knock-out mutants of OsLEA1b had abnormal compound starch granules in endosperm cells and chalky endosperm with significantly decreased grain weight and grain number per panicle. The oslea1b mutants exhibited a lower proportion of short starch chains with degrees of polymerization values from 6 to 13 and a higher proportion of chains with degrees from 14 to 48, as well as significantly lower contents of starch, protein, and lipid compared to the wild type. The difference was exacerbated under high temperature conditions. Moreover, OsLEA1b was induced by drought stress. The survival rate of oslea1b mutants decreased significantly under drought stress treatment, with significant increase in ROS levels. These results indicate that OsLEA1b regulates starch biosynthesis and influences rice grain quality, especially under high temperatures. This provides a valuable resource for genetic improvement in rice grain quality.

Abstract Sulforaphane (SFN) extracted from broccoli sprout has previously been investigated for its potential properties in cancers, however, the underlying mechanisms of the anticancer activity of SFN remain not fully understood. In the present study, we investigate the effects of SFN on cell proliferation, cell cycle, cell apoptosis, and also the expression of several cell cycle and apoptosis-related genes by MTT assay, flow cytometry and western blot analysis in gastric cancer (GC) cells. The results showed that SFN could impair the colony-forming ability in BGC-823 and MGC-803 cell lines compared with the control. In addition, SFN significantly suppressed cell proliferation by arresting the cell cycle at the S phase and enhancing cell apoptosis in GC cells in a dose-dependent manner. Western blot results showed that SFN treatment significantly increased the expression levels of p53, p21 and decreased CDK2 expression, which directly regulated the S phase transition. The Bax and cleaved-caspase-3 genes involved in apoptosis executive functions were significantly increased in a dose-dependent manner in BGC-823 and MGC-803 cells. These results suggested that SFN-induced S phase cell cycle arrest and apoptosis through p53-dependent manner in GC cells, which suggested that SFN has a potential therapeutic application in the treatment and prevention of GC.

Abstract Background The increase in atmospheric CO2 is causing a number of changes in plant growth such as increases in leaf area and number, branching, plant size and biomass, and growth rate. Despite the importance of stomatal responses to CO2, little is known about the genetic and molecular mechanisms that mediate stomatal development and movement in response to CO2 levels. Deciphering the mechanisms that sense changes in CO2 and/or HCO3 − concentration is critical for unraveling the role of CO2 in stomatal development movement. In Arabidopsis, CO2-induced stomatal closure is strongly Ca2+-dependent. To further dissect this signaling pathway and identify new components in the CO2 response pathway, we recorded [Ca2+]cyt changes in mutagenized Arabidopsis leaves and screened for mutants with abnormal guard cell behavior in response to CO2/HCO3 −. Results We observed that 1 mM HCO3 − induces [Ca2+]cys transient changes in guard cells and stomatal closure both in light and darkness. The changes in [Ca2+]cys induced by HCO3 − could be detected by an aequorin-based calcium imaging system. Using this system, we identified a number of Arabidopsis mutants defective in both [Ca2+]cyt changes and the stomatal response to CO2/HCO3 −. Conclusions We provide a sensitive method for isolating stomatal CO2/HCO3 − response genes that function early in stomatal closure and that have a role in regulating [Ca2+]cyt. This method will be helpful in elucidating the Ca2+-dependent regulation of guard cell behavior in response to CO2/HCO3 −.

High levels of ammonium (NH4+) in soils inhibit plant growth and nitrogen utilization efficiency. Elucidating the underlying mechanisms of NH4+ toxicity is essential for alleviating the growth inhibition caused by high NH4+. Our previous work showed that [Ca2+]cyt-associated protein kinase 1 (CAP1) regulates root hair growth in response to NH4+ in Arabidopsis thaliana, and the cap1-1 mutant produces short root hairs under NH4+ stress conditions. However, it is unclear whether CAP1 functions in other physiological processes in response to NH4+. In the present study, we found that CAP1 also plays a role in attenuating NH4+ toxicity to promote shoot growth. The cap1-1 mutant produced smaller shoots with smaller epidermal cells compared with the wild type in response to NH4+ stress. Disruption of CAP1 enhanced the NH4+-mediated inhibition of the expression of cell enlargement-related genes. The cap1-1 mutant showed elevated reactive oxygen species (ROS) levels under NH4+ stress, as well as increased expression of respiratory burst oxidase homologue genes and decreased expression of catalase genes compared with the wild type. Our data reveal that CAP1 attenuates NH4+-induced shoot growth inhibition by promoting cell wall extensibility and ROS homeostasis, thereby highlighting the role of CAP1 in the NH4+ signal transduction pathway.

Background Gastric gland mucin is important for maintaining the basic function of the gastric mucosa, protecting it from foreign substances and reducing the occurrence of gastric diseases. Exploring the phenotype of gastric gland mucus changes during the progression of gastric disease is of great clinical significance. Methods A total of 483 patients with different gastric diseases were collected in this study, including 82 superficial gastritis (SG), 81 atrophic gastritis (AG), 168 dysplasia (GD), and 152 gastric cancer (GC). Mucin staining was performed using HID-ABpH2.5-PAS method and was further grouped according to the mucin coloration. Results The phenotypic characteristics of mucin during disease progression were divided into neutral, acidic, and mucus-free types. Furthermore, acidic mucus can be divided into type I, type II, and type III. Abstract Gastric tumorigenesis is a multistep process initiated by chronic superficial gastritis (SG), followed by atrophic gastritis (AG), then intestinal metaplasia (IM), and finally by dysplasia and adenocarcinoma according to the Correa model. Pepsinogen C (PGC) decreases gradually during progression of cancer, which makes PGC an ideal negative marker for GC. To explore the correlation between PGC and other positive tumor markers in different gastric diseases, we observed the expression of PGC, MG7‐Ag, MMP9, NM23, Ki‐67, and E‐cadherin by immunohistochemistry, quantitative RT‐PCR, and immunoblot analysis. Our results showed that in SG, PGC was highly expressed while malignant phenotype markers were rarely expressed. In contrast with SG, malignant phenotype markers were highly expressed while the positive rate of PGC reached only 1.44% in GC. So there was no coexpression of PGC and malignant phenotype markers in SG or GC tissues. Only in the AG group, which is well‐known to be gastric precancerous disease, coexpression of PGC and malignant phenotype markers was detected. Our results suggested that the expression of PGC in AG was negatively correlated with that of MG7‐Ag and MMP9. Of all AG, those with low expression of PGC and high expression of MG7‐Ag and MMP9 may possess a greater potential of malignant transformation. Combined detection of negative marker PGC and positive markers MG7‐Ag and MMP9 could be used as a potential follow‐up panel for monitoring dynamical progression of AG and improving the detection efficiency of high‐risk individuals of gastric cancer, and then taking necessary interventions on the target population.

Abstract Helicobacter pylori infection is the most important risk factor for gastric intestinal metaplasia (IM). Our previous study demonstrated that infection with H. pylori HpslyD-positive strains associated with IM. To further investigate the signalling pathway involved in HpSlyD-induced IM, CDX2 and VIL1 expressions were determined before and after HpSlyD application. TCTP was knocked down by siRNA or overexpressed by plasmid transfection. An HpSlyD binding protein was used to block HpSlyD’s enzymatic activity. The expression of CDX2 and TCTP in gastric diseases was measured by immunohistochemistry. Our results showed HpSlyD induced CDX2 and VIL1 expressions. TCTP protein expression was markedly increased after application of HpSlyD and in an HpSlyD-expressing stable cell line. Downregulation of TCTP protein led to decreased HpSlyD-induced CDX2 and VIL1. Overexpression of TCTP protein improved the expression of CDX2 and VIL1. Co-application of HpSlyD and FK506 led to significant reductions in CDX2, VIL1, and TCTP expression. Immunohistochemistry demonstrated that CDX2 and TCTP expression was higher in HpslyD-positive specimens compared with HpslyD-negative ones. Expression of CDX2 was positively correlated with TCTP in HpslyD-positive cells. Our study is the first to show that HpSlyD induction of CDX2 and VIL1 expression mediated through TCTP may contribute to IM in the stomach.

The impact of phase structure on piezoelectric performances of textured Na0.5Bi0.5TiO3 (NBT) based lead-free ceramics was studied, including 0.88NBT-0.08K0.5Bi0.5TiO3-0.04BaTiO3 (88NBT) with morphotropic phase boundary (MPB) composition and 0.90NBT-0.07K0.5Bi0.5TiO3-0.03BaTiO3 (90NBT) with rhombohedral phase. Both textured ceramics exhibit a high Lotgering factor, being on the order of f~96%. The piezoelectric coefficients of the textured 88NBT and 90NBT ceramics are increased by 20% and 60%, respectively, comparing to their randomly oriented ceramics. The piezoelectric enhancement of 90NBT textured ceramic is three times higher than 88NBT, revealing the phase structure plays a significant role in enhancing the piezoelectric performances of textured ceramics. Of particular significance is that the 90NBT textured ceramic exhibits almost hysteresis-free strain behavior. The enhanced piezoelectric property with minimal strain hysteresis is attributed to the poled rhombohedral engineered domain configuration.

This work provides a comparative study of the dielectric and piezoelectric properties of randomly oriented and textured 0.88Na0.5Bi0.5TiO3-0.08K0.5Bi0.5TiO3-0.04BaTiO3 (88NBT) ceramics. Textured ceramics were fabricated by template grain growth (TGG) method using NaNbO3 (NN) for templates. For textured ceramics with 4 wt% NN templates, a Lotgering factor of 96% and piezoelectric coefficient d33 of 185 pC/N were obtained. Compared to the randomly oriented ceramics, textured ceramics show lower strain hysteresis (H = 7.6%), higher unipolar strain of 0.041% with corresponding large signal piezoelectric coefficient d33* of 200 pm/V at applied field of 2 kV/mm. This enhancement can be explained by the grain orientation along direction by texturing, where an engineered domain configuration is formed after polarization, leading to decreased hysteresis and increased piezoelectric property.

Background: Multi-system damage is a hallmark of systemic lupus erythematosus (SLE), a chronic systemic autoimmune disease. The typical initial symptoms of SLE are arthritis and dermatosis, whereas the presence of intracranial mass lesions as the first manifestation of systemic lupus erythematosus is very rare. This report describes an 18-year-old woman with intracranial mass lesions associated with SLE. Case Report: An 18-year-old woman was initially admitted to the hospital because of headache for 3 days, weakness in left arm, and blurred vision for 1 day. Magnetic resonance imaging (MRI) of her brain showed multiple abnormal occupying lesions in the right frontoparietal lobe. However, no evidence of tumor or infection was found. One month later, she was readmitted with right limb weakness and aphasia for 1 day. Brain MRI showed obvious and new abnormal signal shadows in both the right parietal lobe and the left frontotemporal parieto-occipital lobes compared with the previous MRI. She responded positively to immunotherapy, which, in a woman of child-bearing age, supports the diagnosis of SLE. Ultimately, the presence of focal neurological symptoms, abnormal autoantibodies (such as antinuclear antibodies, anti-dsDNA antibodies, anti-SSA autoantibodies, and anti-ribosomal P protein antibodies), as well as her positive response to immunotherapy, contributed to the diagnosis of SLE with intracranial mass lesions. No recurrence was seen during 1 year of follow-up. Conclusions: It is unusual for SLE to present with intracranial mass lesions as the initial symptoms. The pathogenesis of the neurological symptoms of the patient may be small vessel thrombosis or vasculitis leading to cerebral masslike necrosis. [ABSTRACT FROM AUTHOR]

We propose a full-field optical method for the label-free and quantitative mapping of the velocities of red blood cells (RBCs) in capillaries. It integrates spatiotemporal demodulation and an autocorrelation algorithm, and measures RBC velocity according to the ratio of RBC length to lag time. Conventionally, RBC length is assumed to be a constant and lag time is taken as a variable, while our method treats both of them as variables. We use temporal demodulation and the Butterworth spatial filter to separate RBC signal from background signal, based on which we obtain the RBC length by image segmentation and lag time by autocorrelation analysis. The RBC velocity calculated now is more accurate. The validity of our method is verified by an in vivo experiment on a mouse ear. Owing to its higher image signal-to-noise ratio, our method can be used for mapping RBC velocity in the turbid tissue case. [ABSTRACT FROM AUTHOR]

Background: Serum pepsinogen (PG) I/II ratio has been widely used as "serological biopsy" for the screening of gastric cancer (GC) and atrophic gastritis (GA). However, study concerning in situ expression of PGs is currently insufficient, particularly for their relationship with serum PGs levels. This study was designed to investigate in situ expression of PGI and PGII in subjects with normal mucosa (NOR), superficial gastritis (GS), GA and GC, and to evaluate the correlations between PGs expressions in situ and in serum. Methods: 185 subjects were enrolled for the study, including 30 NOR, 70 GS, 54 GA and 31 GC. PGI and PGII expressions in situ and in serum were detected by immunohistochemistry and enzyme-linked immunosorbent assay (ELISA) respectively. H. pylori immunoglobulin (Ig) G was also determined by ELISA. Results: In situ expressions of PGI, PGII and PGI/II ratio consistently decreased in sequence of NOR/GS- > GA- > GC. The expressions of PGI, PGII and PGI/II ratio in situ were statistically higher in youngers than in olders (P < 0.05). In the NOR subjects, PGI staining was statistically higher in males than that in females (p = 0.02). For the correlations between in situ and serum expressions of PGI, PGII and PGI/II ratio, a borderline correlation in the total study sample (r = 0.131, P = 0.076) and a statistical correlation in GA cases (r = 0.307, P = 0.027) were observed for the PGI/II ratio. The PGI expression correlated well with that of PGII in situ and in serum. Conclusions: The in situ levels of PGI, PGII and PGI/II ratio sharply decreased in the GA and GC cases. The youngers exhibited higher levels of PGI, PGII and PGI/II ratios than the olders. The in situ PGI/II ratio rather than PGI and PGII alone showed certain correlation with that in serum, and the PGI expression correlated well with PGII expression. Objective To investigate the changes and significance of Toll-like receptor 7 （TLR7）， TLR9 and type I interferon （IFN-Ⅰ） in children with severe mycoplasma pneumoniae pneumonia （SMPP）. Methods 80 children with mycoplasma pneumoniae pneumonia （MPP） were divided into the non-severe MPP （MPP group） and SMPP groups （SMPP group）. 26 healthy children who underwent outpatient physical examination were chosen as the control group. Serum samples in each group were collected. In the SMPP group， bronchoalveolar lavage fluid （BALF） on the affected and contralateral sides of 26 children with unilateral lung consolidation， atelectasis， lung abscess or lung tissue necrosis on imaging examination were collected. The levels of TLR7， TLR9， myeloid differentiation factor 88 （MyD88）， interferon-α （IFN-α） and IFN-β in the serum and BALF samples were determined by enzyme-linked immunosorbent assay （ELISA）. Results The serum levels of TLR7， TLR9， MyD88， IFN-α and IFN-β in the SMPP and MPP groups were significantly higher than those in the control group， and the levels of all cytokines in the SMPP group were significantly higher compared with those in the MPP group （all P < 0.05）. The levels of TLR7， TLR9， MyD88， IFN-α and IFN-β in the BALF on the affected side were significantly higher than those on the contralateral side （all P < 0.01）. The area under the receiver operating characteristic curve （AUC） of serum TLR7 and MyD88 levels for predicting SMPP was 0.709 and 0.723. Conclusions Over-induced and generated TLR7， TLR9， IFN-α and IFN-β may be pathogenic factors for the incidence of SMPP and severe local lung tissue injury in children with SMPP. TLR7 and MyD88 can be used as predictors of SMPP to guide clinical treatment.
