Myocarditis is a potentially life-threatening inflammatory disease of the myocardium, often resulting from infectious and immune-mediated responses. Clinical presentation in severe cases often results in a devastating illness requiring extracorporeal membrane oxygenation support as a result of cardiogenic shock. Although endomyocardial biopsy is still considered the gold standard for diagnosis, it often reveals nonspecific lymphocytic infiltration. Because the precise cause is usually unknown, the initial treatment typically involves immunosuppression and frequent assessment of myocardial contractility. This report presents 3 rare cases of autoimmune diseases (polymyositis, immunoglobulin G4–related disease, and systemic lupus erythematosus) that require extracorporeal membrane oxygenation support as a result of fulminant myocarditis, including their follow-up periods.

4. Hyperglycemia Stimulates the Irreversible Catabolism of Branched-Chain Amino Acids and Generation of Ketone Bodies by Cultured Human Astrocytes

Astrocytes are considered to possess a noticeable role in brain metabolism and, as a partners in neuron–glia cooperation, to contribute to the synthesis, bioconversion, and regulation of the flux of substrates for neuronal metabolism. With the aim of investigating to what extent human astrocytes are metabolizing amino acids and by which compounds are they enriching their surroundings, we employed a metabolomics analysis of their culture media by 1H-NMR. In addition, we compared the composition of media with either 5 mM or 25 mM glucose. The quantitative analysis of culture media by 1H-NMR revealed that astrocytes readily dispose from their milieu glutamine, branched-chain amino acids, and pyruvate with significantly high rates, while they enrich the culture media with lactate, branched-chain keto acids, citrate, acetate, ketone bodies, and alanine. Hyperglycemia suppressed the capacity of astrocytes to release branched-chain 2-oxo acids, while stimulating the generation of ketone bodies. Our results highlight the active involvement of astrocytes in the metabolism of several amino acids and the regulation of key metabolic intermediates. The observed metabolic activities of astrocytes provide valuable insights into their roles in supporting neuronal function, brain metabolism, and intercellular metabolic interactions within the brain. Understanding the complex metabolic interactions between astrocytes and neurons is essential for elucidating brain homeostasis and the pathophysiology of neurological disorders. The observed metabolic activities of astrocytes provide hints about their putative metabolic roles in brain metabolism.

Despite developing new diagnostics, drugs, and vaccines, treating tuberculosis (TB) remains challenging. Monitoring inflammatory markers can contribute to more precise diagnostics of TB, identifying its active and latent forms, or monitoring its treatment success. We assessed alterations in plasma levels of 48 cytokines in 20 patients (17 males) with active pulmonary TB compared to age-matched healthy controls (n = 18). Blood samples were collected from individuals hospitalised with TB prior to commencing antibiotic therapy, after the first week, and following the third week. The majority of patients received treatment with a combination of four first-line antituberculosis drugs: rifampicin, isoniazid, ethambutol, and pyrazinamide. 6. Application of the Hydrophilic Interaction Liquid Chromatography (HILIC-MS) Novel Protocol to Study the Metabolic Heterogeneity of Glioblastoma Cells

Glioblastoma is a highly malignant brain tumor consisting of a heterogeneous cellular population. The transformed metabolism of glioblastoma cells supports their growth and division on the background of their milieu. One might hypothesize that the transformed metabolism of a primary glioblastoma could be well adapted to limitations in the variety and number of substrates imported into the brain parenchyma and present it their microenvironment. Additionally, the phenotypic heterogeneity of cancer cells could promote the variations among their metabolic capabilities regarding the utilization of available substrates and release of metabolic intermediates. With the aim to identify the putative metabolic footprint of different types of glioblastoma cells, we exploited the possibility for separation of polar and ionic molecules present in culture media or cell lysates by hydrophilic interaction liquid chromatography (HILIC). The mass spectrometry (MS) was then used to identify and quantify the eluted compounds. The introduced method allows the detection and quantification of more than 150 polar and ionic metabolites in a single run, which may be present either in culture media or cell lysates and provide data for polaromic studies within metabolomics. The method was applied to analyze the culture media and cell lysates derived from two types of glioblastoma cells, T98G and U118. The analysis revealed that even both types of glioblastoma cells share several common metabolic aspects, and they also exhibit differences in their metabolic capability. This finding agrees with the hypothesis about metabolic heterogeneity of glioblastoma cells. Furthermore, the combination of both analytical methods, HILIC-MS, provides a valuable tool for metabolomic studies based on the simultaneous identification and quantification of a wide range of polar and ionic metabolites—polaromics.

Adenosine is a multifunctional nucleoside with several roles across various levels in organisms. Beyond its intracellular involvement in cellular metabolism, extracellular adenosine potently influences both physiological and pathological processes. In relation to its blood level, adenosine impacts the cardiovascular system, such as heart beat rate and vasodilation. To exploit the adenosine levels in the blood, we employed the liquid chromatography method coupled with mass spectrometry (LC–MS). Immediately after collection, a blood sample mixed with acetonitrile solution that is either enriched with 13C-labeled adenosine or a newly generated mixture is transferred into the tubes containing the defined amount of 13C-labeled adenosine. The 13C-enriched isotopic adenosine is used as an internal standard, allowing for more accurate quantification of adenosine. This novel protocol for LC–MS-based estimation of adenosine delivers a rapid, highly sensitive, and reproducible means for quantitative estimation of total adenosine in blood. The method also allows for quantification of a few catabolites of adenosine, i.e., inosine, hypoxanthine, and xanthine. Our current setup did not allow for the detection or quantifying of uric acid, which is the final product of adenosine catabolism. This advancement provides an analytical tool that has the potential to enhance our understanding of adenosine’s systemic impact and pave the way for further investigations into its intricate regulatory mechanisms.

The paper deals with the development of the finite element method (FEM) model of piezoelectric beam elements, where the piezoelectric layers are located on the outer surfaces of the beam core, which is made of functionally graded material. The created FEM model of piezoelectric beam structure is reduced using the modal truncation method, which is one of model order reduction (MOR) method. The results obtain from reduced state-space model are compared with results obtain from finite element model. MOR state-space model is also used in the design of the linear quadratic regulator (LQR). Created reduced state-space model with feedback with the LQR controller is analysed and compared with the results from FEM model.

Considering that the lattice construction of the transmission tower consists of hundreds of trusses it is obvious that some auxiliary tool is necessary for preparation of such model in ANSYS APDL. This tool was programmed in SW Wolfram Mathematica.

In the contribution, our 3D FGM Timoshenko beam finite element with 12x12 stiffness and mass matrices for doubly symmetric open and closed cross-section [1] is extended by warping torsion effect (non-uniform torsion) to 14x14 finite element matrices. A longitudinal continuous variation of effective material properties is considered by the finite element equations derivation, which can be obtained by homogenization of the spatial varying material properties in real FGM beam. Results of numerical elastostatic non-uniform torsional analysis of the FGM cantilever beam of I-cross-section are presented and the accuracy and effectiveness of the new FGM beam finite element is discussed and evaluated.

The application of first order shear beam theory in the analysis of beam structures made of functionally graded materials requires the access to homogenized stiffness quantities. These quantities depend on the cross-sectional shape and on the spatial variation of constitutive parameters. Some of these stiffness quantities can be evaluated easily by simple integration, however, the access to transverse shear stiffnesses and to stiffness quantities regarding warping torsion is typically cumbersome. In this contribution a novel approach for their evaluation is proposed, which is based on a reference beam problem. Here, we restrict ourselves to double symmetric cross-sections, however, a generalization of the proposed method to the arbitrary case is obvious. Besides that, a novel approach to cover non-uniform warping torsion is included. The proposed method is efficient, since the discretization of the cross-section suffices, and accurate as can be shown in challenging bench mark problems.

16. Application of Geophysical Methods in Archaeological Survey of Early Medieval Fortifications

As powerful economic and cultural centers, fortified sites played an important role in early medieval society. In Central Europe, early medieval fortified site research has been an essential topic for several generations. However, gradual changes in the landscape are a threat to these cultural heritage monuments. The main task of this paper was to compare the previous results from archaeological excavations with new data acquired by geophysical methods. The presented study is based on the three methods widely used in archaeology: magnetometry, ground-penetrating radar, and electrical resistivity tomography. New surveys provide information about the internal structure and the state of preservation of the fortifications in a non-destructive way. Comparison of the results encourages the evaluation of archaeological excavation and helps determine the suitability and effectiveness of geophysical methods in specific natural conditions.

In this paper, results of numerical simulations and measurements are presented concerning the non-uniform torsion and bending of an angled members of hollow cross-section. In numerical simulation, our linear-elastic 3D Timoshenko warping beam finite element is used, which allows consideration of non-uniform torsion. The finite element is suitable for analysis of spatial structures consisting of beams with constant open and closed cross-sections. The effect of the secondary torsional moment and of the shear forces on the deformation is included in the local finite beam element stiffness matrix. The warping part of the first derivative of the twist angle due to bimoment is considered as an additional degree of freedom at the nodes of the finite elements. Standard beam, shell and solid finite elements are also used in the comparative stress and deformation simulations. Results of the numerical experiments are discussed, compared, and evaluated. Measurements are performed for confirmation of the calculated results.

In this contribution, results of elastostatic analysis of spatial composite beam structures are presented using our new beam finite element of double symmetric cross-section made of a Functionally Graded Material (FGM). Material properties of the real beams vary continuously in the longitudinal direction while variation with respect to the transversal and lateral directions is assumed to be symmetric in a continuous or discontinuous manner. Continuously longitudinal varying spatial Winkler elastic foundations (except the torsional foundation) and the effect of axial and shear forces are considered as well. Homogenization of spatially varying material properties to effective quantities with a longitudinal variation is done by the multilayer method (MLM). For the homogenized beam finite element the local stiffness matrix is established by means of the transfer matrix method. By the conventional finite element procedure, the global element stiffness matrix and the global system of equation for the beam structure are established for calculation of the global displacement vector. The secondary variables (internal forces and moments) are then calculated by means of the transfer relations on the real beams. Further, the mechanical stress in the real beams are calculated. Finally, the numerical experiments are carried out concerning the elastic-static analysis of the single FGM beams and beam structures in order to show the possibilities of our approach.

Pyruvate carboxylase (PC) is a mitochondrial, biotin-containing enzyme catalyzing the ATP-dependent synthesis of oxaloacetate from pyruvate and bicarbonate, with a critical anaplerotic role in sustaining the brain metabolism. Based on the studies performed on animal models, PC expression was assigned to be glia-specific. To study PC distribution among human neural cells, we probed the cultured human astrocytes and brain sections with antibodies against PC. Additionally, we tested the importance of PC for the viability of cultured human astrocytes by applying the PC inhibitor 3-chloropropane-1,2-diol (CPD). Our results establish the expression of PC in mitochondria of human astrocytes in culture and brain tissue and also into a subpopulation of the neurons in situ. CPD negatively affected the viability of astrocytes in culture, which could be partially reversed by supplementing media with malate, 2-oxoglutarate, citrate, or pyruvate. The provided data estimates PC expression in human astrocytes and neurons in human brain parenchyma. In this contribution modeling and simulation of surface acoustic waves (SAW) sensor using finite element method will be presented. SAW sensor is made from piezoelectric GaN layer and SiC substrate. Two different analysis types are investigated - modal and transient. Both analyses are only 2D. The goal of modal analysis, is to determine the eigenfrequency of SAW, which is used in following transient analysis. In transient analysis, wave propagation in SAW sensor is investigated. Both analyses were performed using FEM code ANSYS.

Content of the paper is oriented to calculation of elastic normal stress in the Functionally Graded Beams (FGM). Spatial variation of material properties is considered in the lateral, transversal and longitudinal direction of the straight beam. The displacements and internal forces are calculated using our new FGM finite beam element. Heterogeneous material properties are homogenized by extended mixture rules, laminate theory and reference volume element (RVE). Obtained results by our approach are evaluated and compared with the ones obtained by the 3D solid finite elements.

Abstract A measurement of the inclusive ep scattering cross section is presented in the region of low momentum transfers, 0.2 GeV2 ≤Q 2≤12 GeV2, and low Bjorken x, 5⋅10−6≤x≤0.02. The result is based on two data sets collected in dedicated runs by the H1 Collaboration at HERA at beam energies of 27.6 GeV and 920 GeV for positrons and protons, respectively. A combination with data previously published by H1 leads to a cross section measurement of a few percent accuracy. A kinematic reconstruction method exploiting radiative ep events extends the measurement to lower Q 2 and larger x. The data are compared with theoretical models which apply to the transition region from photoproduction to deep inelastic scattering.

This contribution deals with a two-node straight sandwich composite bar element with constant double symmetric rectangular cross-sectional area. This new bar element (based on the non-linear second-order theory) is intended to perform the non-incremental full geometric non-linear analysis. Stiffness matrix of this composite bar contains transfer constants, which accurately describe polynomial uniaxial variation of the material thermo-physical properties.In the numerical experiments the weak coupled thermo-structural geometric non-linear problem was solved. Obtained results were compared with several analyses made by ANSYS programme. Findings show good accuracy of this new finite element. The results obtained with this element do not depend on the element mesh density.

This contribution deals with a new truss element with varying stiffness intended to geometric and physically nonlinear analysis of composite structures. We present a two-node straight composite truss finite element derived by new nonincremental full geometric nonlinear approach. Stiffness matrix of this composite truss contains transfer constants, which accurately describe the polynomial longitudinal variation of cross-section area and material properties. These variations could be caused by nonhomogenous temperature field or by varying components volume fractions of the composite or/and functionally graded materials (FGM´s). Numerical examples were solved to verify the established relations. The accuracy of the new proposed finite truss element are compared and discused.
