Specific immunotherapy is the only treatment able to modify the natural outcome of the disease by restoring a normal immunity against allergens. The preparation of allergen-solid lipid nanoparticles as delivery vehicles for therapeutic proteins, P. judaica major allergen Par j 2, was investigated. The Par j 2 allergen was expressed in a large amount in Escherichia coli and purified to homogeneity. Its immunological properties were studied by western blotting and enzyme-linked immunosorbent assay inhibition. Solid lipid nanoparticles were obtained by water-in-oil-in-water multiple emulsion method and characterized in terms of mean size and surface charge. These systems (approximately 250 nm diameter and negative surface charge) incorporated recombinant Par j 2 with 40% or greater efficiency. Moreover, the endotoxin level and anaphylactic activity of the empty solid lipid nanoparticles and recombinant Par j 2-loaded solid lipid nanoparticles were evaluated by looking at the overexpression of CD203c marker on human basophils. These results demonstrate that recombinant Par j 2-nanoparticles could be proposed as safe compositions for the development of new therapeutic dosage forms to cure allergic reactions.Keywords: solid lipid nanoparticles, Parietaria judaica (Par j), drug delivery, recombinant allergens, specific immunotherapy, allergic rhinitis

Aims: To assess adherence to guideline recommendations among a large network of Italian cardiology sites in the management of acute and chronic heart failure (HF) and to evaluate if an ad‐hoc educational intervention can improve their performance on several pharmacological and non‐pharmacological indicators. Methods and results: BLITZ‐HF was a cross‐sectional study based on a web‐based recording system with pop‐up reminders on guideline recommendations used during two 3‐month enrolment periods carried out 3 months apart (Phase 1 and 3), interspersed by face‐to‐face macro‐regional benchmark analyses and educational meetings (Phase 2). Overall, 7218 patients with acute and chronic HF were enrolled at 106 cardiology sites. During the enrolment phases, 3920 and 3298 patients were included, respectively, 84% with chronic HF and 16% with acute HF in Phase 1, and 74% with chronic HF and 26% with acute HF in Phase 3. At baseline, adherence to guideline recommendations was already overall high for most indicators. Among acute HF patients, an improvement was obtained in three out of eight indicators, with a significant rise in echocardiographic evaluation. Among chronic HF patients with HF and preserved or mid‐range ejection fraction, performance increased in two out of three indicators: creatinine and echocardiographic evaluations. An overall performance improvement was observed in six out of nine indicators in ambulatory HF with reduced ejection fraction patients with a significant increase in angiotensin receptor–neprilysin inhibitor prescription rates. Conclusions: Within a context of an already elevated level of adherence to HF guideline recommendations, a structured multifaceted educational intervention could be useful to improve performance on specific indicators. Aims: Lower parental education has been linked to adverse youth mental health outcomes. However, the relationship between parental education and youth suicidal behaviours remains unclear. We explored the association between parental education and youth suicidal ideation and attempts, and examined whether sociocultural contexts moderate such associations. Methods: We conducted a systematic review and meta-analysis with a systematic literature search in PubMed, PsycINFO, Medline and Embase from 1900 to December 2020 for studies with participants aged 0–18, and provided quantitative data on the association between parental education and youth suicidal ideation and attempts (death included). Only articles published in English in peer-reviewed journals were considered. Two authors independently assessed eligibility of the articles. One author extracted data [e.g. number of cases and non-cases in each parental education level, effect sizes in forms of odds ratios (ORs) or beta coefficients]. We then calculated pooled ORs using a random-effects model and used moderator analysis to investigate heterogeneity. Results: We included a total of 59 articles (63 study samples, totalling 2 738 374 subjects) in the meta-analysis. Lower parental education was associated with youth suicidal attempts [OR = 1.12, 95% Confidence Interval (CI) = 1.04–1.21] but not with suicidal ideation (OR = 1.05, 95% CI = 0.98–1.12). Geographical region and country income level moderated the associations. Lower parental education was associated with an increased risk of youth suicidal attempts in Northern America (OR = 1.26, 95% CI = 1.10–1.45), but with a decreased risk in Eastern and South-Eastern Asia (OR = 0.72, 95% CI = 0.54–0.96). An association of lower parental education and increased risk of youth suicidal ideation was present in high- income countries (HICs) (OR = 1.14, 95% CI = 1.05–1.25), and absent in low- and middle-income countries (LMICs) (OR = 0.91, 95% CI = 0.77–1.08). Conclusions: The association between youth suicidal behaviours and parental education seems to differ across geographical and economical contexts, suggesting that cultural, psychosocial or biological factors may play a role in explaining this association. Although there was high heterogeneity in the studies reviewed, this evidence suggests that the role of familial sociodemographic characteristics in youth suicidality may not be universal. This highlights the need to consider cultural, as well as familial factors in the clinical assessment and management of youth's suicidal behaviours in our increasingly multicultural societies, as well as in developing prevention and intervention strategies for youth suicide. Two maternal mRNAs, coded in cell surface proteins and synthesized during oogenesis, are asymmetrically arranged in the unfertilized Paracentrotus lividus egg. The RNAs localize during fertilization and are detected in macro- and mesomeres and disappear during the late gastrula stage. The mRNA synthesized proteins exhibit similar localization as the mRNAs.

Ceramic ethnoarchaeology has been used to explore fully the chaîne opératoire and to understand all of the stages and factors involved in pottery production, such as raw material selection or paste recipes used by the potters. This work presents the results of the application of compositional analysis undertaken in the village of Pabillonis (western Sardinia, Italy), the main cooking ware production centre of the island. Pottery and local clays have been characterized using a combination of analytical techniques. By integrating the ethnographic information and the archaeometric approach, it was possible to reconstruct the operational sequence, exploring the relationship between the processing of raw materials and the functionality of the final products, and the intra-production compositional variability. The aim of this work is to examine whether it is possible to find chemical markers that allow a distinction to be made between the imported black glossed 'Campanian A' and the Sicilian imitation (end of fourth to first century bc) of these productions by carrying out quantitative chemical microanalysis of the slip using the SEM- EDS technique. The efficiency of the proposed analytical method has been tested on a set of ceramic samples corresponding to Sicilian black gloss imitations whose ceramic body has already been characterized petrographically by thin-section microscopy and chemically by XRF. The analytical data point to Na2O as a suitable chemical marker to distinguish between original 'Campanian A' imported from the Gulf of Naples area and Sicilian imitations of the same forms of Hellenistic pottery. In order to verify the above result, the enrichment factors ( EFs) between the raw clays, the corresponding ceramic body and black gloss slip were calculated. Some differences in the patterns of EFs between original 'Campanian A' and Sicilian imitations were recognized and explained. Therefore, the obtained results can help to accomplish a first distinction between imported and local material on a firm analytical basis, working on a statistically significant number of individuals. Summary Background Parietaria pollen is one of the major cause of pollinosis in the southern Europe. Specific immunotherapy is the only treatment able to modify the natural outcome of the disease restoring a normal immunity against allergens. Methods We designed a recombinant molecule ( Pj EDloop1) comprised of genetic-engineered variants of the major allergens of the Parietaria pollen ( Par j 2/ Par j 1). Purity and chemical-physical properties of the derivative were analysed by RP- HPLC chromatography and Photon Correlation Spectroscopy. Immunological activity was evaluated by means of Western blotting, ELISA inhibition and PBMC proliferation assay in 10 Parietaria allergic patients. Basophil activation was studied in six subjects. The immunogenicity of the hybrid was studied looking at the immune responses induced in a mouse model of sensitization. Results The Pj EDloop1 hybrid was produced as a purified recombinant protein with high stability in solution. Western blot, ELISA inhibition and basophil activation test showed that the Pj EDloop1 displays a remarkable reduced IgE binding and anaphylactic activity. CD3 reactivity was conserved in all patients. Mice immunization with the r Pj EDloop1 induced antibodies and T cell responses comparable to that obtained by the wild type allergens. Such antibodies shared the specificities to r Par j 1 and r Par j 2 with human IgE antibodies. Conclusion Our results demonstrated that a mutant hybrid expressing genetically engineered forms of the major P. judaica allergens displayed reduced allergenicity and retained T cell reactivity for the induction of protective antibodies in vaccination approaches for the treatment of Parietaria pollinosis. Deep blue glasses coloured by octahedral Fe2+ cations are often reported as textbook examples of blue pigmentation. However, despite the possibility of laboratory synthesis under reducing conditions, to date there are no well-reported occurrences of their production and use in the past. A thorough historical, ethnographic, mineralogical, and chemico-physical investigation of the ‘smaltini di calcara ’ from several baroque churches in Palermo, Sicily, has revealed that the blue enamels widely used for altar decorations in the 17th and 18th centuries are actually a unique case of ancient blue glasses pigmented by divalent iron cations in distorted octahedral coordination. This mixed-alkali glass was accidentally produced under severely reducing conditions in the local kilns during production of lime. [ABSTRACT FROM AUTHOR]

Pantellerian ware is a Late Roman cooking ware whose production centre was established on the island of Pantelleria by the pioneering research of Fulford and Peacock almost 20 years ago ( Peacock 1982 ; Fulford and Peacock 1984 ). Archaeological and archaeometric studies carried out by the authors of the present contribution during the past four years have aimed to fully characterize this ceramic class. Recurrent ceramic forms, their distribution over time and space, their petrographic characteristics and their chemical identity, as well as possible raw materials and their technological properties, were considered. The present paper is a comprehensive review of this archaeometric work and aims to establish a ‘reference group’. Using a representative number of samples of Pantellerian ware that were recently discovered in the island through archaeological field surveys or surface and submarine excavations, it was possible to characterize in detail the compositional variability of this ware in terms of chemistry and petrography. Furthermore, the physical properties of this ceramic type have been defined in order to better understand its performance characteristics, mainly in response to induced thermal stress. In the meantime, the experimental mixing and tempering of locally sampled raw materials have shed light on the ancient manufacturing process and have led to an approximation of the original paste. Samples of Roman thin-walled ware from Segesta (northwestern Sicily), dating back to the early Imperial period, were studied by optical microscopy (OM) and Neutron Activation Analysis (NAA). Up to now, this class of Roman fine tableware has only occasionally been evaluated archaeometrically. Nevertheless, numerous production centres are believed to have been simultaneously active in the western Mediterranean area. Petrographic and chemical data seem to be in agreement with the archaeological hypothesis of local manufacture in Segesta for most of the analysed samples, through a comparison with kiln wasters and local raw materials. The effectiveness of thin-section petrography for determining the provenance of such a tiny tempered class of pottery and the integrated use of two different grouping procedures (petrography and chemistry) were also tested. Objective: To compare results of treatment of adenocardinoma of the prostate using Standard (2D) vs Conformal (3D) treatment planning. Methods: The records of all patients with adenocarcinoma of the prostate treated curatively with radiation therapy alone from July 1991 to June 1994 were reviewed. Acute and late complications were scored by the RTOG criteria. Biochemical failure was defined as a rising PSA of at least 10% on two measurements separated ≥1 month or either a PSA nadir >4 ng/ml or >1 ng/ml. Disease free survival (DFS) was defined as no evidence of local, distant, or biochemical failure. 2D planning included standard simulation with target volume drawn from the treatment planning or diagnostic CT. 3D planning included a CT in the treatment position with computer simulation using beam’s-eye-view for field design. Results: Two-hundred and seventeen 2D and 45 3D patients had similar median age and pre-treatment PSA, T-stage, and dose to the prostate. The median follow-up periods for the 2D and 3D groups were 32.0 and 21.5 months, respectively. The two-year actuarial survival, local or biochemical control, and DFS were not different. The 3D group had a significantly higher incidence of acute bladder side effects of all grades and acute grade 1/2 rectal complications. There were no differences in the incidence of late bladder or rectal complications. Conclusions: Careful 2D planning for the treatment of localized adenocarcinoma of the prostate is an acceptable means of treatment. Within the dose range of 64–70 Gy, this preliminary analysis demonstrated no reduction in complications nor improvement in local or biochemical control, or DFS was seen with the the use of 3D treatment planning. This article presents a study on toxicity of recombinant β-amyloid prefibrillar oligomers on the morphogenic development of sea urchin, Paracentrotus lividus. By using the RNA from human neuroblastoma LAN5 cells and an Escherichia coli expression vector, recombinant peptide can be produced in large quantities. Conclusions and significance of the study are presented.

Introduction The conventional tACS approach involves defining the frequency and phase of stimulation ad hoc and testing them on a cohort of subjects. However, this approach exhibits two limitations: (1) the brain networks targeted by the stimulation cannot be verified without simultaneous functional magnetic resonance imaging (fMRI); (2) those stimulation parameters may vary across subjects due to difference in anatomy or heterogeneity in disease. Yet, there is a combinatorial explosion in the biologically plausible range of stimulation frequencies and phases, resulting in thousands of possibilities. Identifying the optimal stimulation protocol for a given individual is like ‘finding a needle in a haystack’. Therefore, using conventional methodology makes tailoring tACS to an individual highly unfeasible. Methods To address this fundamental challenge, we combine tACS with an innovative framework that utilizes real-time fMRI and machine-learning: The Automatic Neuroscientist ( Lorenz, 2016 ). The framework starts with a target brain state and finds a set of tACS parameters (frequency and phase of stimulation) that maximally activates it. This is done in a closed-loop fashion ( Fig. 1), i.e. real-time fMRI provides instantaneous information on how effective certain stimulation parameters are. Based on this, the Bayesian optimization approach proposes the combination of tACS parameters that will be applied in the next iteration. This cycle continues until the Automatic Neuroscientist identifies the sweet spot within the exhaustive parameter space (Fig. 2 ) that maximally activates the target brain state. Results We could demonstrate the technical feasibility of novel neurotechnology that combines real-time fMRI, machine-learning and tACS in a fully automated manner. Moreover, we showed that our closed-loop framework is far more efficient that the standard approach. Conclusion We envision that our framework will advance personalized treatment by means of non-invasive brain stimulation. This will be of particular importance for neurological and psychiatric deficits that are diffuse and widely heterogeneous in their origin. [ABSTRACT FROM AUTHOR]
