Abstract

Sedimentary ancient DNA (sedaDNA) has rarely been used to obtain population‐level data due to either a lack of taxonomic resolution for the molecular method used, limitations in the reference material or inefficient methods. Here, we present the potential of multiplexing different PCR primers to retrieve population‐level genetic data from sedaDNA samples. Vaccinium uliginosum (Ericaceae) is a widespread species with a circumpolar distribution and three lineages in present‐day populations. We searched 18 plastid genomes for intraspecific variable regions and developed 61 primer sets to target these. Initial multiplex PCR testing resulted in a final set of 38 primer sets. These primer sets were used to analyse 20 lake sedaDNA samples (11,200 cal. yr BP to present) from five different localities in northern Norway, the Alps and the Polar Urals. All known V. uliginosum lineages in these regions and all primer sets could be recovered from the sedaDNA data. For each sample on average 28.1 primer sets, representing 34.15 sequence variants, were recovered. All sediment samples were dominated by a single lineage, except three Alpine samples which had co‐occurrence of two different lineages. Furthermore, lineage turnover was observed in the Alps and northern Norway, suggesting that present‐day phylogeographical studies may overlook past genetic patterns. Multiplexing primer is a promising tool for generating population‐level genetic information from sedaDNA. The relatively simple method, combined with high sensitivity, provides a scalable method which will allow researchers to track populations through time and space using environmental DNA. Abstract

The perioperative setting is a high-risk environment which is particularly susceptible to communication deficits and errors. The situation, background, assessment, recommendation (SBAR) approach provides an intuitive guideline for team communication, which is associated with an improved quality of the handover. The German Society for Anaesthesiology and Intensive Care Medicine (DGAI) has updated its recommendations in March 2022 and continues to endorse the use of the SBAR template. The impact of tools used for structured communication during patient handover are often studied in the context of a larger bundle of measures. The SBAR template is one option for establishing structured communication in clinical practice. Successful implementation is supported by clearly defined standard workflows to promote consistent use. This standardization identifies common communication barriers and assists in resolving them in a high-risk environment. A common understanding of the inherent values, and a shared interest in learning, applying, and training these techniques are paramount in establishing a culture of patient safety. Abstract

Within the product development the need for generating design variants is given in many situations, for example to optimize an existing initial design with respect to new or modified requirements. For an efficient process it is essential that these variations call be done very easily within a small timeframe. In the virtual product development the physical characteristic of a component is determined by numerical simulation. Commercial software products exist for nearly each physical phenomenon. Often these methods are covered under CAE. A fundamental issue for building and analyzing variants easily and fast is a seamless interaction between the CAD and CAE software tools. This paper presents a powerful CAD/CAE sequence to the engineer's community, where in contrast to other approaches results of the CAE analysis directly interact with CAD data. This strategy is supported by describing the product's geometry by parameters. The CAD/CAE sequence is integrated in an optimization loop. The presented application example is an automotive part. We assessed the cardiopulmonary effects of low doses of NO in early allograft dysfunction following lung transplantion. Design: Prospective clinical dose- response study. Setting: Anesthesiological intensive care unit of a university hospital. Patients and participants: 8 patients following a single or double lung transplantation who had a mean pulmonary arterial pressure (PAP) in excess of 4.7 kPa (35 mmHg) or an arterial oxygen tension/fractional inspired oxygen ratio (PaO.sub.2/FIO.sub.2) of less than 13.3 kPa (100 mmHg). Interventions: Gaseous NO was inhaled in increasing concentrations (1, 4 and 8 parts per million, each for 15 min) via a Siemens Servo 300 ventilator. Measurements and results: Cardiorespiratory parameters were assessed at baseline, after each concentration of NO, and 15 min after withdrawal of the agent [statistics: median (25th/75th percentiles: Q1/Q3), rANOVA, Dunnett's test, p &lt 0.05]. Inhaled NO resulted in a significant, reversible, dose-dependent, selective reduction in PAP from 5.5(5.2/6.0) kPa at control to 5.1(4.7/5.6) kPa at 1 ppm, 4.9(4.3/5.3) kPa at 4 ppm, and to 4.7(4.1/5.1) kPa at 8 ppm. PaO.sub.2 increased from 12.7(10.4/17.1) to 19.2(12.4/26.0) kPa at 1 ppm NO, to 23.9(4.67/26.7) kPa at 4 ppm NO and to 24.5(11.9/28.7) kPa at 8 ppm NO. All patients responded to NO inhalation (either with PAP or PaO.sub.2), all were subject to long-term inhalation (1--19 days). All were successfully weaned from NO and were discharged from the intensive care unit. Conclusion: The present study demonstrates that low-dose inhaled NO may be an effective drug for symptomatic treatment of hypoxemia and/or pulmonary hypertension due to allograft dysfunction subsequent to lung transplantation. Abstract

We use sensitive dynamic cantilever magnetometry to measure the magnetic hysteresis of individual magnetic Janus particles. These particles consist of hemispherical caps of magnetic material deposited on micrometer-scale silica spheres. The measurements, combined with corresponding micromagnetic simulations, reveal the magnetic configurations present in these individual curved magnets. In remanence, ferromagnetic Janus particles are found to host a global vortex state with vanishing magnetic moment. In contrast, a remanent onion state with significant moment is recovered by imposing an exchange bias to the system via an additional antiferromagnetic layer in the cap. A robust remanent magnetic moment is crucial for most applications of magnetic Janus particles, in which an external magnetic field actuates their motion. [ABSTRACT FROM AUTHOR]

Abstract

The biomimetic, deterministic aperiodic golden angle Vogel spiral lacks both translational and rotational symmetry and exhibits circularly symmetric scattering rings in Fourier space. We present a versatile and low cost method for maskless fabrication of a Vogel spiral relief grating by illumination of a photoresist with laser light complexly structured using a spatial light modulator and subsequent transfer of its negative into a polymer layer via replica molding. We investigate its diffractive properties and present as one application its implementation as a surface structure, leading to an efficiency enhancement of 18% in an organic solar cell.
