Quality and safety are one of the main concerns of the European Union in food preservation. Using chemical additives extends the shelf-life of fresh foods but raises consumer’s concerns about the potential long-term carcinogenic effects. Using natural substances derived from agro-industrial by-products, which have significant antimicrobial and antioxidant activities, could extend the shelf-life of fresh foods such as meat. Furthermore, they can provide nutritional improvements without modifying organoleptic properties. This study analyzes the antimicrobial activity of pomegranate peel extract (PPE) and the antioxidant activity of olive leaf extract (OLE), added at concentrations of 10 mg g−1 and 0.25 mg g−1, respectively, to minced poultry and rabbit meat. PPE exhibited in vitro antimicrobial activity against foodborne pathogens starting at 10 mg/well. PPE and OLE determined a reduction in colony count over a storage period of 6 days at 4 °C. Additionally, the combination of PPE and OLE showed antioxidant effects, preserving lipid oxidation and maintaining pH levels. The obtained results demonstrate that PPE and OLE can be recommended as food additives to preserve the quality and extend the shelf-life of meat products.

In this study, we present data on the effects of condensed tannins (CTs) and hydrolysable tannins (HTs), polyphenols extracted from plants, at different concentrations on zebrafish development to identify the range of concentrations with toxic effects. Zebrafish embryos were exposed to CTs and HTs at two different concentration ranges (5.0–20.0 μgL−1 and 5.0–20.0 mgL−1) for 72 h. The toxicity parameters were observed up to 72 h of treatment. The uptake of CTs and HTs by the zebrafish larvae was assessed via HPLC analysis. A qRT-PCR analysis was performed to evaluate the expressions of genes cd63, zhe1, and klf4, involved in the hatching process of zebrafish. CTs and HTs at 5.0, 10.0, and 20.0 μgL−1 were not toxic. On the contrary, at 5.0, 10.0, and 20.0 mgL−1, HTs induced a delay in hatching starting from 48 h of treatment, while CTs showed a delay in hatching mainly at 48 h. The analysis of gene expression showed a downregulation in the group exposed to HTs, confirming the hatching data. We believe that this study is important for defining the optimal doses of CTs and HTs to be employed in different application fields such as the chemical industry, the animal feed industry, and medical science.

Keratin extracted (KE) from chicken feathers was used for the production of composite films comprising poly(ε-caprolactone) (PCL) and keratin (PCL/KE films). The process involved the extraction of keratin from chicken feathers using a 0.1 M NaOH solution, followed by characterization via sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). The PCL was synthesized through the ring-opening polymerization (ROP) of ε-caprolactone (ԑ-CL) with Sn(Oct)2 as a catalyst. Films were prepared via solvent casting, including pure PCL films and those enriched with different weight percentages of KE (10%, 15%, 25%, and 30%). The films were characterized by differential scanning calorimetry (DSC), thermogravimetric analysis (TG), and scanning electron microscopy (SEM). SEM analysis revealed a more uniform incorporation of KE within the PCL matrix in the case of the 15% keratin-enriched film (PCL/KE15) as compared to other keratin percentages. The thermal analysis showed a positive influence of keratin on the thermal stability of the films. Keratinocytes viability and proliferation tests on the PCL/KE15 film demonstrated compatibility with cells. Collectively, these results hold relevance for potential biomedical applications of PCL/KE films.

Bioaerosols and pathogens in indoor workplaces and residential environments are the primary culprits of several infections. Techniques for sanitizing air and surfaces typically involve the use of UV rays or chemical sanitizers, which may release chemical residues harmful to human health. Essential oils, natural substances derived from plants, which exhibit broad antimicrobial properties, could be a viable alternative for air and surface sanitation. The objective of this study has been to investigate the efficacy of thyme essential oil (TEO) in environmental sanitation processes. In Vitro assays through agar well diffusion, disk volatilization and tube dilution methods revealed significant antimicrobial activity of TEO 100% against foodborne and environmental isolates, with both bacteriostatic/fungistatic and bactericidal/fungicidal effects. Therefore, aqueous solutions of TEO 2.5% and 5% were formulated for air sanitation through nebulization and surface disinfection via direct contact. Bioaerosol samples and surface swabs were analyzed before and after sanitation, demonstrating the efficacy of aqueous solutions of TEO in reducing mesophilic and psychrophilic bacteria and environmental fungi levels in both air and on surfaces. The obtained results prove the antimicrobial potential of aqueous solutions of TEO in improving indoor air quality and surface cleanliness, suggesting thyme essential oil as an effective and safe natural sanitizer with minimal environmental impact compared to dangerous chemical disinfectants.

Chitosan (CH) shows great potential as an immunostimulatory feed additive in aquaculture. This study evaluates the effects of varying dietary CH levels on the growth, immunity, intestinal morphology, and antioxidant status of Nile tilapia (Oreochromis niloticus) reared in a biofloc system. Tilapia fingerlings (mean weight 13.54 ± 0.05 g) were fed diets supplemented with 0 (CH0), 5 (CH5), 10 (CH10), 20 (CH20), and 40 (CH40) mL·kg−1 of CH for 8 weeks. Parameters were assessed after 4 and 8 weeks. Their final weight was not affected by CH supplementation, but CH at 10 mL·kg−1 significantly improved weight gain (WG) and specific growth rate (SGR) compared to the control (p < 0.05) at 8 weeks. Skin mucus lysozyme and peroxidase activities were lower in the chitosan-treated groups at weeks 4 and 8. Intestinal villi length and width were enhanced by 10 and 20 mL·kg−1 CH compared to the control. However, 40 mL·kg−1 CH caused detrimental impacts on the villi and muscular layer. CH supplementation, especially 5–10 mL·kg−1, increased liver and intestinal expressions of interleukin 1 (IL-1), interleukin 8 (IL-8), LPS-binding protein (LBP), glutathione reductase (GSR), glutathione peroxidase (GPX), and glutathione S-transferase (GST-α) compared to the control group. Overall, dietary CH at 10 mL·kg−1 can effectively promote growth, intestinal morphology, innate immunity, and antioxidant capacity in Nile tilapia fingerlings reared in biofloc systems.

Grape pomace is the main by-product of vine-winery chains. It requires adequate treatment and disposal but is also an economically underused source of bioactive plant secondary metabolites. This study aimed to investigate the antibacterial effects of polyphenolic extracts from Aglianico (Vitis vinifera L.) grape pomace. In particular, hydroethanolic extracts obtained via an ultrasonic-assisted extraction technique were selected for antimicrobial tests. The extracts were screened for their antibacterial effects against foodborne pathogens that were both Gram-positive, in the case of Staphylococcus aureus and Bacillus cereus, and Gram-negative, in the case of Escherichia coli and Salmonella enterica subsp. enterica serovar Typhimurium, showing variable bacteriostatic and bactericidal effects. In addition, our results demonstrated that the tested grape pomace extracts can reduce the inhibitory concentration of standard antibiotics. Interestingly, selected extracts inhibited biofilm development by S. aureus and B. cereus. Overall, these new insights into the antibacterial properties of grape pomace extracts may represent a relevant step in the design of novel therapeutic tools to tackle foodborne diseases, and in the management of resistant biofilm-related infections.

The aim of the present study was to evaluate the wild freshwater fish health status using a vast array of biomarkers as predictive factors of pollutant exposure. The European eel (Anguilla anguilla) and brown trout (Salmo trutta fario), resident in rivers with different degrees of pollution in the South of Italy (Picentino River with good environmental quality and Tusciano River with low environmental quality), were examined using biometric parameters, histopathological and immunohistochemical biomarkers to evaluate the health status and a possible correlation with the water quality. Several alterations identified in the liver positively correlated with water and soil pollutants: hemorrhage (p ≤ 0.05), cytoplasmic vacuolization (p ≤ 0.01), hemosiderosis (p ≤ 0.05), irregular arrangement of hepatocytes (p ≤ 0.01), lipid accumulation (p ≤ 0.05), necrosis (p ≤ 0.01), cellular hyperplasia (p ≤ 0.05), leukocyte infiltration (p ≤ 0.01) and melanomacrophages centers (MMC) (p ≤ 0.01). In the spleen, only hemosiderosis correlated with water and soil pollutants (p ≤ 0.05). The inflammatory biomarker tumor necrosis factor α (TNFα) and ciclooxigenase 2 (COX2) responded to the environmental pollution, as well as the oxidative stress biomarkers superoxide dismutase (SOD2) and 8-Hydroxy-2′-deoxyguanosine (8-OHdG). Erythrocytic nuclear abnormalities and erythrocytic cellular abnormalities were found to be significantly higher in the blood of both the European eel (p < 0.0001) and brown trout (p < 0.001) in the Tusciano River compared with the Picentino River. Taken together, these results outline the need to increase the number of suitable biomarkers to assess fish health and reinforce the importance of employing additional biomarkers in biomonitoring programs that can be applied to evaluate water quality and in environmental assessment around the world.

The present study aimed to investigate the effects of the herbaceous plant, Dracocephalum kotschyi, on the immune response, antioxidant enzymes activity, growth performance, and related genes expression in zebrafish (Danio rerio). An in vivo trial was carried out by feeding zebrafish (initial weight of 0.47 ± 0.04 g) with different levels of powdered D. kotschyi: 0% (C-0%, control), 1% (Dk-1%), 2% (Dk-2%) and 3% (Dk-3%) for 10 weeks. An 8-week feeding trial was carried out to evaluate the effects of grapevine (Vitis vinifera) leaf extract (GLE) on the growth, oxidative enzymatic activities, immunity, and expression of antioxidant genes in zebrafish (Danio rerio). Three hundred and sixty zebrafish were supplied and fed with different levels of GLE: 0, 0.5, 1, and 2 g kg−1. The dietary administration of 1 g kg−1 of GLE significantly increased growth parameters in fish. Fish fed diets with GLE showed increased total protein. The total Ig and lysozyme activity significantly changed in the whole-body serum, but not in skin mucus. GLE significantly increased Catalase (CAT), Superoxide Dismutase (SOD), and Glutathione Peroxidase (GPx) activities compared to the control diet. GLE treatments caused a significant decrease in the malondialdehyde (MDA) content. In the skin mucus, only CAT and SOD activities significantly increased. The highest expression of Toll-like receptor-1 (TLR-1) and Tumor Necrosis Factor-α (TNFα) genes was achieved in fish fed 2 g kg−1 of GLE. CAT and SOD gene expressions were significantly higher in fish fed 1 and 2 g kg−1 of GLE. GPx gene expression was significantly higher in fish fed 1 g kg−1 of GLE. In conclusion, the results of the present study revealed that GLE affects growth performance and regulates antioxidant and immune gene expression. The determination of the optimum dosage merits further research.

Here, the olive leaf extract (OLE) rich in polyphenols was employed as a prebiotic agent, together with Lactobacillus reuteri and Bacillus clausii, to develop synbiotics. The prebiotic effect of olive leaf extract on the probiotic strains was tested at concentrations of 0, 50, 100, 400, and 1000 μg mL−1, and also 20 and 40 mg mL−1. Olive leaf extract at 40 mg mL−1 showed the best prebiotic activity on L. reuteri and B. clausii. A basal diet and two experimental synbiotic-containing diets were prepared. The synbiotic diets were manufactured by adding to the basal diet 5 × 106 CFU g−1 L. reuteri + 5 × 106 CFU g−1 B. clausii + 0.25 mg g−1 OLE and 1 × 107 CFU g−1 L. reuteri + 1 × 107 CFU g−1 B. clausii + 0.25 mg g−1 OLE. The diets were administered to the freshwater crayfish Astacus astacus (1.35 ± 0.04 g) in an 84-day feeding trial. The diet containing 5 × 106 CFU g−1 L. reuteri + 5 × 106 CFU g−1 B. clausii + 0.25 mg g−1 OLE significantly improved (p < 0.05) final weight, specific growth rate, body condition, and survival rate. A significant growth of Enterobacteriaceae, which include strains with proven beneficial activities for intestinal health and general animal welfare, significantly increased in crayfish fed with synbiotics. The obtained results could be suitable for functional feed development in crayfish farming.

This research examined the role of nano curcumin (NC) on growth performances, body composition, and blood parameters of red tilapia (Oreochromis sp.) challenged with Aspergillus flavus. Fish (5.0 g ± 0.30) were randomly distributed in four equal groups (20 fish per pond in triplicates) and fed various concentrations of NC fortified with 0 (Control), 40 mg/kg (NC1), 50 mg/kg (NC2), and 60 mg/kg diet (NC3) of nano curcumin. After eight weeks of the feeding trial, the fish were challenged with A. flavus for 15 days, and the cumulative mortality was recorded. Fish fed with different concentrations of NC improved significantly (p < 0.05) the growth performances, feed utilization, and survival rate. There was no significant (p > 0.05) difference between NC2 and NC3 treatments. However, NC3 exhibited higher performances. Fish feed supplemented with NC decreased the mortality rate when challenged with A. flavus. Hence, dietary supplementation of NC enhanced the growth and health status of Oreochromis sp. and protected it from A. flavus infection. This study suggests the optimum inclusion level of NC is a 50–60 mg/kg diet.

Nowadays, the wide spread of foodborne illness and the growing concerns about the use of synthetic food additives have shifted the focus of researchers towards essential oils (EOs) as possible antimicrobials and preservatives of natural origin. Thanks to their antimicrobial properties against pathogenic and food spoilage microorganisms, EOs have shown good potential for use as alternative food additives, also to counteract biofilm-forming bacterial strains, the spread of which is considered to be among the main causes of the increase in foodborne illness outbreaks. In this context, the aim of this study has been to define the antibacterial and antibiofilm profile of thyme (Thymus vulgaris L.) essential oil (TEO) against widespread foodborne pathogens, Salmonella enterica subsp. enterica serovar Typhimurium and Bacillus cereus. TEO chemical composition was analyzed through gas chromatography-mass spectrometry (GC-MS). Preliminary in vitro antibacterial tests allowed to qualitatively verify TEO efficacy against the tested foodborne pathogens. The subsequent determination of minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) values allowed to quantitatively define the bacteriostatic and bactericidal effects of TEO. To evaluate the ability of essential oils to inhibit biofilm formation, a microplate assay was performed for the bacterial biofilm biomass measurement. Results suggest that TEO, rich in bioactive compounds, is able to inhibit the growth of tested foodborne bacteria. In addition, the highlighted in vitro anti-biofilm properties of TEO suggest the use of this natural agent as a promising food preservative to counteract biofilm-related infections in the food industry.

Diazion is a common pesticide which cultured fish can be exposed. There is limited information available on the possible effects of diazinon on reproductive parameters of fish. Therefore, in this study, we focused on the effects of sub-lethal doses (0, 0.8, 1.6, and 3.2 mg/L) of diazinon, an organophosphate insecticide on sex hormones, ovarian morphology, reproductive, immunity, and antioxidant related genes, in female zebrafish (Danio rerio) after 30 days of treatment. The expression of reproductive-related genes (CYP19a, Vtg, and ERα) significantly decreased, while the expression of the immune (TNFα and Il-1β) and antioxidant (SOD and CAT) related genes increased considerably. Among sex steroids, only progesterone significantly decreased after the treatment with Diazinon sub-lethal doses, while 17-β estradiol and testosterone did not change. The ovarian morphology showed a decrease in oocyte development in diazinon-treated groups. Understanding the function and mechanism of action of diazinon on the female reproductive, immune, and oxidant status may provide new strategies to counteract its adverse effects.

This study aimed to investigate the effects of Amla (Phyllanthus emblica) fruit extract (AFE) on the growth, skin mucosal and serum immunity, and disease resistance of Nile tilapia (Oreochromis niloticus) against Streptococcus agalactiae. Three hundred fish (10.48 ± 0.56 g fish-1) were distributed in 15 tanks with 20 fish per tank and fed basal diet supplemented with 0, 5, 10, 20, and 40 mg kg-1 AFE for eight weeks. The completed randomized design with three replications was applied. Growth performance, peroxidase, alternative complement, phagocytosis, and respiratory burst activities were determined at weeks 4 and 8. The challenge study was conducted at the end of the experiment. Based on the results, Nile tilapia fed the AFE diets significantly enhanced skin mucus lysozyme and peroxidase activities (P ≤ 0.05) post-feeding. The highest levels were observed in the group that received 20 mg kg-1 of AFE (P ≤ 0.05), whereas the control group presented the lowest levels of lysosome and peroxidase activities. Similarly, incorporation of 20 mg kg-1 of AFE brought about the maximum levels of (P ≤ 0.05) serum lysozyme, peroxidase, alternative complement, phagocytosis, and respiratory burst activities in contrast to the other treatments; whereas fish fed the control diet generated the lowest immune parameter values (P ≤ 0.05). The aim of this study was to compare the effects of adding taurine to diet as a mixture with feed and also as spray on growth and feeding performance, body composition and physiological responses of Asian sea bass, Lates calcarifer (13.62 ± 0.2 g). This study was carried out in a completely randomized design as three treatments with triplicates. Fish were fed with feed containing 0 (control group), 10 g/kg taurine in feed as mixture (treatment M) and 10 g/kg as spray on feed (treatment S) for 6 weeks. At the end of the experiment, final weight, weight gain, specific growth rate and feed conversion ratio were significantly higher in treatment S compared to the control and treatment M (p < 0.05). Carcass fat and moisture contents significantly varied in treatment M with the control and with treatment S (p < 0.05). However, carcass protein value was not influenced by supplemented tuarine (p>0.05). The obtained results indicated that dietary taurine did not enhance Asian sea bass blood biochemical factors and non-specific immune response (p>0.05). Overall, it can be concluded that feeding Asian sea bass with the diet supplemented by 10 g taurine as spray for a period of 6 weeks enhances the growth and feeding performance.

Foodborne diseases continue to represent an important public health issue. The control of food spoilage and pathogenic microorganisms is achieved mainly by synthetic chemicals, unfortunately associated to several undesirable aspects. The growing requirement for new and safe alternative strategies has resulted in the research of agents from natural sources with antimicrobial properties, such as essential oils (EOs). This study’s purpose was to define the antibacterial profile of thyme (Thymus vulgaris) and cloves (Syzygium aromaticum) essential oils against both Gram-positive and Gram-negative important foodborne pathogenic bacteria. Gas chromatography mass spectrometry analysis was performed for EOs’ chemical composition. Qualitative in vitro antimicrobial assays (i.e., agar well diffusion method and disk-volatilization method) allowed for verification of the efficacy of EOs, used individually and in binary combination and both in liquid and vapor phase, against Staphylococcus aureus and Escherichia coli food isolates. Minimal inhibitory concentrations and minimal bactericidal concentration values have been used to quantitatively measure the antibacterial activity of EOs, while the fractional inhibitory concentration index has been considered as a predictor of in vitro antibacterial synergistic effects. The microbiological tests suggest that thyme and cloves EOs, rich in bioactive compounds, are able to inhibit the growth of tested foodborne bacteria, especially in vapor phase, also with synergistic effects. Results provide evidence to consider the tested essential oils as promising sources for development of new, broad-spectrum, green food preservatives.

The current study evaluated the effects of hydrolyzable and condensed tannins from chestnut and quebracho wood, respectively (TSP, Silvafeed®), on zebrafish with intestinal inflammation induced by a plant-based diet (basal diet). Four experimental diets were prepared as follows: the basal diet + 0 TSP, the basal diet + TSP at 0.9 g/kg of feed, the basal diet + TSP at 1.7 g/kg of feed, and the basal diet + TSP at 3.4 g/kg of feed. Eighty-four zebrafish (Danio rerio) were fed for 12 days with the experimental diets. In zebrafish fed the basal diet, intestine integrity appeared to be altered, with damaged intestinal villi, high immunoexpression of tumor necrosis factor-α (TNFα) and cyclooxygenase 2 (COX2), and high expression of the cox2, interleukin 1 (il-1b), interleukin 8 (cxcl8-l1), and tnfα genes. The tannin treatment partially restored intestinal morphology and downregulated the expression of cytokines. The best activity was detected with 1.7 and 3.4 g/kg of feed. In the guts of all groups, Proteobacteria, Fusobacteria, Firmicutes, and Bacteroidetes were the most represented phyla. The most represented genera were Plesiomonas and Sphingomonas, belonging to the Proteobacteria phylum; Cetobacterium, belonging to the Fusobacteria phylum; and Lactobacillus, belonging to the Firmicutes phylum. No significant differences were detected among groups, except for a slight decrease in the Fusobacteria phylum and slight increases in the Shewanella and Bacteroides genera with TSP. In conclusion, these results suggest that tannins can improve the zebrafish intestinal inflammation caused by a terrestrial-plant-based diet in a dose-dependent manner.

The growing demand for high-quality food has induced a rapid expansion of the aquaculture sector. On the other hand, this sector has to overcome numerous challenges and problems triggered by the adoption of intensive farming systems, such as stress and high susceptibility to diseases. The improper use of chemicals and antibiotics has led to the development of antibiotic resistance in fish, with consequent health risks for consumers. Natural additives are increasingly used in aquaculture and, among these, medicinal plants are constantly under investigation as safe and environmentally friendly alternatives to chemicals. Great attention has been paid to Lamiaceae plants as feed additives capable of enhancing the growth performance, immune system, and antioxidant status of farmed fish. The aim of this review is to provide an updated picture of the employment of the Lamiaceae species (oregano, rosemary, sage, thyme, and mint) to enhance farmed fish health. The benefits of oregano, rosemary, sage, thyme, and mint feed supplementation on growth performance, immune system, antioxidant status, hemato-biochemical parameters, and resistance to stress, parasites, and bacteria have been described, highlighting weaknesses and drawbacks and proposing possible implementations.

The present study was designed to investigate the effect of Spirulina platensis (SP) supplemented diets on the growth performance, digestive enzymes, hepatic antioxidants and innate immunity biomarkers in juvenile grass carp (Ctenopharyngodon idella). Two hundred and forty grass carp juveniles (average weight 4.81 ± 0.13 g) were divided into four treatment groups in triplicates (20 fish/replicate) and fed with diets containing 0, 1, 5, and 10% Spirulina for 90 days. A significant increase in growth (p < 0.05) was observed in fish fed with diets having 1 and 5% Spirulina. Intestinal protease and lipase activities increased significantly (p < 0.01) in fish fed with a diet having 5% Spirulina while intestinal amylase activity increased significantly (p < 0.01) in fish fed with diets having 1, 5, and 10% Spirulina. Hepatic lipid peroxidation decreased significantly (p < 0.05) in fish fed with a 1 and 5% Spirulina supplemented diet. The activity of catalase, glutathione-S-transferase, and glutathione levels increased significantly (p < 0.05) in the livers of fish fed with 1% Spirulina supplemented diets while no significant difference (p > 0.05) was observed for hepatic superoxide dismutase levels when compared to the control. Significant increases in the skin mucus protease (p < 0.05), antiprotease (p < 0.01), lysozyme (p < 0.001), and peroxidase (p < 0.05) activities were observed in fish fed with 5% Spirulina-supplemented diets. Gene expression analysis of head kidney showed that fish fed with a 1% Spirulina diet had significantly (p < 0.01) higher expression of tnf-α, il-8, and inf-γ. In conclusion, the present study suggested that the inclusion of 5% Spirulina platensis in the diet of grass carp has positive effects on growth, digestive enzymes, antioxidants, and innate immunity.

An 8-week feeding experiment was conducted to investigate the inclusion potential of five levels of Calotropis persica seed powder (CSP) (0, 10, 20, 30, 40 and 50 g/kg of the basal diet) in rainbow trout (Oncorhynchus mykiss) diet, in a completely randomized design. To perform the test, 3600 fries (11.5 ± 3.64 g) were treated for 56 days. According to the results, the increase in milkweed seed powder up to 40 g/kg, resulted in a significant increase in specific growth rate and intestinal lactic acid bacteria count compared to the control (p < 0.05). The best results of survival rate, feed conversion ratio, hepatosomatic and gastrosomatic indices were achieved in the treatments receiving 20, 30, 40, and 50 g/kg CSP (p < 0.05). Antibacterial activity of skin mucus, lysozyme and alkaline phosphatase showed the highest level in the 40 g/kg treatment (p < 0.05). Based on the results, the inclusion of 40 g/kg milkweed seed powder caused positive health effects and could be a suitable herbal feed additive in the rainbow trout diet.

In this study, we tested the effect of two agar-based biocomposites (differing for the fish meal source), on the Paracentrotus lividus gonad cycle progression and biochemical composition, in an offshore pilot scale trial. The purpose of this study was to identify a feeding regime for P. lividus that would allow adequate gonadal growth without accelerating the gamete maturation, in order to extend the market period. The purpose was pursued through the use of agar as a binding agent in the manufacturing of biocomposites to be used as feed for P. lividus and the realization of special offshore cages. The gonad index, histology, biochemical composition of fatty acids (FA) and free amino acids (FAA) and volatile molecule fingerprint were evaluated. Both the gonad index and the histological analysis showed a slower gametogenesis progression in the gonads of caged sea urchins with respect to the field collected ones, consistent with our previous results obtained testing a similar feed composition in a recirculating system. The amount and the relative proportions of FA and FAA in the gonads of wild and cultured gonads were similar, while the analysis of the volatile substances of the gonads of the sea urchin conducted using the electronic nose, shows that they vary as a result of the different feeding treatments. These results show that P. lividus sea urchins can be housed, under this feeding regime during the recovery phase of the gonads, without acceleration of the maturation of the gametes and thus in conditions to allow the extension of the market period. Moreover, the here tested agar-based biocomposites make it possible to reduce the management costs of the rearing system as they need to be replaced only once a week, and its environmental impact as they guarantee a limited nutrient dispersion in the water.

In the last decades, resistant microbial infection rate has dramatically increased, especially infections due to biofilm-producing strains that require increasingly complex treatments and are responsible for the increased mortality percentages compared with other infectious diseases. Considering that biofilms represent a key factor for a wide range of chronic infections with high drug tolerance, the treatment of biofilm-causing bacterial infections represents a great challenge for the future. Among new alternative strategies to conventional antimicrobial agents, the scientific interest has shifted to the study of biologically active compounds from plant-related extracts with known antimicrobial properties, in order to also evaluate their antibiofilm activity. In this regard, the aim of this study has been to assess the antibiofilm activity of polyphenolic extracts from myrtle leaf and pomegranate peel against oral pathogens of dental plaque, an excellent polymicrobial biofilm model. In particular, the in vitro antibiofilm properties of myrtle and pomegranate extracts, also in binary combination, were highlighted. In addition to inhibiting the biofilm formation, the tested polyphenolic extracts have been proven to destroy both preformed single-species and multispecies biofilms formed by Streptococcus mutans, Streptococcus oralis, Streptococcus mitis, and Rothia dentocariosa oral isolates, suggesting that the new natural sources are rich in promising compounds able to counteract biofilm-related infections.

The increasing incidence rate of oral diseases, the wide spread of antimicrobial resistance, and the adverse effects of conventional antibiotics mean alternative prevention and treatment options are needed to counteract oral pathogens. In this regard, our study aims to evaluate the antibacterial activity of polyphenolic extracts prepared from acacia honey, myrtle leaves, and pomegranate peel against cariogenic bacteria, such as Streptococcus mutans and Rothia dentocariosa. The chemical-physical parameters of acacia honey and the RP-HPLC polyphenolic profile of pomegranate peel extract have been previously described in our studies, while the characterization of myrtle extract, performed by HPLC analysis, is reported here. All the extracts were used singly and in binary combinations to highlight any synergistic effects. Moreover, the extracts were tested in association with amoxicillin to evaluate their ability to reduce the effective dose of this drug in vitro. The values of minimal inhibitory concentrations and minimal bactericidal concentrations have been used to quantitatively measure the antibacterial activity of the single extracts, while the fractional inhibitory concentration index has been considered as predictor of in vitro anticariogenic synergistic effects. Finally, a time-kill curve method allowed for the evaluation of the bactericidal efficacy of the combined extracts. The microbiological tests suggest that acacia honey, myrtle, and pomegranate extracts are able to inhibit the cariogenic bacteria, also with synergistic effects. This study provides useful and encouraging results for the use of natural extract combinations alone or in association with antibiotics (adjuvant therapy) as a valid alternative for the prevention and treatment of oral infectious diseases.

The aim of the present study was to investigate the potential effects of dietary synbiotic (SYN) (Pediococcus acidilactici + Galactooligosaccharides; 10 g kg−1), sodium propionate (SP; 5, 10 and 20 g kg−1) and a combination of SYN + SP on the growth performance, humoral immunity, antioxidant responses and disease resistance against Aeromonas hydrophila of African cichlid (Labidochromis lividus) fingerlings (0.52 ± 0.05 g) in a feeding trial lasting 63 days. A completely randomized design was run with eight treatments, including 0 (control) and supplemented diets containing SYN + SP (e.g., 10 + 5, 10 + 10, 10 + 20, 0 + 5, 0 + 10, 0 + 20 and 10 + 10). The lowest feed conversion ratio value was observed in fish fed the 5 g kg−1-SP and 10 g kg−1-SYN (p < 0.05). The highest values of protein efficiency ratio and protein productive value were recorded in fish fed the 10 g kg−1-SYN (p < 0.05). Fish fed the 10 g kg−1-SYN diet had the highest activities of immunity (lysozyme, immunoglobulin) and antioxidant responses (glutathione peroxidase and superoxide dismutase) (p < 0.05). After 28 days post-challenge, the highest survival rate (57%) was recorded in the diet containing 10 g kg−1 SYN and 5 g kg−1 SP. The results indicated that the single administration of SYN or combined with SP, especially at the level of 5 g kg−1 of diet, enhanced the survival and growth performances, humoral immune response, antioxidant and digestive enzymes of African cichlid.

This study was carried out to evaluate combined and singular effects of ethanolic extract of Persian shallot (Allium hirtifolium Boiss) and synbiotic Biomin®IMBO on growth performance, innate immune responses, and antioxidant defense in zebrafish (Danio rerio). Fish with initial weight of 151.90 ± 0.31 mg were allocated in 21 10-L glass aquariums. The experimental groups were as follows: T1, control (without any supplementation); T2, 1% synbiotic; T3, 3% synbiotic; T4, 1% Persian shallot (as a medical plant); T5, 3% Persian shallot; T6, 1% Persian shallot and 1% synbiotic; T7, 3% Persian shallot and 3% synbiotic. At the end of the experiment (60 days), all treatments significantly showed higher final weight (FW), weight gain (WG), WG (%), and specific growth rate (SGR) compared with the fish fed on control diet. Furthermore, both synbiotic Biomin®IMBO and Persian shallot significantly improved intestine immune parameters including lysozyme, alternative complement hemolytic activity (ACH50), total immunoglobulin (total Ig), and myeloperoxidase (MPO) of zebrafish compared to fish fed on control diet (p < 0.05). Also, in all experimental groups, hepatic catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GPx), and glutathione reductase (GR) activities significantly increased compared to the control group. Whereas, the highest MDA level was observed in the control group compared to the treatments (p < 0.05). Moreover, skin mucus immune parameters of zebrafish have been noticeably improved with synbiotic Biomin®IMBO and Persian shallot compared to fish fed on the control diet (p < 0.05). The results indicate that synbiotic or Persian shallot supplemented diet could enhance the general health status of the zebrafish.

Pectins are a group of carbohydrates found in structural parts of terrestrial plants with wide industrial and biomedical applications. This study was designed to investigate the dietary effects of apple pomace-derived pectin (APDP) in rainbow trout (Oncorhynchus mykiss). Four formulated diets were provided with different inclusion levels of APDP for 30 days: 0, 5, 10, and 20 g kg−1; referred to as control, P1, P2, and P3, respectively. In this study, 300 fish (3.56 ± 0.007 g) were randomly distributed into twelve fiberglass tanks and fed 3% of their respective body weight four times a day. At the end of the experiment, growth parameters, including weight gain, specific growth rate, and food conversion ratio (FCR) were significantly improved in P1 and P2 treatments compared to those of the other treatments. Results from proximate composition analysis showed that protein content increased, and lipid decreased in the P2 and P3 groups. Serum lysozyme, complement activity, total immunoglobulin levels, and total protein were significantly enhanced in all treatments compared to those of the control group. Gene expression results showed no significant difference in regulation of interleukin-1β (IL-1β); however, up-regulation of lysozyme, interleukin-8 (IL-8), and tumor necrosis factor-α (TNF-α) was observed in both P1 and P2. Unlike lipase, the activity of protease and amylase significantly increased in fish receiving different levels of APDP compared to the control (p < 0.05). In conclusion, the present findings suggest APDA as a promising feed additive for rainbow trout.

The crucial need for safe and healthy aquatic animals obligates researchers in aquaculture to investigate alternative and beneficial additives. Medicinal herbals and their extracts are compromised with diverse effects on the performances of aquatic animals. These compounds can affect growth performance and stimulate the immune system when used in fish diet. In addition, the use of medicinal herbs and their extracts can reduce oxidative stress induced by several stressors during fish culture. Correspondingly, aquatic animals could gain increased resistance against infectious pathogens and environmental stressors. Nevertheless, the exact mode of action where these additives can affect aquatic animals’ performances is still not well documented. Understanding the mechanistic role of herbal supplements and their derivatives is a vital tool to develop further the strategies and application of these additives for feasible and sustainable aquaculture. Gene-related studies have clarified the detailed information on the herbal supplements’ mode of action when administered orally in aquafeed. Several review articles have presented the potential roles of medicinal herbs on the performances of aquatic animals. However, this review article discusses the outputs of studies conducted on aquatic animals fed dietary, medicinal herbs, focusing on the gene expression related to growth and immune performances. Furthermore, a particular focus is directed to the expected influence of herbal supplements on the reproduction of aquatic animals.

Grape harvesting generates a high amount of wastes, mostly leaves, which represent an economic and ecological problem for farmers. New products can be generated through these wastes, giving environmental, social, and economic advantages while also meeting the industry demand for novel natural ingredients. In this study, aqueous leaf-extracts from two cultivars of Vitis vinifera Aglianico (Agl) and Greco di Tufo (Gre) were produced by microwave-assisted extraction (MAE) and evaluated in composition by ATR- FTIR and HPLC to identify the main phenolic compounds, especially quercetin and kaempferol. The results showed that leaves extracts confirm to be a potential source of phenolic compounds. Dry extracts, although highly functional, show critical handling characteristics, being sticky and unstable in normal post-processing conditions. A stable and easy handling microparticulate ingredient was produced by spray drying containing the most phenolic-rich obtained extract (AGL-28). The microparticle powder form based on pectin/maltodextrin matrix was produced with high process efficiency. The microstructures were able to confer functional and chemical stability to the extract while also showing good technological characteristics (high water dissolution rate and flow properties), transforming the extract into a handling ingredient able to meet new industrial uses.

The aim of the present study was to test the possible ameliorative efficacy of phytochemicals such as tannins on intestinal inflammation and dysbiosis. The effect of a chestnut shell (Castanea sativa) extract (CSE) rich in polyphenols, mainly represented by tannins, on k-carrageenan-induced intestinal inflammation in adult zebrafish (Danio rerio) was tested in a feeding trial. Intestinal inflammation was induced by 0.1% k-carrageenan added to the diet for 10 days. CSE was administered for 10 days after k-carrageenan induced inflammation. The intestinal morphology and histopathology, cytokine expression, and microbiota were analyzed. The k-carrageenan treatment led to gut lumen expansion, reduction of intestinal folds, and increase of the goblet cells number, accompanied by the upregulation of pro-inflammatory factors (TNFα, COX2) and alteration in the number and ratio of taxonomic groups of bacteria. CSE counteracted the inflammatory status enhancing the growth of health helpful bacteria (Enterobacteriaceae and Pseudomonas), decreasing the pro-inflammatory factors, and activating the anti-inflammatory cytokine IL-10. In conclusion, CSE acted as a prebiotic on zebrafish gut microbiota, sustaining the use of tannins as food additives to ameliorate the intestinal inflammation. Our results may be relevant for both aquaculture and medical clinic fields.

Nesfatin-1 (Nesf-1) is an anorexigenic peptide involved in the regulation of homeostatic feeding. Nesf-1 is expressed in the central nervous system and other organs, including pancreas, where it promotes the release of insulin from β-cells. This raises the possibility that Nesf-1 dysfunction could be involved in metabolic disorders, particularly in type 2 diabetes mellitus (T2D). Recently, it has been discovered that dolphins can be a natural animal model that fully replicates human T2D, due to its prolonged glucose tolerance curve and maintenance of a state of hyperglycemia similar to human T2D during fasting. This correspondence suggests that dolphins may be a suitable model for investigating physiological and pathological metabolic disorders. Here, we have characterized Nesf-1 distribution in the pancreas of the common bottlenose dolphin (Tursiops truncatus) and measured plasmatic levels of Nesf-1 and glucose during fasting and post-prandial states. The Mediterranean Marine Mammal Tissue Bank (MMMTB) of the University of Padova provided us with pancreas samples, derived from four animals, and plasma samples, collected before and after the main meal. Interestingly, our results showed that Nesf-1-immunoreactive cells were distributed in Langerhans islets, co-localized with glucagon in α-cells. Similar to humans, dolphin plasma Nesf-1 concentration doesn’t show a statistically significant difference when comparing fasting and post-prandial states. On the other hand, blood glucose levels were significantly higher before than after the main meal. Our data provide a comparative analysis for further studies on the involvement of Nesf-1 in mammalian metabolic disorders.

Fish meal (FM) is no longer a sustainable source for the increasing aquaculture industry. Animal proteins from insects may be used as a FM alternative source as long as they do not create adverse effects in fish. Black soldier fly larvae meal (BSFLM) was tested in a 12-week experiment on Nile tilapia (Oreochromis niloticus). Four hundred and twenty (14.77 ± 2.09 g) fish were divided into seven groups and were fed seven diets: control (0% BSFLM-100% FM), and FM replaced by BSFLM at rates of 10%, 20%, 40%, 60%, 80% and 100%. Growth indexes, feed utilization efficiency indices, feed intake, and survival rate were not significantly different (p > 0.05) between FM and BSFLM fed fish. Values of red blood cell, white blood cells, hemoglobin, hematocrit, mean corpuscular volume and hemoglobin, mean corpuscular hemoglobin concentration, red blood cell distribution width, and platelet values were not affected by BSFLM. Skin, mucus lysozyme, and peroxidase activities were improved in BSFLM fed fish. BSFLM can be used as a substitution for FM in the Nile tilapia (O. niloticus) diet at up to a 100% rate with no adverse effects.

Fresh figs are very sensitive to microbial spoilage, even in cold storage conditions. Thus, fresh figs are high perishable products during postharvest with microbiological decay that induces an unpleasant taste and smell due to rot, and suitable conservation methods must therefore be applied. The fruit usually is consumed fresh locally, dried, or preserved longer term in other transformed forms. A sustainable approach to extend the shelf-life of figs can be constituted by application of an edible coating able to maintain the quality of the fruit during storage. A comparison between fresh figs in a commercial preservation system, with the figs preserved in an edible coating, and an active edible coating to preserve their quality characteristics was carried out. The coating efficacy was enhanced with the addition of pomegranate peel extract at two different concentrations. The inclusion of a component with high antioxidant activity in an edible coating proved to be an excellent method for preserving the quality of this highly perishable fruit. The application of natural products, obtained from renewable sources, represents a simple and economic strategy, but also a tool capable of preserving the quality of the fruit during the postharvest storage, which is often consumed in production areas due to shelf-life problems.

The 3,5-diiodo-L-thyronine (3,5-T2) is an endogenous metabolite of thyroid hormones, whose administration to rodents fed high-fat diet (HFD) prevents body weight increase and reverts the expression pattern of pro-inflammatory factors associated to HFD. The diet-induced obese (D.I.O.) zebrafish (Danio rerio) has been recently used as an experimental model to investigate fundamental processes underlying central and peripheral obesity-driven inflammation. Herein, we aim to understand the role of 3,5-T2 in regulating central and peripheral inflammation in D.I.O. model of zebrafish. 3,5-T2 (10 nM and 100 nM) was administered with the obesity-inducing diet (D.I.O. with 3,5-T2) or after 4 weeks of obesity-inducing diet (D.I.O. flw 3,5-T2). 3,5-T2 significantly increased the body weight and serum triglyceride levels in D.I.O. zebrafish in both conditions. Moreover, 3,5-T2 sustained or increased inflammation in the anterior (AI) and mid (MI) intestine when administered with the obesity-inducing diet, as indicated by the immunoexpression of the inflammatory markers tumor-necrosis factor-α (TNFα), cyclooxygenase 2 (COX2), calnexin, caspase 3, and proliferating cell nuclear antigen (PCNA). On the contrary, when 3,5-T2 was administered after the obesity-inducing diet, partly reverted the intestinal alteration induced by D.I.O. In addition, brain inflammation, as indicated by the increase in the activation of microglia, was detected in D.I.O. zebrafish and D.I.O. treated with 3,5-T2. These findings reveal that the effects of 3,5-T2 on fish intestine and brain can deviate from those shown in obese mammals, opening new avenues to the investigation of the potential impact of this thyroid metabolite in different diseases including obesity.

Algae have multiple similarities with fungi, with both belonging to the Thallophyte, a polyphyletic group of non-mobile organisms grouped together on the basis of similar characteristics, but not sharing a common ancestor. The main difference between algae and fungi is noted in their metabolism. In fact, although algae have chlorophyll-bearing thalloids and are autotrophic organisms, fungi lack chlorophyll and are heterotrophic, not able to synthesize their own nutrients. However, our studies have shown that the extremophilic microalga Galderia sulphuraria (GS) can also grow very well in heterotrophic conditions like fungi. This study was carried out using several approaches such as scanning electron microscope (SEM), gas chromatography/mass spectrometry (GC/MS), and infrared spectrophotometry (ATR-FTIR). Results showed that the GS, strain ACUF 064, cultured in autotrophic (AGS) and heterotrophic (HGS) conditions, produced different biomolecules. In particular, when grown in HGS, the algae (i) was 30% larger, with an increase in carbon mass that was 20% greater than AGS; (ii) produced higher quantities of stearic acid, oleic acid, monounsaturated fatty acids (MUFAs), and ergosterol; (iii) produced lower quantities of fatty acid methyl esters (FAMEs) such as methyl palmytate, and methyl linoleate, saturated fatty acids (SFAs), and poyliunsaturated fatty acids (PUFAs). ATR-FTIR and principal component analysis (PCA) statistical analysis confirmed that the macromolecular content of HGS was significantly different from AGS. The ability to produce different macromolecules by changing the trophic conditions may represent an interesting strategy to induce microalgae to produce different biomolecules that can find applications in several fields such as food, feed, nutraceutical, or energy production.

Hypocretins/Orexins neuropeptides are known to regulate numerous physiological functions, such as energy homeostasis, food intake, sleep/wake cycle, arousal and wakefulness, in vertebrates. Previous studies on mice have revealed an intriguing orexins/endocannabinoids (ECs) signaling interaction at both structural and functional levels, with OX-A behaving as a strong enhancer of 2-arachydonoyl-glycerol (2-AG) biosynthesis. In this study, we describe, for the first time in the brain of zebrafish, the anatomical distribution and co-expression of orexin (OX-2R) and endocannabinoid (CB1R) receptors, suggesting a functional interaction. The immunohistochemical colocalization of these receptors by confocal imaging in the dorsal and ventral telencephalon, suprachiasmatic nucleus (SC), thalamus, hypothalamus, preoptic area (PO) and cerebellum, is reported. Moreover, biochemical quantification of 2-AG levels by LC-MS supports the occurrence of OX-A-induced 2-AG biosynthesis in the zebrafish brain after 3 h of OX-A intraperitoneal (i.p.; 3 pmol/g) or intracerebroventricular (i.c.v.; 0.3 pmol/g) injection. This effect is likely mediated by OX-2R as it is counteracted by i.p./i.c.v administration of OX-2R antagonist (SB334867, 10 pmol/g). This study provides compelling morphological and functional evidence of an OX-2R/CB1R signaling interaction in the brain of adult zebrafish, suggesting the use of this well-established vertebrate animal model for the study of complex and phylogenetically conserved physiological functions.

In this study, polysaccharides of marine origin (agar, alginate and κ-carrageenan) were used to embed nutrients to fabricate biocomposites to be employed in animal feeding. The consistency of biocomposites in water has been evaluated up to 14 days, by several methods: swelling, nutrient release and granulometric analysis. Biocomposites were produced with varying percentages of nutrients (5%–25%) and polysaccharides (1%–2%–3%). All possible biopolymer combinations were tested in order to select those with the best network strength. The best performing biocomposites were those manufactured with agar 2% and nutrients 10%, showing the lowest percentage of water absorption and nutrient release. Biocomposites made of agar 2% and nutrients 10% were the most stable in water and were therefore used to analyze their behavior in water with respect to the release of quercetin, a phenolic compound with demonstrated high antibacterial and antioxidant activities. The leaching of such molecules in water was therefore employed as a further indicator of biocomposite water stability. Altogether, our results confirm the suitability of agar as a binder for biocomposites and provide a positive contribution to aquaculture.

The complexes formed by BCL10, MALT1 and members of the family of CARMA proteins have recently been the focus of much attention because they represent a key mechanism for regulating activation of the transcription factor NF‐κB. Here, we report the functional characterization of a novel isoform of BCL10 in the troutOncorhynchus mykiss, which we named tBCL10. tBCL10 dimerizes, binds to components of the CBM complex and forms cytoplasmic filaments. Functionally, tBCL10 activates NF‐κB transcription factor and is inhibited by the deubiquitinating enzyme A20. Finally, depletion experiments indicate that tBCL10 can functionally replace the human protein. This work demonstrates the evolutionary conservation of the mechanism of NF‐κB activation through the CBM complex, and indicates that the rainbow troutO. mykiss can serve as a model organism to study this pathway.

Leaves of Vitis vinifera cv. Greco di Tufo, a precious waste made in the Campania Region (Italy), after vintage harvest, underwent reduction, lyophilization, and ultrasound-assisted maceration in ethanol. The alcoholic extract, as evidenced by a preliminary UHPLC-HR-MS analysis, showed a high metabolic complexity. Thus, the extract was fractionated, obtaining, among others, a fraction enriched in flavonol glycosides and glycuronides. Myricetin, quercetin, kaempferol, and isorhamnetin derivatives were tentatively identified based on their relative retention time and TOF-MS2 data. As the localization of saccharidic moiety in glycuronide compounds proved to be difficult due to the lack of well-established fragmentation pattern and/or the absence of characteristic key fragments, to obtain useful MS information and to eliminate matrix effect redundancies, the isolation of the most abundant extract’s compound was achieved. HR-MS/MS spectra of the compound, quercetin-3-O-glucuronide, allowed us to thoroughly rationalize its fragmentation pattern, and to unravel the main differences between MS/MS behavior of flavonol glycosides and glycuronides. Furthermore, cytotoxicity assessment on the (poly)phenol rich fraction and the pure isolated compound was carried out using central nervous system cell lines. The chemoprotective effect of both the (poly)phenol fraction and quercetin-3-O-glucuronide was evaluated.

In this study different methods were used to evaluate the effectiveness of a carrageenan coating and carrageenan coating incorporating lemon essential oil (ELO) in preserving the physicochemical and olfactory characteristics of trout fillets stored at 4 °C up to 12 days. The fillet morphological structure was analyzed by histological and immunological methods; lipid peroxidation was performed with the peroxide and thiobarbituric acid reactive substances (TBARS) tests. At the same time, two less time-consuming methods, such as Attenuated Total Reflectance-Fourier Transformed Infrared (ATR-FTIR) spectroscopy and the electronic nose, were used. Uncoated trout fillets (UTF) showed a less compact tissue structure than carrageenan-coated threads (CTF) and coated fillets of carrageenan (active) ELO (ACTF), probably due to the degradation of collagen, as indicated by optical microscopy and ATR-FTIR. UTF showed greater lipid oxidation compared to CTF and ACTF, as indicated by the peroxide and TBARS tests and ATR-FTIR spectroscopy. The carrageenan coating containing ELO preserved the olfactory characteristics of the trout fillets better than the carrageenan coating alone, as indicated by the electronic nose analysis. This study confirms that both carrageenan and ELO containing carrageenan coatings slow down the decay of the physicochemical and olfactory characteristics of fresh trout fillets stored at 4 °C, although the latter is more effective.

In this study, chestnut (Castanea sativa) shell was extracted with different solvents, and immunomodulatory activity was investigated in an in vitro model system using blood and intestinal leukocytes of Oncorhynchus mykiss. Gallic acid (GA) was used as a standard. Chestnut shell extract (CSE) and GA readily entered both blood and intestinal leukocytes. Superoxide anion production and phagocytosis were decreased by low doses of CSE and increased with high doses. CSE and GA differently regulated cytokine expression in blood and intestinal leukocytes. High doses of CSE upregulated IL-1β, TNF-α, and IL-10 in intestinal leukocytes and IL-10 in blood leukocytes. Low doses of CSE upregulated IL-1β and TNF-α in blood leukocytes. GA appeared to be effective only in blood leukocytes. The effects of CSE on pro- and anti-inflammatory cytokines seemed to suggest an alert effect of the immune defense system against a possible infectious agent. The less evident effect of GA in comparison to CSE could have been attributable to the synergistic and/or additive effects of polyphenols in the latter. The immune-stimulating activity of CSE reported here could be useful for future practical applications in fish health.

This study aimed to evaluate the in vitro activity of Zanthoxylum rhoifolium bark (Zr-b) extracts against pathogenic aquatic oomycete/fungal isolates that cause different diseases in native European crayfish resulting in an elevated mortality rate and severe economic repercussions. n-hexane, chloroform, chloroform–methanol (9:1) and methanol extracts of Zr-b were used to evaluate the antifungal activity against the strain UEF88662 of Aphanomyces astaci (oomycete) and the strain SMM2 of Fusarium avenaceum (fungus). The anti-oomycete and antifungal activity was quantitatively evaluated by growth, survival and sporulation microbiological assays. The extracts tested demonstrated a dose-dependent inhibitory effect on oomycete and fungal growth and survival, as well as on the production of oomycete and fungal spores. This work presents alternatives for the treatment and prevention of the spreading of Aphanomyces astaci and Fusarium avenaceum, the etiological agents of the diseases crayfish plague and brown spot disease, respectively. The antifungal properties of Zanthoxylum rhoifolium bark extracts warrant further research on their use in the prevention and treatment of both oomycete and fungal diseases. The antifungal properties of Zanthoxylum rhoifolium bark extracts, shown in vitro, indicate the possibility of their use in new therapeutic and prophylactic strategies, providing perspectives for the design of in vivo studies.

The complexes formed by BCL10, MALT1 and specific members of the family of CARMA proteins (CBM complex), have recently focused much attention because they represent a central hub regulating activation of the transcription factor NF-κB following various cellular stimulations. In this manuscript, we report the functional characterization of a Danio rerio 241 amino acids polypeptide ortholog of the Caspase recruiting domain (CARD)-containing protein BCL10. Biochemical studies show that zebrafish Bcl10 (zBcl10) dimerizes and binds to components of the CBM complex. Fluorescence microscopy observations demonstrate that zBcl10 forms cytoplasmic filaments similar to that formed by human BCL10 (hBCL10). Functionally, in human cells zBcl10 is more effective in activating NF-κB compared to hBCL10, possibly due to the lack of carboxy-terminal inhibitory serine residues present in the human protein. Also, depletion experiments carried out through expression of short hairpin RNAs targeting hBCL10 indicate that zBcl10 can functionally replace the human protein. Finally, we show that the zebrafish cell line PAC2 is suitable to carry out reporter assays for monitoring the activation state of NF- kB transcription factor. In conclusion, this work shows that zebrafish may excellently serve as a model organism to study complex and intricate signal transduction pathways, such as those that control NF-κB activation.

This study provides a general approach to the presence and possible role of orexins and their receptors in the gut (three gastric chambers and intestine) of confined environment bottlenose dolphin. The expression of prepro-orexin, orexin A and B and orexin 1 and 2 receptors were investigated by single immunostaining and western blot analysis. The co-localization of vasoactive intestinal peptide and orexin 1 receptor in the enteric nervous system was examined by double immunostaining. Also, orexin A concentration were measured in plasma samples to assess the possible diurnal variation of the plasma level of peptide in this species. Our results showed that the orexin system is widely distributed in bottlenose dolphin enteric nervous system of the all gastrointestinal tract examined. They are very peculiar and partially differs from that of terrestrial mammals. Orexin peptides and prepro-orexin were expressed in the main stomach, pyloric stomach and proximal intestine; while orexin receptors were expressed in the all examined tracts, with the exception of main stomach where found no evidence of orexin 2 receptor. Co-localization of vasoactive intestinal peptide and orexin 1 receptor were more evident in the pyloric stomach and proximal intestine. These data could suggest a possible role of orexin system on the contractility of bottlenose dolphin gastrointestinal districts. Finally, in agreement with several reports, bottlenose dolphin orexin A plasma level was higher in the morning during fasting. Our results emphasize some common features between bottlenose dolphin and terrestrial mammals. Certainly, further functional investigations may help to better explain the role of the orexin system in the energy balance of bottlenose dolphin and the complex interaction between feeding and digestive physiology.

In this study, a hydroalcoholic chestnut shell extract was characterized and tested on six different human cell lines. Gallic, ellagic, and syringic acids were the most abundant non-condensed compounds in the chestnut extract, as determined by high performance liquid chromatography (HPLC). Tannins were mainly represented by condensed monomeric units of epigallocatechin and catechin/epicatechin. After 48 h of treatment, only the human hepatoblastoma HepG2 cells reached an inhibition corresponding to IC50 with an increase of apoptosis and mitochondrial depolarization. The cytokinome evaluation before and after treatment revealed that the vascular endothelial growth factor (VEGF) and the tumor necrosis factor (TNF)-α decreased after the treatment, suggesting a potential anti-angiogenic and anti-inflammatory effect of this extract. Moreover, the metabolome evaluation by 1H-NMR evidenced that the polyphenols extracted from chestnut shell (PECS) treatment affected the levels of some amino acids and other metabolites. Overall, these data highlight the effects of biomolecules on cell proliferation, apoptosis, cell cycle and mitochondrial depolarization, and on cytokinomics and metabolomics profiles.

We will deal with the partial characterization and the activity of the following digestive enzymes: amylase, pectinase, alginase, lipase, and protease present in the digestive tract of juvenile freshwater crayfish Cherax albidus. Gastric juices, the hepatopancreas, and the intestine were sampled for enzyme analysis. Among carbohydratases, amylase activity was the highest. It was significantly higher in the intestine than in the gastric juice and hepatopancreas. Lipase activity was significantly higher in the hepatopancreas and the intestine compared to the gastric juice. Both alkaline and acid proteases were found. Alkaline proteases were characterized by employing specific protease inhibitors. Both trypsin and chymotrypsin activity was detected. The majority of alkaline protease activity was ascribable to trypsin. Several isoforms of digestive enzymes were identified by gel electrophoresis. This work provides basic information to study the digestive abilities of crayfish.

In this study, we report ghrelin and leptin immunoreactive (ir) cells distribution in the gastrointestinal tract and blood ghrelin and leptin levels in rainbow trout ( Oncorhynchus mykiss) and Murray cod ( Maccullochella peelii peelii) fed diets with different fatty acid compositions. Juvenile rainbow trout and Murray cod were fed five iso-energetic experimental diets containing fish oil (FO) or one of the following vegetable oils (VO): olive oil (OO), sunflower oil (SO), linseed oil (LO), and palm oil (PO); as the added dietary lipid source. The presence and distribution of both ghrelin and leptin ir cells in the gastrointestinal tract were affected by the inclusion of VO. Ghrelin ir cells were found in the gastric glands of rainbow trout and in the mioenteric plexuses of the stomach of Murray cod fed FO. Ghrelin ir cells were localized in the mucosa of the intestine of rainbow trout and Murray cod fed VO. Leptin ir cells were more abundant in the epithelial lining of the mucosa folds and in the glands of the stomach of rainbow trout fed VO. Leptin immunoreactivity was detected in the gastric mioenteric plexus of Murray cod fed FO. No differences were found both in ghrelin and leptin levels in blood plasma or in the growth rates of rainbow trout and Murray cod fed the different experimental diets.
