Abstract Urbanization that occurs across a gradient from low‐ to high‐density development, is a primary driver of landscape change that can affect biodiversity. Animals balance trade‐offs in obtaining resources and avoiding anthropogenic disturbances across the gradient of urbanization to maximize their fitness. However, additional research is necessary to understand seasonal variations in how animals respond to urbanization, particularly in arid regions, where resource availability shifts drastically across seasons. Our objective was to evaluate the response of a suite of bat species to urbanization and whether species shift their response to urbanization across seasons. We predicted that the response of bats to urbanization would differ among species, with some species being more sensitive to urbanization than others. We also predicted that bat species would increase the use of moderate and highly urbanized areas in the summer season where food and water resources were assumed to be greater compared with wildland areas. To evaluate these predictions, we used a stratified random sampling design to sample 50 sites with stationary acoustic bat monitors across the gradient of urbanization in the Phoenix metropolitan area, Arizona, USA during four seasons. We identified a total of 14 bat species during 1000 survey nights. Consistent with predictions, bat species exhibited different responses to urbanization, with most species exhibiting a negative relationship with urbanization, and some species exhibiting a quadratic or positive relationship with urbanization. Counter to predictions, most species did not appear to shift their response to urbanization across seasons. Consistent with predictions, plant productivity and water were important for some species in the summer season. Differences in the response of bat species to urbanization was likely related to species traits (e.g., wing morphology and echolocation call characteristics) and behavioral strategies that influence a species' sensitivity to anthropogenic disturbances and ability to access available resources in urbanized areas. Ultimately, to promote the management and conservation of bats, it is likely important to maintain resources in urbanized areas for bats that are more tolerant of urbanization and to conserve areas of undeveloped high‐quality habitat with low anthropogenic disturbance in wildland areas for bats that are sensitive to urbanization.

High viral tolerance coupled with an extraordinary regulation of the immune response makes bats a great model to study host-pathogen evolution. Although many immune-related gene gains and losses have been previously reported in bats, important gene families such as antimicrobial peptides (AMPs) remain understudied. We built an exhaustive bioinformatic pipeline targeting the major gene families of defensins and cathelicidins to explore AMP diversity and analyze their evolution and distribution across six bat families. A combination of manual and automated procedures identified 29 AMP families across queried species, with α-, β-defensins, and cathelicidins representing around 10% of AMP diversity. Gene duplications were inferred in both α-defensins, which were absent in five species, and three β-defensin gene subfamilies, but cathelicidins did not show significant shifts in gene family size and were absent in Anoura caudifer and the pteropodids. Based on lineage-specific gains and losses, we propose diet and diet-related microbiome evolution may determine the evolution of α- and β-defensins gene families and subfamilies. These results highlight the importance of building species-specific libraries for genome annotation in non-model organisms and shed light on possible drivers responsible for the rapid evolution of AMPs. By focusing on these understudied defenses, we provide a robust framework for explaining bat responses to pathogens.

Abstract Ecological understanding of host–pathogen dynamics is the basis for managing wildlife diseases. Since 2008, federal, state, and provincial agencies and tribal and private organizations have collaborated on bat and white‐nose syndrome (WNS) surveillance and monitoring, research, and management programs. Accordingly, scientists and managers have learned a lot about the hosts, pathogen, and dynamics of WNS. However, effective mitigation measures to combat WNS remain elusive. Host–pathogen systems are complex, and identifying ecological research priorities to improve management, choosing among various actions, and deciding when to implement those actions can be challenging. Through a cross‐disciplinary approach, a group of diverse subject matter experts created an influence diagram used to identify uncertainties and prioritize research needs for WNS management. Critical knowledge gaps were identified, particularly with respect to how WNS dynamics and impacts may differ among bat species. We highlight critical uncertainties and identify targets for WNS research. This tool can be used to maximize the likelihood of achieving bat conservation goals within the context and limitations of specific real‐world scenarios.

White-nose syndrome (WNS) is an emerging infectious disease devastating hibernating North American bat populations that is caused by the psychrophilic fungus Geomyces destructans. Previous histopathological analysis demonstrated little evidence of inflammatory responses in infected bats, however few studies have compared other aspects of immune function between WNS-affected and unaffected bats. We collected bats from confirmed WNS-affected and unaffected sites during the winter of 2008-2009 and compared estimates of their circulating levels of total leukocytes, total immunoglobulins, cytokines and total antioxidants. Bats from affected and unaffected sites did not differ in their total circulating immunoglobulin levels, but significantly higher leukocyte counts were observed in bats from affected sites and particularly in affected bats with elevated body temperatures (above 20°C). Bats from WNS-affected sites exhibited significantly lower antioxidant activity and levels of interleukin-4 (IL-4), a cytokine that induces T cell differentiation. Within affected sites only, bats exhibiting visible fungal infections had significantly lower antioxidant activity and levels of IL-4 compared to bats without visible fungal infections. Overall, bats hibernating in WNS-affected sites showed immunological changes that may be evident of attempted defense against G. destructans. Observed changes, specifically elevated circulating leukocytes, may also be related to the documented changes in thermoregulatory behaviors of affected bats (i.e. increased frequencies in arousal from torpor). Alterations in immune function may reflect expensive energetic costs associated with these processes and intrinsic qualities of the immunocapability of hibernating bats to clear fungal infections. Additionally, lowered antioxidant activity indicates a possible imbalance in the pro- versus antioxidant system, may reflect oxidative tissue damage, and should be investigated as a contributor to WNS-associated morbidity and mortality.

White-nose syndrome (WNS) is the most devastating condition ever reported for hibernating bats, causing widespread mortality in the northeastern United States. The syndrome is characterized by cutaneous lesions caused by a recently identified psychrophilic and keratinophylic fungus (Geomyces destructans), depleted fat reserves, atypical behavior, and damage to wings; however, the proximate cause of mortality is still uncertain. To assess relative levels of immunocompetence in bats hibernating in WNS-affected sites compared with levels in unaffected bats, we describe blood plasma complement protein activity in hibernating little brown myotis (Myotis lucifugus) based on microbicidal competence assays using Escherichia coli, Staphylococcus aureus and Candida albicans. Blood plasma from bats collected during mid-hibernation at WNS-affected sites had higher bactericidal ability against E. coli and S. aureus, but lower fungicidal ability against C. albicans when compared with blood plasma from bats collected at unaffected sites. Within affected sites during mid-hibernation, we observed no difference in microbicidal ability between bats displaying obvious fungal infections compared to those without. Bactericidal ability against E. coli decreased significantly as hibernation progressed in bats collected from an affected site. Bactericidal ability against E. coli and fungicidal ability against C. albicans were positively correlated with body mass index (BMI) during late hibernation. We also compared complement activity against the three microbes within individuals and found that the ability of blood plasma from hibernating M. lucifugus to lyse microbial cells differed as follows: E. coli>S. aureus>C. albicans. Overall, bats affected by WNS experience both relatively elevated and reduced innate immune responses depending on the microbe tested, although the cause of observed immunological changes remains unknown. Additionally, considerable trade-offs may exist between energy conservation and immunological responses. Relationships between immune activity and torpor, including associated energy expenditure, are likely critical components in the development of WNS.
