Abstract

Objectives. This study aimed to compare the effects of wrestling and sumo wrestling as similar combat sports activities in a training program on technical wrestling scores, wrestling performance indexes, and intrinsic motivation in youth practitioners. Materials and methods. A randomized controlled trial using a single-blinded design, and repeated measures of parallel groups (sumo-based group: EG, n = 19, aged = 12.42 ± 0.87 years; and only wrestling group: CG, n = 19, aged = 12.41 ± 0.78 years) was conducted. Three 65-minute weekly sessions for four weeks were dedicated to the interventions. A two-factor mixed analysis of variance (ANOVA) model with repeated measures was performed to assess the group vs. time impact analysis. Results. Abstract

Abstract The KM3NeT/ARCA neutrino detector is currently under construction at 3500 m depth offshore Capo Passero, Sicily, in the Mediterranean Sea. The main science objectives are the detection of high-energy cosmic neutrinos and the discovery of their sources. Simulations were conducted for the full KM3NeT/ARCA detector, instrumenting a volume of 1 km $$^3$$ 3 , to estimate the sensitivity and discovery potential to point-like neutrino sources. This paper covers the reconstruction of track- and shower-like signatures, as well as the criteria employed for neutrino event selection. With an angular resolution below 0.1 $$^\circ $$ ∘ for tracks and under 2 $$^\circ $$ ∘ for showers, the sensitivity to point-like neutrino sources surpasses existing observed limits across the entire sky.

22. The contribution of the Middle Triassic fossil assemblage of Monte San Giorgio to insect evolution

Abstract

Abstract The Triassic represents a critical period for understanding the turnover of insect fauna from the Paleozoic to the Mesozoic following the end-Permian mass extinctions (EPME); however, fossil deposits from the Early-Middle Triassic are scarce. The exceptionally preserved 239 million-year-old fossil insect fauna recorded at Monte San Giorgio (Switzerland), including 248 fossils representing 15 major insect clades is presented here. Besides the exceptional features, including their small size and excellent preservation, the fossils have importance in the evolutionary history of the group. The taxonomic and ecological diversity recovered, including both freshwater (dragonflies and caddisflies) and terrestrial taxa (true bugs and wasps), demonstrates that complex environments sustained a paleocommunity dominated by monurans (thought not to have survived the EPME), midges, and beetles. Interestingly, a blattodean-like fossil bearing an external ootheca was also found, important for understanding Paleozoic roachoids to extant cockroaches’ transition and the evolution of maternal brood care. Moreover, the youngest and first complete specimen of †Permithonidae and the oldest sawfly fossils were discovered. Finally, round-shaped bodies, compatible with seminal capsules or lycophyte spores, were found on the abdomens of several midge-like individuals. If these are spores, non-seed-bearing plants could have been the first entomophilous plants rather than gymnosperms, as recently supposed. Altogether, these fossils contribute substantially to understanding insect evolution and Paleozoic-Mesozoic faunal turnover.

Abstract

The notion of quasi-particles is essential for understanding the behaviour of complex many-body systems. A prototypical example of a quasi-particle, a polaron, is an impurity strongly interacting with a surrounding medium. Fermi polarons, created in a Fermi sea, provide a paradigmatic realization of this concept. As an inherent and important property such quasi-particles interact with each other via modulation of the medium. While quantum simulation experiments with ultracold atoms have significantly improved our understanding of individual polarons, the detection of their interactions has remained elusive in these systems. Here, we report the unambiguous observation of mediated interactions between Fermi polarons consisting of K impurities embedded in a Fermi sea of Li atoms. Our results confirm two landmark predictions of Landau's Fermi-liquid theory: the shift of the polaron energy due to mediated interactions, linear in the concentration of impurities, and its sign inversion with impurity quantum statistics. For weak to moderate interactions between the impurities and the medium, we find excellent agreement with the static (zero-momentum and energy) predictions of Fermi-liquid theory. For stronger impurity-medium interactions, we show that the observed behaviour at negative energies can be explained by a more refined many-body treatment including retardation and molecule formation

Abstract

Abstract The measurement of the flux of muons produced in cosmic ray air showers is essential for the study of primary cosmic rays. Such measurements are important in extensive air shower detectors to assess the energy spectrum and the chemical composition of the cosmic ray flux, complementary to the information provided by fluorescence detectors. Detailed simulations of the cosmic ray air showers are carried out, using codes such as CORSIKA, to estimate the muon flux at sea level. These simulations are based on the choice of hadronic interaction models, for which improvements have been implemented in the post-LHC era. In this work, a deficit in simulations that use state-of-the-art QCD models with respect to the measurement deep underwater with the KM3NeT neutrino detectors is reported. The KM3NeT/ARCA and KM3NeT/ORCA neutrino telescopes are sensitive to TeV muons originating mostly from primary cosmic rays with energies around 10 TeV. The predictions of state-of-the-art QCD models show that the deficit with respect to the data is constant in zenith angle; no dependency on the water overburden is observed. The observed deficit at a depth of several kilometres is compatible with the deficit seen in the comparison of the simulations and measurements at sea level.

Abstract

Abstract Extracellular vesicles (EVs) are promising natural nanocarriers for the delivery of therapeutic agents. As with any other kind of cell, red blood cells (RBCs) produce a limited number of EVs under physiological and pathological conditions. Thus, RBC-derived extracellular vesicles (RBCEVs) have been recently suggested as next-generation delivery systems for therapeutic purposes. In this paper, we show that thanks to their unique biological and physicochemical features, RBCs can be efficiently pre-loaded with several kinds of molecules and further used to generate RBCEVs. A physical vesiculation method, based on “soft extrusion”, was developed, producing an extremely high yield of cargo-loaded RBCEV mimetics. The RBCEVs population has been deeply characterized according to the new guidelines MISEV2023, showing great homogeneity in terms of size, biological features, membrane architecture and cargo. In vitro preliminary results demonstrated that RBCEVs are abundantly internalized by cells and exert peculiar biological effects. Indeed, efficient loading and delivery of miR-210 by RBCEVs to HUVEC has been proven, as well as the inhibition of a known mRNA target. Of note, the bench-scale process can be scaled-up and translated into clinics. In conclusion, this investigation could open the way to a new biomimetic platform for RNA-based therapies and/or other therapeutic cargoes useful in several diseases.

Abstract

Abstract An under-recognised aspect of the current humanitarian catastrophe in Gaza is the impact of the war on the environment and the associated risks for human health. This commentary contextualises these impacts against the background of human suffering produced by the overwhelming violence associated with the use of military force against the general population of Gaza. In calling for an immediate cessation to the violence, the authors draw attention to the urgent need to rebuild the health care system and restore the physical and human infrastructure that makes a liveable environment possible and promotes human health and well-being, especially for the most vulnerable in the population. Environmental remediation should therefore form one of the most important parts of international efforts to assist reconstruction, through which we hope Palestinians and Israelis will achieve lasting peace, health, and sustainable development, all as part of accepted international human rights obligations.

Abstract We introduce a georeferenced dataset of Net Ecosystem Exchange (NEE), Ecosystem Respiration (ER) and meteo-climatic variables (air and soil temperature, air relative humidity, soil volumetric water content, pressure, and solar irradiance) collected at the Nivolet Plain in Gran Paradiso National Park (GPNP), western Italian Alps, from 2017 to 2023. NEE and ER are derived by measuring the temporal variation of CO2 concentration obtained by the enclosed chamber method. We used a customised portable non-steady-state dynamic flux chamber, paired with an InfraRed Gas Analyser (IRGA) and a portable weather station, measuring CO2 fluxes at a number of points (around 20 per site and per day) within five different sites during the snow-free season (June to October). Sites are located within the same hydrological basin and have different geological substrates: carbonate rocks (site CARB), gneiss (GNE), glacial deposits (GLA, EC), alluvial sediments (AL). This dataset provides relevant and often missing information on high-altitude mountain ecosystems and enables new comparisons with other similar sites, modelling developments and validation of remote sensing data.

Abstract The domestication process in grapevines has facilitated the fixation of desired traits. Nowadays, vegetative propagation through cuttings enables easier preservation of these genotypes compared to sexual reproduction. Nonetheless, even with vegetative propagation, various phenotypes are often present within the same vineyard due to the accumulation of somatic mutations. These mutations are not the sole factors influencing phenotype. Alongside somatic variations, epigenetic variation has been proposed as a pivotal player in regulating phenotypic variability acquired during domestication. The emergence of these epialleles might have significantly influenced grapevine domestication over time. This study aims to investigate the impact of domestication on methylation patterns in cultivated grapevines. Reduced-representation bisulfite sequencing was conducted on 18 cultivated and wild accessions. Results revealed that cultivated grapevines exhibited higher methylation levels than their wild counterparts. Differential Methylation Analysis between wild and cultivated grapevines identified a total of 9955 differentially methylated cytosines, of which 78% were hypermethylated in cultivated grapevines. Functional analysis shows that core methylated genes (consistently methylated in both wild and cultivated accessions) are associated with stress response and terpenoid/isoprenoid metabolic processes. Meanwhile, genes with differential methylation are linked to protein targeting to the peroxisome, ethylene regulation, histone modifications, and defense response. Collectively, our results highlight the significant roles that epialleles may have played throughout the domestication history of grapevines.

A pesquisa visa analisar os múltiplos usos do território e os desafios para a conservação dos recursos naturais no Parque Nacional de Brasília (PNB). Foram realizados levantamentos bibliográfico e documental, pesquisa de campo, entrevistas, mapeamento, organização e análise das informações. Os resultados evidenciaram potencialidades socioambientais como fragmentos florestais de Cerrado, diversidade biológica nas nascentes e córregos utilizados para o abastecimento local – Represa Santa Maria –, trilhas ecológicas, piscinas naturais, Núcleo de Educação Ambiental, patrimônio histórico e cultural. Entretanto, várias relações conflitivas e impactos socioambientais dificultam a gestão e a conservação dos recursos naturais no PNB, como regularização fundiária, sobreposição com outras Unidades de Conservação, especulação imobiliária no PNB e entorno, ocupações irregulares, pressão urbana, entre outros. Quanto aos impactos, destacam-se a disposição inadequada de resíduos sólidos e efluentes domésticos, queimadas e incêndios florestais, desmatamento, áreas degradadas, processos erosivos, assoreamento dos cursos d’água e da Represa Santa Maria, presença de espécies exóticas, caça e pesca clandestinas, proximidade do Lixão da Estrutural, entre outros. Esses impactos afetam os recursos naturais fundamentais para a população, como a água. É urgente traçar estratégias a partir da Educação Ambiental, priorizando a gestão participativa para a mediação das relações conflitivas e a redução dos impactos no PNB e em seu entorno.

Abstract

ABSTRACT Pseudomonas aeruginosa, a pathogen capable of causing diseases ranging from mild to life-threatening, has a large arsenal of virulence factors. Notably, extracellular vesicles have emerged as significant players in the pathogenesis of this organism. However, the full range of their functions is still being studied, and difficulties related to vesicle purification (long protocols, low yields, and specialized instruments) have become a major obstacle for their characterization. In this context, the utility of rapid new methods of vesicle isolation from clinical strains is still unknown. Here, we analyze the utility of the ExoBacteria OMV isolation kit for a collection of clinical strains of P. aeruginosa. We first phenotypically characterized 15 P. aeruginosa strains to ensure that our samples were heterogeneous. We then determined the best conditions for purifying vesicles from P. aeruginosa PAO1 reference strain by the rapid method and used them to isolate vesicles from clinical strains. Our results indicated that M9 minimal medium is the best for obtaining high purity with the rapid isolation kit. Although we were able to isolate vesicles from at least four strains, the low yield and the large number of strains with unpurifiable vesicles showed that the kit was not practical or convenient for clinical strains. Our findings suggest that although fast procedures for vesicle purification can be of great utility for Escherichia coli, the more complex phenotypes of clinical isolates of P. aeruginosa are a challenge for these protocols and new alternatives/optimizations need to be developed.IMPORTANCEPseudomonas aeruginosa is recognized as an opportunistic pathogen in humans and animals. It can effectively colonize various environments thanks to a large set of virulence factors that include extracellular vesicles. Different methods were recently developed to reduce the time and effort associated with vesicle purification. However, the utility of rapid vesicle isolation methods for clinical strains of P. aeruginosa (which are recognized as being highly diverse) is not yet known. In this context, we analyzed the utility of the ExoBacteria OMV Isolation kit for vesicle purification in P. aeruginosa clinical strains. Our findings showed that the kit does not seem to be convenient for research on clinical strains due to low vesicle recovery. Our results underscore the importance of developing new rapid vesicle purification protocols/techniques for specific clinical phenotypes.

Abstract

Objectives: This systematic review with meta-analysis aimed to evaluate the effects of elastic band training (EBT) on body composition and physical performance in apparently healthy older people. Methods: A systematic literature search was conducted between October 2023 and May 2024 using the core collection of six generic databases: PubMed, ProQuest, EBSCOhost, CINAHL Complete, Scopus, and Web of Science. The PRISMA, TESTEX, RoB 2, and GRADE tools assessed the evidence's methodological quality and certainty. The protocol was registered in PROSPERO (code: CRD42024547050). Results: Of 5916 records, 9 randomized and non-randomized controlled trials involving 477 healthy older people were included. Six meta-analyses were performed showing significant improvements in 30-second chair stand (SMD = 3.03; 95 % CI = 0.14 to 5.93; I2 = 100 %; p = 0.04), sit-and-reach (SMD = 2.09; 95 % CI = 0.15 to 4.03; I2 = 100 %; p = 0.04) and timed up-and-go (SMD = 3.10; 95 % CI = 1.67 to 4.53; I2 = 98 %; p 0.05) in favor of EBT were reported. Conclusion: EBT improves 30-second chair stand, sit-and-reach, and timed up-and-go in older people. Nevertheless, the certainty of evidence is very low; thus, not definitive recommendations can be made.

Abstract

Nutraceuticals have been described as phytocomplexes when derived from foods of plant origin or a pool of secondary metabolites when derived from foods of animal origin, which are concentrated and administered in an appropriate form and can promote beneficial health effects in the prevention/treatment of diseases. Considering that pharmaceutical medications can cause side effects, there is a growing interest in using nutraceuticals as an adjuvant therapeutic tool for several disorders involving autonomic dysfunction, such as obesity, atherosclerosis and other cardiometabolic diseases. This review summarizes and discusses the evidence from the literature on the effects of various nutraceuticals on autonomic control, addressing the gut microbiota modulation, production of secondary metabolites from bioactive compounds, and improvement of physical and chemical properties of cell membranes. Additionally, the safety of nutraceuticals and prospects are discussed. Probiotics, resveratrol, quercetin, curcumin, nitrate, inositol, L-carnosine, and n-3 polyunsaturated fatty acids (n-3 PUFAs) are among the nutraceuticals most studied to improve autonomic dysfunction in experimental animal models and clinical trials. Further human studies are needed to elucidate the effects of nutraceuticals formulated of multitarget compounds and their underlying mechanisms of action, which could benefit conditions involving autonomic dysfunction.

Abstract: This study aimed to develop and validate an instrument to analyze adults’ knowledge about metabolic syndrome. The instrument was developed via documentary research, resulting in 40 statements with Likert-type responses, organized into three domains: (1) knowledge about obesity and related diseases; (2) knowledge about risk factors; and (3) knowledge about protective factors. Experts’ analysis of the construct and testing on 600 participants were used to validate the instrument. Exploratory factor analysis indicated that six assertions had very low factor loadings and were excluded, resulting in 34 assertions. The final version of the instrument presented satisfactory internal consistency indicators, guaranteeing the first evidence of the instrument accuracy in assessing adults’ knowledge about metabolic syndrome.

Como objetivo secundário, analisamos as áreas de secção transversa do músculo esquelético. Abstract

Background Preeclampsia is associated with increased cardiovascular morbidity and death. Primary care or cardiology follow‐up, in complement to routine postpartum obstetric care, provides an essential opportunity to address cardiovascular risk. Prior studies investigating racial differences in the recommended postpartum follow‐up have incompletely assessed the influence of social factors. We hypothesized that racial and ethnic differences in follow‐up with a primary care provider or cardiologist would be modified by income and education. Methods and Results We identified adult individuals with preeclampsia (September 2014 to September 2019) in a national administrative database. We compared occurrence of a postpartum visit with a primary care provider or cardiologist within 1 year after delivery by race and ethnicity using multivariable logistic regression models. We examined whether education or income modified the association between race and ethnicity and the likelihood of follow‐up. Of 18 050 individuals with preeclampsia (aged 31.8±5.7 years), Black individuals (11.7%) had lower odds of primary care provider or cardiology follow‐up within 1 year after delivery compared with White individuals (adjusted odds ratio, 0.77 [95% CI, 0.70–0.85]) as did Hispanic individuals (14.8%; adjusted odds ratio, 0.79 [95% CI, 0.73–0.87]). Black and Hispanic individuals with higher educational attainment were more likely to have follow‐up than those with lower educational attainment (P for interaction=0.033) as did those in higher income brackets (P for interaction=0.006). Conclusions We identified racial and ethnic differences in primary care or cardiology follow‐up in the year postpartum among individuals diagnosed with preeclampsia, a disparity that may be modified by social factors. Enhanced system‐level interventions are needed to reduce barriers to follow‐up care.

47. Prognostic role of haemodynamics at follow-up in patients with pulmonary arterial hypertension: a challenge to current European Society of Cardiology/European Respiratory Society risk tools

Abstract

Background Haemodynamic variables like right atrial pressure (RAP), cardiac index (CI), stroke volume index (SVI) and mixed venous oxygen saturation (SvO2) predict survival in patients with pulmonary arterial hypertension (PAH). However, there is the need to identify further prognostic haemodynamic parameters as well as to redefine their role in PAH risk stratification compared to current risk tools and non-invasive parameters. Methods This cohort study includes treatment-naïve patients assessed at baseline and after first-line PAH therapy with clinical, functional, exercise, laboratory and haemodynamic evaluations. Using a stepwise multivariate Cox regression analysis, independent prognostic haemodynamic parameters were identified and stratified according to cut-offs already defined in the European Society of Cardiology (ESC)/European Respiratory Society (ERS) risk table or defined based on the highest Chi-squared of the log-rank test. Their discriminatory power was tested for all-cause death and a combined end-point of death, hospitalisation and need of treatment escalation. Results 794 patients with PAH were enrolled. At first follow-up, RAP and pulmonary artery elastance were independently associated with death. Because of high correlations between haemodynamic parameters, different multivariable analyses were done identifying six other variables (pulmonary arterial compliance, cardiac efficiency, pulmonary vascular resistance, SvO2, CI and SVI). Haemodynamic parameters were of no added prognostic value compared to ESC/ERS risk tools for the all-cause death end-point but they showed additional value to non-invasive parameters for the combined end-point and, when taken alone, had a discriminatory capacity comparable to ESC/ERS risk tools. Conclusion Haemodynamics’ discriminative ability for clinical worsening is comparable to current ESC/ERS risk tools and is of added value to non-invasive parameters.

Abstract

Barbershops and beauty salons provide community-specific opportunities to engage in cardiovascular disease screening and prevention. This editorial articulates the advantages of what is termed the “barbershop paradigm,” the community-engaged endeavor that leverages familiarity, trust, and stakeholder engagement to advance cardiovascular health. The authors summarize the neighborhood-based factors that contribute to cardiovascular health, and then identify the strategies implemented by ShopTalk and their specific advantages.
