BackgroundThe inappropriate use of prescription medication has recently garnered worldwide attention, but most national policies do not effectively provide for early detection or timely intervention. ObjectiveThis study aimed to develop and assess the validity of a model that can detect the inappropriate use of prescription medication. This effort combines a multiview and topic matching method. The study also assessed the validity of this approach. MethodsA multiview extension of the latent Dirichlet allocation algorithm for topic modeling was chosen to generate diagnosis-medication topics, with data obtained from the Chinese Monitoring Network for Rational Use of Drugs (CMNRUD) database. Topic mapping allowed for calculating the degree to which diagnoses and medications were similarly distributed and, by setting a threshold, for identifying prescription misuse. The Beijing Regional Prescription Review Database (BRPRD) database was used as the gold standard to assess the model’s validity. We also conducted a sensitivity analysis using random samples of validated prescriptions and evaluated the model’s performance. ResultsA total of 44 million prescriptions were used to generate topics using the diagnoses and medications from the CMNRUD database. A random sample (15,000 prescriptions) from the BRPRD was used for validation, and it was found that the model had a sensitivity of 81.8%, specificity of 47.4%, positive-predictive value of 14.5%, and negative-predictive value of 96.0%. The model showed superior stability under different sampling proportions. ConclusionsA method that combines multiview topic modeling and topic matching can detect the inappropriate use of prescription medication. This model, which has mediocre specificity and moderate sensitivity, can be used as a primary screening tool and will likely complement and improve the process of manually reviewing prescriptions.

Detecting circulating tumor cells has exhibited great significance in treating cancers since its concentration is an index strongly associated with the development and transfer of the tumor. However, the present commercial method for CTC detection is still expensive, because special antibodies and complicated devices must be used for cell separation and imaging. Hence, it is quite necessary to apply alternative materials and methods to decrease the cost of CTC detection. In this article, we coated a cellulose acetate membrane with nanoparticles formed by the polymerization of melamine and furfural, creating a surface with nanoscale roughness for the highly efficient capture of the sparse CTCs in a blood sample. Subsequently, the CTCs on the surface can be quantitatively detected by colorimetry with the aid of a COF-based nanozyme. The detection limit (LOD) can be as low as 3 cells/mL, which is the lowest LOD among the colorimetric methods to our knowledge. Considering the low cost of fabricating the membrane for CTC capture and the robustness of nanozymes compared with natural enzymes, this CTC detection approach displays great potential to decrease the financial burden of commercial CTC detection. In this study, a new strain of bacteria, named Rhodococcus sp. KLW-1, was isolated from farmland soil contaminated by plastic mulch for more than 30 years. To improve the application performance of free bacteria and find more ways to use waste biochar, KLW-1 was immobilised on waste biochar by sodium alginate embedding method to prepare immobilised pellet. Response Surface Method (RSM) predicted that under optimal conditions (3% sodium alginate, 2% biochar and 4% CaCl2), di (2-ethylhexyl) phthalate (DEHP) degradation efficiency of 90.48% can be achieved. Under the adverse environmental conditions of pH 5 and 9, immobilisation increased the degradation efficiency of 100 mg/L DEHP by 16.42% and 11.48% respectively, and under the high-stress condition of 500 mg/L DEHP concentration, immobilisation increased the degradation efficiency from 71.52% to 91.56%, making the immobilised pellets have strong stability and impact load resistance to environmental stress. In addition, immobilisation also enhanced the degradation efficiency of several phthalate esters (PAEs) widely existing in the environment. After four cycles of utilisation, the immobilised particles maintained stable degradation efficiency for different PAEs. Hydrogen is garnering growing attention as a green energy source with zero carbon emissions. However, most hydrogen production technologies still rely on the consumption of fossil fuels and are therefore unsustainable. This has driven the search for more environmentally friendly methods of hydrogen production. In this work, we present an innovative approach to enhance hydrogen generation via electrostatic interaction in the Escherichia coli and defective titanium dioxide (TiO2−x) biohybrids. Our method involves narrowing the forbidden bandwidth of TiO2 while introducing defect bands into its conduction band to facilitate visible light absorption and efficient charge separation. This biohybrid system, consisting of E. coli and TiO2−x, demonstrates a remarkable capability to produce 1.25 mmol of hydrogen within a 3-h timeframe under visible light irradiation. This accomplishment signifies a 3.31-fold rise in hydrogen production in comparison to E. coli, signifying a substantial enhancement in hydrogen production efficiency. Furthermore, we delve into the alterations in biological metabolites associated with hydrogen production and the changes in electron transfer in different biohybrid systems. It provides valuable insights into the understanding of the intrinsic mechanisms that drive the process. This work introduces a novel and promising avenue for achieving this exciting goal. Detection of pathogens is one of the key concerns for hospitals, the food industry, water suppliers, or other environmental engineering practices because pathogens can cause a wide range of infectious risks. Staphylococcus aureus (S. aureus) is one of the most common pathogens that are hazardous to human health and its existence is an important index to the safety of food, environmental sanitation, or medical products. In this study, we prepared an electrode with designed surface multilevel 3D micro/nano protrusions for facile and efficient S. aureus detection. The existence of these multilevel protrusions enhanced the adsorption of S. aureus. Hence, the detection limit could be as low as 10 CFU mL−1. Furthermore, the electrode was also successfully used to detect S. aureus in actual samples, such as milk and artificial human tissue fluid. It was found that the recovery of the reported approach showed no significant difference from that of the traditional plate count method. However, compared with the plate count method, the detection process of our approach is much more time-saving and easy-operating. These advantages of the approach we report, such as high sensitivity, reliability, quickness, and user-friendliness, make it a potential platform for detecting S. aureus in relation to the food industry and clinical diagnosis. Medical titanium-based (Ti-based) implants in the human body are prone to infection by pathogenic bacteria, leading to implantation failure. Constructing antibacterial nanocoatings on Ti-based implants is one of the most effective strategies to solve bacterial contamination. However, single antibacterial function was not sufficient to efficiently kill bacteria, and it is necessary to develop multifunctional antibacterial methods. This study modifies medical Ti foils with Cu-doped Co3O4 rich in oxygen vacancies, and improves their biocompatibility by polydopamine (PDA/Cu-Ov-Co3O4). Under near-infrared (NIR) irradiation, nanocoatings can generate •OH and 1O2 due to Cu+ Fenton-like activity and a photodynamic effect of Cu-Ov-Co3O4, and the total reactive oxygen species (ROS) content inside bacteria significantly increases, causing oxidative stress of bacteria. Further experiments prove that the photothermal process enhances the bacterial membrane permeability, allowing the invasion of ROS and metal ions, as well as the protein leakage. Moreover, PDA/Cu-Ov-Co3O4 can downregulate ATP levels and further reduce bacterial metabolic activity after irradiation. This coating exhibits sterilization ability against both Escherichia coli and Staphylococcus aureus with an antibacterial rate of ca. 100%, significantly higher than that of bare medical Ti foils (ca. 0%). Therefore, multifunctional synergistic antibacterial nanocoating will be a promising strategy for preventing bacterial contamination on medical Ti-based implants. [ABSTRACT FROM AUTHOR]

Cancer cells typically display redox imbalance compared with normal cells due to increased metabolic rate, accumulated mitochondrial dysfunction, elevated cell signaling, and accelerated peroxisomal activities. This redox imbalance may regulate gene expression, alter protein stability, and modulate existing cellular programs, resulting in inefficient treatment modalities. Therapeutic strategies targeting intra‐ or extracellular redox states of cancer cells at varying state of progression may trigger programmed cell death if exceeded a certain threshold, enabling therapeutic selectivity and overcoming cancer resistance to radiotherapy and chemotherapy. Nanotechnology provides new opportunities for modulating redox state in cancer cells due to their excellent designability and high reactivity. Various nanomaterials are widely researched to enhance highly reactive substances (free radicals) production, disrupt the endogenous antioxidant defense systems, or both. Here, the physiological features of redox imbalance in cancer cells are described and the challenges in modulating redox state in cancer cells are illustrated. Then, nanomaterials that regulate redox imbalance are classified and elaborated upon based on their ability to target redox regulations. Finally, the future perspectives in this field are proposed. It is hoped this review provides guidance for the design of nanomaterials‐based approaches involving modulating intra‐ or extracellular redox states for cancer therapy, especially for cancers resistant to radiotherapy or chemotherapy, etc. [ABSTRACT FROM AUTHOR]

Microbial fuel cell (MFC) performance is affected by the metabolic activity of bacteria and the extracellular electron transfer (EET) process. The deficiency of nanostructures on macroporous anode obstructs the enrichment of exoelectrogens and the EET. Herein, a N-doped carbon nanowire-modified macroporous carbon foam was prepared and served as an anode in MFCs. The anode has a hierarchical porous structure, which can solve the problem of biofilm blockage, ensure mass transport, favor exoelectrogen enrichment, and enhance the metabolic activity of bacteria. The microscopic morphology, spectroscopy, and electrochemical characterization of the anode confirm that carbon nanowires can penetrate biofilm, decrease charge resistance, and enhance long-distance electron transfer efficiency. In addition, pyrrolic N can effectively reduce the binding energy and electron transfer distance of bacterial outer membrane hemin. With this hierarchical anode, a maximum power density of 5.32 W/m3 was obtained, about 2.5-fold that of bare carbon cloth. The one-dimensional nanomaterial-modified macroporous anodes in this study are a promising strategy to improve the exoelectrogen enrichment and EET for MFCs. [ABSTRACT FROM AUTHOR]

In response to the trend of drug−resistant and super bacteria, the existing single antibacterial methods are not sufficient to kill bacteria, and the development of multifunctional antibacterial nanomaterials is urgent. Our study aims to construct copper−doped polydopamine−coated Ti3C2Tx (CuPDA@Ti3C2Tx) with an enhanced photothermal property and Fenton−like activity. The nanocomposite hydrogel consisting of CuPDA@Ti3C2Tx and alginate can improve the antioxidant activity of two−dimensional MXene nanosheets by coating them with a thin layer of PDA nanofilm. Meanwhile, Cu ions are adsorbed through the coordination of PDA−rich oxygen−containing functional groups and amino groups. Calcium ions were further used to crosslink sodium alginate to obtain antibacterial hydrogel materials with combined chemotherapy and photothermal therapy properties. The photothermal conversion efficiency of CuPDA@Ti3C2Tx is as high as 57.7% and the antibacterial rate of Escherichia coli reaches 96.12%. The photothermal effect leads to oxidative stress in bacteria, increases cell membrane permeability, and a high amount of ROS and copper ions enter the interior of the bacteria, causing protein denaturation and DNA damage, synergistically leading to bacterial death. Our study involves a multifunctional synergistic antibacterial nanodrug platform, which is conducive to the development of high−performance antibacterial agents and provides important research ideas for solving the problem of drug−resistant bacteria. Comprehensive Summary: Fuel‐driven dissipative self‐assembly, which is a well‐established concept in recent years, refers to out‐of‐equilibrium molecular self‐assembly initiated and supported by the addition of active molecules (chemical fuel). It widely exists in nature since many temporary, active micro‐ or nanostructures in living bodies are generated by the dissipative self‐assembly of biomolecules. Therefore, the study on dissipative self‐assembly provides a good opportunity to have an insight into the microscopic mechanism of living organisms. In the meantime, dissipative assembly is thought to be a potential pathway to achieve dynamic, temporary supramolecular materials. Recently, a number of temporary materials have been developed with the aid of strategies for realizing dissipative self‐assembly. Some of their properties, including solubility, stiffness, turbidity, color, or self‐healing ability, change upon the addition of chemical fuel but spontaneously restore with chemical fuel consumption. The dynamic of these materials brings them various unprecedented functions. In this review, the principles of fabricating a fuel‐driven temporary material are first reviewed. Subsequently, recent examples of fuel‐driven temporary materials are emphatically summarized, including gels, self‐erased inks, nanoreactors, self‐healing materials, nanochannels, and droplets. Finally, the challenges of developing fuel‐driven temporary materials and some perspectives on the function and application of such kind of materials are discussed. [ABSTRACT FROM AUTHOR]

Glaucoma has become the world's leading cause of irreversible blindness, and one of its main characteristics is high intraocular pressure. Currently, the non-surgical drug treatment scheme to reduce intraocular pressure is a priority method for glaucoma treatment. However, the complex and special structure of the eye poses significant challenges to the treatment effect and safety adherence of this drug treatment approach. To address these challenges, the application of polymer-based self-assembled drug delivery systems in glaucoma treatment has emerged. This review focuses on the utilization of polymer-based self-assembled structures or materials as important functional and intelligent carriers for drug delivery in glaucoma treatment. Various drug delivery systems, such as eye drops, hydrogels, and contact lenses, are discussed. Additionally, the review primarily summarizes the design strategies and methods used to enhance the treatment effect and safety compliance of these polymer-based drug delivery systems. Finally, the discussion delves into the new challenges and prospects of employing polymer-based self-assembled drug delivery systems for the treatment of glaucoma. [ABSTRACT FROM AUTHOR]

Optical imaging has a wide range of applications in the biomedical field, allowing the visualization of physiological processes and helping in the diagnosis and treatment of diseases. Unexcited light source imaging technologies, such as chemiluminescence imaging, bioluminescence imaging and afterglow imaging have attracted great attention in recent years because of the absence of excitation light interference in their application and the advantages of high sensitivity and high signal‐to‐noise ratio. In this review, the latest advances in unexcited light source imaging technology for biomedical applications are highlighted. The design strategies of unexcited light source luminescent probes in improving luminescence brightness, penetration depth, quantum yield and targeting, and their applications in inflammation imaging, tumor imaging, liver and kidney injury imaging and bacterial infection imaging are introduced in detail.
