Tianjin has a long history of land subsidence, which has gone through six different stages since 1923. Although as of 2020, the land subsidence in a large area of Tianjin has been mostly controlled, there are still serious subsidence areas with an annual subsidence of over 50mm. From large-scale treatment to precise prevention and control of small areas, the distribution characteristics of ground subsidence in Tianjin have shown a new situation, and the prevention and control of land subsidence is also facing new requirements. To accurately grasp the development law of ground subsidence and implement precise policies under the new situation, this paper summarizes the distribution characteristics and evolution rules of severe land subsidence areas in Tianjin by collecting and analyzing the data of ground subsidence leveling, groundwater level, and groundwater extraction in Tianjin from 2010 to 2020. The results show that the land subsidence of Tianjin has experienced three periods during the ten years from 2010 to 2020: a fluctuation period in subsidence from 2010 to 2012, a steady improvement period from 2013 to 2016, and a rapid slowdown period from 2017 to 2020. During these periods, the average amount of land subsidence dropped by 37%, and the area of severe subsidence decreased by 67%. The changes in subsidence were closely related to the amount of groundwater extraction. As of 2020, five severe subsidence areas are concentrated in the southwest of Tianjin, with a distribution range that is similar to the groundwater funnel of the deep water-bearing group.

In permafrost regions with warm frozen soil, the pile foundation is commonly used, but most currently available models for the WFS foundation pile–soil system are either highly empirical or overcomplicated, without a simplified theoretical manner in engineering. This study derives a novel and simplified calculated model of the WFS pile–soil system. The model is formulated in terms of the shear deformation theory and load transfer method based on the rigorous deformation mechanism of the WFS foundation soil around the pile. Considering the different depth soil features and the equilibrium state of the pile–soil system, dividing warm frozen soil foundation into three regions (TPPR, ER, and BPPR) to calculate the Dp and Ds can simply obtain the total displacement of pile under different loads. The results demonstrate that the present theoretical model can well predict the WFS foundation load–displacement response of the pile. The present model provides a simple, practical, and effective approach for the estimation of the load–displacement behavior of piles installed in the WFS foundation.

In the long-term evolutionary process, Achnatherum inebrians and seed-borne endophytic fungi, Epichloë gansuensis, formed a mutually beneficial symbiosis relationship, and Epichloë gansuensis has an important biological role in improving the tolerance of host grasses to abiotic stress. In this work, we first assessed the effects of Epichloë gansuensis on dry weight, the content of C, N, P and metal ions, and metabolic pathway of amino acids, and phosphorus utilization efficiency (PUE) of Achnatherum inebrians at low P stress. Our results showed that the dry weights, the content of alanine, arginine, aspartic acid, glycine, glutamine, glutamic acid, L-asparagine, lysine, phenylalanine, proline, serine, threonine, and tryptophan were higher in leaves of Epichloë gansuensis-infected (E+) Achnatherum inebrians than Epichloë gansuensis-uninfected (E−) Achnatherum inebrians at low P stress. Further, low P stress increased C content of leaves of E+ Achnatherum inebrians compared to 0.5 mM P; Epichloë gansuensis increased K content of leaves compared to the leaf of E− plant at 0.01 mM P and 0.5 mM P. Epichloë gansuensis reduced Ca content of roots compared to the root of E− plant at 0.01 mM P and 0.5 mM P; Epichloë gansuensis reduced the content of Mg and Fe in leaves compared to the leaf of E− plant at 0.01 mM P and 0.5 mM P. In addition, at low P stress, Epichloë gansuensis most probably influenced aspartate and glutamate metabolism; valine, leucine, and isoleucine biosynthesis in leaves; and arginine and proline metabolism; alanine, aspartate, and glutamate metabolism in roots. Epichloë gansuensis also affected the content of organic acid and stress-related metabolites at low P stress. In conclusion, Epichloë gansuensis improves Achnatherum inebrians growth at low P stress by regulating the metabolic pathway of amino acids, amino acids content, organic acid content, and increasing PUE.

Nuclear factor kappa B (NF-κB), a key nuclear transcription factor, is associated with prognosis in a variety of human cancers. However, the clinical value of NF-κB in non-small cell lung cancer (NSCLC) is still controversial. Therefore, the aim of this meta-analysis was to obtain an accurate evaluation of the relationship between NF-κB expression and survival prognosis of NSCLC patients based on published articles. PubMed, EMBASE and Web of Science databases were systematically searched for potential articles. A total of 1159 patients from 7 eligible studies comparing prognostic significance of NF-κB expression levels in NSCLC were included in our meta-analysis. I2 statistic and P value were performed to evaluate heterogeneity. The results of analysis were presented as hazard ratio (HR) or odds ratios (OR) with 95% confidence interval (95% CI). Subgroup analysis based on ethnicity of NSCLC patients and NF-kB cellular localization within cancer cells were conducted to illustrate the potential discrepancy. Improving the energy efficiency of data center has become a research focus in recent years. Previous works commonly adopted the inlet temperature constraint to optimize the thermal environment in the data center. However, the inlet temperature-aware method cannot prevent the servers from over-cooling. To cope with this issue, we propose a thermal-aware workload allocation strategy with respect to the chip temperature constraint. In this paper, we conducted a comparative evaluation of the performance between the chip and inlet temperature-aware workload allocation strategies. The workload allocation strategies adopt a POD-based heat recirculation model to characterize the thermal environment in data center. The contribution of the temperature-dependent leakage power to server power consumption is also considered. We adopted a sample data center under constant-flow and variable-flow cooling air supply to evaluate the performance of these two different workload allocation strategies. The comparison results show that the chip temperature-aware workload allocation strategy prevents the servers from over-cooling and significantly improves the energy efficiency of data center, especially for the case of variable-flow cooling air supply.

In China, HIV-1-infected patients typically receive antiretroviral therapy (ART) that includes lamivudine (3TC) as a reverse-transcriptase inhibitor (RTI) (ART-3TC). Previous studies from certain developed countries have shown that, in ART-3TC, 3TC-resistant HBV progressively emerges at an annual rate of 15-20% in patients coinfected with HIV-1 and HBV. This scenario in China warrants investigation because >10% of all HIV-infected patients in China are HBV carriers. We measured the occurrence of 3TC-resistant HBV during ART-3TC for HIV-HBV coinfection and also tested the effect of tenofovir disoproxil fumarate (TDF) used as an additional RTI (ART-3TC/TDF) in a cohort study in China. We obtained 200 plasma samples collected from 50 Chinese patients coinfected with HIV-1 and HBV (positive for hepatitis B surface antigen) and examined them for the prevalence of 3TC-resistant HBV by directly sequencing PCR products that covered the HBV reverse-transcriptase gene. We divided the patients into ART-3TC and ART-3TC/TDF groups and compared the efficacy of treatment and incidence of drug-resistance mutation between the groups. HIV RNA and HBV DNA loads drastically decreased in both ART-3TC and ART-3TC/TDF groups. In the ART-3TC group, HBV breakthrough or insufficient suppression of HBV DNA loads was observed in 20% (10/50) of the patients after 96-week treatment, and 8 of these patients harbored 3TC-resistant mutants. By contrast, neither HBV breakthrough nor treatment failure was recorded in the ART-3TC/TDF group. All of the 3TC-resistant HBV mutants emerged from the cases in which HBV DNA loads were high at baseline. Our results clearly demonstrated that ART-3TC is associated with the emergence of 3TC-resistant HBV in patients coinfected with HIV-1 and HBV and that ART-3TC/TDF reduces HBV DNA loads to an undetectable level. These findings support the use of TDF-based treatment regimens for patients coinfected with HIV-1 and HBV.

Minimizing the energy consumption is a dominant problem in data center design and operation. To cope with this issue, the common approach is to optimize the data center layout and the workload distribution among servers. Previous works have mainly adopted the temperature at the server inlet as the optimization constraint. However, the inlet temperature does not properly characterize the server’s thermal state. In this paper, a chip temperature-based workload allocation strategy (CTWA-MTP) is proposed to reduce the holistic power consumption in data centers. Our method adopts an abstract heat-flow model to describe the thermal environment in data centers and uses a thermal resistance model to describe the convective heat transfer of the server. The core optimizes the workload allocation with respect to the chip temperature threshold. In addition, the temperature-dependent leakage power of the server has been considered in our model. The proposed method is described as a constrained nonlinear optimization problem to find the optimal solution by a genetic algorithm (GA). We applied the method to a sample data center constructed with computational fluid dynamics (CFD) software. By comparing the simulation results with other different workload allocation strategies, the proposed method prevents the servers from overcooling and achieves a substantial energy saving by optimizing the workload allocation in an air-cooled data center.

BACKGROUND: Drug resistance (DR) of HIV-1 can be examined genotypically or phenotypically. Although sequencing is the gold standard of the genotypic resistance testing (GRT), high-throughput GRT targeted to the codons responsible for DR may be more appropriate for epidemiological studies and public health research. METHODS: We used a Japanese database to design and synthesize sequence-specific oligonucleotide probes (SSOP) for the detection of wild-type sequences and 6 DR mutations in the clade B HIV-1 reverse transcriptase region. We coupled SSOP to microbeads of the Luminex 100 xMAP system and developed a GRT based on the polymerase chain reaction (PCR)-SSOP-Luminex method. RESULTS: Sixteen oligoprobes for discriminating DR mutations from wild-type sequences at 6 loci were designed and synthesized, and their sensitivity and specificity were confirmed using isogenic plasmids. The PCR-SSOP-Luminex DR assay was then compared to direct sequencing using 74 plasma specimens from treatment-naïve patients or those on failing treatment. In the majority of specimens, the results of the PCR-SSOP-Luminex DR assay were concordant with sequencing results: 62/74 (83.8%) for M41, 43/74 (58.1%) for K65, 70/74 (94.6%) for K70, 55/73 (75.3%) for K103, 63/73 (86.3%) for M184 and 68/73 (93.2%) for T215. There were a number of specimens without any positive signals, especially for K65. The nucleotide position of A2723G, A2747G and C2750T were frequent polymorphisms for the wild-type amino acids K65, K66 and D67, respectively, and 14 specimens had the D67N mutation encoded by G2748A. We synthesized 14 additional oligoprobes for K65, and the sensitivity for K65 loci improved from 43/74 (58.1%) to 68/74 (91.9%). CONCLUSIONS: We developed a rapid high-throughput assay for clade B HIV-1 DR mutations, which could be customized by synthesizing oligoprobes suitable for the circulating viruses. The assay could be a useful tool especially for public health research in both resource-rich and resource-limited settings.

Objective The cortical bone trajectory (CBT) technology is an effective substitute for traditional pedicle screw (PS) technology. However, there is still controversy about the CBT screw technology placement strategy. The objective of this study was to simulate cortical screw placement with the help of three‐dimensional (3D) software, to discuss the differences in screws between genders and vertebral segments, and to explore a safer and more efficient strategy for cortical screw placement. Methods Mimics Medical software was used to construct a 3D model of the lumbar spine, and the placement of CBT screws was simulated. The volume of each vertebral body from L1 to L5, the pedicle isthmus height (IH), the pedicle isthmus width (IW), and the sagittal vertebral distance (SAVD) were measured. The transverse distance (TD) and the longitudinal distance (LD) between the ideal starting point (SP) and the clinical SP (the intersection Q of the midline of the superior articular process and the horizontal line 1 mm below the transverse process) were measured. The cephalad angle (CA), lateral angle (LA), maximum screw diameter (MSD), maximum screw length (MSL) of each trajectory of the L1 to L5 vertebral bodies, and the percentage of the screw insertion depth (PSID) into the vertebral body were measured. Data were statistically analyzed using Student's t‐test, one‐way analysis of variance (ANOVA), and Tukey's test. Results Vertebral anatomical parameters and CBT screw parameters differed between males and females. Female patients had lower IH, IW, SAVD, CA, LA, MSD, and MSL than males. IH was greatest in L1 (male, 17.81 mm; female, 16.12 mm) and the smallest in L5 (male, 14.11 mm; female, 13.05 mm). IW was smallest in L1 (male, 8.89 mm; female, 7.37 mm) and greatest in L5 (male, 16.59 mm; female, 15.43 mm). The MSD of males was smallest in L1 (6.05 mm) and greatest in L3 (7.06 mm); the MSD of females was smallest in L1 (5.13 mm) and greatest in L4 (6.64 mm). MSL was greatest at L3 (male, 33.63 mm; female, 32.28 mm) and smallest at L5 (male, 31.25 mm; female, 29.97 mm). CA was smallest in L1 (male, 22.80°; female, 21.92°) and greatest in L3 (male, 25.29°; female, 24.33°). LA was smallest in L1 (male 12.37°, female 11.27°) and greatest in L5 (male 13.56°, female 12.96°). Among the males, TD was smallest at L1 (−0.51 mm) and greatest at L5 (1.37 mm), while LD was greatest at L2 (3.46 mm) and smallest at L5 (2.40 mm). In females, TD was greatest at L1 (0.12 mm) and smallest at L3 (−0.51 mm), while LD was greatest at L1 (3.69 mm) and smallest at L5 (2.08 mm). In the overall sample, the incidence of SAVD and PSID gradually increased from L1 to L5. Conclusion The optimal screw placement strategy for CBT screws varies significantly according to sex and vertebral body segments, particularly noting the specificity of screw placement at L5. The optimal screw placement strategy should be selected based on the patient's sex and segment characteristics before surgery to maximize the safety and accuracy of CBT screw placement.

To solve the problem of poor thermal stability of the current D-allulose 3-epimerase (DAEase), the ancestor sequences of DAEase with different catalytic domains were reconstructed by big data mining, reasonable modification and ancestor sequence reconstruction (ASR) strategy under the guidance of phylogenetic information. The expression vectors of the ancestor sequences were constructed, and DAEase A13 with significantly enhanced thermal stability was screened by recombinant expression and molecular docking, and its enzymatic properties were characterized. In addition, the molecular mechanism of thermal stability enhancement of DAEase A13 was revealed based on structural analysis and molecular dynamics. The results showed that the half-life of A13 constructed based on ASR strategy could reach 8.4 h at 70 ℃, indicating that its thermal stability was significantly enhanced compared with that of wild-type (WT) enzyme. The maximum conversion rate of A13 reached 31%, indicating that the catalytic activity of A13 was slightly higher than that of WT enzyme. The structural and molecular dynamics analysis revealed that the increase in hydrogen bonding and hydrophobic interaction in ASR A13 was the main factor responsible for maintaining the stability of the enzyme's molecular structure at high temperatures. The results showed that ASR strategy could modify DAEases to enhance the stability, activity or hybridity, which could provide superior biocatalyst sources for various industrial applications of functional sugars.

Abstract Glaesserella parasuis (GPS) is an important bacterial pathogen of swine. Serotype identification has presented a bottleneck in GPS research since it was first identified as the pathogen causing Glässer’s disease in pigs in 1910. This paper presents a systematic review of the history of the development and application of gel immunodiffusion (GID), indirect hemagglutination assay (IHA), and polymerase chain reaction (PCR) typing methods for GPS, and the discovery of their shared antigenic basis. It provides a systematic theoretical overview of the immunology and principles underlying the three typing methods and offers new ideas for research into the prevention and control of Glässer’s disease. In 1992, GPS was first classified into serotypes 1–15 using GID based on GPS heat-stable antigens, but about 25% of the strains were found to be non-typeable, and the composition of their antigens for serotyping was unclear. In 2003, the IHA method was established based on saline-extracted antigens of GPS, whose sensitivity and typing rate were higher than for GID, although about 15% of strains were still found to be non-typeable. The results of IHA and GID typing are roughly consistent, since they share the same GPS surface polysaccharide serotyping antigens, although whether these are capsular polysaccharides, lipopolysaccharides, or other polysaccharides, remains to be determined. In 2013, the Capsular polysaccharide (CPS) synthetic gene clusters from GPS serotypes 1–15 were successfully analyzed, confirming that CPS is essential for the formation of antigens for serotyping. In 2015, primers were designed based on the specific target genes of GPS capsules to establish a PCR typing method (H-PCR) for GPS, which, however, could not identify serotypes 5 and 12. In 2017, a new PCR typing method (J-PCR) was established based on the specific target genes of GPS capsules, which could identify serotypes 5 and 12. A combination of the two PCR typing methods enables the typing of almost all GPS strains, and the consistency with GID and IHA was verified using molecular biological methods. The antigenic basis of the three typing methods was shown to involve the GPS capsule. PCR typing methods are characterized by simple operation, fast speed, and low cost, and can successfully solve many problems in GID and IHA serotyping, and so have become widely adopted.

Abstract Objectives Helping residents select nutrient-dense foods is a strategy to improve their diet quality. However, communication based on the nutrient-dense foods as a positive attribute has not been widely used in nutritional education. This study aimed to develop an educational tool based on the picture and guidance of “Chinese food guide pagoda (2022) “, extend it with the concept of nutrient density, and investigate its acceptance by Chinese residents from Henan province. Methods Three examples (one-day diet with high, medium, and low nutrient-rich food (NRF) 9.2 score, an indicator for evaluating dietary nutrient density) were designed for developing a dietary nutrient density educational tool. A self-designed questionnaire was conducted to investigate the acceptance of the “dietary nutrient density educational tool” among college students from Henan province on the basis of the theory of planned behavior. Results Among the three one-day diets used in the tool, with the decrease in the NRF9.2 score, the energy intake increased from 1686 kcal to 2363 kcal, the dietary fat-to-energy ratio increased from 28 to 42%, and the mean adequacy ratio (MAR) decreased from 0.97 to 0.87. A total of 851 college students completed the acceptance questionnaire. The average score of the acceptance was 4.07, with a total score of 5. This study showed that resident’s intention to use the tool was correlated with family residence, perceptual behavior control, and subjective norms. These three factors accounted for 83.5% of the variation in behavior intention. Conclusion To encourage residents choosing healthier foods, a dietary nutrient density educational tool was developed to expanding the current instructional tool—the Chinese food guide pagoda (2022). The acceptance questionnaire survey revealed that residents had good acceptance of the tool, and family residence, perceptual behavior control, subjective norms may strongly contribute to their acceptance and the intention to use of the tool.

Background: Spinal N-methyl-D-aspartate (NMDA) receptor activation is attributed to remifentanil-induced hyperalgesia (RIH). However, the specific mechanism and subsequent treatment is still unknown. Previous studies have shown that the dynamin-related protein 1 (DRP1)-mitochondria-reactive oxygen species (ROS) pathway plays an important role in neuropathic pain. This study examined whether antisense oligodeoxynucleotides against DRP1 (AS-DRP1) could reverse RIH. Methods: The authors first measured changes in paw withdrawal mechanical threshold (PWMT) and paw withdrawal thermal latency (PWTL) at 24 hours before remifentanil infusion and 4, 8, 24, and 48 hours after infusion. The expression levels of DRP1 and NR2B were measured after behavioral testing using Western blotting. In addition, DRP1 expression was knocked down by intrathecal administration of AS-DRP1 to investigate the effects of DRP1 on RIH. The behavioral testing, the expression levels of spinal DRP1 and NR2B, and dorsal mitochondrial superoxide were measured. Changes in mitochondrial morphology were assessed using electron microscopy. Results: After remifentanil exposure, upregulation of spinal DRP1 and NR2B was observed along with a reduction in PWMT and PWTL. In addition, AS-DRP1 improved RIH-induced PWTL and PWMT (P < 0.001 and P < 0.001) and reduced remifentanil-mediated enhancement of spinal DRP1 and NR2B expression (P = 0.020 and P = 0.022). More importantly, AS-DRP1 reversed RIH-induced mitochondrial fission (P = 0.020) and mitochondrial superoxide upregulation (P = 0.031). Conclusions: These results indicate that AS-DRP1 could modulate NMDA receptor expression to prevent RIH through the DRP1-mitochondria-ROS pathway. [ABSTRACT FROM AUTHOR]

Abstract Pediatric trauma plays a crucial role in pediatric mortality, with traffic injuries and falls frequently cited as leading causes of significant injuries among children. A comprehensive investigation, including geographical factors, is essential for developing effective strategies to prevent injuries and alleviate the burden of pediatric trauma. This study involved a retrospective analysis of clinical data from pediatric patients admitted to our hospital’s intensive care unit (ICU) due to trauma over a 10-year period. Comprehensive analyses were conducted to elucidate trends, demographics, injury patterns, and risk factors associated with these admissions. This retrospective study included 951 pediatric patients (mean age: 4.79 ± 3.24 years; mean weight: 18.45 ± 9.02 kg; median time to ICU admission post-injury: 10.86 ± 14.95 h). Among these patients, 422 (44.4%) underwent emergency surgery, and 466 (49%) required mechanical ventilation support, with a mean duration of 70.19 ± 146.62 h. The mean duration of ICU stay was 6.24 ± 8.01 days, and the overall mean hospitalization duration was 16.08 ± 15.56 days. The predominant cause of unintentional injury was traffic accidents (47.9%), followed by falls (42.5%) and burns/scalds (5.3%). Most incidents involved children aged 0–6 years (70.7%), with males comprising 60.0% of patients. Injury incidents predominantly occurred between 12 and 6 PM (44.5%) and on non-workdays (37.6%). The most common locations where injuries occurred were roadsides (49%) and rural areas (64.35%). Single-site injuries (58.78%) were more prevalent than multiple-site injuries (41.22%), and head injuries were the most common among single-site injuries (81.57%). At ICU admission, the mean injury severity score was 18.49 ± 8.86. Following active intervention, 871 patients (91.59%) showed improvement, while 80 (8.41%) succumbed to their injuries. Traffic injuries remain the primary cause of pediatric trauma leading to ICU admission, underscoring the importance of using appropriate child restraint systems and protective gear as fundamental preventive measures. Abstract We propose a novel miniaturized optical attenuator based on the evanescent-field coupling between two nanofibers. Benefiting from a wavelength-dependent waveguiding property of the coupled structure, a tunable attenuation with maximum extinction ratio of ~ 20 dB is demonstrated with an ultrawide optical bandwidth up to 0.7 μm in experiment. The wavelength-depended waveguiding properties of both one single nanofiber and coupled nanofibers are investigated in both theory and simulation. Using an adiabatic coupling structure, an attenuation range from − 0.16 dB to − 18.46 dB is obtained within a spectrum from 1.2 μm to 1.7 μm experimentally. Moreover, the simulated results indicate that, the attenuation only shows a slight change of ~ 0.1 dB with a lateral misalignment of 0.5 μm between the two nanofibers, indicating a high tolerance of this attenuator to the lateral misalignment. Considering the wide bandwidth as well as the ultracompact structure, this attenuator shows high potential in applications such as all-fiber optical sensing and communicating.

Deserts are ideal places to build photovoltaic (PV) power plants, but this plants often face challenges from strong wind and sand activities during the operation and maintenance period, exploring the effects of PV power plant construction on wind disturbances and the control of wind and sand activities by different sand fixation measures is necessary. This study investigated the wind speed outside the PV plant, inside the plant without sand barriers measures (CK), and under three different sand-protecting barriers (gauze sand barriers (GZ), polylactic acid sand barriers (PLA), and grass grid sand barriers (GG)) inside the plant. Though calculated the surface roughness, friction velocity, wind protection effectiveness, and wind turbulence to determined the effectiveness of the barriers by these indexes comprehensively. The results show that: (1) The construction of desert PV power plant can effectively reduce the wind speed. Compared with CK, all three mechanical sand barriers within the plant reduced wind speed. Especially when the height less than 50cm, the GZ sand barriers reduced the wind speeds the most, with an average reduction rate of 101.5%. (2) All three sand barriers increased soil roughness and friction velocity within the power station. (3) At heights below 50cm, the GZ and GG sand barriers have better wind protection effectiveness than PLA sand barriers, while at hights above 100cm, the wind protection effect of PLA and GG sand barriers became less significant or even negligible (4) The wind disturbance caused by the three sand fixation measures increased with wind speed, the comprehensive performance of GZ and PLA sand barriers was superior than that of GG sand barriers and CK.

Abstract Experimental realization of strong coupling between a single exciton and plasmons remains challenging as it requires deterministic positioning of the single exciton and alignment of its dipole moment with the plasmonic fields. This study aims to combine the host–guest chemistry approach with the cucurbit[7]uril-mediated active self-assembly to precisely integrate a single methylene blue molecule in an Au nanodimer at the deterministic position (gap center of the nanodimer) with the maximum electric field (EFmax) and perfectly align its transition dipole moment with the EFmax, yielding a large spectral Rabi splitting of 116 meV for a single-molecule exciton—matching the analytical model and numerical simulations. Statistical analysis of vibrational spectroscopy and dark-field scattering spectra confirm the realization of the single exciton strong coupling at room temperature. Our work may suggest an approach for achieving the strong coupling between a deterministic single exciton and plasmons, contributing to the development of room-temperature single-qubit quantum devices.

In order to improve the economic value of Panax quinquefolius L., this study used Panax quinquefolius L. as raw material to solid state ferment Panax quinquefolius L. by Schizophyllum commune, increased the content of active components in Panax quinquefolius L., and optimized the fermentation technology. In this study, the total ginsenoside content of Panax quinquefolius L. was used as the indicator. First, strain 2 was selected from three strains of Streptomyces as the solid state fermentation strain for Panax quinquefolius L.. Then single factor experiment was conducted on four factors affecting total ginsenoside content. The four factors included: fermentation temperature, inoculation amount of Schizophyllum commune, pH value and fermentation time. On the basis of these results, the technology of solid state fermentation of Panax quinquefolius L. by Schizophyllum commune was further optimized by response surface Box Behnken experimental design. The optimized process conditions were determined as follows: inoculation amount of Schizophyllum commune 10.90%, fermentation temperature 31 ℃, fermentation time 10 d, pH value 5.9. Under the above conditions, the total ginsenoside content of Panax quinquefolius L. fermented by Schizophyllum commune was 10.28%, the content of crude polysaccharide was 12.85%, and the content of total flavonoids was 0.67%, which were higher than that of unfermented Panax quinquefolius L., and the contents of macromolecules ginsenoside Rb1, Rg2 and Re decreased, while the contents of micromolecules ginsenoside Rg1, Rc, Rb2, Rd, Rg3, s-Rh2, r-Rh2 and pseudo-ginsenoside CK increased. Among them, rare ginsenoside Rg3, s-Rh2, r-Rh2, and pseudo-ginsenoside CK were new saponins. In vitro antioxidant tests showed that the IC50 values of DPPH and ABTS+ free radicals scavenging ability of the fermented products of Panax quinquefolius L. by Schizophyllum commune were 3165 and 10910 μg/mL, respectively, which were stronger than those of unfermented Panax quinquefolius L.. This study proved that the solid state fermentation of Panax quinquefolius L. by Schizophyllum commune increased the contents of total ginsenoside, crude polysaccharide, total flavone and rare ginsenoside, and enhanced the antioxidant capacity, which provided data support for the development and utilization of Panax quinquefolius L..

Introduction: Trauma is a significant global public health concern and the leading cause of morbidity and mortality in children. This study aimed to assess the independent predictors of trauma severity as well as mortality in pediatric patients admitted to the intensive care unit (ICU). Methods: In this cross-sectional study, following the STROBE checklist, we retrospectively analyzed the clinical and baseline characteristics of pediatric patients with trauma injuries admitted to the ICU of Children’s Hospital of Zhejiang University School of Medicine, China, over a decade. Perilla (Perilla frutescens) seeds are emerging as a valuable resource for functional foods and medicines owing to their rich oil and protein content with diverse nutritional and health benefits. Perilla seed oil (PSO) possesses a high level of a-linolenic acid (ALA), a favorable ratio of unsaturated to saturated fatty acids, and other active ingredients such as tocopherols and phytosterols, which contribute to its antioxidant, anti-inflammatory, and cardiovascular protective effects. The balanced amino acid ratio and good functional properties of perilla seed protein make it suitable for a variety of food applications. The chemical composition, health benefits, and potential applications of PSO as well as the structural characterization, functional properties, modification methods, bioactivities, and application scenarios of perilla seed protein are comprehensively presented in this paper. Furthermore, the challenges as well as future prospects and research focus of PSO and perilla seed protein are discussed. The growing interest in plant-based diets and functional foods has made PSO and perilla seed protein promising ingredients for the development of novel foods and health products. The purpose of this paper is to highlight implications for future research and development utilizing these two untapped resources to improve human health and nutrition.

Tunable color emissions, emerging from a single CdTe nanowire, are demonstrated experimentally based on optical transverse nonlinear effects. The pumping light at different wavelengths (e.g., 1064 nm and 980 nm) is coupled to a nanowire at both ends via evanescent-field coupling. The light at different wavelengths (e.g., 510 nm, 532 nm, and 713 nm) can be simultaneously assessed using complex optical transverse nonlinear effects, including transverse sum-frequency generation (TSFG), transverse second-harmonic generation (TSHG), and two-photon absorption (TPA)-induced fluorescence. By changing the wavelength and the power of the pumping lights, the spectra of the transverse light emissions change as well, leading to tunable color emissions at the single-nanowire level with a Rec. 2020 coverage of ~21.6%. The results indicate the potential of transverse nonlinear effects in applications ranging from optical display and spectroscopy to communication.

ObjectiveAlthough extensive research has explored the link between mental disorders and asthma, the characteristics and patterns of this association are still unclear. Our study aims to examine the genetic causal links between common mental disorders (specifically, anxiety and depression) and asthma.MethodsWe conducted genetic analyses including linkage disequilibrium score regression (LDSC) and bidirectional two-sample Mendelian randomization (MR) analyses, and utilized summary statistics from recent large-scale Genome-Wide Association Studies (GWASs) in European populations, covering sensation of anxiety or depression, anxiety sensation, depression sensation, anxiety disorders, major depression disorder (MDD), and asthma.ResultsLDSC revealed significant genetic correlations among sensation of anxiety or depression, MDD and asthma (P < 0.017), highlighting potential genetic correlation between anxiety disorders and asthma (P < 0.05 yet > 0.017). In bidirectional two-sample MR, inverse-variance weighted (IVW) analyses suggested that genetic liability to asthma was significantly associated with an increased risk of sensation of anxiety or depression (OR = 4.760, 95%CI: 1.645–13.777), and MDD (OR = 1.658, 95%CI: 1.477–1.860). Conversely, IVW analyses indicated that genetic liability to anxiety disorders was not associated with an increased risk of asthma (P > 0.01), nor was genetic liability to asthma associated with an increased risk of anxiety disorders (P > 0.01). Furthermore, no significant genetic causal relationships were observed for other studied traits. Multivariate MR, after adjusting for body mass index and alcohol consumption, further corroborated the independent causal effect of genetic predisposition to MDD on the risk of asthma (OR = 1.460, 95% CI: 1.285–1.660).ConclusionOur study establishes MDD as a predisposing factor for asthma. Meanwhile, anxiety disorders are not causal risk factors for asthma, nor is the reverse true. It is recommended to closely monitor asthma symptoms in patients with MDD.

Acanthopanax senticosus (AS) is a geo-authentic crude medicinal plant that grows in China, Korea, Russia, and Japan. AS contains bioactive compounds such as eleutherosides, polysaccharides, and flavonoids. It is also a key traditional herb in the Red List of Chinese Species. AS is mainly distributed in Northeast China, specifically in Heilongjiang, Jilin, and Liaoning provinces. Its active compounds contribute to significant biological activities, including neuroprotective, antioxidant, anti-fatigue, and antitumor effects. However, the extraction methods of active compounds are complex, the extraction efficiency is poor, and the structure–activity relationship is unclear. This study focused on the nutrients in AS, including protein, carbohydrates, and lipids. Particularly, the active ingredients (eleutherosides, polysaccharides, and flavonoids) in AS and their extraction and purification methods were analyzed and summarized. The biological activities of extracts have been reviewed, and the mechanisms of anti-oxidation, antitumor, anti-inflammation, and other activities are introduced in detail. The applications of AS in various domains, such as health foods, medicines, and animal dietary supplements, are then reported. Compared with other extraction methods, ultrasonic or microwave extraction improves efficiency, yet they can damage structures. Challenges arise in the recovery of solvents and in achieving extraction efficiency when using green solvents, such as deep eutectic solvents. Improvements can be made by combining extraction methods and controlling conditions (power, temperature, and time). Bioactive molecules and related activities are exposited clearly. The applications of AS have not been widely popularized, and the corresponding functions require further development.

IntroductionGerminated brown rice is a functional food with a promising potential for alleviating metabolic diseases. This study aimed to explore the hypolipidemic effects of autoclaving-treated germinated brown rice (AGBR) and the underlying mechanisms involving gut microbiota.MethodsDietary intervention with AGBR or polished rice (PR) was implemented in patients with hyperlipidemia for 3 months, and blood lipids were analyzed. Nutritional characteristics of AGBR and PR were measured and compared. Additionally, 16S rDNA sequencing was performed to reveal the differences in gut microbiota between the AGBR and PR groups.ResultsAGBR relieves hyperlipidemia in patients, as evidenced by reduced levels of triglycerides, total cholesterol, low-density lipoprotein cholesterol, and apolipoprotein-B, and elevated levels of high-density lipoprotein cholesterol and apolipoprotein-A1. In terms of nutrition, AGBR had significantly higher concentrations of free amino acids (10/16 species), γ-aminobutyric acid, resistant starch, soluble dietary fiber, and flavonoids (11/13 species) than PR. In addition, higher microbial abundance, diversity, and uniformity were observed in the AGBR group than in the PR group. At the phylum level, AGBR reduced Firmicutes, Proteobacteria, Desulfobacterota, and Synergistota, and elevated Bacteroidota and Verrucomicrobiota. At the genus level, AGBR elevated Bacteroides, Faecalibacterium, Dialister, Prevotella, and Bifidobacterium, and reduced Escherichia-Shigella, Blautia, Romboutsia, and Turicibacter.DiscussionAGBR contributes to the remission of hyperlipidemia by modulating the gut microbiota.

Cardiovascular diseases (CVDs) are currently the leading cause of death worldwide. In 2022, the CVDs contributed to 19.8 million deaths globally, accounting for one-third of all global deaths. With an aging population and changing lifestyles, CVDs pose a major threat to human health. Mitochondria-associated endoplasmic reticulum membranes (MAMs) are communication platforms between cellular organelles and regulate cellular physiological functions, including apoptosis, autophagy, and programmed necrosis. Further research has shown that MAMs play a critical role in the pathogenesis of CVDs, including myocardial ischemia and reperfusion injury, heart failure, pulmonary hypertension, and coronary atherosclerosis. This suggests that MAMs could be an important therapeutic target for managing CVDs. The goal of this study is to summarize the protein complex of MAMs, discuss its role in the pathological mechanisms of CVDs in terms of its functions such as Ca2+ transport, apoptotic signaling, and lipid metabolism, and suggest the possibility of MAMs as a potential therapeutic approach.

To investigate the soil improvement effects of different vegetation restoration measures during the operation and maintenance of photovoltaic power plants in the Hobq Desert. This study determined the soil grain size composition and soil nutrient content of 0–5 cm under four vegetation restoration measures and within the mechanical sand barriers by laser diffraction techniques and chemical experiments. The results showed that: (1) Soil particle size composition in the study area is predominantly sandy, with 1.02%, 6.63%, 5.34% and 2.61% less sand in Leymus chinensis (YC), Glycyrrhiza uralensis (GC), Artemisia ordosica Krasch (YH) and Hedysarum scoparium (HB), respectively, compared to mechanical sand barriers (CK). Soil particle distributions of YC, GC, and HB showed better sorting and more concentrated patterns, and all four vegetation restoration measures had higher fractal dimensions than CK. (2) AN (Alkali-hydrolyzable Nitrogen) content performance: The content of YC, GC, and HB was significantly higher than that of YH and CK, AP (Available Phosphorus) content did not differ significantly between measures, AK (Available Potassium) content was higher than that of CK in all measures, with YC content being the highest and SOM content being the highest for GC. (3) Soil total nutrient is unstable across vegetation restoration measures. Overall, the performance showed that the three vegetation restoration measures, YC, GC, and HB, were more evident in the soil total nutrient content improvement. Each particle size characteristic parameter and soil nutrient response vegetation restoration measures ameliorate sandy soil in PV power plants during operation and maintenance. YC and GC have the most apparent effect on soil amelioration.

Perilla frutescens (L.) Britton is an annual herb plant of the Perilla genus in the Labiatae family, which is commonly utilized as an edible and medicinal resource. Polysaccharides are among the major components and essential bioactive compounds of P. frutescens, which exhibit a multitude of biological activities, including antioxidant, antitumor, anti-fatigue, immunoregulation, hepatoprotective, anti-inflammatory, and lipid-lowering effects. As a natural carbohydrate, P. frutescens polysaccharide has the potential to be utilized in the development of drugs and functional materials. In this paper, we provide an overview of progress made on the extraction, purification, structural characterization, and bioactivity of polysaccharides from different parts of P. frutescens. The challenges and opportunities for research are discussed, along with the potential development prospects and future areas of focus in the study of P. frutescens polysaccharides.

D-Allulose has become a promising alternative sweetener due to its unique properties of low caloric content, moderate sweetness, and physiological effects. D-Allulose 3-epimerase (DAEase) is a promising enzyme for D-Allulose production. However, the low catalytic efficiency limited its large-scale industrial applications. To obtain a more effective biocatalyst, a putative DAEase from Christensenellaceae bacterium (CbDAE) was identified and characterized. The recombinant CbDAE exhibited optimum activity at pH 7.5°C and 55°C, retaining more than 60% relative activity from 40°C to 70°C, and the catalytic activity could be significantly increased by Co2+ supplementation. These enzymatic properties of purified CbDAE were compared with other DAEases. CbDAE was also found to possess desirable thermal stability at 55°C with a half-life of 12.4 h. CbDAE performed the highest relative activity towards D-allulose and strong affinity for D-fructose but relatively low catalytic efficiency towards D-fructose. Based on the structure-guided design, the best double-mutation variant G36N/W112E was obtained which reached up to 4.21-fold enhancement of catalytic activity compared with wild-type (WT) CbDAE. The catalytic production of G36N/W112E with 500 g/L D-fructose was at a medium to a higher level among the DAEases in 3.5 h, reducing 40% catalytic reaction time compared to the WT CbDAE. In addition, the G36N/W112E variant was also applied in honey and apple juice for D-allulose conversion. Our research offers an extra biocatalyst for D-allulose production, and the comprehensive report of this enzyme makes it potentially interesting for industrial applications and will aid the development of industrial biocatalysts for D-allulose.

Introduction Percutaneous coronary intervention (PCI) guided by coronary angiography-derived fractional flow reserve (FFR) or intravascular ultrasound (IVUS) has shown improved clinical outcomes compared with angiography-only-guided PCI. In patients with intermediate stenoses, FFR resulted in fewer coronary interventions and was non-inferior to IVUS with respect to clinical outcomes. However, whether this finding can be applied to angiography-derived FFR in significant coronary artery disease (CAD) remains unclear.Method and analysis The comparison of angiography-derived FFR-guided and IVUS-guided intervention strategies for clinical outcomes in patients with coronary artery disease (FLAVOUR II) trial is a multicentre, prospective, randomised controlled trial. A total of 1872 patients with angiographically significant CAD (stenoses of at least 50% as estimated visually through angiography) in a major epicardial coronary artery will be randomised 1:1 to receive either angiography-derived FFR-guided or IVUS-guided PCI. Patients will be treated with second-generation drug-eluting stent according to the predefined criteria for revascularisation: angiography-derived FFR≤0.8 and minimal lumen area (MLA)≤3 mm2 or 3 mm270%. The primary endpoint is a composite of all-cause death, myocardial infarction and revascularisation at 12 months after randomisation. We will test the non-inferiority of the angiography-derived FFR-guided strategy compared with the IVUS-guided decision for PCI and the stent optimisation strategy.The FLAVOUR II trial will provide new insights into optimal evaluation and treatment strategies for patients with CAD.Ethics and dissemination FLAVOUR II was approved by the institutional review board at each participating site (The Second Affiliated Hospital of Zhejiang University School of Medicine Approval No: 2020LSYD410) and will be conducted in line with the Declaration of Helsinki. Informed consent would be obtained from each patient before their participation. The study results will be submitted to a scientific journal.Trial registration number NCT04397211.

In order to explore the differences in the molecular mechanism of the Pueraria cultivars between isoflavone metabolic enzyme gene PtCHI, and to preliminarily reveal the difference content causes of the isoflavones. The materials of the study were Pueraria montana var. lobata and P. montana var. thomsonii. Puerarin and total flavonoids of P. montana var. lobata and P. montana var. thomsonii were extracted by ethanol, and their contents were measured by high-performance liquid chromatography. Based on the reported CHI gene of Pueraria montana var. lobata, the PtCHI gene from P. montana var. thomsonii was isolated by homologous cloning method, and the protein was expressed in vitro. At the same time, the location of the PtCHI gene was studied in Arabidopsis thaliana protoplasts. The results were as follows: (1)The content of puerarin in P. montana var. lobata was significantly higher than the P. montana var. thomsonii, and the content of total flavonoids was also higher but not significant.（2) The gene PtCHI was successfully isolated from P. montana var. thomsonii. The gene was 742 bp in length, containing a complete ORF frame of 672 bp, encoding 223 amino acids, and had up to 99% homology with P. montana var. lobata. (3)This study found that the expression of CHI gene in P. montana var. thomsonii was stem>root>leaf, P. montana var. lobata was root>stem>leaf. The expression of CHI gene from P. montana var. lobata was significantly higher than P. montana var. thomsonii besides in leaves. (4) Through the online tool prediction analysis, PtCHI was found to be stable hydrophilic protein and the size was 27.8 kD. The secondary and tertiary structures were based on α-helix, with 25 phosphorylation sites, closely relating P. montana var. lobata, Glycine max and Glycyrrhiza uralensis, and were more likely to interact with F3H2, F3H, 4CL4, DFR2 and CHS. (5)At the same time, the protein of PtCHI was successfully induced and isolated in vitro, with a single protein of 27.8 kD. (6) Through the Arabidopsis thaliana protoplasts revealed that PtCHI was mainly located in the chloroplasts. This study further analyzed the difference in flavonoids in P. montana var. lobata and P. montana var. thomsonii, as well as provides the reference for the functional verification of P. montana var. thomsonii PtCHI and the research on the mechanism of isoflavone metabolism.

Abstract Information leakage has become an urgent problem in multiple Online Social Networks (OSNs). The interactive communication of users has raised several privacy concerns. However, the current related work on privacy measurement only considers the privacy disclosure of user profile settings, ignoring the importance of profile attributes. To solve the efficient measurement problem, we consider the influence of attribute weight on privacy disclosure scores and propose a privacy measurement method by quantifying users’ privacy disclosure scores in social networks. Through introducing Technique for Order Preference by Similarity to an Ideal Solution (TOPSIS), we propose a Privacy Scores calculation model based on Fuzzy TOPSIS decision method (PSFT), that is more accurate calculate users’ privacy disclosure scores and that can improve users’ privacy awareness in multiple OSNs. Users can reasonably set the attribute file configuration based on privacy scores and attribute weight. We conduct extensive experiments on synthetic data set and real data set. The results of the experiments demonstrate the effectiveness of our model.

In order to improve the economic value of industrial hemp, an active ingredient cannabinoid (CBD) with medicinal value from industrial hemp leaves was extracted by ionic liquid assisted ultrasonic method and the extraction process was optimized. In this study, [C8mim]NTF2 was selected as the best extraction solvent from 16 kinds of imidazole ionic liquids based on the yield of CBD. Then single factor experiment was conducted on six factors affecting the yield of CBD extraction by ionic liquid assisted ultrasonic method. The six factors included: Ultrasonic power, ultrasonic temperature, ultrasonic time, ethanol concentration, molar concentration of ionic liquid and liquid material ratio. And the ethanol concentration was 65%. The ionic liquid molar concentration was 0.3 mol/L (65% ethanol solution preparation). On the basis of these results, four significant factors (ultrasonic power, ultrasonic temperature, ultrasonic time and liquid material ratio) were screened out by Plackett-Burman experimental design. The extraction process was further optimized by response surface Box-Behnken experimental design. The optimum extraction process conditions of CBD significant factors were as follows: Ultrasonic power 280 W, ultrasonic temperature 50 ℃, ultrasonic time 62.5 min, liquid material ratio 25 mL/g. Under the above conditions, the yield of CBD extracted by ionic liquid [C8mim]NTF2 was 7.66%±0.2%, the yield of CBD extracted by methanol was 6.42%±0.3%, and the yield of CBD extracted by 65% ethanol solution was 5.81%±0.2%. The extraction ability of CBD was ionic liquid [C8mim]NTF2>methanol>65% ethanol solution. The CBD degradation experiment showed that the CBD degradation rate of ionic liquid [C8mim]NTF2 was 16.40%±0.3%, which was smaller than that of methanol (24.65%±0.6%) and 65% ethanol solution (32.88%±0.5%). The results showed that adding 65% ethanol solution into ionic liquid [C8mim]NTF2 could not only reduce the degradation rate of CBD, but also increase the yield of CBD. This study proved that ionic liquid was not only a protective agent for CBD, but also an excellent solvent for CBD extraction. The technology of extracting CBD by ionic liquid assisted ultrasonic method was developed, which provided data support for the development and utilization of industrial hemp CBD.

The question of the formation of gerontolinguistics in China as a relatively new scientific branch for the country is considered. The relevance of the study is due to the high growth rate of the elderly Chinese population. The authors of the article offer an overview of gerontolinguistic works in China, selected on the basis of an analysis of journal articles, monographic studies, dissertations placed in Chinese libraries. It is noted that gerontolinguistics in China began its formation only in the 21st century, while in Western science the problems of working with the elderly began to be intensively studied in the second half of the 20th century. It is argued that Chinese gerontolinguistics is based on the traditions of Western science, taking into account Chinese realities. An overview of the works of Chinese scientists on gerontolinguistics is given, from 2003 to the present. It is shown that during this period, multimodal studies of the language and speech of various geront groups, including people suffering from Alzheimer’s disease, are especially actively carried out. The tasks that gerontological research solves are described, including the prevention and treatment of Alzheimer’s disease, improving the quality of life of older people, and identifying their preferences in the sociocultural sphere, for example, advertising. To solve these problems, the possibilities of artificial intelligence are actively used.

Abstract Background Herpesviruses are a class of double‐stranded DNA viruses found in both vertebrates and invertebrates. They are usually highly host‐specific and do not easily spread across species. Chinchillas have gradually entered the Chinese pet market in recent years, but references to viral infections in chinchillas are extremely scarce, and only two reports about the herpesvirus in chinchillas are available at present. Objectives The aim of this study was to present the first report of FHV‐1 infection in chinchillas. Methods A total of 130 nasopharyngeal swab samples of chinchillas and three nasopharyngeal swabs of domestic cats collected from a chinchillas farm were investigated by nested PCR for FHV‐1. Results Four chinchillas were infected with FHV‐1, the positive rate was 3.08% (4/130), and two domestic cats were FHV‐1 positive (2/3). The 253 bp fragments of FHV‐1 gD gene from four chinchillas and two domestic cats were 100% identical, respectively, and the homology between chinchillas and domestic cat was 99.21%, but they all shared nearly 98.81% homology with the reference strain sequences. Phylogenetic tree analysis showed that these four chinchillas strains were clustered together with FHV‐1. Conclusions This is the first time that FHV‐1 was detected in chinchillas and suggested chinchillas are susceptible to FHV‐1 and may play a role as a temporary reservoir for FHV‐1.

Abstract Objectives Sarcopenia is a clinically relevant syndrome with health, social, and economic implications. Alcohol consumption is one of the risk factors for sarcopenia, but it has not been fully investigated in Chinese populations. The purpose of the present study was to assess the relationship between alcohol consumption patterns (including the volume and frequency of alcohol consumption) and sarcopenia or its elements among Chinese women and men from Henan Province. Method A cross-sectional study was designed to collect information about nutrition and health in Henan Province, China, and a total of 680 individuals were studied. Sarcopenia was defined according to the Asian Working Group for Sarcopenia (AWGS) criteria updated in 2019. Alcohol consumption was calculated in grams per day and divided into three groups. Drinking frequency was divided into four groups. We assessed the likelihood that sarcopenia was associated with drinking patterns using multiple logistic regression analysis by odds ratios (ORs) with 95% confidence interval (CIs) after multiple adjustments. Results We found that men who consumed > 25 g of ethanol per day were approximately three times more likely to have low muscle mass (OR, 3.99; 95% CI, 1.10–17.05) or low grip strength (OR, 3.39; 95% CI, 1.33–9.92) than nondrinkers after multiple adjustments. In addition, males who consumed alcohol more than 4 times per week were approximately threefold to fivefold more likely to have low muscle mass (OR, 4.99; 95% CI, 1.24–25.55) or low grip strength (OR, 3.37; 95% CI, 1.40–8.74) than nondrinkers. Unfortunately, we did not find a relationship between alcohol consumption patterns and sarcopenia or gait speed among males, and there was no association between alcohol consumption patterns and sarcopenia or any elements. African swine fever (ASF) is a highly contagious and hemorrhagic disease caused by infection with the African swine fever virus (ASFV), resulting in a mortality rate of up to 100%. Currently, there are no effective treatments and commercially available vaccines for ASF. Therefore, it is crucial to identify biochemicals derived from host cells that can impede ASFV replication, with the aim of preventing and controlling ASF. The ASFV is an acellular organism that promotes self-replication by hijacking the metabolic machinery and biochemical resources of host cells. ASFV specifically alters the utilization of glucose and glutamine, which are the primary metabolic sources in mammalian cells. This study aimed to investigate the impact of glucose and glutamine metabolic dynamics on the rate of ASFV replication. Our findings demonstrate that ASFV infection favors using glutamine as a metabolic fuel to facilitate self-replication. ASFV replication can be substantially inhibited by blocking glutamine metabolism. The metabolomics analysis of the host cell after late-stage ASFV infection revealed a significant disruption of normal glutamine metabolic pathways due to the abundant expression of PLA (phenyllactic acid). Pretreatment with PLA also inhibited ASFV proliferation and glutamine consumption following infection. The metabolomic analysis also showed that PLA pretreatment greatly slowed down the metabolism of amino acids and nucleotides that depend on glutamine. The depletion of these building blocks directly hindered the replication of ASFV by decreasing the biosynthetic precursors produced during the replication of ASFV’s progeny virus. These findings provide valuable insight into the possibility of pursuing the development of antiviral drugs against ASFV that selectively target metabolic pathways.

The enhanced hydrothermal stability of leather, imparted by little Cr(III), has traditionally been ascribed to strong coordinate bonds. However, this explanation falls short when considering that the heat-induced shrinking of collagen fiber is predominantly driven by rupturing weak H-bonds. This study explored the stability source via adsorption thermodynamics using collagen fiber as an adsorbent. Eleven isotherm models were fitted with the equilibrium dataset. Nine of these models aptly described Cr(III) adsorption based on the physical interpretations of model parameters and error functions. The adsorption equilibrium constants from six models could be transformed into dimensionless thermodynamic equilibrium constants. Based on the higher R2 of the van’t Hoff equation, thermodynamic parameters (∆G°, ∆H°, ∆S°) from the Fritz–Shluender isotherm model revealed that the adsorption process typifies endothermic and spontaneous chemisorption, emphasizing entropy increase as the primary driver of Cr(III) bonding with collagen. Thus, the release of bound H2O from collagen is identified as the stability source of collagen fiber modified by Cr(III). This research not only clarifies the selection and applicability of the isotherm model in a specific aqueous system but also identifies entropy, rather than enthalpy, as the principal stability source of Cr-leather. These insights facilitate the development of novel methods to obtain stable collagen-based material.

BackgroundMyocardial ischemia/reperfusion injury (MIRI) refers to the more severe damage that occurs in the previously ischemic myocardium after a short-term interruption of myocardial blood supply followed by restoration of blood flow within a certain period of time. MIRI has become a major challenge affecting the therapeutic efficacy of cardiovascular surgery.MethodsA scientific literature search on MIRI-related papers published from 2000 to 2023 in the Web of Science Core Collection database was conducted. VOSviewer was used for bibliometric analysis to understand the scientific development and research hotspots in this field.ResultsA total of 5,595 papers from 81 countries/regions, 3,840 research institutions, and 26,202 authors were included. China published the most papers, but the United States had the most significant influence. Harvard University was the leading research institution, and influential authors included Lefer David J., Hausenloy Derek J., Yellon Derek M., and others. All keywords can be divided into four different directions: risk factors, poor prognosis, mechanisms and cardioprotection.ConclusionResearch on MIRI is flourishing. It is necessary to conduct an in-depth investigation of the interaction between different mechanisms and multi-target therapy will be the focus and hotspot of MIRI research in the future.

Highly manufacturing process of chestnut paste leaves a considerable space for Economically Motivated Adulteration (EMA) with cheaper ingredients such as mung bean. In this paper a novel quantitative detection of mung bean in chestnut paste using duplex digital PCR was reported. Two sets of primers and probes were designed according to mung bean and chestnut specific genomic genes suitable for duplex droplet digital PCR (ddPCR) and duplex chip digital PCR (cdPCR) to set up a mass ratio quantitative detection method for mung bean, a common alternative plant-derived ingredient in chestnut paste products. The manufacturing process of chestnut paste products was considered to establish the linear relationship formula between mass ratio and gene copy number (CN) ratio of the two ingredients. The limits of quantification for gene CN concentrations (LOQcopy) of mung bean and chestnut were both 6 copies/μL, at the same time a mass ratio of mung bean in chestnut paste range from 5% to 80% was able to be quantified accurately to provide technical support for the identification of fraudulent substitution or adventitious contamination.

Abstract Background Metagenomics technology can directly extract microbial genetic material from the environmental samples to obtain their sequencing reads, which can be further assembled into contigs through assembly tools. Clustering methods of contigs are subsequently applied to recover complete genomes from environmental samples. The main problems with current clustering methods are that they cannot recover more high-quality genes from complex environments. Firstly, there are multiple strains under the same species, resulting in assembly of chimeras. Secondly, different strains under the same species are difficult to be classified. Thirdly, it is difficult to determine the number of strains during the clustering process. Results In view of the shortcomings of current clustering methods, we propose an unsupervised clustering method which can improve the ability to recover genes from complex environments and a new method for selecting the number of sample’s strains in clustering process. The sequence composition characteristics (tetranucleotide frequency) and co-abundance are combined to train the probability model for clustering. A new recursive method that can continuously reduce the complexity of the samples is proposed to improve the ability to recover genes from complex environments. The new clustering method was tested on both simulated and real metagenomic datasets, and compared with five state-of-the-art methods including CONCOCT, Maxbin2.0, MetaBAT, MyCC and COCACOLA. In terms of the number and quality of recovered genes from metagenomic datasets, the results show that our proposed method is more effective. Conclusions A new contigs clustering method is proposed, which can recover more high-quality genes from complex environmental samples.

The accurate understanding of the origin of overpressure is the basis of formation pressure research and the core work of determining the distribution of formation pore pressure in drilling design. The deep water area of Baiyun Sag in the Pearl River Mouth Basin in the northern margin of the South China Sea has been proved to be a hydrocarbon rich depression. In recent years, overpressure has been encountered in the middle and deep layers, but the geological origin of the overpressure in the deep water area are still unclear. Therefore, the pore pressure profile of the Baiyun Sag was evaluated using the data of drilling and seismic velocity, and the relationship between overpressure and the generation mechanism was established. Most of the surrounding and shallow strata of Baiyun Sag show normal pressure, while the middle and deep strata generally show overpressure, with the maximum pressure coefficient up to 1.55. The overpressure zone mainly develops from the middle of Zhujiang Formation to the upper member of Enping Formation. The mudstone resistivity and density values of the two typical Wells deviated from the normal trend. The density showed a constant trend with the increase of depth, while the resistivity showed a certain degree of reversal. The upper layer of the overpressure zone showed abnormally high resistivity values, corresponding to the overgland and transition zone formed by high carbonate content. There is no obvious increase of porosity in the overpressured sandstone strata of Baiyun sag, which does not have the characteristics of undercompaction. The velocity in the overpressure section of the two Wells varies greatly, and the density is basically unchanged or slightly decreased. It is concluded that the overpressure is caused by fluid expansion from hydrocarbon-generation. The large-scale and long-term fluid expansion caused by hydrocarbon generation and expulsion of source rocks provides the power source for the formation of overpressure in Baiyun sag. The distribution of effective source rocks has a controlling effect on the distribution range of overpressure. The tight sandstone strata in the Zhuhai, Wenchang and Enping formations become the cover layer of the overpressure system in the longitudinal direction.

Offshore heavy oil resources are abundant, but they have greater difficulty and higher costs compared to onshore extraction. When crude oil flows through the seawater section, the temperature of the crude oil decreases faster, making it susceptible to solidification in the wellbore and resulting in lower well production. The cooling of crude oil becomes more pronounced in deep-water wellbore. However, the injection of low-temperature gas will have a cooling effect on the formation production fluid, which will have a negative effect. The model analyzes the effects of coiled tubing running depth, gas injection temperature, gas injection volume, coiled tubing diameter, and crude oil production on the temperature distribution of heavy oil in deep-water and shallow water wellbores. We propose recommendations for the selection of each parameter for deep and shallow water environments by analyzing and summarizing the laws.

The article considers a new for the Chinese society philosophy of young people’s lifestyle represented by the concept of TANG PING (to lie flat). The study relevance is due to the tendency for emergence of words and expressions that represent new phenomena of reality in the Chinese language. It is noted that in 2021, the expression to lie flat has become popular in China, and the phenomenon that it designates has been actively discussed not only in everyday Internet communication, but also in the media. It is shown that several publications on this topic have also appeared in the Russian Internet discourse. The article analyzes the perception of the precisely translated Chinese expression to lie flat as a symbol of the contemporary Chinese culture by the Russians based on Internet texts and the online survey results. It is shown that the Chinese concept is closely related to the Russian literary phenomenon “Oblomovism”. There are presented the views of Chinese researchers on “Oblomovism” and its similarities and differences with the new phenomenon in the life of young generations of the Chinese society. The practical significance of the study lies in clarifying the semantic volume of the expression tang ping (to lie flat) as a linguistic unit naming a current phenomenon in the life of a modern developed society which has become widespread not only in China, but also in other countries.
