Active droplets are artificial microswimmers built from a liquid dispersion by microfluidic tools and showing self-propelled motion. These systems hold particular interest for mimicking biological phenomena, such as some aspects of cell locomotion and collective behaviors of bacterial colonies, as well as for the design of droplet-based biologically inspired materials, such as engineered tissues. Growing evidence suggests that geometrical confinement crucially affects their morphology and motility, but the driving physical mechanisms are still poorly understood. Here, we study the effect of activity on a droplet containing a contractile polar fluid confined within microfluidic channels of various sizes. We find a surprising wealth of shapes and dynamic regimes, whose mechanics is regulated by a subtle interplay between contractile stress, droplet elasticity, and microchannel width. They range from worm-like and cell-like shaped droplets displaying an oscillating behavior within wider channels to bullet-shaped droplets exhibiting rectilinear motion in narrower slits. Our findings support the view that geometrical confinement can provide a viable strategy to control and predict the propulsion direction of active droplets. This work presents a microscale approach for simulating the dielectrophoresis assembly of polarizable particles under an external electric field. The model is shown to capture interesting dynamical and topological features, such as the formation of chains of particles and their incipient aggregation into hierarchical structures. A quantitative characterization in terms of the number and size of these structures is also discussed. This computational model could represent a viable numerical tool to study the mechanical properties of particle-based hierarchical materials and suggest new strategies for enhancing their design and manufacture. We numerically study the dynamics of a polydisperse double emulsion under a symmetric shear flow. We show that both dispersity and shear rate crucially affect the behavior of the innermost drops and of the surrounding shell. While at low/moderate values of shear rates, the inner drops rotate periodically around a common center of mass triggered by the fluid vortex formed within the emulsion generally regardless of their polydispersity; at higher values, such dynamics occurs only at increasing polydispersity, since monodisperse drops are found to align along the shear flow and become approximately motionless at late times. Our simulations also suggest that increasing polydispersity favors close-range contacts among cores and persistent collisions, while hindering shape deformations of the external droplet. Bicontinuous interfacially jammed emulsion gels ("bijels") represent a new class of soft materials made of a densely packed monolayer of solid particles sequestered at the interface of a bicontinuous fluid. Their mechanical properties are relevant to many applications, such as catalysis, energy conversion, soft robotics, and scaffolds for tissue engineering. While their stationary bulk properties have been covered in depth, much less is known about their behavior in the presence of an external shear. In this paper, we numerically study the dynamics of a bijel confined within a three-dimensional rectangular domain and subject to a symmetric shear flow sufficiently intense to break the material. Extensive numerical simulations reveal that the shear flow generally promotes the detachment of a sizable amount of particles from the fluid interface and their accumulation in the bulk. Fluid interfaces undergo large stretching and deformations along the flow direction, an effect that reduces their capability of entrapping particles. These results are supported by a series of quantitative indicators such as (i) curvature of the fluid interface, (ii) spatial distribution of the colloidal particles, and (iii) fluid flow structure within the microchannel. Summary: In modern engineering systems, reliability and safety can be conferred by efficient automatic monitoring and fault detection algorithms, allowing for the early identification and isolation of incipient faults. In case of large‐scale and complex systems, scalability issues and computational limitations make centralized monitoring and fault detection methods unapplicable. Research is therefore currently focusing on the development of distributed methods, where the computational complexity is divided among different units. In this paper, we propose a partition‐based model‐based fault detection and isolation scheme based on moving horizon estimation, able to estimate both the state variables and the possible faults, modeled as additive signals on the state and/or output equations. We numerically investigate the rheological response of a noncoalescing multiple emulsion under a symmetric shear flow. We find that the dynamics significantly depends on the magnitude of the shear rate and on the number of the encapsulated droplets, two key parameters whose control is fundamental to accurately select the resulting nonequilibrium steady states. The double emulsion, for instance, attains a static steady state in which the external droplet stretches under flow and achieves an elliptical shape (closely resembling the one observed in a sheared isolated fluid droplet), while the internal one remains essentially unaffected. Novel nonequilibrium steady states arise in a multiple emulsion. Under low/moderate shear rates, for instance, the encapsulated droplets display a nontrivial planetarylike motion that considerably affects the shape of the external droplet. Some features of this dynamic behavior are partially captured by the Taylor deformation parameter and the stress tensor. Besides a theoretical interest on its own, our results can potentially stimulate further experiments, as most of the predictions could be tested in the lab by monitoring droplets' shapes and position over time. The dissociation energies of the intermetallic molecules AuSr and AuBa were for the first time determined by the Knudsen effusion mass spectrometry method. The two species were produced in the vapor phase equilibrated with apt mixtures of the constituent elements, and the dissociation equilibria were monitored mass-spectrometrically in the temperature range 1406-1971 K (AuSr) and 1505-1971 K (AuBa). The third-law analysis of the equilibrium data gives the following dissociation energies (D0°, in kJ/mol): 244.4 ± 4.8 (AuSr) and 273.3 ± 6.3 (AuBa), so completing the series of D0°s for the AuAE (AE = group 2 element) diatomics. The AuAE species were also studied computationally at the coupled cluster including single, double and perturbative triple excitation [CCSD(T)] level with basis sets of increasing zeta quality, and various complete basis set limit extrapolations were performed to calculate the dissociation energies. Furthermore, the entire series of the heteronuclear diatomic species formed from one group 11 (Cu, Ag) and one group 2 (Be, Mg, Ca, Sr, Ba) metal was studied by DFT with the hybrid meta-GGA TPSSh functional and the def2-QZVPP basis set, selected after screening a number of functional-basis set combinations using the AuAE species as benchmark. Dissociation energies, internuclear distances, vibrational frequencies, and anharmonic constants were determined for the CuAE and AgAE species and their thermal functions evaluated therefrom. Computer simulations of bi-continuous two-phase fluids with interspersed dumbbells show that, unlike rigid colloids, soft dumbbells do not lead to arrested coarsening. However, they significantly alter the curvature dynamics of the fluid–fluid interface, whose probability density distributions are shown to exhibit (i) a universal spontaneous transition (observed even in the absence of colloids) from an initial broad-shape distribution towards a highly localized one and (ii) super-diffusive dynamics with long-range effects. Electrospinning is a nanotechnology process whereby an external electric field is used to accelerate and stretch a charged polymer jet, so as to produce fibers with nanoscale diameters. In quest of a further reduction in the cross section of electrified jets hence of a better control on the morphology of the resulting electrospun fibers, we explore the effects of an external rotating electric field orthogonal to the jet direction. Through intensive particle simulations, it is shown that by a proper tuning of the electric field amplitude and frequency, a reduction of up to a 30% in the aforementioned radius can be obtained, thereby opening new perspectives in the design of future ultra-thin electrospun fibers. Abstract: SAHA, an inhibitor of histone deacetylase activity, has been shown to sensitize tumor cells to apoptosis induced by TRAIL, a member of TNF-family. In this paper we investigated the effect of SAHA/TRAIL combination in two breast cancer cell lines, the ERα−positive MCF-7 and the ERα−negative MDA-MB231. Treatment of MDA-MB231 and MCF-7 cells with SAHA in combination with TRAIL caused detachment of cells followed by anoikis, a form of apoptosis which occurs after cell detachment, while treatment with SAHA or TRAIL alone did not produce these effects. The effects were more evident in MDA-MB231 cells, which were chosen for ascertaining the mechanism of SAHA/TRAIL action. Our results show that SAHA decreased the level of c-FLIP, thus favouring the interaction of TRAIL with the specific death receptors DR4 and DR5 and the consequent activation of caspase-8. These effects increased when the cells were treated with SAHA/TRAIL combination. Because z-IEDT-fmk, an inhibitor of caspase-8, prevented both the cleavage of the focal adhesion-kinase FAK and cell detachment, we suggest that activation of caspase-8 can be responsible for both the decrement of FAK and the consequent cell detachment. In addition, treatment with SAHA/TRAIL combination caused dissipation of ΔΨm, activation of caspase-3 and decrement of both phospho-EGFR and phospho-ERK1/2, a kinase which is involved in the phosphorylation of BimEL. Therefore, co-treatment also induced decrement of phospho-BimEL and a concomitant increase in the dephosphorylated form of BimEL, which plays an important role in the induction of anoikis. Dogs, the main domestic reservoirs of Trypanosoma cruzi in the Argentine chaco, may be useful as sentinels of vector-mediated transmission of Trypanosoma cruzi in control programmes if canine infections acquired by all other routes could be excluded. In the present study, in the province of Santiago del Estero, Argentina, the dog populations in the neighbouring villages of Amama, Trinidad, Mercedes, Villa Matilde and Pampa Pozo were studied immediately before spraying of residual insecticides (in 1992 in Amama, Trinidad and Mercedes and 1993-1994 in the other villages) and twice during a subsequent programme of triatomine surveillance (in 1994 and 1996). Overall seropositivity for Trypanosoma cruzi infection (i.e. the proportion of dogs found positive by at least two different serological tests in any one survey) steadily declined from 65% (54/83) in 1992 to 39% (70/182) in 1994 and 15% (36/237) in 1996. No seroconversion was detected between the 1994 and 1996 surveys in any dog aged 2 years. However, among the dogs aged 2 years (which were born after virtual elimination of Triatoma infestans from the villages), seropositivity fell from 15% (12/81) in 1994 to 5% (6/131) in 1996. Thirteen demographic, behavioural and entomological variables were analysed by logistic multipleregression analysis for their association with seropositivity in dogs born locally during the surveillance programme. The total number of Triatoma guasayana collected in the bedroom areas of the owner's house, having a mother which was seropositive for Trypanosoma cruzi , and the number of seropositive dogs with which it cohabited were all found to be significant predictors of seropositivity in any dog. The results are the first indication that Triatoma guasayana is the vector responsible for the new cases of Trypanosoma cruzi infection seen in dogs in domestic or peridomestic sites during the surveillance programme. Butyrate can promote programmed cell death in a number of tumour cells in vitro. This paper provides evidence that butyrate induces apoptosis in human hepatoma HuH-6 and HepG2 cells but is ineffective in Chang liver cells, an immortalised non-tumour cell line. In both HuH-6 and HepG2 cells, apoptosis appeared after a lag period of approximately 16 h and increased rapidly during the second day of treatment. In particular, the effect was stronger in HuH-6 cells, which were, therefore, chosen for ascertaining the mechanism of butyrate action. In HuH-6 cells, β-catenin seemed to exert an important protective role against apoptosis, since pretreatment with β-catenin antisense ODN reduced the content of β-catenin and anticipated the onset of apoptosis at 8 h of exposure to butyrate. Moreover, in HuH-6 cells, butyrate induced loss of mitochondrial membrane potential, release of cytochrome c from mitochondria, activation of caspase 9 and caspase 3, and degradation of poly(ADP-ribose) polymerase. In addition, during the second day of treatment, β-catenin, pRb, and cyclins D and E were diminished and the phosphorylated form of pRb disappeared. Also, the content of the anti-apoptotic factor Bcl-XL fell markedly during this period, while that of the pro-apoptotic factor Bcl-Xs increased. These effects were accompanied by an increase in both Bcl-XL and Bcl-Xs mRNA transcripts, as ascertained by reverse transcriptase-polymerase chain reaction. Our results suggest that caspases have a crucial role in butyrate-induced apoptosis. This conclusion is supported by the observation that the inhibitors of caspases, benzyloxy carbonyl-Val-Ala-Asp-fluoromethylketone and benzyloxy carbonyl-Asp-Glu-Val-Asp-fluoromethylketone, prevented apoptosis and the decrease in Bcl-XL, pRb, cyclins and β-catenin. These effects were most probably responsible for the increased sensitivity of the cells to butyrate-induced apoptosis, which was observed on the second day of treatment. [Copyright &y& Elsevier]

The dynamic interaction of complex fluid interfaces is highly sensitive to near-contact interactions occurring at the scale of ten of nanometers. Such interactions are difficult to analyze because they couple self-consistently to the dynamic morphology of the evolving interface, as well as to the hydrodynamics of the interstitial fluid film. In this work, we show that, above a given magnitude threshold, near-contact interactions trigger nontrivial microvorticity patterns, which in turn affect the effective near-contact interactions, giving rise to persistent fluctuating ripples at the fluid interface. In such a regime, near-contact interactions may significantly affect the macroscopic arrangement of emulsion configurations, such as those arising in soft-flowing microfluidic crystals.
