The emergence of nanotechnology and surface engineering techniques provides new opportunities for designing self-lubricant coatings with enhanced properties. In recent years, green coating technologies have played a vital role in environmental preservation. This article mainly reviews five typical types of self-lubricant coatings including MoN coatings, VN coatings, WN coatings and TMN (Transition Metal Nitride) soft-metal coatings, and DLC (Diamond-like Carbon) with lubricant agents deposited by PVD (Physical Vapor Deposition) for the demanding tribological applications, which is the latest research into the green lubricant coatings. In this paper, a temperature measurement system with NTC (Negative Temperature Coefficient) thermistors was designed. An MCU (Micro Control Unit) primarily operates by converting the voltage value collected by an ADC (Analog-to-Digital Converter) into the resistance value. The temperature value is then calculated, and a DAC (Digital-to-Analog Converter) outputs a current of 4 to 20 mA that is linearly related to the temperature value. The nonlinear characteristics of NTC thermistors pose a challenging problem. The nonlinear characteristics of NTC thermistors were to a great extent solved by using a resistance ratio model. The high precision of the NTC thermistor is obtained by fitting it with the Hoge equation. The results of actual measurements suggest that each module works properly, and the temperature measurement accuracy of 0.067 °C in the range from −40 °C to 120 °C has been achieved. The uncertainty of the output current is analyzed and calculated with the uncertainty of 0.0014 mA. Little information exists on the variation in morphological characteristics, nutritional value, ruminal degradability, and molecular structural makeup of diverse whole-plant silage corn (WPSC) cultivars among different growing regions. This study investigated the between-regions (Beijing, Urumchi, Cangzhou, Liaoyuan, Tianjin) discrepancies in five widely used WPSC cultivars in China (FKBN, YQ889, YQ23, DK301 and ZD958), in terms of 1) morphological characteristics; 2) crude protein (CP) chemical profile; 3) Cornell Net Carbohydrate and Protein System (CNCPS) CP subfractions; 4) in situ CP degradation kinetics; and 5) CP molecular structures. Our results revealed significant growing region and WPSC cultivar interaction for all estimated morphological characteristics (P < 0.001), CP chemical profile (P < 0.001), CNCPS subfractions (P < 0.001) and CP molecular structural features (P < 0.05). Except ear weight (P = 0.18), all measured morphological characteristics varied among different growing regions (P < 0.001). Besides, WPSC cultivars planted in different areas had remarkably different CP chemical profiles and CNCPS subfractions (P < 0.001). All spectral parameters of protein primary structure of WPSC differed (P < 0.05) due to the growing regions, except amide II area (P = 0.28). Finally, the area ratio of amide I to II was negatively correlated with the contents of soluble CP (δ = -0.66; P = 0.002), CP (δ = -0.61; P = 0.006), non-protein nitrogen (δ = -0.56; P = 0.004) and acid detergent insoluble CP (δ = -0.43; P = 0.008), in conjunction with a positive correlation with moderately degradable CP (PB1; δ = 0.58; P = 0.01). In conclusion, the cultivar of DK301 exhibited high and stable CP content. The WPSC planted in Beijing showed high CP, SCP and NPN. The low rumen degradable protein of WPSC was observed in Urumchi. A comprehensive understanding of public participation is critical for delivering justice in the energy transition. Waste-to-energy (WTE) is a practical approach toward sustainable energy transitions, which may threaten the realization of justice. Previous studies on WTE facilities have mostly discussed the siting stage under the environmental impact assessment (EIA) framework, and few have focused on whether public participation differs when different types of pollution occur. In this paper, we take a WTE project in Kunming, China as an example and, based on semi-structured interviews and questionnaires, explore the influence of differential public participation. The findings indicate that residents were more sensitive when faced with obvious pollution, which became the dominant factor in participation; meanwhile, they were inactively engaged in hidden pollution. Lower levels of health participation partly affect the realization of outcome justice. The shift from procedural justice to outcome justice is not inevitable and is subject to many factors. From an activism perspective, different perceptions of pollution risks and constraints on activities contribute to this state. We call for the government to complete the relevant engagement design in WTE projects and improve citizens' awareness to achieve better environmental justice. [ABSTRACT FROM AUTHOR]

Simple indigestion in cows leads to substantial economic losses in the dairy industry. Despite ongoing efforts, an effective treatment for this issue remains elusive. Previous studies have emphasized the vital role of rumen microbes in maintaining ruminant health. To deepen our comprehension of the intricate interplay between rumen microbiota and simple indigestion, we undertook a study involving the analysis of rumen fluid from eight cows with simple indigestion and ten healthy cows. Additionally, we collected data pertaining to milk production, rumination behavior, and rumen characteristics. The results showed that cows with simple indigestion displayed significantly lower milk yield, reduced rumination duration, and weakened rumen contraction when contrasted with the healthy cows (p < 0.05). However, no significant difference in microbiota α-diversity emerged (p > 0.05). Principal coordinate analysis (PCoA) illuminated substantial variations in rumen microbial structure among the two groups (p < 0.05). Further analysis spotlighted distinctive bacteria in the rumen of the cows with indigestion, including Allisonella, Synergistes, Megasphaera, Clostridium_XIVb, Campylobacter, and Acidaminococcus. In contrast, Coraliomargarita, Syntrophococcus, and Coprococcus are the dominant bacterial genera in the rumen of healthy dairy cows. Importantly, these key bacterial genera also dominated the overarching microbial interaction network. The observation suggests that changes in the abundance of these dominant bacterial genera potentially underlie the principal etiology of cows with simple indigestion. The present study investigates the effect of 50 mL AMCB taken daily as a dietary supplement on the rumen fermentation, microbiota, and production performance of 40 Holstein dairy cows in the transition period with 2.76 ± 0.48 parity and 650 ± 25 kg body weight. AMCB supplementation stabilized rumen pH, improved rumen microbiota richness and partial probiotic colonization, and considerably increased dry matter intake, milk production, protein content, and yield. Moreover, after calving, AMCB supplementation considerably reduced the serum blood urea nitrogen, malondialdehyde, hydrogen peroxide, alanine aminotransferase, and aspartate transaminase levels and increased the serum immunoglobulin G and A levels. The results indicated that AMCB dietary supplementation improved postpartum dry matter intake, production performance, partial immune function, antioxidant capacity, and rumen microbiota richness in Holstein dairy cows in the transition period. Supplementation with saccharomyces cerevisiae culture products (SCs) has shown effectiveness in alleviating or improving the health and productivity of ruminants at a high risk of digestive and metabolic problems as a consequence of their physiological state and feeding system (i.e., Holstein cows during peak lactation). However, the effects of SC supplementation on ruminal digestion and microbial population are not yet well-understood. Hence, this study aimed to contribute to the knowledge of the effects of in vitro SC supplementation on ruminal nutrient digestibility and microbial community. This study included three treatment groups: a control group (CON, 0% SC proportion of substrate DM), a low-dose SC group (LSC, 0.10% SC proportion of substrate DM), and a high-dose SC group (HSC, 0.30% SC proportion of substrate DM). The SC product contained 7.0 × 109 CFU/g. After 48 h of fermentation at 39 °C, the incubation fluid and residue were collected to measure the ruminal nutrient digestibility and microbial community. The results showed that supplemental SC tended (p = 0.096) to increase DM digestibility due to an increase (9.6%, p = 0.03) in CP digestibility and via a tendency (0.05 < p < 0.08) to increase the fiber fraction. Additionally, the 16S rRNA high-throughput sequencing results revealed that the richness and diversity of the microbiota were unchanged by SC supplementation, while the abundances of Spirochaetes, Tenericutes, and Spirochaetaceae were lower in the SC groups than those in the CON group (p < 0.05). At the genus level, the abundances of Selenomonas and Succinivibrio were increased by SC supplementation (p < 0.05), while SC supplementation decreased the abundances of Ruminococcaceae_UCG-014 and Treponema_2 (p < 0.05). Furthermore, the predicted function of the microbiota showed that carbohydrate metabolism and lipid metabolism were enriched in the SC groups compared with the CON group (p < 0.05). Except for the increases in ADF digestibility (p = 0.032) and pH (p = 0.076) at 0.30%, the supplemental level did not result in additional effects. Simple Summary: The nutrient contents and rumen degradation of alfalfa hay rely highly on cultivars and grown regions. Moreover, the evaluation of alfalfa hay with respect to nutritional components in conjunction with the selection of feed resources according to local condition is necessary and essential for dairy farms to obtain an efficient and economic formula. Thus, this study was conducted to investigate the variations of different alfalfa hay cultivars grown in China and the US in terms of nutritional components, ruminal degradation characteristics and feed value. The results showed that the cultivar of American Golden Empress (GE) had the greatest rumen degradation characteristics; meanwhile, the indicators of total digestible nutrients (TDN), dry matter intake (DMI), digestible dry matter (DDM), relative feed value (RFV) and relative forage quality (RFQ) of GE were better than China Zhongmu No. 1 (ZM1) and China Gongnong No. 1 (GN1) cultivars. The objective of this study was to evaluate the effects of different cultivars of alfalfa hay, including American Anderson (AA), American Golden Empress (GE), China Zhongmu No. 1 (ZM1) and China Gongnong No. 1 (GN1), on conventional nutrient composition, rumen degradation characteristics and feed value. Four healthy Holstein cows (137 ± 14 days in milk, 2.40 ± 0.50 parity) equipped with permanent ruminal cannulas were examined for the nylon-bag technique. The alfalfa hay samples were incubated in the rumen for 0, 4, 8, 12, 24, 36, 48 and 72 h according to the "gradual in/all out" schedule to detect the ruminal nutrients' degradability. Our results showed that various cultivars of alfalfa hay from different planting regions had significant differences on nutrient contents, rumen degradability and feed value. For nutritional components of alfalfa hay, the highest dry matter (DM) content was found in GE and the lowest in GN1 (p < 0.001); however, GN1 had the greatest concentration of ether extract (EE, p = 0.01), Ca (p < 0.001) and the lowest Ash (p < 0.001). Additionally, the lowest neutral detergent fiber (NDF), acid detergent fiber (ADF) and highest starch contents were observed in AA and GE (p < 0.001). Meanwhile, the cultivar of ZM1 represented the highest NDF, ADF and Ash contents, in conjunction with minimal CP and P concentrations (p < 0.001). In terms of rumen degradation characteristics, the effective degradation rate (ED) of DM in GE and ZM1 was significantly higher than that in AA and GN1 (p = 0.013). The NDF effective degradation was lower in ZM1 than the other three cultivars (p = 0.002), and in addition ZM1 also showed lower CP and ADF effective degradation than GE (p < 0.001). As far as feed value was concerned, the cultivar of alfalfa hay imported from the US, including AA and GE, exhibited higher relative feed value (RFV) and relative forage quality (RFQ) than Chinese alfalfa based on ZM1 and GN1 (p < 0.001). Forages are vital in maintaining the dietary structures of ruminants, and reducing their costs is important for improving dairy production efficiency. Thus, this study investigated the effects of dietary forage types on dry matter intake, production, rumen fermentation, and the microbial profile in peak-lactating cows. Eight cows (600 ± 25 kg) with days in milk (60 ± 10 days) were assigned to four groups using a replicated 4 × 4 Latin square design: OG (oat hay + alfalfa hay + corn silage + concentrate), CW (Leymus chinensis + alfalfa hay + corn silage + concentrate), AS (alfalfa silage + oat hay + corn silage + concentrate), and AC (alkali-treated corn straw + alfalfa hay + corn silage + concentrate). The ruminal butyrate acid concentration was lower in the OG group than in the AS and AC groups post-feeding (12 h; p < 0.05). Ruminal NH3-N content was higher in the AS group than in the AC and CW groups post-feeding (9 h; p < 0.05). The percentage of ruminal Oscillospira and unknown microbes was higher in the CW group than in the other groups (p < 0.05). The total rumination time and rumination time per dry matter intake of AC were significantly higher than those of the other groups (p < 0.05). Milk lactose content in the AS group was highest among the groups (p < 0.05), and milk fat content was higher in the OG group than in the CW group (all p < 0.05). Overall, our results suggested that the butyrate acid content of forage from oat hay in rumen fermentation was significantly lower than that of calcium oxide and alfalfa silage sources when the dietary nutrition level was similar. The diet derived from L. chinensis increased the number of fibrillation helices related to fiber-decomposing bacteria and simultaneously increased unknown strains. Forage derived from alfalfa silage tended to increase milk protein levels. Alkali-treated corn straw could significantly increase the total rumination time and unit dry matter rumination time of dairy cows, which plays an important role in maintaining rumen health. Rosemary extract (RE) is characterized as an antioxidant, and it has the potential to reduce methane emission and change microbial fermentation. Hence, to the extent of the evaluation of RE in ruminant nutrition, the in vitro fermentation technique was used to investigate the effects of RE on the fermentation characteristics of a total mixed ration (TMR) fed to dairy cows. Different doses of RE were added to the TMR to obtain different concentrations of antioxidants, including 0 (CON), 0.05 (LRE), and 0.10 g/kg (HRE). A total of 500 mg ground TMR was incubated in buffer solution and rumen fluid for 48 h at 39 °C. Nutrient degradability, gas production parameters, gas composition, fermentation parameters, and microbial composition were analyzed. The results showed that nutrient degradability and total volatile fatty acid concentration were not affected by the treatments. Furthermore, total methane production and proportion were depressed in a dose-dependent way. The RE increased the propionate concentration and proportion linearly and decreased the acetate concentration and proportion linearly. Finally, microbial diversity analysis showed that the richness and evenness indexes were unchanged by different treatments, while Prevotella_1 was decreased and Prevotella_7 was increased with RE supplementation. In conclusion, RE is an effective inhibitor of methane emission of microbial fermentation and changed the profile of volatile fatty acids with no disadvantageous effects on diet utilization. Rosemary extract (RE) has been used as an antioxidant in cosmetics and food additives, indicating its potential as a feed additive to improve adaptation in high-producing dairy cows. Here, we investigated the effects of RE supplementation on lactation performance and rumen fermentation in high-producing dairy cows. Thirty multiparous cows were blocked into 15 groups based on milk production and were randomly assigned to one of two treatments: 0 or 28 g/d of RE supplementation to the basic diet per cow. The experiment was conducted over a 74-day period, which included an initial two-week adaptation period. We observed significant increases in milk and milk lactose yields following RE supplementation. Somatic cell count tended to decrease by treatment. Additionally, superoxide dismutase concentration significantly increased and malonaldehyde level decreased after RE supplementation. Sequencing of 16S rRNA revealed that RE supplementation significantly affected the microbial composition and decreased the richness of the microbiota. Specifically, the abundance of the genus Prevotella was significantly decreased by RE supplementation and was correlated with volatile fatty acids in the Mantel test, whereas no significant correlation was found for other genera. Simple Summary: The transition period (3 wks before to 3 wks after calving) is an important period in the lactation cycle of dairy cows. During this period, dairy cows undergo a series of physiological and metabolic changes due to the demands of pregnancy, parturition and postpartum lactation, which can easily lead to a negative energy balance (NEB). Therefore, the energy balance and nutritional regulation mechanism of dairy cows during the transition period are the focus of dairy cow nutrition and physiology research. However, crossbred cows have received less attention to date. Our study showed that feeding a prenatal concentrate mix at a rate of 0.6% of the body weight of Montbéliarde–Holstein crossbred cows has no negative effect on the performance and rumen fermentation of postpartum dairy cows and can satisfy the energy needs of dairy cows in the prepartum period. Furthermore, our data provide a theoretical basis for further revisions to the feeding standard and to alleviate the NEB of Montbéliarde–Holstein crossbred cows in China. This study was conducted to investigate the effect of three rates of prepartum dietary concentrate feeding on the milk performance, energy balance, and rumen fermentation of Montbéliarde–Holstein crossbred cows. Eighteen transition Montbéliarde–Holstein crossbred cows with similar days of gestation (258 ± 12 day) and body weights (622 ± 44 kg) were selected and randomly divided into three groups. In the prepartum period, the addition of concentrates accounted for 0.3% (low concentrate, LC), 0.6% (medium concentrate, MC), and 0.9% (high concentrate, HC) of the cow's body weight. The forage was corn stover, which was fed to the cows ad libitum with free access to water. Postpartum, all of the cows were fed a common lactation total mixed ration. The experimental period lasted from 21 days prepartum to 28 days postpartum. The energy balance (EB), net energy intake (NEI), and dry matter intake (DMI) of the HC group were greater than those of the other groups (p < 0.05). Likewise, the non-esterified fatty acids (NEFA), β-hydroxybutyric acid (BHBA), and total bilirubin (TBIL) in the blood of the LC group had significantly higher concentrations than they did in the other groups (p < 0.05). Moreover, the increase in the level of dietary concentrate had no significant effect on the rumen fermentation parameters (p > 0.05), and the total intestinal digestibility of dry matter (DM), crude protein (CP), and ether extract (EE) in the HC group was significantly higher (p < 0.05) than it was in the other groups during the prepartum period. In conclusion, the administration of the MC diet in the prepartum period had no negative effect on the performance and rumen fermentation of postpartum dairy cows and can satisfy the energy needs of prepartum dairy cows. Weaning stress affects the health and performance of calves. L-glutamine (L-Gln) is commonly used as a functional antioxidant and energy supplement in the body. However, dietary L-Gln supplementation improving weaning stress of calves is unclear. Thus, we aimed to explore the effects of L-Gln (provided by rumen-protected L-Gln) on calves during weaning. Seventy-five Holstein calves (54.0 ± 2.68 kg; 42 ± 2.1 d of age) were assigned to five groups: no supplementation and L-Gln with 1%, 2%, 3%, and 4% dry matter daily intake (DMI) supplementation groups, respectively. The experiment lasted for 28 days (42–70 d of age of calves), and the calves were weaned at 15 d of experiment. DMI and body weekly weight of all calves were recorded. Blood samples of nine healthy calves with similar body weight were collected from each group at 0, 7, 14, 16, 18, 21, and 28 d of experiment for detecting serum L-Gln, glucose, insulin, urea nitrogen, D-lactate, cortisol, haptoglobin, interleukin-8, immunoglobulin (Ig) G, IgA, IgM, total antioxidant capacity, superoxide dismutase, glutathione peroxidase, catalase, and malondialdehyde. At the end of the experiment, six healthy calves with similar body weight from each group were selected for slaughter and morphological analysis of small intestine tissue. The results showed that the L-Gln supplementation in the diets improved the negative effects of sudden weaning in calves. Furthermore, compared to the higher-level L-Gln supple-mentation (3 and 4% of DMI) groups, the dietary lower-level L-Gln supplementation (1 and 2% of DMI) had higher average daily gain, glutathione peroxidase and IgG concentration, and villus height/crypt depth of the duodenum and jejunum, as well as lower cortisol, haptoglobin, and interleukin-8 concentration of weaned calves. Simple Summary: The rumen microbiota plays an important role in maintaining microbiota homeostasis and promoting milk production synthesis through utilizing amino acids and non-protein nitrogen. Furthermore, various nitrogen sources have shown distinct effects on microbial growth rates. The methionine dipeptide (MD) is a bioactive peptide consisting of two methionine (Met) residues linked by a peptide bond. Although the role of MD in milk protein synthesis is established, little is known about its role in bacterial fermentation. The present study demonstrates that the various nitrogen sources could reshape microbiota differently, and MD could be more efficient than free Met in the rumen to support acetate producer growth. Our study provides some new insights into the relationship between ruminal microbiota of dairy cows and small peptides and points to potential strategies to effectively enhance the health condition and digestion ability of dairy cows. It is well known that the methionine dipeptide (MD) could enhance the dairy cows milking performance. However, there is still a knowledge gap of the effects of MD on the rumen fermentation characteristics, microbiota composition, and digestibility. This experiment was conducted to determine the effect of different nitrogen sources with a total mixed ration on in vitro nutrient digestibility, fermentation characteristics, and bacterial composition. The treatments included 5 mg urea (UR), 25.08 mg methionine (Met), 23.57 mg MD, and no additive (CON) in fermentation culture medium composed of buffer solution, filtrated Holstein dairy cow rumen fluid, and substrate (1 g total mixed ration). Nutrient digestibility was measured after 24 h and 48 h fermentation, and fermentation parameters and microbial composition were measured after 48 h fermentation. Digestibility of dry matter, crude protein, neutral detergent fiber (NDF), and acid detergent fiber (ADF) in the MD group at 48 h were significantly higher than in the CON and UR groups. The total volatile fatty acid concentration was higher in the MD group than in the other groups. In addition, 16S rRNA microbial sequencing results showed MD significantly improved the relative abundances of Succinivibrio, Anaerotruncus, and Treponema_2, whereas there was no significant difference between Met and UR groups. Spearman's correlation analysis showed the relative abundance of Succinivibrio and Anaerotruncus were positively correlated with gas production, NDF digestibility, ADF digestibility, and acetate, propionate, butyrate, and total volatile fatty acid concentrations. Overall, our results suggested that the microbiota in the fermentation system could be affected by additional nitrogen supplementation and MD could effectively enhance the nutrient utilization in dairy cows. Simple Summary: Corn straw (Zea mays, CS), rice straw (Oryza sativa, RS), and wheat straw (Triticum aestivum, WS) are the three main crop straws worldwide. Few studies on indigestible neutral detergent fiber (iNDF) and total-tract digestibility (TTD) of crude protein (CP), neutral detergent fiber (NDF), and acid detergent fiber (ADF) of these crop straws are available, which limits their utilization in dairy diets. Here, we compared the ruminal degradability, iNDF288 content, intestinal digestibility, and TTD for the CP, NDF, and ADF of these three crop straws with alfalfa hay (Medicago sativa, AH) and corn silage (Zea mays, CSil). The results showed that CS, RS, and WS had higher ruminal potential NDF degradation, intestinal digestible CP, and lower iNDF288 content compared to AH. Greater accuracies for regression equations capable of predicting the iNDF288 content and TTD were also generated based on chemical composition and ruminal degradation kinetics. Incorporating this information into rations could improve our ability to optimize the utilization of main crop straws in balanced dairy diets. Three main crop straws including corn straw (Zea mays, CS), rice straw (Oryza sativa, RS), and wheat straw (Triticum aestivum, WS), and two forages including alfalfa hay (Medicago sativa, AH) and corn silage (Zea mays, CSil) were analyzed in order to compare their ruminal degradability, indigestible neutral detergent fiber (iNDF), intestinal digestibility (ID), and their total-tract digestibility (TTD) of crude protein (CP), neutral detergent fiber (NDF), and acid detergent fiber (ADF) using both an in situ nylon bag technique and a mobile nylon bag technique. The forage samples were incubated in the rumen for 6, 12, 16, 24, 36, 48, 72, and 288 h, respectively, to determine their ruminal degradability. Prior to intestinal incubation, forage samples were incubated in the rumen for 12 h and 24 h to determine the ruminal degradable content of CP, NDF, and ADF, respectively, and for 288 h to determine their iNDF288 content. Residues from the ruminal undegradable fractions (12 h for CP, 24 h for NDF and ADF) were subsequently inserted into the duodenum through a cannula to determine their intestinal digestible content. Here, the TTD of CP, NDF, and ADF were determined as the ruminal degradable content + intestinal digestible content. The results showed that AH had the highest iNDF2.4 (calculated as acid detergent lignin content × 2.4) and iNDF288 values (379.42 and 473.40 g/kg of NDF), while CS and CSil had the lowest iNDF2.4 values (177.44 and 179.43 g/kg of NDF). The ruminal degradability of CP, NDF, and ADF for CS, RS, and WS were lower than those of AH and Csil during the first 48 h of incubation. The potential degradation fraction of CP, NDF, and ADF for CSil was the highest; CS, RS, and WS were intermediate; and AH was the lowest (p < 0.05). CS, RS, and WS had a lower intestinal digestibility with respect to their rumen undegradable content of NDF (p < 0.05), and lower TTD of CP, NDF, and ADF (p < 0.05) compared to AH and CSil. General regression equations with satisfactory accuracy (R2 ≥ 0.828) were derived to predict iNDF288 and TTD based on their chemical compositions and the ruminal degradation kinetics of different forages. Simple Summary: The rumen is a large fermentation chamber that enables dairy cows to utilize otherwise indigestible plant polymers and compounds for their nutrition, highlighting a crucial difference between ruminants and monogastrics. The key point in this process is the pool of enzymes secreted by microorganisms. Hence, exogenous enzymes from additives are important for high-production dairy cows to improve the utilization of feeds. In this study, we used the Aspergillus oryzae and Aspergillus niger co-cultivation extract (AOAN) to improve the digestibility of roughage and total mixed ration in vitro. Our results indicated that the digestibility of nutrients in feeds was significantly increased by AOAN supplementation, except for the digestibility of crude protein in the total mixed ratio (TMR). Furthermore, the diversity of the bacteria in TMR and oat hay was increased with AOAN supplementation. Although broad effectiveness of AOAN was established, regardless of roughage types, the mechanism may be different according to roughage types. AOAN may provide enzymes to improve the digestibility of feeds and enhance rumen fermentation. This study determined the effects of AOAN on digestibility, fermentation characteristics, and bacterial composition using in vitro gas recording fermentation system. A total of 30 mg of AOAN was supplemented into 500 mg of TMR, corn silage, oat hay, and alfalfa hay. Fermentation parameters and bacterial communities were determined after 48 h fermentation, and digestibility was determined after 7, 24, 30, and 48 h fermentation. Gas production and dry matter (DM), crude protein (CP), neutral detergent fiber (NDF), and acid detergent fiber (ADF) digestibility were significantly increased by AOAN supplementation at 48 h (p < 0.05), except for digestibility of CP of the TMR (p > 0.05). AOAN increased starch digestibility in corn silage (p < 0.05) and tended to increase that in TMR (0.05 < p < 0.10). AOAN supplementation increased total volatile fatty acid production (p < 0.05). The molar proportions of acetate and acetate to propionate ratio of oat hay and alfalfa hay were increased (p < 0.05). The 16S rRNA analysis revealed that the microbial richness of TMR and oat hay, and microbial evenness of TMR were increased (p < 0.05). AOAN did not affect the α diversity, β diversity, and bacterial composition of the corn silage. The relative abundance of Prevotella was increased and Ruminococcus was decreased in TMR, oat hay, and alfalfa hay. In conclusion, results suggest that AOAN has the potential to improve the utilization of diets differently, including providing enzymes with changing microbiota (TMR, oat hay, and alfalfa hay) or providing enzymes alone (corn silage). Simple Summary: Research on the amino acid nutrition of cattle is limited, particularly research on the amino acid patterns of growing heifer. This lack of research has made it difficult to minimize the costs and reduce nitrogen emission of dairy heifers. Lysine might be the first limiting amino acid for seven- to nine-month-old Holstein heifers that are fed a corn–soybean meal-based diet, followed by methionine and threonine. The appropriate ratio of lysine, methionine, and threonine—calculated based on the nitrogen retention of seven- to nine-month-old Holstein heifers—were 100:32:57. We expect to reduce the input of protein feed and nitrogen emissions for dairy farms by using this ratio. An "Amino acid (AA) partial deletion method" was used in this experiment to study the limiting sequences and appropriate ratio of lysine (Lys), methionine (Met), and threonine (Thr) in the diets of 7- to 9-month-old Holstein heifers. The experiment was conducted for three months with 72 Holstein heifers (age = 22 ± 0.5 weeks old; BW = 200 ± 9.0 kg; mean ± standard deviation). Following an initial two weeks adaptation period, heifers were allocated to one of four treatments: a theoretically balanced amino acid diet (positive control [PC]; 1.00% Lys, 0.33% Met, and 0.72% Thr), a 30% Lys deleted diet (partially deleted Lys [PD–Lys]; 0.66% Lys, 0.33% Met, and 0.72% Thr), a 30% Met deleted diet (partially deleted Met, [PD–Met]; 1.00% Lys, 0.22% Met, and 0.72% Thr), and a 30% Thr deleted diet (partially deleted Thr [PD–Thr]; 1.00% Lys, 0.33% Met, and 0.45% Thr). Experimental animals were fed a corn–soybean meal-based concentrate and alfalfa hay. In addition, the animals were provided with supplemental Lys, Met, and Thr (ruminal bypass). The results found no differences in the growth performance and nitrogen retention between PD–Thr treatment and PC treatment (p > 0.05). The average daily gain (p = 0.0013) and feed conversion efficiency (p = 0.0057) of eight- to ninr-month-old heifers were lower in both PD–Lys and PD–Met treatment than those in PC treatment. According to growth performance, Lys was the first limiting AA, followed by Met and Thr. Moreover, nine-month-old Holstein heifers in PD–Lys treatment and PD–Met treatment had higher levels of serum urea nitrogen (p = 0.0021), urea nitrogen (p = 0.0011) and total excreted N (p = 0.0324) than those in PC treatment, which showed that nitrogen retention significantly decreased (p = 0.0048) as dietary Lys and Met levels decreased. The limiting sequence based on nitrogen retention was the same as that based on growth performance. The appropriate ratio of Lys, Met, and Thr in the diet based on nitrogen retention was 100:32:57. In summary, the limiting sequence and appropriate amino acid ratio of Lys, Met, and Thr for seven- to nine-month-old Holstein heifers fed a corn–soybean meal-based diet were Lys > Met > Thr and 100:32:57, respectively.
