The Hurricane Weather Research and Forecasting Model (HWRF) was the flagship hurricane model at NOAA's National Centers for Environmental Prediction for 16 years and a state-of-the-art tool for tropical cyclone (TC) intensity prediction at the National Weather Service and across the globe. HWRF was a joint development between NOAA research and operations, specifically the Environmental Modeling Center and the Atlantic Oceanographic and Meteorological Laboratory. Significant support also came from the National Hurricane Center, Developmental Testbed Center, University Corporation for Atmospheric Research, universities, cooperative institutes, and the TC community. In the North Atlantic basin, where most improvement efforts focused, HWRF intensity forecast errors decreased by 45%-50% at many lead times between 2007 and 2022. These large improvements resulted from increases in horizontal and vertical resolution, as well as advances in model physics and data assimilation. HWRF intensity forecasts performed particularly well over the Gulf of Mexico in recent years, providing useful guidance for a large number of impactful landfalling hurricanes. Such advances were made possible not only by significant gains in computing but also through substantial investment from the Hurricane Forecast Improvement Program. SIGNIFICANCE STATEMENT: HWRF is considered one of the most important and successful ocean-coupled, regional numerical weather prediction models. This work documents HWRF forecast performance for TCs in the North Atlantic since its operational inception (2007-22). Track, intensity, and storm size (wind radii) forecasts all showed significant error reductions and skill improvements, including 45%-50% lower intensity errors. HWRF intensity forecasts showed marked improvement for rapid intensification events, especially in the Gulf of Mexico, where numerous impactful TCs made landfall throughout HWRF's lifetime. HWRF's impressive performance led to its expanded use as guidance for official TC forecasts and as a research tool across the TC community. These successes paved the way for NOAA's next-generation hurricane model, the Hurricane Analysis and Forecast System. Gender conflict, which is emerging as a serious concern in Korean society, is also a subject of discourse in the field of education. A vast majority of university students are affected by economic downturn and are required to compete in an uncertain situation. This study proposes an educational model from the perspective of understanding the younger generation, which is struggling in uncertain situations, rather than treating gender conflict merely as hatred between individual women and men. This paper also proposes an educational environment wherein students feel encouraged to deal with gender conflicts by understanding their liminal spaces and crossing borders for gender equity.

The COVID-19 pandemic has had a huge impact on every facet of life around the world. For many, the world has become a place of doubt and uncertainty as people struggle to cope with the changes coming in the wake of COVID-19 and seek to determine how best to respond. With ecological, economic, and ideological crises occurring alongside COVID-19, this study proposes religious education that emphasizes the value of life, promotes equality, and fosters global interconnectedness. This religious education provides an open, equal, and interconnected environment where learners can understand, discuss, and confront global problems for the common future.

Coupling a three-dimensional ocean circulation model to an atmospheric model can significantly improve forecasting of tropical cyclones (TCs). This is particularly true of forecasts for TC intensity (maximum sustained surface wind and minimum central pressure), but also for structure (e.g., surface wind-field sizes). This study seeks to explore the physical mechanisms by which a dynamic ocean influences TC evolution, using an operational TC model. The authors evaluated impacts of ocean-coupling on TC intensity and structure forecasts from NOAA's Hurricane Analysis and Forecast System v1.0 B (HFSB), which became operational at the NOAA National Weather Service in 2023. The study compared existing HFSB coupled simulations with simulations using an identical model configuration in which the dynamic ocean coupling was replaced by a simple diurnally varying sea surface temperature model. The authors analyzed TCs of interest from the 2020-2022 Atlantic hurricane seasons, selecting forecast cycles with small coupled track-forecast errors for detailed analysis. The results show the link between the dynamic, coupled ocean response to TCs and coincident TC structural changes directly related to changing intensity and surface wind-field size. These results show the importance of coupling in forecasting slower-moving TCs and those with larger surface wind fields. However, there are unexpected instances where coupling impacts the nearTC atmospheric environment (e.g., mid-level moisture intrusion), ultimately affecting intensity forecasts. These results suggest that, even for more rapidly moving and smaller TCs, the influence of the ocean response to the wind field in the near-TC atmospheric environment is important for TC forecasting. The authors also examined cases where coupling degrades forecast performance. Statistical comparisons of coupled versus uncoupled HFSB further show an interesting tendency: high biases in peak surface winds for the uncoupled forecasts contrast with corresponding low biases, contrary to expectations, in coupled forecasts; the coupled forecasts also show a significant negative bias in the radii of 34 kt winds relative to National Hurricane Center best track estimates. By contrast, coupled forecasts show very small bias in minimum central pressure compared with a strong negative bias in uncoupled. Possible explanations for these discrepancies are discussed. The first operational version of the coupled Hurricane Analysis and Forecast System (HAFSv1) launched in 2023 consists of the HYbrid Coordinate Ocean Model (HYCOM) and finite-volume cubed-sphere (FV3) dynamic atmosphere model. This system is a product of efforts involving improvements and updates over a 4-year period (2019-2022) through extensive collaborations between the Environmental Modeling Center at the US National Centers for Environmental Prediction (NCEP) and NOAA Atlantic Oceanography and Meteorology Laboratory. To provide two sets of numerical guidance, the initial operational capability of HAFSv1 was configured to two systems -HFSA and HFSB. In this study, we present in-depth analysis of the forecast skills of the upper ocean that was co-evolved by the HFSA and HFSB. We chose hurricane Laura (2020) as an example to demonstrate the interactions between the storm and oceanic mesoscale features. Comparisons performed with the available in situ observations from gliders as well as Argos and National Data Buoy Center moorings show that the HYCOM simulations have better agreement for weak winds than high winds (greater than Category 2). The skill metrics indicate that the model sea-surface temperature (SST) and mixed layer depth (MLD) have a relatively low correlation. The SST, MLD, mixed layer temperature (MLT), and ocean heat content (OHC) are negatively biased. For high winds, SST and MLT are more negative, while MLD is closer to the observations with improvements of about 8%-19%. The OHC discrepancy is proportional to predicted wind intensity. Contrarily, the mixed layer salinity (MLS) uncertainties are smaller and positive for higher winds, probably owing to the higher MLD. The less-negative bias of MLD for high winds implies that the wind-force mixing is less effective owing to the higher MLD and high buoyancy stability (approx. 1.5-1.7 times) than the observations. The heat budget analysis suggests that the maximum heat loss by hurricane Laura was 0(< 3°C per day). The main contributor here is advection, followed by entrainment, which act against or with each other depending on the storm quadrant. We also found relatively large unaccountable heat residuals for the in-storm period, and the residuals notably led the heat tendency, meaning that further improvements of the subscale simulations are warranted. The exosome multiprotein complex plays a critical role in RNA processing and degradation. This system governs the regulation of mRNA quality, degradation in the cytoplasm, the processing of short noncoding RNA, and the breakdown of RNA fragments. We determined two crystal structures of exosome components from Thermoplasma acidophilum (Taci): one with a resolution of 2.3 Å that reveals the central components (TaciRrp41 and TaciRrp42), and another with a resolution of 3.5 Å that displays the whole exosome (TaciRrp41, TaciRrp42, and TaciRrp4). The fundamental exosome structure revealed the presence of a heterodimeric complex consisting of TaciRrp41 and TaciRrp42. The structure comprises nine subunits, with TaciRrp41 and TaciRrp42 arranged in a circular configuration, while TaciRrp4 is located at the apex. The RNA degradation capabilities of the TaciRrp4:41:42 complex were verified by RNA degradation assays, consistent with prior findings in other archaeal exosomes. The resemblance between archaeal exosomes and bacterial PNPase suggests a common mechanism for RNA degradation. Despite sharing comparable topologies, the surface charge distributions of TaciRrp4 and other archaea structures are surprisingly distinct. Different RNA breakdown substrates may be responsible for this variation. These newfound structural findings enhance our comprehension of RNA processing and degradation in biological systems. Aging and menopause are associated with oxidative stress and inflammation. Here, we evaluated the antioxidant properties of pumpkin (Cucurbita pepo L.) seed extract and assessed its ameliorative effects on aging- and menopause-related diseases using Saos-2 cells and ovariectomized rats. The seed extract had bioactive components that exhibited antioxidant activity. The extract increased the alkaline phosphatase (ALP) activity of Saos-2 cells. The oral administration of the extract to ovariectomized rats for 12 weeks decreased their body weight, fat weight, and cardiac risk indices. It also contributed to reductions in the levels of reactive oxygen species, oxidative stress, and inflammation, as assessed by measuring the serum levels of malondialdehyde and analyzing gene expression in rats. Furthermore, the administration of the extract also promoted an enhancement of the transcription of nuclear factor erythroid 2-related factor (Nrf2), heme oxygenase-1 (Ho-1), and catalase (Cat), involved in antioxidant activity; endothelial nitric oxide synthase (eNos), involved in vasculoprotective activity; and PR/SET domain 16 (Prdm16) and peroxisome proliferator-activated receptor-gamma coactivator (Pgc1α), involved in brown adipogenesis and thermogenesis. Our results using ovariectomized rats show that pumpkin seed extract may have ameliorative effects on menopause-related diseases by increasing ALP activity, evaluating the antioxidant system, ameliorating oxidative stress and thermogenesis, and enhancing lipid profiles. The gut microbiome is well known for its influence on human physiology and aging. Therefore, we speculate that the gut microbiome may affect muscle strength in the same way as the host's own genes. To demonstrate candidates for gut microbes affecting muscle strength, we remodeled the original gut microbiome of mice into human intestinal microbiome through fecal microbiome transplantation (FMT), using human feces and compared the changes in muscle strength in the same mice before and three months after FMT. After comparing before and after FMT, the mice were divided into three groups based on the observed changes in muscle strength: positive, none, and negative changes in muscle strength. As a result of analyzing the α-diversity, β-diversity, and co-occurrence network of the intestinal microbial community before and after FMT, it was observed that a more diverse intestinal microbial community was established after FMT in all groups. In particular, the group with increased muscle strength had more gut microbiome species and communities than the other groups. Fold-change comparison showed that Eisenbergiella massiliensis and Anaeroplasma abactoclasticum from the gut microbiome had positive contributions to muscle strength, while Ileibacterium valens and Ethanoligenens harbinense had negative effects. Flavonoids and phenolic acid are two of the rich polyphenols found in cinnamon (Cinnamomum zeylanicum). The effects of cinnamon extract on the inhibition of adipocyte differentiation in 3T3-L1 fibroblast cells and prohibitory lipid accumulation in male mice fed a high-fat diet were examined. Upon treating 3T3-L1 cells with cinnamon for 3 days, the cinnamon inhibited lipid accumulation and increased gene expression levels, such as those of adiponectin and leptin. In in vivo experiments, mice were randomized into four groups after a one-week acclimation period, as follows: normal diet, normal diet + 1% cinnamon extract, high-fat diet, and high-fat diet + 1% cinnamon extract. After 14 weeks of supplementation, we found that cinnamon extract increased the expression of lipolysis-related proteins, such as AMPK, p-ACC, and CPT-1, and reduced the expression of lipid-synthesis-related proteins, such as SREBP-1c and FAS, in liver tissue. Our results show that cinnamon extract may exhibit anti-obesity effects via the inhibition of lipid synthesis and adipogenesis and the induction of lipolysis in both 3T3-L1 fibroblast cells and mice fed a high-fat diet. Christianity as a world religion was propagated from Europe and North America to Africa and Asia. Global Christianity spread to East Asia when Robert Morrison (1782-1843) arrived in Canton, China in 1807, and later in the late 19th-century Protestant missionaries from North America arrived in Japan and Korea. This Christianity experienced a modern transformation characterized by a shift from a universal religious system to multi-centered localizations in a non-Christian world. Based on the recognition that the interplay between national and global forces has a long history in East Asia, this article introduces the different developments of Christian education in East Asia, and pays specific attention to Christian education in Korea. The article will point out three issues that Christian education in both East Asia and Korea may face in the process of modern transformation. These issues can help Christians move beyond simple encounters toward global reciprocity in the process of globalization, secularization, and privatization of Christianity.

During the last two decades, globalization has come to occupy an important position in popular and academic discourses. Globalization has provided opportunities to produce possibilities of global awareness and at the same time crises to perpetuate a culture of fear. This article asks how church and religious education can provide a global education and help Christians continue to be faithful to their hope for God while engaging others for the common good. This also demands new kinds of global religious education directed toward social justice, cultural unity in diversity, and global responsibility, which can adequately address the challenges of neo-liberalism, fundamentalism, and transnationalism. Based on a dialectical relationship with hope and global education, instead of fear, religious education can be an important enterprise for achieving social justice, maintaining cultural unity in diversity, and equipping citizens with global responsibility, moving toward critical hope in a global age.

On 30 September 2021, a saildrone uncrewed surface vehicle (USV) was steered into category 4 Hurricane Sam, the most intense storm of the 2021 Atlantic hurricane season. It measured significant wave heights up to 14 m (maximum wave height = 27 m) and near-surface winds exceeding 55 m s−1. This was the first time in more than seven decades of hurricane observations that in real time a USV transmitted scientific data, images, and videos of the dynamic ocean surface near a hurricane's eyewall. The saildrone was part of a five-saildrone deployment of the NOAA 2021 Atlantic Hurricane Observations Mission. These saildrones observed the atmospheric and oceanic near-surface conditions of five other tropical storms, of which two became hurricanes. Such observations inside tropical cyclones help to advance the understanding and prediction of hurricanes, with the ultimate goal of saving lives and protecting property. The 2021 deployment pioneered a new practice of coordinating measurements by saildrones, underwater gliders, and airborne dropsondes to make simultaneous and near-collocated observations of the air–sea interface, the ocean immediately below, and the atmosphere immediately above. This experimental deployment opened the door to a new era of using remotely piloted uncrewed systems to observe one of the most extreme phenomena on Earth in a way previously impossible. This article provides an overview of this saildrone hurricane observations mission, describes how the saildrones were coordinated with other observing platforms, presents preliminary scientific results from these observations to demonstrate their potential utility and motivate further data analysis, and offers a vision of future hurricane observations using combined uncrewed platforms. The Pivot Mandu is an innovative device featuring a leak-tight adjustable 3D balloon spacer, incorporating inner mesh support, an outer e-PTFE layer, and a compliant balloon in the middle layer with a specialized detachable system. To assess its feasibility, proof of concept was rigorously evaluated through bench testing and survival porcine animal experiments. The results demonstrated successful remote inflation of the balloon system, with the balloon spacer exhibiting sustained patent and functional integrity over an extended observation period of up to 6 months. A noteworthy feature of the newly designed 3D balloon spacer is its capability for easy size adjustment during procedures, enhancing its adaptability and practicality in clinical settings. This three-layered 3D balloon spacer, with its established long-term patency, exhibits highly encouraging outcomes that hold promise in overcoming the current limitations of spacer devices for heart valve diseases. Realistic ocean initial conditions are essential for coupled hurricane forecasts. This study focuses on the impact of assimilating high-resolution ocean observations for initialization of the Modular Ocean Model (MOM6) in a coupled configuration with the Hurricane Analysis and Forecast System (HAFS). Based on the Joint Effort for Data Assimilation Integration (JEDI) framework, numerical experiments were performed for the Hurricane Isaias (2020) case, a category-1 hurricane, with use of underwater glider datasets and satellite observations. Assimilation of ocean glider data together with satellite observations provides opportunity to further advance understanding of ocean conditions and air–sea interactions in coupled model initialization and hurricane forecast systems. This comprehensive data assimilation approach has led to a more accurate prediction of the salinity-induced barrier layer thickness that suppresses vertical mixing and sea surface temperature cooling during the storm. Increased barrier layer thickness enhances ocean enthalpy flux into the lower atmosphere and potentially increases tropical cyclone intensity. Assimilation of satellite observations demonstrates improvement in Hurricane Isaias's intensity forecast. Assimilating glider observations with broad spatial and temporal coverage along Isaias's track in addition to satellite observations further increase Isaias's intensity forecast. Overall, this case study demonstrates the importance of assimilating comprehensive marine observations to a more robust ocean and hurricane forecast under a unified JEDI–HAFS hurricane forecast system. Significance Statement: This is the first comprehensive study of marine observations' impact on hurricane forecast using marine JEDI. This study found that assimilating satellite observations increases upper-ocean stratification during the prestorm period of Isaias. Assimilating preprocessed observations from six gliders increases salinity-induced upper ocean barrier layer thickness, which reduces sea surface temperature cooling and increases enthalpy flux during the storm. This mechanism eventually enhances hurricane intensity forecast. Overall, this study demonstrates a positive impact of assimilating comprehensive marine observations to a successful ocean and hurricane forecast under a unified JEDI–HAFS hurricane forecast system. Purpose: This study aimed to evaluate the validity and reliability of the Korean version of the Quality of Life in Life-Threatening Illness - Family Carer Version 3 (QOLLTI-F v3) among Korean family caregivers of patients with terminal cancers. Methods: A total of 198 family caregivers of terminal cancer patients were investigated using the Korean version of QOLLTI-F v3. Its psychometric properties were examined in terms of factor structure, convergent validity, discriminant validity, criterion validity and internal consistency. Results: The convergent validity of the Korean version of QOLLTI-F v3 showed that the standardized regression weights for each item were above .50 except for two out of 16 items, the average variance extracted was above .50 for four out of five factors, and the critical ratio ranged from .50 to .85. The discriminant validity of the items across all factors was confirmed. The value for Cronbach's α of 16 items was .84. Conclusion: The Korean version of the QOLLTI-F v3 is a reliable instrument with acceptable convergent and high discriminant validity for measuring the quality of life of families of people with life-threatening illnesses, including families of people with terminal cancers. Defective apoptotic death of activated macrophages has been implicated in the pathogenesis of rheumatoid arthritis (RA). However, the molecular signatures defining apoptotic resistance of RA macrophages are not fully understood. Here, global transcriptome profiling of RA macrophages revealed that the osmoprotective transcription factor nuclear factor of activated T cells 5 (NFAT5) critically regulates diverse pathologic processes in synovial macrophages including the cell cycle, apoptosis, and proliferation. Transcriptomic analysis of NFAT5-deficient macrophages revealed the molecular networks defining cell survival and proliferation. Proinflammatory M1-polarizing stimuli and hypoxic conditions were responsible for enhanced NFAT5 expression in RA macrophages. An in vitro functional study demonstrated that NFAT5-deficient macrophages were more susceptible to apoptotic death. Specifically, CCL2 secretion in an NFAT5-dependent fashion bestowed apoptotic resistance to RA macrophages in vitro. Injection of recombinant CCL2 into one of the affected joints of [Nfat5.sup.+/-] mice increased joint destruction and macrophage infiltration, demonstrating the essential role of the NFAT5/CCL2 axis in arthritis progression in vivo. Moreover, after intra-articular injection, NFAT5-deficient macrophages were more susceptible to apoptosis and less efficient at promoting joint destruction than were NFAT5-sufficient macrophages. Thus, NFAT5 regulates macrophage survival by inducing CCL2 secretion.
