Objectives: FGF-8, a member of the FGF family, plays a crucial role in cellular processes and has been implicated in cancer progression. The study aims to comprehend FGF-8's involvement in bone metastasis, emphasizing its potential as a diagnostic marker and focusing on its association with Bone-Alkaline Phosphatase (B-ALP) and other biochemical parameters. Methods: The case-control study spans 12 months, involving 60 participants, including 30 with secondary bone metastases and an equal number without metastasis. FGF-8 levels were quantified using ELISA, and B-ALP, serum ALP, and various biochemical parameters were assessed. The study employed standardized procedures to minimize bias, including matching cases and controls, and obtaining ethical approval. Results: In patients with bone metastasis, serum ALP levels, particularly B-ALP, were significantly higher. The metastatic group exhibited elevated FGF-8 concentrations, showcasing a positive correlation with B-ALP and serum calcium levels. The study successfully differentiated ALP isoenzymes through heat inactivation and L-phenylalanine inhibition. Additionally, serum calcium levels were markedly elevated in the metastatic group. Conclusion: The findings suggest that FGF-8 is a potential diagnostic marker for bone metastasis, particularly in breast and prostate cancers. Elevated FGF-8 levels correlate with increased B-ALP and serum calcium, indicating its role in osteoblastic differentiation in metastasis. The study proposes the utility of ELISA-based kits for FGF-8 in serum as a practical and efficient method for assessing bone tumor progression. [ABSTRACT FROM AUTHOR]

Aim Of Study: The present study was planned to analyze serum heme oxygenase-1 levels in osteosarcoma patients.Materials and Methods: Twenty five histopathologically confirmed cases of osteosarcoma localized without metastasis of all the ages attending the Orthopedic Clinics were included in the study group and twenty five patients having musculoskeletal pain (age and sex matched) served as control. Five ml of venous blood was collected aseptically from antecubital vein and serum was be separated by centrifugation and analyzed the same day. Routine biochemistry investigations were performed as per standard enzymatic methods by autoanalyzer. Serum Heme oxygenase-1 was analyzed by enzyme-linked immunosorbent assay.Results: In osteosarcoma patients, serum HO-1 levels were increased as compared to patients having musculoskeletal pain (P < 0.05). Workers have found that HO-1 induction in prostate cancer cell lines (PC3) cells restored the proliferation of osteoblasts, which was inhibited during co-culture with parental prostate cancer cell line PC3 cells. However, no concrete data are available on blood levels of HO in osteosarcoma. Major role of HO-1 is the protection against oxidative injury, additionally, it regulates cell proliferation, modulates inflammatory response and facilitates angiogenesis.Conclusion: Findings of the present study suggests that pharmacological agents that regulate HO activity or HO-1 gene silencing may become powerful tools for preventing the onset or progression of various cancers and sensitize them to anticancer therapies. Preeclampsia occurs approximately in 10% of pregnancies and remains a leading cause of maternal and neonatal mortality and morbidity worldwide, particularly in developing countries. The condition is usually diagnosed in late pregnancy by the presence of hypertension with proteinuria and/ or edema. Prevention of any disease process requires knowledge of its etiology and pathogenesis, as well as the availability of methods for prediction of those at high risk for this disorder. Numerous clinical, biophysical, and biochemical tests have been proposed for prediction or early detection of preeclampsia. Aim of Study: The present study was planned to analyze the status of serum homocysteine (hcy), folate, and Vitamin B12 in patients with osteosarcoma. Materials and Methods: The present study was conducted on 60 orthopedics patients who were divided into two groups, group I with 30 patients of osteosarcoma and group II with 30 patients with musculoskeletal pain. Results: The present study indicated that serum folate and hcy levels were increased in osteosarcoma patients and decreased Vitamin B12 in osteosarcoma patients when compared to controls. Conclusion: Lowered serum Vitamin B12 levels are possibly due to increased metabolic demand of the tumor. Raised hcy levels could be due to the contribution from rapidly proliferating cells. Objective: The present study is designed to measure sialic acid in pleural effusion and in serum samples of patients to differentiate between malignant and nonmalignant diseases. Materials and Methods: The present study was conducted on 30 patients with malignant pleural effusion and 30 patients with nonmalignant pleural effusion. Pleural fluid and blood samples were taken at the time of admission, before starting any treatment. Sialic acid levels were estimated in serum and pleural fluid by Warren's Thiobarbituric Acid (TBA) method. Results: In the present study, serum sialic acid levels were higher in group II as compared to group I. In the present study, pleural fluid (PF) sialic acid levels and PF/serum(S) ratio was higher in malignant pleural effusion (though difference was not statistically significant). Smokers in group II had higher serum sialic acid as compared to group 1 (P < 0.05). The sensitivity and specificity of PF/S sialic acid ratio with cut off value of 0.7 were 76.67% and 20%, respectively, whereas taking the cut off value of 70mg/dl for pleural fluid sialic acid in malignant pleural effusions, the sensitivity was 63.33%, specificity 60%, and positive predictive value 46.34%. Conclusion: These findings indicate that determination of sialic acid levels in pleural fluid has diagnostic value as a cheap, simple, and reliable marker for malignant pleural effusion. Lipid peroxidation may be involved in cancer and essential nutrients that can scavenge free radicals, such as vitamins E and C, operate in concert. Levels of antioxidant vitamins E and C were estimated in 50 patients with oral cancer and 24 healthy persons served as control. Significantly lower levels of vitamins E and C were observed in oral cancer patients as compared to controls (P < 0.011). Antioxidant nutrients may be utilized to a greater extent in oral cancer patients to counteract free radical-mediated cell disturbances, resulting in a reduction in salivary antioxidant levels.
