Oct-4/ Pou5f1/ Oct-3, a POU domain family protein acts as a crucial transcription factor duringembryonic development. It helps in maintenance of self renewal as well as pluripotentialstate of embryonic stem (ES) cells. Its expression starts right from 2 cell stage especiallyprior to 8 cell stage till the blastocyst stage where it is strongly expressed in inner cell mass(ICM). Thereafter, it is located predominantly in primordial germ cells (PGCs) till the birth. Ittargets particularly those genes which bear an octameric motif ATGCAAAT in promoter orenhancer region. Most of the target genes of Oct-4 are expressed in undifferentiated ES cellsand knockdown of Oct-4 results in ES cell differentiation as a result of down regulation oftargets of Oct-4 which are expressed in ES cells. Since, Oct-4 is crucial for embryo survivalits expression needs tight regulation. Oct-4 is carefully regulated epigenetically as well asby several other factors. DNA methylation and histone modification play an important role inexpression of Oct-4 while proximal promoter, enhancer and distal enhancer are the crucialregulatory elements present on Oct-4 upstream region. There is increasing evidence thatOct-4 is expressed in adult stem cells and these stem cells can get converted to cancer stemcells. Since, it is expressed in germ cells, immunohistochemical localization of Oct-4 andthereby its role as a marker for germ cell tumor detection is increasing. Thus, role of Oct-4 isnot only restricted as a marker for undifferentiated cells; it is also proving to be crucial factorwhich targets several genes involved in ES cell survival as well as help in establishing germcell origin of metastatic tumors.

Purpose: To study the clinical presentation and treatment outcomes of indocyanine green-enhanced transpupillary thermotherapy (ICG-TTT) for treatment-naïve juxtapapillary retinal capillary hemangioblastoma (JRCH). Methods: A prospective interventional case series. The technique involved ICG dye infusion 45 seconds prior to application of TTT. The main study outcomes were local tumor control, resolution of subretinal fluid (SRF), and improvement in best-corrected visual acuity (BCVA). Results: Eight eyes of seven patients (5 males and 2 females) were included. The mean age was 26 years (range: 5–56 years). Systemic evaluation revealed von-Hippel Lindau (VHL) disease in five patients. The most common location was the temporal aspect of the optic disc (5 eyes). The mean basal diameter was 2.9 mm (range: 1–8 mm), and tumor thickness was 1.4 mm (range: 1–4 mm). All eight eyes were treated with multiple sessions of ICG-TTT (mean: 3 sessions). Six eyes received adjuvant intravitreal injection of dexamethasone implant (4 eyes) and/or bevacizumab (4 eyes). Post treatment, six eyes (75%) had tumor regression with reduction of SRF. One eye had a partial response with persisting SRF, and one eye showed poor response to TTT for which external beam radiotherapy was performed. At the last follow-up (median: 11 months; range: 6–29 months), the BCVA remained stable in seven eyes and improved in one eye (hand motion to 20/40). Conclusion: Multiple ICG-TTT sessions can be considered as an alternative treatment option for JRCH with effective local tumor control and SRF resolution.

In the project, an emotion recognition system from speech is proposed using deep learning. The goal of this project is to classify a speech signal into one of the five emotions listed below: anger, boredom, fear, happiness, and sadness. Snippets below from numerous Marathi movies and TV shows were used to construct the dataset for Marathi language samples which include 20 audio samples for anger, 19 for boredom, 5 for fear, and 11 for happiness. The proposed system first processes a speech signal from the time domain to the frequency domain using Discrete Time Fourier Transform (DTFT). Then, data augmentation is performed which includes noise injection, stretching, shifting, and pitch scaling of the speech signal. Next, feature extraction is performed in which 5 features were selected, which include Mel Frequency Cepstral Coefficients (MFCC), Zero Crossing Rate (ZCR), Chroma STFT, Mel Spectrogram, and Root mean square value. These features were then fed to a Convolutional Neural Network (CNN). The efficiency of the suggested system employing the CNNs is supported by experimental findings. This model’s accuracy on the test data is 80.33%, and its f1 values for anger, boredom, fear, happiness, and sadness are 0.85, 0.83, 0.50, 0.62, and 0.84, respectively.

Abstract The abnormal tumor microenvironment (TME) often dictates the therapeutic response of cancer to chemo- and immuno-therapy. Aberrant expression of pericentromeric satellite repeats has been reported for epithelial cancers, including lung cancer. However, the transcription of tandemly repetitive elements in stromal cells of the TME has been unappreciated, limiting the optimal use of satellite transcripts as biomarkers or anti-cancer targets. We found that transcription of pericentromeric satellite DNA (satDNA) in mouse and human lung adenocarcinoma was observed in cancer-associated fibroblasts (CAFs). In vivo, lung fibroblasts expressed pericentromeric satellite repeats HS2/HS3 specifically in tumors. In vitro, transcription of satDNA was induced in lung fibroblasts in response to TGFβ, IL1α, matrix stiffness, direct contact with tumor cells and treatment with chemotherapeutic drugs. Single-cell transcriptome analysis of human lung adenocarcinoma confirmed that CAFs were the cell type with the highest number of satellite transcripts. Human HS2/HS3 pericentromeric transcripts were detected in the nucleus, cytoplasm, extracellularly and co-localized with extracellular vesicles in situ in human biopsies and activated fibroblasts in vitro. The transcripts were transmitted into recipient cells and entered their nuclei. Knock-down of satellite transcripts in human lung fibroblasts attenuated cellular senescence and blocked the formation of an inflammatory CAFs phenotype which resulted in the inhibition of their pro-tumorigenic functions. In sum, our data suggest that satellite long non-coding (lnc) RNAs are induced in CAFs, regulate expression of inflammatory genes and can be secreted from the cells, which potentially might present a new element of cell-cell communication in the TME.

Keshavasut (1866–1905), one of the earliest modern Marathi poets ‘translated’ the famous Edgar Allen Poe poem ‘The Raven’ into Marathi as ‘ghubad’ or the owl. Can the owl be an ‘equivalent’ for a raven? Can this process of transforming the raven into an owl be called translation? Whatever name you give for such a process of transformation, they definitely have played a crucial role in the development of the language of Marathi poetry towards the end of the twentieth century. Almost half a century later, the noted bilingual poet and translator Dilip Chitre (1938–2009) translated Baudelaire’s poem ‘The Owl ’into Marathi and discussed it by explicitly invoking the Keshasuta’s bird as well as Baudelaire’s fascination for Poe. Chitre called his translations of international modernist poets into Marathi ‘apabhranshas’. These apabhranshas’ were particularly influential and played a considerable role in the development of the language of modernist Marathi poetry. No history of literature or culture is complete if we do not take this process of transforming ravens into owls, the process of apabhransha, into account, as this is the process of artistic innovation and the process of reconstitution of cultural memory. In order to engage with the evolution of Marathi literature and culture, it is critical to move beyond the restrictive understanding of literature as a written or printed object, the notion which itself seems to be a translation of the colonial notion of literature. Literature and primarily poetry in India, implied performance, music, retelling, improvisation, and transmission of texts through oral traditions. Besides, it was constituted by intensive inter-medial, cross-lingual and cross-cultural intercourse, in short, by translational activity. We also need to revise our restrictive understanding of translation and equivalence as setting up of inter-lingual synonymy hounded by questions of fidelity and freedom. This restrictive notion of translation, too, seems to be a colonial import. We need a theoretical framework that allows us to see translation as an inter-medial, intercultural phenomenon inseparable from the questions of cultural memory, artistic innovation, and cultural change. The Soviet school of cultural studies, known as the Tartu Moscow School of cultural semiotics (or semiotics of culture), developed a full-fledged theory of culture under the leadership of Yuri Lotman (1922–1993) from the late 1960s which seems to have theoretically addressed these complex questions. In this paper, rather than merely providing a chronological account of the information regarding translation activity in Maharashtra, I intend to propose and lay out the methodological framework for writing and theorising the history of translation using the model of cultural semiotics.

36. Recessive loss‐of‐function variants in DPH1 identified as the molecular cause in a sibling pair previously diagnosed with Fine–Lubinsky syndrome.

- Abstract
Fine–Lubinsky syndrome is a rare clinically defined syndrome sometimes referred to as brachycephaly, deafness, cataract, microstomia, and impaired intellectual development syndrome. Here we provide a clinical and molecular update for a sibling pair diagnosed with Fine–Lubinsky syndrome. An extensive genetic work‐up, including chromosomal microarray analysis and quad exome sequencing, was nondiagnostic. However, a research reanalysis of their exome sequencing data revealed that both were homozygous for an intronic c.749+39G>A [NM_001383.6] variant in DPH1. RNAseq analysis performed on RNA from fibroblasts revealed significantly reduced expression of DPH1 transcripts suggestive of abnormal splicing followed by nonsense mediated mRNA decay. Since the phenotypes of this sibling pair were consistent with those associated with the inheritance of biallelic pathogenic variants in DPH1, they were given a diagnosis of developmental delay with short stature, dysmorphic facial features, and sparse hair 1 (DEDSSH1). This leads us to recommend that all individuals with a clinical diagnosis of Fine–Lubinsky syndrome be screened for variants in DPH1. The clinical histories of this sibling pair emphasize that hearing loss associated with DEDSSH1 may remit over time and that individuals with DEDSSH1 should be monitored for the development of cardiomyopathy. This case also demonstrates the clinical utility of RNAseq as a means of functionally validating the effects of intronic variants that may affect splicing. [ABSTRACT FROM AUTHOR]

- Abstract
Objective The All of Us Evenings with Genetics (EwG) Research Program at Baylor College of Medicine (BCM), funded to engage research scholars to work with the All of Us data, developed a training curriculum for the Researcher Workbench, the platform to access and analyze All of Us data. All of Us EwG developed the curriculum so that it could teach scholars regardless of their skills and background in programming languages and cloud computing. All of Us EwG delivered this curriculum at the first annual All of Us EwG Faculty Summit in May 2022. The curriculum was evaluated both during and after the Faculty Summit so that it could be improved for future training. Materials and Methods Surveys were administered to assess scholars' familiarity with the programming languages and computational tools required to use the Researcher Workbench. The curriculum was developed using backward design and was informed by the survey results, a review of available resources for training users on the Researcher Workbench, and All of Us EwG members' collective experience training students. The curriculum was evaluated using feedback surveys during the Faculty Summit as well as virtual meetings and emails following the Faculty Summit. Results The evaluation results demonstrated the success of the curriculum and identified areas for improvement. Discussion and Conclusion The curriculum has been adapted and improved in response to evaluations and in response to changes to the All of Us data and infrastructure to train more researchers through this program and other scholarly programs. Objective Educational offerings to fill the bioinformatics knowledge gap are a key component to enhancing access and use of health data from the All of Us Research Program. We developed a Train the Trainer-based, innovative training series including project-based learning, modular on-demand demonstrations, and unstructured tutorial time as a model for educational engagement in the All of Us community. Materials and Methods We highlight our training modules and content, with training survey data informing cycles of development in the creation of a 6-module training series with modular demonstrations. Results We have conducted 2 public iterations of the Train the Trainer (Tx3) Series based on survey feedback while training over 300 registered researchers to access and analyze data on the All of Us Researcher Workbench. Discussion and Conclusion Future directions of the Tx3 Series include enhanced focus on project-based learning and learner requests for modularity and asynchronous materials access. Age-related illnesses, including hypertension and accompanying metabolic disorders, compromise immunity and exacerbate infection-associated fatalities. Renin-angiotensin system (RAS) is the key mechanism that controls blood pressure. Upregulation of RAS through angiotensin receptor type 1 (AT1R), a G-protein coupled receptor, contributes to the pathophysiological consequences leading to vascular remodeling, hypertension, and end-organ damage. Genetic variations that increase the expression of human AT1R may cause the above pathological outcomes associated with hypertension. Previously we have shown that our chronically hypertensive transgenic (TG) mice containing the haplotype-I variant (Hap-I, hypertensive genotype) of human AT1R (hAT1R) gene are more prone to develop the metabolic syndrome-related disorders as compared to the TG mice containing the haplotype-II variant (Hap-II, normotensive genotype). Since aging and an increased risk of hypertension can impact multiple organ systems in a complex manner, including susceptibility to various infections, the current study investigated the susceptibility and potential effect of acute bacterial infection using a Gram-negative intracellular bacterial pathogen, Francisella tularensis in our hAT1R TG mice. Our results show that compared to Hap-II, F. tularensis-infected aged Hap-I TG mice have significantly higher mortality post-infection, higher bacterial load and lung pathology, elevated inflammatory cytokines and altered gene expression profile favoring hypertension and inflammation. Consistent with worsened phenotype in aged Hap-I mice post-Francisella infection, gene expression profiles from their lungs revealed significantly altered expression of more than 1,400 genes. Furthermore, bioinformatics analysis identified genes associated with RAS and IFN-γ pathways regulating blood pressure and inflammation. These studies demonstrate that haplotype-dependent over-expression of the hAT1R gene leads to enhanced susceptibility and lethality due to F. tularensis LVS infection, which gets aggravated in aged animals. Clinically, these findings will help in exploring the role of AT1R-induced hypertension and enhanced susceptibility to infection-related respiratory diseases.

Introduction: Medical education aims at training an individual with respect to the knowledge, clinical skills, communication skills, and soft skills and building the highest standards of morale. While majority of training programs offer considerable technical skills only a few incorporate the remaining assets. Methods: A curriculum innovation project “Foundation Course” aiming to achieve the above goals, designed for the students entering a medical college was conducted for 74 students joining M. B. B. S. course in a Government Medical College in Port Blair in the year 2015. The feedback from students and teachers was collected at the end of entire activity in the form of a specially designed structured questionnaire with open as well as close-ended questions. Qualitative analysis was performed for students' and teachers' feedback on the open-ended questions. Results: The response rate was 100%. Students perceived foundation course as an excellent opportunity to acquire basic knowledge, attitude, and skills required for subsequent phases in MBBS course. The students opined that teachers who were friendly and approachable enhanced their learning. The attitude of teachers, the methodology of teaching, and the cooperation of peers all played a significant role in enhancing the learning. This course was helpful and increased their orientation to the curriculum and eased out the apprehension to cope up with the challenges. Conclusion: This foundation course enhanced their orientation to the curriculum and campus, boosted confidence to cope up with the challenging syllabus, helped them develop time and stress management skills, and sensitized them to achieve the highest standards of morale required to ensure ethical practice. Students' feedback revealed that how the faculty taught and interacted with the students had more impact than the content.

Wound healing in the skin is a complex physiological process that is a product of a cell state transition from homeostasis to repair. Mechanical cues are increasingly being recognized as important regulators of cellular reprogramming, but the mechanism by which it is translated to changes in gene expression and ultimately cellular behavior remains largely a mystery. To probe the molecular underpinnings of this phenomenon further, we used the down-regulation of caspase-8 as a biomarker of a cell entering the wound healing program. We found that the wound-induced release of tension within the epidermis leads to the alteration of gene expression via the nuclear translocation of the DNA methyltransferase 3A (DNMT3a). This enzyme then methylates promoters of genes that are known to be down-regulated in response to wound stimuli as well as potentially novel players in the repair program. Overall, these findings illuminate the convergence of mechanical and epigenetic signaling modules that are important regulators of the transcriptome landscape required to initiate the tissue repair process in the differentiated layers of the epidermis.

Background: Breast cancer is the leading cancer diagnosed in Indian women. Lifestyle related factors such as high body mass index (BMI) and obesity have been recognized as major risk factors for the development of breast cancer. However, India has higher proportion of underweight population and recently positive correlation has been reported between underweight and increased risk of breast cancer. We have attempted to study an association between low BMI and total body fat percentage with breast cancer risk by performing retrospective analysis on a small sample size of 41 female patients diagnosed with breast cancer. The data was collected from Department of Oncology, Bharati Vidyapeeth Hospital and Research Centre (BVHRC), Pune, India. Methods: Binary logistic regression was performed to estimate odds ratios (ORs) and to examine the predictive effect of each factor on the breast cancer risk. Results: It was observed that underweight population displayed higher risk of breast cancer development based on BMI (OR-15.40) and body fat % (OR-1.33). Conclusion: This pilot study suggests that low body mass index may be related to poor prognosis in breast cancer and thus warrants further studies on a larger sample size to establish a positive correlation.

An asymmetric zinc phthalocyanine dye (ZnPcT3C), bearing three tetraethylene glycol donor groups and one carboxylic acid anchoring group, was synthesized as a photosensitizer dye for dye‐sensitized solar cells (DSSCs). The tetraethylene glycol (TEG), consisting of four ethoxy units, works as an amphiphilic long‐chain donor group that greatly helped in reducing the molecular aggregation. Carboxylic acid group, on the other hand, an acceptor, together with TEG in ZnPcT3C functions as a 'push–pullʼ system suitable for DSSCs. Absorption spectra of ZnPcT3C in DMSO showed a strong sharp Q band in the infrared region 600–700 nm with (λmax = 682 nm) and a less intense soret band appeared in the region 300–400 nm with λmax = 340 nm. The zinc phthalocyanine (ZnPcT3C) exhibited molar extinction coefficient (ε) of 72,727 L mol−1cm−1. The emission was observed at λem = 695 nm upon excitation at 650 nm and exhibited a fluorescence decay of 2.82 ns. The ZnPcT3C dye sensitised solar cell (c = 1 mM) exhibited the power conversion efficiency (η) of 3.52% in chloroform in I−/I3 electrolyte under simulated 100% brightness. Cosensitization of ZnPcT3C with another auxiliary dye D131 in 1:1 ratio in a cocktail‐type DSSC attained the power conversion efficiency of twice as high as η = 6.27% in an identical condition. A mixture of D131 (λmax = 470 nm) dye with the ZnPcT3C phthalocyanine harvests sunlight across the visible spectra, enabling DSSCs to attain a large photocurrent and photovoltage, enhancing power conversion efficiency.
