Abstract

Nitrogen oxides, emitted from air traffic, are of concern due to their impact on climate by changing atmospheric ozone and methane levels. Using the DLR research aircraft Falcon, total reactive nitrogen (NOy) in-flight measurements were carried out at high altitudes to characterize emissions in the fresh aircraft exhaust from the latest-generation Rolls-Royce Trent XWB-84 engine aboard the long-range Airbus A350-941 aircraft during the ECLIF3 (Emission and CLimate Impact of alternative Fuels 3) experiment. The impact of different engine thrust settings, monitored in terms of combustor inlet temperature, pressure and engine fuel flow, was tested for two different fuel types: Jet A-1 and, for the first time, a 100 % sustainable aviation fuel (SAF) under similar atmospheric conditions. In addition, a range of combustor temperatures and an additional blended SAF were tested during ground-based emission measurements. For the data measured during ECLIF3, we confirm that the NOx emission index increases with increasing combustion temperature, pressure and fuel flow. We find that as expected, the fuel type has no measurable effect on the NOx emission index. These measurements are used to compare to cruise NOx emission index estimates from three engine emission prediction methods. Our measurements thus help to understand the ground to cruise correlation of current engine emission prediction methods while serving as input for climate modelling and extending the extremely sparse data set on in-flight aircraft nitrogen oxide emissions to newer engine generations. Abstract

In order to reduce aviation's CO 2 emissions and comply with current climate targets, the European Union plans a mandatory quota of 2 % sustainable aviation fuel (SAF) by 2025, rising up to ≥70 % SAF by 2050. In addition to a reduction of life cycle CO 2 emissions, the use of SAF can also have a positive impact on particle emissions and contrail properties. In this study we present observations from the ECLIF3 (Emission and CLimate Impact of alternative Fuels) aircraft campaign, which investigated exhaust and contrail characteristics of an Airbus A350-941 equipped with Rolls-Royce Trent XWB-84 engines. For the first time, non-volatile and total particle emissions of 100 % HEFA-SPK (hydroprocessed esters and fatty acids–synthetic paraffinic kerosene) SAF, a blended fuel and a reference Jet A-1 fuel were measured in flight. A maximum reduction in non-volatile particle number emissions of ∼41 % compared to the reference Jet A-1 fuel was measured at low cruise engine power settings when using 100 % HEFA-SPK. The reduction decreases to ∼29 % for typical cruise engine settings and to ∼22 % at high cruise engine power settings. The size of non-volatile particles was slightly smaller for HEFA-SPK compared to Jet A-1. We show a comprehensive analysis of the hydrogen content of globally available fuels. Our results demonstrate the impact of the fuel composition in terms of its aromatic, hydrogen, and sulfur content as well as of the effect of engine power settings on particle emissions. We demonstrate that the use of HEFA-SPK can significantly reduce particle emissions and thus contrail ice particles and therefore can provide an aviation climate benefit. [ABSTRACT FROM AUTHOR]

Abstract

Goats are natural hosts of Mycobacterium (M.) bovis, and affected herds can be the cause of significant economic losses. Similarites in disease course and lesions of M. bovis infections in goats and M. tuberculosis in humans make goats good models for human tuberculosis. The aim of this investigation was to characterize M. bovis challenge models in goats. For this, goats were endobronchially inoculated with three doses of M. bovis or culture medium. Clinical signs, shedding, and immune responses were monitored until 146 days post inoculation (dpi). At necropsy, lesions were examined by computed tomography, histology, and bacteriological culture. Infected goats did not develop clinical signs. M. bovis was cultured from feces, but never from nasal swabs. IGRAs were positive from 28 dpi onwards, antibodies at 140 dpi, and SICCT at 146 dpi. The increase in CD25+, IFN-γ+, and IFN-γ-releasing T-cell subpopulations was time-related, but not dose-dependent. All infected goats developed paucibacillary granulomas in the lungs and regional lymph nodes. M. bovis was regularly cultured. Dose-dependent effects included the size of pulmonary lesions, caverns, intestinal lesions, and early generalization in the high-dose group. Abstract

Ferritin, a key regulator of iron homeostasis in macrophages, has been reported to confer host defenses against Mycobacterium tuberculosis (Mtb) infection. Nuclear receptor coactivator 4 (NCOA4) was recently identified as a cargo receptor in ferritin degradation. Here, we show that Mtb infection enhanced NCOA4-mediated ferritin degradation in macrophages, which in turn increased the bioavailability of iron to intracellular Mtb and therefore promoted bacterial growth. Of clinical relevance, the upregulation of FTH1 in macrophages was associated with tuberculosis (TB) disease progression in humans. Mechanistically, Mtb infection enhanced NCOA4-mediated ferritin degradation through p38/AKT1- and TRIM21-mediated proteasomal degradation of HERC2, an E3 ligase of NCOA4. Finally, we confirmed that NCOA4 deficiency in myeloid cells expedites the clearance of Mtb infection in a murine model. Together, our findings revealed a strategy by which Mtb hijacks host ferritin metabolism for its own intracellular survival. Abstract

The mononuclear phagocyte system includes monocytes, macrophages, some dendritic cells, and multinuclear giant cells. These cell populations display marked heterogeneity depending on their differentiation from embryonic and bone marrow hematopoietic progenitors, tissue location, and activation. They contribute to tissue homeostasis by interacting with local and systemic immune and non-immune cells through trophic, clearance, and cytocidal functions. During evolution, they contributed to the innate host defense before effector mechanisms of specific adaptive immunity emerged. Mouse macrophages appear at mid-gestation and are distributed throughout the embryo to facilitate organogenesis and clear cells undergoing programmed cell death. Yolk sac, AGM, and fetal liver-derived tissue-resident macrophages persist throughout postnatal and adult life, supplemented by bone marrow-derived blood monocytes, as required after injury and infection. Nobel awards to Elie Metchnikoff and Paul Ehrlich in 1908 drew attention to cellular phagocytic and humoral immunity, respectively. In 2011, prizes were awarded to Jules Hoffmann and Bruce Beutler for contributions to innate immunity and to Ralph Steinman for the discovery of dendritic cells and their role in antigen presentation to T lymphocytes. Abstract

Carl Flügge is best known for the promotion of studies demonstrating the transmission of all manner of infections, but particularly tuberculosis, by coughed droplets. But it is seldom recognised that Flügge was also influential in a number of other fields comprising the practice of hygiene. One-hundred years following his death in 1923, we review literature related to the studies of Flügge and his colleagues and students and illustrate the particular emphasis he laid upon the environment within which disease and its transmission might be fostered or prevented, embracing and studying aspects essential to the health of any community ranging from fundamental microbiology in the laboratory to subjects as disparate as housing, clean water supply, nutrition, sanitation, socio-economic circumstances and climate. Very early in his career he promoted breast feeding for the prevention of seasonal gastro-enteritis and later the sheltering of cough as a means of preventing the transmission of infected respiratory droplets, not only as regards tuberculosis, but also concerning all manner of other respiratory infections. By the time of Flügge's death the complexification of available scientific methodologies comprising hygiene made it difficult for any individual to comprehend and study the wide range of hygiene-related subjects such as Flügge did. Abstract

The critical role of suppressive myeloid cells in immune regulation has come to the forefront in cancer research, with myeloid-derived suppressor cells (MDSCs) as a main oncology immunotherapeutic target. Recent improvement and standardization of criteria classifying tumor-induced MDSCs have led to unified descriptions and also promoted MDSC research in tuberculosis (TB) and AIDS. Despite convincing evidence on the induction of MDSCs by pathogen-derived molecules and inflammatory mediators in TB and AIDS, very little attention has been given to their therapeutic modulation or roles in vaccination in these diseases. Clinical manifestations in TB are consequences of complex host- pathogen interactions and are substantially affected by HIV infection. Here we summarize the current understanding and knowledge gaps regarding the role of MDSCs in HIV and Mycobacterium tuberculosis (co)infections. We discuss key scientific priorities to enable application of this knowledge to the development of novel strategies to improve vaccine efficacy and/or implementation of enhanced treatment approaches. Abstract

Powering aircraft by sustainable aviation fuels (SAFs) is a pathway to reduce the climate impact of aviation by lowering aviation lifecycle CO2 emissions and by reducing ice crystal numbers and radiative forcing from contrails. While the effect of SAF blends on contrails has been measured previously, here we present novel measurements on particle emission and contrails from 100 % SAF combustion. During the ECLIF3 (Emission and CLimate Impact of alternative Fuels) campaign, a collaboration between the Deutsches Zentrum für Luft- und Raumfahrt (DLR), Airbus, Rolls-Royce, and Neste, the DLR Falcon 20 research aircraft performed in situ measurements following an Airbus A350-941 source aircraft powered by Rolls-Royce Trent XWB-84 engines in 1 to 2 min old contrails at cruise altitudes. Apparent ice emission indices of 100 % HEFA-SPK (hydro-processed esters and fatty acids–synthetic paraffinic kerosene) were measured and compared to Jet A-1 fuel contrails at similar engine and ambient ice-supersaturated conditions within a single flight. A 56 % reduction in ice particle numbers per mass of burned fuel was measured for 100 % HEFA-SPK compared to Jet A-1 under engine cruise conditions. The measured 35 % reduction in soot particle numbers suggests reduced ice activation by the low-sulfur HEFA fuel. Contrail properties are consistently modeled with a contrail plume model. Global climate model simulations for the 2018 fleet conservatively estimate a 26 % decrease in contrail radiative forcing and stronger decreases for larger particle reductions. Our results indicate that higher hydrogen content fuels as well as clean engines with low particle emissions may lead to reduced climate forcing from contrails. [ABSTRACT FROM AUTHOR]
