Introduction: CD138 (syndecan-1) expression has been confirmed in plasma cells and normal tissues, particularly in epithelial cell types. As a cell surface protein, CD138 regulates cell-cell and cell-matrix interactions. CD138 may also regulate cell proliferation, migration, and cytoskeletal organization. In addition, positive CD138 plasma cells can also be detected with extreme infiltration in inflammatory diseases such as chronic endometritis. Purpose: Investigation of the prognostic significance of the infiltration of CD 138 plasma cells in endoscopic material from patients with ulcerative colitis (UC) and Crohn's disease (CD). Materials and methods: The study group included 34 patients with ulcerative colitis and 8 patients diagnosed with CD. The expression of CD138 was performed by immunohistochemistry and assessed as a membrane-cytoplasmic color reaction in plasma cells localized in the lamina propria and glandular epithelium. We also examined the insensitivity of general CD138-positive cells and divided them into 4 groups: 0-absent, 1-mild increase, 2-moderate increase, and 3-marked increase. Results: The study group consists of 16 females and 18 males in UC and 2 female and 6 male of CD. The UC lesions were mainly located in the rectum (19/34 cases), sigmoid (8/34cases), and others (7/34 cases). Crohn's disease was observed in the small intestine (3/8 cases), caecum (4/8 cases) and rectum (1/8 cases). The inflammatory infiltration consists of CD138+ cells in less than 90% of inflammatory cells in UC (median 52.39%) and in less than 60% of inflammatory cells in CD (36.50%). The increase in CD138+ plasma cells in UC was mild in 3 cases, moderate in 10 cases and marked in 15 cases. CD138+ plasma cells in UC were found to correlate with Geboes score (R=0.404), CD15+ neutrophils (R=0.467) and CD15+macrophages (R=0.459). Plasma cells were slightly increased in 2 cases and moderately increased in 5 cases of CD. Infiltration of CD138+ cells in CD sections was associated with age (R= - 0.481) and crypt destruction (R=0.606). Loss of CD138 expression in glandular epithelium was detected in 4/38 cases of UC and 1/8 cases of CD. Conclusion: Our results showed that infiltration of CD138-positive plasma cells is closely related to the degree of inflammation and architectural changes that could allow anti-CD138 treatment in inflammatory bowel disease. Thaumetopoea pityocampa (pine processionary moth) is one of the most important pine pests in the forests of Mediterranean countries, Central Europe, the Middle East and North Africa. Apart from causing significant damage to pinewoods, T. pityocampa occurrence is also an issue for public and animal health, as it is responsible for dermatological reactions in humans and animals by contact with its irritating hairs. High throughput sequencing technologies have allowed the fast and cost-effective generation of genetic information of interest to understand different biological aspects of non-model organisms as well as the identification of potential pathogens. Using these technologies, we have obtained and characterized the transcriptome of T. pityocampa larvae collected in 12 different geographical locations in Turkey. cDNA libraries for Illumina sequencing were prepared from four larval tissues, head, gut, fat body and integument. By pooling the sequences from Illumina platform with those previously published using the Roche 454-FLX and Sanger methods we generated the largest reference transcriptome of T. pityocampa. In addition, this study has also allowed identification of possible viral pathogens with potential application in future biocontrol strategies.

The baculovirus expression vector system (BEVS) has been widely used to produce a large number of recombinant proteins, and is becoming one of the most powerful, robust, and cost-effective systems for the production of eukaryotic proteins. Nevertheless, as in any other protein expression system, it is important to improve the production capabilities of this vector. The orf46 viral gene was identified among the most highly abundant sequences in the transcriptome of Spodoptera exigua larvae infected with its native baculovirus, the S. exigua multiple nucleopolyhedrovirus (SeMNPV). Different sequences upstream of the orf46 gene were cloned, and their promoter activities were tested by the expression of the GFP reporter gene using the Autographa californica nucleopolyhedrovirus (AcMNPV) vector system in different insect cell lines (Sf21, Se301, and Hi5) and in larvae from S. exigua and Trichoplusia ni. The strongest promoter activity was defined by a 120 nt sequence upstream of the ATG start codon for the orf46 gene. On average, GFP expression under this new promoter was more than two fold higher than the expression obtained with the standard polyhedrin (polh) promoter. Additionally, the orf46 promoter was also tested in combination with the polh promoter, revealing an additive effect over the polh promoter activity. In conclusion, this new characterized promoter represents an excellent alternative to the most commonly used baculovirus promoters for the efficient expression of recombinant proteins using the BEVS.

Virus transmission and the prevalence of infection depend on multiple factors, including the interaction with other viral pathogens infecting the same host. In this study, active replication of an iflavirus, Spodoptera exigua iflavirus 1 (order Picornavirales) was observed in the offspring of insects that survived following inoculation with a pathogenic baculovirus, Spodoptera exigua multiple nucleopolyhedrovirus. Tracking the origin of the iflavirus suggested the association of this virus with the occlusion bodies of the baculovirus. Here we investigated the effect of this association on the stability and infectivity of both viruses. A reduction in baculovirus pathogenicity, without affecting its infectivity and productivity, was observed when associated with the iflavirus. In contrast, viral association increased the infectivity of the iflavirus and its resistance to ultraviolet radiation and high temperature, two of the main factors affecting virus stability in the field. In addition, electron microscopy analysis revealed the presence of particles resembling iflavirus virions inside the occlusion bodies of the baculovirus, suggesting the possible co-occlusion of both viruses. Results reported here are indicative of facultative phoresis of a virus and suggest that virus–virus interactions may be more common than currently recognized, and may be influential in the ecology of baculovirus and host populations and in consequence in the use of baculoviruses as biological insecticides.

Bracoviruses are symbiotic viruses associated with tens of thousands of species of parasitic wasps that develop within the body of lepidopteran hosts and that collectively parasitize caterpillars of virtually every lepidopteran species. Viral particles are produced in the wasp ovaries and injected into host larvae with the wasp eggs. Once in the host body, the viral DNA circles enclosed in the particles integrate into lepidopteran host cell DNA. Here we show that bracovirus DNA sequences have been inserted repeatedly into lepidopteran genomes, indicating this viral DNA can also enter germline cells. The original mode of Horizontal Gene Transfer (HGT) unveiled here is based on the integrative properties of an endogenous virus that has evolved as a gene transfer agent within parasitic wasp genomes for ≈100 million years. Among the bracovirus genes thus transferred, a phylogenetic analysis indicated that those encoding C-type-lectins most likely originated from the wasp gene set, showing that a bracovirus-mediated gene flux exists between the 2 insect orders Hymenoptera and Lepidoptera. Furthermore, the acquisition of bracovirus sequences that can be expressed by Lepidoptera has resulted in the domestication of several genes that could result in adaptive advantages for the host. Indeed, functional analyses suggest that two of the acquired genes could have a protective role against a common pathogen in the field, baculovirus. From these results, we hypothesize that bracovirus-mediated HGT has played an important role in the evolutionary arms race between Lepidoptera and their pathogens.

Antimicrobial peptides (AMPs) and lysozymes are the main effectors of the insect immune system, and they are involved in both local and systemic responses. Among local responses, midgut immune reaction plays an important role in fighting pathogens that reach the insect body through the oral route, as do many microorganisms used in pest control. Under this point of view, understanding how insects defend themselves locally during the first phases of infections caused by food-borne pathogens is important to further improve microbial control strategies. In the present study, we analyzed the transcriptional response of AMPs and lysozymes in the midgut of Spodoptera exigua (Lepidoptera: Noctuidae), a polyphagous pest that is commonly controlled by products based on Bacillus thuringiensis (Bt) or baculovirus. First, we comprehensively characterized the transcripts encoding AMPs and lysozymes expressed in S. exigua larval midgut, identifying 35 transcripts that represent the S. exigua arsenal against microbial infection. Secondly, we analyzed their expression in the midgut after ingestion of sub-lethal doses of two different pore-forming B. thuringiensis toxins, Cry1Ca and Vip3Aa, and the S. exigua nucleopolyhedrovirus (SeMNPV). We observed that both Bt toxins triggered a similar, wide and in some cases high transcriptional activation of genes encoding AMPs and lysozymes, which was not reflected in the activation of the classical systemic immune-marker phenoloxidase in hemolymph. Baculovirus ingestion resulted in the opposed reaction: Almost all transcripts coding for AMPs and lysozymes were down-regulated or not induced 96 hours post infection. Our results shed light on midgut response to different virulence factors or pathogens used nowadays as microbial control agents and point out the importance of the midgut immune response contribution to the larval immunity.

Host-pathogen interactions result in complex relationship, many aspects of which are not completely understood. Vip proteins, which are Bacillus thuringensis (Bt) insecticidal toxins produced during the vegetative stage, are selectively effective against specific insect pests. This new group of Bt proteins represents an interesting alternative to the classical Bt Cry toxins because current data suggests that they do not share the same mode of action. We have designed and developed a genome-wide microarray for the beet armyworm Spodoptera exigua, a serious lepidopteran pest of many agricultural crops, and used it to better understand how lepidopteran larvae respond to the treatment with the insecticidal protein Vip3Aa. With this approach, the goal of our study was to evaluate the changes in gene expression levels caused by treatment with sublethal doses of Vip3Aa (causing 99% growth inhibition) at 8 and 24 h after feeding. Results indicated that the toxin provoked a wide transcriptional response, with 19% of the microarray unigenes responding significantly to treatment. The number of up- and down-regulated unigenes was very similar. The number of genes whose expression was regulated at 8 h was similar to the number of genes whose expression was regulated after 24 h of treatment. The up-regulated sequences were enriched for genes involved in innate immune response and in pathogen response such as antimicrobial peptides (AMPs) and repat genes. The down-regulated sequences were mainly unigenes with homology to genes involved in metabolism. Genes related to the mode of action of Bt Cry proteins were found, in general, to be slightly overexpressed. The present study is the first genome-wide analysis of the response of lepidopteran insects to Vip3Aa intoxication. An insight into the molecular mechanisms and components related to Vip intoxication will allow designing of more effective management strategies for pest control.

Spodoptera exigua microarray was used to determine genes differentially expressed in S. exigua cells challenged with the species-specific baculovirus SeMNPV as well as with a generalist baculovirus, AcMNPV. Microarray results revealed that, in contrast to the host transcriptional shut-off that is expected during baculovirus infection, S. exigua cells showed a balanced number of up- and down-regulated genes during the first 36 hours following the infection. Many immune-related genes, including pattern recognition proteins, genes involved in signalling and immune pathways as well as immune effectors and genes coding for proteins involved in the melanization cascade were found to be down-regulated after baculovirus infection. The down-regulation of immune-related genes was confirmed in the larval gut. The expression of immune-related genes in the gut is known to affect the status of gut microorganisms, many of which are responsible for growth and development functions. We therefore asked whether the down-regulation that occurs after baculovirus infection affects the amount of gut microbiota. An increase in the gut bacterial load was observed and we hypothesize this to be as a consequence of viral infection. Subsequent experiments on virus performance in the presence and absence of gut microbiota revealed that gut bacteria enhanced baculovirus virulence, pathogenicity and dispersion. We discuss the host immune response processes and pathways affected by baculoviruses, as well as the role of gut microbiota in viral infection.

Nowadays, Hepatitis B X interacting protein (HBXIP) is an object of scientists' interest worldwide. It is a protein with significant involvement in the development of malignant tumors like breast or ovarian cancer. One of the most important functions of HBXIP is the regulation of cell proliferation, which is related to the progression of a cell cycle. Many studies provide the growing number of evidence that HBXIP plays various important roles, including the regulation of a cell cycle through complexes with survivin, belonging to the inhibitors of apoptosis and interactions with transcriptional factors like STAT4, SP1, TFIID or E2F1. It also has the influence on the promotion of tumor angiogenesis thanks to the association with VEGF and FGF8. Another important role of HBXIP is a reprogramming of glucose metabolism to conditions favorable to growing cancerous cells due to regulating the activation of SCO2 and PDHA1. Furthermore, it impacts on the complementdependent cytotoxicity, also, HBXIP affects on lipid metabolism through disturbing of metabolic pathways of FAS. According to recent studies, HBXIP can be used as a prognostic biomarker in many tumors, including cervical cancer, ovarian cancer, and esophageal squamous cell carcinoma thanks to the high expression of this protein noted exclusively in these tumor tissues. What is even more interesting, it significantly correlates with clinical attributes like metastasis to lymph nodes or grading and in some cases can potentially be used as the indicator of prognosis of treatment effectiveness. Introduction: Expression of Epo, a glycoprotein secreted by the fetal liver and the adult kidney in response to cellular hypoxia and its receptor have been described in human solid tumors, such as colon and breast cancer. Purpose: Since activation of Epo-EpoR signaling pathway in erythroid progenitor and precursor cells leads to promotion of proliferation and differentiation or prevention of programmed cell death through Bcl-xl and Bcl-2 it was of interest to investigate expression of Epo, EpoR, apoptosis regulator - Bax and marker of proliferating cells - Ki-67 and assess correlation between them, with regard to clinicopathological variables of colorectal cancer. Materials and methods: The correlations between expression of Epo, EpoR, Bax and Ki-67 in colorectal cancer were analyzed in regard to patient age, sex, primary localization, histopathological type, grading, staging and lymph node invasion. Statistical analyses were performed by using the Spearman rank correlation test applying a significance level of p<0,05. Results: Correlation between Bax and EpoR is positive and statistically significant at all groups of patients except group pT1+pT2. Positive correlation between Bax and Epo is statistically significant at following groups of patients: all patients, age ≤ 60, age >60, male, female, primary localization in rectum, primary localization in colon, adenocarcinoma, G2, G3. Statistical analysis revealed no significant correlations between expression of neither Ki-67 with Epo nor Ki-67 with EpoR in all groups of patients. Conclusions: Epo seems to be a pleiotropic cytokine, which can exert its biological effect on several cell types, including neoplastic cells. The refraction index of water at megabar pressures was calculated ab initio with the quantum molecular dynamics (QMD) package ABINIT using the projector augmented (PAW) formalism. Calculations were compared to experimental results obtained by laser-driven shocks on water in standard conditions and on water samples statically precompressed at . The refraction index was measured in transparent and opaque states of water using a VISAR diagnostics. We also modelled the data using an extended Lorentz-Drude model. At high compressions, a strong increase of the refraction index is observed both in experimental results and in the theoretical calculations, which is an indication of water approaching band gap closure. In particular, we addressed the generation of hot electrons (HE) (determining their average energy and number), as well as the parametric instabilities which are producing them. We studied D + D fusion reactions at the CELIA/ECLIPSE laser facility. These reactions were triggered by focusing the ECLIPSE laser (up to 100 mJ on target, pulse duration of 40 fs and focal spot of FWHM) on a solid C2D4 target. By detecting the 2.5 MeV neutrons, produced in the D + D fusion reaction, using C6D6 scintillators in a time-of-flight experimental set-up, we measured a rate of a few hundred fusions per shot. At the same time, we detected also energetic ions (up to a few 100 keV) using time-of-flight diamond detectors. The goal of this paper is twofold: first, we demonstrate shock generation with ultra-short (24 fs) and low-energy (J) laser pulse, following the energy deposition in the target by fast electrons; second, we show that such shocks can be used to provide information on compressed matter. For a target with thickness we have clearly inferred the formation of a shock wave with pressure . We have also measured the color temperature of the emitting target rear side at breakout time , which is in good agreement with predictions from equation-of-state models (SESAME tables) and hydrodynamic simulations. We compressed water to megabar pressures by laser-driven shock waves and evidenced transparent, opaque and reflecting phases as pressure increases. The refraction index of water in the first two states was measured using a VISAR system. At high compression a sharp increase of the real and imaginary part of the refraction index is observed. The and 3p–4d inner shell excitation lines in addition to 2p–3s lines have been identified from the spectrum taken by an upgraded high-resolution x-ray spectrometer. It is found from analysis of the absolute intensities of the and lines that W and Mo concentrations are in the range of and , respectively, with a ratio of ∼5% in JET with the ITER-like wall configuration for ELMy H-mode plasmas with a plasma current of 2.0–2.5 MA, a toroidal magnetic field of 2.7 T and a neutral beam injection power of 14–18 MW. For the purpose of checking self-consistency, it is confirmed that the W concentration determined from the line is in agreement with that from the line within 20% and that the plasma effective charge determined from the continuum of the first order reflection spectrum is also in agreement with that from the second order within 50%. Further, the determined plasma effective charge is in agreement with that determined from a visible spectroscopy, confirming that the sensitivity of the x-ray spectrometer is valid and that the W and the Mo concentrations are also likely to be valid.
