Abstract Cognitive dysfunction is not only a common symptom of major depressive disorder, but also a more common residual symptom after antidepressant treatment and a risk factor for chronic and recurrent disease. The disruption of hypocretin regulation is known to be associated with depression, however, their exact correlation is remains to be elucidated. Hypocretin‐1 levels are increased in the plasma and hypothalamus from chronic unpredictable mild stress (CUMS) model mice. Excessive hypocretin‐1 conducted with hypocretin receptor 1 (HCRTR1) reduced lactate production and brain‐derived neurotrophic factor (BDNF) expression by hypoxia‐inducible factor‐1α (HIF‐1α), thus impairing adult hippocampal neuroplasticity, and cognitive impairment in CUMS model. Subsequently, it is found that HCRTR1 antagonists can reverse these changes. The direct effect of hypocretin‐1 on hippocampal lactate production and cognitive behavior is further confirmed by intraventricular injection of hypocretin‐1 and microPET‐CT in rats. In addition, these mechanisms are further validated in astrocytes and neurons in vitro. Moreover, these phenotypes and changes in molecules of lactate transport pathway can be duplicated by specifically knockdown of HCRTR1 in hippocampal astrocytes. In summary, the results provide molecular and functional insights for involvement of hypocretin‐1‐HCRTR1 in altered cognitive function in depression.

With the widespread application of carbon dots (CDs) in fluorescence imaging, their toxicity has become a focal point of concern. The potential toxicity of CDs synthesized from different raw materials remains an unresolved issue. Laver and wakame, which are commonly popular sea vegetable foods rich in nutrients, were utilized to investigate whether synthetic CDs derived from these alga sources retain medicinal value. Herein, two types of fluorescent alga-derived CDs were prepared through hydrothermal synthesis using laver and wakame respectively. Zebrafish were immersed in both types of CDs to observe their fluorescence imaging effects within the zebrafish bodies. It was observed that laver-derived CDs and wakame-derived CDs exhibited similar luminescence properties but differed in terms of fish egg imaging localization. Additionally, intestinal flora sequencing revealed varying degrees of influence on the zebrafish gut microbiota by the two types of CDs, suggesting that both alga-derived CDs could enhance the abundance of intestinal flora in zebrafish.

Abstract Accumulating studies have shown the effects of gut microbiota management tools in improving depression. We conducted a meta-analysis to evaluate the effects of prebiotics, probiotics, and synbiotics on patients with depression. We searched six databases up to July 2022. In total, 13 randomized controlled trials (RCTs) with 786 participants were included. The overall results demonstrated that patients who received prebiotics, probiotics or synbiotics had significantly improved symptoms of depression compared with those in the placebo group. However, subgroup analysis only confirmed the significant antidepressant effects of agents that contained probiotics. In addition, patients with mild or moderate depression could both benefit from the treatment. Studies with a lower proportion of females reported stronger effects for alleviating depressive symptoms. In conclusion, agents that manipulate gut microbiota might improve mild-to-moderate depression. It is necessary to further investigate the benefits of prebiotic, probiotic and synbiotic treatments relative to antidepressants and follow up with individuals over a longer time before these therapies are implemented in clinical practice.

Aeschynomene indica rhizobia (AIRs) are special classes of bacteria capable of nodulating without nodulation factors and have photosynthetic capacity. With an aim to characterize the structural variations in Bradyrhizobium genomes during its evolution, the genomes of AIRs and the reference Bradyrhizobium strains were compared utilizing molecular biology, bioinformatics, and biochemistry techniques. The presence of symbiotic nitrogen fixation (nif) genes and photosynthetic genes, as well as components of the T3SS (Type III secretion system) and T3CP (Type III chaperone) in the genome of AIRs, was also assessed. Additionally, the origin, evolutionary history, and genes associated with the NF-independent nodulation pattern in AIRs were explored. The results indicate that horizontal gene transfer events have occurred in AIRs, and three distinct origins of AIRs were estimated: early differentiated AIRs, non-symbiotic Bradyrhizobium, and non-AIRs. In contrast to the significant genetic transformations observed in the second and third groups, the first group of AIRs displays a rich evolutionary history, exhibits high species diversity, and primarily relies on vertical transmission of nitrogen fixation and photosynthetic genes. Overall, the findings provide a fundamental theoretical foundation for gaining a deeper understanding of the phylogeny and genealogy of AIRs.

Internet access for adolescents is becoming more prevalent around the world. Although video game playing has been verified to be negatively related to adolescent academic achievement, the mechanisms underlying this relationship are also unknown. Using a nationally representative sample of adolescents from the China Education Panel Survey (2014-2015), this study aims to explore the parallel mediation roles of self-educational expectation and learning attitude in the link between video game playing and academic achievement, and whether the direct and indirect effects are moderated by parent-child relationship. The results indicate that video game playing in adolescents is both directly and indirectly related to their academic achievement, and self-educational expectation and learning attitude partially mediate this association. Moreover, the results reveal that parent-child relationship moderates the direct association between video game playing and academic achievement as well as the indirect association of video game playing on academic achievement via self-educational expectation, respectively. By showing empirical evidence for the usefulness of social cognitive theory to adolescents' academics in the Internet Age, our research provides a supplement to existing literature.

Consuming a high-fat diet (HFD) is widely recognized to cause obesity and result in chronic brain inflammation that impairs cognitive function. Repetitive transcranial magnetic stimulation (rTMS) has shown effectiveness in both weight loss and cognitive improvement, although the exact mechanism is still unknown. Our study examined the effects of rTMS on the brain and intestinal microecological dysfunction. rTMS successfully reduced cognitive decline caused by an HFD in behavioral assessments involving the Y maze and novel object recognition. This was accompanied by an increase in the number of new neurons and the transcription level of genes related to synaptic plasticity (spindlin 1, synaptophysin, and postsynaptic protein-95) in the hippocampus. It was reached that rTMS decreased the release of high mobility group box 1, activation of microglia, and inflammation in the brains of HFD rats. rTMS also reduced hypothalamic hypocretin levels and improved peripheral blood lipid metabolism. In addition, rTMS recovered the HFD-induced gut microbiome imbalances, metabolic disorders, and, in particular, reduced levels of the microvirus. Our research emphasized that rTMS enhanced cognitive abilities, resulting in positive impacts on brain inflammation, neurodegeneration, and the microbiota in the gut, indicating the potential connection between the brain and gut, proposing that rTMS could be a new approach to addressing cognitive deficits linked to obesity.

General well-being is a positive evaluation of one’s mental health, which is an important topic in mental health. General well-being is fundamental to the positive development of young people. A thorough understanding of the factors that influence happiness have not yet been developed for students at normal universities in Ningxia Province in China. This study examined the mediation effects of time management on the relationship between achievement motivation, self-efficacy, and general well-being among students at normal universities in Ningxia Province in China. Using a random sampling, 163 participants (68 males and 95 females) completed the Achievement Motivation Scale (AMS), General Self-efficacy Scale (GSES), Time Management Questionnaire (TMQ) and General Well-being Scale (GWBS). Achievement motivation, self-efficacy, and time management were positively correlated with general well-being. The mediation effects of time management on the relationships between achievement motivation, self-efficacy, and general wellbeing (0.169 and 0.447) were demonstrated. These results add to the nuanced relationship between self-efficacy, achievement motivation, and general well-being. According to the Achievement Motivation Theory and the Self-efficacy Theory, this study reveals the role of achievement motivation, self-efficacy on general well-being and the mediating effect of time management in the relationship between achievement motivation, self-efficacy, and general well-being. According to the findings, the school can carry out a series of lectures and tutoring activities to enhance students’ achievement motivation, self-efficacy, time management, and general well-being.

Human activities and global climate change have greatly increased nitrogen (N) and phosphorus (P) inputs and altered precipitation patterns in alpine meadows. Functional genes are important indicators of microorganisms that drive the nitrogen cycling process; however, the functional gene responses of soil nitrogen cycling to soil N and P availability and drought remain unclear. Separate or combined treatments of nitrogen and phosphorus fertilization and drought were conducted on the Zoige Wetland in the Qinghai-Tibet Plateau, and the abundances of nitrification functional genes AOA amoA and AOB amoA and denitrification functional genes nirS, nirK, and nosZ were measured to explore the response of functional genes to these treatments. Seven treatments, including control (CK), N addition (N), P addition (P), 50% reduction in precipitation (D), N and P addition (NP), N addition with drought (ND), and NP addition with drought (NPD), were investigated. The results indicated that N application significantly increased AOB amoA abundance, while P application and drought had no significant effects on the abundance of functional genes. The combined treatment of N and P addition and drought increased AOB amoA abundance but did not significantly affect AOA amoA abundance, suggesting that AOB amoA was more responsive to soil N and P availability and moisture change than AOA amoA. However, the abundance of denitrification functional genes was not affected by these treatments. Denitrification functional genes were less sensitive to soil N and P availability and moisture change than nitrification functional genes. The integrated effects of N addition, P addition, and drought did not affect the abundance of the above N cycling functional genes. These results indicate that AOB amoA may play a more critical role in the process of ammonia oxidation than AOA amoA in alpine meadows, and the denitrification genes (nirK, nirS, and nosZ) were better than ammoxidation genes (AOA and AOB) at adapting to the soil environmental changes caused by increasing N and P deposition and drought in alpine meadows.

Soil inorganic carbon (SIC) is an essential component of the soil carbon pool and plays a vital role in the global carbon cycle and climate change. However, few studies have examined the effects of different agronomic practices on the SIC content. This paper aims to study the effects of different agronomic practices on the SIC content in the 0- to 40-cm soil layer of dryland fields; the innovations are intended to explore the relative importance and synergistic effects of different agronomic practices on the SIC content of the plough layer of dryland fields. We screened out 136 peer-reviewed articles worldwide from 1990–2022, with a total of 2612 valid data pairs, using meta-analysis to assess the effects of different agronomic practices on the SIC content of the plough layer of dryland fields. Compared to conventional tillage (CT), both no-tillage (NT) and plastic film mulching (PM) were able to increase the SIC content in the 0- to 40-cm soil layer of dryland fields. NT increased the SIC content by 15.07 ± 3.48%, while PM gradually increased SIC accumulation as the soil layer deepened, with the greatest increase in SIC content in the 30- to 40-cm soil layer at 11.61 ± 5.89%. When organic manure application (M) and straw return mulching (SM) were applied, the SIC content in the 0- to 40-cm soil layer of dryland fields showed a non-significant reduction trend, with the largest changes in SIC content in the 0- to 10-cm soil layer, at 5.23 ± 2% and 4.69 ± 3.53% reductions, respectively. No-tillage straw return (NTS) significantly increased the SIC content in the 0- to 40-cm soil layer of dryland fields by 77.34 ± 5.6%, which was significantly higher than the independent effects of NT and SM, showing a more substantial synergistic effect. Different agronomic practices have different effects on the SIC content of dryland fields in the 0- to 40-cm soil layer, with NTS > NT > PM > SM > M. Among practices, NTS, NT and PM can increase the SIC content of the plough layer of dryland fields; in particular, NTS can increase the SIC content of 0- to 40-cm in dryland fields to the greatest extent through a synergistic effect. At the same time, SM and M showed an insignificant reduction effect.

Increases in nitrogen (N) deposition affect the nitrification and denitrification processes that are regulated by microorganisms. With the aim of understanding what happened to the soil nitrification and denitrification rates under the N application gradients, we set a field experiment treated with N at 6 different rates (0, 2, 4, 8, 16, and 32 g N·m−2·yr−1) in 2014. We determined the physico-chemical properties, abundances and community structures of the nitrifiers and denitrifiers, the net nitrification rate (NNR) and the potential denitrification rate (PDR) of soil samples that were collected in 2020. We found that the abundances of ammonia-oxidizing bacteria amoA (AOB amoA), the sum of ammonia-oxidizing archaea amoA (AOA amoA) and AOB amoA increased with the increase in N application rate, but the abundances of nosZ decreased with that. The microorganisms that dominate the ammonia oxidation process could shift from AOA to AOB under high N application rates. Furthermore, the soil microorganisms respond to the N addition preferentially with the abundance changes rather than the community composition changes. Moreover, the NNR increased with the N input, while the decrease in the PDR was due to the decrease in the pH value caused by high N application. The results also showed that the amoA gene abundance explained most (46.3%) of the variation in the NNR. Moreover, the soil moisture and pH explained 44.0% and 27.1% of the variation in the PDR, respectively. The results demonstrated that the NNR and PDR were mainly explained by functional genes abundances and environmental factors, respectively, in alpine meadow soil under sustained N deposition.

Background: This study aimed to explore the main effects of environmental risk factors as well as their interaction effects with miRNA on the risk of autism spectrum disorder (ASD).Methods: One hundred fifty-nine ASD children (ASD group) and 159 healthy children (control group), aged 2–6 years, were included in this study. ASD diagnoses were based on DSM-5 criteria. The extensive medical and demographic characterization of the two groups were recorded. MicroRNAs (miRNAs) in serum were detected by qRT-PCR.Results: Compared with the control group, the ASD group had significantly higher rates of maternal stress during pregnancy (p < 0.001), maternal drinking during pregnancy (p = 0.006), threatened abortion (p = 0.011), pregnancy-induced hypertension (p = 0.032), gestational diabetes (p = 0.039), maternal anemia during pregnancy (p < 0.001), umbilical cord knot (p < 0.001), neonatal jaundice (p < 0.001), family psychiatric history (p = 0.001), and much lower birth weight (p = 0.012). Furthermore, the ASD group had much lower expression levels of hsa-miR-181b-5p (p < 0.001) and hsa-miR-320a (p < 0.001) and significantly higher levels of hsa-miR-19b-3p (p < 0.001). The interactions of hsa-miR-320a and maternal stress during pregnancy (OR = 39.42, p < 0.001), hsa-miR-19b-3p and neonatal jaundice (OR = 2.44, p < 0.001), and hsa-miR-181b-5p and family psychiatric history (OR = 8.65, p = 0.001) could increase ASD risk.Conclusions: The dysregulation of hsa-miR-181b-5p, hsa-miR-320a, and hsa-miR-19b-3p could interact with environmental factors, such as maternal stress during pregnancy, neonatal jaundice, and family psychiatric history, to impact the risk of ASD.

On the Qinghai–Tibet Plateau, nitrogen (N) cycling, such as nitrification and denitrification, in the alpine meadow soils have been considerably affected by grazing, with possible consequences for nitrous oxide (N2O) emissions. However, there is a lack of understanding about how the potential nitrification activity (PNA) and the potential denitrification activity (PDA) might be affected by the grazing intensity. We collected the soil samples in alpine meadow in the east of the Qinghai–Tibet Plateau that was grazed at different intensities from 2015 in peak growing season 2021. We determined the soil physical and chemical properties, the functional gene abundances of nitrifiers and denitrifiers, and the soil PNA and PDA to explore the relationships between a range of abiotic and biotic factors and the PNA and PDA. We found that the PNA and the nitrifiers were significantly affected by the grazing intensity but that the PDA and the denitrifiers were not. The ammonia-oxidizing archaea (AOA) abundance was highest but the ammonia-oxidizing bacteria (AOB)abundance was lower than the control significantly at the highest grazing intensity. The AOA abundance and the soil NH4+-N explained most of the variation in the PNA. The pH was the main predictor of the PDA and controlled the nirS abundance but not the nirK and nosZ abundances. Overall, the PNA was more responsive to the grazing intensity than the PDA. These findings can improve estimations of the nitrification and denitrification process and N2O emissions in alpine meadow.

Hypotrich ciliates with evolutionary novelties are continually being discovered, challenging the current taxonomic system and attracting increased attention. In the present work, two new urostylid ciliates, Heterobakuella bergeri gen. nov., sp. nov. and Anteholosticha perezuzae sp. nov., isolated from Chinese wetland samples, were identified based on morphology and 18S rRNA gene sequences. Heterobakuella gen. nov. is defined by three frontal cirri, single buccal cirrus, one parabuccal cirrus, midventral complex composed of cirral pairs and one cirral row, one left and two right marginal cirral rows, transverse and pretransverse cirri present, caudal and frontoterminal cirri absent. Heterobakuella can be easily distinguished from the morphologically most similar genus, Apobakuella, mainly by the single buccal cirrus (vs. one buccal cirral row) and one parabuccal cirrus (vs. several parabuccal cirral rows originated from different anlagen). Phylogenetic analyses show that H. bergeri branches within the clade formed by Bergeriella ovata, Monocoronella carnea, Anteholosticha gracilis, and Neourostylopsis spp., rather than the clade represented by Apobakuella. The other species, A. perezuzae, is mainly characterized by a distinctly slender body shape with an average length:width ratio about 7, distinctively shaped biconcave and greenish cortical granules, as well as one or two pretransverse cirri. Phylogenetic analyses indicate the genus Anteholosticha is non-monophyletic.

The ciliate genus Uronychia is a marine group with extremely differentiated cortical and ciliary structures. These structures define its unique evolutionary position in the whole subclass Euplotia. However, to date, few data about the ultrastructure of this genus and related taxa is available. In the present work, a dominant species, Uronychia binucleata, was investigated using scanning electron microscopy and transmission electron microscopy. The findings are as follows: (i) this species lacks the typical alveolar plate in its cortex, whereas the abundant electron-lucent vesicular structures occurred densely; (ii) the subpellicular microtubules form a triad configuration in the dorsal side, while appearing in a single configuration in the ventral side; (iii) the cortical granules are extrusomes, which represent a kind of mucocyst instead of ampules; (iv) two kinetosomes in different rows of one cirrus are linked by the single longitudinal connection; (v) the undulating membrane is highly developed and their insides and outsides are partially covered by the cortical flap; (vi) the single-membrane-bound pharyngeal disks interposed with microtubular sheets, and are distributed in three distinct zones. This first detailed report about the ultrastructural features of the genus Uronychia will be a key to improve the diagnosis and systematics of this widely distributed and ecologically important genus and its family Uronychiidae.

In the present study, based on both morphologic and phylogenetic analyses, a new genus, Monourostylopsis n. gen., and new species, Metaurostylopsis alrasheidi n. sp. as well as a new combination, Monourostylopsis antarctica (Jung et al., 2011) n. comb. (original combination: Metaurostylopsis antarcticaJung et al., 2011), are suggested. The new genus is diagnosed mainly by having three or more frontoterminal cirri, a midventral complex with midventral pairs and a single midventral row, one right marginal row and two or more left marginal rows. The new genus can be easily separated from the morphologically similar genera mainly by having single right marginal row (vs. two or more right marginal rows). Based on live observation and protargol staining, the morphology and morphogenesis of a new species, M. alrasheidi n. sp. isolated from China, were investigated. The new species can be characterized by: two types of cortical granules; about 22 adoral membranelles; three or four frontoterminal, four or five transverse cirri; about eight midventral pairs and a midventral row of three or four unpaired midventral cirri; three or four left and right marginal rows. The main morphogenetic features of Metaurostylopsis alrasheidi n. sp. can be summarized as: (1) the entire parental ciliature, including the oral apparatus, is renewed; (2) the oral primordium of the proter probably originates within a pouch; (3) the oral primordium of the opisthe forms de novo on the cell surface; (4) the anlagen of marginal rows and dorsal kineties are formed intrakinetally, and (5) the fusion of macronuclear nodules results in an irregular branched mass prior to karyokinesis. In the phylogenetic trees, all the available Metaurostylopsis sequences cluster together in a clade with full support (ML/BI: 100/1.00) revealing that the genus is monophyletic within the large group of core urostylids.

Cilia are highly conserved organelles present in almost all types of eukaryotic cells, and defects in cilia structure and/or function are related to many human genetic disorders. Single-celled ciliated protists, which possess diverse types of cilia, are remarkable model organisms for studying cilia structures and functions. Euplotes vannus is a representative ciliate with many intriguing features; for example, it possesses extensively fragmented somatic genomes and a high frequency of + 1 programmed ribosomal frameshifting. However, the molecular mechanisms underlying these remarkable traits remain largely unknown, mainly due to the lack of efficient genetic manipulation tools. Here, we describe the first application of a morpholino-based strategy to knockdown gene expression in E. vannus. Through interfering with the function of two ciliary genes, ZMYND10 and C21ORF59, we showed that these two genes are essential for the ciliary motility and proliferation of E. vannus cells. Strikingly, both ZMYND10- and C21ORF59-knockdown cells developed shorter cilia in the ventral cirri, a special type of ciliary tuft, suggesting a novel role for these genes in the regulation of cilia length. Our data provide a new method to explore gene function in E. vannus, which may help us to understand the functions of evolutionarily conserved cilia-related genes as well as other biological processes in this intriguing model.

Aiming at the problem that the solution of GM (2,1) whitening equation affects its prediction accuracy,a new prediction model dubbed GM (2,1) recursive prediction model of equal dimension new information was pro-posed.The model was deduced from the grey differential equation of GM (2,1) mode,which could avoid solving the second-order whitening equation,solve the problem that the errors between equations and differential equations for conversion,and update the model parameters combining the idea of equal dimension and new information.Both the simulation and analysis of the example demonstrate that the proposed method is more effective and practical.

Background: Over 10 million robotic surgeries have been performed. However, the cost and benefit of robotic surgery need to be evaluated to help hospitals, surgeons, patients, and payers make proper choices, making a health economic analysis necessary. The authors revealed the bibliometric profile in the field of health economics of robotic surgery to prompt research development and guide future studies. Materials and methods: The Web of Science Core Collection scientific database was searched for documents indexed from 2003 to 31 December 2022. Document types, years, authors, countries, institutions, journal sources, references, and keywords were analyzed and visualized using the Bibliometrix package, WPS Office software, Microsoft PowerPoint 2019, VOSviewer software (version 1.6.18), ggplot2, and Scimago Graphica. Results: The development of the health economics of robotic surgery can be divided into three phases: slow-growing (2003-2009), developing (2010-2018), and fast-developing (2019-2022). J.C.H. and S.L.C. were the most active and influential authors, respectively. The USA produced the most documents, followed by China, and Italy. Korea had the highest number of citations per document. Surgical Endoscopy and Other Interventional Techniques accepted most documents, whereas Annals of Surgery, European Urology, and Journal of Minimally Invasive Gynecology had the highest number of citations per document. The Journal of Robotic Surgery is promising. The most-cited document in this field is New Technology and Health Care Costs - The Case of Robot-Assisted Surgery in 2010. The proportion of documents on urology is decreasing, while documents in the field of arthrology are emerging and flourishing. Conclusion: Research on the health economics of robotic surgery has been unbalanced. Areas awaiting exploration have been identified. Collaboration between scholars and coverage with provisions for evidence development by the government is needed to learn more comprehensively about the health economics of robotic surgery. This paper is focused on the tension between Chinese peasants' family capital reproduction strategies and the existing higher education system. Under an institutionalised urban-rural segregation, most rural residents are not entitled to some of the basic social welfare guarantees available to urban residents, such as pensions. As a preparation for the future of their children as well as for themselves, peasants have a strong urge to invest in their children's higher education as their family capital reproduction strategy. However, higher education institutions, without sufficient funding from the government, have to increase enrolment size and charge high tuition fees. Increase in the cost of higher education has strongly impacted on the family economy of rural residents, and the rapid expansion of the system has resulted in an employment crisis for college graduates. Bourdieu's theory of capital reproduction will be applied in the analysis of several cases of rural parents and their children who have participated in higher education. The research eventually leads to the conclusion that, under urban-rural segregation, Chinese peasants are compelled to convert their limited economic capital into academic capital, while the reversibility of this capital conversion is hardly objectively guaranteed.
