Abstract Background Continuous cropping of the same crop leads to land degradation. This is also called the continuous-cropping obstacle. Currently, intercropping tobacco with other crops can serve as an effective strategy to alleviate continuous cropping obstacles. Results In this study, tobacco K326 and insectary floral plants were used as materials, and seven treatments of tobacco monoculture (CK), tobacco intercropped with Tagetes erecta, Vicia villosa, Fagopyrum esculentum, Lobularia maritima, Trifolium repens, and Argyranthemum frutescens respectively, were set up to study their effects on rhizosphere soil chemical properties and composition and structure of rhizosphere soil microbial community of tobacco. The 16 S rRNA gene and ITS amplicons were sequenced using Illumina high-throughput sequencing. tobacco/insectary floral plants intercropping can influence rhizosphere soil chemical properties, which also change rhizosphere microbial communities. The CK and treatment groups tobacco rhizosphere soil microorganisms had significantly different genera, such as tobacco intercropping with T. repens and A. frutescens significantly increased the number of Fusarium and intercropping T. erecta, V. villosa, L. maritima, T. repens, and A. frutescens significantly increased the number of Sphingomonas and unknown Gemmatimonadaceae. Additionally, intercropping T. erecta, V. villosa and L. maritima changed the rhizosphere fungal and bacteria community and composition of tobacco and the positive correlation between tobacco rhizosphere the genera of fungi and bacterial were greater than CK. The pathway of the carbohydrate metabolism, amino acid metabolism, and energy metabolism in rhizosphere bacteria were significantly decreased after continuous cropping. Fungal symbiotic trophic and saprophytic trophic were significantly increased after intercropping V. villosa, L. maritima and plant pathogen and animal pathogen were increased after intercropping T. repens and A. frutescens. Additionally, bacterial and fungal communities significantly correlated with soil chemical properties, respectively. Conclusion This study reveals that intercropping tobacco with insectary floral plants, particularly T. erecta, V. villosa, L. maritima and A. frutescens significantly affects soil chemical properties and alters rhizosphere microbial communities, increasing the abundance of certain microbial genera. Additionally, intercropping enhances pathways related to carbohydrate, amino acid, and energy metabolism in rhizosphere bacteria. These findings suggest that intercropping could provide a promising strategy to overcome challenges associated with continuous tobacco cropping by regulating the rhizosphere environment.

Objective At present, there are relatively few studies on the safety assessment of maintenance and construction of urban rail transit operation lines. In order to reduce the operation safety risk of urban rail transit lines, the safety level of urban rail transit line maintenance construction should be comprehensively evaluated to guide the operation production and management. Method Targeting the 19 maintenance work areas on 4 lines in Shanghai rail transit network, a 4 hierarchy-structured safety effectiveness evaluation index system for urban rail transit line maintenance and construction is established. Based on the actual situation of line maintenance construction indexes, the entropy weight method is improved and combined with the mutation progression theory, and a linear combination evaluation method based on improved entropy weight and mutation progression is proposed as the result. The safety effectiveness of maintenance construction in 19 mainten-ance work areas is evaluated and analyzed with this method. Result & Conclusion The linear combination evaluation method based on improved entropy weight-mutation level passes the consistency test, and the evaluation results are stable and reliable. This method effectively expands the applicability of entropy weight method and mutation level theory in the evaluation of urban rail transit maintenance construction, and realizes the dynamic comprehensive evaluation in the field of rail transit line maintenance construction.

Abstract Due to intermittent renewable energy and fluctuating load demand, distribution networks with renewable distributed generation (DG) installations are more likely to suffer voltage issues and significant power losses. The performance of conservation voltage regulation (CVR) schemes may be adversely affected by the undesirable voltage profile at specific nodes. This paper aims to reduce power losses in CVR‐implemented networks by optimally planning new renewable DGs without changing the existing ones. A scenario‐based optimal renewable DG planning model is proposed with a novel scenario formation method. The uncertainties of load demand and renewables are captured jointly and formed into a finite number of scenarios based on a multivariate Gaussian mixture model (MultiGMM). The locations and capacities of different types of new renewable DGs are optimally planned for CVR performance improvements on power loss saving by aggregating the operation status and probabilities of the scenarios using mixed‐integer non‐linear programming (MINLP). A time‐series simulation is formulated for accuracy verification. The results of case studies show that the proposed model can significantly reduce power losses, active load demand, and reactive load demand. The accuracy of the planning results can be guaranteed with fewer scenarios compared to a widely used classical scenario‐based planning method.

Objective: Illumination of the integrative effects of herbs in a formula is a bottleneck that limits the development of traditional Chinese medicine (TCM). In the present study, we developed a transcriptome-based multi-scale network pharmacology model to explore the combined effects of different herbs. Materials and Methods: First, we curated gene signatures at different biological scales, from the molecular to higher tissue levels, including tissues, cells, pathological processes, biological processes, pathways, and targets. Second, using the Xiexin Tang (XXT) formula as an example, we collected transcriptomic data in response to the treatment of XXT or its three compositive herbs on Michigan cancer foundation7 cells. Third, we linked each herbal drug to different biological scales by calculating the correlation scores between herb-induced gene expression profiles and gene signatures. Finally, the combined mechanisms of the three constituent herbs in XXT were deciphered by comparing their multi-scale effects with those of the formula. Results: The results showed that although XXT or single herbs regulated a large number of signatures on each biological scale, the phenotypic effects of these herbal drugs are concentrated onto the “Blood” tissue, types of hemocytes, and hemorrhagic injury-related pathological processes. At the molecular level, these herbs consistently regulate processes such as the cell cycle and blood coagulation-related pathways, as well as protein targets related to the immunoinflammatory response and blood coagulation, such as proteinase-activated receptor 2, integrin beta-3, inhibitor of nuclear factor kappa-B kinase subunit beta, and coagulation factor XII. The analysis of the combinational modes demonstrated that different herbs can cooperate by acting on the same objects and/or regulating different objects in related functions, and cooperative behaviors change at different biological scales. Conclusions: Our model can dissect the combined effects of herbal formulae from a multi-scale perspective and should be beneficial for the development and exploitation of TCM.

Abstract Conventional macrolide-lincosamide-streptogramin B-ketolide (MLSBK) antibiotics are unable to counter the growing challenge of antibiotic resistance that is conferred by the constitutive methylation of rRNA base A2058 or its G2058 mutation, while the presence of unmodified A2058 is crucial for high selectivity of traditional MLSBK in targeting pathogens over human cells. The absence of effective modes of action reinforces the prevailing belief that constitutively antibiotic-resistant Staphylococcus aureus remains impervious to existing macrolides including telithromycin. Here, we report the design and synthesis of a novel series of macrolides, featuring the strategic fusion of ketolide and quinolone moieties. Our effort led to the discovery of two potent compounds, MCX-219 and MCX-190, demonstrating enhanced antibacterial efficacy against a broad spectrum of formidable pathogens, including A2058-methylated Staphylococcus aureus, Streptococcus pneumoniae, Streptococcus pyogenes, and notably, the clinical Mycoplasma pneumoniae isolates harboring A2058G mutations which are implicated in the recent pneumonia outbreak in China. Mechanistic studies reveal that the modified quinolone moiety of MCX-190 establishes a distinctive secondary binding site within the nascent peptide exit tunnel. Structure-activity relationship analysis underscores the importance of this secondary binding, maintained by a sandwich-like π–π stacking interaction and a water–magnesium bridge, for effective engagement with A2058-methylated ribosomes rather than topoisomerases targeted by quinolone antibiotics. Our findings not only highlight MCX-219 and MCX-190 as promising candidates for next-generation MLSBK antibiotics to combat antibiotic resistance, but also pave the way for the future rational design of the class of MLSBK antibiotics, offering a strategic framework to overcome the challenges posed by escalating antibiotic resistance.

Dielectric metasurfaces have made significant advancements in the past decade for enhancing light–matter interaction at the nanoscale. Particularly, bound states in the continuum (BICs) based on dielectric metasurfaces have been employed to enhance nonlinear harmonic generation. However, conventional nonlinear metasurfaces are typically fixed in their operating wavelength after fabrication. In this work, we numerically demonstrate tunable third harmonic generation (THG) by integrating a dielectric metasurface with the phase-change material Ge2Sb2Te5 (GST). The hybrid phase-change metasurface can support two BICs with different electromagnetic origins, which are transformed into two high-Q quasi-BICs through the introduction of structural asymmetry. The two quasi-BICs are selectively excited by controlling the polarization of incident light, and their wavelengths are tunable due to the phase transition of GST. Notably, the efficiency of THG is significantly enhanced at the fundamental wavelengths corresponding to the two quasi-BICs, and the operating wavelength for THG enhancement can be dynamically tuned through the GST phase transition. Furthermore, the wavelength of THG enhancement can be further tuned by manipulating the polarization of pump light. Additionally, a high-Q analog of electromagnetically induced transparency is numerically achieved through the interaction between a low-Q Mie resonance and a quasi-BIC mode, which also improves the THG efficiency. The high-Q polarization-controlled hybrid phase-change metasurface holds promise for applications in dynamically tunable nonlinear optical devices.

Abstract Background Elucidating the mechanism of fiber transformation underlying microbial metabolism is critical for improving fiber-rich silage digestibility and preserving silage energy for ruminant nutrient absorption. However, few studies have combined quantitative microbial function and transformation products in silage to explain this mechanism. Here, we constructed a workflow to detect the substrates and products of fiber transformation in mixed silage of Sesbania cannabina and sweet sorghum (SS) and combined the absolute quantification 16S rRNA sequencing to reveal this mechanism. Results The synergistic effect of Lactobacillus cocktail and cellulase (LC) simplified the microbial diversity and minimized the microbial quantity, making Lentilactobacillus buchneri the dominant species in SS silage. As a result, the LC-treated silage had greater lactic acid content, lower pH value, and less NH3-N content. The indigestible fibers were significantly decreased due to the synergistic effect of the Lactobacillus cocktail and cellulase. Changes in microbial structure during ensiling also resulted in metabolic alterations. The increased levels of microbial enzymes, including β-glucosidase and sucrose phosphorylase, involved in starch and sucrose metabolism led to the enrichment of monosaccharides (including glucose, xylose, mannose, galactose, ribose, rhamnose, and arabinose) in the LC-treated silage. We found that L. buchneri was positively associated with β-glucosidase and sucrose phosphorylase, reflecting the crucial contribution of L. buchneri to fiber decomposition in SS silage. Conclusion Using an absolute quantitative microbiome, we found that LC treatment decreased the microbial biomass in SS silage, which in turn promoted the energy preservation in the SS silage. The cooperative interaction of the Lactobacillus cocktail and cellulase improved the fiber decomposition and in vitro dry matter digestibility rate by changing the microbiome structure and function in the SS silage, providing guidance and support for future fiber-rich silage production in the saline-alkaline region. BackgroundThe relationship between white matter hyperintensities (WMH) and the core features of Alzheimer’s disease (AD) remains controversial. Further, due to the prevalence of co-pathologies, the precise role of WMH in cognition and neurodegeneration also remains uncertain.MethodsHerein, we analyzed 1803 participants with available WMH volume data, extracted from the ADNI database, including 756 cognitively normal controls, 783 patients with mild cognitive impairment (MCI), and 264 patients with dementia. Participants were grouped according to cerebrospinal fluid (CSF) pathology (A/T profile) severity. Linear regression analysis was applied to evaluate the factors associated with WMH volume. Modeled by linear mixed-effects, the increase rates (Δ) of the WMH volume, cognition, and typical neurodegenerative markers were assessed. The predictive effectiveness of WMH volume was subsequently tested using Cox regression analysis, and the relationship between WMH/ΔWMH and other indicators such as cognition was explored through linear regression analyses. Furthermore, we explored the interrelationship among amyloid-β deposition, cognition, and WMH using mediation analysis.ResultsHigher WMH volume was associated with older age, lower CSF amyloid-β levels, hypertension, and smoking history (all p ≤ 0.001), as well as cognitive status (MCI, p

Lassa virus (LASV), a risk-group 4 pathogen, must be handled in biosafety level-4 (BSL-4) conditions, thereby limiting its research and antiviral development. Here, we developed a novel LASV reverse genetics system which, to our knowledge, is the first to study the complete LASV life cycle under BSL-2 conditions. Viral particles can be produced efficiently when LASV minigenomic RNA harbouring minimal viral cis-elements and reporter genes is transfected into a helper cell line stably expressing viral NP, GP, Z and L proteins. The resulting defective virions, named LASVmg, can propagate only in the helper cell line, providing a BSL-2 model to study the complete LASV life cycle. Using this model, we found that a previously reported cellular receptor α-dystroglycan is dispensable for LASVmg infection. Furthermore, we showed that ribavirin can inhibit LASVmg infection by inducing viral mutations. This new BSL-2 system should facilitate studying the LASV life cycle and screening antivirals.

ABSTRACTIndole in the gut is formed from dietary tryptophan by a bacterial tryptophan-indole lyase. Indole not only triggers biofilm formation and antibiotic resistance in gut microbes but also contributes to the progression of kidney dysfunction after absorption by the intestine and sulfation in the liver. As tryptophan is an essential amino acid for humans, these events seem inevitable. Despite this, we show in a proof-of-concept study that exogenous indole can be converted to an immunomodulatory tryptophan metabolite, indole-3-lactic acid (ILA), by a previously unknown microbial metabolic pathway that involves tryptophan synthase β subunit and aromatic lactate dehydrogenase. Selected bifidobacterial strains converted exogenous indole to ILA via tryptophan (Trp), which was demonstrated by incubating the bacterial cells in the presence of (2-13C)-labeled indole and l-serine. Disruption of the responsible genes variedly affected the efficiency of indole bioconversion to Trp and ILA, depending on the strains. Database searches against 11,943 bacterial genomes representing 960 human-associated species revealed that the co-occurrence of tryptophan synthase β subunit and aromatic lactate dehydrogenase is a specific feature of human gut-associated Bifidobacterium species, thus unveiling a new facet of bifidobacteria as probiotics. Indole, which has been assumed to be an end-product of tryptophan metabolism, may thus act as a precursor for the synthesis of a host-interacting metabolite with possible beneficial activities in the complex gut microbial ecosystem.

Abstract Vector-like quarks (VLQs) are a common feature of many scenarios of new physics beyond the Standard Model (SM), which generally decay into a SM third-generation quark with a SM gauge boson or a Higgs boson. The presence of a new exotic decay mode of VLQs will reduce the branching ratios of these standard decay modes and thus relax the current mass exclusion limits from LHC experiments. Based on a model-independent framework, we investigate the prospect of discovering the pair production of the weak-singlet VLQ-B at the future 3-TeV Compact Linear Collider (CLIC), by focusing on the final states including one Z boson and four b-jets via two types of modes: $$Z\rightarrow \ell ^{+}\ell ^{-}$$ Z → ℓ + ℓ - and $$Z\rightarrow \nu {\bar{\nu }}$$ Z → ν ν ¯ . By performing a rapid detector simulation of the signal and background events, and considering the initial state radiation and beamstrahlung effects, the exclusion limit at the 95% confidence level and the $$5\sigma $$ 5 σ discovery prospects are respectively obtained on the branching ratio of $$B\rightarrow bZ$$ B → b Z and the VLQ-B masses at the future 3-TeV CLIC with an integrated luminosity of 5 ab $$^{-1}$$ - 1 .

Abstract Autophagy receptor NDP52 triggers bacterial autophagy against infection. However, the ability of NDP52 to protect against viral infection has not been established. We show that NDP52 binds to envelope proteins of hepatitis B virus (HBV) and triggers a degradation process that promotes HBV clearance. Inactivating NDP52 in hepatocytes results in decreased targeting of viral envelopes in the lysosome and increased levels of viral replication. NDP52 inhibits HBV at both viral entry and late replication stages. In contrast to NDP52-mediated bacterial autophagy, lysosomal degradation of HBV envelopes is independent of galectin 8 and ATG5. NDP52 forms complex with Rab9 and viral envelope proteins and links HBV to Rab9-dependent lysosomal degradation pathway. These findings reveal that NDP52 acts as a sensor for HBV infection, which mediates a unique antiviral response to eliminate the virus. This work also suggests direct roles for autophagy receptors in other lysosomal degradation pathways than canonical autophagy.

Eight new transition-based coordination compounds were hydrothermally fabricated using metal chlorides (M = Co(II), Ni(II), Mn(II), Zn(II) and Cd(II)) with main ligand 4′-(carboxymethoxy)-[1,1′-biphenyl]-4-carboxylic acid (H2cbca) as well as coligands (phen: 1,10-phenanthroline; 2,2′-bipy: 2,2′-bipyridine; 4,4′-bipy: 4,4′-bipyridine or dpe: 1,2-di(4-pyridyl)ethane). The successful synthesis obtained one 0D dimer, three 1D and four 2D coordination polymers, were formulated as [Cd(μ3-cbca)(H2O)]n (1), [Zn(μ-cbca)(phen)]n (2), [Mn(μ3-cbca)(phen)(H2O)]n (3), [Cd(μ3-cbca)(2,2′-bipy)]n (4), [Co(µ-cbca)(μ-4,4′-bipy)(H2O)2]n (5), [Co(μ3-cbca)2(μ-dpe)0.5(H2O)]n (6), [Ni2(cbca)2(μ-dpe)(H2O)8] (7), and [Zn(μ3-cbca)(μ-dpe)0.5(H2O)]n (8). Comprehensive characterisations were performed through standard methods. Topological types and catalytic performance of these compounds were researched. Especially, polymer 2 serves as a stable, highly effective, and recyclable heterogeneous catalyst for the Knoevenagel condensation and cyanosilylation of benzaldehydes, giving a high condensation product yields (up to 100 %) under optimized conditions. Various reaction parameters, substrate scope, and catalyst recycling were also studied. The present work explores the application of H2cbca as the flexible dicarboxylate linker for the preparation of functional coordination polymers.

A new species of Anoectochilus (Orchidaceae) from Guangxi, China, A. zhongshanensis, is described here, which was identified based on phylogenetic studies adopting combined plastid markers (rbcL-matK-trnL-F), morphological observation and chemical analysis. Molecular phylogenetic results support the systematic status of A. zhongshanensis as a new species in Anoectochilus genus. Morphologically, this new species is similar to A. zhejiangensis and A. malipoensis, but differs by its characteristic labellum and column, including the hastate or scalpel-shaped lobes of epichile, forward curved and pinnately divided cristate lobes at both sides of the mesochile and inverted triangle column wings. Furthermore, HPLC-ELSD analysis of these three species revealed the interesting chemotaxonomic difference that the principle and characteristic lactone glycoside in this new species was kinsenoside, rather than its diastereoisomer, goodyeroside A, a major glycoside in A. zhejiangensis and A. malipoensis.

Abstract Background To compare short-term and long-term clinical effects of modified overlap anastomosis and conventional incision-assisted anastomosis for laparoscopic total gastrectomy. Methods This retrospective cohort study included patients with gastric cancer admitted to the Second Affiliated Hospital of Fujian Medical University from January 2016 to March 2020. Quality of life, intraoperative and postoperative conditions were analyzed. Results Compared with the conventional assisted group, the modified overlap group showed a shorter auxiliary incision, milder postoperative pain, shorter time to the first postoperative anal exhaust, shorter time to the first postoperative liquid food intake, and shorter postoperative stay. There were no differences between the two groups regarding operation time, esophagus-jejunum anastomosis time, intraoperative blood loss, number of lymph nodes dissected, and length of the upper incision margin. There were no differences between the two groups regarding postoperative early and late complications. There were no differences between the two groups regarding the QLQ-C30 scale three years after the operation. The scores of the QLQ-STO22 scale 3 years after the operation showed significantly lower scores for dysphagia and feeding limit in the modified overlap group than those in the conventional assisted anastomosis group. There was no recurrence in the modified overlap group but one patient in the conventional assisted group. Conclusions Patients undergoing totally laparoscopic total gastrectomy with modified overlap anastomosis have better minimal invasiveness and faster post-operative recovery than conventional incision-assisted anastomosis.
